One-pot bottom-up fabrication of biocompatible PEGylated WS2 nanoparticles for CT-guided photothermal therapy of tumors in vivo

Jiang-Tao Wang,Wei Zhang,Wan-Bi Wang,Yong-Juan Wu,Ling Zhou,Feng Cao

Background Tungsten disulfide (WS 2 ), which enjoyed a good potential to be a promising clinical theranostic agent for cancer treatment, is still subject to the tedious synthesis procedure. Methods Here, we reported a one-pot ‘bottom-up’ hydrothermal strategy for the fabrication of PEGylated WS 2 nanoparticles (NPs). The WS 2 -PEG nanoparticles were characterized systematically. The CT imaging and photothermal therapy against tumor as well as biosafety in vitro and in vivo were also investigated. Results The obtained WS 2 -PEG NPs enjoyed obvious merits of good solubility and favorable photothermal performance. WS 2 -PEG NPs exhibited desirable photothermal ablation ability against cancer cells and cancer cell-bearing mice in vitro and in vivo. MTT assay and histological analysis demonstrated the low cytotoxicity and biotoxicity of WS 2 -PEG NPs, providing a valid biosafety guarantee for the coming biomedical applications. In addition, thanks to the obvious X-ray attenuation of W atom, the WS 2 -PEG NPs can also be served as a favorable contrast agent for CT imaging of tumors. Conclusion WS 2 -PEG NPs has enjoyed a good potential to be a promising clinical CT-guided photothermal therapeutic agent against cancers.

Active targeted dual-modal CT/MR imaging of VX2 tumors using PEGylated BaGdF5 nanoparticles conjugated with RGD

Tao Wang,Guorong Jia,Chao Cheng,Qiuhu Wang,Xiao Li,Yanyan Liu,Chaofan He,Luguang Chen,Gaofeng Sun,Changjing Zuo

Multimodal imaging can improve the accuracy of cancer diagnosis by combining two or more imaging modalities into one system. In this study, PEGylated BaGdF5 nanoparticles (NPs) were synthesized by a hydrothermal method, and the formed PEG-BaGdF5 NPs showed good X-ray attenuation properties and positive MR contrast effects in vitro. Considering that the overexpression of integrin αvβ3 in the endothelial cells of angiogenic tumor vasculature is universal, RGD peptide, which can recognize and bind integrin αvβ3 specifically, was connected to the surface of PEG-BaGdF5 NPs. The resulting RGD-PEG-BaGdF5 NPs had good biocompatibility and a long half-life in vivo. In VX2 tumor-bearing mice, the HU values and MR signal enhancement of tumor sites in the targeted group were both higher than that in the non-targeted group after intravenous injection of RGD-PEG-BaGdF5 NPs or PEG-BaGdF5 NPs, respectively. Therefore, the RGD-PEG-BaGdF5 NPs may be used as dual-modal CT/MR contrast agents of different types of solid tumors via an active RGD-mediated vasculature targeting pathway.

Composite silica coated gold nanosphere and quantum dots nanoparticles for X-ray CT and fluorescence bimodal imaging

Ji-Tao Song,Xiao-Quan Yang,Xiao-Shuai Zhang,Dong-Mei Yan,Ming-Hao Yao,Meng-Yao Qin,Yuan-Di Zhao

In this study, silica coated Au nanospheres (Au@SiO2) were prepared by a reverse microemulsion method; subsequently, a layer of fluorescent quantum dots (QDs) were adsorbed onto it and then it was coated with silica again. After modifying with PVP, the composite silica coated gold nanosphere and quantum dots nanoparticle (Au@SiO2-QDs/SiO2-PVP) was obtained. This composite structure contained Au and QDs, and it could be used for contrast-enhanced X-ray CT imaging and fluorescence imaging. Characterization showed that the composite nanoparticle had good dispersity, a high fluorescence intensity and a good effect of X-ray absorption, and it was suitable for using as a bimodal imaging probe.

Synthesis of PEGylated BaGdF5 Nanoparticles as Efficient CT/MRI Dual-modal Contrast Agents for Gastrointestinal Tract Imaging

Feng-Zhi CUI,Jian-Hua LIU,Yang LIU,Bi-Ying YUAN,Xue GONG,Qing-Hai YUAN,Ting-Ting GONG,Lei WANG

Molecular imaging (MI) techniques have extremely significant effect on the diagnosis and prognosis of diseases, such as gastrointestinal (GI) tract-related diseases. Furthermore, nanomaterials as contrast agents have attracted a great deal of research attention, due to their rational fabrication and biomedical. Herein, we reported a facile process to synthesis PEGylated BaGdF 5 nanoparticles via a one-pot solvothermal route as CT/MRI dual-modal contrast agents for in vivo imaging of GI tract. Compared to the commercially used iodine contrast agents, the well-prepared nanoparticles exhibited enhancement in computed tomography (CT) imaging. In the presence of Gd, nanoparticles could also be used in magnetic resonance imaging (MRI). PEGylated BaGdF 5 nanoparticles were characterized by transmission electron microscopy (TEM), X-ray powder diffraction analysis (XRD), Fourier transform infrared (FT-IR) spectroscopy and thermogravimetric analysis (TGA). Besides, MTT assay revealed that the prepared nanoparticles proved to be low toxicity and high biocompatibility for Hela cells. Hemolytic assay illustrated nanoparticles possess favorable hemocompatibility. H & E (Hematoxylin and Eosin) histological staining assessed long-term toxicity of oral PEGylated BaGdF 5 nanoparticles, and the results showed that the nanoparticles had overall safety for GI tissue with comparatively high compatibility. Based on above results, PEGylated BaGdF 5 nanoparticles were admired multi-functional contrast agents to apply in CT and MRI imaging with prominent prospects.

Stimuli responsive PEGylated bismuth selenide hollow nanocapsules for fluorescence/CT imaging and light-driven multimodal tumor therapy

Lihong Sun,Qian Li,Lei Zhang,Huihui Chai,Ling Yu,Zhigang Xu,Yuejun Kang,Peng Xue

Stimuli-responsive therapeutic nanosystems exhibit enhanced selectivity and higher biosafety for cancer theranostics via recognizing exogenous or endogenous tumor-associated factors. Herein, we developed a multifunctional nanocomplex (Bi2Se3@PEG/DOX/Ce6 nanocapsules, or BPDC NCs in brief) which was constructed by loading chlorin e6 (Ce6) and doxorubicin (DOX) into PEGylated hollow bismuth selenide nanocapsules. Upon administration of BPDC NCs, composite laser irradiation can effectively activate the local hyperthermia generation and the yield of cytotoxic reactive oxygen species (ROS). In another aspect, on-demand drug release can be triggered in a mild acidic tumor microenvironment or by thermal shock. Moreover, imaging and navigation with respect to infrared thermography, computed tomography (CT) and fluorescence imaging may potentially monitor the biodistribution of BPDC NCs, thanks to the local hyperthermia generation, fluorescence emission of Ce6 and high Z-element of bismuth. Finally, we demonstrated tumor site-specific photothermal therapy (PTT), photodynamic therapy (PDT) and chemotherapeutic effects for highly efficacious tumor suppression with minimized systemic toxicity. Taken together, these findings indeed provide insights that can broaden the application of Bi2Se3 nanocapsules for cancer management and precision medicine.

Large-scale synthesis of PEGylated lutetium hydroxycarbonates as nanoparticulate contrast agents for X-ray CT imaging

Zhaogui Ba,Yumin Zhang,Junpei Wei,Jiwu Han,Zhenqiang Wang,Guangrui Shao

Nanoparticulate contrast agents have drawn considerable attention and interest because of their potential application in medical diagnosis and prognosis. In the present study, we designed and constructed high-performance nanoparticulate contrast agents based on PEGylated lutetium hydroxycarbonate nanoparticles (PEG-LuNPs) for X-ray computed tomography imaging, which were synthesized via a green and large-scale route. Under the daily clinical voltage, our PEG-LuNPs provided much more enhanced contrast than the routinely used iodine-based molecules. More importantly, PEG-LuNPs could act as liver-targeted contrast agents for the further detection of hepatic metastases. Both in vitro and in vivo toxicity studies indicated that these nanoparticles possess extremely high biocompatibility, revealing their overall safety. Based on these results, PEG-LuNPs with intrinsic physicochemical properties and excellent imaging capability demonstrated a useful nanoplatform for biomedical applications.

A novel CT contrast agent for intestinal-targeted imaging through rectal administration

Jingyao Zhu,Hao Weng,Shichen Xie,Jiejun Cheng,Jun Zhu

In this study, a novel CT contrast agent used by rectal administration is developed for targeting intestinal imaging. Iopamidol, an iodinated contrast agent, is loaded in chitosan (CS) nanospheres modified by Anti-5-HT3R (AH) antibody. The obtained AH-CS-I nanospheres (AH-CS-I Ns) would combine to 5-HT3 receptors highly expressed on the gastrointestinal mucosal, enhancing the intestinal-targeting ability of the contrast agent. The AH-CS-I Ns were administered by the rectal route for intestinal CT imaging, and FITC-labeled AH-CS-I Ns were prepared for investigating the in vivo distribution of the contrast agent. As a result, obvious contrast enhancement could still be observed at 6 h post administration because of the poorly absorption of enteral AH-CS-I Ns. Unlike the intravascularly administered agents, AH-CS-I Ns would not accumulate in the kidney and induce adverse reactions. Therefore, this technology has potential applications in the examination of intestinal diseases and could reduce the side effect of commercial iopamidol.

A novel surface modification strategy of CdTe/CdS QDs and its application for sensitive detection of ct-DNA

Wanjun Liang,Zhengqing Liu,Shaopu Liu,Jidong Yang,Youqiu He

A kind of novel Glyp-functionalized-CdTe/CdS Quantum dots (QDs) was successfully synthesized by coating Glyp on the surface of the CdTe/CdS QDs. The fluorescence emission of the modified QDs was enhanced greatly but with a low resonance Rayleigh scattering (RRS). When it interacted with calf thymus DNA (ct-DNA), the fluorescence intensity of the system was obviously quenched and the RRS was enhanced, both in good linear relations. Based on the above, a novel fluorescence biosensor for simple, sensitive and selective detection of ct-DNA was developed. Under the optimized conditions, the linear ranges of the functionalized QDs fluorescence intensity and RRS intensity versus the concentration of ct-DNA were 0.109–70   μg   mL −1 and 0.482–90   μg   mL −1 , respectively. The corresponding detection limits by fluorescence quenching and by RRS enhancing methods were 0.0327   μg   mL −1 and 0.146   μg   mL −1 , respectively. The proposed method was applied to detect ct-DNA in synthetic sample with satisfactory results.

Encapsulating tantalum oxide into polypyrrole nanoparticles for X-ray CT/photoacoustic bimodal imaging-guided photothermal ablation of cancer

Yushen Jin,Yanyan Li,Xibo Ma,Zhengbao Zha,Liangliang Shi,Jie Tian,Zhifei Dai

A nanotheranostic agent has been readily fabricated by encapsulating tantalum oxide (TaO x ) nanoparticles (NPs) into polypyrrole (PPy) NPs via a facile one-step chemical oxidation polymerization for bimodal imaging guided photothermal ablation of tumor. It was proved that the obtained composite nanoparticles (TaO x @PPy NPs) with an average diameter around 45 nm could operate as an efficient bimodal contrast agent to simultaneously enhance X-ray CT and photoacoustic (PA) imaging greatly in vivo . Systemically administered TaO x @PPy NPs could passively accumulate at the tumor site during the blood circulation, which was proved by both CT and PA imaging. In addition, the in vivo therapeutic examinations showed that TaO x @PPy NPs exhibited significant photothermal cytotoxicity under near infrared laser irradiation. The tumor growth inhibition was evaluated to be 66.5% for intravenously injection and 100% for intratumoral injection, respectively. This versatile agent can be developed as a smart and promising nanoplatform that integrates multiple capabilities for both accurate diagnosing and precise locating of cancerous tissue, as well as effective photoablation of tumor.

One-pot synthesis of flower-like Bi2S3 nanoparticles for spectral CT imaging and photothermal therapy in vivo

Cai Zhang,Li Zhou,Jing Zhang,Rui Dai,Pengrui Zhuang,Zhaoxiang Ye

Spectral computed tomography (CT) imaging agents are the key to spectral CT imaging with an unprecedented signal-to-noise ratio in vivo. However, there is still extreme lack of high-performance contrast agents. Herein, we present a facile and green strategy for the fabrication of uniform flower-like Bi2S3 nanoparticles (NPs) for spectral CT imaging and photothermal therapy in vivo for the first time. High-quality Bi2S3 NPs are obtained by simply heating the mixture of Bi(NO3)3 and glutathione in ethylene glycol without the need for adding toxic agents and inert gas protection. The proposed Bi2S3 NPs exhibit good monodispersity, high Bi content (60%), favorable water solubility, strong absorption in near-infrared windows, good biocompatibility and excellent CT and spectral CT imaging abilities due to the high K-edge energy of Bi (91 keV). Not only the accurate gastrointestinal tract spectral CT imaging but also the evaluation of the effect of intestinal obstruction surgery was achieved using the Bi2S3 NPs. Then effective tumor photothermal therapy in vivo was realized. This study proposes a facile and green method for the synthesis of high-quality Bi2S3 NPs, and proves the promising potential of Bi2S3 nanomaterial as a high-performance spectral CT imaging agent.

Au/Mn nanodot platform for in vivo CT/MRI/FI multimodal bioimaging and photothermal therapy against tongue cancer

Zhe Yang,Yueqi Zhao,Yang Li,Lei Song,Yangliu Lin,Kaimeng Liu,Yujia Zhang,Andrei V. Zvyagin,Linan Fang,Yuanqing Sun,Bai Yang,Quan Lin

Surgical resection is the main method for oral tongue squamous cell carcinoma (OTSCC) treatment. However, the oral physiological function and aesthetics may be seriously damaged during the operation with a high risk of recurrence. Therefore, it is important to develop an alternative strategy with precise guidance for OTSCC treatment. Herein, multifunctional Au/Mn nanodots (NDs) are designed and synthesized. They can perform multimodal bioimaging, including computed tomography (CT) and magnetic resonance imaging (MRI) simultaneously, and exhibit bright near-infrared fluorescence imaging (FI) for navigation, and even integrate photothermal therapy (PTT) property. The localization of OTSCC relies on visual and tactile cues of surgeons while lacking noninvasive pretreament labeling and guidance. Au/Mn NDs provide CT/MRI imaging, giving two means of accurate positioning pretherapy. Meanwhile, the fluorescence of the Au/Mn NDs in the near-infrared region (NIR) is beneficial for noninvasive labeling and intuitive observation with the naked eye to determine the tumor boundary during PTT. Further, Au/Mn NDs showed excellent results in ablating tumors in vivo. Above all, the Au/Mn NDs provide a key platform for multimodal bioimaging and PTT in a single nanoagent, which demonstrated attractive performance for OTSCC treatment.

Core–shell BaYbF5:Tm@BaGdF5:Yb,Tm nanocrystals for in vivo trimodal UCL/CT/MR imaging

Peng Zhang,Yangyang He,Jianhua Liu,Jing Feng,Zhiqiang Sun,Pengpeng Lei,Qinghai Yuan,Hongjie Zhang

Lanthanide-doped nanocrystals have been researched extensively and used for bioimaging because of their optical properties, magnetic properties and X-ray absorption. Core–shell-structured lanthanide-doped nanocrystals have been developed and characterized by TEM and XRD analysis. The nanocrystals are composed of BaYbF5:0.5% Tm as the core and BaGdF5:20% Yb, 0.5% Tm as the shell. Apart from characterization of the nanocrystals, evaluation of both their cytotoxicity via MTT assays and their long-term toxicity via histological analysis showed that their cytotoxicity was low, indicating the possibility of further in vivo imaging. This work combined the functions of trimodal imaging into one nanoplatform and then UCL, CT, and MR imaging with core–shell-structured nanocrystals were investigated both in vitro and in vivo. Taking into consideration its structural characteristics and trimodal imaging abilities, it is expected that the developed multifunctional nanoplatform may be potentially useful for diagnosing diseases at an early stage.
