Photodynamic therapy (PDT) and ferroptosis show significant potential in tumor treatment. However, their therapeutic efficacy is often hindered by the oxygen-deficient tumor microenvironment and the challenges associated with efficient intracellular drug delivery into tumor cells. Toward this end, this work synthesized perfluorocarbon (PFC)-modified Pluronic F127 (PFC-F127), and then exploits it as a carrier for codelivery of photosensitizer Chlorin e6 (Ce6) and the ferroptosis promoter sorafenib (Sor), yielding an oxygen self-supplying nanoplatform denoted as Ce6-Sor@PFC-F127. The PFCs on the surface of the micelle play a crucial role in efficiently solubilizing and delivering oxygen as well as increasing the hydrophobicity of the micelle surface, giving rise to enhanced endocytosis by cancer cells. The incorporation of an oxygen-carrying moiety into the micelles enhances the therapeutic impact of PDT and ferroptosis, leading to amplified endocytosis and cytotoxicity of tumor cells. Hypotonic saline technology was developed to enhance the cargo encapsulation efficiency. Notably, in a murine tumor model, Ce6-Sor@PFC-F127 effectively inhibited tumor growth through the combined use of oxygen-enhanced PDT and ferroptosis. Taken together, this work underscores the promising potential of Ce6-Sor@PFC-F127 as a multifunctional therapeutic nanoplatform for the codelivery of multiple cargos such as oxygen, photosensitizers, and ferroptosis inducers.

Prostate cancer (PCa) is the most common malignant tumor of male genitourinary system, ranking second among all male malignant tumors and showing a trend toward younger age. However, for metastatic prostate cancer, commonly used androgen deprivation therapy combined with docetaxel chemotherapy or neoendocrine therapy is prone to complications (incontinence, erectile dysfunction, and rectal damage), resulting in reduced quality of life or serious side effects for patients. Therefore, the designed GO-DAA-Ce6 nanoparticles were constructed to couple graphene oxide and Ce6 via diacylhydrazine adipate (DAA) as a bridge. The combination therapy of PTT and PDT is made possible by the outstanding photothermal conversion ability of GO and the high singlet oxygen generation of Ce6. GO-DAA-Ce6 demonstrated low toxicity and great biocompatibility in cytotoxicity and hemolysis assays. According to the outcomes of in vitro anti-tumor tests, GO-DAA-Ce6 greatly outperformed the photothermal and photodynamic efficacy of GO alone and Ce6 alone in terms of tumor suppression rates. The findings of the anticancer mechanism study point to the possibility that GO-DAA-Ce6 may cause cancer cells to undergo apoptosis and necrosis by initially damaging suborganelles such mitochondria. The straightforward design of the photothermal and photodynamic collaboration platform kills prostate cancer effectively and offers a general method for the treatment of various conditions including inflammation and skin infections.

Accurate diagnosis of cancer cells directly affects the clinical treatment of cancer and can significantly improve the therapeutic effect of cancer patients. Cancer cells have a unique microenvironment with a large amount of peroxide inside, effectively differentiated from relevant microenvironment normal cells. Therefore, designing the high-sensitive probes to recognize and distinguish the special physiological microenvironment of cancer cells can shed light on the early diagnosis of cancers. In this article, we design and construct a fluorescence (FL) contrast agent for cancer cell recognition and imaging analysis. Firstly, luminol-gold NPs (Lum-AuNPs) have been initially built, and then successfully loaded with the fluorescent receptor Chlorin e6 (Ce6) to prepare the luminescent nanoprobes (Ce6@Lum-AuNPs) with green synthesis, i.e., with biocompatible agents and mild temperature. The as-prepared fluorescent Ce6@Lum-AuNPs can efficiently and sensitively realize FL bioimaging of cancer cells. The relevant bio-sensing mechanism pertains to the presence of hypochlorite (ClO−); hydrogen peroxide (H2O2) in cancer cells could readily interact with luminol to produce chemiluminescence, which can activate the Ce6 component to emit near-infrared (NIR) FL. Therefore, this raises the possibility of utilizing the Ce6@Lum-AuNPs as efficient fluorescent nanoprobes for promising cancer early diagnosis and other relevant disease bioanalysis. Keywords:hydrogen peroxide-triggered fluorescent nanoclusters;Ce6@Lum-AuNPs;FL nanoprobes;cancer cell bioimaging

Photomediated cancer therapy, mainly including photothermal (PT) therapy (PTT) and photodynamic therapy (PDT), has attracted tremendous attention in recent years thanks to its noninvasive and stimuli-responsive features. The single mode of PTT or PDT, however, has obvious drawbacks, either requiring high-power laser irradiation to generate enough heat or only providing limited efficacy due to the hypoxia nature inside tumors. In addition, the reported synergistic PTT/PDT generally utilized two excitation sources to separately activate PTT and PDT, and the problem of high-power laser irradiation for PTT was still not well solved. Herein, a new concept, loading a small amount of photosensitizers onto a PTT agent (both of them can be triggered by a single-near-infrared (NIR) laser), was proposed to evade the shortcomings of PTT and PDT. To validate this idea, minute quantities of photosensitizer chlorin e6 (Ce6) (0.56% of mass) were anchored onto amino-rich red emissive carbon dots (RCDs) that possess superior photothermal (PT) character under 671 nm NIR laser (PT conversion efficiency to be 46%), and meanwhile the PDT of Ce6 can be activated by this laser irradiation as well. The findings demonstrate that Ce6-modified RCDs (named Ce6-RCDs) offer much higher cancer therapy efficacy under a reduced laser power density (i.e., 0.50 W cm–2 at 671 nm) in vitro and in vivo than the equivalent RCDs or Ce6 under the same irradiation conditions. Besides, the Ce6-RCDs also exhibit multimodal imaging capabilities (i.e., fluorescence (FL), photoacoustic (PA), and PT), which can be employed for guidance of the phototherapy process. This study suggests not only a strategy to enhance cancer phototherapy efficacy but also a promising candidate (i.e., Ce6-RCDs) for multimodal FL/PA/PT imaging-guided and single-NIR-laser-triggered synergistic PTT/PDT for cancers by a reduced irradiation power.

Metastasis is the main cause of treatment failure in breast cancer, and integrated phototheranostics is a promising strategy to achieve both precision theranostics and metastasis inhibition. In this work, a multifunctional phototheranostic nanoprobe composed of chlorin e6 (Ce6)-conjugated and polydopamine (PDA)-coated gold nanostars (AuNSs) was synthesized for simultaneous photoacoustic (PA) imaging, photothermal therapy (PTT) and photodynamic therapy (PDT). Under the irradiation of near infrared laser, AuNSs@PDA showed enhanced photothermal conversion and amplified PA imaging performance, compared with single AuNSs. By the covalent conjugation of Ce6, the AuNSs@PDA-Ce6 nanoprobe showed robust stability and excellent singlet oxygen (1O2) generation ability. Under the combination of PTT/PDT, the AuNSs@PDA-Ce6 nanoprobes significantly reduced the growth of 4T1 tumors and suppressed their lung metastasis. All the results demonstrated the considerable potential of AuNSs@PDA-Ce6 phototheranostic nanoprobes for precision theranostics and metastasis inhibition of breast cancer.

Many drug controlled release methods have been integrated in multifunctional nanoparticles, such as pH-, redox-, temperature-, enzyme-, and light-responsive release. However, few report is associated with the ROS responsive drug controlled release. Herein, a thioketal linker-based ROS responsive drug (camptothecin conjugated with thioketal linker, abbreviated as TL-CPT) was prepared and the thioketal linker could be cleaved by ROS(reactive oxygen species). To achieve cancer simultaneous optical imaging, photodynamic therapy and chemotherapy, the photosensitizer Chlorin e6(Ce6), TL-CPT and carboxyl-mPEG were loaded on the upconversion nanoparticles (UCNPs), which were named as Ce6-CPT-UCNPs. Under 980 nm laser irradiation, Ce6-CPT-UCNPs emitted a narrow emission band at 645-675 nm which was overlapped with Ce6 absorption peak. Ce6 absorbed the light to produce ROS, which was used for photodynamic therapy and to cleave the thioketal linker in Ce6-CPT-UCNPs to release camptothecin for chemotherapy. Meanwhile, Ce6 absorbed the light, was used for near-infrared fluorescence imaging. The in vivo biodistribution studies showed that the prepared nanoparticles had high orthotopic lung cancer targeting efficiency. The in vivo therapeutic results demonstrated that NCI-H460 lung cancers could be completely eliminated by combining chemo- and photodynamic therapy under 980 nm laser irradiation. The prepared multifunctional Ce6-CPT-UCNPs have great potential in applications such as cancer targeted fluorescent imaging, simultaneous ROS activated chemo- and photodynamic therapy in near future.

Wound infection remains a significant clinical challenge, exacerbated by the growing prevalence of bacterial resistance due to the overuse of conventional antibiotics. Photodynamic therapy (PDT) offers a promising approach for wound sterilization that circumvents the issue of antibiotic resistance. However, conventional photosensitizers are prone to inactivation and exhibit poor retention at the wound site, limiting their clinical efficacy. To overcome these limitations, we developed a biomimetic mineralization approach to encapsulate the small-molecule photosensitizer chlorin e6 (Ce6) within magnesium phosphate nanoparticles (CMP NPs), preserving its photodynamic activity. This mineralized CMP NPs were further integrated with gelatin and natural moisturizing factor to fabricate a composite hydrogel dressing (CMP/Gel) for infected wound healing. Gelatin functions as a structural matrix that prolongs the local retention of CMP NPs, while the natural moisturizing factor enhances the hydration capacity and mechanical integrity of the dressing. Notably, magnesium ions released during the degradation of CMP NPs contribute to accelerated wound healing through multiple therapeutic effects, including antioxidant, anti-inflammatory, and pro-angiogenic activities following PDT-mediated bacterial eradication. Both in vitro and in vivo experimental results demonstrate that CMP/Gel effectively promotes infected wound repair, highlighting its potential as a novel and multifunctional therapeutic strategy for infected wound management.

Background Singlet oxygen ( 1 O 2 ) is highly reactive to biological components such as lipids, proteins and DNA, which induces oxidative damage to cells and tissues. Natural antioxidants may function as 1 O 2 quencher to prevent 1 O 2 involved photosensitized oxidation in biological system. Methods Time-resolved measurement of 1 O 2 luminescence was employed to evaluate the 1 O 2 quenching abilities of natural antioxidants in air-statured phosphate buffered saline (PBS), including (-)-Epigallocatechin gallate (EGCG), Proanthocyanidins, L-carnosine and Vitamin C. The 1 O 2 quenching effects and rate constant of EGCG were investigated by detecting the absorption, fluorescence and 1 H-NMR spectroscopy and 1 O 2 luminescence decay curves, respectively. In addition, the protective activity of EGCG against 1 O 2 oxidative damage caused by Ce6-mediated photodynamic therapy (PDT) was verified in cells. Results EGCG, proanthocyanidins, L-carnosine and Vitamin C efficiently quenched 1 O 2 luminescence at 1270 nm. The triplet-state quenching rate constants of EGCG for Rose Bengal (RB), Chlorin e6, AlPcS and HiPorfin are 2.21 × 10 9 , 4.90 × 10 8 , 3.30 × 10 8 , 1.78 × 10 9 M −1 s −1 , while the 1 O 2 quenching rate constants are 2.80 × 10 8 , 1.50 × 10 8 , 1.30 × 10 8 , 1.70 × 10 8 M −1 s −1 , respectively. Furthermore, EGCG could effectively quench 1 O 2 production to prevent NIH/3T3 cells oxidative damage induced by Ce6-mediated PDT. Conclusions EGCG is an efficient quencher for both triplet-state photosensitizers and 1 O 2 . The quenching ability of EGCG during photosensitization for selected photosensitizers is: RB > HiPorfin > Ce6 > AlPcS. EGCG could be used to protect normal cells and tissue against oxidative damage.

Chemotherapy as a primary treatment for breast cancer is limited due to drug resistance in clinical treatment. The rapid proliferation and invasive biological properties of breast cancer, resulting in intratumoral hypoxia and adaptive antioxidation, are common manifestations of drug-resistant breast cancer. Photodynamic therapy is a potential strategy to overcome drug resistance, but its effectiveness is often limited by tumor hypoxia. In order to overcome the problem of tumor hypoxia and improve the effectiveness of photodynamic therapy, a strategy that employs ZIF-90 nanoparticles simultaneously delivering a hypoxia-induced drug (TPZ) and photosensitizer Ce6 labeled antisense nucleic acid (Ce6-G3139) into breast cancer cells was developed. Owing to the pH responsiveness of ZIF-90, TPZ and Ce6-G3139 are able to be efficiently released inside the tumor microenvironment. First, Ce6-G3139 can perform gene therapy, downregulating the expression of antiapoptotic protein BCL-2 and accelerating cell apoptosis. On the other hand, photosensitizer Ce6 can undergo photodynamic therapy (PDT), producing reactive oxygen species that interfere with the apoptosis mechanism of cells. More importantly, although the continuous consumption of oxygen in PDT aggravates the hypoxic environment of the tumor, the hypoxic-induced chemotherapy drug was further activated. Our research results indicate that the designed strategy targeting the hypoxic microenvironment of drug-resistant breast cancer exhibits significant anticancer effects both in vitro and in vivo, favoring the development of a PDT treatment paradigm that is effective against drug resistance under hypoxic conditions.

Cuproptosis is a copper-dependent programmed cell death triggered by mitochondrial dysfunction, offers significant anti-tumor potential but requires tumor-specific copper delivery to avoid systemic toxicity. Here, we developed a synergistic nanoplatform (Cu₂O@SiO₂-Ce6, CSC) integrating cuproptosis induction with photodynamic therapy (PDT). Cuprous oxide (Cu₂O) core was encapsulated in silicon dioxide and covalently linked to the photosensitizer Ce6. After accumulating in tumors regions via the EPR effect, CSC releases a surge of copper ions upon encountering the acidic tumor microenvironment and lysosomes. These ions induce mitochondrial cuproptosis by aggregating lipoylated TCA cycle proteins and depleting Fe-S cluster proteins. Concurrently, laser irradiation activates Ce6 to generate cytotoxic reactive oxygen species (ROS), actively disrupting tumor membranes. The experiments showed that the nanoplatform significantly enhanced the tumor cell killing effect and reduced the toxicity to normal tissues through a dual mechanism, which provided a new strategy for overcoming the drug resistance of PDT and the precise regulation of cuproptosis.

Fe-based metal–organic frameworks (MOFs) can be used for chemodynamic therapy (CDT) for tumors due to their unique Fenton-like effects and porous and biodegradable nature. The adsorption and transport of small molecule drugs by their structure has attracted much attention. Herein, MnO2@NH2-MIL101(Fe)@Ce6-F127 nanoparticles (MNMCF NPs) were synthesized using a facile solvothermal strategy. The small molecule photosensitizer Ce6 was adsorbed by MOFs to improve the biocompatibility of Ce6 and give it high bioavailability when injected intravenously. When the MNMCF NPs reached the tumor site, Fe-based MOFs exhibited Fenton-like properties, producing ˙OH and showing CDT effects. MnO2 could specifically respond to produce O2 in a tumor microenvironment, thereby improving the tumor hypoxia state and enhancing the efficacy of photodynamic therapy (PDT) by Ce6. Both the in vitro and in vivo experiments showed that the MNMCF-guided CDT/PDT combination therapy could effectively ablate tumors without the drawbacks of poor tolerability and potential long-term side effects. Therefore, the prepared MNMCF NPs can be used as promising candidates for synergistic CDT/PDT tumor therapy.

Angiogenesis is an irreplaceable therapeutic cancer target, where anti-angiogenesis are drugs that are limited by their hydrophobicity and low therapeutic effects. What is more, the long-term shutdown of tumor blood vessel density also aggravates hypoxia and causes immunosuppression in the tumor microenvironment (TME). In order to solve these shortcomings, we developed a single therapeutic agent based on a bovine serum albumin nanocarrier that can co-deliver the anti-angiogenic drug Sorafenib (“S”) and the photosensitizer Ce6 (“C”) along with a molecular oxygen supply based on MnO2 (“M”) as a convenient one-pot formulated nanoscale agent (SCM@BSA). Compared with anti-angiogenesis monotherapy, SCM@BSA can not only improve upon the solubility and therapeutic effects of anti-angiogenesis agents, but it also reshapes the immunosuppressive TME during anti-angiogenic therapy. Together, these results point out that SCM@BSA synthesized via a very simple method can solve the shortcomings usually experienced during long-term anti-angiogenic therapy.
