In this work, we demonstrated a sensor array with multiple fluorescence channels using dendritic mesoporous silica nanoparticles embedded with three quantum dots for the determination of four heavy metal ions (Hg 2+ , Cu 2+ , Cr 3+ , and Ag + ). Carboxyl-modified CdTe QDs with three different fluorescence emission wavelengths were loaded onto a dendritic mesoporous supporter by an amidation reaction. The fluorescence sensor array exhibited excellent analytical performance for discrimination and semi-quantification of heavy metal ions from a single test, which simplified detection procedures. The four heavy metal ions exhibited different degrees of quenching of the fluorescence emission intensities of the three quantum dots and resulted in a variant data matrix for linear discriminant analysis . Under optimized conditions, the fluorescence sensor array discriminated the four heavy metal ions in a concentration range of 0.05–5 μmol/L, and semi-quantified Hg 2+ , Cu 2+ , Cr 3+ , and Ag + with a limit of detection of 2.51 nmol/L, 5.15 nmol/L, 3.81 nmol/L, and 5.74 nmol/L, respectively. The fluorescence sensor array integrated the sensing units into a single nanoparticle instead of the complex multiple detection steps used in traditional sensor arrays, providing an alternative strategy for constructing a single-well sensing platform. Furthermore, the fluorescence sensor array showed great practical potential for distinguishing heavy metal ions in raw water and crayfish samples.

Simultaneous detection of active and inactive proteases is clinically meaningful for improving diagnostic specificity. In this work, we reported an electrochemical method for simultaneous immunoassays of active and total proteases. Magnetic beads (MBs) were used as the solid supports for immobilization of capture antibodies and enrichment of targets. For the detection of active protease, the proteolytic-reaction-based analysis was carried out by the generation of Cu 2+ -binding peptide, in which a label-free peptide was used as the proteolytic substrate. The redox potential of the resulting peptide-Cu 2+ complex was intrinsically distinguished from that of free Cu 2+ , thus allowing the “signal-on” detection of active protease. For the immunoassay of total protease in a sandwich-like format, electroactive metal-organic frameworks (Cu-MOFs) were used as the signal tags. The captured Cu-MOFs could directly produce a well-defined electrochemical signal from the reduction of Cu 2+ ions. The analytical performances of the immunoplatform were evaluated by determining the model analytes of free and total prostate-specific antigen (fPSA and tPSA) in buffer and serum. The detection limits were found to be 0.3 pM for fPSA and 2 pM for tPSA. This work proposed a new strategy for simultaneous detection of active and total proteases, which should be evaluable for clinical diagnosis and treatment of protease-relative diseases.

Background Multi-component nano-delivery systems based on chemotherapy (chemo)- photodynamic therapy (PDT)- chemodynamic therapy (CDT) have gained increased attention as a promising strategy to improve clinical outcomes in cancer treatment. However, there remains a challenge in developing biodegradable, biocompatible, less toxic, yet highly efficient multicomponent nanobased drug delivery systems (DDS). Here, our study presents the screening and development of a novel DDS based on co-assemblies natural small molecule (NSMs). These molecules (oleanolic acid, and betulinic acid) are combined with photosensitizers Chlorine6 (Ce6) and Cu 2+ that are encapsulated by tumor cell membranes. This nanocarrier encapsulated in tumor cell membranes achieved good tumor targeting and a significant improvement in tumor accumulation. Methods A reprecipitation method was used to prepare the co-assembled nanocarrier, followed by the introduction of Cu 2 + into the DDS (OABACe6 NPs). Then, by wrapping the surface of NPs with the cell membranes of 4T1 which is a kind of mouse breast cancer cells (CM@OABACe6/Cu NPs). and analysis of its structure and size distribution with UV–Vis, XPS, FT-IR, SEM, TEM, and DLS. The synergistic effects of in vitro chemotherapy, CDT and PDT and targeting were also validated by cellular and animal studies. Results It was shown that CM@OABACe6/Cu NPs achieved good tumor targeting and a significant improvement in tumor accumulation. In the composite nano-assembly, the NSMs work together with the Ce6 to provide effective and safe chemo and PDT. Moreover, the effect of reduced PDT due to the depletion of reactive oxygen species (ROS) by excess glutathione (GSH) in the tumor can be counteracted when Cu 2 + is introduced. More importantly, it also confers CDT through a Fenton-like catalytic reaction with H 2 O overexpressed at the tumor site. Conclusions By constructing CM@OABACe6/Cu NPs with homologous targeting, we create a triple synergistic platform for cancer therapy using PDT, chemo, and CDT. We propose here a novel combinatorial strategy for designing more naturally co-assembled small molecules, especially for the development of multifunctional synergistic therapies that utilize NSMs.

Porphyrinic metal–organic frameworks (MOFs) had promising applications in the photodynamic therapy (PDT) and chemodynamic therapy (CDT) of cancers, but the insufficient reactive oxygen species restricted the efficacy. Herein, by coordinating Cu 2+ and coating ultrasmall gold nanoparticles (AuNPs), a bi-functional porphyrinic MOFs (PCN-224) nanoplatform (PCN-224(Cu)-Au) was designed, achieving the dual-enhancement of PDT/CDT. The results indicated that the Cu 2+ was doped in the core of porphyrin ring, and the singlet oxygen ( 1 O 2 ) generation increased from 79.9% to 85.7%, owing to the enhanced capture capability of photogenerated electrons by Cu 2+ . Using the glucose oxidase-like property of ultrasmall AuNPs, the produced hydrogen peroxide (H 2 O 2 ) promoted the Fenton-like reaction of Cu + , by which the hydroxyl radicals ( OH) generation increased from 63.2% to 80.3%, exhibiting the enhanced CDT performance. Under the combination of enhanced PDT/CDT, the cell viability reduced to 9.1%, and the tumors disappeared completely, demonstrating the dual-enhancement effect of PCN-224(Cu)-Au nanoprobe .

Improving the mechanical properties of hydrogels is a prime example of their large-scale, diverse applications. Herein, we report a one-pot method for preparing a double network system hydrogel where the polyvinyl alcohol served as the first polymer backbone, acrylamide as the second network, and N, N′-Methylenebisacrylamide as the cross-linker, and the prepared hydrogels presented excellent mechanical properties with 1168% tensile strain and 598 kPa compressive strength. Through the metal–ligand bonds, an electrolyte solution containing Cu 2+ was introduced into the hydrogel, which exhibits higher water retention than other electrolyte-containing hydrogels. Specially, the hydrogel was able to retain water for 8 h under extreme dry conditions at 60°C. The GF value was calculated to be 0.124 when the strain was 0%–64.2%. Furthermore, the hydrogel flexible sensor can detect changes in ambient temperature. When the ambient temperature rises, its relative resistance also tends to rise. In conclusion, this hydrogel sensor offers great potential applications in flexible sensors.

Cu 2+ are found to greatly reduce the photoinduced oxidase activity of fluorescein and then inhibit the chromogenic reaction catalyzed by fluorescein. A simple colorimetric assay for Cu 2+ is established. Based on this, bifunctional nanocomposites of α-fetoprotein (AFP) antibody (Ab) and copper-based metal-organic framework (Ab2@Cu-MOF) are synthesized by the simple self-assembly of AFP Ab2, Cu 2+ , and 4,4′-dipyridyl: the binding site of AFP Ab2 exposed on the surface of the nanocomposites can specifically recognize AFP antigen; Cu 2+ in nanocomposites can inhibit the visible light-induced activity of fluorescein. The structure of Ab2@Cu-MOF disintegrate and Cu 2+ is released in an acetate buffer solution. The higher the amount of AFP antigens, the more significant the inhibitory effect. Thus, the Ab2@Cu-MOF immunoassay for AFP determination is established using 3,3′,5,5′-tetramethylbenzidine as chromogenic substrate with a detection limit of 35 pg.mL −1 . This simple, cheap, and sensitive method sheds substantial light on practical clinical diagnosis. Meanwhile, the mechanism of inhibition is revealed to facilitate the targeted selection of enzyme regulators.

Nowadays, effluent treatment is a severe challenge mainly because of its complex composition, which includes oil, heavy metal ions, and dyes. Developing new intelligent membranes is one of the strategies to tackle these significant challenges in wastewater treatment . In this study, we fabricated asymmetric polyethylene glycol terephthalate (PET) membranes by grafting cross-linked poly (itaconic anhydride) (CL-PITA) nanoparticles onto the irradiated face. These nanoparticles were then functionalized with polyethyleneimine (PEI) and protonated with HCl to introduce numerous active electropositive amine groups. The fundamental purpose was to increase surface roughness, introduce numerous hydrophilic groups, and modify it to create a multi-functional PET membrane to separate complex environments. The promising results demonstrated that the protonated PET- g -ITA/DVB (10) -cat membrane exhibited excellent separation efficiencies (SE) for water/light oil, water/heavy oil and oil-in-water (O/W) emulsion. Compared to PET- g -ITA/DVB (0) -cat, it showed superior performance in SE for O/W emulsion and flux decay for water/light oil after 10 cycles. More interestingly, owing to numerous positively charged active amino groups and negativley charged carboxylate groups, the intelligent membrane exhibited a high removal rate of ca. 90 % for anionic dye (congo red) and heavy metals (Cu 2+ and Co 2+ ), showing great potential in complex water treatment environments.

Tumor cells can survive when detached from the extracellular matrix (ECM) or lose cell–cell connections, a phenomenon known as anoikis-resistance (AR). AR is closely associated with tumor cell metastasis and recurrence, enabling tumor cells to disseminate, migrate, and invade after detachment. To address this issue, a novel intervention method combining intraoperative hemostasis with multifunctional nanozyme driven-enhanced chemodynamic therapy (ECDT) has been proposed, which holds the potential to weaken the AR capability of tumor cells and suppress tumor recurrence. Here, a nanocomposite containing a dendritic mesoporous nanoframework with Cu 2+ was developed using an anion-assisted approach after surface PEG grafting and glucose oxidase (GOx) anchoring (DMSN-Cu@GOx/PEG). DMSN-Cu@GOx/PEG was further encapsulated in a thermal-sensitive hydrogel (H@DMSN-Cu@GOx/PEG). DMSN-Cu@GOx/PEG utilizes its high peroxidase (POD) activity to elevate intracellular ROS levels, thereby weakening the AR capability of bladder cancer cells. Additionally, through its excellent catalase (CAT) activity, DMSN-Cu@GOx/PEG converts the high level of hydrogen peroxide (H 2 O 2 ) catalyzed by intracellular GOx into oxygen (O 2 ), effectively alleviating tumor hypoxia, downregulating hypoxia-inducible factor-1α (HIF-1α) expression, inhibiting epithelial-mesenchymal transition (EMT) processes, and ultimately suppressing the migration and invasion of bladder cancer cells. Interestingly, in vivo results showed that the thermosensitive hydrogel H@DMSN-Cu@GOx/PEG could rapidly gel at body temperature, forming a gel film on wounds to eliminate residual tumor tissue after tumor resection surgery. Importantly, H@DMSN-Cu@GOx/PEG exhibited excellent hemostatic capabilities, effectively enhancing tissue coagulation during post-tumor resection surgery and mitigating the risk of cancer cell dissemination and recurrence due to surgical bleeding. Such hydrogels undoubtedly possess strong surgical application. Our developed novel nanosystem and hydrogel can inhibit the AR capability of tumor cells and prevent recurrence post-surgery. This study represents the first report of using dendritic mesoporous silica-based nanoreactors for inhibiting the AR capability of bladder cancer cells and suppressing tumor recurrence post-surgery, providing a new avenue for developing strategies to impede tumor recurrence after surgery. Smart responsive nanomedicines serving as carriers can take advantage of the specific tumor microenvironment (TME) to convert non-toxic drugs into effective anticarcinogens and release chemotherapeutic drugs in situ to improve therapeutic efficiency against tumors without inducing off-target toxicity to surrounding normal tissues/cells. Herein, a pH-responsive metal-organic framework nanoparticle is constructed by encapsulating disulfiram (DSF) and doxorubicin (DOX) into the zeolitic imidazolate framework-8 (ZIF-8) and then coating with copper ion (Cu 2+ )-tannic acid (TA) complex (denoted as DSF/ [email protected] @Cu-TA) to achieve enhanced combination chemotherapy. The release of Cu 2+ from the outer shell of DSF/ [email protected] @Cu-TA and the cooperative exposure of DSF and DOX are triggered at multiple stages by mild acidity TME. The accumulation of DSF (non-toxic drugs) and Cu 2+ results in the rapid formation of high cytotoxic bis(N, N-diethyl dithiocarbamato)-Cu 2+ complexes (CuL 2 ) in situ via DSF and Cu 2+ chelating reaction, which accompany the production of ROS via Cu + -based Fenton-like reaction. Besides, the anticancer effect of DOX is augmented by CuL 2 -modulated ROS-MAPK and NF-κB signal pathways. The constructed DSF/ [email protected] @Cu-TA present significantly improved therapeutic efficiency as demonstrated both in vitro and in vivo . The strategy of nontoxicity-to-toxicity transitions with the addictive effect of chemotherapeutics provides a promising paradigm to design novel nanoplatforms for synergistic cancer chemotherapy.

Mitochondrial dysfunction of neurons is the core pathogenesis of incurable Parkinson's disease (PD). It is crucial to ameliorate the mitochondrial dysfunction of neurons for boosting the therapy of PD. Herein, the remarkable promotion of mitochondrial biogenesis to ameliorate mitochondrial dysfunction of neurons and improve the treatment of PD by using mitochondria-targeted biomimetic nanoparticles, which are Cu 2- x Se-based nanoparticles functionalized with curcumin and wrapped with DSPE-PEG 2000 -TPP-modified macrophage membrane (denoted as CSCCT NPs), is reported. These nanoparticles can efficiently target mitochondria of damaged neurons in an inflammatory environment, and mediate the signaling pathway of NAD + /SIRT1/PGC-1 α /PPAR γ /NRF1/TFAM to alleviate 1-methyl-4-phenylpyridinium (MPP + )-induced neuronal toxicity. They can reduce the mitochondrial reactive oxygen species, restore mitochondrial membrane potential (MMP), protect the integrity of mitochondrial respiratory chain, and ameliorate mitochondrial dysfunction via promoting mitochondrial biogenesis, which synergistically improve the motor disorders and anxiety behavior of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD mice. This study demonstrates that targeting mitochondrial biogenesis to ameliorate mitochondrial dysfunction has a great potential in the treatment of PD and mitochondria-related diseases.
