Docosahexaenoic acid (DHA) is essential
for human health, and sustainable
production of DHA lipids by Schizochytrium sp. is industrially important. Unlike conventional bottom-up engineering
of individual pathway enzymes, this study applied a top-down strategy
using heterologous transcription factors. Screening identified soybean
GmDof4 as the most potent regulator. Overexpression significantly
boosted total lipid yield by 31.7% in a 5-L bioreactor and improved
glucose conversion efficiency by 22.4%. Although the DHA proportion
decreased slightly, absolute DHA lipid titer and yield rose by 20.8%
and 14.4%, respectively. Mechanistically, GmDof4 primarily upregulated
acetyl-CoA carboxylase (ACCase) while coordinately modulating fatty
acid synthesis and triacylglycerol assembly. Chemical inhibition of
ACCase abolished this enhancement, confirming its centrality. These
findings validate transcription factor engineering as a powerful strategy
for developing high-performance microbial DHA platforms.

Developing an appropriate concentration strategy based on the characteristics of algal oil feedstock is crucial for preparing high-concentration premium glyceride-type docosahexaenoic acid (DHA). This study employed Aspergillus Oryzae (AO) lipase hydrolysis combined with molecular distillation (MD) to concentrate DHA in microalgae oil. Under the optimal reaction conditions for AO lipase, the hydrolysis reached equilibrium after 8 h, with the stable lipid composition, in which TAG and DAG accounted for 50.1 % and 21.9 % of the total lipids, respectively, with DHA content in TAG and DAG reaching 86.8 %. A finite difference model was employed to optimize the evaporation temperature, identifying the optimal temperature as 185 °C. At this temperature, MD experiments on the enzymatic hydrolysis products increased the DHA content in the retentate from 41 % in the initial oil to 49 %, with a concentration factor of 1.21 as well as a DHA recovery rate of 93.8 %. Furthermore, a second-stage enzymatic hydrolysis combined with MD on the first-stage retentate product elevated the DHA content to 60.9 %, achieving a final concentration factor of 1.49. Lipid as well as fatty acid composition analysis exhibited DHA content variation on the glycerol backbone, and the hydrolysis mechanism of AO lipase was preliminarily clarified. The enzyme hydrolyzes small molecular fatty acids, enriching large molecular polyunsaturated fatty acids (DHA) in TAG and DAG, which are then collected in the retentate via MD, achieving high DHA concentration.

Head and neck cancer is the sixth most common cancer worldwide. Dihydroartemisinin (DHA), a semi-synthetic derivative of artemisinin, exhibits a wide range of biological roles including a highly efficient and specific anti-tumor activity. Here, we aimed to examine the effect of DHA on head and neck carcinoma cells and elucidate the potential mechanisms. We used five head and neck carcinoma cell lines and two non-tumorigenic normal epithelial cell lines to achieve our goals. Cells were exposed to DHA and subjected to cellular activity assays including viability, cell cycle analysis, cell death, and angiogenic phenotype. Our results show that DHA causes cell cycle arrest which is mediated through Forkhead box protein M1 (FOXM1). We also demonstrate that DHA induces ferroptosis and apoptosis in head and neck carcinoma cells. Lastly, our results show that DHA alters the angiogenic phenotype of cancer cells by reducing the expression of angiogenic factors and the ability of cancer cells to support endothelial cell tubule formation. Our study suggests that DHA specifically causes head and neck cancer cell death through contribution from both ferroptosis and apoptosis. DHA may represent an effective strategy in head and neck cancer treatment.

Home cooking has been considered as an indoor pollution problem since cooking oil fumes contain various toxic chemicals such as aldehydes. Fortifying edible oils with docosahexaenoic acid (DHA) has been applied to enhance the nutritional value of oils. This study designed a frying simulation system and examined the effect of oil type, DHA fortification, heating time, and addition of natural antioxidant on the emissions of aldehydes from heated oils. Results showed that linseed oil had the highest total aldehyde emissions, followed by soybean oil, peanut oil, and palm oil. Fortifying soybean oil with DHA increased the toxic aldehydes emitted. Quercetin, a flavonoid , significantly reduced aldehydes emitted from DHA-fortified soybean oil (by up to 39.80%) to levels similar to those of normal soybean oil. Further analysis showed that DHA-fortified soybean oil with quercetin had a significantly higher DHA and unsaturated fatty acids (UFAs) content than the control oil at each heating time point. The result indicated that quercetin inhibited emissions of aldehydes, at least in part, by protecting UFAs from oxidation . Collectively, quercetin could be used as a natural additive in DHA-fortified and normal cooking oils to reduce aldehyde emissions, indoor air pollution , and preserve functional DHA and other UFAs.

BACKGROUND Algae oil has garnered widespread acclaim due as a result of its high purity of docosahexaenoic acid (DHA) and excellent safety profile. The present study aimed to develop stable nanoemulsions (NEs) systems containing DHA from algae oil through thermal sterilization by combining modified whey protein concentrate (WPC) with low methoxyl pectin (LMP), as well as to investigate the impact of LMP concentration on the thermal stability and the gastrointestinal delivery efficiency of DHA NEs. RESULTS The addition of LMP enhanced the stability of the emulsion after sterilization, at the same time as improving the protective and sustained release effects of DHA in the gastrointestinal tract. Optimal effect was achieved at a LMP concentration of 1% (10 g kg −1 sample), the stability of the emulsion after centrifugation increased by 17.21 ± 5.65% compared to the group without LMP, and the loss of DHA after sterilization decreased by only 0.92 ± 0.09%. Furthermore, the addition of 1% LMP resulted in a substantial reduction in the release of fatty acids from the NEs after gastrointestinal digestion simulation, achieving the desired sustained-release effect. However, excessive addition of 2% (20 g kg −1 sample) LMP negatively impacted all aspects of the NEs system, primarily because of the occurrence of depletion effects. CONCLUSION The construction of the LMP/WPC-NEs system is conducive to the protection of DHA in algae oil and its sustained-release in the gastrointestinal tract. The results of the present study can provide reference guidance for the application of algae oil NEs in the food field. © 2024 Society of Chemical Industry.

para-Xylene (PX) is a basic building block of polyethylene terephthalate, which is currently produced from petroleum resources. Developing a renewable route to PX is highly desirable to address both economic and environmental concerns. Several attempts used noble metal catalysts, e.g. Pd/Al2O3, to synthesize PX from biomass-derived 4-methyl-3-cyclohexene-1-carboxaldehyde (4-MCHCA), but suffered from a severe decarbonylation reaction, resulting in toluene as the main product. In this paper, we report an FeOx modification strategy to suppress the decarbonylation reaction on a Pd/Al2O3 catalyst, leading to a drastic shift in selectivity towards PX with a yield up to 81% via a cascade dehydroaromatization–hydrodeoxygenation (DHA–HDO) pathway. Characterization and control experiments revealed that the electron density of Pd sites decreased in an FeOx-modified Pd/Al2O3 catalyst compared to Pd/Al2O3, thus tuning the preferential adsorption mode of the substrate from η2-(C,O), the key transition state of the decarbonylation reaction, to the η1-(O) mode that favors the hydrodeoxygenation process. Notably, this designed catalyst is highly stable and is readily applicable in the selective synthesis of a broad range of desired aromatic chemicals via the same DHA–HDO pathway from cyclohex-3-enecarbaldehyde derivatives. Overall, this work develops a controllable catalyst modification strategy that tailors an efficient catalyst for petroleum-independent bio-PX synthesis.

Ethnopharmacological relevance The traditional Chinese medicine formula Danggui-Shaoyao-San (DSS) has been reported to show therapeutic effect on alleviating the symptoms of Alzheimer's disease (AD). Aim of the study The present study aims to investigate the relation between DSS treatment of AD and DHA metabolism and evaluates its neuroprotective effect on cognitive in APP/PS1 mice. Material and methods DSS (1.6, 3.2, 6.4 g/kg/day) or Aricept (3 mg/kg/day) was orally administered (i.g.) to APP/PS1 mice, and saline was orally administered to Wild-type (WT) male mice as control group. Then, the Morris water maze (MWM) test, Y-maze spontaneous alternation test, open filed test and fear conditioning test were conducted for evaluation of learning and memory abilities. The DHA content was assessed by HPLC-MS/MS. Physiological indices were determined, including triglyceride (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), ROS level, activity of superoxide dismutase (SOD), glutathione (GSH), malondialdehyde (MDA), PEG2, TXB2 and LTB4. The expressions of COX-1, COX-2, cPLA2 , iPLA2, 15-LOX, and were assessed by Western blot . Results APP/PS1 mice showed serious cognitive impairment in behavioral tests. However, treatment of DSS extract significantly ameliorated the cognitive deficits of APP/PS1 mice. Biochemical measurements showed the increases in TG, TC, LDL-c and the decrease in HDL-c in APP/PS1 mice compared with WT mice, and DSS extract significantly retarded these changes. Low content of DHA, low expression of iPLA2 and 15-LOX were observed both in hippocampus and cortex of APP/PS1 mice, while DSS extract significantly restored these changes. Additionally, the abnormal activity of SOD and ROS level, the decreased levels of MDA and GSH were observed in APP/PS1 mice, while DSS extract prominently lessened these changes. Moreover, DSS extract decreased the level of PEG2, TXB2 and LTB4 and also attenuated the expression of cPLA2, COX-1 and COX-2 in hippocampus as well as cortex of APP/PS1 mice. Conclusions Based on these results, we suggest that DSS play a positive effective role in increasing DHA content by up-regulating iPLA2 and 15-LOX, resulting in ameliorating oxidative stress and inflammation and finally ameliorating cognition deficits in APP/PS1 mice.

Docosahexaenoic acid (DHA) glycerides are regarded as ideal functional lipids due to their high bioavailability and safety, although their efficient enzymatic synthesis faces major challenges. Herein, crescent-shaped amphiphilic semi-Janus-poly(acrylic acid)/poly(styrene) particles (JPP) and semi-Janus-poly(vinylidene imine)/octadecane particles (JPO) were prepared and used as carriers for Candida antarctica lipase B (CALB) to improve its performance in the synthesis of DHA glycerides. The surface properties of the enzyme carrier could significantly affect enzymatic performance. Immobilization of CALB on the hydrophilic side of JPO (CALB@JPO(W)) significantly enhanced its activity and stability. Compared with free enzyme, CALB@JPO(W) exhibited a 2.19-fold enhancement in catalytic efficiency, increased optimal reaction temperature from 70 to 80 °C, and substantially improved thermal stability (retaining 70% of its initial activity after 1 h of incubation at 80 °C). JPO was simultaneously employed as a solid stabilizer and a carrier to enrich the biocatalyst at the interface, forming a Pickering emulsion interfacial catalysis system for the production of DHA glycerides. The yield of DHA glycerides catalyzed by CALB@JPO(W) reached 21.35%, representing a 72.66% improvement over free CALB. These results demonstrate that the reported Pickering emulsion interfacial catalysis system provides a potential strategy for the efficient production of functional lipids.

Metal-organic frameworks (MOFs) with tunable porosity are promising carriers for enzyme immobilization, but their practical application is limited by structural instability and difficult recovery. We previously developed an oxidation-reduction (O-R) pyrolysis strategy for mesoporous MIL-88 A, yielding a magnetic carrier (O-R-Meso) with excellent lipase immobilization and recyclable catalysis. Herein, we verify the applicability of this O-R pathway for Fe-based MIL-88 A with tailored pore structures (micro-, meso-, macroporous) by extending it to microporous C-MIL-88 A and macroporous SOM-MIL-88 A. The O-R derivatives (O-R C, O-R-Meso and O-R-SOM) retained the original morphology and pore channel integrity, with improved specific surface area and sufficient magnetization (13–23 emu/g) for magnetic separation. Notably, after O-R pyrolysis, MIL-88 A with various pore sizes essentially maintained or even enhanced the specific activity of immobilized lipase ( Candida antarctica lipase B, CalB), successfully deriving into magnetic carriers with enhanced catalytic efficiency and recyclability. This superiority originates from the preserved pore structure and biocompatible γ-Fe 2 O 3 surface of the O-R derivatives, which effectively retain the native conformation of CalB. Besides, this work systematically compares the secondary structure of free and immobilized CalB on MOF-derived carriers with distinct pore architectures, providing direct structural evidence for the superior enzyme-carrier compatibility of mesoporous materials. In the transesterification synthesis of phosphatidyl EPA/DHA, CalB@O-R-Meso showed the highest EPA/DHA total incorporation, prominent sn -1 positional selectivity, and optimal reusability, outperforming CalB@O-R-C and CalB@O-R-SOM. Furthermore, kinetic studies on the CalB@O-R-Meso-catalyzed system confirmed a Ping-Pong Bi Bi mechanism and revealed competitive product inhibition by phosphatidyl EPA/DHA ( K i = 29.8 mM) with negligible internal mass transfer resistance (η ≈ 1). This work establishes a O-R pyrolysis strategy for tunable porosity Fe-MOFs, clarifies pore size-dependent enzyme -carrier compatibility, and provides a fundamental kinetic basis for phosphatidyl EPA/DHA synthesis.

Objective Docosahexaenoic acid (DHA) is indispensable for neurological health, yet its therapeutic potential is hampered by poor bioavailability and non-specific brain distribution. We hypothesized that co-administering DHA with specific molecular carriers – eicosapentaenoic acid (EPA) or phosphatidylserine (PS) – would exploit distinct cellular transport pathways to achieve region-specific brain enrichment and associated neuroprotection. Methods By dietary intervention using C57BL/6J mice, we employed regional lipidomics, ELISA, and western blotting to assess brain fatty acid incorporation, neurotrophic factor levels, inflammatory signaling, and transporter expression following supplementation with DHA alone or in combination with EPA or PS.Results Lipidomic analyses revealed striking, carrier-dependent spatial modulation of DHA. Co-administration with EPA enriched the cortex and striatum, while PS co-administration preferentially targeted the hippocampus and cortex. Mechanistically, both carrier-DHA complexes enhanced the expression of the key blood–brain barrier (BBB) transporter MFSD2A. Functionally, this precision delivery activated distinct neuroprotective programs. PS + DHA robustly upregulated the CREB-BDNF neurotrophic pathway, while EPA + DHA uniquely suppressed the NF-κB pathway, demonstrating potent anti-inflammatory effects. These results demonstrate that the choice of molecular carrier dictates both the spatial distribution of DHA and the nature of the ensuing neuroprotective response.Discussion Our findings establish that dietary co-supplementation with specific lipid carriers enables precise spatial delivery of DHA by engaging specific transporters, thereby activating distinct neuroprotective programs in a region-specific manner. This work provides a mechanistic framework for a precision nutrition strategy, tailoring DHA formulations to target specific neuroanatomical and cellular vulnerabilities in neurological disorders.

In order to investigate the amelioration of docosahexaenoic acid-enriched phosphatidylserine (DHA-PS) on bisphenol A (BPA)-induced nephrotoxicity , the murine nephrotoxicity model was established by intragastric administration of BPA (5 mg/kg/B.W.) for 6 weeks. The biochemical indices, hematoxylin-eosin (H&E) staining, kidney metabolomics, and related protein expression levels of SIRT1-AMPK pathway were then determined. Our results indicated that DHA-PS (100 mg/kg/B.W.) ameliorated the BPA-induced nephrotoxicity after 6 weeks of intragastric administration, primarily by decreasing the serum creatinine (CRE) and blood urea nitrogen (BUN), renal inflammatory cytokines and lipid levels, and increasing the antioxidant enzyme activities . In addition, the untargeted metabolomics of the kidney indicated that BPA perturbed the tryptophan metabolism, pyridine metabolism, and valine , leucine , and isoleucine biosynthesis, while DHA-PS administration significantly affected the glycerophospholipid metabolism, valine, leucine, and isoleucine biosynthesis to ameliorate the BPA-induced metabolic disorder . Moreover, DHA-PS administration could ameliorate the BPA-induced lipid disturbance by upregulating the expressions of AMPKα1, SIRT1, and PPARα while downregulating the expression of SREBP-1c through the SIRT1-AMPK pathway. This is the first time that the amelioration effects of DHA-PS on BPA-induced nephrotoxicity have been investigated from multiple perspectives, suggesting that DHA-PS might be a potential dietary supplement for reducing BPA-induced nephrotoxicity.

Algal oil is a sustainable source of docosahexaenoic acid (DHA), but its high susceptibility to oxidation and hydrophobic nature limits its applications. To address these challenges, biopolymer nanocapsules (40.87–169.27 nm) were fabricated from shellac and phospholipids using microfluidic-mediated co-precipitation for algal oil delivery. Nanocapsule size increased from 78.51 nm to 152.96 nm with higher poor solvent ratios and decreased from 169.27 nm to 61.65 nm as total fluid velocity increased. Additionally, particle size was influenced by the phospholipid fraction, ranging from 40.87 nm to 61.65 nm, with the smallest size achieved at a shellac-to-phospholipid ratio of 2:3. Nanocapsule composition further affected particle stability, oxidative stability, and DHA bioaccessibility. A higher phospholipid fraction enhanced stability against pH fluctuations and thermal stress, while all nanocapsules exhibited stability during centrifugation and static storage, retaining over 93.01% DHA after 28 days at 25 °C. During accelerated storage (37 °C, 20 days), nanocapsules reduced hydroperoxide levels by 5.44- to 3.30-fold and secondary oxidation products by 2.62- to 1.79-fold compared to pure algal oil, with higher shellac content providing superior oxidative protection. Furthermore, the nanocapsules enhanced DHA bioaccessibility by a factor of 5.43–8.23 compared to pure algal oil, achieving a maximum DHA bioaccessibility of 54.31% at the highest phospholipid level. These findings provide valuable insights into designing advanced nano-colloids for enhancing the stability and bioavailability of bioactive lipids.
