Preparation of BMP-2/PDA-BCP Bioceramic Scaffold by DLP 3D Printing and its Ability for Inducing Continuous Bone Formation

Ziyang Yang,Li Xie,Boqing Zhang,Gang Zhang,Fangjun Huo,Changchun Zhou,Xi Liang,Yujiang Fan,Weidong Tian,Yinghui Tan

Digital light processing (DLP)-based 3D printing is suitable to fabricate bone scaffolds with small size and high precision. However, the published literature mainly deals with the fabrication procedure and parameters of DLP printed bioceramic scaffold, but lacks the subsequent systematic biological evaluations for bone regeneration application. In this work, a biphasic calcium phosphate (BCP) macroporous scaffold was constructed by DLP-based 3D printing technique. Furthermore, bone morphogenetic protein-2 (BMP-2) was facilely incorporated into this scaffold through a facile polydopamine (PDA) modification process. The resultant scaffold presents an interconnected porous structure with pore size of ∼570 μm, compressive strength (∼3.6 MPa), and the self-assembly Ca-P/PDA nanocoating exhibited excellent sustained-release property for BMP-2. Notably, this BMP-2/PDA-BCP scaffold presents favorable effects on the adhesion, proliferation, osteogenic differentiation, and mineralization of bone marrow stromal cells (BMSCs). Furthermore, in vivo experiments conducted on rats demonstrated that the scaffolds could induce cell layer aggregation adjacent to the scaffolds and continuous new bone generation within the scaffold. Collectively, this work demonstrated that the BMP-2/PDA-BCP scaffold is of immense potential to treat small craniofacial bone defects in demand of high accuracy.

Effect of particle size on rheology, curing kinetics, and corresponding mechanical and thermal properties of aluminum nitride (AlN) ceramic by digital light processing (DLP)-based vat photopolymerization

Lifu Lin,Haidong Wu,Yehua Li,Junye Wang,Shanghua Wu

In this work, the influence of AlN particle size (250 nm, 1.2 µm, and 5 µm) on the curing behavior, stability and viscosity of ceramic suspensions was evaluated. It was revealed that an increase in the particle size of AlN powders reduced the stability and viscosity of the suspension. For curing behavior, the ceramic suspension prepared with 250 nm AlN powder exhibited the smallest curing depth and excess width, whereas the 5-μm ceramic suspension had the largest curing depth. The analysis of the scattering length revealed that a decrease in AlN powder particle size reduced the scattering length. After sintering, both the 250-nm and 1.2-μm samples could be sintered densely at temperatures more than 1800 °C with the relative density greater than 99%. Finally, following a 5 h sintering at 1850 °C, the 250-nm and 1.2-μm samples obtained thermal conductivities of 171 and 174 W/mK, respectively.

Low-temperature DLP 3D printing of low-concentration collagen methacryloyl for the fabrication of durable and bioactive personalized scaffolds

Xiaxia Yang,Linyan Yao,Xiuxia Sun,Lili Wang,Jianxi Xiao

Digital Light Processing (DLP) bioprinting has revolutionized tissue engineering and regenerative medicine with its speed and precision. Although commonly used in DLP printing, Gelatin Methacrylate (GelMA) lacks the mechanical strength and complex biological signaling of collagen, owing to the absence of collagen’s unique triple helix structure. However, collagen faces challenges such as denaturation at high temperatures and aggregation at neutral pH, limiting its use as a bioink. Herein, we present for the first time the development of a low-temperature DLP 3D printing technique using low-concentration Collagen Methacryloyl (ColMA) for fabricating durable and bioactive personalized scaffolds. Our study shows that pH significantly influences ColMA printing, with pH 3 producing low-concentration scaffolds that exhibit enhanced mechanical properties, minimal swelling, and prolonged resistance to enzymatic hydrolysis compared to high-concentration GelMA scaffolds, while low-concentration GelMA cannot be printed independently. ColMA scaffolds notably boost the proliferation, adhesion, migration, and differentiation of human foreskin fibroblasts. In vivo rat model tests demonstrate that ColMA scaffolds reduce inflammation, enhance collagen deposition, and accelerate epidermal regeneration. This method offers a robust solution for low-temperature DLP 3D printing of collagen ink, facilitating the creation of personalized scaffolds with superior bioactivity and representing a significant advancement in tissue engineering applications.

An effective DLP 3D printing strategy of high strength and toughness cellulose hydrogel towards strain sensing

Zhengqiang Guo,Chengdong Ma,Weigui Xie,Aimin Tang,Wangyu Liu

Photocurable 3D printing technology has outperformed extrusion-based 3D printing technology in material adaptability, resolution, and printing rate, yet is still limited by the insecure preparation and selection of photoinitiators and thus less reported. In this work, we developed a printable hydrogel that can effectively facilitate various solid or hollow structures and even lattice structures. The chemical and physical dual-crosslinking strategy combined with cellulose nanofibers (CNF) significantly improved the strength and toughness of photocurable 3D printed hydrogels. In this study, the tensile breaking strength, Young's modulus , and toughness of poly(acrylamide- co -acrylic acid) D /cellulose nanofiber (PAM-co-PAA) D /CNF hydrogels were 375 %, 203 % and 544 % higher than those of the traditional single chemical crosslinked (PAM-co-PAA) S hydrogels, respectively. Notably, its outstanding compressive elasticity enabled it to recover under 90 % strain compression (about 4.12 MPa). Resultantly, the proposed hydrogel can be utilized as a flexible strain sensor to monitor the motions of human movements, such as the bending of fingers, wrists, and arms, and even the vibration of a speaking throat. The output of electrical signals can still be collected through strain even under the condition of energy shortage. In addition, photocurable 3D printing technology can provide customized services for hydrogel-based e-skin, such as hydrogel-based bracelets, fingerstall, and finger joint sleeves.

Multi-Material Digital Light Processing (DLP) Bioprinting of Heterogeneous Hydrogel Constructs with Perfusable Networks

Ming Yang,Lang Chu,Yanfeng Zhuang,Cheng Qi,Si Meng,Zhou Liu,Tiantian Kong

The challenges of replicating the complex mechanical and structural diversity of natural tissues in vitro by leveraging multi-material digital light processing (DLP) bioprinting are addressed. This technique utilizes PEGDA-AAm bio-ink to develop multi-component, cell-laden hydrogel constructs with varying moduli. These constructs not only possess heterogeneous mechanical properties but also feature complex architectures and precisely engineered surface microstructures. The hydrogel microfluidic chips are successfully fabricated with perfusable microchannels, embedding various cell types within the matrix. This approach enables the bioprinting of intricate cell-laden structures with unique surface topologies, such as spiral grooves and triply periodic minimal surfaces (TPMS), which effectively influence cell alignment, spreading, and migration. By integrating various cell-laden PA hydrogels, the diverse mechanical moduli of biological tissues, including bone, liver lobules, and vascular networks are replicated. This technique ensures high-fidelity differentiation between cell types and regions. These findings provide valuable insights into the impact of substrate modulus and structure on cell behavior, underscoring the potential of multi-component, multi-modulus hydrogel constructs in creating sophisticated structures with custom-tailored mechanical properties. This study significantly advances the field by demonstrating the feasibility and effectiveness of multi-material DLP bioprinting in developing complex, functionally relevant tissue models for tissue engineering and regenerative medicine.

Advantages of photo-curable collagen-based cell-laden bioinks compared to methacrylated gelatin (GelMA) in digital light processing (DLP) and extrusion bioprinting

Huimin Shi,Yang Li,Kailei Xu,Jun Yin

The development of cell-laden bioinks that possess high biocompatibility and printability is crucial in the field of bioprinting for the creation of cell-embedded tissue engineering scaffolds. As widely known, methacrylated gelatin (GelMA) is one of the most commonly used photo-crosslinkable bioink for cell-laden bioprinting with different printing methods, but GelMA is the derivative of gelatin, so it loses the unique triple-helix molecular structure of collagen and may not be able to successfully activate the cellular pathways or facilitate cell-matrix interaction as effectively as collagen. Recently, methacrylated collagen (CMA) was developed to be an alternative photocrosslinkable bioink with a good bioactivity, but its low printability and biocompatibility limited that application in tissue engineering. In this study, the synthetic process for CMA was improved by synthesizing under 4 °C and using acidic aqueous solution as solvent. Our CMA bioinks were demonstrated a similar printability as GelMA in extrusion bioprinting, while a better formability in digital light processing (DLP). To further analyze the bioactive properties, CMA bioinks were encapsulated with Schwann cells (SCs) and bone mesenchymal stem cells (BMSCs) for printing. SCs-laden CMA bioinks had a significantly higher proliferation rate and expression of neural stem cell-associated genes than GelMA in DLP bioprinting. While, BMSCs-laden CMA bioinks demonstrated >95% cellular viability, better cell spreading and higher expression of osteogenesis-related genes than that of GelMA. Overall, we speculate that the CMA-based bioink developed in this study could be potential bioinks for 3D cell-laden bioprinting in the future.

3D Printing of Thermo-Sensitive Drugs

Sadikalmahdi Abdella,Souha H. Youssef,Franklin Afinjuomo,Yunmei Song,Paris Fouladian,Richard Upton,Sanjay Garg

Three-dimensional (3D) printing is among the rapidly evolving technologies with applications in many sectors. The pharmaceutical industry is no exception, and the approval of the first 3D-printed tablet (Spiratam®) marked a revolution in the field. Several studies reported the fabrication of different dosage forms using a range of 3D printing techniques. Thermosensitive drugs compose a considerable segment of available medications in the market requiring strict temperature control during processing to ensure their efficacy and safety. Heating involved in some of the 3D printing technologies raises concerns regarding the feasibility of the techniques for printing thermolabile drugs. Studies reported that semi-solid extrusion (SSE) is the commonly used printing technique to fabricate thermosensitive drugs. Digital light processing (DLP), binder jetting (BJ), and stereolithography (SLA) can also be used for the fabrication of thermosensitive drugs as they do not involve heating elements. Nonetheless, degradation of some drugs by light source used in the techniques was reported. Interestingly, fused deposition modelling (FDM) coupled with filling techniques offered protection against thermal degradation. Concepts such as selection of low melting point polymers, adjustment of printing parameters, and coupling of more than one printing technique were exploited in printing thermosensitive drugs. This systematic review presents challenges, 3DP procedures, and future directions of 3D printing of thermo-sensitive formulations.Keywords:3D printing;thermolabile;thermosensitive;FDM;SSE;SLA;DLP

High-precision freeform DLP bioprinting by thermo-reversible gelation

Daobo Han,Mingshan Zhang,Junfang Xie,Zhaoxuan Dong,Shuang Xia,Yifei Zhan,Yipeng Dong,Yonghe Li,Jin Wang,Zhichao Deng,Jianchun Mei,Jianguo Tian,Qing Ye

Light-based 3D bioprinting technology can be used to prepare complex hydrogel structures for tissue engineering, but it is limited by reduced precision due to radical diffusion and light scattering, as well as geometric restriction. We propose a reversible gelation suspension stereolithography technique, which combines DLP printing with temperature-controlled reversible gelation, gelling the bioink prior to photocuring. This strategy suppresses diffusion and scattering, achieving 6.8 μm precision, and provides in situ support for suspended structures, overcoming traditional geometric restriction. We demonstrated the feasibility in unrestricted geometry by printing various structures and demonstrated multi-material printing. Cell-loaded hexagonal hepatic lobule bio-inspired structure and multi-scale branched vessels seeded with HUVEC were then constructed, high survival rates and functional performance demonstrated its superior biocompatibility, thereby providing possibilities for new tissue engineering applications.

Siloxane-Reinforced GelMA/Methacrylated Chitosan Hydrogels for DLP Printing

Yufan Zhang,Jing Wang,Bo Liu,Xueqin Zhang,Yunxuan Weng

Photocrosslinked gelatin methacryloyl/methacrylated chitosan (GelMA/CSMA) hydrogels are attractive for biomedical applications but are limited by insufficient mechanical strength. Here, γ-methacryloxypropyltrimethoxysilane (MPTS) was prehydrolyzed and condensed to generate siloxane species retaining polymerizable methacrylate groups, which were subsequently incorporated into the GelMA/CSMA network as a reinforcing phase. The effects of MPTS precursor concentration (2–5%w/v) and prehydrolysis time (12–48 h) on the morphology, viscoelasticity, compressive behavior, cytocompatibility, and DLP printability of the resulting GCM hydrogels were systematically investigated. FTIR confirmed methoxy-group consumption while retaining C=C functionality. SEM showed that increasing MPTS precursor concentration increased the apparent abundance of granular features, whereas prolonged prehydrolysis produced larger structures. XRD indicated a predominantly amorphous MPTS-derived phase. GCM-48 h-5% achieved a storage modulus of 23.5 kPa, compressive modulus of 0.682 MPa, and maximum compressive stress of 1.081 MPa, corresponding to 7.4-, 16.9-, and 4.0-fold increases over the GelMA/CSMA control, respectively. The hydrogels exhibited favorable L929 cytocompatibility and retained DLP printability under 405 nm irradiation. These findings demonstrate that regulating MPTS precursor concentration and prehydrolysis time provides a simple route to mechanically reinforce DLP-printable GelMA/CSMA hydrogels without directly incorporating preformed silica particles.

Fabrication of High-Density Microarchitected Tungsten via DLP 3D Printing

Junyu Cai,Songhua Ma,Wenbin Yi,Jieping Wang

Current additive manufacturing (AM) techniques for tungsten, such as powder bed fusion and directed energy deposition, often generate parts with rough surfaces. Vat photopolymerization presents a promising alternative for fabricating tungsten structures with high shape fidelity and low surface roughness. However, existing vat photopolymerization approaches suffer from surface defects and low final density, leading to compromised mechanical properties. Therefore, achieving high-density tungsten structures using vat photopolymerization remains a crucial challenge. This work presents a straightforward and reliable method for fabricating complex, micro-architected tungsten structures with superior density and hardness. The approach utilizes a water-based photoresin with exceptional tungsten ion loading capacity. The photoresin is then patterned using digital light processing (DLP) to create tungsten-laden precursors. A three-step debinding and sintering process subsequently achieves 3D tungsten structures with dense surface morphology and minimal internal defects. The microstructures achieve a minimum feature size of 35 µm, a low surface roughness of 2.86 µm, and demonstrate exceptional mechanical properties. This new method for structuring tungsten opens doors to a broad range of applications, including micromachining, collimators, detectors, and metamaterials.

DLP 3D printed hydrophobic polymer for passive daytime radiative cooling

Yuxuan Ma,Xun Zhang,Feiyue Zhou,Lixuan Yang,Jingxing Gui,Yuting Xue,Chunyan Hu,Baojiang Liu,Dan Yu,Wei Wang

Rapid urbanization has exacerbated the urban heat island (UHI) effect and increased building cooling energy demand, posing significant challenges to public health and energy sustainability. Herein, we developed a 3D-printed spectrally engineered polymer (PPHP) for building radiative cooling via UV-initiated thiol–ene click chemistry combined with chemical vapor deposition (CVD), achieving high solar reflectance, strong emissivity within the atmospheric window, and stable performance under humid conditions. PPHP consists of a crosslinked organic network formed by PETMP and PETA/HDDA, with PVDF-HFP incorporated to generate a porous fluorinated structure. The multiscale scattering interfaces provided a high solar spectral reflectance of 94.24%, effectively suppressing solar absorption. Meanwhile, the strong vibrational characteristics of the–CF 2 – and C–F bonds in the 8–13 μm range enable a near-blackbody emissivity of 99.16%, facilitating efficient thermal radiation to outer space. MTMS chemical vapor deposition further constructs a hydrophobic interface (contact angle of 136°), ensuring spectral stability across a humidity range of 11–98% relative humidity (RH). As a result, PPHP 15 -C/T 5 exhibited a theoretically simulated subambient cooling potential of 6–10 °C under a solar irradiance of 900 W m −2 and achieved an average subambient cooling of 6.88 °C in the rooftop model-house test. EnergyPlus simulations further indicated that PPHP 15 -C/T 5 could reduce the annual cooling load in cooling-dominated regions when applied to roofs and opaque exterior walls. This study provides a scalable and manufacturable strategy for passive radiative cooling of building envelopes under complex climatic conditions.

DLP-Printed Microstructured Sensors with Enhanced Sensitivity for Wearable Physiological

Yi Ru,Lijun Dong,Ru Jia,Jing Wang,Lanlan Dong,Aiyiti Wurikaixi,Cijun Shuai

Flexible capacitive pressure sensors have attracted significant attention owing to their simple structure and performance. However, the limited volumetric deformation of traditional planar dielectric layers under pressure severely restricts sensitivity improvement. In this study, we developed a high-performance flexible sensor based a 3D-printed microstructured hydrogel composed of polyethylene glycol diacrylate (PEGDA), N-hydroxyethyl acrylamide (HEAA), and lithium chloride (LiCl). An exposure regulation strategy maintained optimal curing energy (100–200 mJ/cm 2 ) by reducing exposure time by 30–50% with every 5 mW/cm 2 increase in light intensity. By precisely engineering the microneedle aspect ratio (H/D = 2:1), synergistic optimization of sensitivity across broad pressure ranges was achieved: 0.31 kPa -1 in the low-pressure regime (0–6 kPa) and 0.11 kPa -1 in the high-pressure regime (30–160 kPa), which was 3-5 times higher than that of the flat structure. Notably, the cross-sectional shape has a significantly impacts on sensitivity. The triangular cross-sectional design exhibited an enhanced sensitivity of 1.03 kPa -1 in the low-pressure range and maintained 0.56 kPa -1 in the high-pressure region—representing a 400% enhancement over elliptical or square structures. The sensor also demonstrated environmental adaptability from -40 °C to 40 °C, long-term durability (600 cycles), and fast response times (65 ms). These features enable real-time monitoring of diverse physiological signals (e.g., pulse, respiration, and muscle motion), as well as individual identification through handwriting pattern recognition—highlighting its potential for personalized health monitoring, smart wearables, human-machine interfaces, and secure authentication systems.
