Dual drive acute lethal toxicity of methylene blue to Daphnia magna by polystyrene microplastics and light

Xiangmei Xiang,Juanjuan Zhou,Shaochen Lin,Nan Zhang,Gulizhabaier Abulipizi,Guikui Chen,Zhanjun Li

Microplastics (MPs) can adsorb and influence the toxicity of traditional pollutants significantly. Although the complex toxicity of MPs and molecular pollutants were frequently reported, rare work has been done on the influence of MPs on the phototoxicity of photosensitive pollutants under light illumination condition. Herein, polystyrene microplastics (PS) (~1 μm in diameter, 5.0 mg/L) was used as a model MP to investigate its influence on the phototoxicity of a soluble blue dye, methylene blue (MB) using Daphnia magna as a model organism. The results indicate that PS could adsorb MB effectively and quickly, thus led to concentrated MB on PS/water interface. D. magna ingested MB-adsorbed PS very quickly within tens of minutes. Although MB or PS alone led to negligible lethal phototoxicity to D. magna , PS significantly enhanced the lethal phototoxicity of MB (0.25 mg/L) to D. magna after light illumination (10 h) with the survival rate decreased by 63.3 % compared with the control in the dark. Further, the phototoxicity of MB was found positively consistent with PS concentration from 0.50 mg/L to 7.50 mg/L. The singlet oxygen fluorescence assay indicates that the presence of PS did not increase the total amount of singlet oxygen in the aquatic environment but increased the local concentration in the gut area via non-selective ingestion of D. magna . High level singlet oxygen generated in the gut might possibly be the main reason that led to the massive death of D. magna . Surface adsorption of photosensitive pollutants may transform inert MPs into persistent solid sources of singlet oxygen production and become a new potential lethal threat to aquatic small organisms and ecological equilibrium. This kind of MPs and light dual drive phototoxicity of photosensitive pollutants needs to paid more attention in understanding the uncertain ecological risk of MPs.

Triazophos exposure on maternal Daphnia magna at environmental-related concentrations revealed toxic effects to its offspring

Shaoquan Liu,Jianlu Zhang,Ruijiao Li,Chunyun Zhang,Lixin Wang,Hao Liang,Guangpeng Feng,Dongmei Xiong

Due to chemical and photochemical stability, triazophos has been frequently detected in rivers and oceans over the years with extensive use for pest control in agriculture , and it has become a worldwide ecological concern to the aquatic environment . Until now, fewer data are available regarding the potential long-term adverse effects of triazophos on aquatic invertebrates , which plays an essential role in aquatic food webs , as a key group for water ecosystems. In this experiment, the F 1 - and F 2 progenies of Daphnia magna were recovered when daphnias (F 0 ) exposure to triazophos at environmental-related concentrations (0.1 and 1.0 μg/L) for 21 d; and the indexes related to phenotypic traits, reproduction and gene expression were measured in tested animals. The results showed that heart rate and total number of neonates in exposed F 0 -daphnias were significantly lower than those of control group, and the detoxification genes ( HR96 and P-gp ) were up-regulated while genes related reproduction ( Vtg ) and molting ( Nvd and Shd ) were significantly down-regulated. The heart rate and individual size of F 1 -daphnias (<24 h) were significantly reduced in the treatment group. After 21-d recovery, the heart rate and expression of HR96 , P-gp, Vtg, Nvd and Shd were declined in F 1 -daphnias. There was no obvious difference of morphological traits and heart rate between treatment and control in F 2 -daphnias (<24 h). In summary, daphnias (F 0 ) exposure to triazophos with environmental dose could raise toxic effects on its offspring (F 1 ), which is mainly manifested by reduced heart rate, the accumulated number and individual size of offspring and decreased expression of genes related to molting and reproduction.

Combined exposure of polystyrene microplastics and carbamazepine induced transgenerational effects on the reproduction of Daphnia magna

He Yide,Zhang Yiqun,Zhou Wei,Freitas Rosa,Zhang Yunhai,Zhang Yongjun

Polystyrene microplastics (PS MPs) and carbamazepine (CBZ) are frequently detected in freshwater ecosystems. However, the transgenerational effects of PS MPs and CBZ on the reproduction of aquatic organisms and the corresponding mechanisms are still unclear. In the present study, Daphnia magna was used to evaluate the reproductive toxicity in two consecutive generations (F0, F1). The molting and reproduction parameters, the expression of reproduction, and the toxic metabolism genes were examined after 21-day exposure. A significantly enhanced toxicity was observed in the presence of 5 μm PS MPs and CBZ. Chronic exposure results showed that the 5 μm PS MPs alone, CBZ alone, and their mixtures exerted significant reproductive toxicity of D. magna . The results of RT-qPCR showed transcripts of genes related to reproduction ( cyp314 , ecr-b , cut , vtg1 , vtg2 , dmrt93b ) and toxic metabolism ( cyp4 , gst ) were altered in both the F0 and F1. In addition, for the F0, gene transcriptional changes of reproduction were not fully translated into physiological performance, probably due to the compensatory responses caused by the low dose of PS MPs alone, CBZ alone, and their mixtures. Whereas for the F1, the trade-off between reproduction and toxic metabolism at gene levels was observed, which translated into a significant reduction in the total neonate number of F1. These findings suggest that long-term exposure to MPs and CBZ can cause serious reproduction damage to aquatic animals, which needs to be given sufficient attention. Graphical Abstract

Evaluating the Impact of Individual and Combined Toxicity of Imidacloprid, Cycloxaprid, and Tebuconazole on Daphnia magna

Yanli Man,Tian Sun,Chi Wu,Xingang Liu,Mingyuan He

The risks posed by chemicals in the environment are typically assessed on a substance-by-substance basis, often neglecting the effects of mixtures. This may lead to an underestimation of the actual risk. In our study, we investigated the effects of three commonly used pesticides—imidacloprid (IMI), cycloxaprid (CYC), and tebuconazole (TBZ)—both individually and in combination, using various biomarkers to assess their impact on daphnia. Our findings indicated that the order of toxicity, from highest to lowest, was TBZ, IMI, and CYC, as determined by acute toxicity as well as reproduction. The effects of the ITmix (IMI and TBZ) and CTmix (CYC and TBZ) combinations on immobilization and reproduction were evaluated by MIXTOX, revealing a higher risk of immobilization at low concentrations for ITmix. The effect on reproduction differed depending on the ration of pesticides in the mixture, with synergism observed, which may be caused mainly by IMI. However, CTmix showed antagonism for acute toxicity, with the effect on reproduction depending upon the composition of the mixture. The response surface also exhibited a switch between antagonism and synergism. Additionally, the pesticides extended the body length and inhibited the development period. The activities of superoxide dismutase (SOD) and catalase (CAT) content was also significantly induced at different dosage points in both the single and combination groups, indicating changes in the metabolic capabilities of detoxifying enzymes and target site sensitivity. These findings highlight the need for more attention to be focused on the effects of pesticide mixtures. Keywords:ecotoxicity;pesticide mixture;combined toxicity;MIXTOX;risk assessment

Polystyrene microplastics facilitating reproductive toxicity and transgenerational transfer of ZnO nanoparticles in daphnia under chronic dietary exposure

Jingyi Guo,Na Liu,Jingfu Liu,Jianjian Wu,Fei Ge

Microplastics (MPs) act as vectors for co-existing pollutants, altering their environmental behavior and ecological risks. Yet the impact of MPs on the chronic effects of co-existing pollutants remains poorly understood. Herein, we explored the role of polystyrene (PS) MPs in facilitating the transgenerational transfer (F 0 -F 4 ) and reproductive toxicity of zinc oxide nanoparticles (ZnO NPs) in Daphnia magna ( D. magna ) under chronic dietary exposure. Results showed that PS MPs facilitated the transgenerational transfer of ZnO NPs, resulting in prolonged transport of ZnO NPs by one generation. This was attributed to that PS MPs impaired Zn excretion of D. magna via significantly inhibiting the molting rates by 40% ( p  < 0.05), promoting the transfer of Zn to offspring. Besides, confocal microscopy confirmed Zn enrichment in the ovum and umbilical cords. Molecular docking simulation revealed stable Zn 2 + binding to vitellogenin (VTG) protein via hydrogen bonds (-1200 kJ/mol), indicating VTG-mediated maternal transfer of Zn. Furthermore, PS MPs facilitated the multi-generational reproductive impact of ZnO NPs to extend to the fourth generation (F 3 ). Transcriptomic analysis identified significant inhibition of prolactin signaling pathways, with the key genes Signal Transducer and Activator of Transcription ( STAT ) and VTG decreasing by 80.4% and 7.8% in the female generation (F 0 ), respectively, inhibiting oocyte development. Overall, these findings confirmed that MPs facilitated the reproductive toxicity and transgenerational transfer of co-existing pollutants, providing critical insights for long-term ecological risk assessment of MPs and co-existing pollutants combined exposure in aquatic systems.

Evaluation of the subtle effects and oxidative stress response of chloramphenicol, thiamphenicol, and florfenicol in Daphnia magna

Zhang Yuxuan,Guo Peiyong,Wu Yanmei,Zhang Xiaoyan,Wang Meixian,Yang Simin,Sun Yinshi,Deng Jun,Su Haitao

Phenicol antibiotics, such as chloramphenicol, thiamphenicol, and florfenicol, are commonly used in the veterinary and aquaculture fields to treat diseases and have frequently been detected in aquatic environments. Nevertheless, there is limited information regarding the effects of phenicol antibiotics on aquatic nontarget species. Thus, the present study aims to investigate the long-term (21-d) influence on the reproduction and growth of and the acute (24-h) oxidative response and tissue damage in the crustacean Daphnia magna after exposure to phenicol drugs, including their environmental concentrations. The results indicate that D. magna exposed to florfenicol are likely to cause more adverse effects than those exposed to chloramphenicol and thiamphenicol over long-term (21-d) exposures. Furthermore, changes in biochemical biomarkers such as malondialdehyde (MDA), catalase (CAT), and reduced glutathione (GSH) induced by individual and mixtures of phenicol antibiotics were also observed. Low concentrations of chloramphenicol, thiamphenicol + florfenicol, and chloramphenicol + thiamphenicol significantly increased the MDA levels of D. magna after 24-h exposures, causing cellular oxidative damage in the animals. In addition, discrepancies between CAT activities and GSH levels were observed, underscoring the need to evaluate multiple indicators of oxidative stress in toxicological studies using D. magna as a model. Acute and Chronic Toxicity of Carbamazepine on the Release of Chitobiase, Molting, and Reproduction in Daphnia similis

Huihui Chen,Xiaohong Gu,Qingfei Zeng,Zhigang Mao

As one of the most frequently detected pharmaceutical compounds in aquatic environments, carbamazepine (CBZ) has recently been shown to cause acute and chronic toxicity in a variety of non-target aquatic organisms. However, little is known about the ecotoxicological effects it has on the molting and reproduction of crustaceans. The aim of the present work was to evaluate the acute and chronic toxic responses to CBZ in the crustaceanDaphnia similis. After acute exposure (4 days), CBZ did not cause lethal toxicity at the tested concentrations. However, CBZ did inhibit the molting and release of chitobiase at concentrations higher than 6.25 μg/L, with 96 h EC50(median effective concentration) values of 864.38 and 306.17 μg/L, respectively. The results of chronic exposure showed that the mean number of molts, size of the first brood, mean number of offspring per brood, mean number of broods per female, and total offspring per female decreased significantly with increasing CBZ concentrations. Significant effects of CBZ on the molting or fecundity inD. similiswere observed even at concentrations as low as 0.03 μg/L. In conclusion, CBZ can cause inhibition of molting, delayed reproduction, and reduced fecundity inD. similis. CBZ toxicity toD. similisdepends on the timing and duration of the exposure. Moreover, our results indicated that CBZ would act as an endocrine disrupter inD. similis, as with vertebrates (e.g., fish). Keywords:carbamazepine;molting;Daphnia similis;chitobiase;chronic toxicity;reproduction

Effects of suspended common-scale and nanoscale particles on the survival, growth and reproduction of Daphnia magna

Xiaoyan Zhang,Peiyong Guo,Jinxiu Huang,Xiufu Hou

Suspended particles are a natural component of aquatic ecosystems. This study provides a report on the survival, growth and reproduction of common-scale and nanoscale particles of Daphnia magna Straus exposed to five types of particles (i.e. KN (kaolinite), MN (montmorillonite), MNn (nanoscale MN), NP (natural particles), and NPn (nanoscale NP)). The results of the study show that the suspended particles elicited a dose-dependent toxicity in KN, MN and MNn, with the following toxicity pattern: MN   >   KN   >   MNn. On the contrary, NP and NPn did not show any harmful effects on the animals. Instead, NP and NPn, especially NPn, contributed to the survivorship of the animals. The animals were able to survive throughout the 21-day period of bioassays, and when the particle concentration reached 600   mg   L −1 , they produced many neonates without any addition of food. This experimental results also indicated that the nanoscale particles were less toxic than the common-scale ones, both for MN and NP. Moreover, the results of the 21-day period of bioassays indicated that when the organisms were exposed to particles of different size scales, they showed different selection patterns for allocating resources. This may be due to the generation of different assimilation and digestion patterns.

Altered life history traits and transcripts of molting- and reproduction-related genes by cadmium in Daphnia magna

Wei Xinrong,Li Xiao,Liu Hongsong,Lei Haojun,Sun Weijun,Li Dan,Dong Wu,Chen Hongxing,Xie Lingtian

Cadmium (Cd) is a non-essential element and can be toxic to aquatic organisms at low concentrations. Despite its well-known toxicity to Daphnia magna , the effects of Cd on physiological parameters (heart rate and thoracic limb activity) and molting- and reproduction-related genes are relatively understudied. In this study, D. magna were exposed to 0 (control), 25, 50 and 75 μg L −1 of Cd for 7 d and 21 d to determine the toxicity of Cd. The results showed that the Cd body burden in D. magna was significantly increased with elevated Cd concentrations, up to 13.4 μg Cd/g dry weight (dw) after exposure to 75 μg L −1 for 21 d. After 21 d of exposure, the body length and body weight of D. magna were significantly decreased in all Cd treatments compared to the control. The heart rate and thoracic limb activity were reduced by 4.3–11.7 and 5.0–10.3%, respectively. The levels of malondialdehyde (MDA) were increased by ~24–37% and the activity of catalase (CAT) was inhibited by ~50% compared to the control. The reproductive parameters (i.e., size of the first brood, the total number of offspring per female and the number of offspring per brood) were remarkably reduced, causing adverse effects on the population dynamics. In addition, the transcripts of genes ( cyp314 , cyp18a1 , ecra , usp , hr3 , cut , cht and cht3 ) related to the molting of D. magna were altered, whereas the transcripts of genes ( vtg1 , vtg2 and vmo1 ) related to reproduction were down-regulated. This study helps better understand the effects of Cd at different biological levels.

Evaluation of the Acute Effects and Oxidative Stress Responses of Phenicol Antibiotics and Suspended Particles in Daphnia magna

Yuxuan Zhang,Peiyong Guo,Yanmei Wu,Meixian Wang,Jun Deng,Haitao Su,Yinshi Sun

Suspended particles (SP) exist widely in various water systems and are able to adsorb other pollutants in water, producing ecotoxic effects on aquatic nontarget species. Until now, however, few studies have focused on the effects of SP on antibiotics. Therefore, the present study investigated the effects of the mixtures of SP and phenicol antibiotics (chloramphenicol [CAP], thiamphenicol [TAP]) on acute toxicity and oxidative stress responses in Daphnia magna . The results indicated that the acute toxicity of phenicol antibiotics in D. magna was increased when combined with SP. Besides, the immobilization of daphnids caused by phenicol drugs in the presence of 10 mg/L of SP was more intense than that with 200 mg/L of SP. Furthermore, the impact of SP with diverse concentrations on the activity of catalase and the level of reduced glutathione in D. magna was different. Notably, almost all CAP + TAP + SP treatments markedly increased malondialdehyde content in D. magna , causing potential cellular oxidative damage in D. magna . In summary, the present study provides insights into the toxic effects of phenicol antibiotic and SP mixtures on aquatic organisms. Environ Toxicol Chem 2021;40:2463–2473. © 2021 SETAC

Individual and binary mixture effects of bisphenol A and lignin-derived bisphenol in Daphnia magna under chronic exposure

Dan Li,Hongxing Chen,Ran Bi,Haibo Xie,Yu Zhou,Yongju Luo,Lingtian Xie

In recent years, many new chemicals have been synthesized from biomass with an aim for sustainable development by replacing the existing toxic chemicals with those having similar properties and applications. However, the effects of these new chemicals on aquatic organisms remain relatively unknown. In this study, the effects of bisphenol A (BPA) and lignin-derived bisphenol (LD-BP, a BPA analogue) on Daphnia magna were evaluated. The animals were exposed to BPA, LD-BP, and their binary mixture at concentrations (2–2000 μg L −1 ) for 21 days. The expression of various biochemical markers and the effects on growth, molting, and reproduction parameters were examined. The results showed that the weight of daphnids significantly increased after exposure to BPA, LD-BP, and the binary mixture relative to that of the control animals. The activity of superoxide dismutase was significantly inhibited by LD-BP and the binary mixture. At the highest exposure concentration of the binary mixture, the activities of acetylcholinesterase and α-glucosidase, fecundity, and the number of neonates per brood were significantly altered. Our results showed that the effects of BPA and LD-BP on D. magna were generally comparable, except for the effect on the weight at their environmentally relevant concentrations (e.g., <20 μg L −1 ). The effects on the reproduction of D. magna could be mainly due to the shift in energy redistribution under BPA and LD-BP exposures. Our results implied that exposures to both BPA and LD-BP could potentially cause deleterious effects at the population level in D. magna .

Joint effect of nanoplastics and humic acid on the uptake of PAHs for Daphnia magna: A model study

Wei Lin,Ruifen Jiang,Xiaoying Xiao,Jiayi Wu,Songbo Wei,Yan Liu,Derek C.G. Muir,Gangfeng Ouyang

Nanoplastics (NPs) are emerging pollutants which can adsorb large amounts of hydrophobic organic compounds (HOCs) and be ingested by aquatic organisms. NPs interact with dissolved organic matter (DOM) and result in significant impacts on the bioaccumulation of HOCs in the actual environment. For the first time, the joint effects of two complex matrices on the bioaccumulation of polycyclic aromatic hydrocarbons (PAHs) to Daphnia magna ( D. magna ) were studied by modeling calculation. The complex matrices, nano-sized polystyrene (PS) and/or humic acid (HA), were under environmentally realistic concentrations. A biodynamic model was modified and the uptake fluxes from all exposure pathways were quantified using the experimental data. A flux estimation showed that the bioaccumulation amounts at equilibrium were mostly dependent on dermal uptake (≥99.3 % of the total). The PS matrix would retard the intestinal uptake process in D. magna , especially for the less hydrophobic PAHs; while the HA or the HA-PS matrix would facilitate the mass transfer of PAHs from the matrix to lipids in the gut. Moreover, the biota matrix accumulation factor (BMAF) were calculated to verify the biodynamic model. This work is helpful to clarify the bioaccumulation effects of PAHs in complex environmental systems.
