Remarkable Enhancement of Antioxidant Activity of the Ovalbumin–EGCG Conjugate through a Novel Preceding Selective Protection Grafting Strategy

Yumin Wang,Songbai Liu

Conventional radical grafting of proteins with catechins consumed the most antioxidant-active hydroxyls during grafting, thus failing to effectively retain antioxidant activity in conjugates. In this study, a novel strategy of selective protection of the most reactive hydroxyls before grafting was developed to preserve the most reactive hydroxyls and effectively retain antioxidant activity in conjugates. Selective protection of the most reactive hydroxyls of (−)-epigallocatechin-3-gallate (EGCG) was successfully realized in a yield of 87% applying trimethyl orthopropionate and catalytic calcium triflate at 40 °C. The novel ovalbumin (OVA)–EGCG conjugate with 93% grafting ratio was prepared by radical grafting with the selectively protected EGCG and subsequent deprotection. Substantially enhanced antioxidant performance of the novel OVA–EGCG conjugate in liposomes was unveiled with notably reduced curcumin degradation and leakage. The strategy and approaches developed in this study will be valuable to effectively improve the antioxidant activities of protein–catechin grafting conjugates.

Multifunctional Liposomes Co-encapsulating Epigallocatechin-3-gallate (EGCG) and miRNA for Atherosclerosis Lesion Elimination

Chunying Cui,Shuang Zhang,Dandan Li,Danni Liu,Yaoqi Wang,Qi Sun,Ran Sun,Jie Zhang,Xiaoxuan Hong,Ran Huo

Atherosclerosis is a chronic inflammatory disease, characterized by a lipid accumulated plaque. Anti-oxidative and anti-inflammation as well as lipid metabolism promoting therapeutic strategies have been applied for atherosclerosis treatment. However, the therapeutic effect by a single therapeutic method is limited. Combination of the two strategies could for one thing prevent lipid accumulation cause by inflammation and oxidative stress, for another thing promote lipid efflux from atherosclerotic plaque, to normalize arteries maximumly. Hence, a multifunctional liposome co-encapsulating an antioxidant and anti-inflammatory drug epigallocatechin-3-gallate(EGCG) as well as a lipid-efflux promoting gene miR-223 was established. The system (lip@EGCG/miR-223) could encapsulate miR-223 in core areas of the liposomes to provide an effective protective effect for gene drugs. Moreover, lip@EGCG/miR-223 was smaller in size (91.28±2.28 nm characterized by DLS), making it easier to target to AS lesions which had a smaller vascular endothelial spaces. After internalized into cellular efficiently, lip@EGCG/miR-223 exhibited excellent antioxidant and anti-inflammatory effects in vitro by eliminating overproduced ROS and decreasing the level of inflammatory cytokines (TNF-α, IL-1β and MCP-1), which is due to the effect of EGCG. Besides, the lipid-efflux promoting protein ABCA1 was up-regulated when treated with lip@EGCG/miR-223. By the two therapies mentioned, lip@EGCG/miR-223 could effectively inhibit the formation of foam cells, which is a main component of atherosclerotic plaques. In AS model mice, after intravenous (i.v.) administration, lip@EGCG/miR-223 was effectively accumulated in atherosclerotic plaques, and the distribution of drugs in heart and aorta to that in kidney was significantly increased when compared with free drugs (the ratio was 6.27% for free miR-223 treated group, and was increased to 66.10% for lip@EGCG/miR-223 treated group.). By decreasing inflammation level and lipid accumulation, the arterial vessels in AS were normalized with less macrophages and micro-angiogenesis when treated with lip@EGCG/miR-223. Overall, this study demonstrates lip@EGCG/miR-223 can be developed as a potential system for atherosclerosis treatment by a combined treatment of antioxidant, anti-inflammatory and lipid-efflux promoting effects, which provides a novel strategy for safe and efficient management of atherosclerosis.

Enhancement of rheological properties and oxidative stability of OSA starch oleogels by chitosan-EGCG conjugates: Influence of chitosan viscosity

Wenbo Miao,David Julian McClements,Junqing Han,Hangyan Ji,Zhengyu Jin,Chao Qiu

The rheology and oxidative stability of oleogels are critical factors impacting their application as solid fat substitutes in the food industry. For this reason, the influence of chitosan-EGCG conjugates on the rheology and oxidative stability of OSA-starch (OS) oleogels was examined. Conjugates were prepared from chitosan ingredients with different viscosities (low viscosity: <200 mPa s and high viscosity: >400 mPa s). Chitosan-EGCG conjugates (HCE) prepared from the high viscosity chitosan provided the highest antioxidant activity. Moreover, oleogel containing a high concentration of HCE (OS-HCE 1 ) had the highest viscoelasticity, thixotropic recovery (91.79 %), and oil-holding capacity (90.95 %), mainly attributed to the formation of more stable protective coatings. The oxidative stability analysis showed that HCE was most effective in delaying lipid oxidation of OS oleogels during storage. Finally, the potential of using the optimized oleogels (OS-HCE 1 ) as fat substitutes in bread products was evaluated by partially or fully substituting fat at levels of 0 %, 25 %, 50 %, 75 %, and 100 %. The results indicated that up to 50 % of commercial margarine could be replaced with oleogel in bread formulations without adversely affecting key quality parameters, including color, shape, texture, and sensory acceptability. In conclusion, this study offers an effective approach for improving both the structural and oxidative stability of OSA starch-based oleogels and provides valuable guidance for the formulation of healthier lipid systems in bakery applications.

Stability of glycosylated complexes loaded with Epigallocatechin 3-gallate (EGCG)

Jianyong Zhang,Hongchun Cui,Jiahuan Qiu,Xiaoqing Wang,Yixin Zhong,Caiping Yao,Lanying Yao,Qunxiong Zheng,ChunHua Xiong

The application of Epigallocatechin-3-gallate (EGCG) in food industry was limited by its low stability in aqueous solutions and poor bioavailability in vivo . The novel EGCG glycosylated arachin nanoparticles (Ara-CMCS-EGCG) and EGCG glycosylated casein nanoparticles (Cas-CMCS-EGCG) were prepared to improve the stability and bioavailability of EGCG. The effect of different variables on the storage stability and the slow-release behavior of novel glycosylation complexes in nanoparticle background solution and artificial gastrointestinal fluid were investigated. The results showed that the DPPH scavenging activity of Ara-CMCS-EGCG and Cas-CMCS-EGCG were stable in temperature (25 ∼ 70 °C). EGCG could enhance the crosslinking effect of molecular particles in glycosylation complexes solution. The glycosylated protein nanoparticles were stable to acid-base and enzymolysis in simulated gastrointestinal fluid. The release rate of EGCG in simulated intestinal fluid was higher than that in simulated gastric fluid. The glycosylated protein carrier can not only release EGCG slowly, but also significantly improve the stability and bioavailability of EGCG in simulated gastrointestinal fluid.

Preparation of β-lactoglobulin/gum arabic complex nanoparticles for encapsulation and controlled release of EGCG in simulated gastrointestinal digestion model

Jian Gao,Yuezhong Mao,Chuyue Xiang,Mengna Cao,Gerui Ren,Kuiwu Wang,Xiangjuan Ma,Di Wu,Hujun Xie

In this work, the β-lactoglobulin/gum arabic (β-Lg-GA) complexes were prepared to encapsulate epigallocatechin gallate (EGCG), forming β-Lg-GA-EGCG complex nanoparticles with an average particle size of 133 nm. The β-Lg-GA complexes exhibited excellent encapsulation efficiency (84.5%), and the antioxidant performance of EGCG in vitro was improved after encapsulation. It was recorded that 86% of EGCG could be released in simulated intestinal fluid after 3 h of digestion, much faster than that in simulated gastric fluid, indicating that the β-Lg-GA complexes were effective in enhancing EGCG stability, which was confirmed using SDS-PAGE and SEM. Further spectrum results demonstrated that various intramolecular interactions including electrostatic, hydrophobic and hydrogen bonding interactions contribute to the formation of β-Lg-GA-EGCG complex nanoparticles. Also, XRD   experiments indicated that EGCG was successfully encapsulated by β-Lg-GA complexes. Therefore, the β-Lg-GA complexes hold great potentials in the protective delivery of sensitive bioactives.

Antioxidant activity, storage stability and in vitro release of epigallocatechin-3-gallate (EGCG) encapsulated in hordein nanoparticles

Hongdong Song,Qingyu Wang,Aijing He,Sen Li,Xiao Guan,Yawen Hu,Siyi Feng

Hordein nanoparticles have been successfully fabricated to encapsulate EGCG. The present study aimed to investigate the effect of hordein nanoparticles encapsulation on antioxidant activity, storage stability and in vitro release of EGCG. The antioxidant activities of EGCG before and after encapsulation by hordein nanoparticles had no significant difference. The 28-day storage experiments indicated that EGCG-hordein nanoparticles had a good storage stability in terms of average diameter and zeta potential. Meanwhile, the stability of EGCG was significantly improved by hordein nanoparticles encapsulation. The remaining amount of encapsulated EGCG was 1.31-fold and 1.52-fold higher than that of free EGCG at 4 °C and 25 °C, respectively. The release study in the simulated gastric or intestinal fluids suggested that hordein nanoparticles could slow down the release of EGCG in the simulated gastric or intestinal fluids. These results showed that encapsulation of EGCG using hordein nanoparticles could be a promising approach to improve its stability.

Epigallocatechin gallate (EGCG) attenuates myocardial hypertrophy and fibrosis induced by transverse aortic constriction via inhibiting the Akt/mTOR pathway

Yue Cui,Yongqiang Wang,Gang Liu

Context Epigallocatechin gallate (EGCG) is the most abundant catechin from tea. Previous studies have indicated EGCG has a cardioprotective effect. Objective This manuscript mainly explores the role of EGCG in pressure-overload cardiac hypertrophy and its mechanism related to the Akt/mTOR pathway. Methods and methods Transverse aortic constriction (TAC) was utilized to establish the cardiac hypertrophy mice model. C57BL/6 mice were assigned into 6 groups. Starting from the first day after surgery, mice received different doses of EGCG (20, 40, 80 mg/kg) or vehicle orally for four weeks. Heart weight to body weight (HW/BW) ratio and heart weight to tibia length (HW/TL) ratio as well as hematoxylin-eosin staining were utilized to evaluate cardiac hypertrophy. Masson’s trichrome and Sirius red staining were used to depict cardiac fibrosis. The expressions of fibrosis and hypertrophy-related markers and Akt/mTOR pathway were quantified by western blot and qRT-PCR.Results EGCG significantly attenuated cardiac function shown by decreased HW/BW (TAC, 6.82 ± 0.44 vs. 20 mg/kg EGCG, 5.53 ± 0.45; 40 mg/kg EGCG, 4.79 ± 0.32; 80 mg/kg EGCG, 4.81 ± 0.38) and HW/TL (TAC, 11.94 ± 0.69 vs. 20 mg/kg EGCG, 11.44 ± 0.49; 40 mg/kg EGCG, 8.83 ± 0.58; 80 mg/kg EGCG, 8.98 ± 0.63) ratios as well as alleviated cardiac histology. After treatment, hemodynamics was improved, cardiac fibrosis was attenuated. The activated Akt/mTOR pathway was inhibited by EGCG.Discussion and conclusions EGCG plays a protective role in the TAC model by regulating the Akt/mTOR pathway, which provides a theoretical basis for its clinical treatment.

Pulsed electric field improves the EGCG binding ability of pea protein isolate unraveled by multi-spectroscopy and computer simulation

Ze-Ling Chen,Ying Li,Jin-Hua Wang,Rui Wang,Yong-Xin Teng,Jia-Wei Lin,Xin-An Zeng,Meng-Wai Woo,Ling Wang,Zhong Han

Understanding molecular mechanisms during protein modification is critical for expanding the application of plant proteins. This study investigated the conformational change and molecular mechanism of pea protein isolate (PPI) under pulsed electric field (PEF)-assisted (-)-Epigallocatechin-Gallate (EGCG) modification. The flexibility of PPI was significantly enhanced after PEF treatment (10 kV/cm) with decrease (23.25 %) in α-helix and increase (117.25 %) in random coil. The binding constant and sites of PEF-treated PPI with EGCG were increased by 2.35 times and 10.00 % (308 K), respectively. Molecular docking verified that PEF-treated PPI had more binding sites with EGCG (from 4 to 10). The number of amino acid residues involved in hydrophobic interactions in PEF-treated PPI-EGCG increased from 5 to 13. PEF-treated PPI-EGCG showed a significantly increased antioxidant activity compared to non-PEF-treated group. This work revealed the molecular level of PEF-assisted EGCG modification of PPI, which will be significant for the application of PPI in food industry.

EGCG inhibits CTGF expression via blocking NF-κB activation in cardiac fibroblast

Yi Cai,Shan-Shan Yu,Ting-Ting Chen,Si Gao,Biao Geng,Yang Yu,Jian-Tao Ye,Pei-Qing Liu

Connective tissue growth factor (CTGF) has been reported to play an important role in tissue fibrosis and presents a promising therapeutic target for fibrotic diseases . In heart, inappropriate increase in level of CTGF promotes fibroblast proliferation and extracellular matrix (ECM) accumulation, thereby exacerbating cardiac hypertrophy and subsequent failure. Epigallocatechin-3-gallate (EGCG), the major polyphenol found in green tea, possesses multiple protective effects on the cardiovascular system including cardiac fibrosis . However, the molecular mechanism by which EGCG exerts its anti-fibrotic effects has not been well investigated. In this study, we found that EGCG could significantly reduce collagen synthesis , fibronectin (FN) expression and cell proliferation in rat cardiac fibroblasts stimulated with angiotensinII (AngII). It also ameliorated cardiac fibrosis in rats submitted to abdominal aortic constriction (AAC). Moreover, EGCG attenuated the excessive expression of CTGF induced by AAC or AngII, and reduced the nuclear translocation of NF-κB p65 subunit and degradation of IκB-α. Subsequently, we demonstrated that in cardiac fibroblasts NF-κB inhibition could suppress AngII-induced CTGF expression. Taken together, these findings provide the first evidence that the effect of EGCG against cardiac fibrosis may be attributed to its inhibition on NF-κB activation and subsequent CTGF overexpression, suggesting the therapeutic potential of EGCG on the prevention of cardiac remodeling in patients with pressure overload hypertrophy.

A Supramolecular Assembly of EGCG for Long-Term Treatment of Allergic Rhinitis

Fu Zhong Zhang,Min Tan,Jing Zeng,Xu-Wei Qi,Ye-Tao Zhang,Yu-Ting Che,Sheng Zhang,Bang-Jing Li

Allergic rhinitis (AR) is a type-I hypersensitivity disease mediated by immunoglobulin E (IgE). Although antihistamines, glucocorticoids, leukotriene receptor antagonists, and other drugs are widely used to treat AR, the various adverse side effects of long-term use of these drugs should not be ignored. Therefore, more effective and safe natural alternative strategies are urgently needed. To this end, this study designed a nanosupramolecular delivery system composed of β-cyclodextrin supramolecular polymer (PCD), thiolated chitosan (TCS), and natural polyphenol epigallocatechin gallate (EGCG) for intranasal topical continuous treatment of AR. The TCS/PCD@EGCG nanocarriers exhibited an excellent performance in terms of retention and permeability in the nasal mucosa and released the vast majority of EGCG responsively in the nasal microenvironment, thus resulting in the significantly high antibacterial and antioxidant capacities. According to the in vitro model, compared with free EGCG, TCS/PCD@EGCG inhibited mast cell activity and abnormal histamine secretion in a more long-term and sustained manner. According to the in vivo model, whether in the presence of continuous or intermittent administration, TCS/PCD@EGCG substantially inhibited the secretion of allergenic factors and inflammatory factors, mitigated the pathological changes of nasal mucosa, alleviated the symptoms of rhinitis in mice, and produced a satisfactory therapeutic effect on AR. In particular, the therapeutic effect of TCS/PCD@EGCG systems were even superior to that of budesonide during intermittent treatment. Therefore, the TCS/PCD@EGCG nanocarrier is a potential long-lasting antiallergic medicine for the treatment of AR.

EGCG-NPs inhibition HO-1-mediated reprogram iron metabolism against ferroptosis after subarachnoid hemorrhage

Liyong Huang,Xue Wang,Yanning Zheng,Dongcen Lang,Jian Wang,Shuaiguo Yan,Ying Chen

Subarachnoid hemorrhage (SAH), a devastating disease with a high mortality rate and poor outcomes, tightly associated with the dysregulation of iron metabolism and ferroptosis. (-)-Epigallocatechin-3-gallate (EGCG) is one of major bioactive compounds of tea catechin because of its well-known iron-chelating and antioxidative activities. However, the findings of iron-induced cell injuries after SAH remain controversial and the underlying therapeutic mechanisms of EGCG in ferroptosis is limited. Here, the ability of EGCG to inhibit iron-induced cell death following the alleviation of neurological function deficits was investigated by using in vivo SAH models. As expected, EGCG inhibited oxyhemoglobin (OxyHb)-induced the over-expression of HO-1, which mainly distributed in astrocytes and microglial cells. Subsequently, EGCG blocked ferrous iron accumulation through HO-1-mediated iron metabolic reprogramming. Therefore, oxidative stress and mitochondrial dysfunction was rescued by EGCG, which resulted in the downregulation of ferroptosis and ferritinophagy rather than apoptosis after SAH. As a result, EGCG exerted the superior therapeutic effects in the maintenance of iron homeostasis in glial cells, such as astrocytes and microglial cells, as well as in the improvement of functional outcomes after SAH. These findings highlighted that glial cells were not only the iron-rich cells in the brain but also susceptible to ferroptosis and ferritinophagy after SAH. The detrimental role of HO-1-mediated ferroptosis in glial cells can be regarded as an effective therapeutic target of EGCG in the prevention and treatment of SAH.

Ultrasound-assisted preparation of lactoferrin-EGCG conjugates and their application in forming and stabilizing algae oil emulsions

Sairui Zhang,Xueqi Li,Xiaojia Yan,David Julian McClements,Cuicui Ma,Xuebo Liu,Fuguo Liu

The aim of this study was to prepare lactoferrin-epigallocatechin-3-gallate (LF–EGCG) conjugates and to determine their ability to protect emulsified algal oil against aggregation and oxidation. LF–EGCG conjugates were formed using an ultrasound-assisted alkaline treatment. The ultrasonic treatment significantly improved the grafting efficiency of LF and EGCG and shortened the reaction time from 24 h to 40 min. Fourier transform infrared spectroscopy and circular dichroism spectroscopy analyses showed that the covalent/non-covalent complexes could be formed between LF and EGCG, with the C O and C N groups playing an important role. The formation of the conjugates reduced the α-helix content and increased the random coil content of the LF. Moreover, the antioxidant activity of LF was significantly enhanced after conjugation with EGCG. LF–EGCG conjugates as emulsifiers were better at inhibiting oil droplet aggregation and oxidation than LF alone. This study demonstrates that ultrasound-assisted formation of protein–polyphenol conjugates can enhance the functional properties of the proteins, thereby extending their application as functional ingredients in nutritionally fortified foods.
