Orange emissive carbon dots for fluorescent determination of hypoxanthine in fish

Zehuai Mou,Zhijin Gao,Yaoping Hu

Sensitive determination of hypoxanthine (HX), an indicator reflecting the degradation of meat and fish, is significantly important in monitoring food freshness. Herein, we design a novel sensor consisting of orange emissive carbon dots (O-CDs), nitrotetrazolium blue chloride (NTBC), and xanthine oxidase (XOD) for fluorescence turn-off detection of HX. O-CDs, possessing a high fluorescence quantum yield of 37%, are synthesized by hydrothermal treatment of 2,3-diaminopyridine in sulfuric acid. NTBC can react with HX/XOD-generated H 2 O 2 and O 2 – to yield a violet-colored formazan, which remarkably quenches the orange fluorescence of O-CDs through inner filter effect. There is a linearity between the quenching efficiency and HX concentration in the range of 2–250 μM, and the limit of detection is 0.61 μM, lower than those of most reported HX sensors. In addition, the proposed method exhibits excellent selectivity, and can be applied to quantify HX in fish samples with satisfactory results.

Preparation and properties of caffeic-chitosan grafting fish bone collagen peptide

Xuqin Li,Zhou Lei,Jie Sheng,Yishan Song

In this study, a novel peptide grafted chitosan (CACS-FBP) with high peptide content, excellent moisture-absorption and moisture-retention abilities was prepared. Caffeic acid (CA) was used to modify chitosan, the highly water-soluble intermediate further reacted with fish bone collagen peptide to obtain the final product, and the synthesis of CACS-FBP was confirmed by the Fourier transform infrared spectroscopy (FT-IR), NMR, and UV-vis. The single-factor experiments indicated that the degree of substitution (DS) of CACS-FBP depended on the reaction temperature, reaction time, the mass ratio of fish bone collagen peptide to CACS (mFBP/mCACS) and the mass ratio of MTGase to CACS (mMTGase/mCACS). In addition, the antioxidant assay indicated that CACS-FBP had an excellent antioxidant capacity, and the CACS-FBP showed no cytotoxicity toward L929 mouse fibroblasts, all the results mean that the prepared peptide-containing chitosan derivative has potential application in pharmaceutical and biomedical fields.

Discovery of mass spectral peak markers and protein biomarkers in fish muscle exudates for rapid and precise recognition of fish species via magnetic beads (MBs) and mass spectrometry

Weijiao Chen,Winnie C. Soko,Jing Xie,Hongyan Bi

In this study, muscle exudates from five fishes belonging to the family Sciaenidae, in the order Perciformes, were analyzed as models for the discovery of biomarkers by matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS). MagSi-weak cation exchange magnetic beads (WCX-MBs) were utilized for the enrichment of proteins from fish exudate samples, allowing protein biomarkers to be identified and subsequently used for fish species differentiation. Buffers with pH ranging from 4.0 to 9.0 can provide an environment for proteins in fish muscle exudate to bind to the WCX-MBs. The optimal enrichment based on WCX-MBs can be achieved when the exudate samples are diluted 100folds. More species-specific biomarkers in mass spectra can be identified when using WCX-MBs. The number of ions that can be considered as peak markers and can differentiate the analyzed fishes increases from 38 to 121 when using WCX-MBs to isolate peptides/protein in fish muscle exudate. Particularly, eight peak markers in mass spectra were assigned to be specific to Nibea albiflora (NA), three peak markers specific to Larimichthys crocea (LC), two peak markers specific to Miichthys miiuy (MM), seven peak markers specific to Collichthys lucidus (CL), and six peak markers specific to Larimichthys polyactis (LP). Furthermore, five proteins were identified based on the characterization of tryptic peptides and their potential to be biomarkers, of which four proteins specific to CL and one specific to LC were identified. The single-blind samples analysis demonstrated that these species-specific peak markers and protein biomarkers can be successfully utilized for corresponding fish recognition. The utilization of WCX-MBs can improve the discovery of fish species-specific biomarkers in fish muscle exudate samples. The present protocol holds potential of being a rapid and accurate identification tool for recognition of fish species.

Divergent Antiviral Mechanisms of Two Viperin Homeologs in a Recurrent Polyploid Fish

Mou Cheng-Yan,Li Shun,Lu Long-Feng,Wang Yang,Yu Peng,Li Zhi,Tong Jin-Feng,Zhang Qi-Ya,Wang Zhong-Wei,Zhang Xiao-Juan,Wang Guang-Xin,Zhou Li,Gui Jian-Fang

Polyploidy and subsequent diploidization provide genomic opportunities for evolutionary innovations and adaptation. The researches on duplicated gene evolutionary fates in recurrent polyploids have seriously lagged behind that in paleopolyploids with diploidized genomes. Moreover, the antiviral mechanisms of Viperin remain largely unclear in fish. Here, we elaborate the distinct antiviral mechanisms of two viperin homeologs (Cgviperin-A and Cgviperin-B) in auto-allo-hexaploid gibel carp (Carassius gibelio). First, Cgviperin-A and Cgviperin-B showed differential and biased expression patterns in gibel carp adult tissues. Subsequently, using co-immunoprecipitation (Co-IP) screening analysis, both CgViperin-A and CgViperin-B were found to interact with crucian carp (C. auratus) herpesvirus (CaHV) open reading frame 46 right (ORF46R) protein, a negative herpesvirus regulator of host interferon (IFN) production, and to promote the proteasomal degradation of ORF46R via decreasing K63-linked ubiquitination. Additionally, CgViperin-B also mediated ORF46R degradation through autophagosome pathway, which was absent in CgViperin-A. Moreover, we found that the N-terminal α-helix domain was necessary for the localization of CgViperin-A and CgViperin-B at the endoplasmic reticulum (ER), and the C-terminal domain of CgViperin-A and CgViperin-B was indispensable for the interaction with degradation of ORF46R. Therefore, the current findings clarify the divergent antiviral mechanisms of the duplicated viperin homeologs in a recurrent polyploid fish, which will shed light on the evolution of teleost duplicated genes.

Repurposing auranofin combined with colistin to effectively combat fish-pathogenic Edwardsiella piscicida

Jinfang Lu,Yangbin Shi,Yajing Pan,Hans-Peter Grossart,Luyu Mei,Danli Xie,Wanchun Guan,He Zhang,Yongliang Lou

Edwardsiella piscicida ( E. piscicida ) is an important opportunistic fish pathogen, and infects various species of cultured fish posing a great challenge to the aquaculture industry globally. To address the frequent emergence of multi-antibiotics resistant E. piscicida and the shortage of effective antimicrobial treatments, this study aimed to develop a novel combination therapy against E. piscicida . The combination of the FDA approved drug auranofin (originally used to treat rheumatoid arthritis) with colistin exerted great bactericidal effects against E. piscicida both in vitro and in vivo . Auranofin disrupted the thioredoxin system, and decreased the minimum inhibitory concentration (MIC colistin ) of E. piscicida . Further, auranofin combined with colistin significantly inhibited the growth of E. piscicida isolates, even resulting in cell death. A mechanistic study revealed that the combination remarkably damaged the bacterial membrane increasing its permeability, and it also promoted a decrease of membrane potential, thereby exacerbating oxidative stress and the bactericidal effects of colistin. Importantly, auranofin combined with colistin strongly attenuated the essential bacterial two-component system, lipopolysaccharide modification, motility and virulence pathways. Moreover, the combination of both chemicals also efficiently reduced E. piscicida burden in the gill, intestine and head kidney of infected zebrafish ( Danio rerio ), and significantly improved their survival rate. Overall, these results suggested that the thioredoxin-mediated redox cascade is a potential target for novel antimicrobials, and that auranofin combined with colistin could be repurposed to effectively combat Edwardsiella infections.

Improved oxidative stability of fish oil emulsion by grafted ovalbumin-catechin conjugates

Jin Feng,He Cai,Hua Wang,Chunyang Li,Songbai Liu

Conjugates of ovalbumin (OVA) with (+)-catechin (C), (−)-epigallocatechin (EGC) or (−)-epigallocatechin gallate (EGCG) were synthesized through a free-radical grafting approach. The covalent binding between OVA and catechins was confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). OVA-catechin conjugates were further characterized by a combination of steady-state fluorescence spectroscopy, differential scanning calorimetry (DSC), Fourier transform infrared (FTIR) spectroscopy and circular dichroism (CD) analyses. Results suggested that the conjugates contained less α-helix but more β-sheet secondary structure than OVA. The catechin-grafting reaction led to improved surface hydrophobicity and decreased tertiary conformation stability of OVA. Compared with that emulsified by OVA, fish oil emulsion coated by conjugates demonstrated smaller droplet size, better storage stability and less viscosity. Besides, conjugates inhibited the lipid oxidation in fish oil emulsion more effectively than OVA because of their higher antioxidant activity and interfacial accumulation.

Improving foaming properties of fish gelatin by neutral protease and gum arabic

Chen Yu,Pan Yi,Hu Yu-Ting,Yang Le-Ying,Wang Lin-Ru,Sha Xiao-Mei,Tu Zong-Cai

Neutral protease and gum arabic (GA) were combined to enhance foaming ability (FA) and foaming stability (FS) of fish gelatin. Meanwhile, structural changes of pure, single-modified and combined-modified fish gelatins were evaluated in detail to investigate the mechanism. FA and FS of combined-modified fish gelatin were boosted to the maximum of 158% and 55%, respectively. The enzymolysis enhanced the surface hydrophobicity (increased by 40%) and destroyed the microstructure. On the basis, hydrogen bond and electrostatic forces of combined-modified fish gelatin were strengthened by GA, which elevated viscosity and interfacial protein adsorption content (6 mg/g), enhancing FS and FA. The redshift of amide I and II wavenumbers indicated more stable secondary structure of combined-modified fish gelatin, reflected in the SEM-verified denser foam. The underlying mechanism for improving foaming properties of fish gelatin was related to the opened structure by neutral protease and further noncovalent interaction by GA. New insights for the development of sustainable foaming agents were provided by these findings.

An immunochromatographic sensor for ultrasensitive and direct detection of histamine in fish

Lu Zeng,Xinxin Xu,Lingling Guo,Zhongxing Wang,Hongliu Ding,Shanshan Song,Liguang Xu,Hua Kuang,Liqiang Liu,Chuanlai Xu

To ensure food quality and prevent histamine (HA) toxicity, a rapid and direct method of detecting HA is required. In this work, we prepared a monoclonal antibody (mAb) against HA using a hapten produced by the introduction of a phenyl-containing linker. The novel mAb exhibited high sensitivity against HA as determined by ELISA, with a half-maximal inhibitory concentration of 21.51 ng/mL. A gold nanoparticle-based immunosensor was fabricated for rapid detection of HA in fish samples. After optimizing the immunosensor, a visual limit of detection (LOD) and a calculated LOD were 0.25 mg/kg and 10.48 μg/kg for HA, respectively. Recovery rates from the spiked fish samples ranged from 87.33% to 104.67% with the coefficient of variation below 10.82%. Concurrently, the whole process in testing real sample was completed within 15 min, and all results were well confirmed and comparable by liquid chromatography-mass spectrometry and the commercial test strip. These data revealed that the proposed immunosensor could be used as a monitoring tool for the rapid and direct detection of HA in fish samples.

Enhancing the Antioxidant Activity of Fish Scale Collagen Hydrolysates Through Plastein Reaction

Xu Chengzhi,Cai Chaonan,Liu Tianyi,Kang Jizhen,Li Sheng,Zhang Juntao,Wei Benmei,Wang Haibo

An important area of focus for developing antioxidant collagen hydrolysates (peptides) involves enhancing the antioxidant properties of collagen hydrolysates. In this study, fish scale collagen hydrolysate (FH) was used as the raw material for plastein reaction. Various enzymes, including papain, alcalase, flavourzyme, and the combination of alcalase and flavourzyme, were employed for this purpose. The plastein reaction significantly improved the thermal stability, chemical antioxidant activity, and capacity to scavenge cellular reactive oxygen species of FH. Notably, the plastein reaction catalyzed by alcalase exhibited the most significant improvement, increasing the hydroxyl radical scavenging rate from 72.3 to 93.4% and restoring the viability of the oxidative stress-induced HepG2 cell model from 50.8 ± 1.7 to 74.9 ± 1.7%. During the plastein reaction, condensation and hydrolysis reactions occurred simultaneously, with condensation being the dominant process. These reactions, along with physical aggregation, facilitated the formation of larger yet more concentrated collagen peptide aggregates, leading to increased exposure of hydrophobic groups. This enhanced the uptake of collagen hydrolysates by the cells and contributed to the enhancement of their antioxidant properties. Thus, the plastein reaction is an effective method for enhancing the antioxidant properties of collagen hydrolysates, with its simplicity of operation and promising application potential.

Phosphate structure-dependent modification tailors gelation and interfacial properties of fish gelatin

Jinghong Xu,Xinjing Zhang,Zongcai Tu,Hui Wang

TSPP, STPP, and STMP induced distinct molecular rearrangement pathways. • TSPP reinforced gel networks through enhanced molecular organization. • STPP promoted stable interfaces and improved emulsion stability. • STMP enhanced emulsifying activity via rapid interfacial adsorption. Fish gelatin (FG) is a promising functional biopolymer, but its application in structured food systems is restricted by insufficient gel strength, thermal stability, and interfacial stabilization ability. Although phosphorylation has been widely applied to improve protein functionality, how phosphate structures regulate molecular rearrangement and determine different functional outcomes remains unclear. This study investigated the structure-dependent effects of three phosphate reagents, sodium pyrophosphate (TSPP), sodium tripolyphosphate (STPP), and sodium trimetaphosphate (STMP), on the multiscale structure and functionality of FG. All phosphate treatments successfully introduced phosphorus-containing groups into FG while largely preserving gelatin chain integrity; however, the extent and pattern of molecular restructuring varied among phosphate reagents. TSPP treatment resulted in the highest phosphorus content and was associated with the most pronounced conformational rearrangement, as evidenced by increased ordered secondary structures, altered tertiary microenvironments, enhanced surface hydrophobicity, and formation of a denser gel network with improved mechanical properties. In contrast, functional enhancement was not directly proportional to phosphorus content. STPP generated a more balanced structural state, resulting in improved thermal stability and superior emulsion stabilization, whereas STMP mainly enhanced interfacial adsorption ability but provided limited improvement in long-term emulsion stability. These results reveal that phosphate structure plays a critical role in regulating the structural evolution and functional direction of modified FG, whereas phosphorus content alone cannot fully predict functional performance. Aggregation-disaggregation cycling: A physical strategy for low-viscosity fish protein particles

Xueer Yu,Xinyao Zhao,Bowen Zou,Aijun Jian,Jiaxin Li,Xianbing Xu,Ming Du,Chao Wu,Xiaokang Na

Cyclical physical modification sharply reduces cod protein viscosity. • 62.6% adhesion reduction (AFM: 3.96 → 1.48 nN) yields Newtonian flow at 6% w/v. • Depleted reactive groups drive structural shift to dense molten globules. The development of low-viscosity, high-concentration protein systems is hindered by shear-thickening behavior arising from intermolecular entanglement. We report a cyclical preheating-ultrasonication strategy that reconstitutes cod protein (CPs) into low-adhesion particles. Pre-heating (100 °C, 10 min) partially unfolds CPs to expose hydrophobic nucleation sites, followed by ultrasonication (400 W, 10 min) fragmenting aggregates into metastable subunits via cavitation shear while redistributing surface charges. Force curve analysis validated the adhesion force reduction (62.6%, 3.96 → 1.48 nN) and its direct correlation with significant viscosity reduction at high concentration. The modified CPs exhibit Newtonian fluid behavior at 6% w / v concentration, with the viscosity was reduced to approximately 0.1 Pa·s at a scanning frequency of nearly 1 s −1 . Small-angle X-ray scattering (SAXS) shows a structural shift from loose aggregates to dense molten spheres. This physical reprocessing approach provides a sustainable pathway to engineer high-density protein colloids for injectable formulations and 3D food printing.
