A novel Michael/alkylation cascade reaction of N-unprotected 3-bromooxindoles with α,β-unsaturated acyl phosphonates using DABCO as a robust catalyst followed by the derivatization of the acyl phosphonate intermediates in situ has been developed. This scenario enables rapid access to a diverse set of highly functionalized spirocyclopropyl oxindoles in moderate yields with good to excellent diastereoselectivities, which are analogues of a high active non-nucleoside reverse transcriptase inhibitor against HIV-1. The synthetic potential of this tactic has been highlighted by a gram-scale reaction and Suzuki cross-coupling reactions of the product. Moreover, the reaction mechanism has been tentatively elucidated by control experiments and dynamic high-resolution mass spectrometry studies, which indicates that the Michael/alkylation cascade reaction involves DABCO-derived α-substituted ammonium ylides.

Curcumin is a molecule found in turmeric root that has anti-inflammatory, antioxidant, and anti-tumor properties and has been widely used as both an herbal drug and a food additive to treat or prevent neurodegenerative diseases. To explore whether curcumin is able to ameliorate HIV-1-associated neurotoxicity, we treated a murine microglial cell line (N9) and primary rat cortical neurons with curcumin in the presence or absence of neurotoxic HIV-1 gp120 (V3 loop) protein. We found that HIV-1 gp120 profoundly induced N9 cells to produce reactive oxygen species (ROS), tumor necrosis factor-α (TNF-α) and monocyte chemoattractant protein-1 (MCP-1). HIV-1 gp120 also induced apoptosis of primary rat cortical neurons. Curcumin exerted a powerful inhibitory effect against HIV-1 gp120-induced neuronal damage, reducing the production of ROS, TNF-α and MCP-1 by N9 cells and inhibiting apoptosis of primary rat cortical neurons. Curcumin may exert its biological activities through inhibition of the delayed rectification and transient outward potassium (K + ) current, as curcumin effectively reduced HIV-1 gp120-mediated elevation of the delayed rectification and transient outward K + channel current in neurons. We conclude that HIV-1 gp120 increases ROS, TNF-α and MCP-1 production in microglia, and induces cortical neuron apoptosis by affecting the delayed rectification and transient outward K + channel current. Curcumin reduces production of ROS and inflammatory mediators in HIV-1-gp120-stimulated microglia, and protects cortical neurons against HIV-1-mediated apoptosis, most likely through inhibition of HIV-1 gp120-induced elevation of the delayed rectification and transient outward K + current.

Up to now, the colloidal gold labeling immunochromatographic test strip is a mature and applicable technology. However, different from the conventional gold nanoparticle , quantum dot (QD) possesses larger specific surface area and better biocompatibility. So, as a novel nanomaterial , QD is capable of assembling more biomolecule which could enhance the sensitivity and accuracy of strips by rationality. Besides, strand displacement amplification was drawn into our test strips in this paper, this assumption made HIV-DNA recycling many times and converting it to plentiful QD-dsDNA (double-stranded deoxyribonucleic acid), where after these nano-structures would be captured by test zone. Meanwhile, the suggested scheme eliminated the hook effect owing to the target drop out of the incorporation on test zone, and any nucleotide sequence or substance which has aptamers can work as the target, such as carcinoembryonic antigen or mycotoxin . This assay realized the detection limit of as low as 0.76 pM (S/N = 3) and the detection range of 1 pM to 10 nM. In the end, we made use of this fluorescent lateral flow assay strips with great reproducibility for detecting HIV-DNA in human serum, that attested this method could be applied to practical application prospectively.

An ultrasensitive portable electrochemical immunosensor for human immunodeficiency virus p24 (HIV p24) antigen detection has been developed, whereby the detection sensitivity was 1000 times higher than that of the ELISA method. Firstly, a novel HRP enzyme–antibody copolymer (EV-p24 Ab2) was synthesized through an EnVision regent (EV, a dextrin amine skeleton anchoring more than 100 molecules of HRP and 15 molecules of anti IgG), then incubated in the secondary antibody of p24. Secondly, the copolymer was immobilized on the gold nanocolloids (AuNPs) to fabricate a novel signal tag (AuNPs/EV-p24 Ab2). Subsequently, a sandwich-type immunoreaction would take place between the capture probe (silicon dioxide-coated magnetic Fe3O4nanoparticles (MNPs) labeled with the primary p24 antibody (MNPs-p24 Ab1)), p24 (different concentrations) and the signal tag [AuNPs/EV-p24 Ab2)] to form the immunocomplex. Finally, the immunocomplex was absorbed on the surface of screen printed carbon electrode (SPCE) by a magnet and immersed in the o-hydroxyl phenol (HQ) and H2O2. The large amounts of HRP on the signal tag can catalyze the oxidation of HQ by H2O2, which can induce an amplified reductive current. Moreover, the capture probe could improve the accumulation ability of p24 and facilitate its separation from the substrate through the magnet. Under optimal conditions, the proposed immunoassay exhibited good sensitivity to p24 within a certain concentration range from 0.001 to 10.00 ng/mL, with a detection limit of 0.5 pg/mL (S/N = 3). The proposed method can be used for real-time and early detection of HIV-infected people.

Human immunodeficiency virus (HIV) is still a significant global health concern. Hence, developing a rapid and sensitive method for the detection of HIV is important in early detection. In this paper, we focus on the preparation of a sensitive electrochemical luminescence (ECL) biosensor that uses Ti 3 C 2 T x modified ZIF-8 as an ECL emitter to detect HIV-1 protein. This ECL biosensor used 0.05 M K 2 S 2 O 8 as the co-reactant and conductive carbon black combined with magnetic nanoparticles as the quenching agent. Under the optimized experimental conditions, the relationship was linear in the range from 1 fM to 1 nM (R 2 = 0.993) and the detection limit was 0.3 fM (S/N = 3). Finally, we used the constructed Ti 3 C 2 T x /ZIF-8 nanocomposites to detect HIV-1 in real serum samples and obtained great recoveries. The results demonstrated that this biosensor, which not only exhibited improved electrical conductivity but also demonstrated an excellent ECL response, could be a promising candidate for HIV-1 detection.

The authors describe a dual-signal electrochemical biosensor for highly sensitive determination of the HIV-1 related gene. This method is based on the application of cascaded toehold-mediated strand displacement reactions (TMSDRs) in combination with non-enzymatic target recycling amplification (TRA). A DNA machine with two TMSDRs was designed, and this resulted in reusable target and an output of two oligonucleotides, referred to as strand A (AS) labeled with the redox tag methylene blue (MB) and as untagged strand B (BS). A ferrocene (Fc)-modified signal probe (Fc-P 1 ) is immobilized on the gold electrode surface by hybridizing with a thiolated probe (P 2 ). The labeled AS causes the dissociation of Fc molecules and the gathering of MB molecules via strand displacement reaction. The target gene triggers TMSDRs and TRA. This leads to an increase in the distance changes between the redox tags and the gold electrode. The assay works in the 1 pM to 10 nM concentration range. On account of target recycling and dual recognition, the limit of detection is as low as 0.88 pM (at an S/N ratio of 3). The assay also has a remarkable selectivity which is ascribed to the use of both cascaded TMSDRs and dual recognition. In our perception, this assay represents a robust means of wide scope in that it may be applied to the detection of various kinds of nucleic acid even in complex samples.
Schematic of a dual-signal electrochemical biosensor based on cascaded toehold-mediated strand displacement and non-enzymatic target recycling amplification strategy for the HIV-1 related gene detection. Two electroactive molecules are used to produce electrochemical signals in this “signal-on/off” sensing system.

Human immunodeficiency virus (HIV) infection inflicts significant economic and social burdens on many countries worldwide. Given the substantial morbidity and mortality from HIV infection, there is an urgent need for accurate and early detection of the virus. In this study, immunofluorescence and visual techniques are described that detect the HIV-1 p24 antigen, which relied on selective recognition of Ag + /Ag nanoparticles (Ag NPs) and Cu 2+ /Cu + using cadmium telluride quantum dots (CdTe QDs). After the sandwich immunoreactions were accomplished, the alkaline phosphatase (ALP) hydrolyzed L-ascorbic acid 2-phosphate (AAP) to form ascorbic acid (AA) that further reduces Ag + and Cu 2+ to Ag NPs and Cu + , respectively. This method was highly sensitive and selective and could detect as low as 1 pg/mL of p24 antigen by naked eyes and had a good linearity in the concentration range 1–100 pg/mL. When using Ag + and Cu 2+ as media, the limit of detection (LOD) of the new method was 0.3 pg/mL and 0.2 pg/mL, respectively. Compared with clinical electrochemiluminescence immunoassay (ECLIA) results and clinical data, this method demonstrated good consistency for the quantification of HIV-1 p24 antigen in 34 clinical serum samples. In addition, this method could accurately distinguish HIV from other viruses and infections such as hepatitis B virus , systemic lupus erythematosus , hepatitis C virus , Epstein-Barr virus , cytomegalovirus , lipemia , and hemolysis . Therefore, our dual-mode analysis method may provide additional solutions to identify clinical HIV infection. Graphical abstract An immunofluorescence and visualization dual-mode strategy for the detection of p24 antigen was constructed based on immune recognition reaction and a phenomenon that cadmium telluride quantum dots (CdTe QDs) can selectively recognize Ag + /Ag nanoparticles (Ag NPs) and Cu 2+ /Cu + .

Background China's significant population affected by HIV poses a substantial threat to blood transfusion safety. Despite advancements in blood testing techniques, a residual risk of HIV transmission persists. Accurately assessing HIV epidemic and the residual risk is vital for monitoring blood supply safety and evaluating the effectiveness of new screening tests. Methods We conducted a retrospective analysis of HIV detection results among voluntary blood donors from 2003 to 2022. The study included data on HIV-confirmed positive donors, HIV prevalence, infection risk factors, and an incidence-window period mathematical model to estimate the residual risk of HIV. Results Between 2003 and 2022, HIV prevalence among blood donors in Guangzhou showed a peak-shaped trend, initially increasing before declining. The overall HIV prevalence was 18.9 infections per 100,000 donations. Male donors had a significantly higher prevalence compared with female donors. Donors aged 26–35 years had the highest prevalence. Ethnic minority donors had a higher prevalence compared with Han donors. Repeat donors had a lower prevalence compared with first-time donors. Donors from other provinces had a higher prevalence compared with local donors. During the period of 2003 to 2022, the residual risk of HIV in Guangzhou steadily decreased, reaching a notable 1 in 526,316 donations in the past two years. Conclusion The HIV epidemic among blood donors in Guangzhou remains severe, but the residual risk of HIV is decreasing. Novel detection methods have proven advantageous in reducing this residual risk. Implementing additional effective measures is imperative to ensure blood safety and curb the spread of HIV.

Human Immunodeficiency Virus (HIV) compromises the human immune system and remains one of the deadliest infectious diseases globally. Nucleic acid testing is a powerful tool for the early detection of HIV infection, capable of real-time HIV DNA detection without the limitation of a window period. In this study, a DNA nanostructure-based biosensor that leveraged interactions between functionalized gold nanoparticles (AuNPs) and three-dimensional (3D) DNA walkers were developed for ultrasensitive fluorescence detection of HIV DNA. The biosensor comprised blocked-walking particles (BPs) with locked DNAzyme and substrate particles (SPs) with quenched carboxyfluorescein (FAM) fluorophores. Upon encountering HIV DNA, the blocking strands hybridized more strongly with the HIV DNA, exposing the DNAzyme, which subsequently cleaved FAM on the SPs from the AuNPs and restoring fluorescence. As all substrate strands on SPs were cleaved, the BPs interacted with new SPs, significantly amplifying the fluorescent signal. The fluorescence intensity showed a linear correlation with HIV DNA concentration ranging from 0.02 to 6.00 nM, with a detection limit of 12.8 pM. The performance of biosensor was evaluated using spiked serum samples, demonstrating good precision and recovery rates. The DNA walker biosensor leveraged particle-to-particle interactions for rapid signal amplification, offering great potential for sensitive biosensors.

At present, AIDS remains a significant global health issue that requires ongoing attention. Although there is no practical cure for AIDS, hemoadsorption can effectively remove the virus from the blood and may serve as a promising treatment strategy. In this research, cellulose acetate/metal–organic framework composite beads (CA/MIL-125) with interconnected macropores were first fabricated using a one-step phase inversion method and then applied as a viral hemoadsorbent for the treatment of AIDS. The results showed that CA/MIL-125 exhibited excellent specific surface areas (221.17 m2 g–1) and an interconnected macropore structure, which allows HIV molecules to be transported freely within the pores of the beads. The CA/MIL-125 composite beads reached a 97.37% adsorption rate of HIV in plasma, which was 90 times higher than that of pure cellulose acetate (CA) microspheres, demonstrating that MIL-125 played a dominant role in HIV adsorption. Furthermore, the adsorption of CA/MIL-125 on HIV primarily obeys the pseudo-second-order kinetics and Freundlich model, with a maximum adsorption capacity (Qm) of 642.2 ng g–1. Tests for hemolysis, coagulation time, and cytotoxicity confirmed that CA/MIL-125 had good blood compatibility and biocompatibility. This study presents an effective strategy for HIV adsorption through hemoperfusion, suggesting a promising new treatment approach for AIDS.

Hydride generation atomic fluorescence spectrometry (HG-AFS) has been widely used for highly sensitive determination of more than 10 elements that are capable of hydride generation in geological, environmental and biological samples, but its bioanalytical potential has not been well-exploited yet. With CdS quantum dots (QDs) as both the signal labels for DNA sandwiching and analytes for HG-AFS (acid-released Cd2+ from QDs), here we proposed an ultrasensitive indirect method for HIV DNA analysis by HG-AFS. After a conventional sandwich-type hybridization reaction between capture DNA, target DNA and signal DNA labelled with CdS QD assembled silica microspheres, nitric acid was added to dissolve QDs and release Cd2+ from the QD-labels, which were then detected by HG-AFS. This efficient signal amplification strategy, coupled with the highly sensitive HG-AFS detection, gave rise to an impressive limit of detection of 0.8 aM and a wide dynamic concentration range (1 aM to 1 fM). This HG-AFS-based DNA biosensing platform holds great promise in future tumor marker detection and early cancerous diseases diagnosis.

Due to variations in virus structures and their high variability, treating viral infections remains challenging. Moreover, developing vaccines and medications for newly emerged viruses demands a significant time investment. Efficient methods are required to prevent dangerous infections caused by unknown pathogens. Hemoperfusion, with its low cost, can effectively remove toxins from the blood quickly, thus achieving therapeutic effects. The core of the hemoperfusion technology lies in the adsorbent. Therefore, the development of a viral adsorbent aiming to combat viruses through hemoperfusion was envisioned, following general principles to effectively save lives and buy time for the research and development of vaccines and medications. Towards this goal, a facile, green, and low-priced magnetic biochar/quaternary ammonium salt (Fe 3 O 4 /C-2) was constructed using the hydrothermal carbonization (HTC) method. HIV lentiviruses were used as a model pathogen to quantify the adsorption capacity and elucidate the adsorption mechanism. Benefiting from a macroporous structure and electrostatic interactions, fabricated Fe 3 O 4 /C-2 has a high adsorption capacity (965 ng g −1 ) and strong suppression of infectious activity (99.99%). Moreover, a low hemolysis ratio (<2%), high cell viability (>100%), high activated partial thromboplastin time (APTT) (67.29 s), and low protein adsorption (<5%) indicate outstanding biocompatibility and good anticoagulant activity. Therefore, Fe 3 O 4 /C-2 is expected to serve as a simple and efficient pathogen sorbent for viral diseases treatment.
