There are many instances of hollow-structure morphogenesis in the development of tissues. Thus, the fabrication of hollow structures in a simple, high-throughput, and homogeneous manner with proper natural biomaterial combination is valuable for developmental studies and tissue engineering, while it is a significant challenge in biofabrication field. We present a novel method for the fabrication of a hollow cell module using a coaxial co-flow capillary microfluidic device. Sacrificial gelatin laden with cells in the inner layer and GelMa in the outer layer are used via a coaxial co-flow capillary microfluidic device to produce homogenous micro-beads. The overall and core sizes of core-shell microbeads were well controlled. When using human vein vascular endothelial cells (HUVECs) to demonstrate how cells line the inner surface of core shell beads, as the core liquifies, a hollow cell ball with asymmetric features is fabricated. After release from the GelMa shell, individual cell balls are obtained and deformed cell balls can self-recover. This platform paves way for complex hollow tissue modelling in vitro, and further modulation of matrix stiffness, curvature, and biochemical composition to mimic in vivo microenvironments.

Human umbilical vein endothelial cells (HUVECs) have great potential in tissue engineering, regenerative medicine, and clinical applications. There is an ever-increasing demand to provide living HUVECs and HUVECs-hydrogel constructs to end users when needed in cell-based therapy and clinical applications. However, current methods to provide living cells and their constructs are mainly continuous culture and cryopreservation, which are high-cost, labor-intensive, time-consuming and inflexible. The research about hypothermic storage of HUVECs and their hydrogel constructs is still limited. Here, we studied the cell survival of HUVECs without encapsulation (W/O Encap) or with encapsulation (alginate, alginate with carboxymethyl chitosan [CMCH]) at 4°C and 25°C during 7 days, respectively. Also, we explored the optimal CMCH concentration for hypothermic storage, which is 0.5% (w/v) at 4°C and 25°C. Moreover, we evaluated the cell attachment after hypothermic storage. Our results enable the hypothermic storage of HUVECs and HUVEC-hydrogel constructs, and facilitate their application in tissue engineering and clinical medicine.

Hypospadias is a common congenital malformation of the male urogenital system. As a recently arisen reconstruction method for hypospadias, polymer tissue engineering urethral scaffold often results in necrosis and other complications due to the lack of bioactivity. To improve the bioactivity of polymer, in this work, we prepared biodegradable Mg/poly(lactic-co-glycolic acid) (PLGA) composite nanofiber scaffolds by electrospun with 3, 6, and 9 wt.% of micron-scale Mg particles. The mechanical properties of the PLGA scaffold gradually decrease with the addition of Mg particles but still at an acceptable level. On the other hand, these composite scaffolds show very good biocompatibility and promotional bioactivity, namely non-cytotoxicity, gradual proliferation enhancement, and good adhesion state of human adipose stem cells (HADSCs) on the scaffold surface. Besides, through quantitative real-time polymerase chain reaction (qRT-PCR) test, different genes expression levels of human umbilical vein endothelial cells (HUVECs) were also gradually up-regulated with the increase in Mg content. The present results indicate that Mg containing PLGA scaffolds might not only have the potential to promote tissue regeneration related to urethral repair, but also stimulate endothelial cells to achieve vascularization and anti-inflammatory functions. Consequently, considering both the mechanical properties and bioactivity for clinical demand, the Mg/PLGA composite scaffolds with suitable Mg contents are promising for the regeneration of defective urethral tissues.

Cryopreservation is essential to store living cells and tissues for future use while maintaining the proper levels of cell functions. The use of cryoprotective agents (CPAs) to inhibit intracellular ice formation during cryopreservation is vital for cell survival, but the addition and removal of CPAs and ice recrystallization during rewarming will cause fatal injury to cells. The conventional CPA loading and unloading methods generate osmotic shocks and cause mechanical injury to biological samples, and the conventional method of rewarming using a water bath also leads to ice recrystallization and devitrification. A new CPA-loaded microparticle-based method for loading and photothermal rewarming under near-infrared (NIR) laser irradiation was proposed to overcome these difficulties. We have successfully achieved the controlled release of CPAs (2 M EG, 2 M PG, and 0.5 M trehalose) with a graphene oxide (GO, 0.04% w/v) core from a 1.5% (w/v) sodium alginate shell to the human umbilical vein endothelial cells (HUVECs) within 60 s using NIR laser irradiation (808 nm Lasever at 5000 mW/cm2) and successfully recovered the CPA-loaded cells with 0.04% (w/v) GO in 8–10 s using the same NIR irradiation. The results show that this method achieved 25% higher viability of HUVECs compared to the conventional method. In short, this study proposes a new approach for achieving controlled CPA loading to cells with a photothermal-induced strategy for cell cryopreservation.

Background: Inflammatory bowel disease (IBD), characterized by chronic inflammation of the digestive tract, involves angiogenesis as a key pathogenic mechanism. Ginsenoside Rg3, derived from the traditional Chinese herb ginseng, is recognized for its anti-angiogenic properties but is limited by low oral bioavailability. This necessitates the development of an alternative delivery system to improve its therapeutic effectiveness. Methods: Pluronic F-127 (F127) and Pluronic F-68 (F68) were used to construct Rg3-loaded thermosensitive hydrogel Gel-Rg3. Meanwhile, a series of physicochemical properties were determined. Then the safety and pharmacological activity of Gel-Rg3 were evaluated in vitro and in vivo using human umbilical vein endothelial cells (HUVECs) and colitis mouse model, in order to initially validate the potential of Gel-Rg3 for the treatment of IBD.Results: We engineered a rectally administrable, thermosensitive Gel-Rg3 hydrogel using F127 and F68, which forms at body temperature, enhancing Rg3’s intestinal retention and slowly releasing the drug. In vitro, Gel-Rg3 demonstrated superior anti-angiogenic activity by inhibiting HUVEC proliferation, migration, and tube formation. It also proved safer and better suited for IBD’s delicate intestinal environment than unformulated Rg3. In vivo assessments confirmed increased intestinal adhesion and anti-angiogenic efficacy. Conclusions: The Gel-Rg3 hydrogel shows promise for IBD therapy by effectively inhibiting angiogenesis via rectal delivery, overcoming Rg3’s bioavailability limitations with improved safety and efficacy. This study provides new inspiration and data support for the design of treatment strategies for IBD.

Repairing damaged tissue caused by bacterial infection poses a significant challenge. Traditional antibacterial hydrogels typically incorporate various components such as metal antimicrobials, inorganic antimicrobials, organic antimicrobials, and more. However, drawbacks such as the emergence of multi-drug resistance to antibiotics, the low antibacterial efficacy of natural agents, and the potential cytotoxicity associated with metal antibacterial nanoparticles in hydrogels hindered their broader clinical application. In this study, we successfully developed imidazolium poly(ionic liquids) (PILs) polymer microspheres (APMs) through emulsion polymerization. These APMs exhibited notable antibacterial effectiveness and demonstrated minimal cell toxicity. Subsequently, we integrated the APMs into a gelatin methacryloyl (GelMA)—polyethylene glycol (PEG) hydrogel. This composite hydrogel not only showcased strong antibacterial and anti-inflammatory properties but also facilitated the migration of human skin fibroblasts (HSF) and human umbilical vein endothelial cells (HUVECs) and promoted osteogenic differentiation in vitro.

Nonclinical toxicity testing in rodent and nonrodent models often fails to predict human pharmacological responses, and the reliance on poorly characterized biomaterials hinders clinical translation in drug development. To address these limitations, we engineered a human serum albumin methacryloyl (HSAMA) cryogel system incorporating key liver extracellular matrix (ECM) components, collagen I (C) and fibronectin (F), to mimic the liver's architectural complexity. By co-culturing HepG2 and HUVECs within the HSAMA-CF cryogel, we created a three-dimensional, large-pore scaffold that facilitates critical hepatocyte-endothelial cell interactions essential for liver function and drug metabolism. Over extended culture periods, the HSAMA-CF cryogel significantly enhanced liver-specific functions, including elevated albumin production, increased urea secretion, and upregulated expression of hepatic functional genes. Notably, the co-culture model successfully detected multiple hepatotoxic phenotypes—such as elevated transaminase levels, hepatocyte damage, and steatosis—following exposure to known hepatotoxins like fialuridine and acetaminophen. This HSAMA-CF co-culture system may represent a robust in vitro platform for liver toxicity assessment. By closely mimicking human liver physiology and drug responses, it might offer a promising alternative to traditional animal models, potentially improving drug safety evaluations, reducing reliance on preclinical animal testing, and minimizing risks associated with clinical drug development.

Angiogenesis plays a pivotal role in tissue development and regeneration, but few safe and efficient pro-angiogenic factors are available for long-term clinical therapy. Although stem cell-derived exosomes exhibit excellent biocompatibility and considerable pro-angiogenic capacity, their low yield extremely limits their clinical potential. Exosome-mimetics (EMs) employ similar features to exosomes and can be produced in a higher yield. In this study, the HY-EMs were produced to enhance in vitro and in vivo pro-angiogenic ability from hypoxia-preconditioned bone marrow mesenchymal stem cells (BMSCs) using a serial extrusion method. The yield of EMs was over 10 times greater than that of exosomes. Furthermore, compared with the EMs group, the tube number of human umbilical vein endothelial cells (HUVECs) in vitro doubled in the HY-EMs treated group, and the number of blood vessels increased by 186% in subcutaneously implanted scaffold with HY-EMs. More importantly, the HY-EMs effectively promoted vascularized osteogenesis in a critical-sized cranial defect model, with the new bone volume and vessel number in regenerated bone increased to 120% and 175%, respectively. Consequently, our current work provides a promising strategy to customize and mass-produce functional EMs for vascularized osteogenesis.

Diabetics face multiple long-term health complications and diabetic foot ulcers affect approximately 15–20% of the diabetic population. The high disability rate of these patients results in substantial mental burden and economic pressure to the individuals as well as the societies. Here, we report a therapeutic integrin-binding pro-survival peptide-engineered silk fibroin nanosheet that can regulate angiogenesis and promote diabetic ulcer healing. The nanosheet with thickness less than 100 nm is highly transparent and flexible, and it can firmly adhere to the biological tissue surfaces in underwater environment. The presence of integrin-binding pro-survival peptide in the nanosheet significantly upregulates the expression of angiogenesis genes and proteins of human umbilical vein endothelial cells (HUVECs). In the diabetic mouse model, the peptide-engineered nanosheet reveals significant acceleration of wound closure, epithelialization and granulation tissue formation. These findings suggest that this functional nanosheet could be a promising therapeutic candidate for complicated diabetic ulcer healing.

The purpose of this study was to construct a biomimetic urethral repair substitute. The nano-Laponite/polylactic acid–glycolic acid copolymer (PLGA) fiber scaffolds were produced to replicate the natural human urethra tissue microenvironment. PLGA (molar ratio 50:50) and Laponite were used in this study as raw materials. The nano-Laponite/PLGA scaffolds were fabricated via electrospinning technology. After preparing the material, the microstructural and mechanical properties of the nano-Laponite/PLGA scaffold were tested via scanning electron microscopy and electronic universal testing. The effects of different amounts of Laponite on the degradation of the nano-Laponite/PLGA scaffold were studied. Human umbilical vein endothelial cells (HUVECs) were co-cultured with PLGA and nano-Laponite/PLGA scaffolds for 24, 48, or 72 h. Scanning electron microscopy results illustrated that the microstructure of the scaffold fabricated by electrospinning was similar to that of the natural extracellular matrix. When the electrospinning liquid contained 10% Laponite, the nano-Laponite/PLGA stress–strain curve illustrated that the scaffold has strong elastic deformation ability. HUVECs exhibited good growth on the nano-Laponite/PLGA scaffold. When the scaffold contained 1% Laponite, the cell proliferation rate in the CCK-8 test was significantly better than that for the other three materials, displaying good cell culture characteristics. The 1% nano-Laponite/PLGA composite scaffold can be used as a suitable urethral repair material, but its performance requires further development and research.

Owing to its superior mechanical and physicochemical characteristics, polyetheretherketone (PEEK) is considered a potent orthopedic material. Yet, poor bactericidal, osseointegrative and angiogenic properties of bioinert PEEK collectively hamper its clinical adoption. Herein, we present a strategy for developing multifunctional hierarchical coatings bearing a synthetic fusion peptide and polydopamine/silver nanoparticles (pDA/AgNPs) bio-heterojunctions (bio-HJs), in an attempt to empower PEEK with triple-modal abilities, which prevents bacterial infection, enhances ossification and promotes re-vascularization for physiological osseointegrative fixation. By harnessing the adhesion of bioinspired pDA, AgNPs are facilely immobilized onto peripheral surface and incorporated into interior cavity of micro/nano-porous sulfonated PEEK (SP) to form hierarchical pDA/AgNPs heterostructures. The bio-HJs coatings exhibit robust and repeatable bacteria-eradicating ability, assisted by near-infrared illumination, due to the synergistic effects of the metal ion/photothermal therapy. The fusion peptide-engineered PEEK exhibits osteogenesis activity in terms of promoting osteoblast proliferation, spreading, boosting the synthesis and secretion of osteogenesis-related genes. Besides, the fusion peptide-engineered PEEK also has the ability to promote HUVECs growth, reproduction, and tube formation, facilitating the synthesis and secretion of angiogenesis-associated genes. Moreover, the results of the in vivo experiments further corroborate the in vivo osteointegration and angiogenic abilities of materials. Expectantly, the multifunctional coatings with the presented triple-therapeutic effects represent a prospective clinical solution to address the current clinical challenges associated with the use of PEEK implants.

The development of three-dimensional (3D) in vitro model to recapitulate the in vivo tumor tissue is essential for studying tumor biology, discovering anti-cancer drugs, and evaluating anti-cancer drug efficacy . However, most of the previous models lack the involvement of vascular barrier. Here, we proposed an in vitro 3D cocultured tumor-vascular barrier model by the combination of alginate hydrogels beads and Transwell system. PC-3 cells and NIH/3T3 cells were encapsulated in alginate hydrogel beads, which were cultured in the bottom chamber of Transwell, while human umbilical vein endothelial cells (HUVECs) were cultured on the porous membrane in the upper chamber to form vascular barrier. The effect of the concentration of alginate sodium on the morphology, diameter and swelling ratio of the beads was studied. The alginate sodium content and cell seeding density were further optimized according to cell proliferation ability. The formation of endothelial barrier was verified by immunostaining with tight junction protein VE-cadherin and transendothelial electrical resistance (TEER) monitoring. Finally, the drug response of 3D cocultured tumor-vascular barrier model to curcumin was evaluated. Compared with two-dimensional (2D) coculture model and 3D coculture spheroid model, 3D tumor-vascular barrier model showed the highest activity of cancer cells and the strongest drug resistance. The developed 3D cocultured tumor-vascular barrier model possesses great potential to be applied for in vitro evaluation of anti-tumor drugs.
