Cervical cancer remains one of the leading cause of cancer death in women. As the development of nanomedicine, multifunctional nanoplatforms which integrate imaging and therapy for effective diagnostics and therapeutics of cancer have attracted great attention in recent years. In this work, we have designed and developed a kind of multimodal imaging and treatment strategy which combined near-infrared (NIR) fluorescence imaging with magnetic resonance imaging (MRI) as well as photothermal therapy (PTT) via a multifunctional nanoplatform. The poly lactic-co-glycolic acid (PLGA) was utilized to encapsulate magnetic Fe 3 O 4 nanoparticles (NPs) and NIR fluorescent dye clinical indocyanine green (ICG), by which integrated the advantages of Fe 3 O 4 NPs and clinical ICG under 808-nm laser excitation, to achieve highly efficient dual-modal imaging-guided cervical tumor ablation. This work provides a promising approach for multimodal imaging diagnosis and PTT of cervical cancer.

Photothermal therapy (PTT) is a rapidly developing approach for cancer therapy, which has been widely recognized to exert high efficacy as compared to chemotherapy. However, the limited tumour homing property of currently available drug delivery systems (DDSs) is the bottleneck for the efficient delivery of photothermal agents. Here in this study, we surface modified silica nanoparticles (SLN) with the cell membrane (CM) derived from 143B cells to construct a platform (CM/SLN) capable of targeting the homogenous 143B cells. In addition, indocyanine green (ICG) as a photothermal agent was encapsulated into CM/SLN to finally construct a DDS suitable for tumour-targeted PTT of osteosarcoma. Our results revealed that CM/SLN/ICG was mono-dispersed core-shell nanoparticles with advanced stability in a physiological environment. Moreover, due to the modification of CM, CM/SLN/ICG could specifically target the homogenous 143B cells both in vitro and in vivo, which demonstrated superior anticancer efficacy when compared with either SLN/ICG or free ICG. Hence, CM/SLN/ICG could be a promising DDS for tumour targeted PTT of osteosarcoma.

Photothermal therapy (PTT) is a safe and efficient anti-tumor treatment. A photothermal agent (PTA) with good biocompatibility and strong photothermal properties is of great importance for PTT. In this study, near-infrared (NIR) excitable clinical indocyanine green (ICG) was utilized as a PTA and further encapsulated by another PTA polydopamine (PDA) to form highly stable and efficient ICG@PDA nanoparticles (NPs). Then the ICG@PDA NPs were modified with methoxy polyethylene glycol amine (mPEG2000-NH2) to form biocompatible ICG@PDA@PEG NPs. ICG@PDA@PEG NPs showed good water solubility and a spherical shape with an average size of 140 nm. Furthermore, the photothermal properties of ICG@PDA@PEG NPs were studied and excellent photothermal performance with a photothermal conversion efficiency of 43.7% under 808 nm laser irradiation was achieved. Then, the PTT properties of ICG@PDA@PEG NPs were confirmed on HeLa cells with an efficiency of 86.1%. Meanwhile, the in vivo biocompatibility and toxicity of ICG@PDA@PEG NPs were evaluated. No apparent in vivo toxicity was observed in 24 hours and 7 days. Next, in vivo PTT analysis was conducted for cervical tumor-bearing nude mice under 808 nm laser excitation. It showed a good anti-tumor effect in vivo. Thus, ICG@PDA@PEG NPs exhibited great potential for safe and efficient photothermal therapy in anti-tumor therapy.

Sustainable polysaccharide-based carriers offer a promising route to overcome the instability and aggregation issues of indocyanine green (ICG), a clinically approved phototherapy dye. In this study, a scalable approach is presented to construct quaternized microcrystalline cellulose (QMCC) carriers with tunable surface charge for stabilized and high-capacity ICG loading. By modulating the degree of quaternary ammonium substitution, QMCC effectively modulated ICG aggregation behavior, promoting the formation of broadened and red-shifted near-infrared absorption bands with enhanced photostability. Among them, QMCC2-ICG exhibited a balanced electrostatic and steric environment through the π-π stacking interaction, favoring the formation of hybrid aggregates with broad-spectrum absorption profiles and superior optical stability. This enabled high ICG loading (388.3 mg/g), elevated photothermal conversion efficiency (55.0 %), and robust singlet oxygen generation. 4 T1 in vitro experiments demonstrated improved cell phototoxicity, ROS-mediated cytotoxicity, and tumor cell migration inhibition. In vivo , QMCC2-ICG also showed excellent hemocompatibility and enabled strong photoacoustic imaging and tumor ablation in 4 T1-bearing mice. Histological and molecular assays confirmed apoptosis induction, proliferation inhibition, and biosafety. This work presents a sustainable polysaccharide platform that enables aggregation-state control and excited-state tuning for improved multimodal phototherapy, highlighting the role of surface charge and morphology in driving green therapeutic material design.

Nanoparticles (NPs) have proven to be effective drug carriers in diagnosis and therapy of cancer. But, they faced a contradictory issue that NPs with large size appear weak tumor penetration, meanwhile small size resulted in poor tumor retention. Herein, we fabricated doxorubicin conjugated carbon dots (CDs-DOX) and indocyanine green (ICG)-loaded liposomes (ICG-LPs) named CDs-ICG-LPs using a modified reverse phase evaporation process, and with high incorporation in the aqueous core. The CDs-ICG-LPs exhibited good monodispersity, excellent fluorescence/size stability, and consistent spectra characteristics compared with free ICG or DOX. Moreover, the CDs-ICG-LPs showed higher temperature response, faster DOX release under laser irradiation. In the meantime, the fluorescence of DOX and ICG in CDs-ICG-LPs was also visualized for the process of subcellular location in vitro. In comparison with chemo or photothermal treatment alone, the combined treatment of CDs-ICG-LPs with laser irradiation synergistically induced the apoptosis and death of DOX-sensitive HepG2 cells. In vivo antitumor activities demonstrated CDs-ICG-LPs could reach higher antitumor activity compared with CDs-DOX and ICG-LPs for H22 tumor cells, and suppressed H22 tumor growth in vivo. Notably, no systemic toxicity occurrence was observed after repeated dose of CDs-ICG-LPs with laser irradiation. Hence, the well-defined CDs-ICG-LPs exhibited great potential in targeting cancer imaging and chemo-photothermal therapy.

Graphical GNRs@mSiO 2 core–shell nanoparticles have been synthesized as host to load ICG and DOX molecules by the layer-by-layer assembly method through electrostatic interactions. A sensitive Se–Se linker was synthesized and grafted to ensure controlled release. Under NIR irradiation, the enhanced cytotoxicity to cancer cells is derived from the synergistic effect of chemotherapy, PTT, and PDT. A novel GNRs@mSiO 2 core–shell nanocomposite has been synthesized to integrate sensitive chemotherapy with photodynamic therapy (PDT) and photothermal therapy (PTT) for enhanced antitumor treatment by using gold nanorods (GNRs) as the core and mesoporous silica (mSiO 2 ) as the shell. After modifying with -NH 2 , Indocyanine Green (ICG, negative charge) and doxorubicin hydrochloride (DOX, positive charge) molecules could be stored in the mesopores by the layer-by-layer assembly method through electrostatic interactions. To inhibit leakage from the mesopores, a sensitive Se–Se linker was synthesized and adopted as a nanovalve coated on the outside of the nanoparticles (GNRs@mSiO 2 -ICG-DOX@Se-Se). Under NIR (808 nm) irradiation, heat elevation (derived from GNR/ICG) and generation of reactive oxygen species (ROS, derived from ICG) can also be detected in vitro. Owing to the redox sensitivity of Se–Se, GNRs@mSiO 2 -ICG-DOX@Se-Se showed reduction (GSH)- as well as oxidation (ROS)-triggered DOX release. To further ensure specific targeting, folic acid (FA) was grafted to the outside of the nanoparticles, because of the high expression of FA receptors on tumor cells. Detailed cell experiments performed with the nanoparticles and HepG2 as typical cancer cells showed enhanced cytotoxicity towards the cancer cells owing to the synergistic effect of chemotherapy, PTT, and PDT. On account of the different GSH concentrations in cancer and normal cells and the switch-on/off application of NIR irradiation, the GNRs@mSiO 2 -ICG-DOX@Se-Se-FA nanoparticles show potential for applications in cancer therapy.

Dye-sensitized upconversion nanoparticles (UCNPs) have gained significant interest in biological applications. However, previous attempts to utilize these dye-sensitized UCNPs in the aqueous environment suffer from dramatic decline in the upconversion luminescence (UCL) intensity and problems with nanoparticles aggregation and water quenching. By employing indocyanine green (ICG) as a hydrophobic amplifier and energy harvester for UCNPs, we use a general, facile, and kinetically controlled mixing technology—flash nanoprecipitation (FNP)—to prepare aqueous stable ICG-sensitized UCNPs encapsulated by a biocompatible block copolymer poly (ethylene glycol)- b -poly (lactic- co -glycolic acid (PEG- b -PLGA) to efficiently activate ROS generation for simultaneous cancer cell imaging and treatment. As compared to NPs obtained from traditional drip and stir method (TM), the NPs under high turbulent conditions (high Re ) exhibit smaller size, narrower size distribution, and much higher stability, for the UCNPs hydrophobicity enhanced by ICG which was bound on the surface of UCNPs due to the coordination effect between sulfonates of ICG molecules and naked lanthanide metal ions of UCNPs. The remained UCL intensity was significantly boosted as well as ROS generation through FNP in the presence of ICG, but not for TM samples indicating superior protection of upconversion NPs from water quenching and the energy transfer between ICG and UCNP was well retained. The efficiency of ROS generation of NPs via FNP evidently increases with the stronger red-light emission of UCNPs, resulting in high cytotoxicity for SMMC-7721 cells in vitro and good suppression of tumor growth in vivo . The success of NPs by FNP in biological application demonstrates a bright prospect of FNP for synthesis of efficient antitumor nanoparticles.

The infections associated with dental and orthopedic implants always result from the biofilm of bacterial colonization, leading to implant failure, prolonged hospitalization, and increased healthcare costs. Iron oxide (Fe 3 O 4 ) nanoparticles and indocyanine green (ICG) are both reported to have good antibacterial properties. However, the anti-biofilm efficiency of either component alone is still unsatisfactory. In this paper, octahedral Fe 3 O 4 nanoparticles (O-Fe 3 O 4 ) were synthesized and integrated with ICG to achieve the best biofilm removal effect, and the optimized ratio between the Fe 3 O 4 and ICG was also investigated. The results showed that ICG inhibited the magnetic properties of Fe 3 O 4 slightly (from 92 emu/g to 76 emu/g); however, it endowed Fe 3 O 4 with significantly improved photothermal, photodynamic, and peroxidase-like activities, achieving excellent antibacterial property (eradicating more than 99% of S. aureus under the combination of H 2 O 2 treatment and laser irradiation). Combined with the good cytocompatibility (cell viability > 80% at a concentration of 250 μg/mL), the synthesized O-Fe 3 O 4 @ICG nanoparticles (especially O-Fe 3 O 4 @ICG-10) have strong application potential for synergistic therapy against biofilm infection associated with dental and orthopedic implants. This study presents a promising nano-antibacterial system for combating resistant biofilm infections, offering insights into the rational design of synergistic nanotherapeutics for clinical applications.

Developing effective therapy to inhibit postoperative recurrence and metastasis of colorectal cancer (CRC) is challenging and significant to reduce mortality and morbidity. Here, a granular hydrogel, assembled from gelatin microgels by dialdehyde starch and interpenetrated with in situ polymerized poly(sulfobetaine methacrylate- co -N-isopropylacrylamide) (P(SBMA- co -NIPAM)), is prepared to load and lock Food and Drug Administration (FDA)-approved indocyanine green (ICG) with definite photothermal function and biosafety for photothermal therapy (PTT) combining with checkpoint inhibitor. The presence of P(SBMA- co -NIPAM) endows granular hydrogel with high retention to water-soluble ICG, preventing easy diffusion and rapid scavenging of ICG. The ICG-locking granular hydrogel can be spread and adhered onto the surgery site at wet state in vivo, exerting a persistent and stable PTT effect. Combined with αPD-L1 treatment, ICG-locking granular hydrogel-mediated PTT can eradicate postsurgery residual and metastatic tumors, and prevent long-term tumor recurrence. Further mechanistic studies indicate that combination treatment effectively promotes dendritic cells maturation in lymph nodes, enhances the number and infiltration of CD8 + T and CD4 + T cells in tumor tissue, and improves memory T cell number in spleen, thus activating the antitumor immune response. Overall, ICG-locking gel-mediated PTT is expected to exhibit broad clinical applications in postoperative treatment of cancers, like CRC.

Traditional chemotherapy for breast cancer faces limitations, including poor drug bioavailability, multidrug resistance, and severe systemic side effects. Therefore, a carrier-free nanodrug composed of Mastoparan M (Mast-M, derived from Wasp toxin), FDA-approved near infrared fluorescence dye (indocyanine green, ICG), and biosafe ion Fe 3+ was developed to achieve photothermal (PTT) and synergistic chemodynamic therapy (CDT)-assisted oxidation therapy. Once accumulated within tumor sites by enhanced permeability and retention (EPR) effects, the Mast-M/Fe 3+ /ICG rapidly dissociates in response to elevated glutathione (GSH), releasing Fe 2+ , ICG, and Mast-M. Mast-M efficiently triggers reactive oxygen species (ROS) generation to induce mitochondrial membrane potential disruption. Fe 2+ -mediated CDT further amplifies oxidative stress, accompanied by GSH consumption and lipid peroxide (LPO) accumulation. Under laser irradiation, ICG generates hyperthermia to enhance oxidative stress. In vitro and in vivo studies demonstrated enhanced antitumor efficacy with reduced systemic toxicity. Together, these findings highlight the potential of carrier-free nanodrugs for oxidative stress-based breast cancer therapy.

Recurrence and metastasis are the primary causes of mortality in hepatocellular carcinoma (HCC), primarily driven by an immunosuppressive tumor microenvironment. This study developed a multifunctional flexible patch (MnO 2 -P-ICG NFs) based on electrospun SiO 2 gel fibers whose surface is modified with a layer of “nest-like” MnO 2 . The patch enables the co-delivery of indocyanine green (ICG) and pachymaran directly to the tumor site via a laparoscopic system. Under near-infrared light irradiation, ICG serves as a photosensitizer for photodynamic therapy (PDT), effectively ablating tumors and inducing immunogenic cell death (ICD), thereby activating antitumor immunity. Meanwhile, pachymaran, with its immunomodulatory effects, preferential activates natural killer (NK) cells to exert antitumor effects. Additionally, PDT further enhances this immune response by activating the cGAS-STING pathway. In mouse models, implantation of the patch significantly inhibited primary liver tumor progression and ascites formation. In a malignant liver tumor model, MnO 2 -P-ICG NFs prevented local recurrence in 80% of treated mice. Notably, both the depletion of NK cells and the blockade of the cGAS-STING pathway compromised the therapeutic efficacy of MnO 2 -P-ICG NFs. Clinically, tumors with higher NK cell infiltration were associated with improved patient outcomes. In conclusion, this versatile patch provides a promising therapeutic strategy for patients with advanced HCC.

Background Phototherapy is a potential new candidate for glioblastoma (GBM) treatment. However inadequate phototherapy due to stability of the photosensitizer and low target specificity induces the proliferation of neovascular endothelial cells for angiogenesis and causes poor prognosis. Methods In this study, we constructed c(RGDfk)-modified glycolipid-like micelles (cRGD-CSOSA) encapsulating indocyanine green (ICG) for dual-targeting neovascular endothelial cells and tumor cells, and cRGD-CSOSA/ICG mediated dual effect of PDT/PTT with NIR irradiation. Results In vitro, cRGD-CSOSA/ICG inhibited cell proliferation and blocked angiogenesis with NIR irradiation. In vivo, cRGD-CSOSA/ICG exhibited increased accumulation in neovascular endothelial cells and tumor cells. Compared with that of CSOSA, the accumulation of cRGD-CSOSA in tumor tissue was further improved after dual-targeted phototherapy pretreatment. With NIR irradiation, the tumor-inhibition rate of cRGD-CSOSA/ICG was 80.00%, significantly higher than that of ICG (9.08%) and CSOSA/ICG (42.42%). Histological evaluation showed that the tumor vessels were reduced and that the apoptosis of tumor cells increased in the cRGD-CSOSA/ICG group with NIR irradiation. Conclusion The cRGD-CSOSA/ICG nanoparticle-mediated dual-targeting phototherapy could enhance drug delivery to neovascular endothelial cells and tumor cells for anti-angiogenesis and improve the phototherapy effect of glioblastoma, providing a new strategy for glioblastoma treatment.
