Effect of sturgeon protein in promoting the adhesion of Lactobacillus plantarum and Lactobacillus rhamnosus

Erdong Yuan,Miao Zhou,Zhengyang Liang,William Kwame Amakye,Chuanli Hou,Jiaoyan Ren

Dietary protein could serve as primary nitrogen source for intestinal flora to enhance the intestinal environment by influencing probiotic adhesion. Our work aims to demonstrate a framework for probiotic activity guided isolation of potential effective fractions from the sturgeon protein. We confirmed the isolated fractions of sturgeon protein as having probiotic adhesion activity against Lactobacillus plantarum 45 (LP45) and Lactobacillus rhamnosus GG (LGG). Three protein fractions (0%, 40%, and 60% NaCl elution fractions) were obtained through anion exchange resin column. The molecular weights of the three fractions were identified by polyacrylamide gel electrophoresis in the ranges of 20.1–66.4 kDa (Fr1), 14.3–44.3 kDa (Fr2) and 14.3–97.2 kDa (Fr3), respectively. A detailed study was performed using an in vitro adhesion model to obtain the bioactive protein. In this model, probiotics were incubated on isolated intestinal segments of mice, and the adhesion effect of probiotics was evaluated through polysaccharide analysis, biofilm quantification, viable count measurement and electron microscopy . Notably, Fr2 was identified as the most effective fractions, significantly increasing the intestinal adhesion rates of LP45 and LGG by 144.8% and 53.7%, respectively. Ultimately, the primary structure of the protein 14.3 kDa in Fr2 was identified by mass spectrometry . Partial peptide amino acid sequences of the protein were confirmed to be W-W-Y-C-F (m/z = 804), K-N-H-D-I/L-Q-A-D-D-I/L-T-C (m/z = 1510) and Q-M-V-H-K-I/L-E-D-E-E-I/L-C-T-S-A-D-K-D (m/z = 2300). In conclusion, we found evidence that purified fractions of sturgeon protein facilitates the adhesion of intestinal probiotics and can be used as an active protein beneficial for intestinal health.

Lactobacillus salivarius and Lactobacillus agilis feeding regulates intestinal stem cells activity by modulating crypt niche in hens

Hong Yi,Zhou Zhou,Yu Lingzi,Jiang Keyang,Xia Jiamiao,Mi Yuling,Zhang Caiqiao,Li Jian

Previously, we found that Lactobacillus salivarius , Lactobacillus agilis , and Lactobacillus aviarius were associated with excellent egg nutrition in native chicken. Next, the optimal Lactobacillus combination is worth studying. Here, a total of 120 HyLine hens (30 hens per group contained 3 replicate cells, 10 hens/cell) in the laying peak period were randomly divided into (1) control, (2) L. salivarius  +  L. agilis , (3) L. salivarius  +  L. aviarius , and (4) L. agilis  +  L. aviarius groups, fed with diet only or with corresponding Lactobacilli (10 8 colony-forming units/hen/day) for 30 days. As a result, L. salivarius  +  L. agilis feeding could (1) improve egg-laying rate, egg weight, and albumen’s amino acid levels; (2) increase Lactobacillus abundance, decrease Escherichia coli abundance, upregulate the tryptophan metabolism pathway-related molecules, and downregulate the primary bile acid biosynthesis pathway-related molecules in intestinal contents; and (3) upregulate oxidative-phosphorylation pathway-related genes, reactive oxygen species levels, and mRNA abundance of Wnt3a , Dll1 , Lgr5 , CCDN1 , and CDK2 in the crypt. Collectively, L. salivarius  +  L. agilis feeding in hens could improve intestinal microflora and metabolism profile, promote crypt’s local energy metabolism and reactive oxygen species levels, and thus enhance Paneth cells and intestinal stem cells activity. Key points • Lactobacilli co-feeding could improve laying performance and egg nutrition. • Lactobacilli co-feeding could improve intestinal microflora and metabolism profile. • Lactobacilli co-feeding could enhance Paneth cells and intestinal stem cells activity.

Naturally Occurring Exopolysaccharide Nanoparticles: Formation Process and Their Application in Glutathione Detection

Chengcheng Li,Xiaodong Zhang,Yuxin Guo,Farzad Seidi,Xiaotong Shi,Huining Xiao

Naturally occurring nanoscale exopolysaccharide (EPS) has attracted much attention in recent years. In this research, we obtained a new kind of naturally occurring spherical EPS nanoparticles (EPS-R503) from Lactobacillus plantarum R503. The secretion, self-assembly process, morphological structure, and surface characteristics of the as-prepared nanoparticles were comprehensively revealed with transmission electron microscopy (TEM) and atomic force microscope (AFM) for the first time. It was found that the EPS-R503 nanoparticles consist of negatively charged heteropolysaccharide composed of mannose, glucose, galactose, and glucuronide with several functional groups including −OH, −COOH, and −NH2. When different solvents were used to treat the EPS-R503 nanoparticles, the morphological structure and surface properties could be changed or manipulated. The forming mechanism of EPS-R503 was elucidated based on the aggregation processes from a fundamental point of view. Furthermore, EPS-R503 can serve as reducing and stabilizing agents for the biosynthesis of manganese dioxide nanosheets (MnO2 NSs), leading to EPS-MnO2 nanocomposite. The as-prepared nanocomposites can absorb fluorescein (FL) to form EPS-MnO2-FL, which can be used to detect glutathione (GSH) with a low limit of detection (0.16 μM) and a wide detection range from 0.05 to 4 mM. The excellent biocompatibility of EPS-MnO2-FL endows the feasibility of in vivo detection of GSH as well. Overall, the findings from this work not only benefit the exploitation of naturally occurring EPS nanomaterials but also provide a novel strategy for the green synthesis of metal-containing nanosheets for GSH detection.

Lactobacillus gasseri LG 2361 from infant feces synergizes with Lactobacillus acidophilus ATCC 43121 to lower lipids in high-fat fed mice

Xi Lu,Xirui Cheng,Guohua Li

Hyperlipidemia impose a substantial burden on human health. Recent studies have demonstrated that probiotics can regulate lipid metabolism by modulating gut microbiota composition or producing bioactive metabolites, with human-derived strains potentially exhibiting superior host colonization capacity. This study isolated Lactobacillus gasseri LG 2361 from 84 healthy infant fecal samples. Safety assessments confirmed non-hemolytic activity, antibiotic susceptibility (except natural resistance), and gastrointestinal adaption (81 % survival at pH 3.0; 60 % survival at 0.1 % bile). The strain demonstrated lipid-lowering capacity in vitro , degrading 43.0 % cholesterol and 34.6 % triglycerides. In high-fat diet mice, LG 2361 reduced hepatic lipid accumulation (11.6 % total cholesterol, p < 0.05), attenuated abdominal adiposity (32.9 % subcutaneous fat reduction, p < 0.01), and may enhance antioxidant status (22.5 % glutathione peroxidase increase, p < 0.01). Synergy with L . acidophilus ATCC 43121 amplified efficacy, achieving 61.6 % abdominal fat reduction (p < 0.01) while suppressing TNF-α (0.46-fold) and IL-1β (0.29-fold, p < 0.001). Mechanistically, LG 2361 downregulated PPARγ (0.59-fold, p < 0.01) and modulated FAT/CD36 pathways, indicating dual regulation of lipogenesis and lipid transport. These findings suggest that LG 2361 may be a probiotic candidate for hyperlipidemia management.

Polysaccharides from Lyophyllum decastes reduce obesity by altering gut microbiota and increasing energy expenditure

Tao Wang,Junjie Han,Huanqin Dai,Jingzu Sun,Jinwei Ren,Wenzhao Wang,Shanshan Qiao,Chang Liu,Li Sun,Shuangjiang Liu,Dianpeng Li,Shenglong Wei,Hongwei Liu

Polysaccharides are known to confer protection against obesity via modulation of gut microbiota . To expand our knowledge of mushroom-derived prebiotics , we investigated the structural characteristics and anti-obesity effects of Lyophyllum decastes polysaccharides. Two heteroglycans were purified and characterized. The isolated polysaccharides effectively reduced obesity and the related disorders in the diet-induced obese (DIO) mice. An altered gut microbiota with enrichments of Bacteroides intestinalis and Lactobacillus johnsonii and an increase of secondary bile acids were detected in the polysaccharide-treated mice. Supplementation of B. intestinalis and L. johnsonii prevented the obesity and hyperlipidemia in DIO mice, demonstrating their causal linkage to the efficacy of polysaccharides. An enhancement of energy expenditure in the brown adipose tissues due to up-regulation of the secondary bile acids-activated TGR5 pathway was deduced to be one of the mechanisms underlying the effect of polysaccharides. These results confirmed Lyophyllum decastes -derived polysaccharides as new prebiotics for preventing and treating obesity.

Lactobacillus plantarum enhances cognition through acetate-modulated microglia function

Tingyi Zhou,Yi Yue,Juncai Leng,Yiming Jiang,Han Liu,Shiqi Zhang,Zhengchao Yang,Wei Zhao,Li Li

Lactobacillus plantarum enhances cognition in both normal and antibiotic-treated mice. • Microbial modulation elevates fecal acetate and reshapes brain immune homeostasis • Acetate supplementation alone improves cognition and restores microglial homeostasis. • Acetate enhances microglial homeostasis by reducing mitochondrial oxidative stress. • Acetate activates ACSS2-dependent acetyl-CoA production to reduce microglial inflammation. Gut microbiota profoundly influences cognition through immune and metabolic pathways, but how specific strains contribute to these processes remain unclear. Here, we show that supplementation with Lactobacillus plantarum C067 enhanced spatial and fear-associated memory, while restoring hippocampal cytokine expression and microglial morphology in both healthy and antibiotic-treated mice. L. plantarum also reshaped microbial composition and elevated short-chain fatty acids, particularly acetate. Acetate supplementation replicated the cognitive benefits and restored microglial homeostasis. Mechanistically, acetate exerted anti-inflammatory effects via acetyl-CoA synthetase 2 (ACSS2)–dependent acetyl-CoA production in microglia, reducing mitochondrial oxidative stress and cytokine release. Together, these findings identify an ACSS2-mediated microbe–metabolite–microglia axis that underlies the cognitive benefits of L. plantarum , providing a mechanistic framework for probiotic- and metabolite-based interventions against cognitive decline. Characterisation of five alcohol dehydrogenases from Lactobacillus reuteri DSM20016

Zhenghong Hu,Pu Jia,Yajun Bai,Tai-ping Fan,Xiaohui Zheng,Yujie Cai

Alcohol dehydrogenases convert aldehydes causing volatile aromas in cheese into corresponding alcohols. In this study, we cloned and recombinantly produced five novel ADHs from Lactobacillus reuteri DSM20016 in Escherichia coli BL21 (DE3). The resulting proteins have molecular weights between 35 and 39 kDa. The optimum pH for enzyme activity of ADH-1, ADH-2 and ADH-3 was weakly acidic (pH 5, 6 and 5, respectively), compared with pH 7 for ADH-4 and ADH-5. The optimum temperature range for ADH-1, ADH-3, ADH-4 and ADH-5 was 40 °C ―55 °C, compared with 35 °C for ADH-2. These enzymes reduced a broad range of substrates; the highest catalytic efficiency and substrate affinity for ADH-1 was observed with heptaldehyde (K cat /K m  = 39.4 s −1 ·mM −1 , K m  = 0.459 mM), while 3-methylbutanal was the best substrate for ADH-5 (K cat /K m  = 24.0 s −1 ·mM −1 , K m  = 1.35 × 10 −2  mM). ADH-2, ADH-3 and ADH-4 exhibited the highest substrate affinity for valeraldehyde, propionaldehyde, and hexanal, respectively (K m  = 1.19 × 10 −2  mM, 2.81 × 10 −2  mM, 1.35 × 10 −2  mM). The enzymatic properties indicated that these enzymes effectively converted fragrant compounds during cheese ripening.

Purification and characterization of bacteriocin produced by Lactobacillus rhamnosus zrx01

Ruixiang Zhao,Yanqing Lu,Junjian Ran,Gang Li,Shuang Lei,Yang Zhu,Baocheng Xu

The bacteriocin-zrx01 produced by Lactobacillus rhamnosus zrx01 was purified, its basic biochemical characteristics and bacterial antibacterial sensitivity were studied. The bacteriocin-zrx01 was separated and purified using ethyl acetate, AKTA purifier™ 10 protein purifier with HiTrap™ Capto™ Q anion exchange chromatography and Superdex™ 75 10/300 GL gel chromatography. The purity of bacteriocin-zrx01 was analyzed using high performance liquid chromatography . The sensitivity to temperature, pH and enzymes of the purified bacteriocin-zrx01 was measured as the diameter of the antimicrobial ring. The results showed that the purified bacteriocin-zrx01 bacterial antibacterial activity was good, and the specific vitality was raised from the original fermentation broth at 625 to 5330 AU/mg. The bacteriocin-zrx01 had a higher stability at pH 3–9, and the antibacterial activity was the strongest at pH 4. When pH was 9, it still retained 86% of the antibacterial activity of the control group. Bacteriocin-zrx01 was cold-stable at −70 and −20 °C for 30 min. It still retained 57% antimicrobial activity compared with the control after being autoclaved at 121 °C for 30 min. Some enzymes, such as protease K, neutral protease, trypsin, thrombin, alkaline protease and papain , affected its antibacterial activity, but it was sensitive to pepsin , and the diameter of the antimicrobial ring decreased by 50% after 2 h of pepsin treatment. These results showed that the bacteriocin-zrx01 produced by Lactobacillus rhamnosus zrx01 might have a potential as a natural food preservative , although in vivo studies are needed.

Antimicrobial Substances and Mechanisms of Lactobacillus rhamnosus against Gardnerella vaginalis

Huang Yu-Ping,Shi Jie-Yan,Luo Si-Chen,Xu Shao-Yan,Zhang Jia-Dong,Molnár István,Yang Qiong-Qiong,Zhang Bo-Bo

Bacterial vaginosis (BV) is a common vaginal disease associated with abnormal changes in the vaginal microbiome. Our previous study found that Lactobacillus rhamnosus has a good therapeutic effect on bacterial vaginosis by inhibiting the most prominent bacterium associated with BV, Gardnerella vaginalis . In this study, we show that acetic acid and lactic acid are the main substances in the cell-free supernatant (CFS) of L. rhamnosus that inhibit the growth of G. vaginalis. Further study on the mechanism showed that acetic acid and lactic acid alter the morphology of the G. vaginalis cells, eventually causing the cells to shrink or burst, resulting in exudation of their intracellular contents. In addition, these two organic acids also dissipate the membrane potential of bacterial cells, affecting their synthesis of ATP. A reduced activity of the Na + /K + -ATPase leads to abnormal ATP metabolism, and ultimately inhibits the growth and reproduction of G. vaginalis . Our study provides valuable information for the widespread application of L. rhamnosus in the treatment of bacterial vaginosis.

Lactobacillus gasseri postbiotics ameliorate age-related decline in endometrial receptivity

Meng Li,Shangqi Li,Danlei Zheng,Baoying Liao,Hongjuan Niu,Yue Wang,Xiyao Liu,Heng Pan,Yang Yu,Ping Zhou,Rong Li

Endometrial senescence contributes to a decline in fertility in women of advanced reproductive age, with emerging evidence linking it to remodeling of the uterine microbiome, particularly a reduced Lactobacillus abundance. Here, using a cross-sectional human cohort with clinical follow-up, we identify an age-related shift in the endometrial microbiome, characterized by the loss of beneficial Lactobacillus -enriched states and increased Lactobacillus iners . This shift is associated with impaired endometrial receptivity and less favorable embryo transfer outcomes. Functional screening of reproductive tract isolates highlights Lactobacillus gasseri as a candidate strain with strong antioxidant, anti-inflammatory, and adhesion-related activities. In cells and mouse models, L. gasseri and its exopolysaccharides (EPSs) attenuate endometrial senescence and improve implantation in aged mice. Mechanistically, these effects are mediated, at least partly, through the modulation of Hippo-YAP signaling. These findings suggest that restoring beneficial Lactobacillus and applying EPSs may represent complementary strategies to improve endometrial function and fertility outcomes in women of advanced reproductive age.

Lactobacillus reuteri E9 Regulates Sleep Disorders Through Its Metabolite GABA

Yongcheng Jiang,Lina Guo,Houde He,Haonan Chen,Tao Chen,Yan Liu,Wang Zhao

Background:Insomnia, the most prevalent sleep disorder, is clinically defined as difficulty initiating or maintaining sleep. Although many medications are effective for insomnia treatment, they carry risks of drug dependence and abuse. The microbiota-gut-brain axis (MGBA) facilitates bidirectional signaling between the gastrointestinal tract and the central nervous system via gut microbes. Probiotics that provide mental and behavioral benefits through MGBA (psychobiotics) offer broad therapeutic potential. Methods:A non-toxic, drug-resistant strain of Lactobacillus reuteri E9 was isolated and characterized. Its effects were evaluated in a pentylenetetrazol (PTZ)-induced zebrafish model of sleep disorder. Neurotransmitter levels (glycine, serine, taurine, γ-aminobutyric acid (GABA)) and gene expression of GABA/melatonin receptors were analyzed.Results:E9 significantly upregulated inhibitory neurotransmitters, including GABA, taurine, glycine, and serine (p < 0.05). In PTZ-induced zebrafish, E9 exerted sedative effects by reducing seizures and hyperactivity. Concurrently, E9 upregulated the expression of GABA receptor genes and melatonin receptor (Mtnr1aa) genes in zebrafish neural tissue. Conclusions:Lactobacillus reuteri E9 demonstrates potential as a psychobiotic for sleep disorder management by modulating key inhibitory neurotransmitters and sleep-related receptor expression via the MGBA pathway, offering a non-pharmacological alternative to conventional treatments.

Lactobacillus helveticus alleviates cognitive impairment via microbiota-regulated phosphatidylcholine metabolism

Yuting Zhou,Yang Jiang,Aonan Xia,Bo Yang,Xiaoming Liu,Peng Zhou,Jianxin Zhao,Wei Chen

The microbiota-gut-brain axis emerges as a potential therapeutic strategy. In this study, the effect of Lactobacillus helveticus CCFM1096 on alleviation of cognitive impairment and influence on the gut microbiota and metabolites in scopolamine-induced mice were investigated. Our findings indicated that CCFM1096 significantly improved cognitive behavior and brain histopathology in mice, and altered the structure of the gut microbiota and modulated relevant metabolites in cognition impaired mice. In particular, phosphatidylcholine and its metabolites levels were significantly regulated and the expression of genes related to the acetylcholine/Brain-Derived Neurotrophic Factor signaling pathway was significantly up-regulated in hippocampus. Furthermore, phosphatidylcholine and glycerophosphocholine effectively alleviated scopolamine-induced PC12 neuronal cell impairment, increased acetylcholine level and up-regulated the expression of neurotrophic-related genes. In conclusion, CCFM1096, a potential probiotic interfering with cognitive function in mice, attenuated scopolamine-induced cognitive dysfunction in mice via the gut-brain axis and exerted a neuroprotective effect by up-regulating phosphatidylcholine, glycerophosphocholine, and acetylcholine/Brain-Derived Neurotrophic Factor levels.
