Synergy of Free Radicals in Microcystis aeruginosa Inactivation within an Electrolysis/Persulfate/Chlorine System

Ze Zhang,Wankui Shao,Qiaoling Xu,Shuying Shen,Huaman Sun,Jiehao Zhan,Beini Gong,Lihua Cui,Zhujian Huang

In this study, an electrochemical system combined with peroxymonosulfate (PMS) and chlorine was developed to enhance the removal efficiency of Microcystis aeruginosa. The almost complete inactivation of algal cells can be achieved in 30 min at a chlorine concentration of 30.0 μM, PMS concentration of 0.75 mM, and current density of 15 mA/cm2. Quenching experiments indicate the involvement of •OH, SO4•–, Cl•, •O2–, and 1O2 in the inactivation of M. aeruginosa, and SO4•– plays a crucial role. Besides the inherent contribution of SO4•– in fortifying the electrolysis/PMS/chlorine system, the ability of the system to control algal proliferation is also bolstered by the SO4•– indirect activation of chlorine species. The alterations in superoxide dismutase, CAT, and malondialdehyde activities substantiate the induction of apoptosis in algal cells. The production of free radicals precipitates a cascade of oxidative impairment within algal cells, which is characterized by marked transformations in cell morphology and structural integrity. Further, microcystin-LR in actual water body also could be effectively removed in the electrolysis/PMS/chlorine system. The findings of this study convincingly establish the electrolysis/PMS/chlorine system as an efficient and environmentally friendly electrochemical application in alga wastewater treatment.

Sustained and enhanced inhibitory effects of allelochemicals on Microcystis Aeruginosa during its recruitment stage

An Lao,Hanqi Wu,Xuhui Huang,Kaili Huang,Xingzhang Luo,Zheng Zheng

Microcystis aeruginosa is the main toxic strain in cyanobacterial blooms, and the recruitment stage in its temperature-dependent seasonal succession is considered as the key to its subsequent growth. In this study, a protocol with specific temperature settings was developed as the simulated recruitment stage in order to investigate and confirm the superior inhibitory effects of allelochemicals on M. aeruginosa at that stage of recruitment. One of the most common allelochemicals, gallic acid (GA) (10 mg/L, 20 mg/L) was employed to treat M. aeruginosa under initially low temperature condition (15 °C), then intermediate (20 °C) and last normal (26 °C), which corresponds to the critical temperatures for cyanobacterial recruitment and growth. Growth, metabolism, photosynthetic activity, extracellular polysaccharides (EPS) and microcystins (MCs) release were analyzed and discussed in this study, and a more sustained and better inhibitory effect over a 20-day period was achieved. Notably, GA (10 mg/L) markedly delayed the recruitment of M. aeruginosa from low temperature, with an inhibition efficiency of 85.71 %, and suppressing Fv/Fm and photosynthetic pigments production. It is also observed that M. aeruginosa at recruitment stage exhibited higher sensitivity and poorer resistance to allelochemical treatment, with variable responses suggesting that optimal dosages may alter. The antioxidant enzyme activities remained high under prolonged stress, and the secretion of EPS was stimulated, indicating that cyanobacteria were more inclined to form colonies. While the laboratory-based inhibitory mechanism appeared to increase the release of microcystins in individual cells, the actual concentration of microcystins in natural aquatic environments requires further investigation.

Effects of Sb(V) on Growth and Chlorophyll Fluorescence of Microcystis aeruginosa (FACHB-905)

Wang Shuzhi,Pan Xiangliang

In this study, effects of antimony Sb(V) on growth, pigments content, oxygen evolution, and photosystem II (PSII) activity of Microcystis aeruginosa were investigated. JIP-test, Q − A reoxidation kinetic test and S-state test were used in this study to study the energy distribution and electron transport in PSII. Treatment with Sb(V) at various concentrations ranging from 5 to 100 mg/l had long-term effects on growth, pigments content, and oxygen evolution of M . aeruginosa . Low concentration of Sb(V) had no significant inhibition of the biomass production and PSII activity but inhibited the pigment synthesis. Growth, pigments content, oxygen evolution, and PSII activity were seriously inhibited when treated by high concentration of Sb(V) (100 mg/l). The target sites of Sb(V) toxic effect on the PSII of M . aeruginosa were mainly on the donor side and the apparatus in the light-dependent reaction. The quantum yield for photochemistry, density of reaction centers and photosynthesis performance index decreased, whereas the dissipated energy increased. PSII activity of M . aeruginosa was promoted when exposure to 50 mg/l Sb(V) by increasing the density of active reaction centers and electron transport after Q − A .

The effects of exogenous amino acids on production of microcystin variants in Microcystis aeruginosa

Yaxin Guo,Xiating Zhao,Zongcheng Yao,Zongyao Qian,Yuting Wang,Qiming Xian

Dissolved free amino acids are a significant component of dissolved organic nitrogen (DON) in natural waterbodies. The effects of four amino acids (glutamic acid, phenylalanine, leucine, and arginine) on the growth and microcystins (MCs) production of  Microcystis aeruginosa  were studied in batch culture. The profiles of five MCs variants and the expression levels of target genes involved in MCs biosynthesis and nitrogen metabolism were measured. When amino acids were used as the sole nitrogen source instead of nitrate at different levels (0.5, 2.0 and 8.0 mg/L based on N) in BG-11 medium, algal cell growth and intracellular MCs quotas were inhibited slightly by the treatments with glutamic acid and arginine. The treatments with phenylalanine and leucine, on the other hand, had a strong inhibitory effect on algal cell growth and MCs production. Moreover, the concentrations of Chlorophyll a , phycocyanin and allophycocyanin in cells cultured in glutamic acid, leucine and phenylalanine were lower than those in the control group with nitrate as nitrogen source. The existence of leucine or phenylalanine can lead to a significant increase in the relative abundance of MCs variants structured with the corresponding amino acids. The expression of microcystin-producing gene mcyD was downregulated while the gene pipX associated with nitrogen metabolism was upregulated during the cultivation of M. aeruginosa with amino acids as sole nitrogen source. M. aeruginosa undergoes significant alterations due to exogenous amino acids and exhibits advanced strategies for MCs production.

Single and combined effects of microplastics and lead on the freshwater algae Microcystis aeruginosa

Shan Wang,Qi Li,Suzhen Huang,Wei Zhao,Zheng Zheng

Recently, the pollution of microplastics (MPs) in the global freshwater environment has become increasingly problematic, but there are few studies on the freshwater environment risks of MPs. The present study, therefore, has investigated the single and combined effects of MPs and lead (Pb) on the freshwater algal Microcystis aeruginosa. Results showed that Pb-only (>0.05 mg·L −1 ) promoted the growth of algal cells, while MPs-only (1 mg L −1 ) resulted in growth inhibition. However, compared with the corresponding concentration of Pb-only groups, the growth of algal cells was promoted in MPs + Pb treatments. MPs-only and Pb-only (0.5 mg L −1 ) both reduced the content of photosynthetic pigments and affected algal photosynthesis. The MPs-only treatment and MPs + Pb 2+ (no pretreatment, 0.5 mg L −1 Pb 2+ ) treatments showed significant cell aggregation. At the same time, MPs-only caused a significant increase in bound extracellular polysaccharides (bEPS), while 0.5 mg L −1 Pb reduced bEPS. Furthermore, under high Pb stress (0.5 mg L −1 ), the effects of combined MPs and Pb on chlorophyll content, antioxidant enzyme activity (peroxidase (POD), catalase (CAT)), and damage to algal cells were less compared to individual effects, and the combination of MPs and Pb had a synergistic effect on promoting aggregations of M. aeruginosa . These results demonstrate that single and combined effects of MPs and Pb can induce differential responses in the freshwater algal M. aeruginosa , which can have a significant impact on aquatic ecosystems.

Toxic effects of terpinolene on Microcystis aeruginosa: Physiological, metabolism, gene transcription, and growth effects

Pengcheng Zhao,Yingmu Wang,Wei Huang,Lei He,Ziyuan Lin,Jian Zhou,Qiang He

Bioherbicide terpinolene is widely employed in the agricultural field because of its unique phytotoxic properties. However, little is known about the toxicity of bioherbicide on harmful algal blooms (HABs) and its mechanisms. Therefore, in this study, the inhibiting effect of bioherbicide terpinolene on the growth and physiological change of Microcystis aeruginosa was determined. Results showed that the cell density and photosynthetic activity of microalgae were significantly inhibited by terpinolene. Activities of nitrate reductase (NR) and glutamine synthetase (GS) were decreased separately by 25.38% and 42.75% after 4 d of exposure to 1.47 mM terpinolene, suggesting the inhibiting effect of terpinolene on algal nitrogen metabolism. However, the transcript abundance of genes related to membrane protein cytochrome c oxidase subunit II (COX II) and ATP-binding cassette transporters (ABC transporter) were enhanced separately by 3.15 and 1.69-fold compared with control, suggesting the resistance response of cells to terpinolene stress. Additionally, terpinolene not only increased the content of endogenous phytohormones including indole-3-acetic acid (IAA), zeatin, and brassinolide, but also inhibited the expression of genes related to calcium-binding protein (CaBPs), one kind of calcium (Ca 2+ ) sensors, suggesting its regulation on algal signal molecules. These findings helped to understand the ecotoxicity of terpinolene and guide the rational use of bioherbicide in agriculture.

Contributions of Zn Ions to ZnO Nanoparticle Toxicity on Microcystis aeruginosa During Chronic Exposure

Du Jingjing,Guo Ruilin,Li Ke,Ma Bingbing,Chen Yan,Lv Yanna

In this work, we assessed the toxic effects of ZnO nanoparticles (NPs; 1, 10, and 50 mg L −1 ) and the corresponding dissoluble Zn ions (0.71, 8.66, and 35.59 mg L −1 ) on Microcystis aeruginosa . After chronic exposure (28 days), significantly higher growth inhibition was observed under ZnO NPs at 1 mg L −1 (47%) than under Zn ions at 0.71 mg L −1 (−15%). The opposite effect pattern was observed for ZnO NPs at 10 (71% vs. 80%) and 50 mg L −1 (73% vs. 95%) compared to Zn ions at the corresponding concentrations. After 7 days of exposure, ZnO NPs at 10 and 50 mg L −1 led to an increase of 83 and 53% in malondialdehyde content, as well as an increase of 106 and 61% in superoxide dismutase activity, respectively. However, Zn ions at the corresponding concentrations showed negligible impacts on the two parameters. The different results indicate that the insoluble NPs during the initial exposure mostly account for lipid peroxidation, which further lead to microalgal antioxidant response. During the subsequent exposure, the contributors of ZnO NP toxicity shift with the concentration and exposure time of ZnO NPs. In conclusion, the study presents new insights into the different contributions of insoluble NPs and dissoluble metallic ions to metallic NP toxicity.

Integrated transcriptomic and metabolomic analysis of Microcystis aeruginosa exposed to artemisinin sustained-release microspheres

Cunhao Du,Wenlu Sang,Chu Xu,Zhiyun Jiang,Jiajia Wang,Yuanyi Fang,Chengjie Zhu,Jakpa Wizi,Muhammad Asif Akram,Lixiao Ni,Shiyin Li

Artemisinin sustained-release microspheres (ASMs) have been shown to inhibit Microcystis aeruginosa ( M. aeruginosa ) blooms. Previous studies have focused on inhibitory mechanism of ASMs on the physiological level of M. aeruginosa , but the algal inhibitory mechanism of ASMs has not been comprehensively and profoundly revealed. The study proposed to reveal the toxicity mechanism of ASMs on M. aeruginosa based on transcriptomics and metabolomics. After exposure to 0.2 g·L −1 ASMs for 7 days, M. aeruginosa biomass was significantly inhibited, with an inhibition rate (IR) of 47 % on day 7. Transcriptomic and metabolomic results showed that: (1) 478 differentially expressed genes (DEGs) and 251 differential metabolites (DMs) were obtained; (2) ASMs inhibited photosynthesis by blocking photosynthetic pigment synthesis, destroying photoreaction centers and photosynthetic carbon reactions; (3) ASMs reduced L -glutamic acid content and blocked glutathione (GSH) synthesis, leading to an imbalance in the antioxidant system; (4) ASM disrupted nitrogen metabolism and the hindered synthesis of various amino acids; (5) ASMs inhibited glyoxylate cycle and TCA cycle. This study provides an important prerequisite for the practical application of ASMs and a new perspective for the management of harmful algal blooms (HABs).

Unraveling the inhibitory mechanism of composite allelochemicals on Microcystis aeruginosa: Integrated transcriptomic and metabolomic insights

Jie Li,Liu Shao,Anglu Shen,Peimin He

Harmful cyanobacterial blooms (HCBs), driven by eutrophication, pose critical threats to aquatic ecosystems and public health. Current studies focused on inhibitory effect of single-component allelochemical on HCBs, yet cyanobacteria in natural environments are simultaneously exposed to multiple allelochemicals. Thus, it is more meaningful to study composite allelochemicals formulation for HCBs control. Our previous study found that the composite allelochemicals agent of 4-tert-butylcatechol and l -lysine (CAA-TL) can synergistically inhibit the growth of Microcystis aeruginosa . In this study, we investigated the inhibitory mechanism of CAA-TL against M. aeruginosa , integrating physiological, transcriptomic, and metabolomic approaches. CAA-TL reduced the cell density of M. aeruginosa by more than 90 % within four days, and significantly decreased the Chl- a content and chlorophyll fluorescence parameters. The integrated transcriptomic and metabolomic results showed that: (1) 471 upregulated and 312 downregulated differential expressed genes (DEGs) and 433 upregulated and 215 downregulated differential metabolites (DMs) were identified; (2) CAA-TL markedly impacted the expression of photosynthesis related genes (e.g., PsbA , PsbL , PsaB and PsaK ) indicating the inhibition of photosynthetic activity; (3) CAA-TL impacted DNA and protein synthesis through downregulation of nucleotide metabolism; (4) CAA-TL disrupted energy supply via the dysregulation of energy metabolism pathways. These findings reveal the inhibitory mechanism of CAA-TL via an integrated omics approach and provide theoretical support for their eco-friendly application in HCBs control.

Effects of Prometryn Exposure Scenarios on Microcystis aeruginosa Growth and Water Qualities in Incubator Experiments

Zhenjiang Yang,Suiliang Huang,Xiaowen Huang,Xiaofu Liang,Waseem Akram,Zobia Khatoon

Although multiple herbicide exposures are more prospective to occur in water, many previous studies were carried out as single herbicide exposure. To investigate the toxic effect of prometryn on cyanobacteria and water qualities, single and double prometryn exposures (at different growth phases) onMicrocystis aeruginosagrowth and concentrations of nutrients were compared after a 44-day experiment. Results indicated that under single exposure, maximum inhibition rates were 4.7–12.0% higher than those under double exposures. Correspondingly, the maximumMicrocystis aeruginosadensities and growth rates under single exposure were 10.3–21.1% and 19.5–37.7% lower than those under double exposures (p< 0.05), respectively. These findings revealed that repeated prometryn exposures resulted in a reduction in biological effects, because the time of application and the concentration injected during the first application were both significant factors in the biological effects of prometryn. Prometryn exposure scenarios did not have a significant effect on nutrient or nutrient consumption concentrations (p> 0.05). In general, the pattern of nutrient limitation showed a shift from phosphorus to nitrogen limitation. The quantified relationships betweenMicrocystis aeruginosagrowth rates and consumed nutrients were studied. Based on the above findings, we believe that a high-dose and single prometryn exposure is a more effective exposure pattern for limiting cyanobacteria growth.

Effects of PET and Gallic Acid on Growth, Photosynthesis, and Oxidative Stress of Microcystis aeruginosa

Wang Tiantian,Liu Haicheng

Utilizing the allelopathic effect of plants to inhibit algae growth has gained significant attention from researchers in recent years. However, there are large amounts of microplastics in the aquatic environment, which may influence the inhibitory effects of allelochemicals on algae. Consequently, this research aimed to examine the individual and combined toxicity of gallic acid (GA) and polyethylene glycol terephthalate (PET) microplastics on Microcystis aeruginosa. At a concentration of 50 mg/L, PET significantly enhanced algal cell growth and chlorophyll a synthesis, while inducing oxidative stress without damaging the cell membrane. On the other hand, low concentrations of GA promote algal cell growth. However, high concentrations of GA not only inhibit growth and photosynthesis but also result in cellular abnormalities and severe oxidative damage, as well as membrane disruption. Moreover, it was observed that the exposure of PET diminished the toxicity of GA to M. aeruginosa . The combined exposure to GA and PET resulted in a slight increase in chlorophyll a content, as well as phycocyanin (PC), alloxyphycocyanin (APC), and phycoerythrin (PE) content, along with maximum photochemical efficiency (Fv/Fm), compared to GA exposure alone. In the combined system of GA and PET, the contents of superoxide dismutase (SOD) and malondialdehyde (MDA) were lower than those in the single GA system. Additionally, both systems showed a degree of improvement compared to the control group. Scanning electron microscopy (SEM) observed that the presence of PET resulted in reduced damage to algal cells compared to exposure to GA alone. This research establishes a foundation for understanding the synergistic impacts of allelochemicals and microplastics on M. aeruginosa , thereby providing a reference for managing cyanobacteria in water blooms.

Effects of fish feed addition scenarios with prometryn on Microcystis aeruginosa growth and water qualities

Zhenjiang Yang,Suiliang Huang,Wenwen Kong,Liang Chen,Priakhina Ekaterina,Zobia Khatoon,Muhammad Nabil Ashraf,Waseem Akram

The number of undesirable environmental impacts of fish feed has been reported widely. Although repeated fish feed exposures are more prospective to occur in water, previous studies were mostly conducted as a single exposure of fish feed. In order to fill these gaps, a 40 days incubator experiment was conducted to explore the effects of fish feed addition scenarios during the lag phase with prometryn on both Microcystis aeruginosa growth and concentrations of nutrients. The maximum algae densities in groups of single exposure were 6.0–26.2% and 8.8–74.4% higher than those in groups of double and triple exposures, respectively ( P  < 0.05). At the beginning of the experiment, concentrations of nutrients in groups with different feed exposure scenarios were significantly different. The pattern of nutrient limitation showed a transformation from phosphorus limitation to nitrogen limitation generally. Furthermore, the average inhibition rates of algae by prometryn in the case of a single fish feed exposure were 4.6–9.4% lower than those under double exposures, and 22.0–26.8% lower than those under triple exposures ( P  < 0.05). In addition, algae growth rates have been developed as a function of concentrations of consumed nutrients ( R 2 = 0.410–0.932). Based on the above results, we concluded that in terms of limiting algae growth multiple low-dosage additions of fish feed were considered as a better addition pattern. By optimizing feed addition scenarios, there is considerable potential to increase the environmental sustainability of aquaculture.
