Naringenin prolongs lifespan and delays aging mediated by IIS and MAPK in Caenorhabditis elegans

Yue Ge,Huibin Chen,Jilite Wang,Guishan Liu,Steve W. Cui,Ji Kang,Yumei Jiang,Hao Wang

Naringenin (NN) is one of the most abundant flavonoids in citrus and grapefruits and has been shown to have antioxidant properties in vitro. The purpose of the study is to examine the antioxidant and anti-aging activities of NN in C. elegans, and to further explore the molecular mechanism. The results showed that NN enhanced the lifespan under normal and oxidative stress induced by H2O2. After treatment with NN, locomotion capability was improved and aging pigment accumulation was suppressed. NN also delayed the paralysis and reversed the defective chemotaxis behavior induced by Aβ protein. Meanwhile, the treatment with NN enhanced the activities of antioxidant enzymes and reduced the accumulation of reactive oxygen species (ROS) and malondialdehyde (MDA) content. The possible targets and pathways interacting with NN were predicted by network pharmacology. Real-time PCR analysis indicated that NN upregulated the expression levels of daf-16, sek-1 and skn-1, downregulated the expression levels of daf-2, age-1 and akt-1, and further activated sod-3, ctl-1, ctl-2, gst-4 and mtl-1. Moreover, the selected mutant strains were used and molecular docking was conducted to further suggest that IIS and MAPK pathways could be involved in the NN-mediated longevity-promoting effect.

pH-shifted quinoa protein isolate-based nanocarriers for naringenin: Structure characterization and stability evaluation

Ying-Ying Chen,Qiang-Ming Li,Li-Hua Pan,Jian-Ping Luo,Xue-Ying Li,Xue-Qiang Zha

Naringenin (Nar) is an important natural flavonoid compound with multiple health functions, but stability issues and poor solubility greatly limit its bioavailability. To enhance the water-solubility, chemical stability and bioavailability of Nar, quinoa protein isolate (QPI)-Nar and pH-shifted QPI (PQPI)-Nar nanoparticles were constructed in this study. The results of ultraviolet–visible and fluorescence spectra showed there existed the interactions between QPI or PQPI and Nar, with stronger interaction between PQPI and Nar. Thermodynamic analysis indicated the molecular interactions between QPI or PQPI and Nar were mainly driven by hydrogen bonds. Circular dichroism (CD) and surface hydrophobicity results revealed that the binding of Nar changed the secondary structures and reduced surface hydrophobicity of QPI and PQPI. The measured DLS, LC and EE results suggested that PQPI-Nar nanoparticles formed a more stable system with higher encapsulation capacity. Additionally, PQPI-Nar nanoparticles exhibited higher antioxidant activity , superior slow-release property and biocompatibility, as well as better thermal and ultraviolet stability than QPI-Nar nanoparticles. These results indicated that PQPI-Nar nanoparticles could effectively improve the bioavailability of Nar. This study may provide a theoretical basis for the promising application of PQPI-Nar nanoparticle delivery systems in the food industry.

Obtention, interaction, and characterization of the soy protein isolate-glycyrrhizin nanocomplex for encapsulating naringenin

Qingchen Cui,Xixi Song,Mengshuang Li,Yanjun Wei,Juan Dong,Xianggen Wu

Naringenin (NAR) has several potential benefits as a bioactive nutrient, but its low water solubility and poor bioavailability have limited its application. Here, a soy protein isolate-naringenin complex (NAR@SPI) was prepared for NAR delivery. Furthermore, to improve the encapsulation capacity of SPI for hydrophobic bioactive nutrients and their complex physiochemical properties, dipotassium glycyrrhizinate (DG) was used to modify SPI with a simple stirring process to obtain the SPI-DG complex, and then this complex was used to load the NAR. The final obtained nanocomplex was named NAR@SPI-DG. This novel NAR@SPI-DG has shown higher encapsulation efficiency (93.20 ± 0.27% versus 62.09 ± 3.20%) and loading capacity (8.53 ± 0.02% versus 5.89 ± 0.30%) for NAR as compared to the NAR@SPI. Spectral evaluations and molecular simulations revealed that the SPI's and DG's interaction in SPI-DG involved the combination of a spontaneous exothermic reaction, and the mechanisms of interaction were dominated by a hydrogen bond and van der Waals forces. NAR@SPI-DG has demonstrated significant improvements in solubility, in vitro antioxidant activity, in vitro anti-diabetes activity, and in vitro bioaccessibility as compared with bare NAR and NAR@SPI. Consequently, the use of DG is promising in the modification of SPI protein to construct a nano-delivery system for hydrophobic bioactive nutrients, such as NAR, in functional foods.

Natural product flavonoid of naringenin based thin-film composite polyester membranes preparation for antibiotic separation

Chao Wang,Peijuan Chu,Tengfei He,Cheng Yi,Yue Yao,Lijia Liu,Chunhong Zhang

The application of membrane separation technology in wastewater treatment and pharmaceutical purification has attracted considerable attention. The development of a membrane with effective separation performance and antibacterial properties is of considerable interest for the separation and purification of antibiotics. Here, a thin-film composite (TFC) polyester membrane with effective antibiotic removal and antibacterial properties was fabricated via a facile one-step interfacial polymerization (IP) strategy. Naringenin (Nar), a natural product of flavonoid material derived from the pericarp, was utilized as the aqueous monomer. The suitable reactivity of Nar afforded the resulting membrane with a highly negative charged surface. The rejection performances for the antibiotics cephalothin, cefuroxime sodium, ceftriaxone sodium, and cefoperazone sodium were 96.3 %, 97.7 %, 99.2 %, and 99.8 %, respectively. The pure water permeance was 7.5 L m −2 h −1 bar −1 at a transmembrane pressure of 5 bar. The antibacterial efficacy was greater than 99.0 % against Escherichia coli and Staphylococcus aureus , attributable to the flavonoid structure of Nar. The present work proposed a feasible strategy to prepare TFC membranes with high nanofiltration performance and intrinsic antibacterial efficacy.

Potato starch/naringenin complexes for high-stability Pickering emulsions: Structure, properties, and emulsion stabilization mechanism

Hong Feng,Ting Li,Lian Zhou,Lei Chen,Qingyun Lyu,Gang Liu,Xuedong Wang,Xi Chen

In this study, potato starch (PS)/naringenin (NAR) complex was prepared, and its properties and emulsification behavior were evaluated. The experimental results demonstrated that NAR successfully formed a complex with PS molecules through hydrogen bonds and other non-covalent interactions. The emulsifying capacity (ROV) of PS/NAR complex with 16 % composite ratio was 0.9999, which was higher than PS (ROV = 0.3329) ( p  < 0.05). Based on particle property analysis and molecular dynamics simulation, the mechanism of improving the emulsification performance might be the action of the benzene ring of NAR and intermolecular hydrogen bonding. In addition, the stability of the Pickering emulsions with PS/NAR complexes as emulgators was significantly improved. The emulsifying and rheological behavior of starch-based Pickering emulsions could be adjusted by changing the proportion of the complexes. Results demonstrated that the PS/NAR complexes might be a prospective stabilizer of Pickering emulsions based on starch material and might expand the use of PS in edible products.

Molecularly imprinted polymer combined with multiple-component covalent organic frameworks for extraction of naringenin enantiomers

Juanqiong Ma,Na Yang,Xingyuan Zhang,Manyi Xiao,Cheng Cao,Zhiyong Xie,Pei Li,Qiongfeng Liao,Yanlong Chen

Naringenin is a plant flavonoid found in citrus fruits and is known for its anti-inflammatory and antioxidant activities. However, extracting high-purity naringenin from citrus fruits and separating its enantiomers remains challenging. This study synthesized a molecularly imprinted multiple-component covalent organic framework (MI COF) with abundant imprinted recognition sites for the first time. This MI COF combines the advantages of molecular imprinting and covalent organic frameworks into an innovative adsorbent capable of specifically recognizing and efficiently adsorbing naringenin. MI COF exhibited an excellent adsorption capacity of 121 mg/g and a superior selectivity with an imprinting factor of 2.8 for naringenin. As a result, remarkably selective extraction of naringenin from citrus fruit extracts was achieved, with recoveries and purities of 85.7 % and 87.5 %, respectively. In addition, the vitro immunological and anticancer activities of naringenin enantiomers from citrus fruit was preliminarily investigated. These findings demonstrate that MI COF is an effective sorbent for the selective adsorption and separation of naringenin and its enantiomers from plants and fruits, holding significant potential for practical applications in food and clinical fields.

α-Tocopherol and naringenin in whey protein isolate particles: Partition, antioxidant activity, stability and bioaccessibility

Xin Yin,Xiaojun Fu,Hao Cheng,Wusigale,Li Liang

Food proteins are being used as carrier materials for the co-encapsulation of bioactive components. Partition of bioactive components in a carrier depends on their solubility and interaction with carrier materials. In this study, hydrophobic α-tocopherol and amphiphilic naringenin were chosen as model bioactive components to construct their complex nanoparticles with whey protein isolate (WPI). Naringenin in the molecular level was bound to WPI particles, while α-tocopherol was both bound in the molecular level and encapsulated as the aggregate in the particles. In ternary complex particles, the increased antioxidant activity of WPI-naringenin particles counteracted the protein shielding effect on the activity of α-tocopherol. Oxidation of α-tocopherol and naringenin did not cause any damage to WPI, but slightly changed the size and ζ-potential of protein particles during storage. Addition of naringenin improved the stability and bioaccessibility of α-tocopherol. The data gathered here would provide guidance for the development of protein-based co-encapsulating carriers.

Naringenin protects swine testis cells from bisphenol A-induced apoptosis via Keap1/Nrf2 signaling pathway

Huijie Chen,Jianqing Chen,Xu Shi,Lu Li,Shiwen Xu

Bisphenol A (BPA) has caused serious pathologies in varying organs of humans and animals, especially reproductive organs. Naringenin (NRG) is a flavanone compound that has shown protective effects against several environmental chemicals through suppression of oxidative stress and activation of nuclear factor erythroid 2-related factor 2 (Nrf2) pathway. Herein, we described the discovery path of NRG inhibition on apoptosis in BPA exposed swine testis (ST) cells through targeting Kelch-like ech-associated protein (Keap1). We found that NRG could specifically bound to the active residues of DGR domain in Keap1, thereby activating Nrf2 signaling pathway, and then increasing the levels of SOD, GPx and CAT, and finally inhibiting oxidative stress and mitochondrial apoptosis induced by BPA in ST cells. Altogether, our results showed that NRG inhibits oxidative stress and mitochondrial apoptosis induced by BPA in ST cells by targeting Keap1/Nrf2 signaling pathway, indicating that NRG could serve as an antagonistic therapy against BPA.

Design and preparation of naringenin loaded functional biomimetic nano-drug delivery system for Alzheimer’s disease

Chang Yan,Jinlian Gu,Shun Yin,Hao Wu,Xia Lei,Fang Geng,Ning Zhang,Xiaodan Wu

Efficient brain drug delivery has been a challenge in the treatment of Alzheimer’s Disease and other brain disorders as blood-brain barrier (BBB) impedes most drugs to reach brain. To overcome this obstacle, we developed a novel TGN decorated erythrocyte membrane-coated poly (lactic-co-glycolic acid) nanoparticle (TRNNs). The nanoparticle significantly boosted the penetration (7.3 times) in a U-118MG and HCMEC/D3 cell co-culture BBB model in vitro. Living image was performed to assess the TRNNs distribution in vivo. The fluorescence intensity in the isolated brain of TRDNs-treated mice was about 8 times that of the DNs-treated. In the novel object recognition test, the mice after administration of TRDNs showed higher recognition index (0.414 ± 0.016) than the model group (0.275 ± 0.019). A significant increase in the number of dendritic spines from TRNNs administrated mice hippocampi neurons was observed after Golgi stain. This improvement of neurons was also confirmed by the significant high expression of PSD95 protein level in hippocampi. We measured the OD values of Aβ25-35 induced PC12 cells that pre-treatment with different nanoparticles and concluded that TRNNs had a robust neuroprotection effect. Above all, functional biomimetic nanoparticles could increase the accumulation of naringenin into brain, thereby enable the drug to exert greater therapeutic effects.

Efficient multi-enzyme system for icaritin production from naringenin with synchronous prenol utilization in Escherichia coli

Anyi Wu,Junping Zhou,Yinan Xue,Yun Liu,Jidong Shen,Xin Gao,Zhiqiang Liu,Yuguo Zheng

Icaritin (ICT) shows various pharmacological activities and is used as an anti-hepatocellular carcinoma drug. While most of the studies focused on constructing yeast chassis cells for ICT production to replace the ICT extraction of Epimedium species, an efficient multi-enzyme system was first constructed here in the prokaryote Escherichia coli , for the successful biotransformation from naringenin (NAR) and prenol to ICT. The plant-derived flavonoid 3-hydroxylase (F3H) and flavonol synthase (FLS) were screened, and then fused together with protein linkers to efficiently catalyze the conversion of NAR to kaempferol (KAE). The isopentenol utilization (IU) pathway was subsequently introduced to increase dimethylallyl diphosphate (DMAPP) supply, along with catalytic activity improvement of prenyltransferase (PT) using N -terminal truncations and active site pocket engineering. Furthermore, by screening O -methytransferases (OMT) from different sources, EpOMT 4 from Epimedium pseudowushanense was selected for the ICT production. With the combination of these strategies, 80.8 ± 3.6 mg/L ICT could be produced from NAR and prenol in the recombinant E. coli , from the original 0 % degree of conversion. The systematic engineering of multi-enzyme system for ICT biosynthesis has laid the foundation for the large-scale ICT production using the industrially important E. coli chassis cell.

Stability-enhanced liposomal system co-loaded with γ-aminobutyric acid and naringenin: outstanding sleep-promoting efficacy

Ming Wu,Yu Liang,Hao Lei,Lin Li,Xia Zhang,Yi Yuan,Bing Li

BACKGROUND Due to weak stability and bioavailability, γ -aminobutyric acid (GABA) and naringenin (Nar), two agents with sleep-promoting activities, are limited in practical application. To improve their sleep-promoting efficacy, this study developed a stability-enhanced co-loaded liposomal system (GABA-Nar-Lip) as a lyophilized powder. RESULTS The non-lyophilized GABA-Nar-Lip exhibited good dispersibility, with an average particle size of 161.34 ± 1.68 nm, and achieved encapsulation efficiencies of 46.18 ± 0.32% (GABA) and 82.01 ± 1.33% (Nar), respectively. The stability of GABA-Nar-Lip was significantly enhanced after lyophilization with sugars (especially 0.10 g mL −1 trehalose) as cryoprotectants (CPAs), and maintained targeted release in the small intestine. The CPAs exerted cryoprotective effects by replacing water molecules and forming hydrogen bonds with soy phosphatidylcholine, thereby preserving the spherical structure and unilamellar vesicle morphology of the liposomes. In caffeine-induced zebrafish models, lyophilized GABA-Nar-Lip showed an inferior but still significant effect in sleep latency reduction. Moreover, it decreased nocturnal twitching activity and enhanced daytime locomotor activity on the following day, without the adverse effects associated with melatonin, such as poor sleep quality and reduced daytime activity. CONCLUSION The stabilized lyophilized GABA-Nar-Lip powder is a promising, safe sleep aid with balanced efficacy and no compromise to next-day performance, supporting its potential application in dietary sleep interventions. © 2026 Society of Chemical Industry.

Naringenin Attenuates Ox-LDL-induced Injury in HUVECs by Regulating Lipophagy Partly through the SOCE Pathway

Linhong Jiang,Dandan Bai,Yunjun Yang,Yingying Wu,Yanran Chen,Mengyuan Wang,Wenxin Wang,Haixia Wang,Yanzhe Xiong,Siqi Zhu,Xinyu He,Jun Long,Dongping Yuan,Jingwei Chen

ObjectiveNaringenin (NAR) is a naturally occurring tiny molecule that has a significant role in lipid metabolism. However, the molecular mechanism by which NAR is involved in lipid metabolism to protect ECs is not clear. This study aims to investigate the effect of NAR on autophagy in oxidized low-density lipoprotein (ox-LDL)-treated human umbilical vein endothelial cells (HUVECs) and its potential molecular mechanisms.MethodsOxidized LDL-induced HUVECs injury in vitro was treated with NAR. Chloroquine was used as an autophagy inhibitor. Ionomycin (Iono) and 2-aminoethoxydiphenylborate (2-APB) were used as an SOCE pathway agonist and an inhibitor, respectively. The autophagy levels in HUVECs were determined by quantitative real-time PCR, Western blot, and immunofluorescence methods. The concentration of calcium ions in HUVECs was measured by flow cytometry.ResultsThe findings revealed that NAR increased the viability of ox-LDL-impaired HUVECs. NAR increased the level of autophagy and decreased lipid accumulation in ox-LDL-treated HUVECs, which could interfere with chloroquine. Moreover, NAR significantly downregulated the expression of STIM1 and ORAI1 proteins and Ca2+ levels in the SOCE, which could be interfered with Iono or 2-APB, respectively.ConclusionIn summary, NAR can increase autophagy levels and decrease lipid accumulation in HUVECs, eventually protecting against ox-LDL-induced injury in HUVECs, which is associated with inhibition of the SOCE pathway.
