Complement component 1q (C1q) is the initial protein of the classical complement pathway and plays an important role in immune response against bacterial infection. In this study, the full-length of C1q subcomponent A, B and C chain genes ( C1qA , C1qB and C1qC ) were identified and characterized from Nile tilapia ( Oreochromis niloticus ). Molecular characterization of these three C1q subcomponents ( OnC1qs ) harbored conserved amino acids through analyses of multiple sequence alignment and phylogenetic tree , which were homologous to other teleost species. Expression analysis revealed that the OnC1qs were highly expressed in liver. After the in vivo challenges of Streptococcus agalactiae and LPS , the mRNA expressions of OnC1qs were significantly up-regulated in liver. Meanwhile, the concentration variation of OnC1qs at the protein level from tilapia serum after challenge with S. agalactiae were measured by a competitive-inhibition ELISA . In addition, the up-regulation expressions of OnC1qs were also demonstrated in head kidney adherent leukocytes and the cell culture medium in vitro stimulation with S. agalactiae , Aeromonas hydrophila and LPS, respectively. Moreover, the recombinant OnC1qs enhanced expression of cytokines IL-6, IL-8 and IL-10 in head kidney adherent leukocytes, and were able to bind both heat-aggregated mouse IgG and IgM. Taken together, the results of this study indicated that OnC1qs might be involved in host defense against bacterial infection in Nile tilapia.

The present study was conducted to administer a commercial multi-strain probiotic (MP), Yilibao ( Bacillus velezensis : Bacillus cereus : Lactobacillus casei = 2:2:1), in Nile tilapia, Oreochromis niloticus . In terms of aerobic Bacillus spp. counts, we produced five diets containing 0, 0.34, 1.68, 3.36 and 6.72 g/kg of MP dry product (Control, T1, T2, T3, T4). Seven hundred and fifty tilapia juveniles (13.26 ± 0.01 g) distributed into 25 tanks in five replications were fed their diet for eight weeks. Results showed that fish fed T3 and T4 diets displayed significantly higher final body weight and weight gain ( p < .05). Further, all MP-treated fish exhibited remarkably decreased plasma lipid profiles (cholesterol, triglycerides) ( p < .05) and fish fed T3 and T4 diets displayed significantly higher plasma myeloperoxidase activity and complement C3 content ( p < .05). Additionally, fish fed T3 and T4 diets exhibited significantly promoted total antioxidant capacity, glutathione peroxidase activity, glutathione level and declined malondialdehyde content in the plasma and/or liver ( p < .05). Moreover, significantly elevated chymotrypsin activity, villus height and intraepithelial lymphocytes counts were found in fish fed T3 and T4 diets ( p < .05). As fish fed T3 and T4 diets surpassed the other treatments in growth, immune–antioxidative status and gut health, the recommended dose for tilapia is 3.36–6.72 g/kg.

BACKGROUND In order to utilize tilapia skin gelatin hydrolysate protein, which is normally discarded as industrial waste in the process of fish manufacture, we study the in vivo and in vitro angiotensin-I-converting enzyme (ACE) inhibitory activity of the peptide Leu-Ser-Gly-Tyr-Gly-Pro (LSGYGP). The aim was to provide a pharmacological basis of the development of minimal side effects of ACE inhibitors by comparative analysis with captopril in molecular docking. RESULTS This peptide from protein-rich wastes showed excellent ACE inhibitory activity (IC 50 = 2.577 μmol L −1 ) and exhibited a mixed noncompetitive inhibitory pattern with Lineweaver–Burk plots. Furthermore, LSGYGP and captopril groups both showed significant decreases in blood pressure after 6 h and maintained good digestive stability over 4 h. Molecular bond interactions differentiate competitive captopril upon hydrogen bond interactions and Zn(II) interaction. The C-terminal Pro generates three interactions (hydrogen bonds, hydrophilic interactions and Van der Waals interactions) in the peptide and effectively interacts with the S1 and S2 pockets of ACE. CONCLUSION LSGYGP, with an IC 50 value of 2.577 μmol L −1 , has an antihypertensive effect in spontaneously hypertensive rats. Through comparison with captopril, this study revealed that LSGYGP may be a potential food-derived ACE inhibitory peptide and could act as a functional food ingredient to prevent hypertension. © 2019 Society of Chemical Industry

This study aimed to gain a better understanding of the long-chain (≥C 20 ) polyunsaturated fatty acid (LC-PUFA) biosynthetic metabolism, with a specific focus on the metabolic fate of linolenic acid (18:3n-3, LNA) and linoleic acid (18:2n-6, LA) and the relationship between these two substrates in the process of LC-PUFA biosynthesis in juvenile Nile tilapia , Oreochromis niloticus . Fish were fed four isoproteic (37.5%) and isolipidic (6.2%) diets formulated with fixed levels of LA (0.60%) and gradual levels of LNA (0.10, 0.63, 1.56 and 2.04% of dry weight, respectively) for 10 weeks. The whole-body fatty acid balance method was used to evaluate fish in vivo fatty acid metabolism . The results showed that most of dietary LNA was bioconverted to LC-PUFA when tilapia receiving not enough LNA for optimal growth. In contrast, when fish receiving sufficient dietary LNA, most of the LNA was β-oxidized, with only <10% LNA being bioconverted. The in vivo apparent △-5/-6 desaturase and elongase activities on n-3 and n-6 PUFA, respectively, exhibited a significant linear increase and decrease with the increasing dietary LNA levels, clearly indicating that for all of the enzyme activities , n-3 PUFA is a preferred substrate and the LC-PUFA biosynthetic pathway is substrate limited in Nile tilapia. Dietary higher inclusion of LNA could block or at least slow down n-6 LC-PUFA biosynthesis from LA, indicating that a direct substrate competition for accessing the Δ-6 desaturase and elongase exists between these two substrates. Furthermore, Nile tilapia were relatively efficient in bioconversion from LNA to DHA, and the DHA biosynthesis from EPA may be more direct or faster than the production of EPA from LNA. Despite of the active bioconversion of LNA up to DHA, this metabolic effort was still insufficient to compensate for the significant reduced dietary intake of n-3 LC-PUFA. No significant difference was observed in the gene expression of fatty acyl desaturase ( fads2 ) and fatty acid elongase ( elovl5 ) among all groups, which suggests that the total in vivo apparent enzyme activities are directly and positively affected by substrate availability rather than the extent of the enzyme gene expression.
