Background Glioma is a highly aggressive brain tumor with a poor prognosis. Photodynamic therapy (PDT) induces antitumor immunity, a key mechanism of its efficacy. Dendritic cell-derived exosomes (Dexs) are crucial for tumor antigen presentation and T-cell activation. Dexs represent a promising cell-free strategy for cancer immunotherapy. Understanding the role of Dexs in PDT-induced immunity may enhance PDT efficacy. Methods GL261 glioma cells were subjected to PDT, followed by co-culture with dendritic cells (DC) to obtain PDT-Dexs. Flow cytometry and ELISA were used to assess the effects of PDT-Dexs on DC maturation and T-cell activation. MicroRNA sequencing was performed on PDT-Dexs. The effect of PDT-Dexs on tumor growth and survival was evaluated in immune-competent and immune-deficient glioma model mice. Results Co-incubation with PDT-Dexs significantly increased DC maturation and T-cell activation. Sequencing revealed 156 up-regulated microRNAs, miR-152-3p was validated as a key functional miRNA that promotes DC maturation by targeting DNMT1 to upregulate MyD88 expression. PDT-Dexs entered the systemic circulation, increasing the serum IL-2, IFN-γ, and TNF-α levels. In immunocompetent mice, PDT-Dexs increased CD3 + CD4+ and CD3 + CD8+ T-cell percentages, slowed tumor growth, prolonged survival, and were associated with enhanced CD8 + T-cell infiltration and tumor cell apoptosis. Crucially, PDT-Dexs failed to inhibit tumor growth or improve survival in immunodeficient nude mice. Conclusions PDT-Dexs promote DC maturation and T-cell activation, improving antitumor immunity and PDT efficacy. Combining of PDT with PDT-Dexs further improves antitumor immunity and PDT efficacy. PDT-Dexs warrants further investigation as a potential strategy for enhancing antitumor immunity in glioma. Graphical The alternative text for this image may have been generated using AI.

Pathological scars are fibrotic disorders arising from aberrant wound healing. PDT is effective but limited by a dense matrix, hypoxia, and post-PDT excessive ROS-induced secondary inflammation. To address this, a NIR-switchable ROS-regulatory liposome (ICC@Lip) with CaO2 NPs (oxygenation to strengthen PDT) and Cu5.4O nanozymes (ROS scavenging) was developed. In addition, inspired by Cyperaceae stems and triangular arrows, a highly penetrable biomimetic microneedle platform loaded with ICC@Lip and collagenase I (ICC@Lip/HPMN) was constructed to realize efficient transdermal cargo delivery and diffusion into a rigid pathological scar matrix. The puncture resistance of ICC@Lip/HPMN was only 56.4% that of conventional rectangular pyramid microneedles, with an effective puncture rate of 96% in mouse skin. In vitro, under NIR irradiation-mediated PDT/PTT, ICC@Lip significantly alleviated intracellular hypoxia and related fibrotic indicators, achieving a cytotoxicity rate of 93.71% against myofibroblasts─1.49-fold higher than that of the IR808 alone group (62.74%)─while reducing M2 macrophage polarization and TGF-β secretion. Notably, timely post-PDT ROS clearance decreased M1 polarization of macrophages and reduced the release of over 50% of inflammatory cytokines. In rabbit ear, a hypertrophic scar model exhibited that ICC@Lip/HPMN reduced the scar elevation index from 3 to 1.7, significantly alleviating collagen deposition and inflammation. The antifibrotic efficacies were majorly exerted through cross-downregulation of TGF-β, IL-4, and toll-like receptor signaling pathways. This study provided a promising adaptive therapeutic strategy of self-strengthened PDT coupling ROS precise regulation for pathological scars.

Antibacterial photodynamic therapy (aPDT) has emerged as an attractive treatment option for efficient removal of pathogenic bacteria. However, aPDT in deep tissue will encounter difficulties such as limited light penetration depth, insufficient oxygen (O 2 ) supply and inability to eliminate inflammation introduced by bacteria, which hinders its clinical application. Herein, the near infrared (NIR) strategy of simultaneously generating O 2 and CO was developed for aPDT based antibacterial therapy and mitigation of deep infection inflammation. Methods: We prepared NIR-mediated multifunctional aPDT nanoplatform (POS-UCNPs/ICG) producing therapeutic gas of O 2 and CO. The CO, O 2 and ROS generation of the nanoplatform were characterized by dye probes, respectively. The antibacterial activity and anti-inflammation of POS-UCNPs/ICG were demonstrated in vitro and in vivo . In addition, the therapeutic effects in vivo were serially analyzed by immunofluorescence staining, Masson's staining, hematoxylin and eosin staining, colony formation units (CFU) and so on. Results: NIR-mediated multifunctional aPDT nanoplatform was realized by combining the up-conversion nanoparticles (UCNPs) and partially oxidized SnS 2 (POS) nanosheets (NSs) as well as indocyanine green (ICG). Using a single 808 nm light, aPDT can be achieved via ICG molecules, meanwhile, O 2 /CO can be generated by POS NSs through upconversion light excitation. During the aPDT process, O 2 can enhance aPDT, while CO can regulate inflammation through the PI3K/NF-κB pathway. Therefore, POS-UCNPs/ICG groups had a highest percentage of healing area up to 91.55±1.26% in mouse abscess model. Conclusion: Due to enhanced aPDT and anti-inflammatory collaborative therapy, the POS-UCNPs/ICG composites showed remarkably accelerated recovery in animal abscess models. Such NIR light responsive nanoplatform with optimized antibacterial capacity and immunomodulatory functions is promising for clinical therapeutics of bacteria-induced infections.

We designed and synthesized three new berberine-based compounds, namely, pyridine-2,6-dimethyl-/2,2′-bipyridine-3,3′-dimethyl-tethered berberine dimers BD1 and BD2, and a tetrakis(4-benzyl)ethylene linked berberine tetramer BD4. We identified that the dimer BD2 and tetramer BD4, as well as 1,10-phenanthroline-2,9-dimethyl-linked berberine dimer BD3 previously reported by us, showed remarkable aggregation-induced emission (AIE) properties which endowed them with higher singlet oxygen (1O2) production ability than berberine. Of the four compounds, BD3 exhibits the lowest ΔEST energy with the highest 1O2 generation ability and thus was selected for further construction of AuNSs–BD3@HA (denoted as ABH, AuNSs = gold nanostars; HA = hyaluronic acid). The nanosystem of ABH shows a remarkable therapeutic effect toward breast cancer by combining photodynamic therapy (PDT) from BD3, photothermal therapy (PTT) from AuNSs, and the CD44-targeting capability of HA. The synergistically enhanced PDT and PTT induce superior cancer cell apoptosis/necrosis in vitro and anti-breast cancer activity in vivo. This study provides a new concept for PDT using natural product derivatives and their combination with PTT for efficient treatment of tumors.

Choroidal melanoma, the most prevalent subtype of uveal melanoma with a bleak prognosis, necessitates safer and more efficient treatment methods. Tumor cell membrane-modified nanodelivery systems have exhibited significant enhancement in photodynamic therapy (PDT). Currently, photosensitizers utilized in PDT encompass porphyrin-related organic molecules and other types, requiring complex and intricate systems construction. This study focuses on harnessing the naturally occurring photodynamic active photosynthetic unit-thylakoid which serve as a major source of singlet oxygen. Initially, thylakoids extracted from fresh leaves were identified based on their absorbance value, morphology, and characteristic proteins. The tumor cell membrane-thylakoid membrane (CM-Thy) photodynamic therapy system was prepared through ultrasonic fragmentation and liposomal extrusion technology. Due to its membrane targeting ability, CM-Thy demonstrated excellent uptake performance towards OCM-1 cells within intraocular tissues. Both in vitro and in vivo experiments confirmed CM-Thy's photodynamic activity. Specifically, Thy in the system generates singlet oxygen, after cellular internalization, the elevated intracellular reactive oxygen species (ROS) triggered by this process induce cell lipid peroxidation and increased membrane permeability, while singlet oxygen further mediates DNA oxidative damage, reduced cell proliferation capacity, and NLRP3 inflammasome-mediated pyroptosis, all these effects synergistically contribute to ultimate tumor cell apoptosis. Furthermore, the anti-tumor effect of CM-Thy was validated through various mechanisms involved in tumor formation such as angiogenesis and vasculogenic mimicry. Interestingly, the results from visible light experiments conducted in vitro also substantiated the remarkable therapeutic efficacy of this system for refractive eye disorders while providing innovative ideas for cross-species biological interventions.

Based on the properties of photodynamic therapy (PDT) and magnetic resonance imaging (MRI), combining them to construct multifunctional nanotheranostics can leverage strengths and avoid weaknesses for tumor diagnosis and treatment. However, certain problems remain unsolved, notably the short observation window caused by insufficient retention time. In this study, a GSH-responsive shape-transformable nanotheranostics Gd-Ce6-FFVLGGGC-SS-PEG are designed (abbreviated as GdCFS) by combining a single metal Gd with Ce6, and peptide (Phe-Phe-Val-Leu-Gly-Gly-Gly-Cys) disulfide-conjugated with polyethylene glycol (PEG) to perform dual-modal T 1 /T 2 MRI specifically at the tumor site. Due to its amphiphilic features, GdCFS can self-assemble to spherical nanoparticles, while transforming to nanofibers (NFs) in the presence of intracellular overexpressed GSH stimuli. After NFs formation, the hydrophobic core of GdCFS is exposed, resulting in T 1 positive contrast enhancement, whereas NFs can simultaneously achieve T 2 negative contrast enhancement. Furthermore, GSH depletion can cause imbalance in intracellular redox state, ultimately improving PDT efficacy. GdCFS benefits from the shape-transformation in terms of sufficient retention time, thus positively minimizing the toxicity risk. On the tumor model of nasopharyngeal carcinoma, in vivo and in vitro outcomes confirm that GdCFS can be a promising candidate for early diagnosis, real-time monitoring, and precise treatment of tumors with great biocompatibility.

The combinations of phototherapeutic modalities with chemotherapy represent an innovative antitumor strategy. Herein, we constructed a NIR laser regulated nanoplatform (PTE@TPP-NM(DOX/IR780)) via the self-assembly process of PEG-b-PCL modified with mitochondrial targeting triphenylphosphine (TPP), reactive oxygen species (ROS)-sensitive polymer (PTE), DOX, and IR780. Under 808 nm laser irradiation, IR780 in PTE@TPP-NM(DOX/IR780) generated abundant reactive oxygen species (ROS) and heat for photothermal therapy (PTT) and photodynamic therapy (PDT), while ROS oxidized PTE to release DOX for chemotherapy. The signal 808 nm laser could effectively regulate PTT, PDT, and chemotherapy in real-time, enhancing the synergistic efficacy. PTE@TPP-NM(DOX/IR780) could result in depolarization of the mitochondrial membrane and inhibition of the expression of heat shock proteins to amplify antitumor effect. Enhanced therapeutic effect was demonstrated via in vivo and in vitro experiment. This study provided a promising strategy for the design of multifunctional nanocarriers to address the limitation of combined therapy.

Phototherapy has attracted widespread attention for cancer treatment due to its noninvasiveness and high selectivity. However, severe hypoxia, overexpressed glutathione and high levels of hydrogen peroxide (H 2 O 2 ) of tumor microenvironment limit the antitumor efficiency of phototherapy. Herein, inspired by the specific response of nanozymes to the tumor microenvironment, a simple and versatile nanozyme-mediated synergistic dual phototherapy nanoplatform is constructed. In this study, tin ferrite (SnFe 2 O 4 , SFO) nanozyme as a photosensitizer was surface modified with polydopamine (denoted as P-SFO) and incorporated into poly( l -lactide) to fabricate an antitumor scaffold fabricated by selective laser sintering. On one hand, SFO nanozyme could act as a photoabsorber to convert light energy into heat for photothermal therapy (PTT). On the other hand, it played a role of photosensitizer in transferring the photon energy to generate reactive oxygen species (ROS) for photodynamic therapy (PDT). Importantly, its multivalent metal ions redox couples would decompose H 2 O 2 into O 2 for enhancing O 2 -dependent PDT and consume glutathione to relieve antioxidant capability of the tumors. Besides, polydopamine as a photothermal conversion agent further enhanced the photothermal performance of SFO. The results revealed the PLLA/P-SFO scaffold possessed a photothermal conversion efficiency of 43.52% for PTT and a high ROS generation capacity of highly toxic ·O 2 − and ·OH for PDT. Consequently, the scaffold displayed a prominent phototherapeutic effect with antitumor rate of 96.3%. In addition, the PLLA/P-SFO scaffolds possessed good biocompatibility for cell growth. These advantages endow PLLA/P-SFO scaffold with extensive applications in biomedical fields and opened up new avenue towards nanozyme-mediated synergistic phototherapy.

To improve the therapeutic effect of tumors, developing well-defined hybrid metal-organic frameworks (MOFs) through MOF-on-MOF growth has attracted much attention. In this work, an in situ growth strategy was reported to assemble doxorubicin (DOX)-encapsulated zeolitic imidazolate frameworks (ZIF-8) on the surface of Zr(IV)-based porphyrinic MOFs. Such an approach offers a high controllability in the thickness of the ZIF-8 layer without any requirement of surface modifications on porphyrinic MOFs. Impressively, the acidic pH-response degradation of ZIF-8 promoted the release of DOX in the acidic tumor microenvironment. Upon light irradiation, porphyrinic MOFs can harvest photons to produce plentiful singlet oxygen, thus leading to amplified chemo/PDT therapeutic efficacy. This work paves the way to utilize the hybrid engineering of MOFs for synergistic cancer therapy.

In this study, biosynthetic Ag@AgCl plasma is used as a photosensitizer for the first time to construct a photothermal/photodynamic/chemotherapy and pH-responsive synergistic antitumor nanomedicine delivery system CS-Ag@AgCl/C. The Ag@AgCl and AgNPs synthesized by the extracts of alpine banana, C itrus reticulata , and Citrus sinensis are characterized. The Ag@AgCl NPs synthesized with banana extract have a regular crystal shape and a granular size of 12 nm, and they are more stable than AgNPs synthesized with the other two extracts. In addition, Ag@AgCl plasma used as a photosensitizer can reduce the dependence of CS-Ag@AgCl/C on oxygen in the photodynamic process. After coating with chitosan, though the photothermal conversion efficiency of CS-Ag@AgCl/C decreases from 44.06% to 29.51%, the pH response character of it increases, and the pH response plays an important role in drug release experiments. Under an acidic environment of pH = 5.0 phosphate-buffered saline buffer, the drug release rate of the samples increases significantly. In the synergistic antitumor experiment, the cell survival rate increases from 36.83% to 78.69% after chitosan-coated Ag@AgCl/C NPs. When CS-Ag@AgCl/C-DOX+808 nm is co-cultured with HeLa cells, the cell survival rate is only 14.89%, which indicates that the constructed plasma drug delivery system has excellent photothermal/photodynamic/chemotherapy combined treatment capability.

DIPF was fabricated via drug self-assembly, with high encapsulation efficiency and loading capacity of both DOX and ICG. • DIPF exhibits strong ability to penetrate the BBB, to accumulate in gliomas, and enables real-time in vivo imaging. • DIPF integrates chemotherapy, PTT and PDT to inhibit gliomas, overcome drug resistance and reduce DOX-induced cardiotoxicity. • DIPF is a promising delivery platform for multi‑therapy glioma treatment. Combination therapy tactics that incorporate numerous therapies via innovative multi-drug co-loaded nanomedicine delivery systems have shown considerable benefits in crossing the blood-brain barrier (BBB) and slowing glioma growth. However, efficiently developing multi-drug co-loaded delivery systems with high drug loading capacity, the ability to alleviate drug resistance, and minimal in vivo toxicity remains a significant problem. In this study, we created a self-assembling nanomedicine delivery system using a DIPF composite encapsulated with polydopamine and folate. The nanoparticles have an internally coated self-assembled structure that is assembled via electrostatic interactions, π-π stacking, and hydrophobic interactions between doxorubicin (DOX) and indocyanine green (ICG). This achieves high encapsulation efficiency and loading content for the co-delivery of multiple drugs. In this delivery system, the encapsulation efficiencies of DOX and ICG reached 94.08% and 94.01%, respectively, while the inherent cardiotoxicity of DOX was reduced by 64.33%. H&E staining results and anti-tumor experimental outcomes—including expression of TNF-α, Ki67, Caspase-3, and GFAP cytokines—clearly demonstrated that combined DIPF+NIR therapy significantly inhibited tumor growth and substantially extended survival. The highly efficient multi-drug co-loaded delivery system developed here demonstrates significant application potential for active targeting, controlled drug release, treatment visualization, and combined chemotherapy, photothermal (PTT), and photodynamic (PDT) therapy for brain tumors. A highly efficient multi-drug co-loaded drug delivery system that can cross the BBB and delivery combined chemotherapy, photothermal and photodynamic therapy for brain tumors target was established in a self-assembling way via electrostatic interactions, π-π stacking, and hydrophobic interactions between doxorubicin and indocyanine green. Photodynamic therapy (PDT) induces apoptosis in cancer cells by generating cytotoxic reactive oxygen species (ROS); however, this effect is constrained by the intrinsic antioxidant mechanisms of the cells and the hypoxic conditions of the tumor microenvironment (TME). To overcome these limitations, a novel ferroptosis-boosted nanophotosensitizer (FCE NPs) is developed after loading alkaloid evodiamine based on an acid-responsive nanoplatform (FC NPs) composed of ferric ions and Chlorin e6 (Ce6). Upon exposure to laser irradiation, the photosensitizer Ce6 efficiently generates singlet oxygen (1O2), which depletes glutathione (GSH), thereby disrupting the redox homeostasis within tumor cells and leading to the oxidation of cellular unsaturated lipid into lipid peroxides (LPO). Additionally, the Fenton reaction between released Fe(III) ions and H2O2 in the TME produces highly cytotoxic hydroxyl radicals (•OH), a form of ROS, causing the accumulation of LPO together with 1O2 to trigger ferroptosis. Following the incorporation of evodiamine, the integrated nanoplatform FCE NPs exhibited excellent antitumor activity through high-performance ferroptosis by inhibiting a glutathione peroxidase 4 (GPX4) level. It was also observed that the key regulator of Wnt signaling pathway β-catenin was downregulated, suggesting the inhibition of FCE NPs on the Wnt signaling pathway. Therefore, this study demonstrated that the FCE NPs could lead to the explosive production of ROS and simultaneous inhibition of GPX4/GSH pathways and the Wnt signaling pathway in colorectal cancer, significantly amplifying the tumoricidal efficacy of PDT through the synergistical sensitization of ferroptosis. This approach offers a promising strategy for the clinical application of ferroptosis therapy.
