Perfluorooctanesulfonate (PFOS) is a widespread, persistent environmental pollutant that exerts apparent liver toxicity. Flaxseed oil (FO), a dietary oil rich in α-linolenic acid, has been demonstrated to possess a diverse array of health benefits. However, whether FO protects against PFOS-induced liver injury and its underlying mechanisms remain unclear. C57/BL6 mice were orally treated with different concentrations of FO alone or in combination with 10 mg/kg of PFOS for 28 consecutive days. Blood and liver tissues were collected for proteomic, histopathological, biochemical, immunohistochemical, and molecular examinations. Results demonstrated that FO supplementation reduced PFOS-induced liver injury, as evidenced by a decrease in histopathological changes, serum transaminase (ALT and AST) levels, levels of oxidative stress, and inflammatory cytokine (TNF-α, IL-1β, and IL-6) levels. Proteomic analyses showed that differentially expressed proteins were enriched in cholesterol metabolic pathways when comparing the PFOS group to the FO supplementation groups. The expression of cholesterol metabolism-related proteins was also subsequently measured, revealing that FO supplementation decreased the protein expressions of SREBP2, HMGCR, and LDLR while increasing the expression of CYP7A1. This study demonstrates that FO can alleviate PFOS-induced hepatotoxicity by regulating hepatic cholesterol metabolism, indicating that FO may serve as an effective dietary intervention for preventing liver injury caused by PFOS.

Perfluorooctane sulfonate (PFOS) is a representative persistent organic pollutant that exerts toxic effects on aquatic organisms. As an alternative to PFOS, sodiump-perfluorous nonenoxybenzene sulfonate (OBS) has been frequently detected in aquatic environments and human tissues in recent years. However, its toxic effects on aquatic organisms and potential health risks to humans remain unclear. Zebrafish embryos are transparent and amenable to in vivo manipulation and observation. Therefore, in the present study, we investigated its developmental toxicity in zebrafish embryos, with PFOS as the positive control. We exposed zebrafish embryos to different concentrations of OBS (15, 20, and 25 mg/L) and PFOS (15 mg/L) for 2–168 h post fertilization (hpf) and then examined physiological and gene expression changes. At 24 hpf, spontaneous twitches in the 25 mg/L OBS group decreased to (5 ± 0.34)/min. By 48 hpf, the 20 mg/L OBS group’s hatching rate was (47.78 ± 2.22)%, significantly lower than the control. At 72 hpf, heart rates in both the PFOS and OBS groups were elevated, at 82 ± 0.6, 84.5 ± 0.5, 89.4 ± 0.3, and 93.7 ± 0.4, respectively. Similarly to PFOS, OBS induced developmental toxicity in zebrafish embryos. In addition, both OBS and PFOS exposure downregulated the expression level of anti-apoptotic Bcl-2 in zebrafish embryos, with a notable 0.53-fold decrease observed in the 25 mg/L OBS group. Conversely, they upregulated the expression levels of pro-apoptotic Bax, Caspase-3, and Caspase-9, with Caspase-3 expression increasing 1.14-, 1.5-, and 1.7-fold in the 15 mg/L PFOS, 20 mg/L OBS, and 25 mg/L OBS groups, respectively. These OBS- and PFOS-induced changes in gene expression increased apoptosis, suggesting that OBS can induce developmental toxicity in zebrafish embryos, and that its effect is comparable to that of PFOS. Therefore, considering its aquatic toxicity, measures aimed at limiting or remediating OBS pollution in the environment are necessary.

Various commercial and industrial products widely use highly toxic eight-carbon-chain perfluorooctanesulfonate (PFOS), posing a significant threat to the health of living organisms. In this study, the electrochemical detection of PFOS was achieved by developing a carbon paste electrode (CPE) using the Mo 2 Ti 2 AlC 3 MAX phase. Mo 2 Ti 2 AlC 3 was synthesized and directly used to construct the CPE. The electrochemical performance of the prepared sensor was tested using various electrochemical techniques, such as cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), differential pulse voltammetry (DPV), and amperometric titration. The developed electrochemical sensor exhibited two linear ranges from 0.001 to 0.09 μM and from 1.1 to 62.6 μM, with a detection limit of 0.04 nM. The sensor demonstrated high sensitivity, measuring 145.1 μA μM −1 cm −2 , and a response time of 5 s for PFOS quantification at a working potential of 0.3 V. Additionally, the sensor demonstrated outstanding resistance to typical interfering chemicals. The applicability and reliability of the developed sensor for PFOS determination were further tested in real samples, yielding recoveries in the range of 92.6–108.2%, with relative standard deviation (RSD) values between 1.8% and 3.7%. The Mo 2 Ti 2 AlC 3 MAX phase-based electrochemical sensor is simple, rapid, sensitive, and cost-effective, making it a promising approach for the quantification of PFOS in environmental water and soil samples.

A simple, highly sensitive and selective assay method has been developed for the determination of perfluorooctane sulfonate (PFOS), a kind of emerging persistent organic pollutant, which is bio-accumulative, resistant to degradation and toxic. It was found that PFOS can form complexes with protonated Janus Green B (JGB) through electrostatic interaction in pH 5.8 citric acid–sodium citrate (CA–NaCA) buffer medium resulting in a remarkable enhancement of the resonance light scattering (RLS) intensity of the system. The study shows that the maximum scattering wavelength is located at 319.0 nm and the enhanced RLS intensities are in proportion to the concentration of PFOS in the range of 0.05–9.0 μmol L−1 (R2 = 0.9958), with a detection limit of 5.6 nmol L−1 (3σ). Under the same experimental conditions, various perfluorinated compounds (PFPrA, PFBA, PFPeA, PFHeA, PFHpA, PFOA, PFNA, and PFBS) and JGB interactions were studied and the scattering signal changes very little. Based on the above discussion, a simple, highly sensitive and selective resonance light scattering analysis method was established for the detection of PFOS. In this work, the optimum reaction conditions were investigated. The reaction mechanism between JGB and PFOS was also studied by the absorption spectrum, scanning electron microscopy (SEM), the combination ratio and so on. This method is successfully applied to the selective detection of PFOS in environmental water samples with RSD ≤2.7%.

As one of the most reactive species, hydrated electron (e aq − ) is promising for reductive decomposition of recalcitrant organic pollutants, such as perfluorooctane sulfonate (PFOS). In this study, PFOS decomposition using a vacuum ultraviolet (VUV)/sulfite system was systematically investigated in comparison with sole VUV and ultraviolet (UV)/sulfite systems. A fast and nearly complete (97.3%) PFOS decomposition was observed within 4 h from its initial concentration of 37.2 μM in the VUV/sulfite system. The observed rate constant ( k obs ) for PFOS decomposition in the studied system was 0.87 ± 0.0060 h − 1 , which was nearly 7.5 and 2 folds faster than that in sole VUV and UV/sulfite systems, respectively. Compared to previously studied UV/sulfite system, VUV/sulfite system enhanced PFOS decomposition in both weak acidic and alkaline pH conditions. In weak acidic condition (pH 6.0), PFOS predominantly decomposed via direct VUV photolysis, whereas in alkaline condition (pH > 9.0), PFOS decomposition was mainly induced by e aq − generated from both sulfite and VUV photolytic reactions. At a fixed initial solution pH (pH 10.0), PFOS decomposition kinetics showed a positive linear dependence with sulfite dosage. The co-presence of humic acid (HA) and NO 3 − obviously suppressed PFOS decomposition, whereas HCO 3 – showed marginal inhibition. A few amount of short chain perfluorocarboxylic acids (PFCAs) were detected in PFOS decomposition process, and a high defluorination efficiency (75.4%) was achieved. These results suggested most fluorine atoms in PFOS molecule ultimately mineralized into fluoride ions, and the mechanisms for PFOS decomposition in the VUV/sulfite system were proposed.

Because of the serious harm to animals and the environment associated with perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA), a rapid, sensitive and low-cost method for detecting PFOS and PFOA is of great importance. In this paper, a novel sensing method has been proposed for the highly sensitive detection of PFOS and PFOA in environmental water samples based on the “turn-on” switch of erythrosine B (EB)-hexadecyltrimethylammonium bromide (CTAB) system. In pH 8.55 Britton-Robinson (BR) buffer, EB can react with CTAB by electrostatic attraction, resulting in a strong fluorescence quenching of EB. With a subsequent addition of the CTAB, a red-shift occurred (11 nm), followed by a significant increase in fluorescence at high surfactant concentrations. It was found that PFOS and PFOA can obviously enhance fluorescence intensity of EB-CTAB system. The enhanced fluorescence intensity is proportional to the concentration of PFOS and PFOA in the range of 0.05–10 μM with detection limit of 12.8 nM and 11.8 nM (3σ), respectively. The presented assay has been successfully applied to sensing PFOS and PFOA in real water samples with RSD ≤ 4.3% and 2.9%, respectively.

Background Perfluorooctane sulfonate (PFOS) is a prototypical persistent organic pollutant that has been linked to liver inflammation. Sesamin (Ses), a major lignan in sesame, has been shown to exhibit health-promoting properties. However, the role and specific mechanism of Ses in PFOS-induced liver inflammation remain largely unclear. Purpose This study aimed to investigate the hepatoprotection and potential mechanism of Ses against PFOS-induced inflammation. Methods Wild-type (WT), Mincle knockout (Ko-mincle), and Hepatocyte-specific knockout Foxo1 (Hko-Foxo1) mice were intragastrically administered Ses and/or PFOS (10 mg/kg) for 4 weeks. In vitro, HepG2 cells were pretreated with Ses for 1 h, then exposed to PFOS for 24 h to establish a cell injury model. Results We revealed that Ses markedly alleviated PFOS-induced liver injury and inflammation, as evidenced by noticeable histopathological improvements, increased cell activity, and alterations in serum liver enzyme and inflammatory factor levels. Moreover, Ses pretreatment reduced the expression of Mincle and the phosphorylation of its downstream targets, SYK and p65, in macrophages. The reducing effect of Ses on liver inflammation in wild-type mice is significantly better than that in Mincle knockout mice. We also found that Ses decreased the expression of spliceosome-associated protein 130 (SAP130) and Foxo1. Further experiments revealed that SAP130 may serve as a mediator between hepatocytes and macrophages. Hepatocyte-specific knockout Foxo1 reduced the release of SAP130 from hepatocytes and inhibited Mincle/Syk signaling activation in macrophages. Conclusion This study demonstrates that Ses protects against PFOS-induced liver inflammation in mice by modulating crosstalk between hepatocytes and macrophages. Those findings support the potential of Ses as a promising natural compound for counteracting liver inflammation.

Organic micropollutants (OMPs), including both regulated and emerging contaminants, have been extensively detected in ground and surface waters, posing hazards to organisms and humans. Rapid and efficient removal of OMPs from environmental water by novel sorbents is still faced with severe challenges. Here we developed a magnetic nanoparticles-attached fluorographene-based (MNPs@FG) sorbent with high affinity to OMPs such as perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) compared to powdered activated carbon, the most widespread sorbent for OMPs removal. The removal efficiencies of PFOA and PFOS using the MNPs@FG sorbent were systematically examined at room temperature. The MNPs@FG sorbent showed 92–95% and 94–97% removal efficiencies of PFOA (180 μg/L) and PFOS (180 μg/L) in 2 min respectively, with relatively high sorption capacities for PFOA (50.4 mg/g) and PFOS (17.2 mg/g). The adsorption performance was stable even in the presence of natural organic matter. Moreover, the MNPs@FG sorbent could notably reduce PFOA and PFOS from ∼5 μg/L (an environmentally relevant concentration) to <50 ng/L in environmental water, lower than the U.S. Environmental Protection Agency health advisory level (70 ng/L). Besides PFOA and PFOS, the MNPs@FG sorbent also showed high removal efficiencies (97–99%) of difloxacin hydrochloride (DIF), acetochlor (ACE) and chlorantraniliprole (CHL) in short time (e.g. 10 min). The used MNPs@FG sorbent was facilely regenerated by methanol washing and reused five times without reduction in PFOA/PFOS removal and magnetic performance. Thus, this study provided a novel sorbent for rapid and effective removal of OMPs from environmental water.

The electroplating industry is an important source of per- and polyfluoroalkyl substances (PFAS) contamination, but there is a lack of comprehensive studies on the occurrence, transport, and removal of PFAS in electroplating parks. In this study, we investigated typical electroplating parks in China and conducted the first full-scale removal of PFAS from chromium-plating wastewater using pore-enlarged granular activated carbon (GAC) and hydrophobic anion exchange resin (AER). The results showed that 6:2 fluorotelomer sulfonate (6:2 FTS) gradually replaced perfluorooctanesulfonate (PFOS) in China’s electroplating industry. The conventional reduction-sedimentation process hardly removed 6:2 FTS from chromium-plating wastewater, while the special air flotation process resulted in over 60% of PFOS entering the chromium sludge cake. Based on the full-scale evaluation, GAC and AER adsorption were feasible technologies for removing PFAS from chromium-plating wastewater, among which AER had higher adsorption capacity and removal efficiency for PFAS but poorer selectivity for 6:2 FTS than for PFOS. It is estimated that GAC and AER adsorption have the potential to reduce the discharge of PFAS by 5030–8000 kg/year in China. This study reveals the current status of PFAS contamination in China’s electroplating industry and provides feasible technologies for PFAS control.

Perfluoroalkyl sulfonate (PFOS) is a commonly used chemical compound that often found in materials such as waterproofing agents, food packaging, and fire retardants. Known for its stability and persistence in the environment, PFOS can enter the human body through various pathways, including water and the food chain, raising concerns about its potential harm to human health. Previous studies have suggested a cardiac toxicity of PFOS, but the specific cellular mechanisms remained unclear. Here, by using AC16 cardiomyocyte as a model to investigate the molecular mechanisms potential the cardiac toxicity of PFOS. Our findings revealed that PFOS exposure reduced cell viability and induces apoptosis in human cardiomyocyte. Proteomic analysis and molecular biological techniques showed that the Endoplasmic Reticulum (ER) stress-related pathways were activated, while the cellular autophagy flux was inhibited in PFOS-exposed cells. Subsequently, we employed strategies such as autophagy activation and ER stress inhibition to alleviate the PFOS-induced apoptosis in AC16 cells. These results collectively suggest that PFOS-induced ER stress activation and autophagy flux inhibition contribute to cardiomyocyte apoptosis, providing new insights into the mechanisms of PFOS-induced cardiomyocyte toxicity.

Perfluorooctane sulfonate (PFOS) is an emerging organic pollutant that is prevalent in the environment. The high toxicity, bioaccumulation and environmental persistence of PFOS pose significant risks to ecosystems and human health, and remediation technologies are urgently needed. However, conventional treatment techniques have proven difficult, necessitating the development of more affordable alternatives to adsorbents. Biochar shows great potential as an inexpensive adsorbent for PFOS removal owing to its excellent pore structure, surface properties, and ability to generate persistent free radicals (PFRs). In this study, biochar is prepared from bamboo powder at 300, 500, and 700 °C (BP300, BP500, and BP700). BP500 showed a more developed pore structure, larger specific surface area, and higher PFR signals. The removal efficiency of PFOS on BP500 (49.45 %) was superior to that on BP300 and BP700. The influence of environmental factors (biochar dosage, pH, kinetics, isotherms, and inorganic anions) on the adsorption behavior of PFOS was determined. The removal of PFOS increased at low pH, reaching 63.77 % at pH 2, while the presence of inorganic anions in the solution inhibited PFOS removal by BP500. The presence of oxidants (H 2 O 2 and S 2 O 8 2− ) alongside BP500 in the solution improved PFOS removal efficiency. The oxidant added to biochar could degrade PFOS, as evidenced by the detection of F − , and the removal and degradation rates increased with the oxidant dosage. This study provides a new perspective for analyzing the mechanism of PFOS removal by biochar. In the future, the industrial-scale application of bamboo biochar for contaminant removal needs to be strengthened.

Perfluorooctane sulfonate (PFOS), classified as a persistent organic pollutant, promotes mitochondrial iron overload through voltage-dependent anion channel 2 (VDAC2). However, the source of iron transported through VDAC2 remains unclear. Here, we reported that PFOS enhanced extracellular iron influx into human hepatocytes HepG2, which peaked at 6 h. Under PFOS exposure, total VDAC2 protein levels remained unchanged, whereas its expression increased in mitochondrial lysates but decreased in cytoplasmic lysates, which were observed both in HepG2 cells and mice liver. Meanwhile, the total protein expression of the extracellular iron importer ZRT/IRT-like protein 14 (ZIP14) was upregulated, whereas absent from the mitochondrial lysates. Knockdown of either VDAC2 or ZIP14 inhibited PFOS-induced extracellular iron influx and mitochondrial iron overload, but did not affect cytoplasmic iron levels. Notably, immunofluorescence colocalization analysis revealed that the timeline of mitochondrial redistribution toward the plasma membrane closely paralleled the course of increased extracellular iron influx. Furthermore, the result of molecular docking suggested the potential interaction between VDAC2 and ZIP14. Together, these works pointed out and further advanced our understanding of the extracellular iron transport into mitochondria in hepatocytes under PFOS exposure and revealed a potential therapeutic target for mitigating the disease mediated by iron-related mechanism.
