Dietary chlorogenic acid regulates gut microbiota, serum-free amino acids and colonic serotonin levels in growing pigs

Yi Wu,Wenhui Liu,Qi Li,Yafei Li,Yali Yan,Fang Huang,Xin Wu,Quancheng Zhou,Xugang Shu,Zheng Ruan

Chlorogenic acid (CGA) has many biological properties, including antibacterial, antioxidant and anti‐inflammatory properties, and is one of the most abundant phenolic acids available in the human diet. The aim of this study was to investigate the effects of CGA on regulation of the gut microbiota, and on the levels of free amino acids and 5-hydroxytryptamine (5-HT, serotonin). Ninety-six healthy growing pigs were randomly assigned to two treatment groups: the Ctrl group (control group, standard feed) and the CGA group [standard feed plus 0.05% 3-caffeoylquinic acid (3-CQA)] for 60 days. The diversity of the gut microbiota was increased after CGA supplementation. Changes in these microbes were significantly associated with the serum free amino acid levels and colonic 5-HT level. Compared with the Ctrl group, the levels of serum aspartic acid, threonine, alanine, arginine, and colonic 5-HT were significantly increased (p < .05). These data suggest important roles for CGA in regulating the gut microbiota and increasing the serum free amino acid levels.

An Interlocked Bone/Fiber Construct Recapitulating the Native Soft‒Hard Tissue Interfaces for Periodontal Complex Regeneration in Pigs

Yi Tian,Xuan Li,Yu-Zhe Chen,Dian Gan,Dao-Kun Deng,Lin Jing,Fang Li,Wen-Jie Zhang,Rui-Xin Wu,Fen Liu,Jia Wang,Bei-Min Tian,Fa-Ming Chen,Xiao-Tao He

The repair of periodontal complexes poses a significant challenge due to the lack of functional material constructs that can recapitulate the hierarchically interlocked structures of the soft–hard tissue interfaces within periodontium. Here, a bioinspired construct is fabricated by engineering interlocked bone–fiber interfaces within its hierarchical architectures to mimic the multiscale features found in native periodontal ligament (PDL)‒bone enthesis. The 3D-printed bone module and electrospun fiber module of the bioinspired constructs offered suitable architecture, biomechanics, and cytokine landscapes to recruit reparative cells and promote their osteogenic and fibrogenic differentiation. At the interfaces of the dual-module construct, the electrospun nanofibers of the fiber module are inserted into the bone module owing to various intermolecular interactions, which facilitate strong conjunction between the dual modules and provided biophysical gradients along the inserted nanofibers, similar to the microstructure and micromechanics of PDL‒bone insertions. When the constructs are placed in periodontal defects in pigs for 12 weeks, the alveolar bone is restored to its native level, and newly formed mature collagen fibers are inserted into the regenerated alveolar bone, culminating in superb integration between the soft and hard tissue interfaces. The findings demonstrate that bioinspired constructs with interlocked bone–fiber interfaces can regenerate complex soft‒hard tissue interfaces in pigs.

The effect of Zexie decoction on vestibular and auditory function in DDAVP-induced endolymphatic hydrops of Guinea pigs

Renlong Ji,Yanchang Xu,Kang Li,Wen Jiang,Yuan Li,Jianwei Zeng,Wei Li,Caiji Wang,Zeqi Zhao,Yalan Li,Naveena Konduru,Wen Liu,Yuehua Qiao,Xuanyi Li

Objective: To investigate the effects of Zexie decoction on vestibular and auditory function in guinea pigs with endolymphatic hydrolysis induced by desmopressin. Methods: Sixty guinea pigs were randomly and evenly divided into four groups, each group has 15 guinea pigs: normal control group, DDAVP group, DDAVP modeling combined with Zexie Decoction group, and DDAVP combined with Double Zexie group. At 7 and 14 days, bone-conducted cervical vestibular evoked myogenic potential tests, auditory brainstem responses, and distortion-product otoacoustic emissions were conducted on each group of guinea pigs to evaluate their vestibular and auditory function quantitatively. After functional testing, the outer hair cells were observed by scanning electron microscope. On day 14, one guinea pig was randomly selected from both the normal control group and DDAVP group to verify the successful establishment of the model using gadolinium-enhanced magnetic resonance imaging of the inner ear. Results: We conducted BC-cVEMP, ABR, and DPOAE tests on guinea pigs, and the results showed that DDAVP did affect vestibular function and hearing in guinea pigs. Analyses were performed from those results that were statistically significant, Zexie Decoction improved DDAVP-induced vestibular dysfunction and hearing loss dose-dependently, though complete reversal was not achieved. About scanning electron microscopy, outer hair cells of the DDAVP group showed significant loss and cilia lodging, however, treatment with Zexie decoction can alleviate the loss of outer hair cells and the lodging of cilia. When the outer hair cells were exposed to DDAVP for a long time, the improvement effect of Zexie decoction was not as obvious as before. Conclusion: The extent of improvement correlates with the concentration and dosage of Zexie Decoction. Even at double the dosage, Zexie Decoction only partially mitigates the decline in vestibular and auditory function induced by DDAVP, falling short of complete reversal.

LC-MS-based metabolomics approach to investigate the taste compounds formation of Jinhua ham prepared using Taihu pigs

Li Huanhuan,Zhao Ke,Chen Lihong,Huang Qicheng,Ma Xiaozhong,Ge Shengyuan,Su Mingwei,Tang Honggang

In the present study, we evaluated the physicochemical properties, free amino acids concentration, metabolomics, and the relationship between main metabolites and volatile compounds of Jinhua ham prepared using Taihu black pigs during a ripening period of 210 days to elucidate the distinguishing features and formation process of the quality of this ham. During the eight timepoints, the color parameters redness and yellowness exhibited significant changes. Water activity gradually decreased. The tastes of saltiness, sourness and umami were main taste characteristics of Jinhua ham. Carnosine and glutamic acid were the main umami taste compounds and their contents reached to 1785 ± 92.2 and 699 ± 65.1 mg/100 g respectively. Metabolomics-based analysis showed that the dominant biochemical reaction was the metabolism of carboxylic acids and their derivatives. Total 153 significantly differential metabolites were identified. The main metabolic pathways were different in the three different periods respectively. Lipid metabolism was the predominant pathway in low–temperature stage, while amino acid formation and protein metabolism were the predominant pathways in the moderate–temperature and high–temperature stage. The dominant flavor compounds were closely related to the key differential metabolites. Our study findings revealed the quality, significantly differential metabolites, and formation process of Jinhua ham prepared using Taihu black pigs.

Effect of Epigallocatechin-3-Gallate on Growth Performance and Intestinal Epithelial Function in Weaned Pigs Exposed to Oxidative Stress

Qiming Duan,Xiang Li,Kunhong Xie,Aibing Yu,Yuheng Luo,Ping Zheng,Xiangbing Mao,Hui Yan,Jun He

Oxidative stress is a major challenge in weaned piglet production because it impairs intestinal health, nutrient utilization, and growth performance; however, the effects of dietary epigallocatechin-3-gallate (EGCG) on intestinal dysfunction under oxidative stress conditions have not been fully elucidated. This study examined whether dietary EGCG supplementation could improve intestinal function in weaned pigs subjected to Diquat-induced oxidative stress. Thirty-two weaned piglets were allocated to four treatments according to a 2 × 2 factorial arrangement involving dietary EGCG supplementation (0 or 50 mg/kg) and exposure to saline or Diquat (50 mg/kg body weight). Diquat exposure impaired intestinal morphology and antioxidant status and altered the transcription of genes associated with epithelial barrier integrity and redox regulation. Dietary EGCG supplementation partially attenuated these alterations, as reflected by improvements in villus architecture, intestinal antioxidant indices, and the mRNA expression of barrier- and antioxidant-related genes. These findings indicate that dietary EGCG supplementation was associated with improved intestinal function in weaned pigs exposed to oxidative stress. These beneficial effects may be related, at least in part, to changes in redox status, inflammatory responses, epithelial barrier-related gene expression, and selected gut microbial populations. This study supports EGCG as a potential bioactive dietary factor for maintaining intestinal health under oxidative stress conditions.

Transplantation of Elderly Human Gut Microbiota Into Pigs Reprograms Intestinal Barrier Function, Plasma Metabolome, and Gut Mucosal Transcriptomic Landscape

Jiaxin Wang,Lingyu Li,Lihua Mei,Zhining Tang,Baorui Liao,Qianye Zhao,Yuzeng Wang,Qingyao Fu,Liu Ren,Zhian Zhai,Yifei Xu,Aoyu Yang,Shuai Duan,Zhengyuan Zhai,Yanling Hao,Yalin Zhou,Yajun Xu,Ying Yang,Zhenlong Wu,Yun Ji

The gut microbiota plays a pivotal role in maintaining intestinal homeostasis and regulating host metabolism, yet its composition and function undergo substantial alterations with aging. However, the relationship between age-associated microbial changes and host intestinal physiology remains not fully elucidated. Here, we employed Bama miniature pigs, a model with close gastrointestinal similarity to humans, to investigate the impact of fecal microbiota transplantation (FMT) from young or elderly human donors on gut structure, barrier integrity, plasma metabolites, and intestinal mucosal transcriptomics. FMT resulted in distinct gut microbial profiles, with elderly-donor FMT reducing ileal villus height and tight junction proteins (ZO-1, claudin-1, and occludin) across multiple intestinal segments. At the species level, Phocea massiliensis predominated in young-donor pigs, while Blautia obeum was enriched in elderly-donor pigs. Plasma metabolomic analysis revealed increased 3-methyloxindole, prostaglandin E3, and 2-hydroxybutanoic acid but reduced Tyr-Phe and 2-hydroxyoctadecanoic acid in the elderly group. Among B. obeum -associated metabolites, Tyr-Phe and 2-hydroxyoctadecanoic acid exhibited a positive correlation with certain intestinal tight junction protein levels, whereas prostaglandin E3 showed an inverse correlation. Further validation performed in IPEC-1 cells demonstrated that Tyr-Phe elevated transepithelial electrical resistance (TEER), while prostaglandin E3 lowered this parameter. Transcriptomic profiling identified seven hub genes ( MX2 , ISG15 , IFI6 , IFIT1 , OAS1 , DHX58 , ISG12(A) ) that were consistently downregulated in the ileal mucosa of pigs receiving elderly-donor microbiota. These findings link age-associated microbiota alterations to coordinated changes in intestinal architecture, barrier integrity, and host metabolic and transcriptional profiles, providing insights into microbial and metabolic features associated with age-related intestinal decline.

Polyethylene glycol improves in vitro digestibility and fermentability of nutrients for pigs by binding with tannins in sorghum grain

Yao Huang,Mingmei Shen,Weiyun Zhu,Long Pan

This experiment was conducted to determine the effects of polyethylene glycol (PEG) supplementation on the digestion and fermentation characteristics of sorghum nutrients in the in-vitro simulated porcine gastrointestinal tract. In Exp. 1, high-tannin sorghum grain without or with 50 mg/g PEG was digested by porcine pepsin and pancreatin to measure in-vitro digestibility of nutrients. In Exp. 2, the lyophilised porcine ileal digesta from barrows fed the high-tannin sorghum grain without or with 50 mg/g PEG and the undigested residues from Exp. 1 were, individually, incubated with fresh pig caecal digesta as inoculums for 48 h to simulate the porcine hindgut fermentation. PEG reduced the detectable condensed tannin content by 32% (p < .05) and increased in-vitro digestibility of nutrients especially for crude protein(p < .05). The in-vitro fermentability of nutrients and microbial metabolites including the accumulative gas production excluding the first 12 h, total short-chain fatty acid and microbial protein content in the fermented solutions were improved (p < .05). The relative abundance of Lachnospiraceae NK4A136 was both increased by PEG supplementation regardless of unhydrolyzed residues (p < .05) or ileal digesta (p = .06) as fermentation substrates. In conclusion, PEG supplementation not only directly increased the chemical enzymatic digestion of nutrients in the simulated anterior intestine, but also directly promoted the microbial fermentation including microbial diversities and metabolites in the simulated posterior intestine of porcine. It implies that the increased abundances of Lachnospiraceae by PEG in the hindgut may improve the fermentation capacity of microbiota and thus increase the nutrient digestion in the hindgut.

PacBio sequencing combined GC–IMS approach to investigate the flavor formation of Jinhua ham prepared using Taihu black pigs

Huanhuan Li,Ke Zhao,Jin Zhang,Lihong Chen,Qicheng Huang,Xiaozhong Ma,Shengyuan Ge,Honggang Tang

In the present study, we evaluated the physicochemical and microbial properties, microbial diversity, and volatile organic compounds (VOCs) of Jinhua ham prepared using Taihu black pigs during a ripening period of 210 days. During the eight timepoints, the protein oxidation, proteolysis indexes, and TBARS values of Jinhua ham had different change trends. The growth of total bacteria and total Staphylococcus showed a decline, while the growth trend of total fungi was parabolic. Staphylococcus equorum was the dominant bacterial species, and the relative abundance were 19.3% (day 180)-76.7% (day 120). Debaryomyces hansenii and Eurotium sp. were the dominant fungus species, with opposite change trends. GC–IMS analysis detected 84 volatile compounds, with alcohols and aldehydes being the dominant volatile compounds in the late ripening period. The correlation analysis between microbial diversity and VOCs showed that Staphylococcus equorum was significantly negatively correlated with eight VOCs, while Debaryomyces hansenii and Eurotium sp. were significantly correlated with 31 and 29 VOCs respectively. Our findings reveal the quality, microbial diversity at species level, volatile compound characteristics, and relationship between microbial communities and flavor formation of Jinhua ham prepared using Taihu black pigs.

Different copper sources and levels affect growth performance, copper content, carcass characteristics, intestinal microorganism and metabolism of finishing pigs

Yang Wen,Runxian Li,Xiangshu Piao,Gang Lin,Pingli He

Copper (Cu) is an essential trace element in the production of swine. This study was conducted to investigate the effect of 3 different sources of Cu on growth performance, Cu metabolism, and intestinal microorganisms of finishing pigs, so as to estimate the bioavailability of the 3 sources for pigs. A total of 42 male finishing pigs (88.74 ± 5.74 kg) were randomly allocated to 7 treatments. The factors were 3 sources (CuSO 4 , Cu-glycine, Cu-proteinate) and 2 levels (5 and 20 mg/kg) of Cu, plus one negative control treatment (0 mg/kg added Cu level) for the entire 28-d experiment. The average daily gain (ADG) and feed to gain ratio (F:G) both increased when Cu was added. The Cu level in liver, bile, kidney, serum, lung, urine and feces rose ( P  < 0.001) with increasing dietary Cu level regardless of the source. Meanwhile, pigs receiving organic Cu (glycinate or proteinate) retained more Cu and excreted less Cu than those receiving inorganic Cu (CuSO 4 ), which showed that organic forms were more bioavailable. At the transcriptional level, changes in the level and source of dietary Cu resulted in modulation of transporters. In the jejunal mucosa, import transporter high affinity copper uptake protein 1 ( CTR1 ) and export transporter ATPase copper transporting alpha ( ATP7A ) in supplemental Cu treatments were down-regulated compared to the control. Also, peptide transporter 1 ( PepT1 ) and lanine-serine-cysteine transporter, type-2 ( ASCT2 ) were significantly ( P  < 0.01) up-regulated in 20 mg/kg Cu-proteinate and Cu-glycinate treatments, respectively. Microbial diversity was lowest in the 20 mg/kg CuSO 4 treatment, and the ratio of Firmicutes to Bacteroidetes was higher in added Cu treatments, especially Cu-glycinate treatment. These results indicate that uptake of different Cu forms is facilitated by different transporters and transport mechanisms, and compared with inorganic Cu, organic Cu provides benefits to intestinal microflora and reduces Cu excretion.

Ultra-fast self-gelling self-expanding self-propelling high-adhesion procoagulant hemostatic powder for non-compressible hemorrhage hemostasis in pigs

Huang Shengfei,Li Meng,Xu Huiru,Shu Tianyu,Jia Honglin,Li Zhenlong,Yang Yutong,Zhao Xin,Guo Baolin

Non-compressible hemorrhage remains the chief cause of battlefield mortality and civilian trauma death. Here, we propose a hemostatic strategy based on ultrafast self-gelling self-expanding powder of polyacrylic acid (PAA), polyethyleneimine (PEI) and foaming agent, which achieves non-compressible hemorrhage hemostasis through the multiple synergistic effects of expansion plugging, self-gelling adhesion and activating the coagulation factors. The hemostatic powder integrates rapid physical crosslinking with spontaneous gas foaming, exhibits a fast gelation rate, high expansion ratio, strong tissue adhesion, and activation of red blood cells, platelets, and fibrin. The optimized formulation (PP/PT5-TXA30) achieves superior hemostatic performance compared to commercial powders in rat liver volumetric defect, femoral artery and vein transection, as well as complete transection of the subclavian artery and vein in rabbits. Notably, in a lethal porcine model of complete subclavian artery and vein transection, PP/PT5-TXA30 achieves superior performance of non-compressible hemorrhage compared to gauze and XStat™. Additionally, PP/PT5-TXA30 accelerates full-thickness skin wound healing. This work demonstrates a strategy based on ultra-fast self-gelling and self-expanding mechanisms for developing hemostatic materials for non-compressible hemorrhage.

Vitamin A with L-ascorbic acid sodium salt improves the growth performance, immune function and antioxidant capacity of weaned pigs

H.B. Zhou,X.Y. Huang,Z. Bi,Y.H. Hu,F.Q. Wang,X.X. Wang,Y.Z. Wang,Z.Q. Lu

Vitamin A is easily degraded by environmental factors. Therefore, it is very important to add antioxidants during Vitamin A production. In the past, ethoxyquin ( EQ ) was widely used, but recent studies have found that it has potential toxicity. Therefore, in this study, we evaluated the antioxidant activities of 4 antioxidants in vitro : EQ, butylated hydroxytoluene, α-tocopherol and L-ascorbic acid sodium salt (Vitamin C sodium). In vitro experiments showed that Vitamin C sodium had better antioxidant capacity. Then, we explored the effects of different antioxidant types of Vitamin A on the growth performance, immune function and antioxidant capacity of weaned pigs. In total, 288 weaned piglets with an initial mean BW of 8.34 ± 0.02 kg at 30 days old were randomly divided into three groups with four replicates and 24 piglets per replicate for 35 days of feeding. The experimental diets were as follows: i) basal diet without external Vitamin A ( NC ); ii) basal diet supplemented with 12   000 IU/kg EQ Vitamin A and iii) basal diet supplemented with 12   000 IU/kg Vitamin C sodium Vitamin A. On day 36, two pigs from each replicate were selected to collect serum samples. The in vivo results showed that pigs in the EQ Vitamin A and Vitamin C sodium Vitamin A groups had significantly higher final weight and average daily gain ( P  < 0.05). During the trial, the levels of IgG and glutathione peroxidase in the EQ Vitamin A and Vitamin C sodium Vitamin A groups were significantly higher than those in the NC group ( P  < 0.05), and the malondialdehyde content was significantly lower ( P  < 0.05). On the 36th day, the levels of IgA and total antioxidant capacity in the Vitamin C sodium Vitamin A group were significantly higher than those in the EQ Vitamin A and NC ( P  < 0.05) groups. Thus, Vitamin C sodium Vitamin A can significantly improve the growth performance, antioxidant capacity and immune function of weaned pigs. Meanwhile, Vitamin C sodium may replace EQ as an antioxidant additive for Vitamin A.

Combination and efficiency: preparation of dissolving microneedles array loaded with two active ingredients and its anti-pigmentation effects on guinea pigs

Yu-shuai Wang,Wei-han Yang,Wei Gao,Lu Zhang,Fang Wei,Hang Liu,Shu-ya Wang,Ying-ying Li,Wei-man Zhao,Tao Ma,Qingqing Wang

Hyperpigmentation is a common skin disorder caused by excessive melanogenesis and uneven dispersion of melanin in the skin. To combine multiple active agents with an efficient transdermal drug delivery system is an effective strategy to combat UV induced skin pigmentation. In this work, Arbutin (Arb) and Vitamin C (Vc) mixed in 1:1 were found to have the greatest inhibition effects on melanogenesis and tyrosinase activity in B16 murine melanoma cells. And hyaluronic acid (HA) based dissolving microneedles array (DMNA) was employed to overcome the skin barriers for improved topical drug delivery, which exhibited the most desirable features, including morphology, mechanical properties, dissolving ability, and the highest drug loading. Furthermore, DMNA could greatly increase the stability of Vc during storage without adding any antioxidant which is an important issue for Vc administration. Pharmacodynamics study showed that DMNA loaded with Arb and Vc could synergistically suppress UVB-induced hyperpigmentation in guinea pig skin. This work provides a promising treatment strategy and solution for skin pigmentation and other skin problems.
