A novel ion-imprinted membrane induced by amphiphilic block copolymer for selective separation of Pt(IV) from aqueous solutions

Jianxian Zeng,Chaoqiang Lv,Guoqing Liu,Zhe Zhang,Zhihui Dong,JinYan Liu,Ying Wang

A novel ion-imprinted blend membrane was fabricated to separate selectively platinum(IV) from aqueous media. An amphiphilic copolymer poly(methyl methacrylate)-b-poly(4-vinylpyridine) (PMMA- b -P4VP) was firstly synthesized via the reversible addition fragmentation chain transfer polymerization , and characterized by 1 H NMR, FT-IR and GPC. Subsequently, poly(vinylidene fluoride) (PVDF) blend membranes were prepared using PMMA- b -P4VP as a blending additive. Effects of the following factors on performance of blend membranes were systematically examined: P4VP content of PMMA- b -P4VP, solvent type, and PMMA- b -P4VP/PVDF ratio. Further, platinum(IV) ion-imprinted membrane (Pt(IV)-IIM) was fabricated by blending polymer-Pt(IV) complexes with PVDF. SEM, FTIR-ATR, contact angle, water flux and adsorption capacity were employed to characterize all membranes. Results indicated that the membrane, which was prepared using the copolymer with high P4VP content (polymerization time 10 h) as a functional polymer , N,N-dimethylacetamide as a solvent and PMMA- b -P4VP/PVDF ratio of 30 wt%, exhibited an intriguing morphology and much higher performance. Pt(IV)-IIM showed higher adsorption and selectivity for Pt(IV) (selectivity coefficients of 27.66 ± 1.38 and 77.16 ± 3.86 for Pt(IV)/Cu(II) and Pt(IV)/Ni(II), respectively) compared with a non-imprinted membrane. During membrane filtration, Pt(IV)-IIM was effective and selective for separation of Pt(IV) in the presence of Cu(II) and Ni(II) ions. Also, Pt(IV)-IIM was chemical stable, and had a good regeneration performance.

A pH-responsive liposomal nanoplatform for co-delivery of a Pt(IV) prodrug and cinnamaldehyde for effective tumor therapy

Tang Ting,Gong Yufang,Gao Yuan,Pang Xinlong,Liu Shuangqing,Xia Yulan,Liu Dongsheng,Zhu Lin,Fan Qing,Sun Xiao

Introduction: The tumor microenvironment (TME) is mainly characterized by abnormally elevated intracellular redox levels and excessive oxidative stress. However, the balance of the TME is also very fragile and susceptible to be disturbed by external factors. Therefore, several researchers are now focusing on intervening in redox processes as a therapeutic strategy to treat tumors. Here, we have developed a liposomal drug delivery platform that can load a Pt(IV) prodrug (DSCP) and cinnamaldehyde (CA) into a pH-responsive liposome to enrich more drugs in the tumor region for better therapeutic efficacy through enhanced permeability and retention effect. Methods: Using the glutathione-depleting properties of DSCP together with the ROS-generating properties of cisplatin and CA, we synergistically altered ROS levels in the tumor microenvironment to damage tumor cells and achieve anti-tumor effects in vitro. Results: A liposome loaded with DSCP and CA was successfully established, and this liposome effectively increased the level of ROS in the tumor microenvironment and achieved effective killing of tumor cells in vitro. Conclusion: In this study, novel liposomal nanodrugs loaded with DSCP and CA provided a synergistic strategy between conventional chemotherapy and disruption of TME redox homeostasis, leading to a significant increase in antitumor effects in vitro.

Enhanced separation of Pd(II) and Pt(IV) from hydrochloric acid aqueous solution using 2-((2-methoxyethyl)thio)-1H-benzimidazole

Feng Shao-Ping,Huang Kun

Abstract Separation of palladium (Pd) and platinum (Pt) by solvent extraction is difficult because of their extremely similar physicochemical properties. Development of new extractants with a high extraction efficiency and excellent separation selectivity is the primary focus for enhancing the separation of Pd and Pt by solvent extraction. In this study, a new extractant, 2-((2-methoxyethyl)thio)-1H-benzimidazole (MOBI), which contains N, S, and O atoms, was synthesized and used to separate Pd(II) and Pt(IV) from a hydrochloric acid aqueous solution. The experimental results revealed that Pd(II) and Pt(IV) were effectively separated under the optimal conditions: MOBI concentration of 0.005 mol·L −1 , HCl concentration of 0.2 mol·L −1 , organic/aqueous (O/A) phase ratio of 1.0, and contact time of 15 min. The separation coefficient between Pd(II) and Pt(IV) was 1246.40, which indicates that MOBI has a higher selectivity for Pd(II) than Pt(IV). In addition, the mechanism of coordination between Pd(II) and MOBI was demonstrated through the slope method, Fourier transform infrared spectroscopy (FTIR), hydrogen-1 nuclear magnetic resonance spectroscopy ( 1 H NMR), and carbon-13 nuclear magnetic resonance spectroscopy ( 13 C NMR). The results revealed that MOBI could coordinate with Pd(II) via the N atom in the benzimidazole ring of MOBI molecules to form [Pd(MOBI) 2 Cl 2 ] (O) . Graphic abstract

Multitargeting Pt(IV) Anticancer Prodrugs Bearing Mono- and Bis-Probenecid Ligands in Axial Positions: Synthesis and Evaluation of Biological Activity

Panxing Qiu,Yu Zhang,Yang Dou,Zhijin Cheng,Xiaoqin Wu,Silong Zhang,Fuyi Wang,Kui Wu

Background : To battle the side effects of anticancer Pt(II) drug cisplatin, the development of photoactivatable and/or intracellular reduction-activatable Pt(IV) prodrugs has become a promising strategy. Methods: Herein, two novel Pt(IV) prodrugs, namely, cis , cis , trans -[Pt IV (NH 3 ) 2 (Cl) 2 (OH)(probenecid)]) (SPP) and cis , cis , trans -[Pt IV (NH 3 ) 2 (Cl) 2 (probenecid) 2 ] (DPP) bearing mono- and di-probenecid at the axial positions of oxoplatin have been synthesized via covalently linking of carboxylate group in probenecid, which is a well-established clinic drug by inhibiting organic anion transporter 1 (OAT1) to reduce cisplatin-induced nephrotoxicity, with the axial hydroxyl group(s) in oxoplatin. The promising cytotoxicity of SPP and DPP against MCF-7, T47D breast cancer cells and the MDA-MB-231 triple-negative breast cancer cells was evaluated, and the mechanism of action of the two Pt(IV) prodrugs was investigated by apoptosis assay and Western blot assay. Results : SPP exhibits a comparable cytotoxicity to cisplatin against MCF-7 and T47D breast cancer cells, while it shows 2.1-fold higher cytotoxicity than cisplatin against MDA-MB-231 cells. DPP was shown to be more cytotoxic than SPP, and exhibits 8.7-, 7.5-, and 2.3-fold higher cytotoxicity than cisplatin against MCF-7, T47D, and MDA-MB-231 cells, respectively. Apoptosis assays revealed a similar early-apoptotic cell death mechanism to cisplatin for both SPP and DPP. The enhanced cellular and nuclear uptake of DPP compared to cisplatin contributes to its promising anticancer efficacy. DPP can bind to OAT1 in cancer cells, which may synergistically enhance the cytotoxicity of the Pt(IV) anticancer prodrugs. Conclusions : The direct conjugation of probenecid to the axial positions of oxoplatin confers the resulting Pt(IV) prodrugs a multitargeting property, significantly promoting the cytotoxicity of the resulting Pt(IV) complexes. This finding provides a practical strategy for drug design and cancer treatment based on platinum complexes.

Precision mitochondrial reprogramming via a ROS-amplifying Pt(IV) nanoplatform potentiates tri-modal therapy to overcome Pt resistance in HCC

Ruoning Qian,Junqing Hou,Hongjun Guo,Qiang Zhang,Jiamin Lin,Yitian Zhou,Zhiqiang Bi,Jiajia Shen,Xi Yang,Shanshan Wang,Zhengguang Zhang,Ruogu Qi

Mitochondrial dysfunction plays a critical role in hepatocellular carcinoma (HCC) progression. This study develops a glutathione-responsive Pt(IV)–artesunate (ART) prodrug (Pt–ART) that co-releases cisplatin and ART in the tumor microenvironment, synergizing DNA damage and iron-dependent ROS generation for combined chemotherapy and chemodynamic therapy. The prodrug is encapsulated into a ROS-responsive, fluorinated polyethyleneimine-chlorin e6-based polycationic nanoparticle, coated with polyethylene glycol-chondroitin sulfate for enhanced tumor targeting via the EPR effect, CD44 receptor-mediated uptake, and mitochondrial accumulation. This nanoplatform effectively inhibits Drp1 expression, induces mitochondrial membrane depolarization, and demonstrates potent triple-modality therapy combining chemotherapy, chemodynamic therapy, and photodynamic therapy. Our work provides an innovative strategy integrating herbal medicine with nanotechnology to overcome limitations of conventional Pt-based therapies.

Iodine Conjugated Pt(IV) Nanoparticles for Precise Chemotherapy with Iodine–Pt Guided Computed Tomography Imaging and Biotin-Mediated Tumor-Targeting

Jie Yu,Xidong He,Qingfei Zhang,Dongfang Zhou,Zigui Wang,Yubin Huang

Theranostics of platinum (Pt)-based chemotherapy are able to self-track the biodistribution and pharmacokinetics while performing therapeutic effects. Pt-based CT imaging is expected to visualize and monitor the tumor throughout the entire tumor inhibition stage. However, a sufficient Pt concentration is necessary for CT imaging, which may bring about severe nephrotoxicity. A Bio-Pt-I compound is designed and synthesized by conjugation of iodine and biotin to the structure of Pt and further self-assembles into nanoparticles. The introduction of iodine not only enhances the CT imaging signal with a much lower dose of Pt but also overcomes the resistance of tumor cells to Pt-containing nanomedicine by inhibiting the expression of Bcl-2. Furthermore, biotin-mediated tumor targeting increases drug accumulation in tumors. This work combines CT imaging based self-track with efficient cisplatin-resistance reversion ability, which may promote the clinical transformation of Pt-containing nanomedicine.

Construction of PAMAM-based Nanocomplex Conjugated with Pt(IV)-complex and Lauric Acid Exerting Both Anti-tumor and Antibacterial Effects

Xin You-Tao,Wang Lu-Yao,Chang Huan-Huan,Ma Fei-He,Sun Ming-Lei,Chen Li,Gao Hui

Oxaliplatin is the common chemotherapy drug for the treatment of colorectal cancer (CRC), however, the harmful Fusobacterium nucleatum ( Fn ) that existed in the CRC tumor tissues leads to extreme drug resistance to oxaliplatin, which reduces the anticancer efficiency. Here we fabricated a new dendrimer-based nanocomplex with anti-tumor and antibacterial dual-functions through conjugating Pt(IV)-complex and lauric acid (LA) to the dendritic carrier PAMAM (G=3). The synthesized PAMAM-Pt-LA nanoparticle was further coated with hyaluronic acid (HA) on its surface through electrostatically interaction in order to enhance biocompatibility and tumor targeting. The biocompatibility, anti-tumor activity and antibacterial activity of PAMAM-Pt-LA@HA nanocomplex were evaluated in vitro. The results showed that the PAMAM-Pt-LA@HA nanocomplex ehxibited a good biocompatibility, high anti-colorectal cancer activity and high anti- Fn activity. This dual-functional nanocomplex provides a new method to develop anticancer nanomedicines that will be more therapeutically efficacious and less toxic than oxaliplatin for the treatment of CRC.

Extraction behaviour and mechanism of Pt(IV) and Pd(II) by liquid–liquid extraction with an ionic liquid [HBBIm]Br

Wenhui Liu,Qi Wang,Yan Zheng,Shubin Wang,Yan Yan,Yanzhao Yang

In this study, a method of one-step separation and recycling of high purity Pd(II) and Pt(IV) using an ionic liquid, 1-butyl-3-benzimidazolium bromate ([HBBIm]Br), was investigated. The effects of [HBBIm]Br concentration, initial metal concentration, and loading capacity of [HBBIm]Br were examined in detail. It was observed that [HBBIm]Br was a very effective extractant for selectively extracting Pd(II) and precipitating Pt(IV). Through selectively extracting Pd(II) and precipitating Pt(IV), each metal with high purity was separately obtained from mixed Pd(II) and Pt(IV) multi-metal solution. The method of one-step separation of Pd(II) and Pt(IV) is simple and convenient. The anion exchange mechanism between [HBBIm]Br and Pt(IV) was proven through Job's method and FTIR and 1H NMR spectroscopies. The coordination mechanism between [HBBIm]Br and Pd(II) was demonstrated via single X-ray diffraction and was found to be robust and distinct, as supported by the ab initio quantum-chemical studies. The crystals of the [PdBr2 ·2BBIm] complex were formed first. Moreover, the influence of the concentrations of hydrochloric acid, sodium chloride, and sodium nitrate on the precipitation of Pt(IV) and extraction of Pd(II) was studied herein. It was found that only the concentration of H+ could inhibit the separation of Pt(IV) because H+ could attract the anion PtCl62−; thus, the exchange (anion exchange mechanism) between the anions PtCl62− and Br− was prevented. However, both the concentration of H+ and Cl− can obviously inhibit the extraction of Pd(II) because H+ and Cl− are the reaction products and increasing their concentration can inhibit the progress of the reaction (coordination mechanism).

Separation of Pd(II) and Pt(IV) in concentrated hydrochloric acid using thin-layer oil membrane and comparison with conventional extraction

Dechao Xiao,Haitao Zhou,Kaihui Cui,Kun Huang

This study proposes a new thin-layer oil membrane (TOM) extraction strategy for enhanced mutual separation of Pd(II) and Pt(IV) chlorocomplex anions in concentrated aqueous hydrochloric acid solutions using di- n -butyl sulfoxide (DBSO) as an extractant. It was discovered that Pd(II) could be preferentially extracted even in concentrated hydrochloric acid solutions. The separation coefficient between Pd(II) and Pt(IV) considerably increased than that between them during conventional extraction using a separating funnel. A new extractor was designed to perform TOM extraction by spreading a thin layer of organic extractant liquid membrane on the surface of aqueous feed solutions under laminar flow . A simplified TOM extraction model and controlled experiments were used to investigate the effect of H + and Cl − ion concentrations on the mutual separation between Pd(II) and Pt(IV). Molecular dynamics (MD) simulations were used to understand the microscopic separation mechanisms at the molecular level. Experimental results revealed that the competitive distribution of H + and Cl − ions in the boundary layer along the normal vertical direction of the organic-aqueous interface are crucial in enhancing the separation between Pd(II) and Pt(IV). The competitive adsorption and enrichment of Cl − ions at the interface prevent DBSO molecules from being protonated by H + ions, which is beneficial for the substituted coordination of DBSO molecules with PdCl 4 2− . However, the ionic association of protonated DBSO molecules with PtCl 6 2− remains dominant at the interface. As a result, even in concentrated hydrochloric acid solutions, the difference in mass transfer between PdCl 4 2− and PtCl 6 2− toward the interface was intensified.

A separation strategy of Au(III), Pd(II) and Pt(IV) based on hydrophobic deep eutectic solvent from hydrochloric acid media

Ronghao Liu,Jinjie Hao,Yangyang Wang,Yuanyuan Meng,Yanzhao Yang

A hydrophobic deep eutectic solvent (HDES) based on lidocaine and thymol was examined for selective extraction and separation of Au(III), Pd(II) and Pt(IV) in the acidic leaching solution of secondary resources. At first, the properties of HDES were characterized by 1 H NMR, DSC, TGA, RDF and ESP to prove its suitability for extraction process. In addition, extraction behaviors, maximum extraction capacities and competitive extraction experiment were investigated to explore the extraction ability of HDES. The results show that the maximum extraction capacity of Au(III), Pd(II) and Pt(IV) reached 761.2 g/L, 98.3 g/L and 299.3 g/L, respectively. Meanwhile, their extraction efficiency was affected by the changing acidity of aqueous. And then, ion-association mechanism in the extraction process was proposed through the analysis of FT-IR. Finally, a separation process was proposed for the recovery of Au(III), Pd(II) and Pt(IV) based on acidity control and KSCN, which achieved recovery efficiency of 97.8 % Au(III), 99.9 % Pd(II) and 89.5 % Pt(IV). After collecting and regenerating the used HDES, their extraction performance of Au(III), Pd(II) and Pt(IV) were still maintained.

Metalloselective adsorption of Au(III), Pd(II) and Pt(IV) from acidic solutions by a thiourea-modified Cr-based metal-organic framework

He Baili,Kou Jue,Sun Chunbao,Li Yue,Li Xiaolin,Temirov Uktam,Chang Ziyong

The development of effective adsorbents is crucial for the sustainable recovery of noble metals from secondary resources. In this study, MIL-101(Cr)-Tu, a Cr-based metal–organic framework (MOF) modified with thiourea, was successfully prepared by double-phase encapsulation followed by post-synthetic modification. At 298 K, the largest adsorption capacities of Au(III), Pd(II), and Pt(IV) by MIL-101(Cr)-Tu were (1230.83 ± 8.72), (330.41 ± 7.29), and (315.58 ± 13.67) mg·g −1 , respectively. The uptakes of Au(III), Pd(II), and Pt(IV) increased with temperature. The adsorption data fit well to both the Langmuir and pseudo-second-order kinetic models, indicating that the adsorption process conformed to monolayer adsorption and chemisorption. Mechanistic analysis revealed that the amino and thiourea groups facilitated the adsorption of noble metals through coordination and electrostatic attraction. Notably, Au(III), Pd(II), and Pt(IV) interacted strongly with the thiourea groups owing to their “soft–soft” interactions. Moreover, Au(III) was reduced to Au(I) and Au(0), Pt(IV) was reduced to Pt(II), and the amino groups were oxidized to NO 2 . The adsorption of Pd(II) did not involve redox reactions. Additionally, MIL-101(Cr)-Tu exhibited strong selectivity for noble metals and excellent reusability, demonstrating its great potential for the extraction of noble metals from secondary resources.

Separation of Pt(IV), Pd(II), Ru(III), and Rh(III) from chloride medium using liquid–liquid extraction with mixed imidazolium-based ionic liquids

Yan Yan,Qi Wang,Zeyang Xiang,Yanzhao Yang

A reasonable project was presented for the extraction and separation of Pt(IV), Pd(II), Ru(III), and Rh(III) from the mixed imidazolium-based ionic liquids (ILs) [C6mim]Cl, [C6mim][NTf2], and [C6mim][DDTC]. Pt(IV) was first separated from other three platinum group metals with a mixture of [C6mim]Cl and [C6mim][NTf2]. Then, separation of Pd(II) and Ru(III) from Rh(III) was realized by changing the concentration of [C6mim][DDTC]. Back-extraction of Pt(IV), Pd(II), and Ru(III) from loaded hydrophobic phase and the recovery of Rh(III) in aqueous phase were also investigated. Mechanisms between Pd(II)/Ru(III) and [C6mim][DDTC] were further explored using ultraviolet spectrophotometry (UV-Vis), infrared spectrophotometry (FTIR), the method of continuous variations, ab initio quantum chemical studies, and X-ray crystallographic analysis. The results afford more directions for extracting and separating metals by introducing devisable functionalized group into ILs.
