Multifunctional Injectable Microspheres Containing “Naturally-Derived” Photothermal Transducer for Synergistic Physical and Chemical Treating of Acute Osteomyelitis through Sequential Immunomodulation

Yang Shen,Yaowen Xu,Ziqian Yu,Guo Chen,Bin Chen,Liqiong Liao

Osteomyelitis induced by Staphylococcus aureus (S. aureus) is a persistent and deep-seated infection that affects bone tissue. The main challenges in treating osteomyelitis include antibiotic resistance, systemic toxicity, and the need for multiple recurrent surgeries. An ideal therapeutic strategy involves the development of materials that combine physical, chemical, and immunomodulatory synergistic effects. In this work, we prepared injectable microspheres consisting of an interpenetrating network of ionic-cross-linked sodium alginate (SA) and genipin (Gp)-cross-linked gelatin (Gel) incorporated with tannic acid (TA) and copper ions (Cu2+). The Gp-cross-linked Gel acted as a “naturally−derived” photothermal therapy (PTT) agent. The results showed that the microspheres exhibited efficient and rapid bactericidal effects against both S. aureus and Escherichia coli (E. coli) under the irradiation of near-infrared light at 808 nm wavelength; moreover, the release of Cu2+ also induced sustained inhibitory effects against bacteria during the nonirradiation period. The in vitro cell culture results indicated that when combined with PTT, the microspheres could adaptively modulate macrophage M1 and M2 phenotypes in sequence. Additionally, these microspheres were found to enhance the osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs). In vivo studies conducted in a rat femur osteomyelitis model with bone defects showed that under multiple laser irradiation the microspheres effectively controlled bacterial infection, improved the pathological immune microenvironment, and significantly enhanced the repair and regeneration of bone tissues in the affected area.

Rhamnolipid-Coated Iron Oxide Nanoparticles as a Novel Multitarget Candidate against Major Foodborne E. coli Serotypes and Methicillin-Resistant S. aureus

Mohamed Sharaf,Alaa H. Sewid,H. I. Hamouda,Mohamed G. Elharrif,Azza S. El-Demerdash,Afaf Alharthi,Nada Hashim,Anas Abdullah Hamad,Samy Selim,Dalal Hussien M. Alkhalifah,Wael N. Hozzein,Mohnad Abdalla,Taisir Saber

Surface-growing antibiotic-resistant pathogenic bacteria such as Escherichia coli and Staphylococcus aureus are emerging as a global health challenge due to dilemmas in clinical treatment. Furthermore, their pathogenesis, including increasingly serious antimicrobial resistance and biofilm formation, makes them challenging to treat by conventional therapy. Therefore, the development of novel antivirulence strategies will undoubtedly provide a path forward in combatting these resistant bacterial infections. In this regard, we developed novel biosurfactant-coated nanoparticles to combine the antiadhesive/antibiofilm properties of rhamnolipid (RHL)-coated Fe3O4 nanoparticles (NPs) with each of the p-coumaric acid (p-CoA) and gallic acid (GA) antimicrobial drugs by using the most available polymer common coatings (PVA) to expand the range of effective antibacterial drugs, as well as a mechanism for their synergistic effect via a simple method of preparation. Mechanistically, the average size of bare Fe3O4 NPs was ~15 nm, while RHL-coated Fe3O4@PVA@p-CoA/GA was about ~254 nm, with a drop in zeta potential from −18.7 mV to −34.3 mV, which helped increase stability. Our data show that RHL-Fe3O4@PVA@p-CoA/GA biosurfactant NPs can remarkably interfere with bacterial growth and significantly inhibited biofilm formation to more than 50% via downregulating IcaABCD and CsgBAC operons, which are responsible for slime layer formation and curli fimbriae production in S. aureus and E. coli, respectively. The novelty regarding the activity of RHL-Fe3O4@PVA@p-CoA/GA biosurfactant NPs reveals their potential effect as an alternative multitarget antivirulence candidate to minimize infection severity by inhibiting biofilm development. Therefore, they could be used in antibacterial coatings and wound dressings in the future.IMPORTANCE Antimicrobial resistance poses a great threat and challenge to humanity. Therefore, the search for alternative ways to target and eliminate microbes from plant, animal, and marine microorganisms is one of the world’s concerns today. Furthermore, the extraordinary capacity of S. aureus and E. coli to resist standard antibacterial drugs is the dilemma of all currently used remedies. Methicillin-resistant S. aureus (MRSA) and vancomycin-resistant S. aureus (VRSA) have become widespread, leading to no remedies being able to treat these threatening pathogens. The most widely recognized serotypes that cause severe foodborne illness are E. coli O157:H7, O26:H11, and O78:H10, and they display increasing antimicrobial resistance rates. Therefore, there is an urgent need for an effective therapy that has dual action to inhibit biofilm formation and decrease bacterial growth. In this study, the synthesized RHL-Fe3O4@PVA@p-CoA/GA biosurfactant NPs have interesting properties, making them excellent candidates for targeted drug delivery by inhibiting bacterial growth and downregulating biofilm-associated IcaABCD and CsgBAC gene loci.

Analysis of the Chemical Profiles and Anti-S. aureus Activities of Essential Oils Extracted from Different Parts of Three Oregano Cultivars

Yuanpeng Hao,Jingyi Li,Wenying Zhang,Meiyu Sun,Hui Li,Fei Xia,Hongxia Cui,Hongtong Bai,Lei Shi

The use of antibiotics in the food industry is highly regulated owing to the potential harmful effects of antibiotics on human health. Therefore, it is crucial to seek alternatives for ensuring food safety. Essential oils (EOs) extracted from plants of the genusOriganumexhibit a wide range of chemical and antibacterial activities. Species and tissue factors shape the production and accumulation processes of EOs inOriganumplants, thereby affecting their bactericidal activity. In this study, the morphologies and EO yields from the inflorescences, leaves, and stems of three oregano cultivars were evaluated. In addition, the chemical compositions and antibacterial abilities of oregano EOs (OEOs) were assessed. The results showed that OEOs from the different parts of the plant displayed only minor differences in chemical composition, whereas the yield of EOs varied considerably. Additionally, the chemical profiles of OEOs differed significantly among cultivars. The carvacrol content in the OEOs was closely related to its activity againstStaphylococcus aureus; the antibacterial properties of the OEOs were further verified using carvacrol. These findings suggested that OEOs possessing high antibacterial activity may have the potential to be developed as bactericides in the food industry. Keywords:oregano essential oil;chemical composition;carvacrol;antibacterial activity;food safety

Carbon dots derived from folic acid as an ultra-succinct smart antimicrobial nanosystem for selective killing of S. aureus and biofilm eradication

Meizhe Yu,Xiuzhi Guo,Haojie Lu,Peili Li,Ruobing Huang,Chunning Xu,Xuedong Gong,Yuhong Xiao,Xiaodong Xing

Developing efficient and safe approach to counteract the bacterial infections has become an urgent clinical challenge. In this work, we report folic acid derived carbon dots (FA-CDs) prepared by a one-step pyrolysis method. The FA-CDs can respond to the environmental stimuli of bacterial infections and acquire antimicrobial capacity “on demand” as a simple and effective antimicrobial nanosystem. The assays on antibacterial properties and mechanism show that FA-CDs are able to induce the gradual increase of intracellular reactive oxygen species (ROS) levels in bacteria under the simulated acidic environment and display pH-responsive biocidal activity. In a neutral condition, FA-CDs have no antibacterial activity, which will be conducive to mitigating the rise of bacterial drug resistance. The responsive antibacterial behavior can be attributed to that FA-CDs exhibit distinctive oxidase-like and peroxidase-like activity under acidic stimuli and inhibit the superoxide dismutase activity in bacteria cells, causing the accumulation of intracellular ROS. Meanwhile, FA-CDs can selectively kill Gram-positive bacterium S. aureus and remove mature biofilms. Besides, the in vitro and in vivo experiments demonstrate that FA-CDs possess excellent biocompatibility, and show remarkable therapeutic effect on the S. aureus -infected back wound models of mouse, which are crucial for clinical applications. Thus, this study highlights the potential application of FA-CDs as an ultra-succinct smart antimicrobial nanosystem for coping with infections caused by S. au reus in the clinical setting, and will bring about a new perspective on developing antimicrobial CDs.

Polyadenine-mediated Immobilization of Aptamers on a Gold Substrate for the Direct Detection of Bacterial Pathogens

Wang Wen-Wen,Han Xiao,Chu Li-Qiang

Nucleic acid aptamers have been widely used as synthetic probes for bioanalytical applications. Herein, we carried out a detailed study on the immobilization of a series of aptamers ranging from 37 to 88 bases, which are specific to either Escherichia coli ( E. coli ) or Staphylococcus aureus ( S. aureus ), on a planar gold substrate via a polyadenine-mediated immobilization method. The resultant surfaces were characterized by both surface plasmon resonance spectroscopy (SPR) and X-ray photoelectron spectroscopy. The results clearly show that the aptamer solution at a lower ionic strength gives rise to a higher lateral density of the aptamer when compared to that at a higher ionic strength. The SPR aptasensors are then employed for detecting their corresponding bacteria ( i.e., E. coli and S. aureus , respectively). The data indicate that the SPR aptasensor with a higher density of aptamer exhibits a better capture of target bacteria.

Highly Antibacterial Efficacy of a Cotton Fabric Treated with Piperazinyl Schiff Base

Wen Wen,Zhang Zaixing,Jing Lingxiao,Zhang Tonghua

Due to the structure of hierarchical aligned cellulose fibrils, cotton fabric used in clothing possesses excellent moisture and thermal managements. Such structure yet may retain metabolic excrements and sebum secretions discharged from the human skin, which reproduce microorganisms harmful for human health. However, incorporating the antimicrobial coating into the cotton fabric can sufficiently resist the microorganism growth. In this work, a water-soluble antibacterial coating named N-(4-(allyloxy) benzylidene)-2-(piperazin-1-yl) ethanamine (NABPE) was synthesized to produce a rechargeable and fast sterilization cotton fiber fabric (M-cotton/NABPE). M-cotton/NABPE exhibited a high effective antibacterial activity, and the inhibition ratios against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) were 94 % and 93 %, respectively. Chlorination was performed with sodium hypochlorite solution to form N-Cl bond on the M-cotton/NABPE fabrics, resulting in a high biocidal efficacy of up to 100 % via contact killing for a duration of 5 min. After 25 washing cycles, the antibacterial fabric still maintained an antibacterial rate of 91.95 % and 92.15 % against E. coli and S. aureus , respectively. Furthermore, the fabrics showed integrated properties of excellent UV stability, long-term stability, robust rechargeable biocidal activity (chlorine recharging >5000 ppm) and washing durability. This research provides fundamental insights into the synthesis of the NABPE and prolonged biocidal efficacy of the M-cotton/NABPE, and thereby pave a pathway to incorporate an economic and environmental-friendly antibacterial coating suitable for finishing cotton fabric.

Highly efficient g-C3N4/TiO2/kaolinite composite with novel three-dimensional structure and enhanced visible light responding ability towards ciprofloxacin and S. aureus

Chunquan Li,Zhiming Sun,Wanzhong Zhang,Caihong Yu,Shuilin Zheng

A novel 3D heterogeneous g-C 3 N 4 /TiO 2 /kaolinite composite with enhanced visible light activity was fabricated via a mild sol-gel method associated with chemical stripping and self-assembly. Compared with bare photocatalysts, the g-C 3 N 4 /TiO 2 /kaolinite 3D structure exhibits enhanced adsorption-photocatalytic degradation ability for the removal of ciprofloxacin (CIP) under visible-light irradiation, and also facilitate the recyclability of the photocatalyst as demonstrated from the reusability test. The apparent rate constant of the composite is up to around 5.35 times, 6.35 times and 4.49 times that of bare TiO 2 , g-C 3 N 4 and P25, respectively, and a possible degradation pathway was also proposed. On the other hand, the as-received composite also exhibited enriched disinfection ability towards S. aureus . It is indicated that the superoxide radical ( O 2 − ) is the main active species in the degradation process, and the superior photocatalytic performance of composite should be mainly attributed to both the improvement of light harvesting as well as the enhanced separation and transfer efficiency. It is expected that this novel ternary visible-light responding composite would be a promising candidate material for the organic pollutants degradation and bacteria inactivation.

Cationic Oligo(thiophene ethynylene) with Broad-Spectrum and High Antibacterial Efficiency under White Light and Specific Biocidal Activity against S. aureus in Dark

Qi Zhao,Junting Li,Xiaoqian Zhang,Zhengping Li,Yanli Tang

We designed and synthesized a novel oligo(thiophene ethynylene) (OTE) to investigate the antibacterial activities against Gram-positive (Staphylococcus aureus and Staphylococcus epidermidis) and Gram-negative (Ralstonia solanacearum and Escherichia coli) bacteria in vitro by photodynamic therapy (PDT). Notably, OTE presents broad-spectrum and greatly high antibacterial activities after white light irradiation at nanogram per milliliter concentrations. The half inhibitory concentrations (IC50) values obtained for S. aureus, S. epidermidis, E. coli, and R. solanacearum are 8, 13, 24, and 52 ng/mL after illumination for 30 min, respectively, which are lower than that of other PDT agents. Interestingly, OTE shows the specific and very strong dark killing capability against S. aureus at the concentration of 180 ng/mL for 30 min, which is the highest efficiency biocide against S. aureus without the need of irradiation to date. The antibacterial mechanism investigated demonstrated that reactive oxygen species or singlet-oxygen generated by OTE kills bacteria irreversibly upon white light irradiation, and OTE as a v-type oligomer exerts its toxicity directly on destroying bacterial cytoplasmic membrane in the dark. Importantly, the OTE shows no cell cytotoxicity and excellent biocompatibility. The results indicate that it is potential to provide versatile applications in the efficient control of pathogenic organisms and specific application for killing S. aureus.

Self-powered biosensing system driven by triboelectric nanogenerator for specific detection of Gram-positive bacteria

Congyu Wang,Peng Wang,Junhuan Chen,Liyang Zhu,Dun Zhang,Yi Wan,Shiyun Ai

Gram-positive bacterial concentration , such as Staphylococcus aureus (S. aureus) , is a crucial criterion for evaluating water quality and influencing human health, developing effective and specific detection of Gram-positive bacteria is important for prevention of their threats. In our proof-of-concept work, a self-powered biosensing system based on the vertical contact-separation triboelectric nanogenerator (TENG) was developed to specifically detect Gram-positive bacteria in solution. In the biosensing system, vancomycin was utilized to identify and capture Gram-positive bacterial cells via specific vancomycin-bacteria wall interactions, guanidine functionalized multi-walled carbon nanotubes (CNT-Arg) was adopted as signal amplification material due to its high electrical conductivity , and TENG was adopted as a stable voltage signal source. Thus, the self-powered biosensing system could specifically detect the Gram-positive bacteria in solution by determining the measured voltage variation of biosensor. It was proven that the self-powered biosensing system was applied to detect S. aureus (employed as a model) with a linear relationship, low limit of detection (2 × 10 3 CFU mL −1 ) and high selectivity. Moreover, a warning program of Labview was written in this work, and the warning program could convert the voltage signal into visual signal for convenient observation. This work provides a design concept for self-powered biosensing system with TENG, and by replacing the molecular probes, the system can facilitate the application of TENG in many relevant fields, such as environmental pollution, iatrogenic diseases and microbiological corrosion .

Multifunctional chitin-based barrier membrane with antibacterial and osteogenic activities for the treatment of periodontal disease

Tao Wu,Lin Huang,Jing Sun,Jiahui Sun,Qi Yan,Bo Duan,Lina Zhang,Bin Shi

The guided tissue regeneration technique is an effective approach to repair periodontal defect. However, collagen barrier membranes used clinically lose stability easily, leading to soft tissue invasion, surgical site infection, and failure of osteogenesis. An ideal barrier membrane should possess proper antibacterial, osteoconductive activities, and favorable biodegradation. In this study, zinc oxide nanoparticles were homogeneously incorporated into the chitin hydrogel (ChT-1%ZnO) through one-step dissolution and regeneration method from alkaline/urea solution the first time. The remaining weights of ChT-1%ZnO in 150 μg/mL lysozyme solution was 52% after 5 weeks soaking. ChT-1%ZnO showed statistical antibacterial activities for P. gingivalis and S. aureus at 6 h, 12 h, and 24 h. Moreover, ChT-1%ZnO exhibits osteogenesis promotion in vitro , and it was further evaluated with rat periodontal defect model in vivo . The cemento-enamel junction value in ChT-1%ZnO group is 1.608 mm, presenting a statistical difference compared with no-membrane (1.825 mm) and ChT group (1.685 mm) after 8 weeks postoperatively.

Gut microbial metabolite indole-3-propionic acid inhibits inflammation and restores blood-milk barrier in S. aureus- induced mastitis by targeting aryl hydrocarbon receptor

Lihua Zhao,Lei Jin,Yunhe Fu,Bin Yang

Background: Indole-3-propionic acid (IPA) is a tryptophan metabolite produced by intestinal bacteria, which has functions such as penetrating tissue barriers and reducing tissue inflammatory reactions. In the present study, the therapeutic effect of IPA on Staphylococcus aureus (S. aureus)-induced mastitis was investigated. Methods: A mouse model of mastitis was established using breast injection of S. aureus. Except for the control group, all other mice were given oral administration of IPA. Hematoxylin eosin (H&E) staining was used to detect pathological changes in mouse mammary tissue. ELISA was used to detect TNF-α and IL-1β levels. Western blot was used to detect protein expression. Results: As the results demonstrated, IPA treatment obviously attenuated S. aureus-induced mammary pathological injury, myeloperoxidase (MPO) activity, malondialdehyde (MDA) content, TNF-α, and IL-1β levels. Meanwhile, IPA treatment could restore blood-milk barrier, as confirmed by up-regulating the expression of ZO-1 and occludin. In vitro, IPA could inhibit TNF-α and IL-1β production and the activation of NF-κB and NLRP3 induced by S. aureus. Furthermore, IPA could increase the expression of aryl hydrocarbon receptor (AhR). In addition, the inhibition of IPA on S. aureus-induced inflammation was reversed by AhR inhibitor. Conclusion: In conclusion, the results suggested that IPA inhibited S. aureus-induced mastitis through inhibition inflammation and restoring blood-milk barrier by activating AhR.

Multifunctional nanoplatform for dual-mode sensitive detection of pathogenic bacteria and the real-time bacteria inactivation

Zhe Hao,Xiaodong Lin,Jinjie Li,Yanliang Yin,Xia Gao,Shuo Wang,Yaqing Liu

Bacterial infection is a growing public health concern and causes a huge medical and financial burden. It is of significance to efficiently construct multifunctional platforms for bacterial point-of-care testing (POCT) and elimination. Herein, near-infrared (NIR) light-responded vancomycin-doped prussian blue nanoparticles (PB-VAN NPs ) with high efficient photothermal conversion was synthesized for binding, dual-mode portable detection, and elimination of bacteria. The PB-VAN NPs can bind to the surface of Gram-positive bacteria such as Staphylococcus aureus ( S. aureus ), forming complex of PB-VAN NPs / S. aureus . After being centrifugated, the suspension solution of PB-VAN NPs can stimulate perfluorohexane (PFH) to rapidly release oxygen (O 2 ) under NIR irradiation. Thus, the bacteria can be sensitively detected with portable pressure meter as signal reader, reporting a limit of detection (LOD) of 1.0  CFU  mL -1 . On the other side, the sediment of PB-VAN NPs / S. aureus can be detected via thermal camera, reporting a LOD of 1.0 CFU mL -1 . Interestingly, the bacteria can be effectively inactivated with the local temperature elevation during temperature-based detection. The antibacterial efficiency reaches as high as 99.8%. The developed multifunctional nanoplatform not only provides a straightforward “mix-then-test” way for portable detection of bacteria with high sensitivity, also realizes high efficiency elimination of bacteria simultaneously. The developed strategy was further applied for promoting wound healing of bacteria-infected mice.
