ssDNA-Functionalized Optical Fiber SPR Sensor for Trace Dopamine Detection

Xiang Li,Rui-jie Tong,Yu Wang,Lu-yao Wang

Dopamine, as a neurotransmitter, is essential for maintaining the function of the extrapyramidal nervous system. This article introduces an optical fiber sensor based on surface plasmon resonance (SPR) for detecting trace dopamine. The SPR effect is induced by an Au film coated on a plastic-cladding optical fiber. The specificity for dopamine is achieved through an aptamer, while graphene serves as a mediator between the gold-coated optical fibers and adapters. The sensor shows a linear sensitivity of 0.853 nm/log(M) over a dopamine concentration range of 10 −11 to 10 −4 M, with a detection limit of 1 × 10 −11 M. Compared with existing dopamine detection methods, the proposed sensor exhibits a lower detection limit, a sufficiently wide linear range, and broad application prospects in biosensing.

High-Sensitivity SPR Temperature Sensor Based on Ag/NiO Nanoflowers

Xu Yanpei,Hao Haixing,Wang Qi

This work presents a high-sensitivity surface plasmon resonance (SPR) temperature sensor based on a BK7 prism coated with silver film and NiO nanoflowers. NiO nanoflowers were synthesized via hydrothermal method and deposited onto the Ag film using a drop-casting technique (3 cycles optimized). The resulting BK7/Ag/NiO nanoflowers structure significantly enhances refractive index (RI) sensitivity. Finite element method (FEM) simulations and experimental measurements confirmed the sensor's performance. The Ag/NiO nanoflowers sensor achieved a remarkable RI sensitivity of 4138.34 nm/RIU, representing a 63.83% improvement over a bare Ag film sensor. Leveraging this enhanced sensitivity and the strong thermo-optic coefficient of silicone oil, a temperature sensor (BK7/Ag/NiO nanoflowers /silicone oil) was developed. It exhibited an excellent temperature sensitivity of -1.61 nm/°C over the range of 25 °C to 75 °C, with a low detection limit of 0.075 °C. The sensor also demonstrated good repeatability and stability, making it a promising candidate for highly sensitive temperature sensing applications.

Smart hydrogel-based optical fiber SPR sensor for pH measurements

Yong Zhao,Ming Lei,Shi-Xuan Liu,Qiang Zhao

A hydrogel-coated optical fiber surface plasmon resonance (SPR) sensor based on MMF-SMF-MMF structure has been fabricated for pH measurements. When the solutions with difference pH values contacting the sensing probe, the amount of the dissociated carboxylic ions in the hydrogel will change according to the pH value of the solution. This process will modify the volume and refractive index of the hydrogel, which results in a shift of the SPR wavelength. The experimental sensor is able to indicate pH variations in a wide pH range from 1 to 12, and the maximum pH sensitivity of the proposed sensor can reach 13 nm/pH at higher pH region (8–10 pH). The influence of temperature (in the range of 20 °C–40 °C) on SPR wavelength is found to be weak. Further, the sensor possesses remarkable repeatability and excellent stability. Due to the wide measuring range and simple fabrication process, the sensor is promising to be used for the pH measurements.

High sensitivity fiber optic SPR sensor for selenium ion concentration detection

Chunlan Liu,Ziqiang Zhang,Yong Wei,Puxi Ren,Yixiong Tang,Chao Guo,Zhihai Liu

Balance of selenium content is of great significance to human health, and there is a need for rapid and sensitive detection of selenium ion concentrations in the human body and food. This article proposes a highly sensitive fiber surface plasmon resonance (SPR) sensor for selenium ion concentration detection. By fabricating a fiber U-shaped ring composite structure and modifying Cu3VSe4 material, the sensitivity of the fiber SPR sensor is improved to 6826.82 nm/RIU. Using 2,4-diamino-6phenyl-1,3,5-triazine to modify the surface of the sensor to achieve specific detection of selenium ion concentration. The experiment results indicate that the detection sensitivity of the sensor is 1.858 nm/(lg(pg/ml)) with range of 100 pg/ml to 1 μg/ml, and the LOD is 124.06 pg/ml. The highly sensitive SPR sensor proposed in this article, without labeling, can achieve high sensitivity and rapid detection of selenium ions, providing a new approach for trace element content detection.

Gain Compensation-Enhanced High-Accuracy SPR Biosensor for Glucose Concentration Detection

Ben Li,Qi Wang

A novel high-accuracy surface plasmon resonance (SPR) optical fiber sensor based on the gain compensation method (GCM) for glucose concentration detection is presented in this paper. For the first time, we proposed a method of generating emission peak using fluorescent material rhodamine 6G (R6G) to compensate for radiation loss and ohmic loss. This approach achieves the reduction of the full width at half-maximum (FWHM) without compromising the sensitivity, thereby addressing the critical challenge in the field of SPR sensors which the FWHM widens as the sensor sensitivity increases, and significantly enhancing the detection accuracy (DA) of the sensor. The present work conducted numerical analysis on the working principle of the gain-compensated sensor and the material concentration of R6G, along with other relevant parameters, using the finite element method (FEM). Experimental validation confirmed the effectiveness of the GCM in improving the DA of the sensor. Ultimately, the measurement results indicated that the refractive index detection accuracy of the sensor, enhanced through the GCM, was improved by 79%, while its sensitivity remaining stable and maintaining a high level of 4194 nm/RIU. Simultaneously, the proposed sensor demonstrated the DA of 9.6 × 10−3 nm−1 and limit of detection (LOD) of 0.012 mg/mL when detecting glucose solution. This study not only offers a new technical pathway for optimizing the performance of SPR optical fiber sensors but also provides vital support for high-accuracy biomarker detection.

SPR-Amplified TiO2–NanoMXene Heterojunction for Ultrasensitive Contactless Human Activity Recognition

Yan Wang,Chong Li,Shiya Qi,Xinghong Chen,Jian Yang,Ziqian Chen,Xueming Hong,Jingting Luo,Yong-Qing Fu,Jinghan Zhang,Xuejin Li,Ran Tao,Yuzhi Chen

Contactless human activity recognition (HAR) is a cornerstone of next-generation intelligent systems, but vision/inertial sensor-based progress is limited by reliability and privacy concerns. Monitoring subtle moisture signatures offers a smart alternative, yet constrained by either sensitive material or sensor architecture design and facing the trade-off among ultrasensitivity, rapid response, and scalable multichannel operation. We address this via a new moisture-sensing paradigm: a plasmon-amplified TiO2–nanoMXene heterojunction integrated onto a fiber-optic tip, leveraging light as an active catalyst in a self-reinforcing feedback loop. Surface plasmon resonance (SPR) injects hot electrons, amplifying the built-in electric field of the heterojunction to enhance H2O polarization and boost the trace moisture adsorption. This alters dielectric constant, feeding back to SPR signals. This self-reinforcing cycle translates subtle moisture signatures into a robust optical output, enabling a new class of HAR with ultrahigh sensitivity, fast response/recovery, minimal hysteresis, and robust environmental resilience. Its transformative potential is validated through contactless finger sensing, noninvasive diaper wetness monitoring, and clinical-grade respiratory analysis.

U-shaped fiber optic SPR sensor for trace Copper ion detection

Hua-Long Du,Qi Wang

The accurate detection of heavy metal ions in aqueous environments is of paramount significance for both environmental conservation and the safeguarding of human health. Based on the surface plasmon resonance (SPR) principle, this study presents a U-shaped multimode fiber SPR sensor designed for detecting copper ion (Cu²⁺) concentration. The sensor eliminates traditional splicing processes by employing a single segment of multimode fiber, thereby significantly enhancing structural stability and sensing performance. The U-shaped sensing region was fabricated using an alcohol lamp. It was subsequently functionalized by sputtering an Au film, followed by coating with barium titanate (BaTiO₃), polyacrylic acid (PAA), and chitosan (CS) films. The specific adsorption of Cu²⁺ by the PAA/CS composite film induces a decrease in the sensing region's effective refractive index (RI), leading to a blue shift in the resonance wavelength. Experimental results demonstrate that the sensor exhibits a sensitivity of -686.2 nm/ppm and a limit of detection (LOD) of 0.0146 ppb for Cu²⁺ across a concentration range of 0.01 to 0.30 ppm, highlighting its superior capability for detecting low-concentration analytes. Moreover, it can accurately detect the presence of Cu²⁺ in aqueous solutions. Furthermore, the sensor shows outstanding stability, repeatability, and specificity, with a relative standard deviation (RSD) of 0.22% in stability tests and a maximum error of 0.0075 ppm in Cu²⁺ concentration detection. The sensor is characterized by simple fabrication, low cost, and compact structure, offering a promising approach for the application of fiber optic SPR technology in heavy metal ion detection.

Nondestructive multiparameter quantification of nanoporous films by broadband SPR/PWR spectroscopy

Ziwei Liu,Zhimei Qi,Xiumei Wan,Hongyi Tang

Nondestructive measurement of film thickness (d) and porosity (P) is crucial for the rational design, controllable synthesis and accurate characterization of multifunctional nanoporous thin films (NPTFs). In this work, the d and P of dielectric NPTFs were measured simultaneously by using a new algorithm to process the experimental data obtained by broadband surface plasmon resonance or plasmon waveguide resonance (SPR/PWR) spectroscopy. The NPTF under test is coated on the pristine gold surface of a SPR sensor chip, which enhances the interaction of light with the NPTF and makes the resonance wavelength highly sensitive to both d and P of the NPTF.The measurements are implemented in three steps: (1) to measure two resonance spectra of a NPTF at different angles of incidence or different surrounding media or different polarizations; (2) to create two d versus P curves by fitting each measured spectrum with a combination of Fresnel and Brueggemann equations; (3) to achieve d and P of the NPTF by solving the intersection of the two d-P curves. It is worth emphasizing that if the NPTF is a multimode waveguide layer, its thickness and porosity can be simply derived by fitting the two widely separated resonance peaks in the measured single PWR spectrum. Six NPTFs, including four TiO 2 films and two SiO 2 films, were characterized according to the above steps. The measured results show good agreement with those obtained by other existing techniques, verifying that the method proposed in this work is accurate and reliable.

Algorithm assisted comprehensive optimization of SPR sensors towards single molecule detection

Tingting Liu,Shiqi Hu,Shilei Ding,Lingling Zhang,Chu Yi,Yaofei Chen,Gui-Shi Liu,Zhe Chen,Wei Xiao,Lei Chen,Yunhan Luo,Donglin Cao

Detection of single molecules is a critical aspect of biochemistry, playing a vital role in enhancing our understanding of molecular interactions and functions. Surface plasmon resonance (SPR) biosensors are increasingly used for this purpose due to their ability to enable label-free and rapid monitoring. However, detecting low-concentration analytes presents a substantial challenge for SPR single molecule detection sensors. This work introduces a multi-objective optimization strategy aimed at enhancing the performance of label-free SPR biosensors by improving various sensing metrics, including sensitivity, figure of merit, and depth of resonant dip. These enhancements lead to a significant improvement in the single molecule detection capabilities of SPR sensors. Additionally, the k-means clustering method is applied to identify appropriate design parameters from the optimized parameter set, thereby mitigating the effects of processing errors on sensor performance. The developed SPR single molecule biosensor has demonstrated significant improvements in refractive index S, FOM, DFOM, with increments of 230.22%, 110.94%, and 90.85%, respectively. The optimized device was tested using mouse IgG to validate the improved sensing performance. Experimental findings indicate that this approach achieves a bulk refractive index sensitivity of 24,482.86 nm/RIU, a broad linear dynamic range from femtograms per milliliter (fg/mL) to micrograms per milliliter (μg/mL), and a detection limit as low as 54 ag/mL (0.36 aM). The multi-objective particle swarm optimization method proposed herein offers an efficient, rapid, and straightforward optimization strategy for SPR biosensors, facilitating ultra-sensitive single molecule detection with low limits of detection (LOD).

Transition metal dichalcogenides boost the performance of optical fiber SPR sensors

Lixia Li,Linlin Zhao,Xueyang Zong,Yuli Li,Penglei Li,Yufang Liu

We show the active role that the transition metal dichalcogenide MoS 2 plays in enhancing the sensing performance of optical fiber sensors based on surface plasmon resonance (SPR). This is proved experimentally suggesting that the optical fiber SPR sensor with the MoS 2 has an about 24 percent increase in the bulk refractive index sensitivity compared to the one without the MoS 2 . Also, we investigate the surface sensing performance of the sensor using self-assembly monolayers of polyelectrolytes with well-defined thickness and refractive index.

Boronate Affinity Molecular Imprinting based Sugar and Ovalbumin Fiber Optic SPR Sensor

Xiaozhan Yang,Mengfei Yang,Bin Chen,Juan He

The sugar and ovalbumin sensors are fabricated by the integration of boronate affinity molecular imprinting with fiber optic surface plasmon resonance (SPR) technology. A 1 cm plastic cladding multimode fiber (PCMF) serves as the sensing element and 300 nm silver film is evaporated on the PCMF end face. The silver film is used to enhance the reflection and is encapsulated by UV glue to prevent oxidation. The silver film is also deposited on the surface of the PCMF to excite the SPR phenomenon. The silica-molecularly imprinted polymer (SiO2-MIPs) was deposited on the silver film by dip coating method for glucose measurement. In the concentration range of 0–0.1 ng/mL, the sensitivity is 482.323 nm/(ng/mL) and the detection limit is 3.94 pg/mL. For ovalbumin detection, 4-vinylphenylboronic acid-ethylene glycol dimethacrylate-MIPs (VPBA-EGDMA-MIPs) material was prepared on the surface of the PCMF. In the range of 0.1–800 pg/mL, the sensitivity of ovalbumin is 2.617 nm/[log(pg/mL)] and the detection limit is 0.1 pg/mL. The prepared sensor has the advantages of simple structure, high sensitivity, excellent selectivity, and good stability, and has wide application prospects in the detection of sugar and glycoproteins with high sensitivity and selectivity.

High-Sensitivity Cellulose-Based Fiber-Optic SPR Humidity Sensor With Temperature Compensation

Nailiang Yu,Xuhao Ji,Zhuo Ren,Yifan Qin,Xiang Li,Chunbiao Liu,Hao Lin,He Zhang,Yu Zhang,Zhihai Liu,Libo Yuan

Achieving both high sensitivity and effective temperature compensation is crucial for high-performance humidity sensors. However, integrating temperature compensation often compromises sensitivity. We develop a fiber-optic surface plasmon resonance (SPR) sensor that integrates a cellulose-based humidity-sensing module and a polydimethylsiloxane (PDMS)-based temperature-sensing module on a plastic-coated multimode optical fiber. By utilizing a highly humidity-sensitive cellulose film, we developed a sensor that delivers high sensitivity while incorporating temperature compensation. The sensor exhibits an average humidity sensitivity of -9.15 nm/RH% and a maximum sensitivity of -14.1 nm/RH% within a relative humidity (RH) range of 25%-70%. In addition, it shows a temperature sensitivity of -2.48 nm/°C between 25 °C and 45 °C. By employing a sensitivity coefficient matrix, the system achieves real-time temperature compensation, thereby significantly enhancing humidity measurement accuracy. Due to its high sensitivity, robust performance, and simple structure, the proposed sensor demonstrates strong potential for practical applications, such as human respiration monitoring and health status analysis.
