As a destructive agricultural pest, Spodoptera frugiperda has spread worldwide in the past few years. Azadirachtin , an environmentally friendly and most promising compound, showed adverse effects, including mortality and growth inhibition, against S. frugiperda . While the effects of azadirachtin on the midgut of this pest remain to be determined. In this study, structural damage was observed in the larval midguts of S. frugiperda with azadirachtin exposure. RNA-seq on the larval midguts with different azadirachtin treatments was performed. Compared to the control group, a total of 3344 and 4759 differentially expressed genes (DEGs) were identified in the midguts with 0.1 and 0.5 μg/g azadirachtin exposure, respectively. Among them, the DEGs encoding detoxification enzymes/proteins, immune-related proteins, digestion and absorption-related proteins, and transcript factors were further analyzed. High-throughput sequencing was also used for the identification of differentially expressed microRNAs in different treatments. A total of 153 conserved miRNAs and 147 novel miRNAs were identified, of which 11 and 29 miRNAs were affected by 0.1 and 0.5 μg/g azadirachtin treatments, respectively. The integrated analysis found that 13 and 178 miRNA versus mRNA pairs were acquired in the samples with 0.1 and 0.5 μg/g azadirachtin treatments, respectively. The results of high-throughput sequencing were confirmed by real-time quantitative polymerase chain reaction (RT-qPCR). These results provide useful information for revealing the molecular mechanism of S. frugiperda larval midgut in response to azadirachtin.

The utilization of highly efficient and ecofriendly stimulus-responsive delivery systems for pesticides represents a powerful strategy for enhancing their efficacy and environmental safety. In this study, the application of emamectin benzoate (EB)-loaded amino-functionalized rough mesoporous silica (KMSNs)@poly(glycidyl methacrylate-co-acrylic acid) (PMA) composites (KMSNs@PMA@EB, 6.52 % loading capacity for EB) to control efficacy for Spodoptera frugiperda was investigated in detail. The results showed that, KMSNs@PMA@EB rapidly released EB under acidic and high-temperature conditions. compared with EB and commercial formulations, a solution of KMSNs@PMA@EB with the agricultural organosilicon additive exhibited excellent wetting. Compared with MSNs, KMSNs@PMA significantly enhanced the effective deposition. Moreover, the photostability of KMSNs@PMA@EB was improved by 1.87-fold and 2.25-fold compared to EB and an EB suspension concentrate (EB SC). S. frugiperda larvae preferred corn leaves with low concentrations (0.125 mg/L) of KMSNs@PMA@EB than EB-treated. Bioassays revealed that KMSNs@PMA@EB maintained > 37 % control efficiency after 21 d of spraying, KMSNs@PMA@EB exhibited significantly greater insecticidal activity and persistence against S. frugiperda than commercial formulations. After 21 days of application, the final deposition rates of KMSNs@PMA@EB were lower than the maximum residual limit of EB, spraying KMSNs@PMA@EB has a lower impact on corn plants. Furthermore, biological safety assessments confirmed that KMSNs@PMA@EB did not affect the maize seed germination and growth and posed a low risk to Microplitis manilae . Those results highlight that the advantages of KMSNs@PMA@EB nanoparticles multidimensional enhancement of pesticide efficacy, persistence, and safety to parasitic wasps and has great potential to be used in pest control combining parasitoid and nanopesticides.

Spodoptera frugiperda (S. frugiperda) is an invasive pest that threatens global crop production and food security and poses a serious threat to maize production worldwide. Metal–organic framework (MOF) nanocarriers have great potential for agricultural pest control applications. The present study successfully prepared the chemical cross-linking of iron-based metal–organic framework nanoparticles (MIL-101(Fe)-NH2 NPs) with sodium lignosulfonate (SL) as a pH/laccase double stimuli-responsive pesticide release system. The average particle size of the prepared chlorfenapyr (CF)-loaded nanoparticles (CF@MIL-101-SL NPs) was 161.54 nm, and the loading efficiency was 44.52%. Bioactivity assays showed that CF@MIL-101-SL NPs increased the toxicity of CF to S. frugiperda and caused the rupture of the peritrophic membrane and enlargement of the midgut. Data from 16S rRNA gene sequencing showed that CF@MIL-101-SL treatment reduced the resistance of S. frugiperda to pesticides and pathogens and affected nutrient and energy availability by remodeling the intestinal microbiota of S. frugiperda. The dysregulated microbial community interacted with the broken peritrophic membrane, which exacerbated damage to the host. Nontargeted metabolomic results showed that ABC transporters may be a potential mechanism for the enhanced toxicity of CF@MIL-101-SL to S. frugiperda. In summary, the present study provides effective strategies for toxicological studies of nanopesticides against insects.

Simple SummaryA previous study found that an indigenous endoparasitic waspCotesia ruficrus, has a good control effect on the invasive pestSpodoptera frugiperda. Understanding the molecular mechanism of host recognition ofC. ruficruswould help improve its biological control effect. Chemoreceptor proteins (CSPs) play a crucial role in insect olfactory perception. The CrufCSP1 protein was obtained via prokaryotic expression, and its binding properties were evaluated using fluorescence binding assays. Molecular docking was used to verify the function of CrufCSP1. The findings revealed that CrufCSP1 has the ability to bind with seven host-related compounds. This information serves as a crucial reference for developing natural enemy attractants for pest control.AbstractChemosensory proteins (CSPs) are a class of soluble proteins that facilitate the recognition of chemical signals in insects. While CSP genes have been identified in many insect species, studies investigating their function remain limited.Cotesia ruficrus(Hymenoptera: Braconidae) holds promise as an indigenous biological control agent for managing the invasive pestSpodoptera frugiperda(Lepidoptera: Noctuidae) in China. This study aimed to shed light on the gene expression, ligand binding, and molecular docking of CrufCSP1 inC. ruficrus. A RT-qPCR analysis revealed that the expression of CrufCSP1 was higher in the wings, with male adults exhibiting significantly higher relative expression levels than other developmental stages. A fluorescence competitive binding analysis further demonstrated that CrufCSP1 has a high binding ability with several host-related volatiles, withtrans-2-hexenal, octanal, and benzaldehyde showing the strongest affinity to CrufCSP1. A molecular docking analysis indicated that specific amino acid residues (Phe24, Asp25, Thr53, and Lys81) of CrufCSP1 can bind to these specific ligands. Together, these findings suggest that CrufCSP1 may play a crucial role in the process ofC. ruficruslocating hosts. This knowledge can contribute to the development of more efficient and eco-friendly strategies for protecting crops and managing pests. Keywords:chemosensory protein;Cotesia ruficrus;expression profiles;fluorescence binding assay;molecular docking;Spodoptera frugiperda;biological control
