The type II polyketide synthase (PKS) natural product enterocin ( 1 ) was isolated from a mangrove-derived novel species Streptomyces qinglanensis 172205 guided by genome sequence, and its putative biosynthetic gene cluster was revealed. Its natural analogues 5-deoxyenterocin ( 2 ) and wailupemycin A–C ( 3–5 ) were also identified by tandem mass spectrometry. By feeding experiments with aryl acids, strain 172205 was proved to incorporate partial exogenous starter units into enterocin- and wailupemycin-based analogues, thus being a new and suitable microorganism for engineering unnatural enc -derived polyketide metabolites. In addition, biological assays indicated that enterocin showed obvious inhibitory activity against β-amyloid protein (Aβ 1-42 ) fibrillation and moderate cytotoxicity against HeLa and HepG2 for the first time.

Streptomyces avermitilis is widely recognized for its remarkable ability to produce diverse secondary metabolites with broad-spectrum antifungal activity. In this study, we employed an integrated multi-omics approach to investigate the antifungal mechanisms of S. avermitilis SM12 and explore its potential as a biocontrol agent. Genomic analysis revealed 39 biosynthetic gene clusters (BGCs), nine of which are predicted to encode antimicrobial compounds. Notably, a gene cluster showing high homology to the oligomycin biosynthetic pathway was identified. Metabolomic profiling via LC–MS and HS–SPME–GC–MS detected several antifungal metabolites, including oligomycin, phthoxazolin, ectoine, and the volatile compound 2,3-hexanedione. The cell-free supernatant obtained after 4 d fermentation significantly suppressed the growth of Fusarium solani and Fusarium oxysporum ( P < 0.05). Subsequent in vitro assays validated the potent antifungal activity of oligomycin, ectoine, and 2,3-hexanedione. Integrated transcriptomic analysis demonstrated temporal activation of the oligomycin and phthoxazolin homologous BGC during the peak antifungal production phase, indicating its crucial role in biocontrol activity. Furthermore, time-series transcriptomics uncovered multifaceted regulatory mechanisms operating at post-transcriptional and translational levels. These results provide key insights into the coordination between BGC expression and metabolite synthesis in S. avermitilis SM12, highlighting promising targets for genetic engineering to enhance antifungal efficacy. This study systematically reveals the molecular basis of biocontrol activity in S. avermitilis SM12, supporting its potential application in sustainable agriculture.

Nitrile hydratases have received significant interest both in the large-scale industrial production of acrylamide and nicotinamide, and the remediation of environmental contamination with nitrile-containing pollutants. Almost all known nitrile hydratases include an α-subunit (AnhA) and β-subunit (AnhB), and a specific activator protein is crucial for their maturation and catalytic activity. Many studies exist on nitrile hydratase characteristics and applications, but few have reported their metal insertion and post-translational maturation mechanism. In this study, we investigated the cobalt insertion and maturation mechanism of nitrile hydratase from Streptomyces canus CGMCC 13662 (ScNHase) bearing three subunits (AnhD, AnhE, and AnhA). ScNHase subunits were purified, and the cobalt content and nitrile hydratase activity of the ScNHase subunits were detected. We discovered that cobalt could insert into the cobalt-free AnhA of ScNHase in the absence of activator protein under reduction agent DL-dithiothreitol (DTT) environment. AnhD not only performed the function of AnhB of NHase, but also acted as a metal ion chaperone and self-subunit swapping chaperone, while AnhE did not act as similar performance. A cobalt direct-insertion under reduction condition coordinated self-subunit swapping mechanism is responsible for ScNHase post-translational maturation. Molecular docking of ScNHase and substrates suggested that the substrate specificity of ScNHase was correlated with its structure. ScNHase had a weak hydrophobic interaction with IAN through protein–ligand interaction analysis and, therefore, had no affinity with indole-3-acetonitrile (IAN). The post-translational maturation mechanism and structure characteristics of ScNHase could help guide research on the environmental remediation of nitrile-containing waste contamination and three-subunit nitrile hydratase.

Plastic products made of polyethylene (PE), polypropylene (PP), and polystyrene (PS) are widely used in daily life and industrial production. Polyolefins—which have a very stable structure and do not contain any active molecular groups—are difficult to degrade and pose a serious global environment threat. This study selected latex clearing protein (Lcp K30 ) derived from Streptomyces sp. Strain K30. The natural substrate of the enzyme is rubber ( cis -1, 4-polyisoprene), and the site of action is the carbon‑carbon double bond. Lcp K30 was incubated with UV-irradiated polyolefin PE, PP and PS (UV-PE, UV-PP, and UV-PS containing carbon‑carbon double bonds) for 5 d at 37 °C. The results showed that UV-PE-Lcp K30 was more fragmented than UV-PE-blank; the Fourier transform infrared spectroscopy results showed that UV-PE-Lcp K30 and UV-PP-Lcp K30 produced new active groups (e.g., –OH and –C=O); however, the effect on UV-PS was not significant. Scanning electron microscopy results showed that the treated group had more obvious roughness, cracks, and pits than the control group. The results of high-temperature gel permeation chromatography showed that the average molecular weight (M w ) of UV-PE-Lcp K30 and UV-PP-Lcp K30 decreased; the M w of UV-PE5-Lcp K30 was reduced by 42.02%. The results of gas chromatography–mass spectrometry showed the production of ketones. Therefore, the Lcp K30 latex clearing protein degrade UV-oxidized polyolefin plastics and has great potential for PE and PP degradation but may not be suitable for PS. Furthermore, other Lcps (such as Lcp NRRL , Lcp NVL3 ) can also degrade UV-PE.

Cytochrome P450 enzymes (P450 or CYP) are some of the most versatile biocatalysts , and offer advantages for oxidizing unreactive C–H bonds in mild conditions. In this study, we identified a novel cytochrome P450 154C2 from Streptomyces avermitilis and characterized its function in 2α-hydroxylation of testosterone with regio- and stereoselectivity . To investigate the efficiency of electron transfer , we conducted biotransformation using two different P450 redox partners—RhFRED (RhF reductase domain) from Rhodococcus sp. and Pdx (putidaredoxin)/Pdr (putidaredoxin reductase) from Pseudomonas putida and revealed that RhFRED was more effective than Pdx/Pdr, especially in vivo . The K m and k cat values for testosterone were estimated to be 0.16 ± 0.05 mM and 0.13 ± 0.02 min −1 , and k cat / K m was 0.81 min −1 mM −1 . We also determined the crystal structure of the substrate-free form of CYP154C2 at 1.5 Å resolution. The structure has a closed conformation, and the substrate binding pocket is narrow, which can explain the strict substrate specificity of the enzyme .

Clubroot, caused byPlasmodiophora brassicaeWoronin, poses a major threat to Chinese cabbage (Brassica rapasubsp.pekinensis) production worldwide, significantly impacting crop yield, quality, and economic value. Biological control represents a promising approach since it is non-toxic and eco-friendly, and it reduces the risk of pathogen resistance development. In this study, our objective was to screen for actinomycetes that can effectively inhibit clubroot. We screened 13 actinomycete strains, identifying 2, XDS3-6 and CD1-1, with substantial in vivo inhibitory effects, achieving infection suppression rates above 64% againstP. brassicae. Phylogenetic analysis classified XDS3-6 and CD1-1 asStreptomyces virginiaeandStreptomyces cinnamonensis, respectively. Both strains exhibited protease and glucanase production capabilities, essential for pathogenic suppression. Additionally, these strains induced host defense responses, as evidenced by increased jasmonic acid (JA) and salicylic acid (SA) accumulation and elevated activities of defense-related enzymes. Colonization studies of XDS3-6 and CD1-1 mutant strains in cabbage roots indicated sustained root colonization, with peak colony-forming units (CFUs) at 20 days post-inoculation, reaching 11.0 × 104CFU/g and 8.5 × 104CFU/g, respectively, and persisting for at least 30 days. Overall, these findings underscore the potential ofStreptomycesstrains XDS3-6 and CD1-1 as effective biocontrol agents, providing a theoretical foundation for their application in managing clubroot in Chinese cabbage.

Colletotrichum scovillei is a pathogen that causes anthracnose in pepper fruit, resulting in severe postharvest losses in pepper fruit. Volatile organic compounds (VOCs) produced by microorganisms are regarded as promising environmentally safe fumigants for controlling postharvest diseases. In this study, the Streptomyces lactacystinicus ZZ-84 strain was identified as exhibiting strong antagonistic activity against C. scovillei and effectively controlling anthracnose in pepper fruit. The VOCs generated by ZZ-84 significantly inhibited the mycelial growth and conidial germination of C. scovillei , disrupted mycelial morphology, altered cell membrane permeability, and induced oxidative stress. Transcriptomic analysis revealed that the VOCs primarily affected the metabolic pathways of C. scovillei , particularly those related to protein synthesis, cell membrane integrity, redox homeostasis, and energy conversion. The volatiles 2-methyl-1-butanol, sulphuric acid dibutyl ester, and 6-methyl-2-heptanone were identified as the main antagonistic compounds among the VOCs and exhibited strong antifungal activity against C. scovillei. Overall, strain ZZ-84 has the potential to serve as a microbial fumigation agent for controlling postharvest anthracnose in pepper.

Monensin is a polyether antibiotic produced by Streptomyces cinnamonensis . Due to its highly complex biosynthetic pathway, incomplete characterization of key biosynthetic genes has long hindered improvements in industrial-scale monensin production. Based on our previous transcriptomic analysis that key genes involved in monensin biosynthesis were initially identified in a high-yield S. cinnamonensis strain, this study focused on metabolic engineering of this strain through multi-gene integration strategy, targeting primary metabolism, precursor synthesis, ATP supply, stress resistance, and the monensin biosynthetic gene cluster. Results demonstrated that the co-expression of phosphofructokinase ( pfk ), citrate synthase ( cs ), methylmalonyl-CoA mutase subunit β ( mutB ), and methylmalonyl-CoA mutase ( mcm ) rendered the monensin titer from 12.6 g/L to 20.2 g/L, an approximately 60% increase compared with the original strain. Co-expression of the ATP synthase β subunit ( assβ ), the fungal thiol-coupled enzyme ( mac ), and the monensin biosynthetic enzyme ( monE ) allowed the monensin production from 12.6 g/L to 19.4 g/L, corresponding to an increase of above 50%. Furthermore, simultaneous co-expression of the seven genes using the novel pKHBT1 system increased the monensin titer to 26.2 g/L in a 5-L fermenter, more than double that of the original strain. These newly identified key metabolic genes were indeed associated closely with the monensin biosynthesis in S. cinnamonensis , which supplied a clue to metabolic engineering of the monensin production strain for efficient biosynthesis.

Phospholipase A1 (PLA1) selectively hydrolyzes the sn-1 ester of phospholipids and is ideal for enzymatic oil degumming. To efficiently produce PLA1, this study heterologously expressed the saPLA1 gene from Streptomyces albidoflavus NA297 in the genome-reduced strain Streptomyces mobaraensis smYS1-Δ tg , and analyzed the enzymatic properties and degumming performance of the recombinant enzyme. Results showed that under the control of the strong promoter P gapdh and the transglutaminase (TGase) signal peptide, secretory activity peaked at 39.4 U/mL within 48 h. It exhibited optimal activity at 50°C and pH 6.0, with good stability over pH 5–8. Additionally, saPLA1 was significantly activated by Mn 2 + , Fe 2+ and citrate, and functioned in a Ca 2+ -independent manner. Under optimized process conditions, saPLA1 reduced the phosphorus content in crude soybean oil from 113.01 to 7.27 mg/kg within 2.5 h, meeting the physical refining requirement. This study provides a novel strategy for the industrial production and application of food-grade saPLA1.

Volatile organic compounds (VOCs) produced by microorganisms are considered promising environmental-safety fumigants for controlling postharvest diseases. Ceratocystis fimbriata, the pathogen of black spot disease, seriously affects the quality and yield of sweet potato in the field and postharvest. This study tested the effects of VOCs produced by Streptomyces setonii WY228 on the control of C. fimbriata in vitro and in vivo. The VOCs exhibited strong antifungal activity and significantly inhibited the growth of C. fimbriata. During the 20-day storage, VOC fumigation significantly controlled the occurrence of the pathogen, increased the content of antioxidants and defense-related enzymes and flavonoids, and boosted the starch content so as to maintain the quality of the sweet potatoes. Headspace analysis showed that the volatiles 2-ethyl-5-methylpyrazine and dimethyl disulfide significantly inhibited the mycelial growth and spore germination of C. fimbriata in a dose-dependent manner. Fumigation with 100 μL/L 2-ethyl-5-methylpyrazine completely controlled the pathogen in vivo after 10 days of storage. Transcriptome analysis showed that volatiles mainly downregulated the ribosomal synthesis genes and activated the proteasome system of the pathogen in response to VOC stress, while the genes related to spore development, cell membrane synthesis, mitochondrial function, and hydrolase and toxin synthesis were also downregulated, indicating that WY228-produced VOCs have diverse modes of action for pathogen control. Our study demonstrates that fumigation of sweet potato tuberous roots with S. setonii WY228 or use of formulations based on the VOCs is a promising new strategy to control sweet potato and other food and fruit pathogens during storage and shipment.IMPORTANCE Black spot disease caused by Ceratocystis fimbriata has caused huge economic losses to worldwide sweet potato production. At present, the control of C. fimbriata mainly depends on toxic fungicides, and there is a lack of effective alternative strategies. The research on biological control of sweet potato black spot disease is also very limited. An efficient biocontrol technique against pathogens using microbial volatile organic compounds could be an alternative method to control this disease. Our study revealed the significant biological control effect of volatile organic compounds of Streptomyces setonii WY228 on black spot disease of postharvest sweet potato and the complex antifungal mechanism against C. fimbriata. Our data demonstrated that Streptomyces setonii WY228 and its volatile 2-ethyl-5-methylpyrazine could be a candidate strain and compound for the creation of fumigants and showed the important potential of biotechnology applications in the field of food and agriculture.

A Streptomyces albulus strain NJZJSA2 was isolated from the forest soil sample of Tzu-chin Mountain (Nanjing China) and identified based on its morphological and physiological properties and 16S rDNA gene sequence analysis. The strain S. albulus NJZJSA2 was evaluated for the production of antifungal volatile organic compounds (VOCs) against two fungal pathogens. Results showed that the VOCs generated by S. albulus NJZJSA2 inhibited mycelial growth of Sclerotinia sclerotiorum (SS) and Fusarium oxysporum (FO) by 100 and 56.3%, respectively. The germination of SS sclerotia and FO conidia was completely inhibited in the presence of VOCs produced by S. albulus NJZJSA2 in vitro . In soil, the VOCs delayed the germination of SS sclerotia and inhibited the germination of FO conidia for 45 days. The strain S. albulus NJZJSA2 was able to produce 13 VOCs based on GC/MS analyses. Among those, six compounds were purchased and used for the antifungal activity assay. Three relatively abundant VOCs, 4-methoxystyrene, 2-pentylfuran, and anisole were proved to have antifungal activity. Microscopy analysis showed that the pathogen hyphae were shriveled and damaged after treatment with 4-methoxystyrene. These results suggest that the S. albulus strain NJZJSA2 produce VOCs that not only reduce the growth of SS and FO, but also significantly inhibit the SS sclerotia and FO conidia. The results are useful for the better understanding of biocontrol mechanisms by S. albulus strains and will help to improve the biological control efficiency of lethal plant diseases.
