Core@shell Fe3O4@Mn2+-doped NaYF4:Yb/Tm nanoparticles for triple-modality T1/T2-weighted MRI and NIR-to-NIR upconversion luminescence imaging agents

Yang Luo,Sinan Du,Wei Zhang,Zhengfang Liao,Fang Zuo,Shengtao Yang

Core@shell structures of Fe3O4@Mn2+-doped NaYF4:Yb/Tm nanoparticles (NPs) with multifunctional properties were prepared using a hydrothermal route with a seed-growth procedure. The effect of Mn2+ ions on the phase, shape, and upconversion luminescence (UCL) of Fe3O4@Mn2+-doped NaYF4:Yb/Tm NPs was explored using X-ray diffraction, transmission electron microscopy, dynamic light scattering, and photoluminescence spectroscopy. Furthermore, hydrophobic to hydrophilic surface modification of the Fe3O4@Mn2+-doped NaYF4:Yb/Tm NPs was achieved by coating the NPs with an amphiphilic polymer (poly(maleic anhydride-alt-1-octadecene) (C18PMH)) modified with amine-functionalized methyl ether poly(ethylene glycol). Then, the NIR-to-NIR UCL ascribed to the Tm3+ ions, T1-weighted MRI ascribed to the Mn2+ ions, and T2-weighted MRI ascribed to the Fe3O4 core of the hydrophilic Fe3O4@Mn2+-doped NaYF4:Yb/Tm NPs were evaluated. The results indicate that Mn2+ doping is an effective method to control the size/shape and improve the UCL intensity of Fe3O4@Mn2+-doped NaYF4:Yb/Tm NPs, which are promising as imaging agents for NIR-to-NIR UCL and T1/T2-weighted MRI in biomedical research.

One-pot Synthesis of PEGylated Gd-based Nanoparticles as High-performance and Biocompatibility Contrast Agents for T1-Weighted Magnetic Resonance Imaging In vivo

Huang Sa,Han Qinghe,Wang Lei,Gong Tingting,Yuan Qinghai

Polyethylene glycol modified(PEGylated) NaGdF 4 (PEG-NaGdF 4 ) nanoparticles as a novel T 1 -weighted magnetic resonance imaging(MRI) contrast agent was successfully constructed by a one-pot hydrothermal synthesis method. Because of the functionalization of PEG, the nanoprobes had excellent dispersity, excellent stability and high biocompatibility. More importantly, the as-prepared PEG-NaGdF 4 nanoprobes revealed the high longitudinal relaxivity value and prominent T 1 -weighted MRI contrast performance, which was superior to the commercial MRI contrast agents. With the facile synthesis, excellent dispersity, outstanding stability, remarkable contrast performance and high biocompatibility, the PEGylated NaGdF 4 nanoparticles brought more opportunities to the new generation of nanoparticulate-based T 1 -weighted MRI contrast agents in clinic.

Near-infrared light-activated red-emitting upconverting nanoplatform for T1-weighted magnetic resonance imaging and photodynamic therapy

Xiang-long Tang,Jun Wu,Ben-lan Lin,Sheng Cui,Hong-mei Liu,Ru-tong Yu,Xiao-dong Shen,Ting-wei Wang,Wei Xia

Photodynamic therapy (PDT) has increasingly become an efficient and attractive cancer treatment modality based on reactive oxygen species (ROS) that can induce tumor death after irradiation with ultraviolet or visible light. Herein, to overcome the limited tissue penetration in traditional PDT, a novel near-infrared (NIR) light-activated NaScF 4 : 40% Yb, 2% [email protected] 2 upconversion nanoparticle (rUCNP) is successfully designed and synthesized. Chlorin e6, a photosensitizer and a chelating agent for Mn 2+ , is loaded into human serum albumin (HSA) that further conjugates onto rUCNPs. To increase the ability to target glioma tumor, an acyclic Arg–Gly–Asp peptide (cRGDyK) is linked to [email protected] (Ce6-Mn). This nanoplatform enables efficient adsorption and conversion of NIR light (980 nm) into bright red emission (660 nm), which can trigger the photosensitizer Ce6-Mn complex for PDT and T 1 -weighted magnetic resonance imaging (T 1 -weighted MRI) for glioma diagnosis. Our in vitro and in vivo experiments demonstrate that NIR light-activated and glioma tumor-targeted PDT can generate large amounts of intracellular ROS that induce U87 cell apoptosis and suppress glioma tumor growth owing to the deep tissue penetration of irradiated light and excellent tumor-targeting ability. Thus, this nanoplatform holds potential for applications in T 1 -weighted MRI diagnosis and PDT of glioma for antitumor therapy. Statement of Significance A near-infrared (NIR) light-activated nanoplatform for photodynamic therapy (PDT) was designed and synthesized. The Red-to-Green (R/G) ratio of NaScF 4 : 40% Yb, 2% Er almost reached 9, a value that was much higher than that of a traditional Yb/Er-codoped upconversion nanoparticle (rUCNP). By depositing a CaF 2 shell, the red-emission intensities of the rUCNPs were seven times strong as that of NaScF 4 : 40% Yb, 2% Er. The enhanced red-emitting rUCNPs could be applied in many fields such as bioimaging, controlled release, and real-time diagnosis. The nanoplatform had a strong active glioma-targeting ability, and all results achieved on subcutaneous glioma demonstrated that our NIR light-activated red-emitting upconverting nanoplatform was efficient for PDT. By loading Ce6-Mn complex into [email protected] , the nanoplatform could be used as a T 1 -weighted magnetic resonance imaging agent for tumor diagnosis.

Alpha-Synuclein Oligomers Driven T1–T2 Switchable Nanoprobes for Early and Accurate Diagnosis of Parkinson's Disease

Ying Chen,Zeyu Liang,Qiyue Wang,Lin Xiao,Shangzhi Xie,Shengfei Yang,Xun Liu,Daishun Ling,Fangyuan Li

The alpha-synuclein (α-syn) oligomers hold a central role in the pathology of Parkinson's disease (PD). Achieving accurate detection of α-syn oligomers in vivo presents a promising avenue for early and accurate diagnosis of PD. Magnetic resonance imaging (MRI), with non-invasion and exceptional tissue penetration, offers a potent tool for visualizing α-syn oligomers in vivo. Nonetheless, ensuring diagnostic specificity remains a formidable challenge. Herein, a novel MRI probe (ASOSN) is introduced, which encompasses highly sensitive antiferromagnetic nanoparticles functionalized with single-chain fragment variable antibodies, endowing it with the capacity for discerning recognition and binding to α-syn oligomers and triggering a switchable T1–T2 MRI signal. Significantly, ASOSN possesses the unique capability to accurately discriminate α-syn oligomers from neuroinflammation in vivo. Moreover, ASOSN facilitates the non-invasive and precise visualizing of endogenous α-syn oligomers in living systems. This innovative design heralds the development of a non-invasive visualization strategy for α-syn oligomers, marking a pivotal advancement for early and accurate diagnosis of PD.

Ovalbumin-loaded paramagnetic nano-triangles for enhanced dendritic cell stimulation, T1-MR imaging, and antitumor immunity

Yike Hou,Zhe Tang,Jabeen Farheen,Madiha Saeed,Lijia Luo,Wenzi Ren,Dandan Luo,Asim Mushtaq,Ruibo Zhao,Jian Ge,Zhangsen Yu,Yao Li,M. Zubair Iqbal,Xiangdong Kong

Vaccine-based cancer immunotherapy has demonstrated a significant potential for cancer treatment in clinics. Although the efficiencies of vaccines are limited, they can be enhanced by a well-designed antigen delivery system that promotes sufficient antigen presentation of dendritic cells (DCs) for initiating high T cell immunity. Herein, antigen-loaded manganese oxide (Mn 3 O 4 ) triangular-shaped ultrasmall nanoparticles were prepared to stimulate DC-based immunotherapy under the guidance of T 1 magnetic resonance imaging. The FDA-approved triblock copolymer Pluronic® F-68 was used not only to transfer the phase from hydrophobic to hydrophilic but also to enrich antigen loading and improve the biocompatibility of the prepared nanoparticles. Ovalbumin (OVA), a model antigen, was adsorbed on the surface of polymer-coated nanoparticles through electrostatic interaction to form Mn 3 O 4 @PF68-OVA nanoparticle-antigen complexes to stimulate DC-based immunization and antigen-specific T cell immunity. The Mn 3 O 4 @PF68-OVA nanovaccine (NV) induces negligible toxicity effects against 4T1 and bone marrow-derived dendritic cells (BMDCs) by conventional methods supports the proliferation of intestine organoids, which are an innovative three-dimensional cytotoxicity evaluation system, thereby indicating their potential safety for in vivo cancer therapies. The designed paramagnetic nanovaccine possessed excellent OVA delivery to dendritic-regulated antigen-specific T cells in vitro by stimulating the maturation level of BMDCs. In addition, Mn 3 O 4 @PF68-OVA NVs enhance immunity in vivo by increasing the T-cells and M1 macrophages, which suggests improved immunity. Excitingly, vaccination with Mn 3 O 4 @PF68-OVA offer complete protection in the prophylactic group and significant tumor inhibition in the therapeutic group against B16-OVA tumor. In addition, the designed nanovaccine demonstrated high T 1 -MR imaging in the tumor, further justifying enhanced tumor accumulation and capability to real-time monitor the treatment procedure. This study presents a promising nanosystem to design an effective nanovaccine for T 1 -MR imaging-guided tumor immunotherapy.

Transferrin-Targeted Nanoarchitectonics with Efficient Metastasis Inhibition Enabling Tumor T1-T2 Dual-Mode Magnetic Resonance Contrast Enhancement

Yinuo Yuan,Liya Tian,Xinyu Liu,Kai Guo,Ke Ren,Lei Yan,Mengyao Mu,Shangui Liu,Feifei Teng,Cun Liu,Ping Liang,Xiao Sun

The development of T 1 -T 2 dual-mode contrast agents is expected to improve cancer diagnostic capabilities in response to the limitation of traditional single-mode magnetic resonance imaging (MRI) contrast agents. In this study, a pH-responsive dual-mode MRI nanoprobe, Fe 3 O 4 @MnS-transferrin (FMT), is prepared with the objective of augmenting tumor visualization and chemodynamic therapy (CDT). FMT achieved precise tumor targeting by binding to transferrin receptors on cancer cells via the transferrin molecules on their surface. FMT underwent pH-responsive decomposition in the acidic tumor microenvironment, releasing Fe 3 O 4 and Mn 2+ . This decomposition increased the spatial separation between Fe 3 O 4 and Mn 2+ , attenuating the mutual magnetic shielding effect and activating dual-mode MRI function. Moreover, FMT released Mn 2+ and hydrogen sulfide (H 2 S). Mn 2+ triggered a Fenton-like reaction, generating reactive oxygen species that induced cytotoxicity through CDT. Concurrently, H 2 S inhibited catalase activity, providing additional substrates for the Fenton-like reaction, thereby amplifying CDT in a cascading manner. This synergistic mechanism amplified the cytotoxicity of CDT, enhancing the efficacy of tumor treatment and metastasis inhibition. Therefore, FMT demonstrated significant promise for enhancing both the precision of tumor imaging and the efficacy of treatment.

T1-weighted MRI of targeting atherosclerotic plaque based on CD40 expression on engulfed USPIO's cell surface

Chen Huang,Wentao Huang,Yixuan Meng,Chengqian Zhou,Xiaozhuan Wang,Chunyu Zhang,Yuzhen Tian,Wei Wei,Yongsheng Li,Quan Zhou,Yukuan Tang,Wenli Chen

Atherosclerosis is a chronic inflammatory disease characterized by the accumulation of cholesterol within the arterial wall. Its progression can be monitored via magnetic resonance imaging (MRI). Ultrasmall Superparamagnetic Particles of Iron Oxide (USPIO) (<5 nm) have been employed as T1 contrast agents for MRI applications. In this study, we synthesized USPIO with an average surface carboxylation of approximately 5.28 nm and a Zeta potential of -47.8 mV. These particles were phagocytosed by mouse aortic endothelial cells (USPIO-MAECs) and endothelial progenitor cells (USPIO-EPCs), suggesting that they can be utilized as potential contrast agent and delivery vehicle for the early detection of atherosclerosis. However, the mechanism by which this contrast agent is delivered to the plaque remains undetermined. Our results demonstrated that with increasing USPIO concentration during 10-100 μg/mL, consistent change appeared in signal enhancement on T1-weighted MRI. Similarly, T1-weighted MRI of MAECs and EPCs treated with these concentra-tions exhibited a regular change in signal enhancement. Prussian blue staining of USPIO revealed substantial absorption into MAECs and EPCs after treatment with 50 μg/mL USPIO for 24 h. The iron content in USPIO-EPCs was much higher (5 pg Fe/cell) than in USPIO-MAECs (0.8 pg Fe/cell). Our results confirmed that macrophages are more likely to bind to MAECs treated with AuNPs-PEI-siRNANC (control) for 72 h than to MAECs treated with AuNPs-PEI-siRNACD40 (reduced CD40 expression), thus confirming CD40 targeting at the cellular level. When USPIO-MAECs and MAECs (control) were delivered to mice (high-fat-fed) via tail vein injection respectively, we observed a higher iron accumulation in plaques on blood vessels in high-fat-fed mice treated with USPIO-MAECs. We also demonstrated that USPIO-EPCs, when delivered to high-fat-fed mice via tail vein injection, could indeed label plaques by generating higher T1-weighted MRI signals 72 h post injection compared to controls (PBS, USPIO and EPCs alone). In conclusion, we synthesized a USPIO suitable for T1-weighted MRI. Our results have confirmed separately at the cellular and tissue and in vivo level, that USPIO-MAECs or USPIO-EPCs are more accessible to atherosclerotic plaques in a mouse model. Furthermore, the high expression of CD40 on the cell surface is a key factor for targeting and USPIO-EPCs may have potential therapeutic effects.

Facile synthesis of ultra-small hollow manganese silicate nanoparticles as pH/GSH-responsive T1-MRI contrast agents

Jinyue Zhang,Mingshi Jin,Yong Il Park,Longyi Jin,Bo Quan

Manganese silicate nanoparticles have recently attracted great attention due to the effectiveness of T 1 -weighted magnetic resonance imaging (MRI) contrast agents (CAs) in the mildly acidic and high glutathione (GSH) conditions of the tumor microenvironment. Herein, ultra-small hollow manganese silicate (UHMS) nanoparticles were prepared via a facile, economical, and eco-friendly method based on a hydrothermal process using small silica nanoparticles as both the silicon source and templates. The nanoparticles showed remarkable biocompatibility and dispersion stability after modification with polyethylene glycol (PEG). Owing to their ultra-high longitudinal relaxation rate (r 1 ) at low pH values and high GSH levels, the [email protected] nanoparticles have performed an excellent pH/GSH-responsive T 1 -weighted MRI. Meanwhile, the nanoparticles showed insignificant cytotoxicity in vitro and in vivo. Therefore, the [email protected] nanoparticles show promise as T 1 -MRI CAs in the tumor microenvironment and may be used to assist in clinical cancer diagnosis.

Biomineralization Synthesis of HoMn Nanoparticles for Ultrahigh-Field-Tailored and T1–T2 Dual-Mode MRI-Guided Cancer Theranostics

Qingqing Cheng,Yanzhou Chang,Dong Zhang,Xiangsheng Zhao,Zeyu Xiao,Tianfeng Chen,Changzheng Shi,Liangping Luo

Ultrahigh field magnetic resonance imaging (UHF-MRI) (≥7 T) can dramatically boost image resolution and signal-to-noise ratio, which have distinct advantages in multifunctional imaging. However, their research and application are currently limited by the absence of high-field contrast agents (CAs) and the low sensitivity and accuracy of T1/T2 single-modality CAs. Therefore, the development of T1–T2 dual-mode CAs that respond to UHF-MRI and nanoformulations with therapeutic sensitization can bring ideas for the integrated application of precise and synchronous tumor theranostics. Herein, we present a biomimetic mineralization strategy for synthesizing holmium/manganese oxide–bovine serum albumin–photosensitizer chlorin e6 nanohybrids. The hybrid nanoparticles exhibited better tumor accumulation, a suitable time imaging window, and excellent pH-response T1–T2 dual-mode UHF-MRI performance. The antitumor effect comes from the amelioration of the hypoxic tumor microenvironment to promote the synergistic effect of photodynamic therapy and radiotherapy, along with negligible acute toxicity. Undoubtedly, this work not only provides a different perspective for developing multifunctional nanotherapeutics but also promotes the potential clinical exploitation and translation of UHF CAs.

The Application of Nuclear Magnetic Resonance 1D T2 and 2D T1–T2 Maps in the Research of Condensate Oil Saturation in Condensate Gas Reservoirs

Bowen Sun,Ping Guo

Determination of condensate oil saturation in condensate gas reservoirs is the key parameter for estimating condensate oil blockage and gas well productivity. In PVT experiments, the constant volume depletion (CVD) method is usually adopted to obtain condensate oil saturation. However, in this method, the effect of porous media is not considered. This study applied a low field nuclear magnetic resonance (NMR) test instrument in which 1D T2 and 2D T1–T2 NMR techniques are carried out to investigate the condensate oil saturation of “condensate gas-saturated” core plugs in the depletion process of 29 to 5 MPa at 25 °C. It was observed here that the T1/T2 ratios of condensate oil and gas at the signal peak are approximately 20 and 60, respectively, and the condensate oil and gas could be distinguished by positions in 2D NMR T1–T2 maps. Additionally, a standard equation (the relationship between the signal intensity and the condensate oil saturation) with an excellent linear relationship was established for the calculation of the condensate oil saturation in core plugs. The results indicated that the condensate oil first appears in micropores, and the retrograde condensation phenomenon of the core mainly comes from the contribution of mesopores and macropores; furthermore, the maximum condensate oil saturation in the porous medium is 5.36% higher than that in the PVT cylinder, and the dew point pressure in the porous medium is 11.83% higher than that in the PVT cylinder. The T1–T2 NMR technique may become an efficient approach for assessing fluid distributions and saturations, simultaneously possessing potential applications for characterizing fluid occurrence states in the core.

An intelligent tumor microenvironment responsive nanotheranostic agent for T1/T2 dual-modal magnetic resonance imaging-guided and self-augmented photothermal therapy

Junlie Yao,Fang Zheng,Fang Yang,Chenyang Yao,Jie Xing,Zihou Li,Sijia Sun,Jia Chen,Xiawei Xu,Yi Cao,Norbert Hampp,Aiguo Wu

Photothermal therapy (PTT), as a promising antineoplastic therapeutic strategy, has been harnessed to restrain tumor growth through near-infrared (NIR) irradiation mediated thermal ablation. Nevertheless, its biological applications are hampered by thermal diffusion and up-regulated heat shock proteins (HSPs). Herein, a versatile nanotheranostic agent is developed via integrating Zn0.2Fe2.8O4 nanoparticles (NPs), polydopamine (PDA), and MnO2 NPs for T1/T2 dual-modal magnetic resonance (MR) imaging-guided and self-augmented PTT. The as-designed Zn0.2Fe2.8O4@PDA@MnO2 NPs adequately serve as a PTT agent to realize effective photothermal conversion and obtain local hyperthermia. Additionally, the Zn0.2Fe2.8O4@PDA@MnO2 NPs can significantly consume overexpressed glutathione (GSH) and generate Mn2+ in the tumor microenvironment (TME), thus destroying redox homeostasis and catalytically generating hydroxyl radicals (˙OH) for HSP suppression and PTT enhancement. Meanwhile, Mn2+ and Zn0.2Fe2.8O4 NPs significantly strengthen T1- and T2-weighted MR contrast for tumor imaging and PTT guidance. Hence, this study offers proof of concept for self-augmented PTT and T1/T2 dual-modal MR imaging for tumor elimination.

Ultrasmall Fe3O4 nanoparticles self-assembly induced dual-mode T1/T2-weighted magnetic resonance imaging and enhanced tumor synergetic theranostics.

Qinghua Xie,Xuemei Wang,Gaorui Zhang,Dawei Zhou,Yuxuan Zhao,Hong Liu,Jiazhi Duan,Dexin Yu,Yuanhua Sang

Individual theranostic agents with dual-mode MRI responses and therapeutic efficacy have attracted extensive interest due to the real-time monitor and high effective treatment, which endow the providential treatment and avoid the repeated medication with side effects. However, it is difficult to achieve the integrated strategy of MRI and therapeutic drug due to complicated synthesis route, low efficiency and potential biosafety issues. In this study, novel self-assembled ultrasmall Fe 3 O 4 nanoclusters were developed for tumor-targeted dual-mode T 1 /T 2 -weighted magnetic resonance imaging (MRI) guided synergetic chemodynamic therapy (CDT) and chemotherapy. The self-assembled ultrasmall Fe 3 O 4 nanoclusters synthesized by facilely modifying ultrasmall Fe 3 O 4 nanoparticles with 2,3-dimercaptosuccinic acid (DMSA) molecule possess long-term stability and mass production ability. The proposed ultrasmall Fe 3 O 4 nanoclusters shows excellent dual-mode T 1 and T 2 MRI capacities as well as favorable CDT ability due to the appropriate size effect and the abundant Fe ion on the surface of ultrasmall Fe 3 O 4 nanoclusters. After conjugation with the tumor targeting ligand Arg-Gly-Asp (RGD) and chemotherapy drug doxorubicin (Dox), the functionalized Fe 3 O 4 nanoclusters achieve enhanced tumor accumulation and retention effects and synergetic CDT and chemotherapy function, which serve as a powerful integrated theranostic platform for cancer treatment.
