Personalized radiotherapy strategies enabled by the construction of hypoxia-guided biological target volumes (BTVs) can overcome hypoxia-induced radioresistance by delivering high-dose radiotherapy to targeted hypoxic areas of the tumor. However, the construction of hypoxia-guided BTVs is difficult owing to lack of precise visualization of hypoxic areas. This study synthesizes a hypoxia-responsive T 1 , T 2 , T 2 mapping tri-modal MRI molecular nanoprobe (SPION@ND) and provides precise imaging of hypoxic tumor areas by utilizing the advantageous features of tri-modal magnetic resonance imaging (MRI). SPION@ND exhibits hypoxia-triggered dispersion-aggregation structural transformation. Dispersed SPION@ND can be used for routine clinical BTV construction using T 1 -contrast MRI. Conversely, aggregated SPION@ND can be used for tumor hypoxia imaging assessment using T 2 -contrast MRI. Moreover, by introducing T 2 mapping, this work designs a novel method (adjustable threshold-based hypoxia assessment) for the precise assessment of tumor hypoxia confidence area and hypoxia level. Eventually this work successfully obtains hypoxia tumor target and accurates hypoxia tumor target, and achieves a one-stop hypoxia-guided BTV construction. Compared to the positron emission tomography-based hypoxia assessment, SPION@ND provides a new method that allows safe and convenient imaging of hypoxic tumor areas in clinical settings.

Simple SummaryT-2 toxin, a widespread mycotoxin, induces hepatotoxicity and gut dysbiosis, with limited mitigation strategies. This study demonstrates that lycopene protects against T-2 mycotoxicosis in mice by integrating microbiota restoration with hepatic metabolic reprogramming. Lycopene suppressed pro-inflammatory cytokines, alleviated oxidative stress, and enriched beneficial Muribaculum while suppressing pathogenic Escherichia. Mechanistically, lycopene de-repressed Peroxisome proliferator-activated receptor (PPAR) signaling and restored fatty acid oxidation and phase I detoxification gene expression. A hormetic dose–response was evident: low-dose lycopene outperformed high-dose lycopene, underscoring non-linear efficacy. These findings position lycopene as a candidate feed additive targeting the gut–liver–PPAR axis, though dose optimization remains critical for translation.Exposure to T2 toxin is known to induce hepatotoxicity and gut dysbiosis, yet effective dietary interventions remain underexplored. This study investigates the hepatoprotective and microbiota-modulating effects of lycopene against T2 toxin-induced toxicity in mice. Mice were exposed to T2 toxin with or without lycopene supplementation at low and high doses. The hepatic function, oxidative stress markers, inflammatory gene expression, detoxification pathway activity, and gut microbiota composition were assessed using histological, biochemical, and molecular analyses. T2 toxin exposure resulted in significant weight loss, oxidative liver damage, and gut dysbiosis—marked by a decline in beneficial phyla and an increase in pathogenic bacteria. Hepatic injury was accompanied by upregulated pro-inflammatory genes and downregulated PPAR pathway genes, leading to impaired lipid metabolism and disrupted liver histology. Lycopene supplementation effectively attenuated these effects: it reduced oxidative stress, enhanced antioxidant defense, lowered inflammatory markers, and restored gut microbial balance. Furthermore, lycopene upregulated PPAR pathway and phase I detoxification genes. Notably, the low-dose lycopene regimen demonstrated superior efficacy compared to the high-dose regimen. In conclusion, lycopene, particularly at a low dose, confers significant protection against T2 toxin-induced hepatotoxicity and gut dysbiosis, highlighting its potential as a dietary strategy for mitigating mycotoxin-induced health risks.

Iron oxide nanoparticles (IONPs) have been developed as contrast agents for T1- or T2-weighted magnetic resonance imaging (MRI) on account of their excellent physicochemical and biological properties. However, general strategies to improve longitudinal relaxivity (r1) often decrease transverse relaxivity (r2), thus synchronously strengthening the T1 and T2 enhancement effect of IONPs remains a challenge. Here, we report interface regulation and size tailoring of a group of FePt@Fe3O4 core–shell nanoparticles (NPs), which possess high r1 and r2 relaxivities. The increase of r1 and r2 is due to the enhancement of the saturation magnetization (Ms), which is a result of the strengthened exchange coupling across the core–shell interface. In vivo subcutaneous tumor study and brain glioma imaging revealed that FePt@Fe3O4 NPs can serve as a favorable T1–T2 dual-modal contrast agent. We envision that the core–shell NPs, through interface engineering, have great potential in preclinical and clinical MRI applications.

Two new mid-infrared (MIR) nonlinear optical (NLO) chalcogenide crystals, Ba10Zn7In6S26 (1) and Ba10Zn7Ga6Se26 (2) were discovered by traditional solid state reactions. Compounds 1 and 2 are isostructural, and they represent a novel three-dimensional (3D) framework made by apex-sharing T2 supertetrahedron 2D layers that are stacked alternately by another kind of T2 supertetrahedra though sharing apexes along the c direction, with T1 tetrahedra inserting in the tunnels and Ba2+ cations locating both in the voids and in the tunnels. Among them, 1 shows attractive MIR NLO properties with a wide band gap of 3.0 eV, a strong second harmonic generation (SHG) signal about 4.1 times that of the commercial AgGaS2 at particle sizes of 30–46 μm under a 2.05 μm laser radiation, and a high laser damage threshold (LDT) of 37.2 times that of benchmark AgGaS2 at particle sizes of 150–210 μm under a 1.064 μm laser radiation. This distinguishes 1 as a new promising MIR NLO material. In addition, the density functional theory (DFT) analyses suggest that the SHG response of 1 comes mainly from the electronic transitions from S 3p to Ba 5d, Zn 4s, Zn 3d, In 5s, and In 5p states.

The use of multiparametric magnetic resonance imaging (MRI) to distinguish complex histopathological changes in liver fibrosis has not yet been systematically established. The purpose of this study is to gauge the efficacy of a cutting-edge MRI platform for evaluating ecotoxicologically hazardous carbon tetrachloride (CCl 4 ) induced liver fibrosis, while also scrutinizing the relationship between MRI and its histopathological features. Thirty-six mice were randomly divided into 6 groups, each with 6 mice. Control mice received an intraperitoneal injection of olive oil, while the experimental mice received different doses of intraperitoneal injection of CCl 4 . Both sets underwent this process twice per week over a duration of 5 weeks. MRI measurements encompassed T1WI, T2WI, T1 mapping, T2 mapping, T2* mapping. Liver fibrosis and inflammation were assessed and classified using Metavir and activity scoring systems. CCl 4 successfully induced liver fibrosis in mice, showing an increasing extent of liver fibrosis and liver function damage with the increasing dosage of CCl 4 . Compared with the control group, T1, ΔT1, and T2 in the experimental group were considerably elevated (P < 0.05) than those in the control group. Spearman's correlation showed that the correlation of Native T1 and △T1 with fibrosis (r = 0.712, 0.678) was better than with inflammation (r = 0.688, 0.536). T2 correlation with inflammation (r = 0.803) was superior to fibrosis (r = 0.568). ROC analysis showed that the AUC of Native T1 was highest (0.906), followed by ΔT1 (0.852), while the AUC increased to 0.945 when all relevant MRI parameters were combined. T1 is the most potent MRI parameter for evaluating CCl 4 -induced liver fibrosis, followed by ΔT1. Meanwhile, T2 may not be suitable for evaluating liver fibrosis but is more suitable for evaluating liver inflammation.

The imaging resolution of magnetic resonance imaging (MRI) is influenced by many factors. The development of more effective MRI contrast agents (CAs) is significant for early tumor detection and radical treatment, albeit challenging. In this work, the Hofmeister effect of Fe2O3 nanoparticles within the tumor microenvironment was confirmed for the first time. Based on this discovery, we designed a nanocomposite (FePN) by loading Fe2O3 nanoparticles on black phosphorus nanosheets. After reacting with glutathione, the FePN will undergo two stages in the tumor microenvironment, resulting in the robust enhancement of r1 and r2 based on the Hofmeister effect in the commonly used magnetic field (3.0 T). The glutathione-activated MRI signal of FePN was higher than most of the activatable MRI CAs, enabling a more robust visualization of tumors. Furthermore, benefiting from the long circulation time of FePN in the blood and retention time in tumors, the synergistic therapy of FePN exhibited an outstanding inhibition toward tumors. The FePN with good biosafety and biocompatibility will not only pave a new way for designing a common magnetic field-tailored T1–T2 dual-mode MRI CA but also offer a novel pattern for the accurate clinical diagnosis and therapy of tumors.

Superparamagnetic iron oxides (SPIO) are attractive MRI contrast agents but suffer from limited sensitivity and low tumor specificity. In this study, two sets of SPIO nanoparticles (SPIONPs) were synthesized that were designed to selectively crosslink upon sensing transglutaminase (TGase), which is highly expressed at tumor site, thereby leading to the formation of SPIO nanocluster with T2 signal enhancement properties. For this purpose, SPIONPs with glutamine and lysine surfaces were synthesized. In vitro studies showed that the SPIONPs could crosslink to form aggregates with diameter of 1.3 μm in the presence of TGase and showed a 1.8 fold enhancement in r2 relaxivity compared with the dispersed state. The cellular uptake results illustrated that the SPIONPs could crosslink in high TGase expressing HeLa cells and showed an aggregation-induced retention effect compared with the controlled MCF-7 cell-line (about 1.9 fold). Simultaneously, the administration of the SPIONPs in the HeLa tumor-bearing mice verified the TGase-responsive aggregation and tumor enrichment of the nano contrast agent. MRI imaging also demonstrated the T2 signal enhancing ability of the TGase-responsive SPIONPs; the ΔT2 signal of the SPIONPs in the HeLa tumor-bearing group was about 2.3 fold higher than the MCF-7 group and the TGase-inhibitor treated group.

Hepatocellular carcinoma (HCC) remains one of the leading
causes
of cancer-related mortality worldwide, highlighting the urgent need
for safe and effective imaging tools to improve early diagnosis. In
this study, we developed a hepatocyte-targeted T2 MRI contrast agent
by functionalizing superparamagnetic iron oxide nanoparticles (SPIONs)
with an ethoxybenzyl-polyethylene glycol ligand, yielding Fe 3 O 4 -EOB-PEG 2000 NPs, inspired by the hepatocyte-selective
features of Gd-EOB-DTPA. In vitro studies demonstrated preferential
uptake of Fe 3 O 4 -EOB-PEG 2000 NPs by
hepatocytes over tumor cells, while in vivo imaging in both hepatocellular
carcinoma and colorectal liver metastasis models revealed a distinct
“black-liver/bright-tumor” contrast effect. Compared
with nontargeted Fe 3 O 4 -mPEG 2000 NPs,
Fe 3 O 4 -EOB-PEG 2000 NPs showed significantly
improved tumor visibility in T2-weighted images. Furthermore, Fe 3 O 4 -EOB-PEG 2000 NPs exhibited good biocompatibility
and biosafety. This work highlights a strategy for enhancing T2 MRI
contrast through hepatocyte targeting, providing a promising gadolinium-free
alternative for safe and effective safe and effective diagnosis of
liver tumors.

In this study, spinel-phase Zn 2 Ga 3- x Fe 3+ x Ge 0.75 O 8 ( x = 0, 0.002, 0.004, 0.02, 0.04, 0.08, 0.2) and Zn 2 Ga 2.98- x Fe 3+ x Cr 0.02 Ge 0.75 O 8 ( x = 0, 0.0004, 0.001, 0.002 0.003, 0.004) nanoparticles were prepared by one-step hydrothermal method in combination with vacuum annealing. For ZGGO: Fe 3+ nanoparticles , the near infrared (NIR) emissions (∼725 nm) originate from the 4 T 1 ( 4 G)→ 6 A 1 ( 6 S) transition of Fe 3+ occupying octahedral and tetrahedral sites of Ga 3+ . Furthermore, their afterglow time is more than 15 min and Fe 3+ doping will influence the generation of antisite defects pair ( Ga Zn 0 − Zn Ga ′ ). For Cr 3+ and Fe 3+ co-doped ZGGO nanoparticles with average size of ∼46 nm, the NIR persistent luminescence around ∼700 nm, originating from the 2 E→ 4 A 2 and 4 T 2 → 4 A 2 transitions of Cr 3+ , can be observed and the afterglow time exceeds 850 min. With the increase of Fe 3+ concentration, the afterglow intensity decreases due to the decreased number of ( Ga Zn 0 − Zn Ga ′ ) defects and deeper traps. In particular, autofluorescence-free imaging and MR imaging were successfully realized by using ZGGO: Cr 3+ , Fe 3+ 0.002 nanoparticles dispersed in the simulated biological fluid environments (H 2 O, SLS and HSA). Their clear afterglow images can be acquired within 3 min after the stoppage of the UV irradiation and their transversal relaxivity (r 2 ) is in the range of 227–140 mM −1 s −1 , which is close to the conventional Feridex T 2 agent (108 mM −1 s −1 ). Our results suggest that ZGGO: Cr 3+ , Fe 3+ nanoprobes have potential applications in the diagnosis and therapy of disease.

A lot of nanomaterials are using T1-T2dual mode magnetic resonance (MR) contrast agents (CAs), but multilayer nanowire (NW) with iron (Fe) and manganese (Mn) as T1-T2dual modal CAs has not been reported yet. Herein, we synthesized a Fe/Mn multilayer NW with an adjustable Fe layer, as T1-T2dual-mode CAs. The relaxation performance of Fe/Mn multilayer NW was studied at 1.5 T. Results show that, when the length of the Fe layer is about 10 nm and the Mn is about 5 nm, the r1value (21.8 mM−1s−1) and r2value (74.8 mM−1s−1) of the Fe/Mn multilayer NW are higher than that of Mn NW (3.7 mM−1s−1) and Fe NW (59.3 mM−1s−1), respectively. We predict that our Fe/Mn multilayer NW could be used as T1-T2dual mode MRI CAs in the near future. Keywords:dual modal;MRI;CAs;nanowire;relaxation

The development of nanoplatforms with integrated therapeutic and imaging functions is necessary for highly efficient cancer therapy. Herein, 3D CuS hollow nanoflowers (HNs) consisting of 2D nanoplates are successfully fabricated through the technique of laser ablation in liquids followed by ion-exchange reactions and applied for the first time as a theranostic nanoagent for magnetic resonance imaging (MRI), photothermal therapy (PTT), and chemotherapy simultaneously. Due to the sufficient and immediate contact between the exposed cupric centers of nanoplates and protons from water molecules, CuS HNs are demonstrated to be capable of being a T1 positive contrast agent for efficient MRI of tumors on the T2-weighted fluid-attenuated inversion recovery imaging (T2-FLAIR) sequence. Besides, the hollow structure makes CuS HNs an efficient nanoplatform for drug loading with a laser-triggered drug release. Moreover, CuS HNs exhibit high photothermal conversion efficiency (30%) and good biocompatibility. The combination of PTT and chemotherapy with CuS HNs provides a significant synergistic therapeutic effect, resulting in a higher tumor inhibition ratio than PTT or chemotherapy alone. This study demonstrates a single-component multifunctional theranostic nanoagent for T2-FLAIR MRI guided thermochemotherapy, which has great potential application in theranostics of cancer.

Due to no penetration depth limitation, low cost, and easy control, magnetic nanoparticles mediated magnetic hyperthermia therapy (MHT) has shown great potential in experimental and clinal treatments of various diseases. However, the low heating conversion efficiencies and short circulation times are major drawback for most existing magnetic-thermal materials. Additionally, single MHT treatment always leads to resistance and recurrence. Herein, a highly efficient magnetic-thermal conversion, ferrimagnetic vortex nanoring Fe3O4 coated with hyaluronic acid (HA) nanoparticles (Fe3O4@HA, FVNH NPs) was firstly constructed. Additionally, the doxorubicin (DOX) was successfully enclosed inside the FVNH and released remotely for synergetic magnetic–thermal/chemo cancer therapy. Due to the ferrimagnetic vortex-domain state, the ring shape Fe3O4 displays a high specific absorption rate (SAR) under an external alternating magnetic field (AMF). Additionally, antitumor drug (DOX) can be encapsulated inside the single large hole of FVNH by the hyaluronic acid (HA) shell and quickly released in response the tumor acidic microenvironments and AMF. What’s more, the non-loaded FVNH NPs show good biocompatibility but high cytotoxicity after loading DOX under AMF. Furthermore, the synthesized FVNH can efficiently reduce the transverse relaxation time and enhance negative magnetic resonance imaging (MRI). The impressive in vivo systemic therapeutic efficacy of FVNH was also proved in this work. Taken together, the results of this study demonstrate that the synthesized FVNH NPs offer the promise of serving as multifunctional theranostic nanoplatforms for medical imaging-guided tumor therapies.
