Typical halogenated flame retardants affect human neural stem cell gene expression during proliferation and differentiation via glycogen synthase kinase 3 beta and T3 signaling

Shaojun Liang,Shengxian Liang,Hui Zhou,Nuoya Yin,Francesco Faiola

2′,2′,4,4′-tetrabromo diphenyl ether (BDE-47), one of the most abundant congeners of commercial pentaBDE utilized as flame retardants , has been phased out of production due to its potential neural toxicity and endocrine disrupting activities, and yet still present in the environment. Several alternatives to BDE-47, including tetrabromobisphenol A (TBBPA), tetrabromobisphenol S (TBBPS), tetrachlorobisphenol A (TCBPA) and decabromodiphenyl ether (BDE-209), are presently employed without restrictions and their potential toxic effects on human neural development are still unclear. In this study, we utilized a human neural stem cell (hNSC)-based system to evaluate the potential developmental neurotoxic effects of the above-mentioned five chemicals, at environment and human exposure relevant concentrations. We found that those compounds slightly altered the expression of hNSC identity markers ( SOX2 , SOX3 and NES ), without impairing cell viability or proliferation, in part by either modulating glycogen synthase kinase 3 beta (GSK3β) signaling (TBBPS, TCBPA and BDE-47), and slightly disturbing the NOTCH pathway (TBBPA, TBBPS and TCBPA). Moreover, the five chemicals seemed to alter hNSC differentiation by perturbing triiodothyronine (T3) cellular signaling. Thus, our findings suggest that the five compounds, especially TBBPS, TCBPA, and BDE-47, may affect hNSC self-renewal and differentiation abilities and potentially elicit neural developmental toxicity.

Olfactory toxicity of tetrabromobisphenol A to the goldfish Carassius auratus

Lingzheng Lu,Conghui Shan,Difei Tong,Yingying Yu,Weixia Zhang,Xunyi Zhang,Yang Shu,Weifeng Li,Guangxu Liu,Wei Shi

Tetrabromobisphenol A (TBBPA) is one of the most extensively used brominated flame retardants and its increasing use in consumer products has raised concerns about its ecotoxicity. Given the ubiquity of TBBPA in aquatic environments, it is inevitable that these chemicals will enter the olfactory chambers of fish via water currents. Nevertheless, the olfactory toxicity of TBBPA to aquatic organisms and the underlying toxic mechanisms have yet to be elucidated. Therefore, we investigated the olfactory toxicity of TBBPA in the goldfish Carassius auratus , a model organism widely used in sensory biology. Results showed that exposure to TBBPA resulted in abnormal olfactory-mediated behaviors and diminished electro-olfactogram (EOG) responses, indicating reduced olfactory acuity. To uncover the underlying mechanisms of action, we examined the structural integrity of the olfactory epithelium (OE), expression levels of olfactory G protein-coupled receptors (GPCRs), enzymatic activities of ion transporters, and fluctuations in neurotransmitters. Additionally, comparative transcriptomic analysis was employed to investigate the molecular mechanisms further. Our study demonstrates for the first time that TBBPA at environmentally relevant levels can adversely affect the olfactory sensitivity of aquatic organisms by interfering with the transmission of aqueous stimuli to olfactory receptors, impeding the binding of odorants to their receptors, disrupting the olfactory signal transduction pathway, and ultimately affecting the generation of action potentials.

Ecotoxicity of Caenorhabditis elegans following a step and repeated chronic exposure to tetrabromobisphenol A

Fuwen Liu,Waqas Qamar Zaman,Hongjiang Peng,Chao Li,Xue Cao,Kai Huang,Changzheng Cui,Wei Zhang,Kuangfei Lin,Qishi Luo

To better understand the toxicity of tetrabromobisphenol A (TBBPA), its effects on the model nematode Caenorhabditis elegans were investigated. Following a step and repeated chronic exposure from L4-larvae to day-10 adult, physiology endpoints (growth and locomotion behaviors including head thrashes, body bends and pumping rate), biochemical endpoints (reactive oxygen species, superoxide dismutase activity, catalase activity), and molecular stress-related gene expression were tested at environmentally relevant concentrations of TBBPA (0.01–100 µg/L). The results showed that concentrations of TBBPA greater than 10 µg/L, clearly influenced the physiology behaviors (growth and locomotion endpoints). Under repeated exposure, C. elegans exhibited adaptive responses in head thrashes and pumping rate. Compared to toxicity evaluation following repeated chronic exposure, a significantly greater response was induced at the same concentration following a step chronic exposure. Reactive oxygen species production was significantly enhanced following a step and repeated TBBPA exposure at the concentrations of 1 and 10 µg/L, respectively. qRT-PCR showed that ctl-1 , ctl-2 , ctl-3 and sod-3 expression significantly increased, which was obviously correlated with physiological and biochemical behaviors under both treatment conditions according to Pearson correlation test analysis. sod-3 and ctl-2 mutations were more sensitive than the wild-type N2 under a step chronic TBBPA exposure at a level of 10 µg/L. Thus, chronic exposure to TBBPA induces an oxidative stress response in C. elegans , with ctl-2 and sod-3 playing a vital role in TBBPA-induced toxicity in nematodes.

Highly sensitive and selective fluorescent sensor for tetrabromobisphenol-A in electronic waste samples using molecularly imprinted polymer coated quantum dots

Jingwen Feng,Yun Tao,Xiaoli Shen,Hua Jin,Tingting Zhou,Yusun Zhou,Liqin Hu,Dan Luo,Surong Mei,Yong-Ill Lee

In this study, an optosensing composite was fabricated by coating a molecularly imprinted polymer (MIP) layer onto CdTe quantum dots (MIP-QDs) to develop a fluorescent sensor for the determination of tetrabromobisphenol-A (TBBPA). The resulting MIP-QDs composite was well characterized and had ideal properties of morphology and photoluminescence . Under the optimized experiment conditions, this fluorescent sensor exhibited a good linearity with TBBPA covering the concentration range of 1.0–60.0 ng/mL and the detection limit was as low as 3.6 ng/g. This developed MIP-QDs sensor was successfully employed to detect TBBPA in electronic waste (e-waste) samples for the first time. The mean recoveries were in the range from 89.6% to 107.9% with the relative standard deviation below 6.2%, which were in accordance with the results of HPLC-UV method. The average concentration of TBBPA in electric fan and circuit board samples were 260.20 and 707.30 mg/kg, respectively. This study provided a simple, rapid, highly sensitive and selective method to detect TBBPA in e-waste samples.

Toxic responses of freshwater microalgae Chlorella sorokiniana due to exposure of flame retardants

Dongyang Liu,Mingxiang Qv,Dian Dai,Xu Wang,Liandong Zhu

Flame retardants , such as Tetrabromobisphenol A (TBBPA), Tris(1,3-dichloro-2-propyl) phosphate (TDCPP) and tributyl phosphate (TBP), are frequently detected in surface water. However, the effects of FRs exposure on aquatic organisms especially freshwater microalgae are still unclear. In this study, the toxicities of TBBPA, TDCPP and TBP to microalgae Chlorella sorokiniana , in terms of growth inhibition, photosynthetic activity inhibition and oxidative damage, were investigated, and according ecological risks were assessed. The results showed that TBBPA, TDCPP and TBP had inhibitory effects on C. sorokiniana , with 96 h EC 50 (concentration for 50% of maximal effect) values of 7.606, 41.794 and 49.996 mg/L, respectively. Fv / Fm decreased as the increase of exposure time under 15 mg/L TBBPA. Under 50 mg/L TDCPP and 80 mg/L TBP exposure, Fv / Fm decreased significantly after 24 h. However, Fv / Fm rose after 96 h, indicating that the damaged photosynthetic activity was reversible. The content of chlorophyll a decreased, as the increase of TBBPA concentration from 3 to 15 mg/L. However, chlorophyll a increased first and then decreased, as the increase of TDCPP and TBP concentrations from 0 to 50 mg/L and 0–80 mg/L, respectively. Results indicated that C. sorokiniana could use the phosphorus of TDCPP and TBP to ensure the production of chlorophyll a . The risen content of reactive oxygen species , malondialdehyde as well as superoxide dismutase activity indicated that exposure to FRs induced oxidative stress . Additionally, the risk quotients showed that tested FRs had ecological risks in natural waters or wastewaters. This study provides insights into the toxicological mechanisms of different FRs toward freshwater microalgae for better understanding of according environmental risks.
