N-Acetyl cysteine (NAC)-mediated reinforcement of alpha-tricalcium phosphate/silk fibroin (α-TCP/SF) cement

Tao Feng,Bin Pi,Bin Li,Lei Jiang,Yi-Meng Wang,Xue-Song Zhu,Hui-Lin Yang

Calcium phosphate cements (CPCs) are popular bone filling materials and drug carriers. However poor mechanical properties and lack of osteoinduction restrict their clinical applications. Recent studies suggested the osteogenic properties of NAC. In our study, we incorporated NAC with α-TCP/SF. We found that the compressive strength of α-TCP/SF-NAC composites increased with increase in NAC concentration, possibly due to complex three-dimensional networks of SF induced by NAC, which was large and chemically heterogeneous and induced compact oriented growth of HA crystals. However the setting time increased slightly with the addition of NAC, due to the ruptured disulfide bonds in SF. The α-TCP/SF-NAC composites also showed decent biocompatibility in vitro. As a result, these composites hold great potential as bone filling materials for clinical applications, including minimally invasive surgeries.

Weichang Li,Junpei Kang,Yulin Yuan,Fengcai Xiao,Hang Yao,Sa Liu,Jianxi Lu,Yingjun Wang,Zhen Wang,Li Ren

The poly(vinyl alcohol) (PVA) hydrogel is regarded as a potential articular cartilage replacement for its good biocompatibility , high permeability to fluid and load-bearing properties. This work investigated a novel Poly(vinyl alcohol)-polyetheretherketone/Poly(vinyl alcohol)-β-tricalcium phosphate (PVA-PEEK/PVA-β-TCP) bilayered hydrogels by freezing-thawing with biomimetic properties for articular cartilage and subchondral bone is developed. The bilayered hydrogels microarchitecture consists of a highly porous and dense structure, and the internal structure were analyzed by micro-CT. The morphology of the resulting hydrogels was analyzed by scanning electron microscopy (SEM), the enhancement of the mechanical properties of the PVA-PEEK/PVA-β-TCP bilayered hydrogels were demonstrated by mechanical testing. The bilayered structure indicate that a good bonding exist between the two layers, which is known to be a requisite necessary to assure a good integrity and functionality of the osteochondral construct. In addition, in vitro cell culture studies revealed that the hydrogels has no negative effect on the cell viability and proliferation and possess good biocompatibility. Then, the bilayered hydrogels were implanted into the knee joint defect of rabbits and hematoxylin and eosin and immunohistochemical staining. The PVA-PEEK/PVA-β-TCP bilayered hydrogels show good potential for use in the field of articular cartilage repair.

A Novel Immunoregulatory PEGylated Poly(glycerol sebacate)/β-TCP Membrane for Application in Guided Bone Regeneration

Hongzhou Shen,Chenglong Zhang,Changru Zhang,Jun Shi,Steve GF. Shen,Yuan Yuan,Jiawen Si

The recently developed poly(ethylene glycol)ylated poly(glycerol sebacate)/β-tricalcium phosphate (PEGS/β-TCP) membrane possesses favorable mechanical properties and excellent osteogenic activity. This study is designed to evaluate the feasibility of applying PEGS/β-TCP membrane in guided bone regeneration (GBR) through investigating the in vitro and in vivo activities of macrophages promoted by PEGS/β-TCP membranes during osteogenesis. The behavior of macrophages seeded on PEGS/β-TCP and PEGS membranes is assessed in terms of cell viability, polarization pattern, and release of inflammatory and osteogenic cytokines. The in vitro osteogenic effect of PEGS/β-TCP membrane-associated macrophage polarization is evaluated by incubating mesenchymal stem cells (MSCs) with conditioned medium derived from macrophages stimulated with PEGS/β-TCP or PEGS membranes. The in vivo correlation between macrophage polarization and PEGS/β-TCP membrane-driven osteogenesis is investigated using a rat calvarial defect model. The results show that PEGS/β-TCP and PEGS membranes are capable of downregulating M1 polarization and upregulating M2 polarization. Compared to the PEGS membrane, the PEGS/β-TCP membrane can further alleviate local inflammatory response and significantly promote the in vitro osteogenic differentiation of MSCs and in vivo restoration of bone defects. In conclusion, the novel PEGS/β-TCP membrane can promote the osteogenic commitment of MSCs and restore calvarial bone defects by improving the macrophage polarization pattern.

3D-printed BaTiO3-modified PCL/β-TCP scaffolds with multimodal optimization for enhanced critical-size bone regeneration

Liu Xuanhao,Zhang Jing,Xu Biying,Xu Yang,Wang Yuxin,Huang Zhi,Liu Bin,Fuxiang Song

Multimodal optimization improved scaffold structure, mechanics, and bioactivity. • Low-dose BT generated a detectable but mild local piezoelectric response. • PTB4 achieved the best overall balance of printability and biological performance. • PTB4 enhanced osteogenic differentiation and VEGF-related endothelial responses. • PTB4 promoted osteogenesis–angiogenesis coupling and calvarial bone regeneration. Critical-size bone defects (CSDs) remain a major clinical challenge. Although three-dimensional (3D) printing enables precise structural control and defect-specific scaffold fabrication, conventional polymer-based scaffolds often exhibit insufficient mechanical strength and limited osteogenic activity. In this study, poly(ε-caprolactone)/β-tricalcium phosphate (PCL/β-TCP) scaffolds containing 0, 1, 2, 4, or 8 wt% barium titanate (BaTiO 3 , BT) were fabricated by direct ink writing. Their rheology, microstructure, mechanical properties, local electromechanical response, cytocompatibility, angiogenesis-related activity, osteogenic differentiation, and bone regeneration in vivo were evaluated. All formulations exhibited suitable printability and regular porous structures. Moderate BT incorporation increased surface roughness and mechanical strength, with compressive strength remaining within the range of human cancellous bone. Piezoresponse force microscopy confirmed a detectable but mild local piezoelectric response. However, its direct osteogenic contribution was not isolated from other BT-related material effects and requires further verification. All scaffolds exhibited good cytocompatibility. PTB4 showed the most favorable biological performance, including enhanced MC3T3-E1 cell proliferation, increased ALP-positive area, greater mineralized deposition, and more pronounced VEGF-related fluorescence in HUVECs. After 12 weeks in a rat calvarial critical-size defect model, PTB4 exhibited significantly higher bone volume fraction and bone mineral density than the blank and PT groups. Histological and immunohistochemical analyses showed more extensive new bone formation, collagen deposition, and stronger osteogenesis–angiogenesis coupling. Overall, PTB4 achieved the best balance among printability, mechanical compatibility, cytocompatibility, angiogenesis-related activity, and osteogenic performance, supporting low-dose BT modification as a promising strategy for safe, printable scaffolds for critical-size craniofacial and oral bone defect repair. Biodegradation of TCP in a Sequencing Batch-Fluidized Bed Bioreactor with Waste Coke Particles as the Carrier

Fangming Hu,Huihui Zhou,Zanfang Jin,Qin Sun,Zhiyan Pan,Jesse Zhu

This study investigated the biodegradation of synthetic wastewater containing 2,4,6-trichlorophenol (TCP) in a sequencing batch-fluidized bed bioreactor (SBFBBR) with waste coke particles (WCPs) as the biofilm carrier. The irregular surfaces of WCPs (diameter range from 0.6 to 2.3 mm) rendered them ideal biomass carriers. The attached biomass was 4.0–4.5 mg volatile suspended solids (VSS)/g WCPs 3 weeks after reactor start-up, and the biofilm thicknesses were 35–110 μm. Biodegradation, not adsorption-degradation or volatilization-degradation, was found to be the main TCP removal mechanism. At the initial chemical oxygen demand (COD) concentration of 300  mg L−1, removal of COD was more efficient and faster than that of TCP. The air-water ratio (ranging from 0.280 to 0.58  min−1) exerted negligible effects at dissolved oxygen (DO) concentrations above 2  mg L−1. Maximum TCP degradation (62.1% after 3 h retention time and 75.8% after 6 h) occurred at initial COD concentration of 45  mg L−1. TCP can be removed by biofilm as a sole source of carbon and energy. Furthermore, the TCP biodegradation follows zero-order reaction kinetics, with the zero-order specific biodegradation rate dependent on initial COD concentration.

Isomer-specific cardiotoxicity induced by tricresyl phosphate in zebrafish embryos/larvae

Xun’e Yi,Haiyu Qin,Guangyu Li,Ren Kong,Chunsheng Liu

Tricresyl phosphate (TCP) has received extensive attentions due to its potential adverse effects, while the toxicological information of TCP isomers is limited. In this study, 2 h post-fertilization zebrafish embryos were exposed to tri-o-cresyl phosphate (ToCP), tri-m-cresyl phosphate (TmCP) or tri-p-cresyl phosphate (TpCP) at concentrations of 0, 100, 300 and 600 μg/L until 120 hpf, and the cardiotoxicity and mechanism of TCP isomers in zebrafish embryos/larvae were evaluated. The results showed that ToCP or TmCP exposure induced cardiac morphological defects and dysfunction in zebrafish, characterized by increased distance between sinus venosus and bulbus arteriosis, increased atrium and pericardial sac area, trabecular defects, and decreased heart rate and blood flow velocity, while no adverse effects of TpCP on zebrafish heart were found. Transcriptomic results revealed that extracellular matrix (ECM) and motor proteins, as well as PPAR signaling pathways, were included in the cardiac morphological defects and dysfunction induced by ToCP and TmCP. Co-exposure test with D-mannitol indicated that the inhibition of energy metabolism by ToCP and TmCP affected cardiac morphology and function by decreasing osmoregulation. This study is the first to report the cardiotoxicity induced by TCP in zebrafish from an isomer perspective, providing a new insight into the toxicity of TCP isomers and highlighting the importance of evaluating the toxicity of different isomers.

3D-printed Strontium-incorporated β-TCP Bioceramic Triply Periodic Minimal Surface Scaffolds with Enhanced Angiogenic and Osteogenic Properties

Shan Yanbo,Bai Yang,Zhao Lisheng,Zhou Qing,Yang Shuo,Wang Gang,Lei Ye,Lu Yuzheng,Wu Yanbin,Wei Yu,Peng Jiang,He Rujie,Wen Ning,Gu Bin

Reconstructing bone defects remains a significant challenge in clinical practice, driving the urgent need for advanced artificial grafts that simultaneously promote vascularization and osteogenesis. Addressing the critical trade-off between achieving high porosity/strength and effective bioactivity at safe ion doses, we incorporated strontium (Sr) into β-tricalcium phosphate (β-TCP) scaffolds with a triply periodic minimal surface (TPMS) structure using digital light processing (DLP)-based three-dimensional (3D) printing. Systematically screening Sr concentrations (0-10 mol%), we identified 10 mol% as optimal, leveraging the synergy between the biomimetic TPMS architecture, providing exceptional mechanical strength (up to 1.44 MPa at 80% porosity) and facilitating cell recruitment, and precision Sr-dosing to enhance bioactivity. In vitro assays revealed that the Sr-TCP scaffold dose-dependently stimulated osteogenic differentiation and mineralization in mouse osteoblastic cell line (MC3T3-E1) cells, while also significantly enhancing the angiogenic capacity in human umbilical vein endothelial cells (HUVECs). In vivo studies indicated that the scaffold demonstrated synergistic osteogenic and angiogenic effects in rat femoral condylar defects, leading to marked improvements in bone healing. Collectively, this study establishes a novel design paradigm combining biomimetic topology with optimized ionic doping, resolving key limitations of conventional grafts and advancing the development of safe, highly effective biomaterials for vascularized bone regeneration.

Effects of solidification cooling rate on the corrosion resistance of a biodegradable β-TCP/Mg-Zn-Ca composite

Qiang Yuan,Y. Huang,Debao Liu,Minfang Chen

Biodegradable beta-tricalcium phosphate (β-TCP) particle reinforced magnesium metal matrix composites (Mg-MMC) have attracted increasing interest for application as implant materials . This investigation was conducted to study the effect of cooling rate on the microstructure and corrosion behavior of a biodegradable β-TCP/Mg-Zn-Ca composite. The composite was fabricated under a series of cooling rates using a wedge-shaped casting mold. The microstructure of the composite was examined by optical and scanning electron microscopy, and the corrosion behavior was investigated using an electrochemical workstation and immersion tests in a simulated body fluid (SBF). Faster cooling rates were shown to refine the secondary phase and grain size, and produce a more homogenous microstructure. The refined microstructure resulted in a more uniform distribution of β-TCP particles, which is believed to be beneficial in the formation of a stable and compact corrosion product layer, leading to improved corrosion resistance for the composite.

A mechanically robust and flexible PEGylated poly(glycerol sebacate)/β-TCP nanoparticle composite membrane for guided bone regeneration

Shuang Yu,Jun Shi,Yutong Liu,Jiawen Si,Yuan Yuan,Changsheng Liu

The treatment of bone defects in the field of periodontology and oral implantology has been a major clinical problem to date. Guided bone regeneration (GBR), based on ideal barrier membranes to provide space maintenance and selectively promote the regeneration of the damaged area by prevention of invasion by the fibrous tissues, represents the most promising strategy at present. In this study, the mechanically robust and flexible membrane PEGylated poly(glycerol sebacate) (PEGS) coordinated by β-TCP nanoparticles (PEGS/β-TCP) was developed by a simple prepolymer mixing-in situ crosslinking method. By tailoring the content of β-TCP (0, 10, 50, and 90 wt% relative to the prepolymer), the resultant PEGS/β-TCP composite membranes exhibited controllable degradation rate and reinforced mechanical properties. The maximum tensile strength of 9.58 ± 0.02 MPa was presented by the P20T50 group (weight ratio of β-TCP to PEGS20 prepolymer was 50%), about 1.5-fold higher than that of P20T0 without β-TCP. For cellular responses, the PEGS/β-TCP membranes showed desirable cell attachment and viability on rat bone mesenchymal stem cells (rBMSCs). The incorporation of β-TCP definitely enhanced the alkaline phosphatase activity and promoted mineralization, thus facilitating the osteogenic differentiation. The in vivo result obtained for the rat calvarial defect model reaffirmed the favorable bone regenerative ability of the fabricated membranes, especially for P20T50 with the highest bone volume/tissue volume ratio (BV/TV) at both 4 weeks (17.26 ± 1.49%) and 8 weeks (23.24 ± 2.85%) after the surgery. Therefore, the PEGS/β-TCP composite membranes prepared by this prepolymer mixing-in situ crosslinking process will be a prospective biomaterial for the GBR therapy.

Construction of a micro/nano structured surface on a β-TCP/CaSiO3 bioceramic promotes osteogenic differentiation of mBMSCs

Xiao Liu,Naru Zhao,Xiaoheng Guo,Haibo Duan,Jingjing Diao,Yifan Dong,Yingjun Wang

Cellular behavior is a result of homologous responses to biomaterials, especially material surfaces. An effective way to elicit a desired biological response is to employ micro/nanostructured constructions for biomaterial surface design; however, the brittleness of inorganic materials usually hinders structure regulation. In this study, a micro/nanostructured surface was fabricated using a hydrothermal treatment that did not require crystal regulation reagents. The precursor, β-tricalcium phosphate, was mixed with CaSiO3. The resulting micro/nanostructured surface had a hybrid structure containing nanowires, microrods, and rod clusters. In vitro cellular proliferation, cytotoxicity and osteodifferentiation experiments indicated that the micro/nanostructured surfaces alleviated material cytotoxicity, improved cell proliferation, and effectively promoted osteogenic differentiation of mouse bone mesenchymal stem cells. These results suggest that this simplified surface treatment is an efficient way to improve cell biological response to inorganic biomaterials.

Three-dimensional-printed MPBI@β-TCP scaffold promotes bone regeneration and impedes osteosarcoma under near-infrared laser irradiation

Liangjie Lu,Huihan Wang,Minjie Yang,Lijun Wang,Kaifeng Gan

Beta-tricalcium phosphate (β-TCP) is considered as one of the most promising biomaterials for bone reconstruction. This study generated a functional molybdenum disulfide (MoS 2 )/polydopamine (PDA)/−bone morphogenetic protein 2 (BMP2)-insulin-like growth factor-1 (IGF-1) coating on the β-TCP scaffold and analyzed the outcomes. The MoS 2 /PDA-BMP2-IGF-1@β-TCP (MPBI@β-TCP) scaffold was prepared by 3D printing and physical adsorption, followed by characterization to validate its successful construction. The in vitro osteogenic effect of the MPBI@β-TCP scaffold was evaluated. It was found that MPBI@β-TCP augmented the adhesion, diffusion and proliferation of mesenchymal stem cells (MSCs). The alkaline phosphatase (ALP) activity, collagen secretion and extracellular matrix (ECM) mineralization along with the expression of Runx2, ALP and OCN were also enhanced in the presence of MPBI@β-TCP. Additionally, MPBI@β-TCP stimulated endothelial cells to secrete VEGF and promoted capillary-like tubule formation. We then confirmed the biocompatibility of MPBI@β-TCP to macrophages and its anti-inflammatory effects. Furthermore, under near-infrared (NIR) laser irradiation, MPBI@β-TCP produced photothermal effect to not only kill MG-63 osteosarcoma cells, but also enhance bone regeneration in vivo with biosafety. Overall, this work demonstrates that 3D-printed MPBI@β-TCP with enhanced osteogenic activity under NIR laser irradiation has a vast potential in the field of tissue defects.

3D printed magnesium-doped β-TCP gyroid scaffold with osteogenesis, angiogenesis, immunomodulation properties and bone regeneration capability in vivo

Dahu Qi,Jin Su,Song Li,Hao Zhu,Lijin Cheng,Shuaibin Hua,Xi Yuan,Jiawei Jiang,Zixing Shu,Yusheng Shi,Jun Xiao

Bioceramics have been used in orthopedic surgery for several years. Magnesium (Mg) is an essential element in bone tissue and plays an important role in bone metabolism. Mg-doped bioceramics has attracted the attention of researchers recently. However, the optimal doping amount of Mg in β-TCP and the immunomodulatory property of Mg-doped β-TCP (Mg-TCP) have not been determined yet. In this study, β-TCP scaffolds doped with different contents of magnesium oxide (0 wt%, 1 wt%, 3 wt%, and 5 wt%) with gyroid structure were printed by digital light processing (DLP) method, and the physicochemical and biological functions were then investigated. Mg-doping improved the physicochemical properties of the β-TCP scaffolds. In vitro experiments confirmed that the doping of Mg in β-TCP scaffolds promoted the osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) and angiogenic differentiation of endothelial progenitor cells (EPCs), where the 5Mg-TCP has the optimal properties when using the “one cell type” method. It was also found that all Mg-TCP facilitated the polarization of RAW264.7 cells to the M2 phenotype, especially the 3Mg-TCP. However, 3Mg-TCP displayed the optimal osteogenic and angiogenic potential when using a “multiple cell type” method, which referred to culturing the BMSCs or EPCs in the macrophage-conditioned medium. Finally, the in vivo experiments were conducted and the results confirmed that the 3Mg-TCP scaffolds possessed the satisfying bone defect repair capability both after 6 and 12 weeks of implantation. This study suggests that 3Mg-TCP scaffolds provide the optimal biological performance and thus have the potential for clinical translation.
