A Marine-Derived Sterol, Ergosterol, Mitigates UVB-Induced Skin Photodamage via Dual Inhibition of NF-κB and MAPK Signaling

Junming Zhang,Jiangming Zhong,Yi Li,Qi Zhou,Zhiyun Du,Li Lin,Peng Shu,Ling Jiang,Wei Zhou

Background: Ultraviolet B (UVB) radiation induces oxidative stress, inflammation, and collagen degradation in skin, leading to photodamage. Ergosterol (ERG)—a sterol widely distributed in fungi and algae, including numerous marine species—possesses antioxidant and anti-inflammatory activities, but its photoprotective mechanisms remain unclear. Methods: Using integratedin vitro(UVB-irradiated human keratinocytes) andin vivo(topical ERG in a murine UVB model) approaches, combined with transcriptomic and network pharmacology analyses, we evaluated ERG’s effects on oxidative stress, inflammation, and extracellular matrix integrity. Results: ERG treatment preserved keratinocyte viability, reduced reactive oxygen species, and suppressed pro-inflammatory mediators after UVB exposure. In mice, topical ERG significantly attenuated epidermal hyperplasia, maintained tight-junction integrity, and inhibited collagen matrix degradation. Mechanistically, ERG exerted dual inhibition of the nuclear factor kappa B (NF-κB) pathway, which mediates inflammation, and the mitogen-activated protein kinase (MAPK) pathway, which regulates collagen degradation. Conclusions: These findings identify ERG as a marine-derived sterol with potent photoprotective activity that simultaneously targets oxidative stress, inflammation, and extracellular matrix damage, highlighting its promise as a natural compound for dermatological applications and aligning with ongoing efforts to explore marine-derived agents against skin oxidative stress and inflammation.

A Dual Network Hydrogel Sunscreen Based on Poly-γ-glutamic Acid/Tannic Acid Demonstrates Excellent Anti-UV, Self-Recovery, and Skin-Integration Capacities

Rui Wang,Xuexue Wang,Yijing Zhan,Zheng Xu,Zongqi Xu,Xiaohai Feng,Sha Li,Hong Xu

Novel sunscreen products based on bioadhesive/gel systems that can prevent the skin penetration behaviors of UV filters have attracted increasing attention in recent years. However, integration is very difficult to achieve and control on the wet surface of the skin under sweaty/dynamic physiological conditions, resulting in functional failure. Herein, we demonstrated the fabrication of a novel dual-network hydrogel sunscreen (DNHS) based on poly-γ-glutamic acid (γ-PGA) and tannic acid (TA), which demonstrated prominent UV protection properties across broad UVA and UVB regions (360–275 nm). Due to a three-dimensional network microstructure and a highly hydrated nature that mimics the extracellular matrix of natural skin, DNHS can perfectly match the skin surface without irritation and sensitization. In addition, the intermolecular hydrogen bond interactions of γ-PGA and TA provide an important driving force for coacervation, which endows the DNHS with remarkable self-recovery properties (within 60 s). Moreover, due to the multiple interfacial interactions between γ-PGA/TA and the protein-rich skin tissue surfaces, DNHS simultaneously possesses excellent skin-integration and water-resistance capacities, and it can be readily removed on demand. Our results highlight the potential of the DNHS to be used in next-generation sunscreens by providing long-term and stable UV protection functions even under sweaty/dynamic physiological conditions.

Selenium deficiency sensitizes the skin for UVB-induced oxidative damage and inflammation which involved the activation of p38 MAPK signaling

Xiaokang Zhu,Mingdong Jiang,Erqun Song,Xiaoji Jiang,Yang Song

Ultraviolet B (UVB) radiation causes oxidative damage and inflammation, and ultimately increases the risk of skin carcinogenesis. Selenium is an essential trace element, previous studies indicated selenium deficiency impairs tissue antioxidant capacity in different experimental models. However, the synergistic effect of selenium deficiency and UVB radiation on skin damage is not clear. In the current study, our data revealed selenium deficiency resulted in further increases of reactive oxygen species (ROS), thiobarbituric acid reactive substances (TBARS) and phosphorylated H2AX levels, decreases of GSH level and antioxidant enzyme activities in UVB-irradiated mice. Selenium deficiency also exacerbated UVB-induced cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), tumor necrosis factor-alpha (TNF-α), interleukin-1beta (IL-1β) and IL-6 mRNA expressions. Mechanism studies indicated that UVB-induced p38 signaling was further elevated in the skin of mice maintained with selenium deficiency diet, compared with those maintained with selenium adequate diet. Our investigation suggested that selenium deficiency diet weakens the antioxidant capacity of UVB-irradiated mice skin, which sensitizes to UV radiation-induced oxidative damage and inflammation.

Ameliorating effects and mechanisms of tuna peptides on UVB-induced photoaging and melanogenesis targeting oxidative stress, apoptosis, and tyrosinase inactivation

Yu-Mei Wang,Yu-Hui Zeng,Shuo-Lei Zheng,Bin Wang,Chang-Feng Chi,Qi Zeng

Ultraviolet B (UVB) radiation causes skin photodamage and excessive melanogenesis, but effective multi-target protective agents remain limited. Antioxidant peptides derived from marine by-products are promising; however, it remains unclear whether specific peptides from skipjack tuna skin can simultaneously alleviate UVB-induced photodamage and inhibit melanogenesis, and the underlying molecular mechanisms are poorly defined. Therefore, the primary objective of this study was to systematically investigate the protective efficacy and molecular mechanisms of three antioxidant peptides derived from skipjack tuna skin-Met-Pro-Ala-Gly-Glu (MPAGE), Leu-Asn-Pro-Gly-Ala (LNPGA), and Gly-Thr-Gly-Pro-Ala (GTGPA)—against UVB-induced skin photodamage and excessive melanogenesis. We hypothesized that these peptides exert synergistic photoprotective and anti-melanogenic effects via multi-target regulation of Nrf2-mediated antioxidant pathways, inflammatory and apoptotic cascades, and tyrosinase inhibition. In UVB-irradiated HaCaT cells, MPAGE, LNPGA, and GTGPA significantly upregulate nuclear factor erythroid 2-related factor 2 (Nrf2) protein expression and promote its nuclear translocation, increasing the levels of NAD( P )H quinone dehydrogenase 1 (NQO1), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) levels, thereby scavenging reactive oxygen species (ROS), reducing malondialdehyde (MDA), and alleviating oxidative damage. Mechanistically, molecular docking predictions suggested that these peptides may activate Nrf2 by interacting with the Kelch domain of kelch-like ECH-associated protein 1 (Keap1) and competitively occupying its ligand-binding pockets. Additionally, the peptides also attenuated apoptosis by reducing bcl-2-associated X protein/b-cell lymphoma 2 (Bax/Bcl-2) ratio and cysteine-dependent aspartate-specific protease 3 (Caspase-3) level. Moreover, they potently inhibited the β -galactosidase and collagenase production and reduced matrix metalloproteinase-1 (MMP-1) and matrix metalloproteinase-3 (MMP-3) levels, suppressing UVB-induced collagen degradation and mitigating skin photoaging. Enzyme-Linked Immunosorbent Assay (ELISA) revealed a marked reduction in interleukin-6 (IL-6), interleukin-1 β (IL-1β), and tumor necrosis factor α (TNF-α) levels, demonstrating significant anti-inflammatory effects. Finally, MPAGE, LNPGA, and GTGPA exhibited dose-dependent inhibitory effects on melanogenesis in B16-F10 cells. In silico docking analysis further predicted that these peptides could deeply embed in the active pocket of tyrosinase by hydrogen bondings interactions (<6 Å) with key catalytic residues. In summary, this work provides the first comprehensive mechanistic characterization of MPAGE, LNPGA, and GTGPA, demonstrating that they alleviate UVB-induced photodamage through synergistic antioxidant, anti-aging, anti-inflammatory, and anti-apoptotic mechanisms, while potentially inhibiting melanogenesis through computationally predicted tyrosinase suppression. These findings provide a basis for further experimental validation of these peptides as multi-target skin-protective agents.

Repair Effects of Elastin Peptides From Different Tissue and Recombinant Sources on UVB-Induced Photoaging Damage in Human Dermal Fibroblasts

Hanqiang Liu,Yanhong Sun,Guangling Tian,Yanyan Qin,Xiangmin Meng,Renmin Li

To compare the repair effects of elastin peptides (EPs) from different tissue and recombinant sources on ultraviolet B (UVB)-induced photoaging damage in human dermal fibroblasts (HDFs), four EP preparations were evaluated: bovine ligament-derived elastin peptide (BL), bovine cardiovascular tissue-derived elastin peptide (BC), bonito-derived elastin peptide (Bon), and recombinant human elastin peptide (RH). These preparations represented two bovine tissue-derived EPs, one marine-derived EP, and one recombinant human EP. Their regulatory effects on extracellular matrix (ECM) degradation, inflammatory responses, and antioxidant-related indicators were systematically evaluated. The molecular weight distribution and amino acid composition of the four EPs were first characterized by gel permeation chromatography (GPC) and amino acid analysis. Subsequently, the UVB irradiation dose and working concentrations of EPs were determined using the Cell Counting Kit-8 (CCK-8) assay. The expression levels of matrix metalloproteinase-1 (MMP-1), matrix metalloproteinase-12 (MMP-12), type I collagen (COL I), elastin (ELN), tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and related genes were evaluated by enzyme-linked immunosorbent assay (ELISA) and quantitative real-time polymerase chain reaction (qPCR). The results demonstrated significant differences in molecular weight distribution and amino acid composition among the four EPs. RH exhibited the most concentrated molecular weight distribution, whereas Bon showed a relatively higher proportion of medium-molecular-weight fractions. BL and BC displayed relatively similar overall amino acid compositions. Functional evaluation further revealed distinct anti-photoaging characteristics among the four EPs. BL showed the most pronounced inhibitory effects on MMP-12 and TNF-α expression, BC exhibited the strongest effects in suppressing MMP-1 expression and enhancing antioxidant-related gene expression, Bon demonstrated the greatest restorative effects on COL I and collagen type I alpha 1 chain ( COL1A1 ) mRNA expression, and RH showed the most prominent effects in promoting ELN recovery and suppressing IL-6 expression. Collectively, EPs from different tissue and recombinant sources exhibited distinct functional preferences in the UVB-induced HDF photoaging model, which may be associated with their molecular weight distribution and amino acid composition characteristics. This study provides an experimental basis for the precise development and differential application of elastin peptide materials. Graphical

Structure–Activity Relationships of Cyclodextrin-Included Quercetin and Naringenin and the Protective Effect of the Inclusion Complexes against UVB-Induced Photodamage

Yingying Du,Lin Wang,Yi Ouyang,Qun Chen,Ziwei Liu,Zhimei Liu

Quercetin (Que) and naringenin (Nar) are natural compounds with potent antioxidant and anti-inflammatory activities, showing promise for alleviating ultraviolet B (UVB)-induced skin photoaging. The presence or absence of a C═C bond in the C-ring defines whether the compound is a flavonol or a flavanone. To overcome poor water solubility and low bioavailability of the two flavonoids, we prepared inclusion complexes using β-cyclodextrin (β-CD), hydroxyethyl-β-cyclodextrin (HE-β-CD), and hydroxypropyl-β-CD (HP-β-CD). These complexes leverage the cyclodextrins’ hydrophilic exterior and hydrophobic interior to encapsulate Que and Nar, respectively. Through integrated analyses─including molecular docking, phase solubility studies, characterization (UV, Fourier-transform infrared (FT-IR), X-ray diffraction (XRD), scanning electron microscopy (SEM), and NMR), solubility tests, and antioxidant (2,2-diphenyl-1-picrylhydrazyl (DPPH)) assays─we systematically elucidated the structure–activity relationships of these flavonoid–cyclodextrin interactions. The therapeutic potential was further evaluated in a UVB-induced mouse model of skin photoaging. Results confirmed the spontaneous formation of stable 1:1 molar ratio inclusion complexes between the flavonoids and cyclodextrins. The enhancement of complex stability, water solubility, and antioxidant activity consistently followed the order: HP-β-CD > HE-β-CD > β-CD. Owing to Que’s conjugated planar structure, it exhibited weaker binding to β-CD than did Nar. However, Que formed more stable complexes with HE-β-CD and HP-β-CD than Nar did. In vivo, the Que/HP-β-CD complex demonstrated superior efficacy, most effectively reducing malondialdehyde (MDA) levels, alleviating epidermal hyperplasia, and increasing collagen fiber content. Furthermore, the inclusion complexes amplified the regulatory effects of Que and Nar on mitogen-activated protein kinase (MAPK) and TGF-β/Smad pathways: they enhanced the downregulation of p-p38 and MMP-1 to inhibit collagen degradation and promoted the upregulation of Smad2/3 and COL1A1 to stimulate collagen synthesis. In conclusion, the β-cyclodextrin inclusion complexes of flavonoids, designed based on structure–activity relationship studies, significantly improve the compounds’ solubility, stability, and bioactivity. This leads to enhanced protective efficacy against UVB-induced photodamage through the concurrent modulation of multiple signaling pathways.

Warm temperature thermal stimulation enhanced X-ray induced narrow-band UVB afterglow as long time safe irradiation source for phototherapy of psoriasis

Wei Meng,Ya Gao,Xiaoyan Fu,Yuting Huang,Haonan Li,Guotao Sun,Wenzhi Sun,Jing Jin,Changsheng Wang,Hongwu Zhang

A NB-UVB long persistent luminescence phosphor Ca 2 GeO 4 : Gd 3+ was synthesized, and the remaining luminescence decay lasted for more than 12 h. • After heating treatment, the NB-UVB afterglow intensity of the flexible film increased by three times. • The heat generated by short-term sunlight exposure can also enhance the emission of NB-UVB afterglow. Narrow-band UVB (NB-UVB) is markedly efficacious option for the treatment of psoriasis. However, the NB-UVB therapy still requires specialized medical equipment to avoid the possible risk of skin injury, which has poor portability and then limits its further application. Here, we have prepared a flexible film containing long afterglow NB-UVB phosphor Ca 2 GeO 4 : Gd 3+ , which persistent NB-UVB afterglow emission at 314 nm can be regarded as long time safe irradiation source for the phototherapy of psoriasis. Our results have shown that CGO: Gd 3+ can continuously release NB-UVB photons for more than 12 h, indicating that long time mild UVB irradiation source can be realized. More importantly, after warm temperature treatment, the NB-UVB afterglow emissions of the flexible film are enhanced to 3 times stronger, indicative of the recovery of high phototherapy efficiency. Furthermore, short-time sunlight induced heat also can enhance the NB-UVB afterglow emission intensity obviously, indicating high phototherapy efficiency can be kept when the film is exposed to sunlight. This work provides a low-cost UVB phototherapy method with multiple thermal stimuli to enhance afterglow, which has great potential in the home treatment of psoriasis. Ca 2 GeO 4 : Gd 3+ film can continuously emit NB-UVB afterglow for more than 12 h, which can be considered as a long-term safe and portable irradiation source for psoriasis phototherapy. And after heat treatment, the NB-UVB afterglow emission of the flexible film was enhanced by three times. More importantly, short-term sunlight induced heat can significantly enhance the afterglow emission intensity of NB-UVB, indicating that the film can maintain high phototherapy efficiency when exposed to sunlight. Simultaneously regulating absorption capacities and antioxidant activities of four stilbene derivatives utilizing substitution effect: A theoretical and experimental study against UVB radiation

Yajie Zhang,Changjiao Shang,Chaofan Sun,Lingling Wang

With the continued depletion of the ozone layer, the sun protection consciousness of humans has gradually enhanced. Long-term ultraviolet (UV) rays exposure will lead to skin tanning, even skin cancer in severe cases, and generate free radicals to cause skin aging. To better protect human skin against UV rays, this work explores the absorption capacities and antioxidant activities of four stilbene derivatives (EHDB, EDMB, EAPD, and HPTP) through the computational chemistry method and DPPH radical scavenging experiment. The research results indicate that their absorption spectra cover the entire UV region, and can effectively protect against UVB radiation. Moreover, three prevailing antioxidant mechanisms: hydrogen atom transfer, sequential proton loss electron transfer, and single electron transfer followed by proton transfer mechanisms, were used to evaluate their antioxidant activities in the ground state. It can be concluded that the O 1 H 1 sites of EHDB and HPTP are the most active, and the SPLET mechanism is the most preferred for the four compounds in ethanol solvent. Furthermore, the DPPH radical scavenging experiment compensates for the theoretical calculation deficiency in the excited state, revealing that the EHDB and HPTP are the most suitable for sunscreen due to their excellent performance on antioxidant capacities, whether before or after sunlight. This work will facilitate EHDB and HPTP to be applied in sunscreen and provide a novel idea in sunscreen research.

Advanced technology based on Poly(deep eutectic solvent) core–shell nanomaterials enriched with Fructus Choerospondias phenols for efficient defense against UVB-induced ferroptosis

Luyao Xu,Xiujun Zhang,Wanyu Yang,Haixiang Li,Jiaqi Wang,Lan Wang,Xicheng He,Yan Wang,Xiaoxia Li

Plant phenolic compounds serve as significant natural antioxidants. However, traditional preparation methods face sustainability challenges. In this study, we synthesized a novel deep eutectic solvent (DES) modified core–shell nanomaterial composed of betaine, ethylene glycol and dopamine hydrochloride based on the spatial arrangement formed by the abundant weak interaction forces in the poly(deep eutectic solvent) (PDES). Utilizing the PDES magnetic nanomaterial and combined with ultrasound-assisted matrix solid phase dispersion (UA-MSPD), it was able to efficiently enrich Fructus Choerospondias phenols (FCP) up to 61.468 ± 0.405 mg/g. Density functional theory (DFT) was used to analyze the adsorption mechanism between the material and the target compounds, which involved weak interactions of electrostatics, hydrogen bonds, and π-π stacking, while maintaining the activity of phenols without destroying the molecular structure. Remarkably, the adsorption efficiency of the material remained above 80 % after six repetitions, indicating excellent reusability. FCPs were identified and analyzed using ultra-performance liquid chromatography-tandem triple quadrupole mass spectrometry (UPLC-Q-TOF-MS). The FCP prepared by Fe 3 O 4 @SiO 2 @PDES exhibited superior in vitro antioxidant performance, achieving enrichment and purification in one step. Cell experiment results showed that FCP could scavenge ROS, regulate cytokines, and reduce ferroptosis caused by ultraviolet radiation, making FCP have therapeutic value in practical applications. The technology opens up a new way for the green and sustainable production and enrichment of plant phenolic compounds for anti-aging treatment.

Photoprotective Effects of Dendrobium officinale Protein Hydrolysate Fractions Against UVB-Induced Photoaging Associated with Modulation of MAPK/NF-κB and TGF-β/Smad Signaling

Jinghan Zhang,Yue Sun,Jinhao Zheng,Can Yang,Mingshuo Yang,Liming Pan

Dendrobium officinalehas attracted increasing attention as a functional food because of its diverse biological activities; however, the photoprotective potential of its protein-derived peptides remains poorly understood. In this study,D. officinaleprotein hydrolysates were fractionated by ultrafiltration according to molecular weight, and their protective effects against ultraviolet B (UVB)-induced photoaging were systematically evaluated in HaCaT keratinocytes. Among the tested fractions, low-molecular-weight peptide fractions exhibited relatively stronger antioxidant activity and effectively reduced intracellular reactive oxygen species (ROS) accumulation in UVB-irradiated cells. In addition, the peptide fractions alleviated UVB-induced inflammatory responses and decreased matrix metalloproteinase (MMP) expression, which was associated with modulation of mitogen-activated protein kinase (MAPK) and nuclear factor kappa B (NF-κB) signaling pathways. Higher-molecular-weight fractions showed relatively stronger effects on maintaining skin barrier-related functions and were associated with regulation of transforming growth factor-β/Smad (TGF-β/Smad) signaling and collagen-related protein expression. Overall, these findings demonstrate functional differences amongDendrobium officinalepeptide fractions and suggest their potential application as natural photoprotective ingredients in functional foods and cosmeceutical products.

A nanozyme based wearable device for colorimetric monitoring of UV radiation exposure in sunlight

Guofen Zhou,Linlin Yu,Wenlong Guo,Zhongwei Jiang,Lianzhe Hu,Min Wang

In this study, nanozyme is used for monitoring the dose of ultraviolet (UV) in sunlight for the first time. We find that nitrogen-doped carbon dots (N-doped C-dots) can effectively catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) under different UV region including UVA, UVB, and UVC. In comparison, the N-doped C-dots fail to show the catalytic activity under visible light. The finding is applied for distinguishing UV light and visible light through the color change of TMB. More importantly, the photo-oxidation TMB catalyzed by N-doped C-dots can work well in a near neutral pH, which is beneficial for its application in wearable device due to the reducing of acid corrosion. Finally, a wearable bracelet is fabricated by integrating N-doped C-dots and TMB into hydrogels. The bracelet is used for monitoring the UV exposure in sunlight. The color change of TMB in the bracelet can be clearly observed by naked eyes or quantified by a smartphone. The bracelet is also used for distinguishing different weather condition such as sunny/cloudy or different place such as indoor/outdoor during a sunny day. This study provides the first example of nanozyme in wearable UV sensor and will stimulate the applications of nanozyme in wearable device.

UVB Induces Sympathetic Nervous System Activation and Norepinephrine Secretion to Regulate The Skin Color of Mice Through the β2-AR/AP-1 Pathway in Epidermal Keratinocytes

Deng Qirui,Liu Xinyan,Wen Xiujuan,Huang Hao,Tang Hongfeng

The aim of this study was to investigate how ultraviolet B (UVB) light regulates AP-1 expression via the β2-adrenergic receptor (β2-AR) in epidermal keratinocytes, which in turn regulates melanin synthesis in melanocytes, thereby modulating downstream melanin production in skin hair follicles and altering mouse skin color. We established a UV-irradiated mouse model to investigate the effects of UV radiation on changes in skin color. By measuring changes in the expression of genes related to cutaneous sympathetic nerves, norepinephrine synthesis and melanin synthesis, we investigated the relationship between β2-AR expression and cutaneous melanogenesis and determined the localization of β2-AR in cells. The results of the siRNA-mediated transfection of keratinized cells with downregulated β2-AR expression were further verified in vitro. Our results suggest that UVB alters the color of the dorsal skin in mice by activating the AP-1/IL-6 pathway, which triggers the sympathetic release of norepinephrine, thereby increasing β2-AR expression in keratinocytes. Overall, our study improves the current understanding of how UVB light influences skin color changes and highlights the complex interplay between ultraviolet radiation and skin physiology.
