Preparation of zein-lecithin-EGCG complex nanoparticles stabilized peppermint oil emulsions: Physicochemical properties, stability and intelligent sensory analysis

Hujun Xie,Fangfang Ni,Jian Gao,Chengzhi Liu,Jieyu Shi,Gerui Ren,Shiyi Tian,Qunfang Lei,Wenjun Fang

Peppermint oil emulsions were prepared by using zein-lecithin-EGCG (Z-L/E) complex nanoparticles as emulsifiers. The preparation conditions of emulsions were optimized via measuring the particle size, surface tension and stability of emulsions, and peppermint oil of 3% (particle size = 375 nm, polydispersity index (PDI) = 0.45), the zein:lecithin ratio of 4:1 ( w/w ) (particle size = 396 nm), and the zein:EGCG ratio of 10:1 ( w/w ) (surface tension = 47.32 N/m) was the optimal condition. The rapid stability analysis showed that the instability mechanism of emulsions was ascribed to creaming and stratification, and the stability mechanism of emulsions was explored, indicating that the complex nanoparticles adsorbed on the surface of oil droplets to give Pickering emulsions. Electronic tongue experiments showed that the Z-E/L 4:1 stabilized emulsion was distinguished from the other three samples due to its good stability. The electronic nose experiment could distinguish the emulsions with different droplet sizes.

Encapsulation of curcumin in ZEIN-HTCC complexes: Physicochemical characterization, in vitro sustained release behavior and encapsulation mechanism

Gerui Ren,Ying He,Chengzhi Liu,Fangfang Ni,Xin Luo,Jieyu Shi,Yuling Song,Ting Li,Min Huang,Qing Shen,Hujun Xie

In order to improve the water solubility and stability of curcumin (CUR) and achieve its sustained release, the ZEIN– N -(2-hydroxy) propyl-3-trimethylammonium chitosan chloride (HTCC) complexes were designed to encapsulate CUR using an antisolvent precipitation (ASP) method. The encapsulation efficiency ( EE ) and loading capacity ( LC ) of curcumin in ZEIN-HTCC complexes were 92.2% and 0.6%, respectively. After encapsulation, the resistance of curcumin to thermal, natural light and UV irradiation, as well as the antioxidative activity were improved significantly ( P  < 0.05). ZEIN-HTCC-CUR complexes exhibited good redispersibility and enhanced the water solubility of curcumin. In vitro simulated digestion experiments clarified that the ZEIN-HTCC complexes can control the sustained release behavior of curcumin, and the release kinetics can be fitted by the Higuchi model. Results taken from fluorescence spectra and Fourier transform infrared spectroscopy (FTIR) indicated that hydrophobic interaction, hydrogen bonding and electrostatic interactions were the main driving forces to form the ZEIN-HTCC-CUR complexes. The X-ray diffraction (XRD) results revealed that the encapsulated curcumin was in an amorphous state. The information provided by this study should be of interest to food industry to fabricate robust nanoscale delivery systems with ZEIN-HTCC complexes for CUR and other hydrophobic bioactive compounds.

Fabrication of Zein-Lecithin-EGCG complex nanoparticles: Characterization, controlled release in simulated gastrointestinal digestion

Hujun Xie,Chengzhi Liu,Jian Gao,Jieyu Shi,Fangfang Ni,Xin Luo,Ying He,Gerui Ren,Zisheng Luo

The Zein-Lecithin-Epigallocatechin gallate (EGCG) complex nanoparticles were fabricated by anti-solvent coprecipitation method. The Zein-Lecithin (Z-L) nanocomplexes exhibited great encapsulation efficiency of 68.5% for EGCG, and the encapsulated EGCG still had good antioxidative capacity. The cumulative release of EGCG in simulated gastric fluid (SGF) and simulated intestinal fluid (SIF) were 19% and 92%, respectively, and the release was closest to Fick release in SGF and First release in SIF. Fluorescence spectroscopy (FL), Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) experiments revealed that the EGCG was successfully encapsulated by Z-L nanocomplexes through electrostatic, hydrophobic and hydrogen bonding interactions. The Zein-Lecithin-EGCG complex nanoparticles exhibited excellent stability and great sustained-release performance, which will be the alternative for potential application in the food industry.

Fabrication and characterization of oil-in-water pickering emulsions stabilized by ZEIN-HTCC nanoparticles as a composite layer

Mengna Cao,Chengzhi Liu,Jieyu Shi,Fangfang Ni,Jiaming Qi,Qing Shen,Min Huang,Gerui Ren,Shiyi Tian,Quanquan Lin,Xiaoxin Lu,Qunfang Lei,Wenjun Fang,Hujun Xie

In this work, the ZEIN-HTCC nanoparticles formed by zein and N-(2-hydroxy)propyl-3-trimethylammonium chitosan chloride (HTCC) were used as stabilizers to prepare oil-in-water (O/W) Pickering emulsions. The preparation conditions including shearing time, volume fraction of corn oil, mass ratio of ZEIN:HTCC and total concentration of ZEIN-HTCC of emulsions were optimized by measuring the droplet size, zeta potential, PDI and surface tension of emulsions. The ZEIN-HTCC emulsions are stable at the pH range of 4–9 and in the low salt ion concentrations up to 0.2 mol L −1 , and can keep stable up to 21 d during low temperature storage. Fourier transform infrared spectroscopy (FTIR), the confocal laser scanning microscope (CLSM) and scanning electron microscopy (SEM) were used to analyze the interaction between emulsion components, revealing that zein and HTCC form a composite layer by flocculation to adsorb on the surface of oil droplets, thus preventing emulsion droplets from aggregation. This novel, long-term stable, surfactant-free, and edible zein-based Pickering emulsion could be used as potential carriers for lipophilic nutrients delivery.

Zein structure and its hidden zearalenone: Effect of zein extraction methods

Hongxia Tan,Hongyuan Zhou,Ting Guo,Jiaxin Li,Chi Zhang,Shuo Wang,Yuhao Zhang,Liang Ma

Zein, the plant protein, has received great attention in the last years. However, hidden zearalenone (ZEN), the zein-bound ZEN present in zein would have a great hazard to humans and animals. To date, the fates of hidden ZEN under different extraction methods of zein have not been clarified. In this study, the effect of zein extraction methods on conversion of free ZEN with hidden ZEN and its corresponding mechanism were studied. Results showed that the acid extraction condition promoted the formation of hidden ZEN (up to 92.03%), which was closely related to increased hydrophobic cavity of zein. However, alkaline extraction condition caused the conversion of hidden ZEN (from 58.82% to 13.33%) into free ZEN (from 41.18% to 86.67%), which was attributed to the great denaturation of zein. This study is of great significance for controlling hidden ZEN during zein extraction and maize processing.

Self-Assembly Loading of Schisandra chinensis Nanoparticles and Its Effect on the Malignant Biological Behavior of Ovarian Cancer Cells

Chen Aimin,Hu Ruihua

Schisandra polysaccharide (SCHP) was utilized as the target drug, and Zein (corn prolamine) was utilized as the drug carrier. First, SCHP/Zein nanoparticles (Zein-SCHP NPs) were prepared. Then, a polydopamine (PDP) coating was utilized to enhance the stability and tumor targeting of Zein. Finally, the folic acid (FA)-targeting ligand was covalently attached to the PDP-modified Zein-SCHP@PDP NP surface to form a highly pH-sensitive NP (Zein-SCHP@PDP-FA NPs) with active/passive targeting. In the experiment, apart from physical characterization of Zein-SCHP-based nanomaterials, the in vitro cytotoxicity and cell uptake properties of these materials were tested. In addition, an ovarian cancer cell line, SKOV3, was taken as the research object to explore the photodynamic-mediated killing effect of Zein-SCHP@PDP-FA NPs on this cancer cell. The results demonstrated that the average particle size of Zein-SCHP@PDP-FA NPs was (379.68±1.36) nm, the zeta potential was (−43.68±1.67) mV, the encapsulation efficiency was (97.86±1.38)%, and the drug loading was (11.75±0.16)%. Transmission electron microscope (TEM) suggested that the Zein-SCHP@PDP-FA NPs were spherical, and there was no adhesion between the particles. Differential scanning calorimetry (DSC) and far infrared (FIR) results indicated that SCHP existed in the NPs in the crystal structure and proved that the PDP and FA were successfully modified. The in vitro release of the drug showed that Zein-SCHP@PDP-FA NPs had sustained release, and the CCK-8 test showed that Zein-SCHP@PDP-FA NPs were highly cytotoxic and could notably reduce the IC50 values of SCHP. Furthermore, in vitro uptake experiments revealed that Zein-SCHP@PDP-FA NPs exhibited strong specific recognition in cells expressing different folate receptors. Compared with Zein-SCHP NPs and Zein-SCHP@PDP NPs, Zein-SCHP@PDP-FA NPs combined with photodynamic therapy enhanced reactive oxygen species (ROS) production in cells, downregulated hypoxia-inducible factor (HIF)-1α and interleukin (IL)-6 proteins, and exerted more destructive effects on SKOV3 cells.

Protein-based supramolecular adhesive capable of on-demand adhesion and anti-adhesion for preventing undesired epidural tissue adhesion

Tong Li,Guang Wen,He Zhao,Yang Qu,Han Wu,Yingchuan Sun,Jianwu Zhao,Wen Li

The undesired adhesion between the dura mater and surrounding tissues is a great challenge in the clinical treatment of spinal stenosis and disc herniation, and this condition can typically lead to failed back surgery syndrome (FBSS) and persistent or recurrent lumbosacral pain or sciatica. Here, a protein-based supramolecular adhesive featured with wet adhesion and on-demand anti-adhesion via non-covalent interactions between anionic glycyrrhizic acid (GA) nanofibers and N-α-lauroyl-arginine ethyl ester hydrochloride (LAE) dispersed zein colloid (zein-LAE). By controlling the crosslinking density of the zein-LAE/GA supramolecular polymers, the adhesion and anti-adhesion could be conveniently regulated. In detail, suitable amount of GA improved the adhesion strength of the zein-LAE/GA supramolecular adhesives, whereas excessive GA caused the surface curing and anti-adhesion. It was proposed that the anti-adhesion outcome was attributed to the enhanced cross-linking density and Young’s modulus of the supramolecular polymers, and the declined freedom of the binding groups. The excellent biocompatibility and biodegradation enabled the zein-LAE/GA supramolecular polymers to function as easy-to-implement Janus implants for the in vivo prevention of postoperative epidural adhesion in a rat model. In conclusion, the physical barrier of the Janus implant together with the anti-oxidation and anti-inflammatory effects of the GA component played a synergistic role in achieving epidural anti-adhesion.

Zein-coated versus zein-inserted kaempferol liposomes: enhancing bitterness masking, stability, and bioaccessibility

Haowei Yang,Xiufang Bi,Ruifeng Ying,Khizar Hayat,Xiaohong Sun,Chunlei Ni,Quan "Sophia" He,Meigui Huang

Kaempferol, a plant flavonoid with antioxidant, anti-inflammatory, and anti-allergenic bioactivities, has limited use in functional foods due to its bitterness and low bioavailability. This study compared two zein-based modification strategies, insertion and coating, for kaempferol-loaded liposomes in terms of bitterness masking, stability, and bioaccessibility. At 0.2 mg/mL zein, insertion showed 4.67% higher encapsulation efficiency than coating. ( P   <   0.05 ). E -tongue analysis showed coating reduced bitterness more effectively than insertion. Raman spectroscopy revealed coating involved electrostatic interactions, while insertion involved hydrophobic ones. Fluorescence probing indicated insertion expanded liposome internal space, whereas coating resulted in a compact structure. TG and DSC analyses confirmed coating provided greater thermal stability. Additionally, coated liposomes exhibited superior environmental stability, antioxidant activity, and bioaccessibility. In vitro tests showed zein-coated liposomes released 62.98 ± 1.35% kaempferol, while zein-inserted liposomes released 50.32 ± 0.59%. These findings offer new insights for integrating bitter bioactives into functional foods.

Simulating the behavior of antioxidant to explore the mechanisms of oxidative stability in Pickering emulsion

Zhibin Chen,Zijun Zhao,Wenbo Wang,Qianyi Ye,Jie Xiao

This study explores effective strategies for bolstering emulsion oxidative stability via optimized interfacial distribution of varying hydrophobicity antioxidants (gallic acid, propyl gallate, octyl gallate) in zein nanoparticle (ZP) stabilized Pickering emulsions. Experimental and simulation methods revealed that antioxidant (AO) with higher hydrophobicity or loaded into ZP demonstrated stronger hydrogen bonding and van der Waals interactions with ZP. This increased interfacial loading of antioxidants resulted in improved oxidative stability in Pickering emulsions. The flow, distribution and orientation of AO, as revealed by dissipative dynamics simulations, highlighted the role of hydrophobic interactions during initial AO migration, influenced by varied alkyl chain lengths. Subsequent interface rearrangements arose from conservative force interactions between the AO's phenol hydroxyl ends and ZP. These findings inform effective interfacial engineering to optimize antioxidant efficiency, guiding practical applications in emulsion systems for improved oxidative stability.

Zein-enhanced sodium alginate/thiostannate microsphere adsorbent Zein@SA/KBS for efficient removal of cesium from wastewater

Chi Ma,Wenlei Fan,Wei Qin,Yafei Guo,Lichun Ma,Nelson Belzile,Tianlong Deng

Although efficient removal of Cs from wastewater is of great importance because of the nuclear energy sustainable development and public health, removing cesium is challenging for the high concentrations of sodium and potassium ions coexist. In this work, we synthesized the first novel bismuth-doped layered tin sulfide (KBS) and applied it to the efficient green adsorption of liquid cesium resources. KBS could reach the adsorption equilibrium for Cs + within only 2 min with a theoretical adsorption capacity of 425.55 mg/g. Furthermore, the adsorption performance remained reliable after 10 cycles of use. The structure and properties of KBS were explained at the molecular level by DFT calculations, and the calculated results were in sound agreement with the experimental data. Meanwhile, in this study, the material was reinforced and shaped by the electrostatic interaction of Zein and sodium alginate hydrogel, and finally, Zein@SA/KBS spherical composite adsorbent was obtained. The problem of the difficult recovery of adsorbent and the secondary contamination was solved. The adsorption performance of Zein@SA/KBS on Cs + was better than most composite adsorbents and showed reliable stability. Therefore, the new microspherical adsorbent (Zein@SA/KBS) has great potential for industrial application in removing radioactive cesium from wastewater.

Zein Increases the Cytoaffinity and Biodegradability of Scaffolds 3D-Printed with Zein and Poly(ε-caprolactone) Composite Ink

Linzhi Jing,Xiang Wang,Hang Liu,Yuyun Lu,Jinsong Bian,Jie Sun,Dejian Huang

Electrohydrodynamic printing (EHDP) has attracted extensive interests as a powerful technology to fabricate micro- to nano-scale fibrous scaffolds in a custom-tailored manner for biomedical applications. A few synthetic biopolymer inks are applicable to this EHDP technology, but the fabricated scaffolds suffered from low mechanical strength, biocompatibility, and biodegradability. In this study, a series of poly(ε-caprolactone) (PCL)/zein composite inks were developed and their printability was examined on a solution-based EHDP system for scaffold fabrication. Multilayer grid scaffolds were manufactured by PCL, PCL/zein-10, and PCL/zein-20 inks, respectively and characterized. The mechanical strength of scaffolds printed by PCL/zein composite inks was remarkably enhanced in terms of Young’s modulus and yield stress. The enzyme-accelerated in vitro degradation study demonstrated that zein-containing scaffolds exhibited dose-responsive improvement on the degradation rate as evidenced by surface morphological change of fibers. Moreover, the biocompatibility of PCL/zein scaffolds, tested on mice embryonic fibroblast (NIH/3T3) and human nonsmall lung cancer cell (H1299), manifested better cell affinity. Our findings suggest that scaffolds fabricated by the solution-based EHDP with PCL/zein composite inks can significantly improve Young’s modulus, yield stress, biocompatibility, and biodegradability and have potential applications in drug delivery systems, 3D cell culture modeling, or tissue engineering.

Thiol-modified hyaluronic acid improves the physical stability of curcumin-zein nanoparticles by forming disulfide bonds with zein

Guangshuai Zhang,Mengdi Qin,Mengran Guo,Mo Li,Di Zhang,Yichi Sun,Bingyang Liu,Zhonggui He,Qiang Fu

Zein-based nanoparticles have been developed in the food industry. However, their poor pH stability and unfavorable ionic strength stability remain a challenge even with the use of polysaccharides (such as hyaluronic acid) as stabilizers. To address this shortcoming, an improved strategy based on the disulfide bonds between thiol-modified hyaluronic acid (HASH) and zein was proposed. In this study, curcumin-zein nanoparticles (ZNs-HASH) were prepared with HASH as a stabilizer. The ZNs-HASH displayed similar particle sizes and spherical structures with ZNs and ZNs-HA (HA as a stabilizer). The Fourier transform infrared spectroscopy demonstrated the formation of disulfide bonds between zein and HASH. Among the three formulations tested, ZNs-HASH exhibited the highest pH and salt ion stability and the strongest antioxidant capacity. This study provided new insights for the improvement of physical stability of zein nanoparticles and the development of oral bioactive substances by chemical modification of natural polysaccharides.
