A ratio fluorescent probe to detect Ag + using arginine-naphthalene imide (AN) had been elaborately designed and synthesized. The structure of AN was characterized by 1 HNMR and FTIR. The optical properties of AN were investigated by ultraviolet and fluorescence spectroscopy. The results indicated that the fluorescent intensity of AN was enhanced gradually with the increase of Ag + concentration, which was hardly affected by other metal ions such as Bi 3+ , Cd 2+ , Co 2+ , Cr 3+ , Cu 2+ , Fe 3+ , Hg 2+ , Pb 2+ , K + and Na + . A good linear correlation was displayed between the concentration of Ag + (ranging from 4.30 to 11.40 μmol L −1 ) and the fluorescence intensity ( R 2 = 0.9985). The detection limit was 0.1883 μmol L −1 . The mechanism of the sensor may be based on the principle of intramolecular charge transfer. It was expected that this novel ratio sensor could be used for the detection of silver ions in water.

Putrescine, a biogenic amine, is a highly valued compound in medicine, industry, and agriculture. In this study, we report a whole-cell biocatalytic method inEscherichia colifor the production of putrescine, using L-arginine as the substrate. L-arginine decarboxylase and agmatine ureohydrolase were co-expressed to produce putrescine from L-arginine. Ten plasmids with different copy numbers and ordering of genes were constructed to balance the expression of the two enzymes, and the best strain was pACYCDuet-speB-speA. The optimal concentration of L-arginine was determined to be 20 mM for this strain. The optimum pH of the biotransformation was 9.5, and the optimum temperature was 45 °C; under these conditions, the yield of putrescine was 98%. This whole-cell biocatalytic method appeared to have great potential for the production of putrescine.Keywords:L-arginine;putrescine;Escherichia coli;whole-cell catalysis;co-expression

A fast mass analysis method using digital ion trap technology was reported. The mass analysis was realized by scanning the frequency of digital RF voltage and dipole ion resonance voltage during ion injection period without the conventional ion cooling, ion resonance ejection or ion cleaning process. It can decrease about three fourth of traditional analysis time using ion trap mass spectrometry. The ion signal was optimized through the modification of frequency scanning rate, ion gate voltage and other experimental parameters. For example, an optimal mass spectroscopy of reserpine ( m / z 609) and arginine ( m / z 174) were obtained when the ion gate voltage was 9 V and the digital RF frequency was scanned from 1 MHz to 400 kHz at rate of 2385 Th s −1 . The results indicated that the obtained mass spectra were in very good agreement with conventional analytical method using ion trap mass analyzer.

This work evaluated the effects of l -arginine on the stability of porcine hemoglobin concentrates during storage. The addition of l -arginine increased both the oxyhemoglobin content and a* values but decreased both the methemoglobin content and free iron content ( p < 0.05) during storage. The addition of iron cation decreased both the oxyhemoglobin content and a* values but increased both the methemoglobin content and free iron content ( p < 0.05). CD spectra disclosed that the free iron induced the transformation of the hemoglobin secondary structure from an α-helix to a β-pleated, β-turn, and random-coil arrangement, while l -arginine hindered the behavior of the free iron. Infrared spectra revealed that the l -arginine coordinated with the free Fe 2+ or Fe 3+ to form the corresponding complex at 25 °C and pH 7.3. These results showed that l -arginine enhanced the stability of hemoglobin concentrates by coordinating with free iron.

Arginine-assisted thermal decomposition technique was used to prepare nanosized Co 3 O 4 (A-Co 3 O 4 ) with its supercapacitive performances checked. A-Co 3 O 4 was characterized by different techniques such as X-ray diffraction, X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy and scanning electron microscopy. Brunauer-Emmett-Teller analysis revealed that A-Co 3 O 4 had a larger specific surface area than Co 3 O 4 prepared by conventional calcination without arginine (F-Co 3 O 4 ). Electrochemical performances of A-Co 3 O 4 based electrode were better than that of the F-Co 3 O 4 based electrode. Cyclic voltammetric results showed the specific capacitances of A-Co 3 O 4 and F-Co 3 O 4 were found to be 259.2 and 122.2 F/g at a scan rate of 0.01 V/s. Long-term galvanostatic charge-discharge cycling tests indicated the specific capacitance for A-Co 3 O 4 and F-Co 3 O 4 were 128.6 F/g and 106.4 F/g with 104.7% and 103.3% capacitance retention after 1000 cycles at a current density of 5.0 A/g. Electrochemical impedance spectra analysis showed the charge transfer resistance (Rct) of A-Co 3 O 4 was 7.56 Ω and that of F-Co 3 O 4 was 15.56 Ω. All the results indicated the A-Co 3 O 4 exhibited superiority originated from large surface area with porous structure.

Fluorescent nanosensors based on ligand-engineered gold nanoclusters (Au NCs), guiding antibiotic therapy, offer a promising approach for rapid bacterial detection. Here, we developed dual-ligand-modified Au NCs with 6-azido-2-thiothymidine (ATT) and L-arginine (Arg), in which Arg modification enhanced the photoluminescence (PL) intensity compared to ATT-Au NCs. This enables a linear response (R2 > 0.97) to bacterial concentrations (105–109 CFU mL−1) within 30 minutes. Targeted metabolomics revealed that bacterial Arg deiminase metabolized Arg to citrulline, triggering PL quenching. Furthermore, it demonstrates the quantitative relationship between bacterial metabolic status and alterations in PL signals. The signal changes were visible under UV light and can also be quantified using a smartphone app (Light Meter). The detection limit reached 105 CFU mL−1. In vivo experiments confirmed that the probe can monitor wounds infected with Staphylococcus aureus (S. aureus), with minimal interference from biological matrices. This work highlights a portable, cost-effective strategy for point-of-care bacterial detection, addressing critical needs in clinical diagnostics.

4-nitrophenol (PNP) is generally regarded as a diesel exhaust particle (DEP). Arginine plays an important role as a new feed additive, possessing highly efficient antioxidant activities. Here we investigated the effects of dietary supplementation with arginine against ovarian damage induced by PNP in rats. A total of thirty-two female rats postnatal day 28 (PND 28) were randomly divided into four groups. Two groups were fed with basal diet or 13 g/kg arginine in diet for 4 weeks, respectively; the other two groups were given PNP (100 mg/kg b.w.) daily by subcutaneous injection for 2 weeks following pretreatment with either basal diet or arginine diet for 2 weeks. The values of body weight gain (BWG), average daily gain (ADG) and percentage weight gain (PWG) upon PNP treatment were significantly reduced than those in other groups. The relative liver weight in the PNP group was significantly decreased compared with the control group. Treatment with PNP significant reduced the number of corpora lutea, although serum 17β-estradiol (E2) and progesterone (P4) concentrations were unchanged. The morphology of the ovaries in PNP-treated rats displayed necrosis, follicular deformation and granulosa cells irregular arrangement. Moreover, exposure to PNP enhanced production of malondialdehyde (MDA) and hydrogen peroxide (H2O2), and decreased the activities of total superoxide dismutase (T-SOD) and catalase (CAT), and the co-administration of arginine can attenuate the oxidative stress caused by PNP. These results suggest that arginine may have a protective effect against ovarian damage induced by PNP owing to its antioxidant capacity effect.

Shape and size control of gold (Au) nanocrystals plays important roles in their catalytic and optical properties. Hierarchical Au nanodendrites were directly electrodeposited on a glassy carbon electrode (GCE) using l -arginine as a growth-directing agent, while there is no seed, template, or surfactant involved. It was found that the experimental conditions such as the applied potentials, concentrations of HAuCl 4 and l -arginine, and deposition time were essential to the final morphology of the Au deposits. The as-synthesized Au nanocrystals displayed enhanced electrocatalytic activity and improved stability for ethylene glycol (EG) oxidation in acidic media as compared to Au nanoparticles.

Developing integrated, drug-free therapeutic platforms that simultaneously address oxidative stress, immune dysregulation, and gut microbiota dysbiosis remains a significant challenge in UC management. Although hydrogels offer localized delivery advantages, their clinical versatility is frequently hindered by a reliance on exogenous drug encapsulation. To overcome this limitation, we developed a multifunctional, ROS-responsive hydrogel (ODCAS) that functions as an inherently bioactive platform. The ODCAS network is constructed via dynamic Schiff base crosslinking between ODex and thiolated arginine chitosan, further reinforced by ROS-triggered disulfide bonds leveraging. This sophisticated architecture undergoes ROS-responsive programmed degradation, driving in situ NO generation to trigger a therapeutic cascade. Specifically, the TPA-derived thiol moieties facilitate inflammation-targeted mucoadhesion via disulfide exchange with the mucosal layer, while simultaneously providing potent, direct ROS scavenging (72.21 ± 2.47% DPPH· radical scavenging). Furthermore, the metabolic byproducts of the hydrogel network provide prebiotic support to restore microbial equilibrium. In a DSS-induced murine colitis model, oral administration of ODCAS significantly attenuated disease severity, reducing the DAI by 52.08%, downregulating pro-inflammatory cytokines, and restoring both intestinal barrier integrity and microbial homeostasis. These findings highlight the ODCAS hydrogel as a robust, ROS-responsive biomaterial platform that orchestrates disulfide-mediated anchoring and an arginine-driven NO cascade for the effective management of chronic inflammatory diseases.

As a common opportunistic pathogen, Staphylococcus aureus ( S. aureus ) can rapidly adapt to the host immune system and cause chronic infections. Currently, such chronic infections are extremely difficult to eliminate, severely impairing the function of tissues and organs. We observed localized tissue senescence in the mammary glands of mice with chronic S. aureus infection; however, the mechanism driving this senescence remains unclear. To address this, we employed a mouse model of chronic S. aureus -induced mastitis and an in vitro model of mouse mammary epithelial cells (mMECs) to confirm the inductive effect of S. aureus on tissue and cellular senescence. Through integrated analysis of the transcriptome and metabolome of mouse mammary tissues, it was found that the ornithine cycle was significantly disordered following S. aureus infection, wherein Arginase 1 (Arg1) serves as a key metabolic regulator. Notably, stimulation by S. aureus induces mammary epithelial cells to produce cytokines, thereby promoting the massive release of Arg1 by macrophages into the microenvironment, which constitutes one of the sources of abnormally elevated Arg1 in mammary tissue. This extracellular Arg1 is subsequently taken up by mammary epithelial cells, further accelerating the intracellular conversion of arginine to ornithine. The accumulation of ornithine and its downstream metabolites drives the occurrence of senescence in epithelial cells. Importantly, supplementation with excess arginine or ornithine can mimic this senescence phenotype, while knocking down Arg1 in epithelial cells can reverse this phenomenon. In summary, this study reveals a novel immunometabolic cross-regulation mechanism, specifically that Arg1 released by macrophages—triggered by mammary epithelial cells under S. aureus stimulation—acts as a paracrine senescence-inducing factor that acts back on mammary epithelial cells. The Arg1—ornithine axis holds promise as a potential therapeutic target for chronic infection-associated tissue senescence.
This study found that S. aureus stimulation induces senescence in mouse mammary glands and mMECs. In mMECs, S. aureus directly induces senescence-associated changes, including activation of p53/p21, DNA damage, and SASP-related inflammatory responses, despite reduced endogenous Arg1 expression. Meanwhile, the compensatory upregulation of Arg2 and other ornithine-cycle enzymes suggests that arginine/ornithine metabolism is remodeled in epithelial cells. In the tissue microenvironment, epithelial-derived inflammatory mediators further stimulate macrophages to express and release Arg1. This macrophage-derived Arg1 can be taken up by mMECs and may amplify epithelial senescence by promoting arginine-to-ornithine metabolic dysregulation. Therefore, macrophage-derived Arg1 promotes epithelial senescence but is not the sole initiator of this process.

The persistent escalation of antibiotic resistance poses a severe threat to global public health. As a common opportunistic pathogen, the rapid development of resistance in Escherichia coli (E. coli) poses severe challenges to clinical anti-infective therapy. Against this backdrop, the combined use of amino acids and antibiotics is considered an effective strategy against multidrug-resistant bacterial infections. This study investigated the antibacterial efficacy and mechanism of action of the combination of arginine (Arg) and florfenicol (FFC) against resistant E. coli. Results indicate that Arg significantly enhances the bactericidal activity of FFC against resistant E. coli both in vivo and in vitro, successfully reversing the resistance phenotype. Mechanistic studies reveal that Arg disrupts fatty acid β-oxidation by inhibiting the arginine succinyl-transferase (AST) pathway, leading to excessive free fatty acid (FFA) accumulation. This triggers membrane structural damage, energy metabolism disorders, and oxidative stress. Concurrently, Arg inhibits the expression and function of multidrug efflux pumps, such as AcrAB-TolC and EmrAB-TolC, reducing FFC efflux and increasing its intracellular concentration. This study systematically reveals a novel pathway whereby Arg synergistically enhances the antibacterial activity of FFC through dual mechanisms of metabolic reprogramming and efflux pump inhibition, providing theoretical support for the development and application of amino acid compounds as antibiotic adjuvants.

Ethanol produced during the Huangjiu fermentation can adversely affect the growth and metabolism of Saccharomyces cerevisiae . It has been established that arginine, one of the major amino acids found in Huangjiu , can protect yeast cells against ethanol stress. Previous studies have indicated that BTN2 , which influences arginine metabolism, is associated with ethanol tolerance in yeast. In this study, we evaluated the response of BTN2 to ethanol stress based on analyses of cell growth, morphology, membrane permeability, membrane integrity, ROS and how BTN2 influenced intracellular metabolites, enzymes, and transcriptional genes in the arginine metabolic pathway. Our findings revealed that BTN2 promoted significantly increases in the growth and ethanol tolerance of S. cerevisiae g. Under 10% ethanol stress, RT-qPCR showed that BTN2 Δ ( BTN2 knock out strain) up-regulated CAR1 , CAR2 , GAP1 , DUR1,2 and DUR3 to increase arginine and urea degradation, BTN2 ↑ ( BTN2 over-expressing strain) up-regulated CAR1 , CAN1 and DUR3 to enhance arginine uptake, catabolism and urea reabsorption, while TPS1 was up-regulated to improve ethanol tolerance ability. Yeast up-regulated both CAR1 (Arginase) and DUR3 (urea transporter) genes in BTN2 knockout and over-expression strains to counteract the ethanol effects. Our findings revealed a novel arginine metabolism under ethanol tolerance of a core functional yeast during Huangjiu fermentation. These findings could contribute to the development of an approach for inhibiting toxic ethyl carbamate (EC) production during Huangjiu fermentation.
