Jizhong Yuan,Yaohuang Jiang,Mengle Liu,Peipei Wu,Guoxian Feng,Yanchun Yu,Xiongfa Yang

UV-curable L(-)–borneol-functionalized antibacterial hydrogels for packaging fresh-cut banana and cherry tomato (UV-LBs) were designed from L(-)–borneol-functionalized polyurethane acrylate prepolymers (LB-PUAs) and thiol-functionalized PVA (PVA-SH) using a thiol-ene click reaction initiated by UV light. UV-LBs exhibit unique properties, including excellent thermal stability, high mechanical performance and quite high antibacterial efficiency. The initial thermal decomposition temperature (Td5), tensile strength and elongation at break are in the range of 225–240 °C, 1.38–2.05 MPa and 44.4–68.6%, respectively. The antibacterial efficiency of UV-LBs againstStaphylococcus aureus(S. aureus),Escherichia coli(E. coli), andMonilia albican(M. albican) can reach 67.4%, 75.6% and 83.7%, respectively. The storage time of packaged fresh-cut banana and cherry tomato can be extended from 12 h to 30 h and 4 d to 5 d, respectively.

Enhancing effect and safety assessment of borneol on the oral absorption of sub-100 nm PLGA nanoparticles

Qin Luo,Ge Ru,Tianci Hu,Wenchao Gao,Rouling Song,Danyang Liu,Jing Wu,Jianxin Wang,Jianyong Sheng

Overcoming the gastrointestinal barrier remains a major challenge for oral nanoparticle (NP) drug delivery systems. Here, we evaluate borneol as a permeation enhancer for improving the oral absorption of intact poly(lactic-co-glycolic acid) (PLGA) nanoparticles with different mean sizes (50–300 nm) and assess its long-term safety. In vitro, sub-100 nm nanoparticles (NPS-S) exhibited superior cellular uptake and transport across Caco-2 monolayers compared to their larger counterparts. Borneol selectively and concentration-dependently enhanced the absorption of NPS-S, mechanistically linked to the promotion of caveolae-mediated endocytosis and the reversible modulation of tight junctions, as evidenced by a decrease in transepithelial electrical resistance (TEER) and occludin disruption. Utilizing an aggregation-caused quenching (ACQ) probe for specific tracking of intact NPs, in vivo studies confirmed that borneol significantly enhanced the gastrointestinal retention, absorption and systemic distribution of intact NPS-S. A comprehensive toxicological evaluation, including a 30-day rat study, demonstrated that borneol did not induce cytotoxicity, inflammation, or tissue damage at daily doses up to 270 mg/kg. Our findings establish borneol as a safe and effective oral permeation enhancer for intact nanocarriers, offering a viable strategy to enhance the oral bioavailability of therapeutics, particularly those encapsulated within small-sized NPs.

Borneol-mediated vardenafil hydrochloride patch for pediatric pulmonary arterial hypertension: Preparation, characterization and in vivo study

Dong Jiang,Huajin Tan,Rujiao Zhang,Kaikang Wang,Yujia Zhang,Xiaochuan Tan,Wensheng Zheng

Background Pediatric pulmonary arterial hypertension (PPAH) is a malignant progressive rare disease characterized by restricted pulmonary artery blood flow and progressively increasing blood pressure, which has shorter survival time of only about 10 months as compared to adults. Previous studies have shown that low-dose vardenafil hydrochloride (Var) could significantly improve the symptoms of PPAH. However, Var is currently available only in tablet form in the market for erectile dysfunction , and no special preparation is available for PPAH. Methods In this study, borneol-mediated vardenafil hydrochloride patch (BO-VarP) with sodium polyacrylate as the skeleton material was prepared by coating method, which was characterized by temperature resistance, formability, adhesive force, skin irritation and in vitro permeation. Blood concentration of optimized BO-VarP was measured by LC-MS/MS using intragastric administration (i.g.) as a control, and pharmacodynamic studies were conducted using a rat model of pulmonary arterial hypertension induced by monocrotaline . Results and discussion Optimized BO-VarP showed good appearance, with optimal temperature resistance and formability, appropriate adhesive force and low skin irritation, and its cumulative permeation flux was 14.9 times higher than patch without penetration enhancer . The blood concentration within therapeutic window of BO-VarP lasted longer than i.g., and BO-VarP could improve symptoms of PPAH by reducing pulmonary arterial pressure and right heart hypertrophy index. Conclusion BO-VarP had good therapeutic effect in PPAH rats and suitable compliance in children, which provided a potential industrial transdermal delivery system for the treatment of PPAH.

Enhanced Anti-Glioma Efficacy by Borneol Combined With CGKRK-Modified Paclitaxel Self-Assembled Redox-Sensitive Nanoparticles

Lv Lingyan,Li Xinrui,Qian Wei,Li Shennan,Jiang Yan,Xiong Yaokun,Xu Jianpei,Lv Wei,Liu Xiaoyan,Chen Yun,Tang Yulin,Xin Hongliang

The serious therapeutic obstacles to glioma treatment include poor penetration across the blood-brain barrier (BBB) and low accumulation of therapeutic drugs at tumor sites. In this study, borneol combined with CGKRK peptide (a ligand of the heparan sulfate which overexpress on the glioma cells) modified paclitaxel prodrug self-assembled redox-responsive nanoparticles (CGKRK-PSNPs) were hypothesized to enhance the BBB penetration ability and active tumor targeting efficiency, respectively. The resulting CGKRK-PSNPs possessed a spherical shape with a small particle size (105.61 ± 1.53 nm) and high drug loading for PTX (54.18 ± 1.13%). The drug release behavior proved that CGKRK-PSNPs were highly sensitive to glutathione (GSH) redox environment. The in vitro cell experiments suggested that CGKRK-PSNPs significantly increased the cellular uptake and cytotoxicity of U87MG cells, meanwhile CGKRK-PSNPs showed the low cytotoxicity against BCEC cells. Combined with borneol, CGKRK-PSNPs exhibited enhanced transportation across in vitro BBB model. In intracranial U87MG glioma-bearing nude mice, the higher accumulation of CGKRK-PSNPs combined with borneol was observed through real-time fluorescence image. Moreover, the in vivo anti-glioma results confirmed that CGKRK-PSNPs combined with borneol could improve the anti-glioma efficacy with the prolonged medium survival time (39 days). In conclusion, the collaborative strategy of CGKRK-PSNPs combined with borneol provided a promising drug delivery routine for glioblastoma therapy.

Synergistically Improved Antifouling Efficiency of a Bioinspired Self-renewing Interface via a Borneol/ Boron Acrylate Polymer

Fan Song,Jun Wang,Linlin Zhang,Rongrong Chen,Qi Liu,Jingyuan Liu,Jing Yu,PeiLi Liu,Jizhou Duan

Underwater facilities are often perplexed by severe and ubiquitous biofouling. The widely applied commercial antifouling materials still have several challenges in static applications. Herein, a polymer containing isoborneol and borane (PBABs), the borneol derivative structure and grafted pyridine-triphenylborane (PTPB) as antifouling groups were prepared by radical polymerization. PBABs showed high antibacterial rates for Escherichia coli ( E. coli) and Staphylococcus aureu s ( S. aureus ) of up to 95.1% and 81.1%, respectively, confirming superior antibacterial adhesion propertys. More importantly, PBABs effectively reduced the expression of mussel adhesion protein, indicating superior antifouling propertys, resulting from the synergistic effect of multiple antifouling functional groups on the material’s surface. Therefore, the PBABs have been evaluated as noncytotoxic, low-cost, easily synthesized, and mass-produced, which demonstrates their great potential for actual marine applications.

Borneol-grafted poly(amidoamine) dendrimer with dual antimicrobial and enamel-remineralizing activities against early-stage dental caries

Ziyi QIN,Ying SHA,Siyi SUN,Mengyuan LI,Fei JIANG,Yichen ZONG,Jie LI,Guangdong ZHANG

Early caries is initiated by bacterial colonization and biofilm maturation, which drive localized demineralization of enamel. This study developed a borneol-grafted poly(amidoamine) dendrimer to inhibit bacterial adhesion, suppress biofilm formation, and enhance enamel remineralization. Borneol-grafted poly(amidoamine) dendrimer was synthesized via imine linkage and evaluated for antibacterial activity against Streptococcus mutans and Streptococcus sanguinis assessed by adhesion inhibition, and biofilm biomass/viability assays. Mechanisms were explored by zeta potential analysis and scanning electron microscopy. Remineralization was assessed by microhardness, lesion depth, mineral density, X-ray diffraction, and surface morphology; in vivo efficacy was tested on human enamel samples in an intra-oral model. Borneol-grafted poly(amidoamine) dendrimer reduced bacterial adhesion and viability, neutralized surface charge, and affected membrane integrity. In vitro, borneol-grafted poly(amidoamine) dendrimer improved hardness, reduced lesion depth, and increased mineral density; in vivo, it promoted mineralized layers without systemic toxicity. These findings indicate it as a single-scaffold preventive–restorative agent for early-stage caries.

Enhanced catalytic performance of transition metal-doped Co/TiO2 for continuous-flow synthesis of borneol from isoborneol

Chenyang Li,Zhihan Liu,Huimin Li,Yunsen Xiao,Ying Huang,Yuting Lin,Jie Liu,Bin Wu,Fuweng Zhang,Huidong Zheng,Longfei Yan

Borneol, a pharmaceutically important monoterpenoid, has emerged as a key ingredient in modern medicinal formulations. Here, we report a highly efficient heterogeneous catalytic system using transition metal-doped Co/TiO2 catalysts for the continuous-flow isomerization of isoborneol to borneol. Through systematic screening of bimetallic combinations, we demonstrate that Cu and Ni dopants synergistically enhance the redox properties of Co species, particularly improving hydrogenation–dehydrogenation efficiency in the isomerization of isoborneol. The optimized 1Cu–3Co/TiO2 catalyst (Cu : Co = 1 : 3 mass ratio) enabled a two-stage continuous-flow system, achieving 58% molar yield of borneol with <0.4% camphor byproduct formation after solvent removal. Remarkably, the catalyst maintained >99% metal retention during a total of 200 h of continuous reaction. This work not only provides fundamental insights into promoter effects in bimetallic catalysis but also establishes a commercially viable platform for continuous production of high-purity botanical therapeutics.

Nose-to-brain delivery of borneol modified tanshinone IIA nanoparticles in prevention of cerebral ischemia/reperfusion injury

Luting Wang,Lin Xu,Junfeng Du,Xiao Zhao,Mei Liu,Jianfang Feng,Kaili Hu

Targeted treatment of cerebral ischemia/reperfusion injury (CIRI) remains a problem due to the difficulty in drug delivery across the blood–brain barrier (BBB). In this study, we developed Bo-TSA-NP, a novel tanshinone IIA (TSA) loaded nanoparticles modified by borneol, which has long been proved with the ability to enhance other drugs’ transport across the BBB. The Bo-TSA-NP, with a particle size of about 160 nm, drug loading of 3.6%, showed sustained release and P-glycoprotein (P-gp) inhibition property. It demonstrated a significantly higher uptake by 16HBE cells in vitro through the clathrin/caveolae-mediated endocytosis and micropinocytosis. Following intranasal (IN) administration, Bo-TSA-NP significantly improved the preventive effect on a rat model of CIRI with improved neurological scores, decreased cerebral infarction areas and a reduced content of malondialdehyde (MDA) and increased activity of superoxide dismutase (SOD) in rat brain. In conclusion, these results indicate that Bo-TSA-NP is a promising nose-to-brain delivery system that can enhance the prevention effect of TSA on CIRI.

Pep-1&borneol–Bifunctionalized Carmustine-Loaded Micelles Enhance Anti-Glioma Efficacy Through Tumor-Targeting and BBB-Penetrating

Xiaoyuan Guo,Guojian Wu,Hong Wang,Lukui Chen

Tumor-targeting and blood-brain barrier (BBB)-penetrating are highly desirable for the treatment of glioma. In this study, we developed Pep-1&borneol–bifunctionalized carmustine-loaded micelles (Pep-1/Bor/CMS-M) capable of targeting interleukin-13 receptor–overexpressed glioma and penetrating the brain microvascular endothelial cells–associated physiologic barriers. Pep-1/Bor/CMS-M were nearly spherical particles with a diameter of 32.6 ± 1.1 nm and zeta potential of −21.3 ± 3.1 mV. Carmustine (CMS) released from Pep-1/Bor/CMS-M in pH 7.4 was significantly faster than in acidic environments. In human glioma BT325 cellular studies, Pep-1/Bor/CMS-M remarkably increased the cytotoxicity, notably improved the internalization, and effectively induced the cell apoptosis . Likewise, in human brain microvascular endothelial cells, Pep-1/Bor/CMS-M obviously promoted the cellular uptake, rapidly decreased the transepithelial electrical resistance, and thereby enhanced the ability of penetration. In orthotopic Luc-BT325 glioma tumor-bearing nude mouse models, the stronger fluorescence signal and longer retention were observed in brain tissues compared with other controls, after single administration of DiD-labeled Pep-1/Bor/M (DiD/Pep-1/Bor/M). Importantly, Pep-1/Bor/CMS-M displayed the strongest inhibition of tumor growth, the longest survival period, and low systemic toxicity in treating orthotopic glioma tumor-bearing nude mice. Simultaneous functionalization of Pep-1 and borneol offers a novel strategy for designing CMS-based nanomedicine and precisely treating glioma.

(+)-Borneol inhibits neuroinflammation and M1 phenotype polarization of microglia in epileptogenesis through the TLR4-NFκB signaling pathway

Shuo Li,Alhamdu Adamu,Yucai Ye,Fankai Gao,Rulin Mi,Guofang Xue,Zhaojun Wang

Objective: To investigate the effect of (+)-borneol on neuroinflammation and microglia phenotype polarization in epileptogenesis and its possible mechanism. Methods: Based on mouse models of status epilepticus (SE) induced by pilocarpine, and treated with 15 mg/kg (+)-borneol, western-blot was used to detect the expressions of NeuN, Iba-1, TLR4, p65 and p-p65 in the hippocampus. Immunofluorescence was used to detect the expression of apoptosis-related proteins Bax and Bcl-2. To explore the effect of (+)-borneol on microglia in vitro, we used the kainic acid-induced microglia model and the concentration of (+)-borneol was 25 μM according to CCK-8 results. The levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and interleukin-10 (IL-10) in the supernatant of each group was detected by ELISA. The nitric oxide (NO) content in the supernatant was detected by Griess method. The expressions of Iba-1 and TLR4-NFκB signaling pathway-related proteins (TLR4, p65, p-p65) were detected by Western-Blot. Immunofluorescence was used to detect microglia’s M1 and M2 phenotype polarization and the expression of Iba-1 and TLR4.Results: (+)-borneol reduced hippocampal neuronal injury, apoptosis, and microglia activation by inhibiting the TLR-NFκB signaling pathway in SE mice. TLR4 agonist LPS partially reversed the neuroprotective effect of (+)-borneol. In the KA-induced microglia model, (+)-borneol inhibited microglia activation, M1 phenotype polarization, and secretion of pro-inflammatory cytokines through the TLR4-NFκB signaling pathway. LPS treatment inhibited the therapeutic effects of (+)-borneol.Conclusion: (+)-borneol inhibits microglial neuroinflammation and M1 phenotype polarization through TLR4-NFκB signaling pathway and reduces neuronal damage and apoptosis in SE mice. Therefore, (+)-borneol may be a potential drug for epilepsy modification therapy.

Conjugation of vitamin E-TPGS and guar gum to carry borneol for enhancing blood–brain barrier permeability

Yude Liu,Lian Rao,Hongguan Zhang,Yanyou Cen,Kaili Cheng

Herb borneol is usually used in clinics for the treatment of central nervous system illness, for its ability of blood–brain barrier permeability, although its poor water solubility and poor bioavailability limit its clinical application to some degree. In this study, we developed a novel nanoparticle combining the benefits of vitamin E d-ɑ-tocopheryl poly(ethylene glycol) succinate (E-TPGS) (or TPGS) and guar gum to get TPGS-g-guar gum as a drug delivery system to carry borneol, which could improve the solubility of borneol and increase the drug-loading capacity efficiently. The results showed that TPGS-g-guar gum nanoparticles delivery system was suitable to carry borneol and release the drug effectively, and TPGS-g-guar gum/borneol nanoparticles would be a potential platform for improving the treatment of central nervous system illness and cerebrovascular disease.

Enhanced the treatment of ischemic stroke through intranasal temperature-sensitive hydrogels of edaravone and borneol inclusion complex

Chuanhui Teng,Wei Lv,Yuqin Chen,Linfeng Liu,Jiaqing Yin,Shengnan Li,Zhiyi Min,Qi Zhang,Weichong He,Kunfang Ma,Xinrui Li,Xiang Cao,Hongliang Xin

Since ischemic stroke occurs by a combination of multiple mechanisms, therapies that modulate multiple mechanisms are required for its treatment. The combination of edaravone (EDA) and borneol can significantly ameliorate the symptoms of neurological deficits in cerebral ischemia–reperfusion model in rats. In this study, the solubility of borneol and edaravone was improved by hydroxypropyl-β-cyclodextrin and PEG 400 . Furthermore, a nasal temperature-sensitive hydrogel containing both edaravone and borneol inclusion complex (EDA-BP TSG S ) was developed to overcome the obstacles of ischemic stroke treatment including the obstruction of the blood–brain barrier (BBB) and the unavailability and untimely of intravenous injection. The effectiveness of the thermosensitive hydrogel was investigated in transient middle cerebral artery occlusion/reperfusion model rats (MCAO/R). The results showed that EDA-BP TSG S could significantly alleviate the symptoms of neurological deficits and decrease the cerebral infarct area and the degree of brain damage. In summary, nasal EDA-BP TSG S is a secure and effective brain-targeting formulation that may provide a viable option for the clinical prophylaxis and treatment of ischemic stroke.
