Evaluation of viscosity and printing quality of chitosan-based flexographic inks: The effect of chitosan molecular weight

Hongxia Wang,Ting Guo,Houbin Li

Inks composed of renewable resources are important for the environment protection. We report preparation of a new type of edible chitosan-based flexographic ink. The performance of three inks, containing three different molecular weight (MW) chitosan, was analyzed by the different experimental techniques. The Ink viscosity was investigated from the parameters shear rate, time, temperature, and flexographic printing simulation. The printing quality on coated paper was studied by the scratch resistance, contact angle, print fastness, chromatic aberration, and dot gain. Viscosity recovery rate reached 69% after 6 s, and the print fastness was up to 91%. The experimental Δ E is 0.97, the dot gain is 23%, and contact angle is 69° for the best ink. Experimental results indicate that the developed ink is suitable for the flexographic printing with good application prospects. © 2016 Wiley Periodicals, Inc. J. Appl. Polym. Acceleration of Healing in Full-Thickness Wound by Chitosan-Binding bFGF and Antimicrobial Peptide Modification Chitosan Membrane

Hou Lin,Wang Wei,Wang Mei-Kun,Song Xue-Song

Skin wound healing is an important clinical challenge, and the main treatment points are accelerating epidermal regeneration and preventing infection. Therefore, it is necessary to develop a wound dressing that can simultaneously cure bacterial infections and accelerate wound healing. Here, we report a multifunctional composite wound dressing loaded with chitosan (CS)-binding bFGF (CSBD-bFGF) and antimicrobial peptides (P5S9K). First, CS was used as the dressing matrix material, and P5S9K was encapsulated in CS. Then, CSBD-bFGF was designed by combining recombinant DNA technology and tyrosinase treatment and modified on the dressing material surface. The results show that the binding ability of CSBD-bFGF and CS was significantly improved compared with that of commercial bFGF, and CSBD-bFGF could be controllably released from the CS dressing. More importantly, the prepared dressing material showed excellent antibacterial activity in vivo and in vitro and could effectively inhibit the growth of E. coli and S. aureus. Using NIH3T3 cells as cellular models, the results showed that the CSBD-bFGF@CS/P5S9K composite dressing was a friendly material for cell growth. After cells were seeded on the composite dressing surface, collagen-1 (COL-1) and vascular endothelial growth factor (VEGF) genes expression in cells were significantly upregulated. Finally, the full-thickness wound of the rat dorsal model was applied to analyse the tissue repair ability of the composite dressing. The results showed that the composite dressing containing CSBD-bFGF and P5S9K had the strongest ability to repair skin wounds. Therefore, the CSBD-bFGF@CS/P5S9K composite dressing has good antibacterial and accelerated wound healing abilities and has good application prospects in the treatment of skin wounds.

Ag@MOF-loaded chitosan nanoparticle and polyvinyl alcohol/sodium alginate/chitosan bilayer dressing for wound healing applications

Meng Zhang,Guohui Wang,Dong Wang,Yuqi Zheng,Yanxin Li,Wenqiao Meng,Xin Zhang,Feifan Du,Shaoxiang Lee

In this paper, Ag-Metal−organic framework loaded chitosan nanoparticles (0.1%Ag@MOF/1.5%CSNPs) and polyvinyl alcohol/sodium alginate/chitosan (PACS) were used as the upper and lower layers to successfully prepare a bilayer composite dressing for wound healing. The performance of bilayer dressing was evaluated. The lower layer (PACS) had uniform pore size distribution , good water retention, swelling, water vapor permeability, and biocompatibility while PACS had almost no antibacterial activity . The upper layer (Ag@MOF/CSNPs) possessed excellent antibacterial activity and poor biocompatibility. As the upper layer, it can avoid direct contact with the skin and inhibit microbial invasion. In addition, the bilayer can adhere to a large number of red blood cells and platelets, promoting blood coagulation and cell proliferation . Ag@MOF, CSNPs, Ag@MOF/CSNPs and bilayer showed antibacterial activity in ascending order, due to the synergistic antibacterial action of the upper and lower layer. In vivo evaluation showed that both bilayer and PACS could significantly accelerate the wound healing, and the bilayer dressing showed more complete re-epithelialization with less inflammatory cells. In summary, this new bilayer composite is an ideal dressing for accelerating wound healing.

Covalent and injectable chitosan-chondroitin sulfate hydrogels embedded with chitosan microspheres for drug delivery and tissue engineering

Ming Fan,Ye Ma,Huaping Tan,Yang Jia,Siyue Zou,Shuxuan Guo,Meng Zhao,Hao Huang,Zhonghua Ling,Yong Chen,Xiaohong Hu

Injectable hydrogels and microspheres derived from natural polysaccharides have been extensively investigated as drug delivery systems and cell scaffolds. In this study, we report a preparation of covalent hydrogels basing polysaccharides via the Schiff' base reaction. Water soluble carboxymethyl chitosan (CMC) and oxidized chondroitin sulfate (OCS) were prepared for cross-linking of hydrogels. The mechanism of cross-linking is attributed to the Schiff' base reaction between amino and aldehyde groups of polysaccharides. Furthermore, bovine serum albumin (BSA) loaded chitosan-based microspheres (CMs) with a diameter of 3.8–61.6   μm were fabricated by an emulsion cross-linking method, followed by embedding into CMC-OCS hydrogels to produce a composite CMs/gel scaffold. In the current work, gelation rate, morphology, mechanical properties, swelling ratio, in vitro degradation and BSA release of the CMs/gel scaffolds were examined. The results show that mechanical and bioactive properties of gel scaffolds can be significantly improved by embedding CMs. The solid CMs can serve as a filler to toughen the soft CMC-OCS hydrogels. Compressive modulus of composite gel scaffolds containing 20   mg/ml of microspheres was 13   KPa, which was higher than the control hydrogel without CMs. Cumulative release of BSA during 2   weeks from CMs embedded hydrogel was 30%, which was significantly lower than those of CMs and hydrogels. Moreover, the composite CMs/gel scaffolds exhibited lower swelling ratio and slower degradation rate than the control hydrogel without CMs. The potential of the composite hydrogel as an injectable scaffold was demonstrated by encapsulation of bovine articular chondrocytes in vitro. These results demonstrate the potential of CMs embedded CMC-OCS hydrogels as an injectable drug and cell delivery system in cartilage tissue engineering.

In vivo immuno-reactivity analysis of the porous three-dimensional chitosan/SiO2 and chitosan/SiO2/hydroxyapatite hybrids

Mengxia Guo,Yifan Dong,Jiangwei Xiao,Ruicai Gu,Maochao Ding,Tao Huang,Junhua Li,Naru Zhao,Hua Liao

Inorganic/organic hybrid silica-chitosan (CS) scaffolds have promising potential for bone defect repair, due to the controllable mechanical properties, degradation behavior, and scaffold morphology. However, the precise in vivo immuno-reactivity of silica-CS hybrids with various compositions is still poorly defined. In this study, we fabricated the three-dimensional (3D) interconnected porous chitosan-silica (CS/SiO 2 ) and chitosan-silica-hydroxyapatite (CS/SiO 2 /HA) hybrids, through sol–gel process and 3D plotting skill, followed by the naturally or freeze drying separately. Scanning electron microscopy demonstrated the hybrids possessed the uniform geometric structure, while, transmission electron microscopy displayed nanoscale silica, or HA nanoparticles dispersed homogeneously in the CS matrix, or CS/silica hybrids. After intramuscular implantation, CS/SiO 2 and CS/SiO 2 /HA hybrids triggered a local and limited monocyte/macrophage infiltration and myofiber degeneration. Naturally dried CS/SiO 2 hybrid provoked a more severe inflammation than the freeze-dried ones. Dendritic cells were attracted to invade into the implants embedded-muscle, but not be activated to prime the adaptive immunity, because the absence of cytotoxic T cells and B cells in muscle received the implants. Fluorescence-activated cell sorting (FACS) analysis indicated the implanted hybrids were incapable to initiate splenocytes activation. Plasma complement C3 enzyme linked immunosorbent assay (ELISA) assay showed the hybrids induced C3 levels increase in early implanting phase, and the subsequent striking decrease. Thus, the present results suggest that, in vivo , 3D plotted porous CS/SiO 2 and CS/SiO 2 /HA hybrids are relatively biocompatible in vivo, which initiate a localized inflammatory procedure, instead of a systematic immune response. © 2018 Wiley Periodicals, Inc. Evaluation of the effect of the chitosan/carrageenan ratio on lovastatin release from chitosan/carrageenan based biomaterials

Ha Manh Hung,Vu Quoc Manh,Vu Thi Thu Thao,Dao Thi Phuong Thuy,Pham Tien Dung,Nguyen Thi Bich Viet,Duong Khanh Linh,Nguyen Ngoc Linh,Doan Thi Yen Oanh,Nguyen Thuy Chinh,Thai Hoang,Vu Quoc Trung

The chitosan/carrageenan/lovastatin biomaterials were prepared using a solution technique with 5 % lovastatin and a variable chitosan/carrageenan mass ratio. Fourier transform infrared spectroscopy (FT-IR) and field emission scanning electron microscopy (FESEM) images revealed that the complex's components were well dispersed into each other without changing the chemical structure, forming dipole interactions and hydrogen bonds among them. The abilities to release Lovastatin (Lov) in pH 2 and pH 7.4 solutions, corresponding to the environments in stomach and intestinal fluids, respectively, are both affected by the ratio of polymer components. The best results were obtained with the chitosan/carrageenan/lovastatin (w/w/%w = 10/90/5%) (CsCL195) biomaterial sample in which the particles are evenly dispersed. While the drug release process was poor in the pH 2 solution (gastric juice), a good release observed in the pH 7.4 solution (intestinal fluid) allows facilitating drug absorption through the intestinal mucosa cells into the blood, from which the effects of hypercholesterolemia treatment can be obtained.

Sustainable Chitosan-Dialdehyde Cellulose Nanocrystal Film

Cong Gao,Shuo Wang,Baojie Liu,Shuangquan Yao,Yi Dai,Long Zhou,Chengrong Qin,Pedram Fatehi

In this study, we incorporated 2,3-dialdehyde nanocrystalline cellulose (DANC) into chitosan as a reinforcing agent and manufactured biodegradable films with enhanced gas barrier properties. DANC generated via periodate oxidation of cellulose nanocrystal (CNC) was blended at various concentrations with chitosan, and bionanocomposite films were prepared via casting and characterized systematically. The results showed that DANC developed Schiff based bond with chitosan that improved its properties significantly. The addition of DANC dramatically improved the gas barrier performance of the composite film, with water vapor permeability (WVP) value decreasing from 62.94 g·mm·m−2·atm−1·day−1to 27.97 g·mm·m−2·atm−1·day−1and oxygen permeability (OP) value decreasing from 0.14 cm3·mm·m−2·day−1·atm−1to 0.026 cm3·mm·m−2·day−1·atm−1. Meanwhile, the maximum decomposition temperature (Tdmax) of the film increased from 286 °C to 354 °C, and the tensile strength of the film was increased from 23.60 MPa to 41.12 MPa when incorporating 25 wt.% of DANC. In addition, the chitosan/DANC (75/25, wt/wt) films exhibited superior thermal stability, gas barrier, and mechanical strength compared to the chitosan/CNC (75/25, wt/wt) film. These results confirm that the DANC and chitosan induced films with improved gas barrier, mechanical, and thermal properties for possible use in film packaging.

Water-soluble chlorogenic acid-chitosan and polydatin-chitosan conjugates: antibacterial activity and inhibition of lipid and protein oxidation

Tianfang Duan,Zeru Xu,Shanbai Xiong,Hongying Du

BACKGROUND Chitosan (CS), an abundant alkaline polysaccharide, is valued for its biocompatibility, non-toxicity, and antibacterial properties. However, its limited solubility and modest antioxidant activity constrain its utility. Grafting polyphenols onto chitosan through the use of grafting reactions can enhance both the solubility and bioactivity of chitosan. Among the techniques employed, the free radical grafting method is favored for its simplicity, environmental sustainability, and its effectiveness in preserving biological activity. RESULTS In this study, chlorogenic acid (CGA) and polydatin (PLD) were conjugated successfully to chitosan by a Vc/H 2 O 2 redox system. Analytical techniques such as ultraviolet-visible (UV-visible) spectroscopy, fourier transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD), and proton nuclear magnetic resonance ( 1 H NMR) were employed to confirm the formation of covalent bonding between the polyphenol molecules and the chitosan backbone. The novel conjugates displayed superior antioxidant properties in comparison with pristine chitosan, as evidenced by their enhanced 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical, 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical, and hydroxyl radical scavenging capacities, and Fe 2+ reducing power. Both CGA-CS and PLA-CS exhibited excellent lipid and protein oxidation inhibition capabilities. Furthermore, the conjugates were shown to have significant antibacterial effects against four common pathogenic bacteria: Pseudomonas fluorescens , Pseudomonas aeruginosa , Pseudomonas putida , and Staphylococcus aureus ( P < 0.05). CONCLUSION The newly synthesized water-soluble polyphenol-chitosan conjugates demonstrated remarkable biological activity, particularly CGA-CS. This study offers new insights and a strong theoretical foundation for developing natural food preservation materials with potential applications in the food industry. © 2024 Society of Chemical Industry.

Nanocomposites derived from licorice residues cellulose nanofibril and chitosan nanofibril: Effects of chitosan nanofibril dosage on resultant properties

Wei Liu,Xiaoyu Li,Si Wang,Fengjun Fang,Xiao Wang,Qingxi Hou

Research on nanocomposite film from natural polymers such as cellulose and chitosan is of great importance to promote the development and highly efficient utilization of green and renewable bioresources. In this study, enzymatic pretreatment cellulose nanofibril (ETCNF) derived from licorice residues was prepared, and further processed into nanocomposite film with addition of chitosan nanofibril (CHN). This study focused on the effects of CHN dosage on the main properties of resultant nanocomposite film in terms of crystallinity, thermal stability , light transmittance, hydrophobicity , mechanical properties, and antibacterial activity . The results showed that ETCNF/CHN nanocomposite film exhibited good hydrophobicity especially at higher dosage of CHN, good light transmittance and mechanical properties (tensile strain can reach 39.6 MPa for ETCNF/CHN-10.0%). The as-prepared ETCNF/CHN nanocomposite film also showed good antibacterial activity against Escherichia coli . It was expected that the ETCNF/CHN nanocomposite film would help to realize transformation and high value-added utilization of these biomass residues.

Preparation of α-tocopherol-chitosan nanoparticles/chitosan/montmorillonite film and the antioxidant efficiency on sliced dry-cured ham

Wenjing Yan,Wenbin Chen,Umair Muhammad,Jianhao Zhang,Hong Zhuang,Guanghong Zhou

The aim of this work was to prepare a novel antioxidant film by incorporating α-tocopherol-chitosan nanoparticles (TOC-CSNPs) with chitosan/montmorillonite film (namely, TOC-CSNPs/CS/MMT), and investigate the antioxidant activity of TOC-CSNPs/CS/MMT film on sliced dry-cured ham in a period of 120 days at 4 °C. Positively charged TOC-CSNPs with the average sizes of 214 nm were prepared using TPP-initiated ionic gelation mechanism. The amount of TOC-CSNPs was optimized based on the changes of water vapor permeability (WVP), water solubility and swelling ratio of the film. The TOC-CSNPs/CS/MMT film showed 46.5% radical scavenging activity after 16 days of storage, which was 23 times and 27 times higher than that of CS/MMT and CS film, respectively. The antioxidant activity of TOC-CSNPs/CS/MMT film was evaluated by measuring the peroxide value (POV) and thiobarbituric acid reactive substances (TBARS) values of sliced dry-cured ham coated with the TOC-CSNPs/CS/MMT film-forming solutions. The results indicated that compared with the CS (0.42 g/100 g for POV, 0.65 mg/kg for TBARs) and CS/MMT (0.38 mg/kg for POV, 0.59 mg/kg for TBARs) films, the biceps femoris muscle of the sliced ham coated with TOC-CSNPs/CS/MMT film showed much lower values of POV (0.15 g/100 g) and TBARs (0.15 mg/kg) during 120 days of storage. These results provide a theoretical basis for developing efficient and long-term antioxidant approach, which is crucial for improving and enhancing the quality of food products.
