Structural Characterization and Properties of Pullulan/Curdlan Blend Films and the Coating for Cherry Tomato Preservation

Kai Chen,Yanlin Tian,Hong Qian,Shibo Qiu,Gang Wei,Kao Wu,Junping Yin,Fatang Jiang

Fruits and vegetables (F&V) are prone to moisture loss and deterioration at postharvest. The strategy of a composite coating based on a single chain-triple helix network was applied to inhibit water escape and respiration of F&V. In this study, pullulan (PUL) with a high oxygen barrier was incorporated with the hydrophobic curdlan (CUD) to prepare the PC solutions and films. The results indicated that the linear PUL molecules were entangled with the CUD triple-helix micelle network, forming a continuous, dense three-dimensional network based on hydrogen bonds, and showing good biocompatibility. The addition of CUD enhanced the surface roughness of PC films, and significantly improved the elongation at break by 89%, hydrophobicity, water vapor and oxygen barrier, and thermal stability. Overall, the P 7 C 3 coating was selected for the cherry tomato preservation, significantly delaying weight loss by 2.5% and ascorbic acid decline by 16.8%, maintaining firmness, total solids, and acid content. The coating significantly inhibited respiratory intensity and sustained the sensory attributes (appearance, texture, taste, and smell) of cherry tomatoes at a high level, which suggested that the P 7 C 3 coating can effectively extend the shelf life of cherry tomatoes. This work would provide a reference for the application of pullulan and curdlan complex in F&V preservation and show a good prospect.

Curdlan-polyphenol complexes prepared by pH-driven effectively enhanced their physicochemical stability, antioxidant and prebiotic activities

Huan Li,Saiqing Xu,Ying Xie,Qun Zhang,Shenghua Ding,Rongrong Wang,Fuhua Fu,Xiaobei Zhan

In this study, the curdlan-polyphenol complexes were constructed by a pH-driven method. The interaction between curdlan and various hydrophobic polyphenols (curcumin, quercetin, and chlorogenic acid) was investigated. Curdlan could self-assemble into particles for loading polyphenols through hydrogen bonding and hydrophobic interactions. The three polyphenols were embedded in curdlan in an amorphous state. The curdlan-curcumin complex showed the lowest viscoelasticity but exhibited the highest curcumin loading ability (34.04 ± 1.73 mg/g). However, the curdlan-chlorogenic acid complex emerged the opposite trend, indicating that the loading capacity was associated with the hydrophobicity of polyphenols. The antioxidant activity of curdlan significantly increased after combining with polyphenols, which could be maintained during in vitro simulated gastrointestinal digestion. In particular, the curdlan-quercetin complex exhibited the highest antioxidant activity and short-chain fatty acid concentration, which could influence gut microbiota composition by promoting the proliferation of Prevotella and inhibiting the growth of Escherichia_Shigella . In conclusion, the curdlan-polyphenol complexes prepared by an alcohol-free pH-driven method could effectively enhance the gastrointestinal stability of polyphenols as well as increase the antioxidant and prebiotic activities of curdlan, which could be applied as a functional ingredient to improve gut health.

Curdlan-Decorated Fullerenes Mitigate Immune-Mediated Hepatic Injury for Autoimmune Hepatitis Therapeutics via Reducing Macrophage Infiltration

Chenglong Fei,Lei Liu,Hedong Qi,Yuyang Peng,Jingfen Han,Chunru Wang,Xue Li

Autoimmune hepatitis (AIH) is a severe immune-mediated inflammatory liver disease whose standard of care is immunosuppressive treatment with inevitable undesired outcomes. Macrophage is acknowledged to aggravate liver damage, providing a promising AIH therapeutic target. Accordingly, in this study, a kind of curdlan-decorated fullerene nanoparticle (Cur-F) is fabricated to alleviate immune-mediated hepatic injury for treating AIH via reducing macrophage infiltration in a concanavalin A (Con A)-induced AIH mouse model. After intravenous administration, Cur-F primarily distributes in liver tissues, efficiently eliminates the excessive reactive oxygen species, significantly attenuates oxidative stress, and subsequently suppresses the nuclear factor kappa-B-gene binding (NF-κB) signal pathway, resulting in the lowered production of pro-inflammatory cytokines and the balancing of the immune homeostasis with the prevention of macrophage infiltration in the liver. The regulation of hepatic inflammation contributes to inhibiting inflammatory cytokines-induced hepatocyte apoptosis, decreasing the serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) contents and thus ameliorating immune-mediated hepatic injury. Notably, there is no detectable toxicity to the body. Our findings may open up novel avenues for AIH based on curdlan and fullerene materials.

Effect of curdlan and xanthan polysaccharides on the pasting, rheological and thermal properties of rice starch

Chen Tao,Fang Sheng,Zuo Xiaobo,Liu Yanmei

The effects of microbial polysaccharides xanthan and curdlan on the pasting, viscoelastic properties and retrogradation of rice starch were studied. The additions of xanthan and curdlan increased the peak viscosities and decreased the final viscosities. The breakdown viscosities were increased by the curdlan addition, but were decreased by the xanthan addition. The dynamic viscoelastic results showed that the addition of xanthan in rice starch significantly increased storage modulus and loss modulus. The rice starch gel with xanthan addition exhibited higher resistances to the stress and produced a stronger microstructure network. The creep recovery data were well fitted by a 4-element Burger’s model. Differential scanning calorimetry showed that the addition of curdlan and xanthan decreased the melting enthalpy values and retarded the retrogradation of the rice starch gel during storage at 4 °C for 12 days. The pasting and thermal results demonstrated that the addition of xanthan could significantly inhibited the retrogradation of rice starch. It was concluded that the addition of curdlan and xanthan modified the rheology of rice starch gel in different ways and interacted under different models based on their molecular structures.

Study of alkali-soluble curdlan/bacterial cellulose/cinnamon essential oil blend films with enhanced mechanical properties

Luyao Bian,Jingchao Fu,Tingting Chang,Chuang Zhang,Chong Zhang

In response to the growing demand for biodegraded film with high mechanical properties for complex preservation application scenarios, we developed a curdlan (CD) blended films with exceptional mechanical strength through an alkali dissolution method. Notably, the alkali-soluble CD film exhibited five-fold increase in tensile strength (TS) when compared to its water-soluble counterpart. Furthermore, the inclusion of 2 % bacterial cellulose (BC) resulted in a significant 41.1 % augmentation of the film's TS. Thermal stability improvements were observed through differential scanning calorimetry (DSC) analysis and thermogravimetric analysis (TGA). X-ray diffraction (XRD) and Fourier-transform infrared spectroscopy (FTIR) results provided insights into alterations in film crystallinity and intermolecular interactions. Specifically, the incorporation of 10 % CEO led to an additional improvement in TS. Our experimental investigations involving the packaging of chilled fresh meat with these blended films unveiled their capacity to effectively inhibit microorganism growth, maintain meat color stability, delay protein decomposition and fat oxidation , and extend the storage time up to 9 days. Our study offers a promising solution for food packaging, emphasizing the development of a high-strength degradable CD/BC/CEO blended film, which holds potential for extending the shelf life of food products.

Double network hydrogel based on curdlan and gelatin methacryloyl (GelMA) with enhanced mechanical and tissue adhesive properties

Wenlong Yuan,Yitong Sun,Bo Du,Ruiping Duan,Yanping Shi,Lingrong Liu,Xuemin Li

Gelatin methacryloyl (GelMA) hydrogel based tissue-adhesives have attracted extensive attraction due to its high biocompatibility, however, it suffers from insufficient mechanical properties, low adhesion strength due to the single network. In this study, a novel double-network hydrogel (DN-GelMA-Cur) consisting of GelMA and curdlan (Cur) was developed, where the network composed by triple helix fibers of Cur induced by heating and annealing serves as the hard network, while photo-crosslinked GelMA serves as the flexible network. A series of characterizations of the hydrogels, including hydrogel-forming ability, swelling behavior, morphology, rheological and mechanical properties, tissue adhesion, and biocompatibility were performed. It was found that precursor solution, especially with lower ratios of Cur (Cur: GelMA, 2:8 and 1:9), showed better hydrogel forming ability. Those hydrogels with lower ratios of Cur had better structural maintaining abilities, higher elastic modulus compared with pure-GelMA hydrogel. Particularly, addition of lower ratios of Cur could significantly enhance tissue-adhesion strength of GelMA hydrogel. DN-GelMA-Cur hydrogels showed good cytocompatibilities, especially hydrogels with lower ratios of Cur could promote proliferation of rat peritoneal fibroblasts significantly. Therefore, DN-GelMA-Cur hydrogels with lower ratio of Cur have potential application as novel GelMA hydrogel based tissue-adhesives with enhanced mechanical and bio-adhesive properties.

Preparation and characterization of curdlan-chitosan conjugate nanoparticles as mucosal adjuvants for intranasal influenza H1N1 subunit vaccine

Honglei Jiang,Shu Zhang,Yipan Chen,Fengshan Wang,Wenjie Jiang

Intranasal vaccination offers crucial protection against influenza virus pandemics. However, antigens, especially subunit antigens, often fail to induce effective immune responses without the help of immune adjuvants. Our research has demonstrated that a polyelectrolyte complex, composed of curdlan sulfate/ O -(2-hydroxyl) propyl-3-trimethyl ammonium chitosan chloride (CS/ O -HTCC), effectively triggers both mucosal and systemic immune responses when administrated intranasal. In this study, stable nanoparticles formed by curdlan- O -HTCC conjugate (C O NP) were prepared and characterized. Furthermore, the efficacy of C O NP was evaluated as a mucosal adjuvant in an intranasal influenza H1N1 subunit vaccine. The results revealed that C O NP exhibits uniform and spherical morphology, with a size of 190.53 ± 4.22 nm, and notably, it remains stable in PBS at 4 °C for up to 6 weeks. Biological evaluation demonstrated that C O NP stimulates the activation of antigen-presenting cells (APCs), including macrophages and dendritic cells (DCs), both in vitro and in vivo . Furthermore, intranasal administration of C O NP effectively elicits cellular and humoral immune responses, notably enhancing mucosal immunity. Thus, C O NP emerges as a promising mucosal adjuvant for influenza subunit vaccines.

AMP functionalized curdlan nanoparticles as a siRNA carrier: Synthesis, characterization and targeted delivery via adenosine A2B receptor

Qingming Bao,Tsogzolmaa Ganbold,Qiburi Qiburi,Mingming Bao,Shuqin Han,Huricha Baigude

Receptor-mediated endocytosis has been used for tissue targeted delivery of short interfering RNA (siRNA) drugs. Herein, we investigated adenosine receptor (AR) as a candidate for receptor-mediated siRNA internalization. We synthesized adenosine functionalized cationic curdlan derivatives (denote CuAMP polymers). One of these polymers, CuAMP4, efficiently delivered siRNA to breast cancer cells expressing high level of A 2B receptor. The internalization of siRNA loaded CuAMP4 by cancer cells was inhibited by free AMP as well as endocytosis inhibitors. Moreover, knockdown of A 2B R by siRNA, or pre-treatment of the cells with anti-A 2B R antibody, strongly inhibited the cellular uptake of CuAMP4. Our findings confirmed that A 2B R can be utilized for cell type specific siRNA delivery, and CuAMP4 NP may be a promising delivery system for cancer cell targeted delivery of therapeutic siRNAs.

Construction and characterization of folate-functionalized curdlan-trilysine siRNA delivery platform for in vivo hepatic carcinoma treatment

Yuxuan Qi,Yiwen Pan,Feng Gu,Shuai Wei,Chenglong Fei,Jingfen Han

RNA interference technology is a powerful tool with substantially clinical prospects for carcinoma therapy, in which efficiency and specificity of delivery of dsRNA remains a critical issue. Herein, aiming at delivery of dsRNA in efficient and safe way, we constructed targeting delivery platform (CTL-PEG-FA) by grafting curdlan with trilysine through click reaction, then modifying with PEG linked folic acid. The CTL-PEG-FA vector exhibited excellent gene binding capacity to condense siRNA and dramatically reduced cytotoxicity. Increased cell uptake of CTL-PEG-FA/Bcl-2 siRNA was achieved by the synergism of folate mediated endocytosis and charge interaction, and further causing severe HepG2 cells injury through apoptosis mechanism after down-regulation of Bcl-2 protein. In vivo experiments, CTL-PEG-FA/Bcl-2 siRNA complex distinctly accumulated in tumor site and significantly inhibited the growth of tumor, while no obvious toxicity was observed. Therefore, well-performed CTL-PEG-FA with excellent biocompatibility, has the potential to be the candidate of gene therapy for clinical applications.

Preparation and cell activities of lactosylated curdlan-triornithine nanoparticles for enhanced DNA/siRNA delivery in hepatoma cells

Xia Wang,Yuxuan Qi,Lixia Liu,Tsogzolmaa Ganbold,Huricha Baigude,Jingfen Han

A long-anticipated cancer therapy would deliver the right type of therapeutic agents to the target in control with minimal systemic toxicity. The purpose of this study was to prepare lactosylated curdlan-triornithine nanocarriers (CTOLs), and target deliver gene to hepatoma cells. Structures and biophysical properties had been elucidated with physical and chemical methods. The results revealed that those functionalized polymers can completely condense the gene into spherical nanoparticles. Cytotoxicity assay, GFP-pDNA and siRNA transfection in vitro were implemented successively. Observations showed that CTOL 20% with the highest lactose acid substitution degree targeted delivered gene into HepG2 cells over expressing ASGPR receptors and had pretty gene knockdown efficiency over 70%. Meanwhile, the carriers showed excellent biocompatibility. Our studies demonstrated that CTOLs with lower toxicity and higher gene binding capacity may serve as a potential valuable platform that can be tailored to target the liver cancer cells for therapeutic gene.

Encapsulation of Curcumin using Curdlan-whey Protein Isolate Composite Gels: Gel Properties, Stability and in vitro Release Performance

Hu Wenmei,Qin Xianglin,Huang Kai,Ren Erfang,Miao Yifan,Liao Siru,Zhang Siqi,Chen Shan

This research primarily investigates the encapsulation of curcumin within curdlan-whey protein isolate(CUR-WPI) composite gels and the subsequent release of the encapsulated compound. Results demonstrated that the composite gel could effectively encapsulate curcumin, with the highest encapsulation rate reaching 83.6%. The main force for the gel to encapsulate curcumin was electrostatic interaction and hydrogen bond. Fluorescence spectroscopy showed that curcumin can bind to WPI through hydrophobic interactions and suppress WPI's natural fluorescence. The composite gel exhibits excellent water-holding capacity (WHC) and the favorable texture profile analysis (TPA), which improved the photostability (2.57 times) and storage stability of curcumin (6.09 times). Moreover, the release rate of curcumin from the composite gel in intestinal fluid was higher than that in the stomach, which was related to the non-digestible characteristics of the curdlan and the dense network structure of the gel. The release kinetics results indicated that the main release mechanism of curcumin in the composite gel during the simulated digestion process was the diffusion mechanism. In conclusion, CUR-WPI composite gel is a good delivery carrier for curcumin, indicating its potential value for application in functional dairy products. Graphical

Fabrication and characterization of bioactive curdlan and sodium alginate films for enhancing the shelf life of Volvariella volvacea

Asif Ali Khan,Feng-Jie Cui,Muhammad Wajid Ullah,Abdul Qayum,Ibrahim Khalifa,Syed Asim Shah Bacha,Zheng-Zi Ying,Ismail Khan,Umar Zeb,Abdullah A. Alarfaj,Mohammad Javed Ansari,Yan Yang,Wei Jia,Wen Li

This study aimed to develop bioactive films based on curdlan (CD) and sodium alginate (SA) to enhance the shelf life of the Volvariella volvacea mushroom. A stable nanoemulsion of droplet size of 150.1 ± 5 nm was first formulated using 5% soy protein (Sp) and 10% lavender essential oil (LEO), achieved by ultrasound treatment for 10 min, resulting in small droplet sizes. In parallel, a CD-SA film was prepared by mixing them in a 1:1 ratio. The SpLEO nanoemulsion was then ex situ added to the CD-SA film at varying concentrations (4%, 6%, 8%, and 10%). The resulting bioactive film exhibited desirable properties, including water vapor permeability (1.15 ± 0.05 × 10 −10  g m −1 Pa −1 s −1 ), swelling rate (547%), and tensile strength (10.1 ± 1.3 MPa). Docking and structural analyses suggested that ultrasound treatment enhanced the intermolecular hydrogen bonding, creating a uniform and regular surface and reduced crystallinity. The CD-SA film with ultrasound-treated 8% SpLEO nanoemulsion [i.e., (CD-SA/SpLEO-8)US] showed antibacterial activity against Escherichia coli and Staphylococcus aureus and extended the freshness of straw mushrooms prevention of spoilage during storage. Additionally, the film reduced respiration rate (975 mg CO₂/kg/h at 96 h) and ethylene production (160 ng/kg/s by 96 h) while effectively inhibiting straw mushroom autolysis. This led to a reduction in browning (2.91), total soluble solids (7.5%), weight loss (13.9%), and polyphenol oxidase activity (0.08 ΔOD 420 /min⋅g), thereby extending the shelf life of the mushrooms to 96 h. These findings highlight the potential of incorporating different molecules using ultrasound treatment to develop innovative edible films for food preservation.
