Biodegradation mechanisms and detoxification profiles of zearalenone by Clonostachys and Gliocladium biocontrol strains

Zongping Sun,Yaohuan Zhu,Yonghui Mu,Junjie Yu,Yingying Wang,Juan Lu,Jun Tang

Zearalenone (ZEA) frequently contaminates cereals and poses risks to human and animal health, highlighting the need for effective detoxification approaches. In this study, the biocontrol agents Clonostachys rosea , Gliocladium penicillioides , and G. catenulatum were shown to tolerate 50 μg/mL ZEA, while G. deliquescens and G. viride showed sensitivity to 10 μg/mL ZEA. The tolerant strains exhibited comprehensive degradation capacity against ZEA and its toxic derivatives (α/β-zearalanol and α/β-zearalenol), transforming 20 μg/mL ZEA within 48 h. Notably, C. rosea showed the highest degradation capacity, not only transforming ZEA faster but also degrading over 95.5 % of 50 μg/mL α/β-zearalanol and α-zearalenol, along with 36.2–54.4 % β-zearalenol within 72 h. Additionally, the mRNA levels of zhdr52 and zhdp83 were significantly upregulated during the biotransformation process, suggesting their involvement in this process. ZEA degradation products were identified as hydrolyzed ZEA (H-ZEA) and decarboxylated hydrolyzed ZEA ( D -ZEA) by LC-MS analysis. Importantly, the degradation products H-ZEA and D -ZEA showed markedly reduced cytotoxicity compared to ZEA, as evidenced by the absence of significant cytotoxic effects on normal human hepatocytes and renal cells in CCK-8 assays. This study clarifies the ZEA biodegradation mechanisms of C. rosea , G. penicillioides , and G. catenulatum , identifying three promising fungal candidates for ZEA biocontrol and advancing the development of ZEA detoxification strategies.

Targeting UXS1-Dependent Glucuronate Detoxification Potentiates Metformin's Anti-Tumor Efficacy in Lung Adenocarcinoma

Qihai Sui,Zhencong Chen,Guangyao Shan,Zhengyang Hu,Xing Jin,Jiaqi Liang,Yanjun Yi,Jiacheng Yin,Haochun Shi,Xifei Jiang,Junjie Xi,Zongwu Lin,Cheng Zhan,Fenghao Sun,Wei Jiang

Despite the widely reported experimental anti-tumor effects, metformin's role remains exceedingly complex, with contradictory results in clinical trials. Our study, based on metabolomics analysis of lung adenocarcinoma (LUAD) samples, xenografts, and cells, unveils a novel process that metformin promotes the conversion of UDP-glucose (UDPG) to UDP-glucuronic acid (UDPGA) in glucuronic acid metabolism. Mechanistically, metformin activates UDP-glucose 6-dehydrogenase (UGDH) through AMPK-mediated phosphorylation of UGDH(S476), a previously unstudied phosphorylation site, impeding the binding of UDP-Xyl to UGDH and the subsequent allosteric inhibition. Consequently, metformin-treated cells are more reliant on UXS1, a downstream metabolic enzyme of UGDH, for detoxifying UDPGA based on the “kitchen-sink” model. Through comprehensive virtual screening of a compound library, we identified that plantainoside is a potent UXS1-targeting agent. Remarkably, when combined with metformin, plantainoside exhibits a superior synergistic lethal effect in LUAD cells, organoids, xenografts, and spontaneous models. Moreover, this combination not only directly targets tumor cells but also synergistically boosts CD8+ T cells and suppresses the differentiation of macrophages, thereby significantly enhancing immunotherapy efficacy. Collectively, our results shed light on metformin's complicated role by revealing its novel impact on glucuronic acid metabolism and dependence on UXS1; thus, targeting UXS1 combined with metformin represents a highly promising new strategy.

Zr-MOF/Hydrogel Composite Membrane for Solid-Phase Organophosphate Detoxification under Ambient Conditions

Hong-Bin Luo,Kai-Xin Zhao,Xin-Ru Wu,Jiang-Long Chen,Guo-Qin Zhang,Zhi-Xing Han,Yangyang Liu,Xiao-Ming Ren

Zirconium-based metal–organic frameworks (Zr-MOFs) have shown great promise as effective catalysts for the hydrolytic degradation of organophosphorus nerve agents. However, their high catalytic activity was usually observed in aqueous environments or high humidity conditions, challenging their applicability as chemical protective materials. In this study, we developed hydrogel composite membranes integrating Zr-MOF catalysts with multifunctional polymeric components, enabling the efficient solid-phase degradation of the nerve agent simulant, dimethyl-4-nitrophenyl phosphate (DMNP), under ambient conditions (40% relative humidity). These membranes were prepared by embedding MOF-808 within a hybrid polymer matrix composed of poly(1-vinylimidazole) (PVIM) and poly(vinyl alcohol) (PVA). In this rationally designed material, MOF-808 supplies the catalytic sites for hydrolysis, PVA contributes to water retention, and PVIM acts as a nonvolatile base to buffer the reaction environment. The resulting composite membranes demonstrate high catalytic activity and good recyclability for solid-phase DMNP hydrolysis under ambient conditions, surpassing the performance of many previously reported Zr-MOF-based materials under similar conditions. These findings represent a meaningful advancement toward the deployment of Zr-MOFs in practical chemical defense applications.

Biodegradation and detoxification of neonicotinoid insecticide thiamethoxam by white-rot fungus Phanerochaete chrysosporium

Anwei Chen,Wenjie Li,Xiaoxiao Zhang,Cui Shang,Si Luo,Ruoyu Cao,Doudou Jin

The extensive use of neonicotinoid pesticides in the past two decades caused serious impacts on many kinds of living beings. Therefore, it has been strongly suggested to detoxify and eliminate neonicotinoids’ residual levels in environment. Here, the degradation and detoxification of thiamethoxam (THX) by white-rot fungus Phanerochaete chrysosporium was conducted. Results shown that P. chrysosporium can tolerate THX and degraded 49% of THX after incubation for 15 days, and then 98% for 25 days at the initial concentration of 10 mg/L, which indicates the excellent degradation ability of this fungus to THX. Based on the by-products identified, THX underwent dechlorination , nitrate reduction , and C–N cleavage between the 2-chlorothiazole ring and oxadiazine. ( Z )-N-(3-methyl-1,3,5-oxadiazinan-4-ylidene)nitramide and 3-methyl-1,3,5-oxadiazinan-4-imine were identified as the main metabolites. The impacts of THX and its corresponding degradation intermediates on the growth of E. coil and Microcystis aeruginosa as well as the germination of rape and cabbage demonstrated that P . chrysosporium effectively degrades THX into metabolites and reduces its biotoxicity. The present work demonstrates that P . chrysosporium can be effectively used for degradation and detoxification of THX.

Triclosan detoxification through dechlorination and oxidation via microbial Pd-NPs under aerobic conditions

Yuan Chen,Yuancai Chen,Jianbo Jia,Bing Yan

The present study focuses on the successful preparation of microbial palladium nanoparticles (Pd-NPs). The even distribution of Pd in the periplasmic space of B. megaterium Y-4 cells is characterized using a transmission electronic microscopy (TEM) and scanning electron microscope (SEM). X-Ray photoelectron spectroscopy (XPS) and X-ray diffraction (XRD) revealed the domination of Pd (0) in Pd-NPs. The microbial Pd-NPs were selected to detoxify triclosan (TCS). Liquid chromatography-mass spectrometry (LC-MS) was used to analyze the intermediate products of dechlorination and oxidization. Free radicals quenching and 5,5-dimethyl-1-pyrroline N-oxide (DMPO) capturing experiments confirmed the crucial contribution of atomic H• and O 2 ·– to TCS degradation. Besides, TCS degradation by microbial Pd-NPs could alleviate the cytotoxicity of TCS polluted water. Meanwhile, great circulating utilization of microbial Pd-NPs was obtained in degrading TCS. Corresponding findings in the present study could provide new insight into the role of microbial Pd-NPs in detoxifying pollutants.

Functional Characterization of Two Carboxylesterase Genes Involved in Pyrethroid Detoxification in Helicoverpa armigera

Yong-Qiang Li,Li-Sha Bai,Cai-Xia Zhao,Jing-Jing Xu,Zhong-Juan Sun,Yan-Ling Dong,De-Xian Li,Xi-Li Liu,Zhi-Qing Ma

Insect carboxylesterases are major enzymes involved in metabolism of xenobiotics including insecticides. Two carboxylesterase genes, CarE001A and CarE001H, were cloned from the destructive agricultural pest Helicoverpa armigera. Quantitative real-time polymerase chain reaction showed that CarE001A and CarE001H were predominantly expressed in fat body and midgut, respectively; developmental expression analyses found that the expression levels of both CarEs were significantly higher in fifth-instar larvae than in other life stages. Recombinant CarE001A and CarE001H expressed in the Escherichia coli exhibited high enzymatic activity toward α-naphthyl acetate. Inhibition assays showed that organophosphates had strong inhibition on CarEs activity compared to pyrethroids. Metabolism assays indicated that CarE001A and CarE001H were able to metabolize β-cypermethrin and λ-cyhalothrin. Homology modeling and molecular docking analyses demonstrated that β-cypermethrin could fit nicely into the active pocket of both carboxylesterases. These results suggested that CarE001A and CarE001H could play important roles in the detoxification of pyrehtroids in H. armigera.

Uptake, tolerance, and detoxification mechanisms of antimonite and antimonate in Boehmeria nivea L

Yi Lu,Zhaoxue Zhang,Yingyang Wang,Fangyuan Peng,Zhaoguang Yang,Haipu Li

Boehmeria nivea L. (ramie) is a promising phytoremediation plant for antimony (Sb)-contaminated soils. However, the uptake, tolerance, and detoxification mechanisms of ramie to Sb, which are the basis for finding efficient phytoremediation strategies, remain unclear. In the present study, ramie was exposed to 0, 1, 10, 50, 100, and 200 mg/L of antimonite (Sb(III)) or antimonate (Sb(V)) for 14 days in hydroponic culture . The Sb concentration , speciation, subcellular distribution, and antioxidant and ionomic responses in ramie were investigated. The results illustrated that ramie was more effective in the uptake of Sb(III) than Sb(V). Most of the Sb accumulated in ramie roots, with the highest level reaching 7883.58 mg/kg. Sb(V) was the predominant species in leaves, with 80.77–96.38% and 100% in the Sb(III) and Sb(V) treatments, respectively. Immobilization of Sb on the cell wall and leaf cytosol was the primary mechanism of accumulation. Superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) contributed significantly to root defense against Sb(III), while CAT and glutathione peroxidase (GPX) were the major antioxidants in leaves. CAT and POD played crucial roles in the defense against Sb(V). B, Ca, K, Mg, and Mn in Sb(V)-treated leaves and K and Cu in Sb(III)-treated leaves may be related to the biological processes of Sb toxicity mitigation. This study is the first to investigate the ionomic responses of plants toward Sb and could provide valuable information for the phytoremediation of Sb-polluted soils.

Natural Coumarin Shows Toxicity to Spodoptera litura by Inhibiting Detoxification Enzymes and Glycometabolism

Tao Xia,Yan Liu,Zhanjun Lu,Haizhong Yu

Coumarin and its derivatives are plant-derived compounds that exhibit potent insecticidal properties. In this study, we found that natural coumarin significantly inhibited the growth and development ofSpodoptera lituralarvae through toxicological assay. By transcriptomic sequencing, 80 and 45 differentially expressed genes (DEGs) related to detoxification were identified from 0 to 24 h and 24 to 48 h inS. lituraafter coumarin treatment, respectively. Enzyme activity analysis showed that CYP450 and acetylcholinesterase (AChE) activities significantly decreased at 48 h after coumarin treatment, while glutathioneS-transferases (GST) activity increased at 24 h. Silencing ofSlCYP324A16gene by RNA interference significantly increasedS. lituralarval mortality and decreased individual weight after treatment with coumarin. Additionally, the expression levels of DEGs involved in glycolysis and tricarboxylic acid (TCA) cycle were inhibited at 24 h after coumarin treatment, while their expression levels were upregulated at 48 h. Furthermore, metabonomics analysis identified 391 differential metabolites involved in purine metabolism, amino acid metabolism, and TCA cycle from 0 to 24 h after treated with coumarin and 352 differential metabolites associated with ATP-binding cassette (ABC) transporters and amino acid metabolism. These results provide an in-depth understanding of the toxicological mechanism of coumarin onS. litura. Keywords:Spodoptera litura;coumarin;transcriptome sequencing;metabolome sequencing;RNA interference

A common mechanism of detoxification for lambda-cyhalothrin and abamectin in Cydia pomonella

Ju Di,Hu Chao,Li Peirong,Gao Ping,Li Yuting,Yang Xueqing

Background The primary method utilized by orchard owners to combat Cydia pomonella is the application of various chemical insecticides. However, this has resulted in the development of resistance. The resistance mechanisms to insecticides from different chemical classes are diverse but interconnected. Therefore, it is crucial to comprehend the commonalities in these mechanisms to effectively develop strategies for managing resistance.Materials and methodsTo determine whether target-site insensitivity to LCT and AM plays a role in resistance, the sequences of voltage-gated sodium channel (VGSC) and glutamate-gated chloride channel (GluCl) containing the mutation domains were detected. To validate whether similar mechanisms were involved in the detoxification process of lambda-cyhalothrin (LCT) and abamectin (AM) at sublethal doses (specifically LD10 and LD30), cytochrome P450 monooxygenases (P450), glutathione S-transferases (GST), and carboxylesterases (CarE) activities were evaluated after insecticides exposure; synergistic experiments were conducted using piperonyl butoxide (PBO), diethyl maleate (DEM), and triphenyl phosphate (TPP) as inhibitors of P450, GST, and CarE respectively. RNA-sequencing (RNA-Seq) was performed to compare the expression levels of detoxification-related genes between susceptible (SS) and resistant strains. Results The best known target-site mutations caused by LCT and AM, including L1014F in VGSC and V263I, A309V, I321T, and G326E in GluCl were not occurred. We observed that PBO had a strong synergistic effect on LCT and AM, while DEM on LCT. The activities of detoxification enzymes increased after insecticide exposures, indicating that the detoxification of LCT was primarily carried out by P450 and CarE enzymes, while P450 and GST enzymes played a major role in the detoxification of AM. A total of 72 P450 genes and 75 CarE genes were identified in the C. pomonella transcriptome, with 43 of these genes (including 11 P450, 3 GST, 10 CarE, 11 ABC transporters, and 8 UDP-glycosyl transferases) being over-expressed in response to both insecticides Interestingly, ABC transporters were predominantly induced by AM treatment, while GST showed higher induction levels with LCT treatment. Furthermore, LCT-resistant strains of C. pomonella exhibited higher levels of induction of detoxification-related genes compared to susceptible strains. Conclusion The up-regulation of these detoxification genes is a common metabolic mechanism employed by C. pomonella to counteract the effects of insecticides, although the extent of gene expression change varies depending on the specific insecticide.

Pd0 FOF1-ATPase prone pump modulated-detoxification of oxytetracycline in Bacillus megaterium Y-4

Linya Yi,Shuyan Chen,Yuancai Chen

Bio-remediation are low-cost and environmentally friendly, but microbial enzymes tend to lose their activity when treated antibiotic contaminants due to its high toxicity. In this study, Bacillus megaterium Y-4 was demonstrated to in situ synthesize Pd 0 @cells, and detoxify oxytetracycline (OTC) through enhancing electron transport chain via Pd 0 -dependent F O F 1 -ATPase (proton pumps). Toxicity analyses showed that increasing Pd 0 loading significantly reduced acute and chronic bio-toxicity as well as ecological risk of OTC effluent, as evidenced by the promoting cellular response to oxidative stress with increasing enzymes GSH-Px and IDH activities (4.24 and 1.20-fold). Electrochemical (I-t, LSV, DPV and EDC) showed that the incorporated bio-Pd 0 mainly promoted FDH/Hase-based short chain (gradually took precedence over CoQ-based long chain), diverting OTC extracellular degradation to markedly detoxify OTC to cell. Inhibition (DCCD and TBA) assays and structural equation modelling indicated that bio-Pd 0 upregulated ATP and NADH synthesis. It also acted as transmembrane F O F 1 -ATPase proton pump, which enabled the backward of protons out of the restriction of PMF , eventually maximizing the energy utilization. This study sheds new insights into the detoxification of antibiotics by bio-nanoparticles through proton transmembrane transfer/redistribution.

Simultaneous concentration and detoxification of lignocellulosic hydrolyzates by vacuum membrane distillation coupled with adsorption

Yaqin Zhang,Ming Li,Yafei Wang,Xiaosheng Ji,Lin Zhang,Lian Hou

Low sugar concentration and the presence of various inhibitors are the major challenges associated with lignocellulosic hydrolyzates as a fermentation broth. Vacuum membrane distillation (VMD) process can be used to concentrate sugars and remove inhibitors (furans) efficiently, but it’s not desirable for the removal of less volatile inhibitors such as acetic acid. In this study, a VMD-adsorption process was proposed to improve the removal of acetic acid, achieving simultaneous concentration and detoxification of lignocellulosic hydrolyzates by one step process. Results showed that sugars were concentrated with high rejections (>98%) and little sugar loss (<2%), with the significant reduction in nearly total furans (99.7%) and acetic acid (83.5%) under optimal operation conditions. Fermentation results showed the ethanol production of hydrolyzates concentrated and detoxified using the VMD-adsorption method were approximately 10-fold greater than from untreated hydrolyzates.

Highly Efficient Reductive Detoxification of Sulfamethoxazole by Palladium Nanoparticles Deposited on H2-Transfer Membranes

Qiu-Jin Xu,Ling-Dong Shi,He-Ping Zhao

As one of the most widely used antibiotics, sulfamethoxazole (SMX) is frequently detected in wastewater, imposing a high risk to human health. Current biological and chemical methods for SMX removal are generally lacking in efficiency or are cost-intensive. Here, we demonstrated highly efficient catalytic SMX removal by using a H2-based membrane palladium-film reactor (H2-MPfR), which delivered H2 directly to palladium (Pd0) nanoparticles through bubbleless hollow fibers. The SMX removal rate using this system could reach up to 4.25 mg/L per h, much higher than many other biological or chemical reduction processes. Low H2 partial pressure (e.g., 0.025 MPa under the experimental condition) was proved sufficient for high-rate SMX removal, while appropriate Pd dosage (e.g., 100 mg/dm2) and hydraulic retention time were favorable for maintaining the efficiency of SMX removal. Furthermore, high-performance liquid chromatography and quadrupole-time-of-flight mass spectrometry analyses revealed that the H2-MPfR may remove SMX by destroying the isoxazole ring, and the transformation products have a lower toxicity than SMX. Overall, this study sheds light on catalytic SMX degradation and is helpful in developing cost-effective strategies for removing antibiotics from wastewater.
