The acute effects of erythromycin and oxytetracycline on enhanced biological phosphorus removal system: shift in bacterial community structure

Hu Zhetai,Sun Peide,Han Jingyi,Wang Ruyi,Jiao Liang,Yang Pengfei,Cai Jing

Since extensive application, an increasing amount of antibiotics has been released into wastewater treatment plants. In this study, the enhanced biological phosphorus removal (EBPR) system was fed with synthetic wastewater containing erythromycin (ERY) and oxytetracycline (OTC) for 7 days to evaluate the variations of solution ortho-P (SOP), volatile fatty acid (VFA), poly-bhydroxyalkanoates (PHAs), specific oxygen uptake rater (SOUR), and microbial community in the EBPR system. The obtained results showed that the P-removal efficiency decreased to 0.0%, and at the end of the experiment, only less than 20% of the VFA could be consumed. Besides, the variable processes of P and PHAs were destroyed. Moreover, to better grasp the inhibitory mechanism of antibiotics, microbial community compositions of activated sludge sampled in all reactors were investigated by high-throughput sequencing techniques. Results of comparative and evolutionary analysis revealed that high concentrations (5 and 10 mg/L) of ERY and OTC could seriously shift microbial communities, while combined antibiotics could induce more. Additionally, Accumulibacter and Competibacter were two primary microorganisms at the genus level in the EBPR system. Accumulibacter decreased seriously for exposure to antibiotics, while Competibacter increased in all experimental reactors especially in combined antibiotics reactor.

Effects of erythromycin on biofilm formation and resistance mutation of Escherichia coli on pristine and UV-aged polystyrene microplastics

Xiaofeng Han,Long Fu,Jing Yu,Kunting Li,Ziqing Deng,Ruihao Shu,Dali Wang,Jing You,Eddy Y. Zeng

Microplastics (MPs) and antibiotics co-occur widely in the environment and pose combined risk to microbial communities. The present study investigated the effects of erythromycin on biofilm formation and resistance mutation of a model bacterium, E. coli , on the surface of pristine and UV-aged polystyrene (PS) MPs sized 1–2 mm. The properties of UV-aged PS were significantly altered compared to pristine PS, with notable increases in specific surface area, carbonyl index, hydrophilicity, and hydroxyl radical content. Importantly, the adsorption capacity of UV-aged PS towards erythromycin was approximately 8-fold higher than that of pristine PS. Biofilms colonizing on UV-aged PS had a greater cell count (5.6 × 10 8 CFU mg −1 ) and a higher frequency of resistance mutation (1.0 × 10 −7 ) than those on pristine PS (1.4 × 10 8 CFU mg −1 and 1.4 × 10 −8 , respectively). Moreover, erythromycin at 0.1 and 1.0 mg L − 1 significantly ( p < 0.05) promoted the formation and resistance mutation of biofilm on both pristine and UV-aged PS. DNA sequencing results confirmed that the biofilm resistance was attributed to point mutations in rpoB segment of the bacterial genome. qPCR results demonstrated that both UV aging and erythromycin repressed the expression levels of a global regulator rpoS in biofilm bacteria, as well as two DNA mismatch repair genes mutS and uvrD , which was likely to contribute to increased resistance mutation frequency.

Effect of Various Thermal Treatments on Erythromycin Residues and Degradation Products in Turbot Fish Meat: Implications for Food Safety

Haonan Jiang,Rong Liu,Xuhui Huang,Lei Qin

Erythromycin, a widely utilized antibiotic in aquaculture, poses potential food safety risks through residues in fish products. However, research on the effects of thermal processing on its degradation remains limited. This study developed a sensitive detection method using UHPLC-Q-Exactive HF-X to quantify erythromycin and its degradation products, investigating influences of various thermal treatments and predicting additional degradants. Turbot meat samples spiked with erythromycin were processed via roasting, microwaving, deep frying, and boiling. Post-treatment degradants were identified, with potential metabolites forecasted through structural analysis. The results indicated that erythromycin rapidly degrades under all thermal treatments, with boiling and roasting promoting substantial formation of dehydration products (anhydroerythromycin A and erythromycin A enol ether). The content of N-demethylerythromycin A remained consistently low and stable (<6.67 ng/g). Additionally, five additional erythromycin degradants were screened out. The degradation pathways and product profiles of erythromycin varied depending on the thermal treatment, indicating that thermal processing does not eliminate erythromycin residues but rather transforms them into other substances. This study revealed the specific degradation pattern of erythromycin during thermal processing of fish meat, providing scientific evidence for identifying which harmful substances require priority monitoring in processed fish products, thereby enhancing the level of food safety assessment.

Variations of physical and chemical properties in relation to erythromycin mycelial dreg dewaterability under heat-activated persulfate oxidation conditioning

Yanxiang Zhang,Huiling Liu,Xiaohu Dai,Chen Cai,Jing Wang,Yunpeng Shen,Peng Wang

Erythromycin mycelial dreg (EMD) resulting from erythromycin (ERY) production process may be used as a nutrient resource. However, it has a high moisture content, which makes handling operations complicated. In this study, the EMD dewaterability under heat-activated persulfate (PS) oxidation conditioning was investigated. The variations of physical and chemical properties of EMD were further investigated to uncover the underlying mechanism of EMD dewaterability. Results indicated that EMD dewaterability was worse under single heat process, but significantly improved under heat-activated PS oxidation process. Heat-activated PS oxidation conditioning was capable to increase zeta potential , reduce median particle size, alter EMD flocs morphology, and disrupt mycelial cells. Concurrently, heat-activated PS oxidation conditioning resulted in the decrease of protein in tightly bound (TB) extracellular polymeric substances (EPS) and total EPS, and the decrease of fluorescing constituents (tryptophan protein, tyrosine protein and tryptophan amino acid) in EPS. Furthermore, a possible mechanism was proposed for heat-activated PS oxidation conditioning.

Blue-light photoelectrochemical sensor based on nickel tetra-amined phthalocyanine-graphene oxide covalent compound for ultrasensitive detection of erythromycin

Jinyun Peng,Qing Huang,Wenfeng Zhuge,Yuxia Liu,Cuizong Zhang,Wei Yang,Gang Xiang

In this study, we developed a novel photoelectrochemical (PEC) sensor for the highly sensitive detection of erythromycin by functionalising graphene oxide (GO) with nickel tetra-amined phthalocyanine (NiTAPc) through covalent bonding, which resulted in the formation of NiTAPc-Gr. The fabricated sensor showed a higher PEC efficiency under blue light, exhibiting a peak wavelength of 456 nm, as compared to that of the monomer . Further, the NiTAPc-Gr/indium tin oxide (ITO) sensor exhibited a photocurrent that was 50-fold higher than that for a GO/ITO sensor under the same conditions. Under optimal conditions, the NiTAPc-Gr PEC sensor showed a linear response for erythromycin concentrations ranging from 0.40 to 120.00 μmol L −1 , with the minimum limit for detection being 0.08 μmol L −1 . Thus, the NiTAPc-Gr sensor exhibited superior performance and excellent PEC characteristics, high stability, and good reproducibility with respect to the sensing of erythromycin. Moreover, it is convenient to use, fast, small, and cheap to produce. Hence, it should find wide use in the analysis of erythromycin in real-world applications.

The release of organic matter, nitrogen, phosphorus and heavy metals from erythromycin fermentation residue under heat-activated persulfate oxidation conditioning

Yanxiang Zhang,Huiling Liu,Xiaohu Dai,Jing Wang,Yunpeng Shen,Peng Wang

Erythromycin fermentation residue (EFR) is one kind of biological waste with high organic matter content. The recycling of EFR is not only beneficial to the environmental protection, but also to the economic development. In this study, the release of organic matter, nitrogen (N), phosphorus (P) and heavy metals (HMs) from EFR under heat-activated persulfate (PS) oxidation conditioning was investigated. Results indicated that oxidation conditioning promoted the release of soluble chemical oxygen demand (SCOD). Heat-activated PS oxidation conditioning boosted the release of total nitrogen (TN), ammonia‑nitrogen (NH 4 + -N) and nitrate‑nitrogen (NO 3 − -N) into the supernatant, as well as the decomposition of organic nitrogen (ON). Concurrently, heat-activated PS oxidation conditioning facilitated the release of total phosphorus (TP), orthophosphate (PO 4 3− -P) and organic phosphorus (OP) into the supernatant, and the decomposition of OP. Furthermore, heat-activated PS oxidation conditioning resulted in the increase of release efficiencies of HMs. Therefore, heat-activated PS oxidation conditioning was beneficial to the release of organic matter, nutrients and HMs.

Degradation of macrolide antibiotic erythromycin and reduction of antimicrobial activity using persulfate activated by gamma radiation in different water matrices

Libing Chu,Run Zhuan,Dan Chen,Jianlong Wang,Yunpeng Shen

The degradation of macrolide antibiotic erythromycin (ERY) and the reduction of antimicrobial activity was performed using persulfate activated by gamma radiation in different water matrices. The results showed that the rate constant k obs of ERY degradation in γ ray-activated S 2 O 8 2− (Gamma/PS) system increased by 6.5 times in deionized water, 5.0 times in groundwater and 1.4 times in treated wastewater, respectively. A significant improvement in mineralization rate was observed in Gamma/PS system, with 3.4–52% TOC reduction, compared to 0.9–25% in γ ray system. The results of antimicrobial activity of ERY against E. coli and S . aureus in the three kinds of waters showed that no antimicrobial activity to E. coli was observed at 1 kGy in Gamma/PS system, which required 6 kGy by γ ray alone. The inhibition to S . aureus was still observed at 10 kGy using gamma radiation in groundwater and treated wastewater, which disappeared in Gamma/PS system. The presence of organic substances inhibited ERY degradation more significantly than inorganic anions. In such cases, the ERY degradation rate was promoted by around 2 times and 12 times, respectively in the presence of peptone and glucose in Gamma/PS system. The 14-membered ring of ERY was destroyed by gamma radiation. Formic acid and acetic acid were detected and their yield was higher in Gamma/PS system. The γ ray-activated persulfate process is promising to treat ERY-containing water and wastewater.

The Strength of hERG Inhibition by Erythromycin at Different Temperatures Might Be Due to Its Interacting Features with the Channels

Dongrong Cheng,Xiaofeng Wei,Yanting Zhang,Qian Zhang,Jianwei Xu,Jiaxin Yang,Junjie Yu,Antony Stalin,Huan Liu,Jintao Wang,Dian Zhong,Lanying Pan,Wei Zhao,Yuan Chen

Erythromycin is one of the few compounds that remarkably increase ether-a-go-go-related gene (hERG) inhibition from room temperature (RT) to physiological temperature (PT). Understanding how erythromycin inhibits the hERG could help us to decide which compounds are needed for further studies. The whole-cell patch clamp technique was used to investigate the effects of erythromycin on hERG channels at different temperatures. While erythromycin caused a concentration-dependent inhibition of cardiac hERG channels, it also shifted the steady-state activation and steady-state inactivation of the channel to the left and significantly accelerated the onset of inactivation at both temperatures, although temperature itself caused a profound change in the dynamics of hERG channels. Our data also suggest that the binding pattern to S6 of the channels changes at PT. In contrast, cisapride, a well-known hERG blocker whose inhibition is not affected by temperature, does not change its critical binding sites after the temperature is raised to PT. Our data suggest that erythromycin is unique and that the shift in hERG inhibition may not apply to other compounds.Keywords:erythromycin;temperature;hERG;steady-state activation;steady-state inactivation

Preparation of high-quantum-yield fluorescent carbon dots derived from Paliurus ramosissimus and their dual-functional sensing of erythromycin and Hg2+

Meiling Long,Xinyuan Wang,Shaogui Wu

In this study, sulfur and nitrogen co-doped fluorescent carbon dots (S,N-CDs) were synthesized via a hydrothermal method using Paliurus ramosissimus as a natural and sustainable carbon source. The as-prepared S,N-CDs exhibited a high quantum yield of 73.3% in ethanol, along with pronounced solvent-dependent and excitation-dependent fluorescence properties. Fluorescence sensing investigations demonstrated that the S,N-CDs possessed excellent selectivity toward erythromycin (EM) and Hg 2+ , with limits of detection of 0.27 μM and 38 nM, respectively. The quenching process was attributed to a static quenching mechanism, and satisfactory anti-interference performance was observed. Furthermore, a smartphone-assisted visual detection strategy was established based on the distinct fluorescence color variation of S,N-CDs in response to different EM concentrations, achieving a detection limit as low as 0.41 μM. Spike-and-recovery experiments revealed recoveries of 87.97%–95.52% in rainwater samples and 100.88%–107.11% in milk samples, with relative standard deviations (RSDs) below 3%, indicating favorable practical applicability. Overall, the developed S,N-CDs fluorescent probe combines high sensitivity, strong selectivity, and environmental friendliness, providing a promising platform for the rapid detection of multiple contaminants in environmental and food matrices and broadening the application scope of plant-derived carbon dots.

Distinct effects of nano-polystyrene, erythromycin, and their mixtures on the composition and metabolic profile of intestinal microbiota in Nereis succinea

Dali Wang,Yuheng Zhong,Qi Ding,Xiaofeng Han,Kai Chen,Bo Pan,Jing You

Nanoplastics (NPs) and antibiotics are emerging contaminants that co-occur ubiquitously in coastal environments and pose risks to ecosystems. Here, we report the single and combined effects of nano-sized polystyrene (PS) and erythromycin on a marine benthic invertebrate N. succinea at both individual and intestinal microbial levels. It was found that PS at 10 and 1000 μg L−1 and erythromycin at 249 and 7170 ng L−1, individually and in combination, did not cause mortality of N. succinea, but altered organism weight and behaviors and led to morphological changes and oxidative stresses in the intestines. Synergistic effects on glutathione-S-transferase activity and burrowing behavior were observed for mixtures containing high levels of PS. The results of 16S rRNA sequencing showed that erythromycin at both concentrations did not cause changes in microbial composition at the phylum level, while PS and the mixtures significantly increased the relative abundance of Proteobacteria, and reduced Actinobacteriota, Bacteroidota and Firmicutes. At the genus level, erythromycin decreased some potential pathogens such as Vibrio, Nautella, and Pseudomonas, whereas PS and the mixtures reduced beneficial genera such as Megasphaera and Megamonas. Community-level physiological profiles showed that erythromycin and PS had distinct influences on the metabolism of the cultivable intestinal microbiota, with erythromycin enhancing while PS and the mixtures inhibiting the metabolic capacity and diversity. Collectively, our results illustrated that PS NPs and erythromycin exerted distinct influences on the intestinal microbiota of N. succinea, and their mixture effects were dominated by the PS effect.

Transformation of macrolides residues by a novel erythromycin esterase C (Ere C) and safety evaluation of transformed products on Caenorhabditis elegans

Yulei Zhang,Binbin Ouyang,Yihan Chen,Wenli Zhang,Cuie Guang,Wei Xu,Wanmeng Mu

In this study, the second enzyme of esterase C family from Enterobacter hormaechei ( Enho -Ere) was identified. The optimum pH and temperature of Enho -Ere was determined at pH 7.0 and 50 °C. At the concentration of 0.8 g/L, 100% erythromycin (Ery), 100 % clarithromycin (Clr), and 50 % roxithromycin (Rox) were transformed by Enho -Ere. Based on transformed products (TPs), a hydrolysis-hydroxyl attack-dehydration reaction process was proposed. Firstly, the lactone ring in macrolides were hydrolyzed. The newly formed hydroxyl from hydrolysis then attacked the unsaturated bond at position C9 to generate the intermediate TPs. Finally, the dehydration happened between the position C6 and C9 to generate the formula isomer of macrolides. In addition, the effects of TPs on Caenorhabditis elegans were evaluated after Enho -Ere treatment. The intracellular ROS level, and the expression of antistress/lifespan-genes of C . elegans all showed significant difference after treatment, indicating that transformation and detoxification of macrolides was both accomplished by Enho -Ere.

Enhancement of the heterogeneous photo-Fenton performance of GO/MIL-100(Fe)@Fe3O4 heterostructures for erythromycin degradation through accelerating Fe(II) generation

Lian Yu,Baozi Lu,Zhen Liang,Dabin Wang,Jiarui Lu,Fengyi Wei,Cunzhen Liang

The slow rate of the Fe3+/Fe2+ cycle has decreased the efficiency of photo-Fenton technology for wastewater treatment. Here, a novel GO/MIL-100(Fe)@Fe3O4 heterostructure was constructed by growing Fe3O4 nanoparticles and MIL-100(Fe) onto GO, thus accelerating the Fe3+/Fe2+ cycle and improving catalytic activity, and it was used as a heterogeneous photo-Fenton catalyst for ERY degradation. The catalyst was fully characterized by TEM, XRD, Raman spectroscopy, XPS, N2 adsorption–desorption and UV-vis DRS. The influences of initial solution pH, H2O2 concentration and catalyst dosage on ERY degradation were investigated. Under optimal conditions (initial pH of 4.03, 10.0 mM H2O2, 0.1 g L−1 catalyst), the GO/MIL-100(Fe)@Fe3O4 + H2O2 + vis system removed 97.3% of ERY (0.1 mmol L−1) within 60 min, and the pseudo-first-order rate constant was 0.0575 min−1. Furthermore, the catalyst showed favorable stability, maintaining excellent catalytic activity even after 4 cycles. A possible degradation mechanism of ERY was proposed based on the results of characterization, ERY degradation, and reactive species analysis. Fe3O4, MIL-100(Fe) and GO in the catalyst showed synergistic effects; the photo-induced electrons from MIL-100(Fe) could be immediately transferred to Fe3O4, promoting Fe3+ reduction to Fe2+ in the photo-Fenton reaction. GO in the catalyst could promote electron transfer from MIL-100(Fe) and organics to Fe3O4, significantly promoting the Fe3+/Fe2+ cycle.
