Gastrointestinal dysmotility is a common cause of functional dyspepsia. As two kinds of polysaccharides derived from brown algae, fucoidan and laminarin possess many physiological properties; however, their relative abilities in regulating gastrointestinal motility have not been illustrated yet. In this study, we aimed to investigate the regulatory effect of fucoidan and laminarin on functional dyspepsia mice induced by loperamide. Mice with gastrointestinal dysmotility were treated with fucoidan (100 and 200 mg per kg bw) and laminarin (50 and 100 mg per kg bw). As a result, fucoidan and laminarin reversed the dysfunction mainly through regulating gastrointestinal hormones (motilin and ghrelin), the cholinergic pathway, the total bile acid level, c-kit protein expression, and gastric smooth muscle contraction-related gene expression (ANO1 and RYR3). Moreover, fucoidan and laminarin intervention modulated the gut microbiota profile including the altered richness of Muribaculaceae, Lachnospiraceae, and Streptococcus. The results indicated that fucoidan and laminarin may restore the rhythm of the migrating motor complex and regulate gut microecology. In conclusion, we provided evidence to support that fucoidan and laminarin might have potential abilities to regulate gastrointestinal motility.

Modulating a favorable inflammatory microenvironment that facilitates the recovery of degenerated discs is a key strategy in the treatment of intervertebral disc (IVD) degeneration (IDD). More interestingly, well-mechanized tissue-engineered scaffolds have been proven in recent years to be capable of sensing mechanical transduction to enhance the proliferation and activation of nucleus pulposus cells (NPC) and have demonstrated an increased potential in the treatment and recovery of degenerative discs. Additionally, existing surgical procedures may not be suitable for IDD treatment, warranting the requirement of new regenerative therapies for the restoration of disc structure and function. In this study, a light-sensitive injectable polysaccharide composite hydrogel with excellent mechanical properties was prepared using dextrose methacrylate (DexMA) and fucoidan with inflammation-modulating properties. Through numerous in vivo experiments, it was shown that the co-culture of this composite hydrogel with interleukin-1β-stimulated NPCs was able to promote cell proliferation whilst preventing inflammation. Additionally, activation of the caveolin1-yes-associated protein (CAV1-YAP) mechanotransduction axis promoted extracellular matrix (ECM) metabolism and thus jointly promoted IVD regeneration. After injection into an IDD rat model, the composite hydrogel inhibited the local inflammatory response by inducing macrophage M2 polarization and gradually reducing the ECM degradation. In this study, we propose a fucoidan-DexMA composite hydrogel, which provides an attractive approach for IVD regeneration.

Metal ions play a crucial role in forming hydrogels, and their effects on fucoidan (FUC): κ-carrageenan (KC) mixed gels were investigated. The results indicated that the FUC: KC mixed gels (FC) were promoted by K + and Ca 2+ but destroyed by Fe 3+ . The gel strength of FC was enhanced by K + and Ca 2+ , with G′ and G″ being highest at 50 mmol/L KCl and 25 mmol/L CaCl 2 , respectively. Water mobility was weakened after the addition of KCl and CaCl 2 in accordance with the decrease in T 23 relaxation time (free water, 100–1000 ms). After addition of KCl and CaCl 2 , the FC groups presented a typical three-dimensional network structure in contrast to the lamellar, disordered, and broken structure of FUC. Moreover, the FT-IR spectrum certified the enhancement of hydrogen bonds and the occurrence of electrostatic interactions during gel formation by the red-shift of the O H stretching vibration of the Ca 2+ group and the blue-shift of the C O S vibrations. The XRD results confirmed that the binding of Ca 2+ to FC was tighter than that of K + at the same charge content. These results provide a theoretical basis for understanding the interaction mechanism of FC with metal ions.

Fucoidans represent a type of polyanionic fucose-containing sulfated polysaccharides (FCSPs) that are cleaved by fucoidan-degrading enzymes, producing low-molecular-weight fucoidans with multiple biological activities suitable for pharmacological use. Most of the reported fucoidan-degrading enzymes are glycoside hydrolases, which have been well studied for their structures and catalytic mechanisms. Little is known, however, about the rarer fucoidan lyases, primarily due to the lack of structural information. FdlA fromFlavobacteriumsp. SA-0082 is an endo-type fucoidan-degrading enzyme that cleaves the sulfated fuco-glucuronomannan (SFGM) through a lytic mechanism. Here, we report nine crystal structures of the catalytic N-terminal domain of FdlA (FdlA-NTD), in both its wild type (WT) and mutant forms, at resolutions ranging from 1.30 to 2.25 Å. We show that the FdlA-NTD adopts a right-handed parallel β-helix fold, and possesses a substrate binding site composed of a long groove and a unique alkaline pocket. Our structural, biochemical, and enzymological analyses strongly suggest that FdlA-NTD utilizes catalytic residues different from other β-helix polysaccharide lyases, potentially representing a novel polysaccharide lyase family. Keywords:fucoidan lyase;polysaccharides;crystal structure;catalytic mechanism

The protective effects of fucoidan on alcohol- and carbon tetrachloride-induced liver fibrosis and its potential mechanism were investigated in a mouse model, with a fucus on the gut-bile acid-liver metabolic axis. Fucoidan treatment significantly reduced collagen deposition and downregulated the expression of α-smooth muscle actin (α-SMA) and type III collagen (ColIII). Bile acid profiling demonstrated that fucoidan decreased total bile acids (TBA) levels in both serum and feces, with particularly marked reductions in deoxycholic acid (DCA) and muricholic acid (MCA). Conversely, fucoidan increased the level of taurodeoxycholic acid (TDCA). This treatment restored the metabolic imbalance of bile acids (BAs) caused by alcohol, thereby suppressing YAP-mediated hepatic stellate cells (HSCs) activation and associated inflammation. Mechanistically, fucoidan downregulated G protein-coupled bile acid receptor 1 (TGR5) and its downstream signaling, while also reducing farnesoid X receptor (FXR) expression and modulating the FXR/FGF15/CYP7A1 signaling to improve bile acid metabolism. Additionally, fucoidan alleviated alcohol-induced gut dysbiosis, as evidenced by increased abundance of Alloprevotella and decreased abundance of Lactobacillus . Importantly, when combined with cholestyramine, the protective effect of fucoidan on liver fibrosis significantly weakened. This confirmed that fucoidan exerted its inhibitory effect on the activation of HSCs by regulating the metabolic homeostasis of bile acids. Overall, fucoidan protects against alcoholic liver fibrosis by modulating the gut-bile acid-liver axis and inhibiting HSCs activation through the TGR5/SRC/YAP and FXR/FGF15/CYP7A1 signaling pathways.

Biomaterials derived from natural polymers have raised much attention for use in bone tissue engineering applications because of their high mechanical strength and biocompatibility. However, challenges persist in developing bone materials with the requisite physicochemical and osteogenic properties. In this study, the freeze-drying method fabricated a scaffold using chitosan and fucoidan reinforced with zinc oxide nanoparticles (CSFU@ZnO). The scaffolds were characterized and evaluated for their osteogenic differentiation, proliferation, and viability of pre-osteoblast cells (MC3T3-E1). Field emission-scanning electron microscopy analysis revealed a porous morphology with interconnected pores and hydrogen bond interaction between chitosan and fucoidan, as confirmed by Fourier transform infrared spectroscopy analysis. The combination of fucoidan with chitosan improved the mechanical properties, swelling capacity, and degradability. The CSFU@ZnO scaffold effectively inhibited the growth of S. aureus and E. coli , as shown by a zone of inhibition assay. In vitro studies revealed enhanced MC3T3-E1 viability, proliferation, and adhesion, as validated by SEM analysis. The osteogenic differentiation of the CSFU@ZnO scaffold showed increased alkaline phosphatase activity, better protein adsorption, and improved bone mineralization compared to the chitosan scaffold. These findings suggest that the ZnO nanoparticle-incorporated chitosan and fucoidan scaffold could be a good and viable material for bone tissue engineering.

Silvetia siliquosa , the only species of the family Fucaceae in China, is used as a medicine food homology. Fucoidan from S. siliquosa was extracted by hot water twice thoroughly (13 % of total yield), and a purified fucoidan SSF with a molecular weight of 93 kD was obtained. Chemical composition analysis demonstrated that SSF was primarily composed of sulfate (21.68 wt%) and fucose (84 % of all neutral monosaccharides). IR, methylation analysis, NMR and ESI-MS results indicated SSF had the backbone of mainly (1 → 3)-α-L-fucopyranose and minor (1 → 4)-α-L-fucopyranose, with little 1,3 and 1,4 branched β-D-Xyl p and β-D-Gal p . The in vitro immunomodulatory test on RAW 264.7 cells showed that SSF could up-regulate the expression of immune related factors and proteins in a concentration-dependent manner, but the immunomodulatory effect disappeared from desulfated SSF . This research indicated that highly sulfated fucan possessed immunomodulatory effect and the importance of sulfate groups in the activity of SSF .

Acute lung injury (ALI) is a complex acute respiratory illness with a high mortality rate. Reactive oxygen species (ROS) play a pivotal role in ALI, inducing cellular damage, inflammation, and oxidative stress, thereby exacerbating the severity of the injury. In this study, inspired by the “subtractive” strategy, we developed a fucoidan-based macrophage membrane bio-nanosystem, abbreviated as MF@CB, designed as an anti-inflammatory and antioxidant agent to alleviate lipopolysaccharide (LPS)-induced inflammation in ALI. MF@CB coated with macrophage membrane for effective targeting and accumulation in ALI lesions. In addition, MF@CB activates Nrf2 transcriptional activity in macrophages, inhibiting ROS synthesis at its origin while effectively removing ROS already present in the ALI. This dual-pronged approach demonstrates robust antioxidant properties and restores the macrophage antioxidant defense barrier. In the LPS-induced ALI mouse model, MF@CB significantly mitigated lung inflammatory damage by modulating lung macrophage polarization and inhibiting the over-secretion of pro-inflammatory cytokines by activated immune cells. More importantly, unlike most surface modification strategies because it remove the molecule, this approach is easier to apply and potentially safer and may provide useful insights into the development of more effective therapeutic strategies for ALI.

Background Hypertension influences the inflammatory pathological changes in the retina. The function of the inflammasomes is significant. To see if Sirtuin 1 (SIRT1) regulates angiotensin II (Ang II)-induced hypertensive retinopathy and inflammation by modulating NOD-like receptor thermal protein domain associated protein 3 (NLRP3) inflammasome activation and the potential protective effects of fucoidan (FO) in mouse retinal vascular endothelial cells (mRECs) and mice retina. Methods The diagnosis of hypertensive retinopathy was made after three weeks of Ang II infusion (3000 ng/kg/min). One day prior to the commencement of Ang II infusion, the mice were treatment with NLRP3 inhibitor MCC950 (10 mg/kg/day, intraperitoneal injections) or FO (300 mg/kg/day, oral gavage). A blood pressure was recorded. Hematoxylin and eosin (H&E) staining was used to conduct pathological alterations, dihydroethidium bromide (DHE) was utilized to assess oxidative stress damage in the retina, and fluorescence angiography was used to identify vascular disorders in the eye. Using immunohistochemical labeling, NLRP3 expression was found. Reactive protein and mRNA expression levels in mouse retina and cells were assessed using Western blot and real-time quantitative polymerase chain reaction (RT-qPCR). Results NLRP3 inflammasome activation and SIRT1 decrease were brought about by Ang II infusion. Retinopathy and dysfunction were lessened by MCC950 target-induced NLRP3 inflammasome activation, while overexpression of SIRT1 had the opposite impact on NLRP3 inflammasome activation, indicating that SIRT1 functions as an upstream regulator of NLRP3 activity. FO may improve SIRT1 expression and decrease NLRP3 activation in retinopathy and dysfunction brought on by Ang II, and the effects were consistent across both in vivo and in vitro models. Conclusions SIRT1 adversely regulates the NLRP3 inflammasome pathway, which in turn increases Ang II-induced inflammation and hypertensive retinopathy. FO may mitigate Ang II-induced retinopathy and dysfunction via modulating the expression of SIRT1/NLRP3. This implies practical approaches to the management of hypertensive retinopathy.

Colorectal cancer (CRC) is one of the major malignancies in humans. This study was designed to evaluate the effects of fucoidan on gut flora and tumor prevention in 1,2-dimethylhydrazine-induced colorectal carcinogenesis in rats. We found that dietary fucoidan treatment decreased the tumor incidence and mean tumor weight and increased cell apoptosis . Fucoidan treatment decreased the expression of β-catenin C-Myc, CyclinD1 and Survivin , while the Hippo pathway was activated with increased phosphorylation levels of mammalian sterile 20-like kinase 1 and 2, large tumor suppressor 1 and 2, and Yes-associated protein. Compared with the model group, the levels of interleukin (IL)-17 and IL-23 were decreased, but the levels of interferon-γ, IL-4 and IL-10 were increased, in the fucoidan group. Fucoidan treatment increased natural killer cells in peripheral blood and the proportion of CD4+ T cells . Immunofluorescence detection of colorectal tumor tissues showed decreased expression of Foxp3 and up-regulated expression of CD68 in the fucoidan group. Moreover, fucoidan treatment decreased the levels of diamine oxidase and lipopolysaccharides and up-regulated the levels of tight junction proteins . 16S rDNA high-throughput sequencing revealed that fucoidan treatment decreased the abundance of Prevotella and increased the abundance of Alloprevotella . Fucoidan increased the levels of butyric acid and valeric acid compared to the model group. This study provides experimental evidence that dietary fucoidan may prevent colorectal tumorigenesis by regulating gut microecology and body immunity. Meanwhile, fucoidan activated the Hippo pathway and down-regulated the β-catenin pathway to induce tumor cell apoptosis and suppress tumor growth.

In this study, we used fucoidan (FD), a sulfated polysaccharide , to stabilize zein and obtain a composite nanosystem with better performance that can be used to encapsulate and deliver hydrophobic active substances. The composite nanoparticles of zein and FD with different mass ratios were prepared by the antisolvent precipitation method. The solution (zein/FD = 1:2) could maintain its stable solution state without precipitation under different environmental conditions (pH 2–8, heating at 80 °C for 120 min, salt concentration < 25 mmol/L and storage at 4 °C for 28 days). Subsequently, curcumin (Cur) was used as the model of hydrophobic active substances. When 2 mg Cur was added, the composite nanosystem had a high encapsulation efficiency (81.07%) and could reduce the release of Cur in simulated gastrointestinal digestion. Our results demonstrated that the composite nanosystem prepared by FD and zein, which are both natural biopolymers , has great potential in the encapsulation and delivery of hydrophobic active substances.

Diabetes is a complicated endocrine and metabolic disorder, which has become an epidemic health issue worldwide. Fucoidan is extensively distributed in the brown algae and several marine invertebrates exhibiting diverse biological activities. In the present study, the physicochemical property of Sargassum fusiforme fucoidan (SFF) and its effects on streptozotocin (STZ)-induced diabetic mice and gut microbiota were investigated. Diabetes mice not only showed abnormal blood glucose, but also accompanied by multiple symptoms, such as gradual emaciation, decreased body weight, increased food and water intake. Compared with diabetic mice after 6-week treatment, administration of SFF significantly decreased the fasting blood glucose, diet and water intake. Furthermore, SFF attenuated the pathological change in the heart and liver, improved the liver function, and suppressed oxidative stress in STZ-induced diabetic mice. Simultaneously, SFF significantly altered the gut microbiota in the faeces of diabetic mice, decreased the relative abundances of the diabetes-related intestinal bacteria, which is a potential mechanism for relieving the symptoms of diabetes. Therefore, SFF might be considered as one of the promising complementary and alternative medicines for the management of diabetes mellitus in future.
