Nanomaterial selection is critical for photoelectrochemical (PEC) sensing. In this report, a novel cathodic photoelectrochemical (PEC) strategy was proposed for the detection of doxorubicin hydrochloride (Dox) and gentamicin sulfate (CN). The photocathode was synthesized by noncovalently coupling cadmium sulfide (CdS) to the porphyrin-derived metal–organic framework (CdS@PCN-224). This type of assembly created a pleasant interface for the combination of doxorubicin hydrochloride and gentamicin sulfate, resulting in a good CdS@PCN-224 donor–acceptor system. When compared to a single optoelectronic material, its photocurrent is enhanced by unprecedented nine times. This research could pave the way for the realization of PCN-224’s enormous potential in PEC sensing.

Fishbone-structured SF lifting threads were prototyped. • Glycerol/Ca 2+ regulation improved the mechanical properties of SF threads. • Tannic acid and gentamicin endowed the threads with antioxidant and antibacterial functions. • TA/GEN@SFs showed in vitro antibacterial activity against E. coli and S. aureus. • TA/GEN@SFs attenuated inflammatory infiltration and fibrous capsule formation in vivo . Facial thread lifting is widely employed in minimally invasive rejuvenation for its rapid recovery. However, its clinical acceptance is heavily limited by common complications, particularly local infections and severe inflammatory responses. Herein, a multifunctional silk fibroin (SF) lifting thread incorporating tannic acid (TA) and gentamicin (GEN) was fabricated using a custom-designed mold with a biomimetic fishbone architecture and was denoted as TA/GEN@SFs. TA/GEN@SFs exhibited excellent mechanical properties, with a breaking force of up to 40.00 ± 2.30 N and an elongation at break of 13.20 ± 0.98%. Moreover, TA/GEN@SFs showed significant antibacterial activity against Escherichia coli and Staphylococcus aureus , with inhibition zone diameters of 4.21 ± 0.33 mm and 5.71 ± 0.44 mm, respectively. In addition, TA/GEN@SFs achieved a free radical scavenging rate of 56.12 ± 3.31%, indicating notable antioxidant capability. Results from in vivo subcutaneous implantation further demonstrated that TA/GEN@SFs effectively modulated the wound microenvironment by suppressing excessive fibrosis and shortening the inflammatory phase. Overall, this study offers a promising strategy for creating functional implants in medical aesthetics. Download: Download high-res image (115KB) Download: Download full-size image

Asymmetric mesoporous silica nanoparticles with anisotropic geometry and dual-compartments are highly desired for loading and release of dual-drugs in separated storage spaces. In this study, an asymmetric lollipop-like mesoporous silica nanoparticle Fe 3 O 4 @SiO 2 &EPMO (EPMO = ethane bridged periodic mesoporous organosilica) was successfully developed via an anisotropic epitaxial growth strategy. The asymmetric nanoparticles show a uniform lollipop shape with a head of spherical Fe 3 O 4 @SiO 2 core-shell that is 200 nm in diameter and a tail of EPMO nanorods with a length of ∼90 nm, and a specific surface area of ∼650.3 m 2 g −1 . Most importantly, the asymmetric nanoparticles possess the unique dual independent (hydrophilic/hydrophobic) spaces with good loading capacities and are significantly more efficient for cancer cell killing than pure drug based on in vitro studies. Additionally, the dual-drug-loaded nanoparticles exhibited excellent antibacterial activity.

Herein, a robust photoelectrochemical (PEC) immunosensor was constructed based on an n-n heterojunction between a copper-porphyrin-based metal organic framework and Bi 2 S 3 (Bi 2 S 3 @Cu-TPyP) for efficient detection of gentamicin (GEN). The integration of Cu-TPyP features (enhanced electron transfer capability, improved photoabsorption performance, large specific surface area, and high antibody bioaffinity) with Bi 2 S 3 characteristics (narrow bandgap and visible light absorption capacity) established an efficient interfacial architecture. Enhanced separation of photogenerated electron-hole pairs and improved photoelectric conversion efficiency in the Bi 2 S 3 @Cu-TPyP heterojunction contributed to the amplified photocurrent. The immunosensor exhibited an ultralow detection limit of 0.041 fg mL −1 (1.0 fg mL −1 –1.0 ng mL −1 linear range) with high selectivity and validated applicability in analyzing GEN across diverse food matrices. This study establishes a PEC biosensing paradigm enabling ultrasensitive and rapid antibiotic monitoring in food products due to the synergistic signal amplification mechanisms without compromising specificity.

Infection of the bone (osteomyelitis) remains one of the most challenging problems in the field of orthopedic surgery. The limitations of systemic antibiotics administration include undesired side effects, systemic toxicity, patient discomfort, and development of bacterial resistance. In this study, we developed a bactericidal gentamicin-doped beta-tricalcium phosphate (TCP) scaffold reinforced with a gelatin/genipin hydrogel (G-TCP). Our data showed that the gentamicin-doped G-TCP had a much longer drug releasing period, while the gentamicin was completely released from pure TCP cements (B-TCP) within one day. In addition, the release profile of G-TCP exhibited an initial burst followed by a zero-order release. One standard strain, Staphylococcus aureus (S. aureus, ATCC25923) was selected to evaluate the antibacterial activity and therapeutic effect of this scaffold. G-TCP significantly inhibited growth of S. aureus both in vitro and in vivo. In a rat osteomyelitis model, osteomyelitis could be totally cured after implantation of G-TCP for three weeks. We propose that the gelatin/genipin–gentamicin TCP scaffold represents one of the promising gentamicin releasing bone scaffolds in treating osteomyelitis.

Implant materials are prone to bacterial infections and cause serious consequences, while traditional antibiotic therapy has a long treatment cycle and even causes bacterial resistance. In this work, a photothermal therapy (PTT) assisted drug release system has been developed on the implant surface for in situ rapid disinfection under 808 nm light irradiation within a short time, in which gentamicin (Gent) is loaded by polyethylene glycol (PEG) modified molybdenum disulfide (MoS2) on Ti surface, and then encapsulated with chitosan (CS) (CS/Gent/PEG/MoS2-Ti). The hyperthermia produced by the coatings irradiated by 808 nm near-infrared (NIR) light can not only accelerate the local release of Gent, but also reduce the activity of bacteria, which makes it easy for these locally released drugs to enter the interior of the bacteria to inhibit the protein synthesis and destroy the cell membrane. When maintained at 50 °C for 5 min under NIR irradiation, this system can achieve an antibacterial efficacy of 99.93% and 99.19% against Escherichia coli and Staphylococcus aureus, respectively. By contrast, even after treatment for 120 min, only a 93.79% antibacterial ratio can be obtained for Gent alone. This is because hyperthermia produced from the coatings during irradiation can assist antibiotics in killing bacteria in a short time. Even under a low dose of 2 μg mL−1, the photothermal effect assisted gentamicin can achieve an antibacterial efficacy of 96.86% within 5 min. In vitro cell culture shows that the modified surface can facilitate cell adhesion, spreading and proliferation. The 7 day subcutaneous infection model confirms that the prepared surface system can exhibit a much faster sterilization and tissue reconstruction than the control group with light assistance. Compared with the traditional drug release system, this photothermy controlled drug-loaded implant surface system can not only provide rapid and high-efficiency in situ sterilization, but also offer long-term prevention of local bacterial infection.

In recent years, developments of drug delivery systems of antibiotics have been needed for improving antibacterial performance due to massive usage of antibiotics. With the global growing concept of being green, lignosulfonate (LS) has been deemed as an ecofriendly stabilizer and carrier that holds promise in preparing nanocomposites for antibacterial treatment. At the same time, LS and gentamicin sulfate (GS) have been proven to form a nanocomposite (LS-GS) with highly uniform size and roundness in solution. It also possesses a good loading capacity of GS. Density functional theory (DFT) verified that the interaction forces in LS-GS, a nanocomposite with self-assembly characteristics, were mainly combined via hydrogen bonds, van der Waals forces, and repulsion. The obtained LS-GS has the biocompatible, biodegradable, and low-cost merits of LS. It also exhibited excellent in vitro antibacterial activities against Escherichia coli and Staphylococcus aureus due to the antibacterial properties of GS. Moreover, an in-depth antibacterial study revealed the highly efficient antibacterial and wound-healing applications of LS-GS in vivo. Collectively, LS, a carrier material with low cost, which strikingly increases the antibacterial and wound-healing ability of GS in vitro and in vivo, will contribute to the production of large-scale antibacterial nanocomposites.

In this study, we present a differential intensity-modulated system that comprises a tapered microfiber interferometer functionalized with polydopamine /AuNPs/cysteamine hydrochloride and two fiber Bragg gratings (FBGs) for detecting gentamicin in deionized water and milk. As external refractive index (RI) increases, the interference troughs of microfiber are red-shifted, while Bragg wavelengths remain unchanged, leading to a change in the difference in the reflected intensities of two FBGs, thus resulting in differential intensity modulation of external RI. The microfiber interferometer alone in this system demonstrates a wavelength response of 1314.57 nm/RIU to external RI, while the system exhibits an outstanding response of 2713.74 dB/RIU, highlighting its superior performance. The system demonstrates excellent results and selectivity when detecting gentamicin in deionized water and milk, and the wavelength response and differential intensity response reach 27 nm/μM, 80 dB/μM and 31 nm/μM, 70 dB/μM, respectively, with limit of detection of 1.117 ppb, which is lower than the maximum residue limit of 30 ppb in milk. Furthermore, a novel all-fiber demodulation scheme is presented, which holds great promise for the development of portable field measurement systems. These findings demonstrate the efficiency of differential modulation systems with implications for food safety.

The residue of gentamicin (GEN) in food is a serious threat to human health. Therefore, it is urgent to develop a simple and highly sensitive method for effectively detecting GEN in food. In this study we proposed a lysine-functionalized gold nanoparticles (AuNPs) colorimetric sensor to detect GEN simply and quickly. The detection mechanism as follows: The interaction between lysine and AuNPs causes lysine to adsorb on the surface of AuNPs. After the addition of GEN, the strong hydrogen bond between GEN and lysine promotes the aggregation of AuNPs. The color of AuNPs changed from wine red to blue, which produced obvious color change. UV–vis spectrometer was used to detect it. The results showed that the colorimetric sensor had high selectivity and sensitivity for the detection of GEN, and the linear range and limit of detection were 5–60 nM and 1.22 nM, respectively. In addition, this method has been successfully applied to the quantitative detection of GEN in milk, and the accuracy and recovery were satisfied.

Purpose Antibiotic-resistant bacteria are pathogens that have emerged as a serious public health risk. Thus, there is an urgent need to develop a new generation of anti-bacterial materials to kill antibiotic-resistant bacteria. Methods Nanosilver-decorated mesoporous organosilica nanoparticles (Ag-MONs) were fabricated for co-delivery of gentamicin (GEN) and nanosilver. After investigating the glutathione (GSH)-responsive matrix degradation and controlled release of both GEN and silver ions, the anti-bacterial activities of Ag-MONs@GEN were systematically determined against several antibiotic-susceptible and antibiotic-resistant bacteria including Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Enterococcus faecalis. Furthermore, the cytotoxic profiles of Ag-MONs@GEN were evaluated. Results The GEN-loaded nanoplatform (Ag-MONs@GEN) showed glutathione-responsive matrix degradation, resulting in the simultaneous controlled release of GEN and silver ions. Ag-MONs@GEN exhibited excellent anti-bacterial activities than Ag-MONs and GEN alone via inducing ROS generation, especially enhancing synergetic effects against four antibiotic-resistant bacteria including Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Enterococcus faecalis. Moreover, the IC50 values of Ag-MONs@GEN in L929 and HUVECs cells were 313.6 ± 15.9 and 295.7 ± 12.3 μg/mL, respectively, which were much higher than their corresponding minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values. Conclusion Our study advanced the development of Ag-MONs@GEN for the synergistic and safe treatment of antibiotic-resistant bacteria.

To explore the application of chitosan–gentamicin conjugate (CS-GT) in inhibitingVibrio parahaemolyticus(V. parahaemolyticus), which is an important pathogen in aquatic animals worldwide, the antimicrobial activity of CS-GT and the effects of a CS-GT dose on the intestine histopathology and intestinal flora ofV. parahaemolyticus-infected shrimps were explored. The results showed that CS-GT possessed broad-spectrum antibacterial activity, with minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and half inhibitory concentration (IC50) of 20.00 ± 0.01, 75.00 ± 0.02 and 18.72 ± 3.17 μg/mL forV. parahaemolyticus, respectively. Further scanning electron microscope and cell membrane damage analyses displayed that the electrostatic interaction of CS-GT with cell membrane strengthened after CS grafted GT, resulting in leakage of nucleic acid and electrolytes ofV. parahaemolyticus. On the other hand, histopathology investigation indicated that high (100 mg/kg) and medium (50 mg/kg) doses of CS-GT could alleviate the injury of a shrimp’s intestine caused byV. parahaemolyticus. Further 16S rRNA gene sequencing analysis found high and medium dose of CS-GT could effectively inhabitV. parahaemolyticusinvasion and reduce intestinal dysfunction. In conclusion, CS-GT possesses good antibacterial activity and could protect shrimps from pathogenic bacteria infection. Keywords:chitosan;Vibrio parahaemolyticus;antimicrobial mechanism;intestine microbe

Injectable polymethylmethacrylate (PMMA) bone cement is a widely used bone substitute in cemented arthroplasty, vertebroplasty and osteoporosis fractures. However, due to the inappropriate stiffness, poor bioactivity and high polymerization temperature of PMMA, aseptic loosening of the implanted cement at the bone–cement interface still could be observed in a high rate of patients. To improve the performance of PMMA, artificial extracellular matrices like chitosan–glycerophosphate (CS–GP) thermosensitive hydrogel was introduced into PMMA acting as a pore forming agent and osteoconductive nano-sized hydroxyapatite (nano-HA)/antibiotic gentamicin (GM) as a carrier. It is shown that CS–GP thermosensitive hydrogel can effectively create open pores at the surface of the PMMA cement, which is believed to facilitate bone tissue ingrowth and improve the cement anchorage at the bone–cement interface in future clinical applications. Meanwhile, such a hydrogel effectively decreases the maximum polymerization temperature to below 30 °C, prolongs the working time to more than 720 s and produces cement with a proper modulus of elasticity and a compressive yield strength ranging from 402 to 584 MPa and from 3.1 to 5.9 MPa, respectively. Furthermore, the incorporated nano-HA particles sufficiently increase the mineralization capacity of the cement without compromising its mechanical properties and the incorporated GM remarkably enhances the anti-bacterial activity of the cement. More importantly, nano-HA and GM enriched CS–GP thermosensitive hydrogel effectively improve the overall performance of PMMA cement without influencing the cell survival, suggesting the injectable p-PMMA/CS–GP/nano-HA/GM cement will hold strong promise for future bone reconstruction applications.
