Hemin-loaded amphiphilic nanoparticles enable stabilized luminol chemiluminescence. • Glow-type chemiluminescence allows direct smartphone imaging without excitation. • AChE inhibition is coupled with chemiluminescent readout for OP quantification. • Reliable OP analysis is achieved in food and environmental samples. Hemin-loaded amphiphilic nanoparticles (HeNPs) were engineered to stabilize hemin and enhance its catalytic performance in the luminol–H₂O₂ chemiluminescence (CL) system. The confined hydrophobic microenvironment effectively suppressed hemin aggregation and produced a stable glow-type emission, which could be directly captured by a smartphone without external excitation or optical accessories. By integrating this stabilized chemiluminescent platform with an acetylcholinesterase (AChE) inhibition mechanism, organophosphorus pesticides (OPs) were translated into quantifiable CL responses. The smartphone-based readout enabled sensitive and reliable analysis with good linearity. The proposed method was further validated in lake water and Chinese cabbage samples, where the results obtained by smartphone imaging showed good agreement with those from microplate-based CL detection and GC–MS. Overall, this work demonstrates a smartphone-enabled chemiluminescent sensing strategy for monitoring OP residues in food and environmental samples. The employment of inexpensive metallomacrocycles to create non-precious metal electrocatalysts (NPMEs) with high performance remains a challenge. Herein, we report the self-assembly of low-cost and abundant hemin on carbon black (EC600) under hydrothermal conditions in combination with subsequent pyrolysis, leading to a new NPME. Our NPME exhibits a half-wave potential of 0.89 V vs . reversible hydrogen electrode (RHE), an onset potential of 1.0 V vs . RHE and an average HO 2 − yield below 2% as well as high durability toward oxygen reduction reactions (ORR) in alkaline electrolytes, ranking at the top of all reported NPMEs derived from hemin.

Herein, a smart porous material, Cu-hemin metal-organic-frameworks (Cu-hemin MOFs), was synthesized via assembling of Cu 2+ with hemin to load glucose oxidase (GOD) for electrochemical glucose biosensing for the first time. The formation of the Cu-hemin MOFs was verified by scanning electron microscopy, X-ray powder diffraction, Fourier transform infrared spectroscopy, N 2 adsorption/desorption isotherms, UV-vis absorption spectroscopy, fluorescence spectroscopy, thermal analysis and electrochemical techniques. The results indicated that the Cu-hemin MOFs showed a ball-flower-like hollow cage structure with a large specific surface area and a large number of mesopores. A large number of GOD molecules could be successfully loaded in the pores of Cu-hemin MOFs to keep their bioactivity just like in a solution. The GOD/Cu-hemin MOFs exhibited both good performance toward oxygen reduction reaction via Cu-hemin MOFs and catalytic oxidation of glucose via GOD, superior to other GOD/MOFs and GOD/nanomaterials. Accordingly, the performance of GOD/Cu-hemin MOFs-based electrochemical glucose sensor was enhanced greatly, showing a wide linear range from 9.10 μM to 36.0 mM and a low detection limit of 2.73 μM. Moreover, the sensor showed satisfactory results in detection of glucose in human serum. This work provides a practical design of new electrochemical sensing platform based on MOFs and biomolecules.

It is of great importance to simply and sensitively detect hydrogen sulfide (H 2 S) because of its role in various physiological processes as well as its inherent toxicity. In this work, a colorimetric method for H 2 S detection was developed by employing terbium-G-quadruplex-hemin (Tb/G4-hemin) DNAzyme as a peroxidase mimic, which can catalyze H 2 O 2 -mediated oxidation of 2,2′-azinobis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) to produce radical cation (ABTS + ). Compared with the G4-hemin DNAzyme promoted by K + and Na + , Tb/G4-hemin DNAzyme exhibits a higher catalytic activity. In the presence of Ag + , the peroxidase-like activity of Tb/G4-hemin DNAzyme can be inhibited significantly owing to the disruption of G-quadruplex structure. However, the addition of H 2 S can effectively suppress such negative behavior by competitive binding with Ag + , leading to the recovery of the peroxidase-like activity of Tb/G4-hemin DNAzyme, which can be reflected by an increase in absorbance signal of ABTS + . The absorbance of ABTS + was enhanced linearly with increasing H 2 S concentration from 20 nM to 2 μM. The detection limit for H 2 S is 13 nM, which is much lower than most of previous methods. Moreover, the proposed method possesses the features of simple preparation, easy reproducibility and good biocompatibility. Given the fine performances and striking properties, we believe that the Tb/G4-hemin DNAzyme would have a great promise for analytical applications.

Constructing optical nanoprobes with superior performance is highly desirable for sensitive and accurate assays. Herein, we develop a facile room-temperature strategy for the fabrication of green emissive carbon nanoclusters (CNCs) with dual-exciting centers for the dual-channel sensing of hemin. The formation of the CNCs is attributed to the crosslinking polymerization of the precursors driven by the Schiff base reaction between ethylenediamine and 2,3-dichloro-5,6-dicyano-1,4-benzoquinone. Most importantly, the proposed CNCs have a unique excitation-independent green emission (518 nm) with two excitation centers at 260 nm (channel 1) and 410 nm (channel 2). The dual-exciting central emission can serve as dual-channel fluorescence (FL) signals for highly sensitive and reliable detection of hemin based on the inner filter effect. Because of the great spectral overlap difference between the absorption spectrum of hemin and the excitation lights of the CNCs in the two channels, hemin has a different quenching effect on FL emission from different channels. The dual-channel signals of the CNCs can detect hemin in the range of 0.075–10 μM (channel 1) and 0.25–10 μM (channel 2), respectively. These findings not only offer new guidance for the facile synthesis of dual-exciting central CNCs but also establish a reliable sensing platform for the analysis of hemin in complex matrixes.

Surgical site infections are the most common and serious concern in health care settings. Currently, systemic administration of prophylactic antibiotics or local disinfection treatment is prescribed after surgical procedures. However, prolonged use of antibiotics may pose adverse side effects, bacterial resistance and hospitalization costs. Recently, the use of nanomaterials with peroxidase enzyme-mimic activity has drawn higher consideration due to their high antibacterial potential and minimum biotoxicity. Hemin is a highly active molecule and also the active source of many biological enzymes, such as peroxidase, which could efficiently catalyze various biochemical reactions. However, the insolubility of hemin makes it easy for it to aggregate and lose its catalytic ability. Here, we designed a well-dispersed hemin/graphdiyne (GDY) nanocomposite (GDY–hemin) with high peroxidase activity. GDY–hemin can decompose hydrogen peroxide (H2O2) to generate highly-toxic hydroxyl radicals that impair bacterial cell membranes, ultimately leading to bacterial death in both in vitro and in vivo experiments. The results reveal that GDY–hemin has good biocompatibility and is suitable for wound infection treatment in vivo.

Hemin, the most common biological iron form, serves many biological functions. However, studies on hemin detection are scarce. As an unusual natural peroxide, artemisinin is quite stable in the presence of common species. While, it can decompose to generate superoxide radicals in the presence of hemin. And the as-obtained superoxide radicals can effectively oxidize thiamine to form fluorescent thiochrome. Inspired by this, a simple and novel fluorescent assay for hemin detection based on artemisin-thiamine is developed. Based on the linear relationship between the fluorescence intensity and concentration of hemin, the developed artemisinin-thiamine assay can be utilized for sensitive detection of hemin in the concentration range of 2.0–300.0 nM with a detection limit of 0.68 nM. Moreover, this assay shows high selectivity over other cations and biomolecules. Besides, hemin can be detected in serum samples with satisfactory results, which demonstrates its great potential for real sample analysis. The designed assay is free of any fluorescent nanomaterials or probes by just mixing these reagents together, and all the detection processes is performed within 20 min. Therefore, it can offer a simple, highly sensitive and selective sensing platform for fluorescence detection of hemin.

As a recently developed detection technique, photoelectrochemical (PEC) measurement has received extensive attention. However, owing to the lack of a comprehensive approach to engineer the photo-generated carriers, the performance is still limited. As an example, a significant use of PEC measurement might be in understanding the roles of glutathione in physiological and pathological processes. Here, we developed a new approach for engineering the photo-generated carriers with the aid of the synergic effect of self-doping, surface plasmon resonance, electrical field amplification effect, etc. Thus, a highly sensitive multicomponent PEC platform has been developed, in which Au nanorods are decorated on the surface of self-doped TiO2 nanowires, followed by surface receptor functionalization with hemin for capturing glutathione. The synergic effect effectively increases generation, separation and transfer kinetics of the photo-generated carriers, which can be further increased by using a mixture of Au nanorods with different aspect ratios to tune the absorption wavelength to the entire UV-visible region. As a result, this system exhibits a broad linear range from 10 nM to 17.5 μM and low detection limit down to 8.6 nM for detecting glutathione, about 1–2 orders of magnitude lower than most existing PEC sensors.

The encapsulation of catalytic active centers into polymer hydrogels represents a simple and effective strategy for enzyme mimicry. Current methods only enable the preparation of macroscopic enzyme carriers, which are far from natural enzymes in terms of their sizes, local chemical and physical properties, and catalytic activities. In this work, a nanogel strategy to immobilize hemin, a mimic of the active center for horseradish peroxidase (HRP), into a polyacrylamide (PAAm) matrix is demonstrated. Hemin was modified using N-(3-aminopropyl) methacrylamide hydrochloride (APMA) and the modified hemin together with excess APMA were covalently incorporated into PAAm nanogels. The resulting Hem@Gel shows enhanced catalytic activity and environmental tolerance compared with hemin and unmodified nanogel Hem/Gel, which has been demonstrated via the catalytic oxidation of various organic dyes. Further characterization and molecular simulations reveal the Fe–N coordination between the pendant groups and the enrichment of dye molecules and H2O2 in PAAm nanogels, which together enhance the local reaction rate and realize an overall improvement in catalytic efficiency.

In the present work, a highly sensitive and specific fluorescent biosensor for blood glucose monitoring is developed based on hemin-functionalized graphene quantum dots (GQDs) and glucose oxidase (GOx) system. The GQDs which are simply prepared by pyrolyzing citric acid exhibit strong fluorescence and good water-solubility. Due to the noncovalent assembly between hemin and GQDs, the addition of hemin can make hydrogen peroxide (H 2 O 2 ) to destroy the passivated surface of GQDs, leading to significant fluorescence quenching of GQDs. Based on this effect, a novel fluorescent platform is proposed for the sensing of glucose. Under the optimized conditions, the linear range of glucose is from 9 to 300 μM, and the limit of detection is 0.1 μM. As unique properties of GQDs, the proposed biosensor is green, simple, cost-efficient, and it is successfully applied to the determination of glucose in human serum. In addition, the proposed method provides a new pathway to further design the biosensors based on the assembly of GQDs with hemin for detection of biomolecules.

The traditional G-quadruplex/hemin horseradish peroxidase has poor stability and a strong background signal and is not suitable for the detection of heavy metals such as mercury ions. In this work, we used the previous DNA strand for mercury ion detection and optimized it. Hemin is covalently bound to the 5′ and 3′ ends of the DNA strand. In the covalent binding state of double Hemin, the G-quadruplex has a strong catalytic activity and can catalyze the oxidation of hydrogen peroxide by ABTS. The formation of double chains with the addition of the S2 chain provides a rigid structure for detecting mercury ions. At the same time, after the addition of mercury ions, the formation of T-Hg2+-T structure further hinders the formation of G-quadruplex and reduces the absorbance value. The detection range in ultrapure water was 0–1000 nM, and the limit of detection concentration was 0.61 pM. In the human serum, the detection range is 0–300 nM and the LOD is 0.1 nM. In addition, the biosensor has excellent selective specificity and is expected to be used to detect mercury ions in organic biological environments and industrial environments contaminated by heavy metals.

Hemin/G-quadruplex (hG4) complexes are frequently used as artificial peroxidase-like enzymatic systems (termed G4 DNAzymes) in many biosensing applications, in spite of a rather low efficiency, notably in terms of detection limits. To tackle this issue, we report herein a strategy in which hemin is chemically modified with the amino acids found in the active site of parent horseradish peroxidase (HRP), with the aim of recreating an environment conducive to high catalytic activity. When hemin is conjugated with a single arginine, it associates with G4 to create an arginine–hemin/G4 (R-hG4) DNAzyme that exhibits improved catalytic performances, characterized by kinetic analysis and DFT calculations. The practical relevance of this system was demonstrated with the implementation of biosensing assays enabling the chemiluminescent detection of G4-containing DNA and colorimetry detection of the flap endonuclease 1 (FEN1) enzyme with a high efficiency and sensitivity. Our results thus provide a guide for future enzyme engineering campaigns to create ever more efficient peroxidase-mimicking DNA-based systems.
