Fabrication of hyaluronic acid-inulin coated Enterococcus faecium for colon-targeted delivery to fight Fusobacterium nucleatum

Pengrong Zhu,Lingtong Meng,Yue Shu,Yuqiao Xu,Weiming Liu,Yong Bi,Jian Xu,Lijuan Meng,Yanan Li

The abundance of Fusobacterium nucleatum ( F. nucleatum ) is highly associated with the development and poor prognosis of colorectal cancer (CRC), which is regarded as a promising target for CRC. However, until now, the novel strategy to clear F. nucleatum in the colon and CRC has not been well proposed. Herein, a probiotic strain Enterococcus faecium ( E. faecium , EF47) is verified to secrete various organic acids and bacteriocins to exert superior antimicrobial activity towards F. nucleatum . However, the oral delivery of EF47 is affected by the complex digestive tract environment, so we design the hyaluronic acid-inulin (HA-IN) coated EF47 for colon-targeted delivery to fight F. nucleatum . IN can protect EF47 from the harsh gastrointestinal tract environment and is degraded specifically in the colon, acting as prebiotics to further promote the proliferation of EF47. The exposed HA can also enhance the targeting effect to the tumor area via the interaction with the CD44 receptor on the tumor cells, which is confirmed to increase the adhesive ability in tumor tissues and inhibit the growth of F. nucleatum . Therefore, this colon-targeted delivery system provides a novel platform to realize high-activity and adhesive delivery of probiotics to assist the therapeutic efficiency of CRC.

Inulin-armored hesperetin‑zinc nanodrugs for synergistic redox-immune modulation and barrier repair in ulcerative colitis

Lu Li,Jia Liu,Xiuyan Shi,Chenman Yuan,Zhetong Jin,Xue Rui,Yi Chen,Lingchong Wang,Wei Li,Yugen Chen,Jingjing Wang

Ulcerative colitis (UC) is a chronic relapsing inflammatory bowel disease with a rising global incidence rate, which poses a significant challenge to clinical management due to the limited efficacy and adverse reactions of current treatments. To overcome these limitations, we engineered Hes-Zn@IN, a novel core-shell nanoparticle system combining a hesperetin‑zinc (Hes Zn) metal-polyphenol coordination core encapsulated in an inulin (IN) polysaccharide shell. This nanotherapeutic platform exhibits synergistic therapeutic benefits: superior antioxidant capacity through metal-polyphenol coordination, demonstrating a 38% reduction in ROS and a 47% decrease in NO production in LPS-activated macrophages; robust immunomodulatory effects, significantly lowering pro-inflammatory cytokines (46% reduction in TNF-α, 42% in IL-6) while increasing anti-inflammatory IL-10 by 78%; a pH-dependent release profile enabling 1.85 times greater drug accumulation in inflamed colon tissue. In murine DSS-induced colitis models, oral administration of Hes-Zn@IN produced remarkable therapeutic outcomes, including 19.2% improvement in colon length, restoration of ZO-1 tight junction protein expression, and substantial mitigation of histopathological inflammation. Our findings establish this inulin-armored metal-polyphenol hybrid as a breakthrough nanomedicine that synergistically targets colonic redox-immune remodeling and barrier repair, presenting a comprehensive therapeutic approach for UC management.

Pharmacokinetics, distribution and excretion of inulin-type fructan CPA after oral or intravenous administration to mice

Yao Guo,Yun-yun Shao,Yi-nan Zhao,Xiao Zhang,Zhuang-peng Chang,Yi-fan Sun,Jun-jin Liu,Jianping Gao,Rui-gang Hou

The aim of this work has been to establish and validate a simple and efficient method to detect the concentration of inulin-type fructan CPA from the roots of Codonopsis pilosula (Franch.) Nannf. in biosamples, and then apply it to evaluate the pharmacokinetics behavior, distribution character in tissue and excretion in mice. In this work, fluorescein isothiocyanate (FITC) was used to label CPA. Then FCPA was intravenously and orally administered to mice at different doses. In both i.v and p.o administration, FCPA concentration slowly declined in the circulatory system with a much longer T1/2 and MRT. After p.o administration, the area under the time curve (AUC0-∞) was dose-dependently increased. Taken together, FCPA showed poor absorption and wide tissue distribution. These pharmacokinetic results yield helpful insights into the pharmacological actions of FCPA.

Inulin alleviates offspring asthma by altering maternal intestinal microbiome composition to increase short-chain fatty acids

Guifang Yuan,Song Wen,Xuemei Zhong,Xiaotong Yang,Linrui Xie,Xiuli Wu,Xiaoyu Li

Technically, symptom of offspring asthma is also closely reliant on its maternal high-fiber diet as well as the intestinal microbiome. Fruits and vegetables are abundant in inulin, and this naturally soluble dietary fiber is endowed with a potential value on offspring asthma control through the maternal intake, but the mechanism now remains less studied. In this study, rats were given with inulin-included drinking water, whereas in normal group rats were allowed with normal water. Afterwards, we analyzed both the formations of the offspring intestinal microbiome ahead of asthma model establishment and of the maternal intestinal microbiome through high throughput sequence and the short-chain fatty acids (SCFAs) by metabolomic analysis. Subsequently, lung inflammation indexes were detected by Elisa, and the expression of short-chain fatty acid receptors (GPR41, GPR43) in the offspring of asthma models were evaluated through qPCR assay. Inulin intake resulted in altered maternal intestinal microbiome composition, with a significant increase in SCFAs-producing bacteria (mainly Bifidobacterium ), attenuating the asthmatic inflammatory response in the offspring. Meanwhile, inulin intake during pregnancy modulates the composition of the intestinal microbiome of the offspring, and this alteration appears before the onset of asthma, hence, there should be further studies onto the impacts of offspring’s intestinal microbiome on asthma procession.

Optimized acid hydrolysis conditions for better characterization the structure of inulin-type fructan from Polygonatum sibiricum

Xinyan Zong,Zhe Wang,Shikang Chen,Si Li,Mingyong Xie,Shaoping Nie,Junyi Yin

Polygonatum sibiricum is an edible plant species in China known for its abundant polysaccharides . However, correlations between its analytical methods and fine structure have not been established. This is usually due to incomplete cleavage of the glycosidic linkages and instability of hydrolysis . In this study, a new optimal acid hydrolysis method for monosaccharide composition (2 M H 2 SO 4 for 1 h) and methylation analysis (2 mol TFA hydrolysis at 100 °C for 1 h) was developed for characterization of inulin-type fructans , resulting in significantly improved monosaccharide recovery and providing more reliable methylation data. The effectiveness of this method was demonstrated through its application to the study of polysaccharide from P. sibiricum (IPS-70S). The results showed that IPS-70S with a molecular weight of 3.6 kDa is an inulin-type fructans consisting of fructose and glucose in a molar ratio of 27:1. Methylation and NMR analysis indicated that IPS-70S contains →2)-Fru f -(6 → or →2)-Fru f -(1 → with branching →1,6)-Fru f -(2 → and terminates in Glc p -(1 → or Fru f -(2→. In conclusion, optimal acid hydrolysis applicable to the specific polysaccharides contribute to its structurally characterized. The newly optimized acid hydrolysis method for monosaccharide composition and methylation analysis offers a reliable and effective approach to the structural characterization of inulin-type fructans from P. sibiricum . Providing reliable basis for the overall work of NMR analysis and structural analysis, which have potential significance in the field of polysaccharides structural characterization.

Butyrate-Conjugated Poly(vinyl alcohol) Nanoparticles in an Inulin Hydrogel for Colon-Targeted Drug Delivery in Colitis

Lu Han,Qingqing Pan,Mengting Chen,Xi Fan,Bin He,Yuji Pu

The development of oral drug delivery systems with colon-targeted release and high biocompatibility is of critical importance for the treatment of ulcerative colitis (UC), considering the chronic nature of the disease and the genetic susceptibility of patients. Herein, we report a composite delivery system (UA@PB/Gel) based on Generally Recognized As Safe (GRAS) materials, poly(vinyl alcohol) (PVA) and inulin, designed for colon-specific delivery of ursolic acid (UA). In this system, butyrate-conjugated PVA nanoparticles efficiently encapsulate UA and respond to colonic esterase, enabling the localized release of both UA and butyrate. Embedding these nanoparticles within an inulin hydrogel further enhances colonic retention and provides a sustained release. The composite system demonstrates efficient colon-targeting delivery, prolonged retention, and potent anti-inflammatory effects in vitro and in vivo. In a dextran sulfate sodium-induced colitis mouse model, UA@PB/Gel effectively alleviates colonic inflammation, restores epithelial barrier integrity, reduces proinflammatory cytokine expression, and modulates gut short-chain fatty acid levels, with minimal systemic toxicity. These results highlight the potential of the GRAS materials-based nanoparticle–hydrogel composite as a safe and effective therapeutic platform for UC.

Efficacy of a composite aluminum adjuvant containing inulin in protecting turbot (Scophthalmus maximus) against Aeromonas salmonicida infection

Yingbo Wu,Weiwei Qian,Qin Liu,Xiaohong Liu

An inulin-aluminum composite adjuvant achieved 61.54% RPS against A. salmonicida. • It enhanced lysozyme, total protein, and specific IgM vs. single adjuvants. • It promoted antigen uptake and upregulated Th1-type cytokines and T cell markers. • This adjuvant offers a safe, effective strategy for fish vaccine development. Inulin is a type of plant polysaccharide that is widely distributed in nature. Research has demonstrated that inulin possesses adjuvant activity, stimulating balanced cellular and humoral immune responses. However, the role of inulin as an adjuvant has been relatively underexplored within aquatic vaccine research. In this study, a composite aluminum adjuvant containing inulin was developed, and an inactivated vaccine against Aeromonas salmonicida was formulated based on this adjuvant. Subsequently, the immunoprotective effects of the vaccine in turbot were evaluated to assess the potential application of inulin as an adjuvant in aquaculture vaccines. As a result, the composite of aluminum and inulin significantly promoted immune cell recruitment and antigen uptake at the injection site. Vaccination with the composite adjuvant significantly reduced mortality following A. salmonicida challenge, achieving a relative percent survival (RPS) of 61.54% compared to the control group. Moreover, the vaccine exhibited increased levels of specific IgM along with elevated serum protein levels and lysozyme activity. Furthermore, gene expression analysis revealed significant upregulation of MHC-I , MHC-IIβ , CD4 , CD8 as well as some cytokines including IL-4 , IL-10 , IL-1β , IFN-γ , IL-12 and TNF-α , indicating a robust humoral and cellular immune response. In brief, these findings suggest that inulin acts as a natural immune stimulant and, when combined with aluminum, confers enhanced protective efficacy compared with either adjuvant alone, while balancing both humoral and cellular immune responses. Further studies are needed to clarify the nature of this complementary effect. This novel composite of adjuvants presents a leading candidate strategy for improving fish vaccination practices within aquaculture aimed at bolstering disease prevention efforts. Further studies are warranted to optimize this formulation and investigate the mechanisms underlying immune enhancement provided by the composite adjuvant system for teleost fish.

An inulin-type fructan from Codonopsis pilosula ameliorates cyclophosphamide-induced immunosuppression and intestinal barrier injury in mice

Xingfang Xie,Yao Gu,Yi Liu,Mingyue Shen,Jiaojiao Ji,Jianping Gao,Jiankuan Li

In the present study, an inulin-type fructan (ITF) with the degree polymerization (DP) of 21 was isolated from Codonopsis pilosula roots and its structure was characterized by FT-IR, MALDI-TOF-MS and NMR. The immunomodulatory and intestinal protective effects of ITF were investigated on immunosuppressive mice. Male BALB/c mice were pretreated with cyclophosphamide (Cy) for 3 days to establish an immunosuppressive model followed by ITF treatment. The results demonstrated that compared with the model group, ITF administration significantly increased immune organ index ( P <0.05), alleviated intestinal villus damage, stimulated serum cytokine secretion including Ig G, IL-4, IL-6, IL-2, TNF-α, and INF-γ ( P <0.05), upregulated the expression of Occludin and Claudin-1 ( P <0.05), and increased CD4 + and CD8 + T cells of ileum in Cy-induced mice ( P <0.05). Furthermore, ITF restored the intestinal microbiota dysbiosis caused by Cy by increasing the abundance of Muribaculaceae , Blautia , Odoribacter , Lactobacillus and decreasing the abundance of Lachnospiraceae _ NK4A136 _ group ( P <0.05). Meanwhile, ITF increased the production of short-chain fatty acids (SCFAs) including acetic acid, propionic acid and butyric acid ( P <0.05). These results indicated that ITF can ameliorate cyclophosphamide-induced immunosuppression and intestinal barrier injury, and restore gut microbiota dysbiosis. This study provided important evidences for the immunomodulatory and intestinal protective effects of the ITF from C. pilosula .

Inulin Can Improve Red Blood Cell Cryopreservation by Promoting Vitrification, Stabilizing Cell Membranes, and Inhibiting Ice Recrystallization

Yuying Hu,Xiangjian Liu,Wenqian Zhang,Jiangming Chen,Xiaoxiao Chen,Songwen Tan

In transfusion medicine, the cryopreservation of red blood cells (RBCs) is of major importance. The organic solvent glycerol (Gly) is considered the current gold-standard cryoprotectant (CPA) for RBC cryopreservation, but the deglycerolization procedure is complex and time-consuming, resulting in severe hemolysis. Therefore, it remains a research hotspot to find biocompatible and effective novel CPAs. Herein, the natural and biocompatible inulin, a polysaccharide, was first employed as a CPA for RBC cryopreservation. The presence of inulin could improve the thawed RBC recovery from 11.83 ± 1.40 to 81.86 ± 0.37%. It was found that inulin could promote vitrification because of its relatively high viscosity and glass transition temperature (Tg′), thus reducing the damage during cryopreservation. Inulin possessed membrane stability, which also had beneficial effects on RBC recovery. Moreover, inulin could inhibit the mechanical damage induced by ice recrystallization during thawing. After cryopreservation, the RBC properties were maintained normally. Mathematical modeling analysis was adopted to compare the performance of inulin, Gly, and hydroxyethyl starch (HES) in cryopreservation, and inulin presented the best efficiency. This work provides a promising CPA for RBC cryopreservation and may be beneficial for transfusion therapy in the clinic.

Study of the alleviation effects of a combination of Lactobacillus rhamnosus and inulin on mice with colitis

Zhijing Liu,Fei Liu,Wan Wang,Changbao Sun,Da Gao,Jiage Ma,Muhammad Altaf Hussain,Cong Xu,Zhanmei Jiang,Juncai Hou

Ulcerative colitis (UC) is a common inflammatory bowel disease (IBD) that has serious harmful effects on human health. Lactobacillus rhamnosus, a probiotic, has a strong colonization and adhesion effect and improves the intestinal health of the host. Inulin has good anti-inflammatory effects and can promote the proliferation of beneficial intestinal bacteria. The purpose of this study was to investigate the alleviating effects of L. rhamnosus 1.0320 in combination with inulin on UC, examining the resulting changes in intestinal flora. A UC model was established by having mice freely drink a 3% (w/v) dextran sodium sulphate (DSS) solution for seven days. After successful modeling, the mice were given antibiotics, L. rhamnosus 1.0320 by itself, inulin by itself, and L. rhamnosus 1.0320 combined with inulin as an intragastric intervention for 28 days. The abundance and structural changes of bacteria in the intestinal content of mice were analyzed by 16S rDNA high-throughput sequencing. The study found that male BALB/c mice can successfully establish a typical model of small intestinal inflammation by freely drinking a 3% DSS solution for one week. L. rhamnosus 1.0320 combined with inulin can alleviate DSS-induced colitis, reduce the Disease Activity Index (DAI) score of the pathological damage of colon tissue, decrease myeloperoxidase (MPO) activity, increase hemoglobin content, and regulate the expression levels of inflammatory cytokines IL-1β, IL-6, TNF-α and IL-10. The intestinal flora of mice is reduced after enteritis, and its structure gets disordered. The combination of L. rhamnosus 1.0320 and inulin can increase the abundance and diversity of intestinal flora, and increase the content of beneficial bacteria. Prebiotics promote the colonization ability of probiotics. L. rhamnosus 1.0320 combined with inulin can change the intestinal flora to relieve ulcerative colitis, providing a new theoretical basis for the study of UC mechanism.

Yields of three acids during simulated fermentation of inulin and xylo-oligosaccharides enhanced by six exogenous strains

Kong Lu,Zhao Xin-Huai

Three acids (acetic, propionic and butyric acids) yielded from simulated colonic fermentation of inulin and xylo-oligosaccharides (XOS) in the absence and presence of six exogenous strains were investigated in this study. Both inulin and XOS were fermented by fecal extract (FE) prepared from healthy adults and by an exogenous strain for 48 h, and three acids generated in fermentation products at five time periods were detected by gas chromatography. Inulin and XOS during fermentation were capable of producing more propionic and butyric acids, respectively, whilst inoculation using one strain culture into fermentation systems mostly increased yields of three acids. In inulin-containing fermentation systems, these strains increased yields of propionic and butyric acids by 6–34 and 206–250%, respectively. In XOS-containing fermentation systems, these strains did not enhance yield of propionic acid, but increased yield of butyric acid by 50–96%. In general, Lactobacillus brevis, Lactobacillus plantarum, Lactobacillus rhamnosus, Enterococcus faecalis and Enterococcus faecium were more efficient than Lactobacillus acidophilus to increase yields of propionic and butyric acids. It is thus concluded that the six strains in total displayed beneficial effects on simulated fermentation of inulin and XOS through enhancing yields of three acids especially propionic and butyric acids with healthcare functions.

Dehydration of fructose, sucrose and inulin to 5-hydroxymethylfurfural over yeast-derived carbonaceous microspheres at low temperatures

Xiaofeng Li,Yi Wang,Xiaomin Xie,Changhong Huang,Sen Yang

This work prepared carbonaceous microspheres by hydrothermal carbonization of yeast cells followed by sulfonation with concentrated sulphuric acid (98%) at room temperature. The obtained carbonaceous product (CM-SO3H) had a high acid density (1.80 mmol g−1). We evaluated CM-SO3H as a solid catalyst for the dehydration of fructose-based carbohydrates to 5-hydroxymethylfurfural (5-HMF) in the ionic liquid 1-butyl-3-methylimidazolium chloride ([BMIM][Cl]). The effects of the catalyst and substrate loadings as well as the reaction temperature and time on the yield of 5-HMF were investigated. Under the optimum conditions, a 5-HMF yield of up to 83.5% was obtained from fructose with a reaction temperature of 80 °C for 30 min. Furthermore, 44.8% and 59.2% 5-HMF yields were obtained from sucrose (80 °C for 30 min) and inulin (80 °C for 60 min), respectively. CM-SO3H and [BMIM][Cl] showed high stability and could be recycled between five and eight times without significant loss of catalytic activity. More importantly, the catalytic system could be applied to high substrate concentrations. CM-SO3H combined with [BMIM][Cl] is a promising system for transforming fructose-based carbohydrates into 5-HMF.
