Luteolin-based epoxy resin with exceptional heat resistance, mechanical and flame retardant properties

Tian-Yu Gao,Fen-Dou Wang,Yu Xu,Chun-Xiang Wei,San-E Zhu,Wei Yang,Hong-Dian Lu

Recently, tremendous attentions have been paid to the fabrication of bio-based intrinsic fire-retarded epoxy resins with outstanding thermal and mechanical properties from renewable resources. Herein, the bio-based epoxy monomer of diglycidyl ether luteolin (DGEL) was synthesized via one-step method from sustainable luteolin. The biocompatible epoxy resin was manufactured via the curing reaction between DGEL and 4, 4′-diaminodiphenyl sulfone (DDS). DGEL/DDS showed higher glass transition temperature (T g ) (314.4 °C) than diglycidyl ether bisphenol A (DGEBA)/DDS (217.4 °C), demonstrating the distinguished heat resistance. Tensile testing results exhibited that DGEL/DDS possessed superior tensile strength (69.2 MPa) compared to DGEBA/DDS (57.2 MPa). Thermogravimetric analysis (TGA) illustrated that the char yield of DGEL/DDS was 44.0 wt%, much higher than that of DGEBA/DDS (11.6 wt%). In addition, DGEL/DDS passed the V-0 rating in UL-94 tests with a high limiting oxygen index (LOI) (32.5%) as well as extremely low peak heat release rate (107.5 W/g) indicating the excellent flame retardant properties. Smoke density testing results showed that the peak density of smoke for DGEL/DDS was 304.7, much lower than that for DGEBA/DDS (660.7), indicating the remarkable reduction of toxicity hazards. This work provides a facile approach for the preparation of bio-based epoxy resins with outstanding overall properties that show promising applications in high heat resistance and flame retardant fields.

Molecularly imprinted polymer for the selective extraction of luteolin from Chrysanthemum morifolium Ramat

Die Gao,Fengqing Yang,Zhining Xia,Qihui Zhang

In this work, luteolin-imprinted polymers were prepared by noncovalent precipitation polymerization for the first time. Their structural features and morphologies were analyzed by using Fourier transform infrared spectroscopy and scanning electron microscopy, respectively. The adsorption experiments revealed that the luteolin-imprinted polymers presented high selective recognition property to luteolin. The selectivity experiment showed that the adsorption capacity and selectivity of polymers to luteolin was higher than that of three structural analogs, including quercetin, isorhamnetin, and ombuin. Furthermore, an efficient method based on luteolin-imprinted polymers coupled with solid-phase extraction was developed for the pretreatment of luteolin from Chrysanthemum morifolium Ramat. The results demonstrated that the luteolin-imprinted polymers coupled with solid phase extraction method was proven to be a potentially competitive technique for the separation and enrichment of luteolin in complex samples such as Chinese patent medicines and biological samples.

Ultrasensitive Luteolin Electrochemical Sensor Based on Novel Lamellar CuZn@ Nitrogen-Containing Carbon Nanosheets

Yuhong Li,Yaqi Yang,Jiejun Li,Li Zhang,Pengcheng Zhao,Junjie Fei,Yixi Xie

The Cu/Zn-zeolitic imidazolate framework (Cu/Zn-ZIF) was synthesized using the traditional hydrothermal method, and its surface morphology was controlled by adding polyvinylpyrrolidone (PVP) during its synthesis. It was then calcined at 800 °C to form the nitrogen-containing carbon material[email protected], which improved the electron transfer rate. Scanning electron microscopy (SEM), X-ray crystal diffraction (XRD), and X-ray photoelectron spectroscopy (XPS) were used to investigate the surface morphology and structure. Finally, the electrochemical sensing platform for luteolin was effectively constructed by changing the metal–ion ratio during synthesis to achieve the most suitable electrode material. The sensor platform detects luteolin well, with an operating curve equation of Ip (A) = 0.0571C (nM) − 1.2913 and a minimum detection limit of 15 nM, and the platform has been successfully employed for luteolin detection in real samples. Keywords:lamellar structure;shape control;nitrogen-containing carbon;fast response

Luteolin ameliorates DSS-induced colitis in mice via suppressing macrophage activation and chemotaxis

Liwei Xue,Xiaosheng Jin,Tingting Ji,Rongzhou Li,Xiaoju Zhuge,Fang Xu,Zijiao Quan,Haibin Tong,Weilai Yu

Objectives Luteolin, known for its multifaceted therapeutic properties against inflammatory diseases, holds potential for addressing the unmet need for effective treatments in ulcerative colitis (UC), a prevalent subtype of inflammatory bowel disease (IBD). This study aimed to comprehensively assess luteolin's therapeutic efficacy in a dextran sulfate sodium (DSS)-induced colitis mouse model, shedding light on its anti-UC mechanisms. Methods Our investigation encompassed in vivo assessments of luteolin's therapeutic potential against DSS-induced colitis through rigorous histopathological examination and biochemical analyses. Furthermore, we scrutinized luteolin's anti-inflammatory prowess in vitro using lipopolysaccharide (LPS)-stimulated RAW264.7 cells and primary peritoneal macrophages. Additionally, we quantitatively evaluated the impact of luteolin on C–C motif chemokine ligand 2 (CCL2)-induced macrophage migration employing Transwell and Zigmond chambers. Furthermore, cellular thermal shift assay (CETSA), drug affinity responsive target stability (DARTS) assay, and molecular docking were employed to identify potential therapeutic targets of luteolin and investigate their binding sites and interaction patterns. Results Luteolin demonstrated therapeutic potential against DSS-induced colitis by ameliorating colitis symptoms, restoring intestinal barrier integrity, and inhibiting proinflammatory cytokine production in the colonic tissues. Moreover, luteolin demonstrated robust anti-inflammatory activity in vitro , in lipopolysaccharide (LPS)-stimulated RAW264.7 cells and primary peritoneal macrophages. Notably, luteolin suppressed the phosphorylation of IKKα/β, IκBα, and p65, along with preventing IκBα degradation in LPS-treated RAW264.7 cells and peritoneal macrophages. Furthermore, luteolin impaired the migratory behavior of RAW264.7 cells and peritoneal macrophages, as evidenced by reduced migration distance and velocity of luteolin-treated macrophages. Mechanistically, luteolin was found to antagonize IKKα/β, subsequently inhibiting IKKα/β phosphorylation and the activation of NF-κB signaling. Conclusion Luteolin emerges as a promising lead compound for the clinical therapy of colitis by virtue of its ability to ameliorate DSS-induced colitis, antagonize IKKα/β, suppress NF-κB signaling, and impede macrophage activation and migration.

Fabrication of Luteolin Loaded Zein-Caseinate Nanoparticles and its Bioavailability Enhancement in Rats

Qiao-Ting Xu,Wen-Xuan Zhang,Hai-Xia Xu,Qing-Feng Zhang

Luteolin loaded zein nanoparticles (Lut-ZNP) were prepared by using sodium caseinate as an electrostatic stabilizer. The formulation of the nanoparticles was optimized. Lut-ZNP were spray-dried, and the physicochemical properties were characterized by SEM, XRD , FT-IR and DSC. Then, the bioavailability of luteolin in rats was determined. Under the formulation of luteolin, zein and sodium caseinate with mass ratio of 1:5:15, the particle size, ζ-potential, encapsulation efficiency and loading efficiency of Lut-ZNP were 171.8 nm , -49.05 mV, 85.85% and 3.15%, respectively. Luteolin existed in the nanoparticles with amorphous form. Lut-ZNP exhibited good redispersibility in water after drying. Compared with free luteolin, the solubility, stability and release of luteolin in Lut-ZNP were greatly improved. Besides, the fecal excretion of luteolin in rats was significantly reduced after oral administration of Lut-ZNP. In addition to native luteolin, its metabolites including sulfate, glucuronidate and methylated glucuronidate were found in rat plasma. Lut-ZNP significantly increased the plasma concentrations of luteolin and its metabolites, and the bioavailability of luteolin was enhanced by 2.92 times.

Inhibitory effect of luteolin on the metabolism of vandetanib in vivo and in vitro

Yuxin Shen,Fengsheng Hong,Hualu Wu,Xiaohai Chen,Hailun Xia,Ren-ai Xu,Guanyang Lin,Lu Shi

This study aimed to examine the potential drug-drug interaction (DDI) between vandetanib and luteolin in vivo and in vitro, with the objective of establishing a scientific foundation for their appropriate utilization in clinical settings. Sprague-Dawley (SD) rats were randomly divided into two groups: a control group (vandetanib administered by gavage alone) and an experimental group (vandetanib and luteolin administered together). A series of blood samples were collected at different time intervals. The plasma concentrations of vandetanib and its metabolite N-demethyl vandetanib in rats were determined using an ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS). Incubation systems were set up with rat liver microsomes (RLM) and human liver microsomes (HLM) to measure the Michaelis-Menten constant (Km) and half-maximum inhibitory concentration (IC50) values. Additionally, the inhibitory mechanism of luteolin on vandetanib was also investigated. Ultimately, the molecular mechanism of inhibition was examined through the utilization of molecular docking techniques. In vivo animal experiment results showed that compared with the control group, the AUC(0-t) and Cmax of vandetanib in the experimental group were significantly increased. The findings from the in vitro experiments revealed that luteolin exhibited a moderate inhibitory effect on the metabolism of vandetanib. The IC50 values for RLM and HLM were determined to be 8.56 μM and 15.84 μM, respectively. The identified inhibition mechanism was classified as mixed. This study utilized molecular docking analysis to provide additional evidence supporting the competitive inhibition of luteolin on vandetanib in CYP3A4. The data presented in our study indicated a potential interaction between vandetanib and luteolin, which may necessitate the need for dose adjustment during their co-administration in clinical settings.

NiCoZn Oxide Nanocages on Reduced Graphene Oxide Nanosheets for the Electrochemical Detection of Luteolin

Haixin Wang,Haopeng Zhang,Shuo Huang,Liqiu Zhang,Xin Gao,Jingming Lan,Long Li,Shuai Zhang,Wei Liu,Haoze Wang,Hongyan Yue

Mixed metal oxides derived from a metal–organic framework (MOF) sacrificial template take advantage of the original morphology of MOFs, excellent electrocatalytic properties, mesoporous structure, and more active sites and transport channels, which is favorable to construct a high-performance electrochemical sensor. Here, polyhedral CoZn-MOFs are synthesized by precipitation at room temperature, and then they are etched using Ni2+ to obtain hollow polyhedral NiCoZn hydroxide (NiCoZn-OH) nanocages. After self-assembly with graphene oxide (GO) nanosheets and annealing, the hollow polyhedral NiCoZn mixed metal oxide/reduced graphene oxide (NiCoZnOX/rGO) mixture with a size of ∼700 nm is obtained and used for electrochemically detecting luteolin (LU), which shows a high sensitivity (7.16 μA·μM–1·cm–2 in 0–60 μM) and a low limit of detection (64 nM, S/N = 3). Moreover, it has excellent selectivity in the presence of ascorbic acid and is successfully used to detect LU in diluted human urine, indicating a great potential in clinical applications.

Luteolin-Loaded WPI/Pueraria lobata Amylopectin Composite Gel Improves Adipocyte Thermogenesis and Insulin Sensitivity

Yaxin He,Xin Xie,Yanghui Chen,Wenchen Tao,Junjie Yu,Meng Huang,Yan Lin,Juan Chen,Jian Liu,Xian Zhang

Adipose tissue metabolic dysfunction plays a central role in the development of obesity and related metabolic disease. Luteolin is a natural flavonoid with antioxidant, anti-inflammatory, antidiabetic, and antiobesity activities, while it is limited by poor solubility, bioavailability, and stability. This study developed a whey protein isolate (WPI)–Pueraria lobata amylopectin (PLA) composite gel, characterized by a dense 3D network and enhanced mechanical strength. The WPI-PLA gel efficiently encapsulated luteolin in a stable amorphous form and exhibited uniform particle size and excellent colloidal stability. This encapsulation improved the light, storage, and thermal stabilities of luteolin; enhanced its antioxidant activity due to improved dispersion; and enabled its preferential intestinal release. Encapsulated luteolin promoted the expression of thermogenic genes UCP1, Pgc-1α, Prdm16, and Cidea and protein levels of UCP1 and catalase in brown-differentiated C3H10T1/2 cells and inhibited the expression of lipopolysaccharide-induced inflammatory cytokines Il-6, Tnf-α, and Il-1β in RAW264.7 macrophages. In differentiated white 3T3-L1 adipocytes, WPI-LU-PLA restored insulin sensitivity impaired by macrophage-conditioned medium. This reversal was indicated by the enhanced expression of the insulin sensitivity-related genes GLUT4 and Irs1, along with increases in GLUT4 and p-Akt protein levels. In conclusion, the WPI-PLA composite gel enhances the stability, bioaccessibility, and bioactivity of luteolin, suggesting potential for functional foods targeting metabolic dysfunction in adipose tissues.

Aptamer-functionalized luteolin-loaded albumin nanoplatform for targeted therapy of HER2-positive breast cancer

Caifen Yu,Zhenzhen Chen,Jin Liu,Xiaoyu Shi,Wenbin Xie,Lingjun Zeng,Minxin Zhang,Shenghang Zhang,Aiwen Huang,Shuiliang Wang

A novel anti-solvent ultrasonication method to fabricate luteolin-loaded BSA NPs. • Noncovalent hydrophobic interactions to fabricate aptamer-functionalized Lut-BSA NPs. • Multifunctional nanoplatform integrates natural compounds with nano and aptamer. • Superior tumor inhibition and biosafety highlight clinical translational potential. Luteolin (Lut) is a promising active component derived from traditional Chinese medicine for the treatment of human epidermal growth factor receptor 2 (HER2)-positive breast cancer. However, its clinical translation is substantially hindered by intrinsic physicochemical limitations: poor aqueous solubility and lack of tumor-selective biodistribution. So, we fabricated luteolin-loaded bovine serum albumin (BSA) nanoparticles (Lut-BSA NPs) via a modified anti-solvent ultrasonication method, in which BSA functions simultaneously as a biocompatible drug carrier and stabilizing agent. Then, HApt, the HER2-targeted aptamer, was conjugated to the surface of Lut-BSA NPs to generate the targeted nanoplatform HApt-Lut-BSA NPs, which act as a HER2 navigator to enable precise tumor targeting. HApt-Lut-BSA NPs demonstrated a high encapsulation efficiency of 87.64 ± 1.02% and a loading content of 3.87 ± 0.03%. In vitro release studies indicated that HApt-Lut-BSA NPs exhibited favorable release under acidic conditions, facilitating Lut accumulation in tumors. Cellular uptake assays and MTT analysis demonstrated that HApt-Lut-BSA NPs exhibited excellent targeting and cytotoxicity in vitro . In vivo fluorescence imaging confirmed superior accumulation of HApt-Lut-BSA NPs in SK-BR-3 tumor-bearing NPSG mice, while pharmacodynamic evaluation showed pronounced tumor growth inhibition and satisfactory in vivo safety. Collectively, this multifunctional nanoplatform that uniquely integrates a natural compound with nanotechnology and aptamer technology, not only markedly improves druggability and tumor-targeted delivery efficiency, but also offers a broadly applicable paradigm for the delivery of poorly water-soluble therapeutics and may promote the development of clinically translatable targeted drugs. Investigation on enhanced antimicrobial activity and bactericidal mechanism of photoactivated luteolin against Listeria monocytogenes

Yue Feng,Ying Yang,Yuzhang Zhu,Shuyao Zhao,Khalfan Khalfan Mohd,Cen Li,Jing Jin,Lincheng Zhang,Shuoqiu Tong,Yongjun Wu

This study investigated the antimicrobial mechanism of the natural flavonoid luteolin (LUT) as a novel photosensitizer in photodynamic inactivation (PDI) against Listeria monocytogenes (LM). Following 8-h UV-A activation, luteolin (LUT-UV, 0.8 mg/mL) exhibited markedly improved antimicrobial activity, resulting in 99.93 ± 0.03 % bacterial inactivation after 4-h co-incubation with LM suspension (1 × 10 8  CFU/mL), significantly higher the 87.77 % ± 0.89 % inactivation rate observed with LUT treatment alone. LUT-UV significantly increased reactive oxygen species (ROS) generation, enhanced bacterial membrane permeability, and aggravated intracellular content leakage. Scanning electron microscopy (SEM) observations confirmed that LUT-UV treatment caused significant damage to LM cells, including membrane shrinkage and rupture. Furthermore, RT-qPCR analysis revealed that LUT-UV significantly downregulated genes associated with hemolytic toxicity and biofilm formation in LM. The pepper contamination model experiment demonstrated that LUT-UV could effectively eliminate LM contamination. This study confirmed that LUT functions as both a photosensitizer and photoreceptor in PDI, demonstrated its potential applications in cold chain systems, and provided theoretical foundations for developing efficient and safe natural compound-based sanitizers.

Application of PVA hydrogel loaded with luteolin nanoparticles in anti EMT treatment after GBM

Long Zhou,Qingyu Zhao,Lijuan Gu,Renfu Tian,Yong Li,Xiaoxing Xiong

Background Glioblastoma multiforme (GBM) is the most common and worst-prognosed primary malignant tumor in the central nervous system. Epithelial mesenchymal transition (EMT) is an important cause of postoperative invasion and recurrence in GBM. Due to the presence of the blood-brain barrier, local therapy may be the preferred route for treatment of GBM after surgery. Methods Our objective is to develop an anti- EMT functional hydrogel for post-GBM surgery tamponade. We fabricated self-assembled luteolin nanoparticles (LU NPs) via the solvent evaporation method, and characterized their morphology and particle size using scanning electron microscopy (SEM), transmission electron microscopy (TEM), and dynamic light scattering (DLS). Furthermore, biological evaluations were conducted through CCK-8 assays, EdU staining, flow cytometry for cell cycle analysis, scratch assays, transwell assays and western blot. Ultimately, we constructed a polyvinyl alcohol (PVA) hydrogel loaded with luteolin nanoparticles and validated its therapeutic efficacy in vivo experiments. Results We successfully synthesized LU NPs, which significantly inhibited GBM cell proliferation, as well as GBM cell invasion and migration in vitro. Furthermore, LU NPs downregulated the EMT signaling pathway. We discovered that the observed anti-tumor effects of LU NPs were dependent on the function of β-catenin. Additionally, we successfully constructed a PVA hydrogel loaded with LU NPs (LU@gel). Finally, in the postoperative model of intracranial GBM xenograft in mice, LU@gel effectively suppressed GBM proliferation and EMT, significantly prolonging the survival time of the mice. Conclusions In summary, we have demonstrated that LU@gel exhibits potent anti-GBM effects, primarily attributed to the inhibition of β-catenin-mediated cell proliferation and EMT.

Multifunctional luteolin-Fe@PVA microspheres for combined hepatocellular carcinoma treatment: Embolization/sonodynamic/chemodynamic therapy

Zerong Wang,Ge Yan,Yi Ning,Xiaoli Zhou,Xin Lu,Qianqian Zhou,Qiwei Tian

Transarterial embolization (TAE) is a primary clinical treatment for hepatocellular carcinoma (HCC); however, its therapeutic efficacy is limited by its functional constraints. To enhance treatment outcomes, this study aims to develop a multifunctional embolic microsphere, LUT-Fe@PVA, for the combined treatment of HCC. This approach leverages a synergistic strategy that integrates TAE, sonodynamic therapy (SDT), and chemodynamic therapy (CDT) to improve the therapeutic efficacy. The LUT-Fe@PVA microspheres were synthesized via a microemulsion crosslinking method and loaded with luteolin-iron complexes to enable ultrasound-triggered reactive oxygen species (ROS) generation and Fenton reaction for enhanced tumor destruction. In vitro results demonstrated that the combination therapy significantly kills the tumor cells. In vivo, 18 F-FDG microPET/CT imaging revealed significantly reduced uptake values, while histological analysis confirmed extensive tumor cell necrosis and apoptosis in the combined therapy group. This study presents a promising multifunctional approach for HCC treatment by integrating embolization, SDT, and CDT, offering a novel strategy for effective and selective tumor treatment with minimal side effects.
