An electrochemical aptamer sensor based on nano‑silver-decorated covalent organic framework for the detection of malathion in medicinal herbs

Sai Zhang,Zeyang Liu,Qinqin Gao,Yaping Wang,Jiayi Zhou,Qinglin Sheng

Malathion, a broad-spectrum organophosphorus insecticide widely used in pest control for medicinal herbs, poses risks to human health and ecological balance due to its potential residual toxicity. In this study, we report a green method for the synthesis of silver-nanoparticles modified covalent organic frameworks (AgNPs@COFs) based on static in-situ reduction at room temperature. The material was combined with a malathion aptamer to develop a highly sensitive electrochemical aptasensor. The ferrocene-labeled hairpin aptamer was used as a molecular recognition element, and the sensing performance was evaluated using a differential pulse voltammetry (DPV) system. Under optimized conditions, the sensor exhibited a linear detection range of 10–800 ng/L, a low detection limit of 8.2 ng/L (S/N = 3, Δ I  = 0.01255–0.00774 C Mal , R 2  = 0.995), and excellent repeatability ( RSD  < 2.0 %, n  = 3). The proposed platform was successfully applied to quantify malathion in real herbal samples (e.g., panax ginseng , astragalus membranaceus, lycium barbarum , and lonicera japonica ), achieving recoveries of 98.32 %–101.92 % with corresponding RSDs of 1.25 %–3.28 % ( n  = 3). The sensor is expected to be applied to the rapid screening of malathion residues in medicinal herbs.

A simple colorimetric probe based on anti-aggregation of AuNPs for rapid and sensitive detection of malathion in environmental samples

Li Dongxian,Wang Shun,Wang Ling,Zhang Hao,Hu Jiandong

In this study, a simple colorimetric probe was developed for rapid and highly sensitive detection of malathion based on gold nanoparticles (AuNPs) anti-aggregation mechanism. A certain amount of NaOH can cause the aggregation of citrate-stabilized AuNPs due to the electrostatic interactions, and the color of AuNP solution changes from wine-red to gray. While in the presence of malathion, malathion is easily hydrolyzed in a strong alkali environment (pH > 9), followed by the production of a mass of negative charges, and thus the aggregated AuNPs turns to well-dispersed and the color of AuNP solution changes from gray to wine-red. This characteristic change can be visualized with the naked eye and quantitatively detected by an ultraviolet-visible (UV-Vis) spectrometer. Under optimized conditions, this probe exhibited a linear response to malathion in the concentration range of 0.05–0.8 μM with a limit of detection (LOD) down to 11.8 nM. The probe also showed good specificity for malathion detection in the presence of other interfering pesticide residues. Furthermore, the probe was successfully employed to detect malathion in environmental samples, with a recovery of 94–107% and a relative standard deviation (RSD) less than 8%. The results demonstrated that the proposed colorimetric probe based on anti-aggregation of AuNPs could be used for quantitative analysis of malathion and provided great potential for malathion determination in environmental samples.

An electrochemical aptasensor of malathion based on ferrocene/DNA-hybridized MOF, DNA coupling-gold nanoparticles and competitive DNA strand reaction

Guoli Xu,Danqun Huo,Jingzhou Hou,Chao Zhang,Yanan Zhao,Changjun Hou,Jing Bao,Xin Yao,Mei Yang

A novel electrochemical aptamer sensor platform for malathion detection was successfully constructed based on nanocomposite probe. The zirconium-based metal organic framework (MOF) was synthesized by hydrothermal method, and was coupled with complementary probe (CP) and carboxy-ferrocene (Fc-COOH) by covalent bonds to form CP-MOF-Fc nanocomposite probe. The complex probe hybridized with malathion aptamer on electrode by complementary base-pairing reactions. On account of the stronger affinity of the aptamer to target, CP-MOF-Fc was released into the buffer solution, causing a decrease in the DPV signal of Fc. Under optimized conditions, the prepared sensor showed a linear range of 25–850 ng L −1 toward malathion with a detection limit of 17.18 ng L −1 (S/N = 3) due to the high specific surface area of MOF , showing the excellent ability of signal amplification. In addition, it exhibited good repeatability and excellent selectivity. In the real sample experiment, satisfactory spike recovery results were obtained from 95.00 to 103.16%. The proposed strategy exhibited a rosy prospect in aptamer sensor design for pesticide testing.

A stable colorimetric biosensor for highly selective detection of malathion residue in food based on aptamer-regulated laccase-mimic activity

Linghui Huang,Yue Tang,Jianxun Han,Xiaojuan Niu,Xin Lin,Yuangen Wu

Malathion causes a serious threat to human health due to its widespread use in the environment. Herein, a novel and stable smartphone-integrated colorimetric biosensor for malathion detection is firstly established based on aptamer-enhanced laccase-mimicking activity. The results indicate that the M17-F aptamer can increase the affinity of Ag 2 O nanoparticles to the substrate 2,4-dichlorophenol and enhance their laccase-mimicking activity. Thus, abundant semiquinone radicals are produced in the catalytic system, which are combined with chromogenic agent to generate dark red products. The corresponding RGB values for the colour change of the solution can be easily obtained using smartphones, which is used for the rapid detection of malathion. The established biosensor for malathion has a limit of detection as low as 5.85 nmol·L −1 , and displays good selectivity for other competitive pesticides. Moreover, further studies have verified the applicability of the biosensor in actual samples, indicating that it may have the potential for application in malathion detection in food.

A novel spectral method for determination of trace malathion using EryB as light scattering probe by resonance Rayleigh scattering technique

Yunmei Huang,Jidong Yang,Jiawei Cheng,Yuhui Zhang,Haiyan Yuan

A convenient and sensitive spectrophotometric methods was proposed for determination of Malathion (Mala) using Erythrosin B (EryB) as a probe through the Resonance Rayleigh scattering (RRS) technique. The interaction between EryB, Pd 2+ and malathion in the system was investigated by fluorescence, RRS and UV–Vis spectroscopy. Under the optimum conditions, the RRS intensity of EryB, Pd 2+ and malathion was weak when exist in alone or any two kinds, however, the RRS intensity of the EryB-Pd 2+ -Mala system had an obvious enhancement due to Pd 2+ could interact with the hydrolysis products of Mala and EryB each other form a new complexes. At the same time, the fluorescence intensity of EryB was decreased significantly in the presence of Pd 2+ , and the fluorescence intensity of EryB-Pd 2+ system further decreased when Mala added, interestingly. So it was further proved that there was a forming complex in EryB-Pd 2+ -Mala system. Under the optimal conditions, the RRS enhanced intensity of the system was linearly proportional to the Mala's concentration in the range of 0.012–0.8 μg/mL, and the LOD was 1.7 ng/mL, with the correlation coefficient was R 2  = 0.9960. So, a new method for determination of Mala was established and this method has been demonstrated in real sample with satisfactory results.

Aptamer-based sensor for specific recognition of malathion in fruits and vegetables by surface-enhanced Raman spectroscopy and electrochemistry combination

Yufeng Zhang,Lixia Yang,Chang Sun,Chao Huang,Bixue Zhu,Qianjun Zhang,Dongmei Chen

ABSTRACT: Aptamers are single-stranded DNA or RNA that can specifically bind to a target substance with high affinity. Pesticides have obvious biological effects and are very harmful to the human body and the environment. Surface-enhanced Raman spectroscopy (SERS) can be combined with aptamer for the rapid detection of pesticide residues because of its simple operation and high sensitivity. However, a single analysis method has certain limitations and can't satisfy the research of complex system. In this study, SERS and electrochemical were used to prove the specific recognition of malathion by aptamer. The biochemical sensor directly detected the SERS spectra of the aptamer, malathion, and aptamer-malathion through the SERS method. Malathion could also be verified the specific recognition of aptamer by differential pulse voltammetry (DPV) and cyclic voltammetry (CV). Based on the peak difference before and after the combination of malathion and aptamer, a detection method for the specific recognition of malathion in a complex system was established. The identification mechanism was discussed. Then, the practical detection of malathion demonstrated the practicability of the method. The method was applied for the detection of malathion in spiked apple and bell pepper samples. The recoveries were in the range of 89.80%–106.50%, and the relative standard deviations were from 2.51%–6.92%. The use of aptamers for the specific detection of pesticides will have the potential for broad applications in the future.

“Off-On” non-enzymatic sensor for malathion detection based on fluorescence resonance energy transfer between β-cyclodextrin@Ag and fluorescent probe

Miao Wang,Kun Su,Jing Cao,Yongxin She,A.M. Abd El-Aty,Ahmet Hacımüftüoğlu,Jing Wang,Mengmeng Yan,Sihui Hong,Shuibing Lao,Yanli Wang

Here, we developed a novel non-enzymatic rapid testing method for determination of organophosphate pesticide (malathion) in water. In principle, target molecule can block the Fluorescence resonance energy transfer (FRET) between chemical fluorescent probe (energy donor) and β-cyclodextrin-coated silver nanoparticles (@AgNP) (receptor). The effects of malathion on the dynamics of fluorescent probe and β-cyclodextrin@AgNP were evaluated and their properties were further characterized. The current methodology showed a good sensitivity of 0.01 μg/mL represented as a limit of detection (LOD) and the calibration curve was linear over the concentration range of 0.1–25 μg/mL. Recovery rate from water samples spiked at 3 different concentration levels (0.3, 0.4, and 0.6 μg/mL) showed satisfactory range between 83% and 101%.

A Novel Aggregation-Induced Emission-Based Electrochemiluminescence Aptamer Sensor Utilizing Red-Emissive Sulfur Quantum Dots for Rapid and Sensitive Malathion Detection

Yajun Wu,Dongxiao Ma,Xiaoli Zhu,Fangquan Xia

Rapid, effective, and cost-effective methods for large-scale screening of pesticide residues in the environment and agricultural products are important for assessing potential environmental risks and safeguarding human health. Here, we constructed a novel aggregation-induced emission (AIE) electrochemical aptamer (Apt) sensor based on red-emissive sulfur quantum dots (SQDs), which aimed at the rapid screening and quantitative detection of malathion. SQDs were prepared using a two-step oxidation method with good electrochemiluminescence (ECL) optical properties. These SQDs were modified onto the electrode surface to serve as ECL luminophores. Subsequently, Apt was introduced and modified to form a double-helix structure with the complementary chain (cDNA). The ECL signal was reduced because the biomolecules had poor electrical conductivity and inefficient electron transfer. When the target malathion was added, the double helix structure was unraveled, the malathion Apt fell off the electrode surface, and the ECL signal was restored. The linear range of detection was 1.0 × 10 −13 –1.0 × 10 −8 mol·L −1 , and the detection limit was 0.219 fM. The successful preparation of the sensor not only develops the ECL optical properties of SQDs but also expands the application of SQDs in ECL sensing.

Portable bioactive capsule-integrated electrochemical-colorimetric dual-mode biosensor based on Mn/Ce MOF nanozyme for rapid detection of malathion in water

Li Sirui,Wang Shanshan,Chang Guorui,Yang Yaoxing,Feng Hui,Xue Haoqian,Zhang Li,Luo Manlin,Liu Jing,Li Bowen,Xu Shuxia

To enable rapid and reliable on-site monitoring of organophosphorus pesticides a novel portable dual-mode biosensor has been developed which integrates electrochemical and colorimetric detection into a single freeze-dried bioactive capsule. This innovative design supports instrument-free operation and provides dual-signal cross-validation. The sensor is based on a newly synthesized Mn/Ce bimetallic metal-organic framework (MOF) nanozyme with high oxidase-like activity, co-immobilized with acid phosphatase (ACP) within a mannitol- and sucrose-stabilized lyophilized capsule to ensure long-term stability and field readiness. In the presence of malathion, the pesticide selectively inhibits ACP, thereby restoring the MOF's catalytic activity and enabling the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB). The resulting signal is detected both visually and through chronoamperometry after conversion to a yellow diimine product. This approach achieves a wide linear detection range of 1–2000 nM and an exceptionally low detection limit of 0.06 nM for malathion, along with high accuracy evidenced by recoveries of 93.31%-104.03% in real water samples. This work establishes a field-deployable and user-friendly sensing platform that combines innovative material design with integrated dual-mode detection, offering a robust and practical strategy for on-site environmental monitoring. Dual-functional carbon nitride quantum dots as electron bridges and PMS activation sites on NH2-MIL-125(Ti) for malathion photocatalytic degradation

Xiaoxia Liang,Zewan Qin,Siwei Zhang,Rong Hai,Lingqing Gu,Xia Yang

30CNQDs@NMT/PMS achieved almost complete degradation of malathion within 30 min simulated sunlight irradiation. • CNQDs was integrated successfully to promote the charge separation efficiency as electron bridges and enhance the adsorption energy of PMS for activation revealed by DFT. • •OH and •SO 4 – were generated to attack the identified sites of malathion computed via Fukui function. • The intermediate products in the photocatalytic process were less toxic than malathion assessed by T.E.S.T. Malathion, an organophosphate pesticide widely used in agricultural fields across the world, has raised environmental concerns due to its persistence and toxicity. Thus, a novel photocatalyst was synthesized by integrating carbon nitrogen quantum dots (CNQDs) onto NH 2 -MIL-125(Ti) (NMT) via a solid-phase thermal polycondensation method. It was demonstrated that CNQDs served a dual function as electron bridges to facilitate charge separation and as peroxymonosulfate (PMS) adsorption activation sites substantially enhancing the photocatalytic activity, which was revealed by Density Functional Theory (DFT). Under simulated sunlight, 30CNQDs@NMT achieved nearly 100 % malathion removal within 30 min via PMS activation, exhibiting 8.1 times higher than NMT. While •OH and •SO 4 – acted as the dominant reactive species to attack the malathion sites. The toxicity of the intermediate products assessed by T.E.S.T. (Toxicity Estimation Software Tool) were much lower than malathion. Moreover, 30CNQDs@NMT performed effectively in real water matrices, demonstrating its potential for practical environmental applications. Development of a pH-responsive ratiometric fluoroprobe based on GSH-CuNCs-Ce3+ and N-CQDs for rapid/sensitive malathion detection in fruit juices

He Mei,Jingcheng Huang,Guixin Wang,Kun Chen,Huili Wang,Xuedong Wang,Wenwei Wang

Herein, we engineered a novel pH-responsive ratiometric assay by virtue of the composite system of Ce 3+ -enhanced glutathione-encapsulated copper nanoclusters (GSH-CuNCs-Ce 3+ ) and N-doped carbon quantum dots (N-CQDs). The GSH-CuNCs-Ce 3+ and N-CQDs were fabricated using one-pot chemical reduction and hydrothermal approaches, respectively, and their morphologies as well as physical and chemical properties were testified in detail by a series of characterization techniques. At 350 nm excitation wavelength, the GSH-CuNCs-Ce 3+ /N-CQDs system (GCC-NC) featured the 440/650 nm dual-emitting property. The ratios of fluorescence intensities (F 650 /F 440 ) demonstrated a strong pH-dependence from 3.0 to 5.0 with a coefficient of determination of 0.9982. Consequently, the GCC-NC system was feasible for constructing a pH-responsive dual-emitting fluoroprobe. Based on the catalytic effect of acetylcholinesterase on acetylcholine to yield acetic acid and further trigger varying solution pH, the as-constructed GCC-NC fluoroprobe was satisfactorily applied for detecting malathion . Under optimized conditions, the newly developed GCC-NC fluoroprobe supplied a wider linear range (0.25–200 μM), lower detection limit (0.075 μM), satisfactory recoveries (96.3–106.2%) and higher precision for malathion in several kinds of fruit juice samples. The fortified experiments by malathion’s structural analogues and metal ions provided compelling evidence that this fluoroprobe had strong anti-interference capacity and high specificity for malathion. These findings render us to believe that the as-constructed pH-responsive dual-emitting fluoroprobe holds great promise in trace malathion assay in food matrices.

Analyte-inhibited in-situ formation of Au nanoparticles suppressing their Tyndall effect for simple, ultrasensitive, and portable quantitative colorimetric detection of nanomolar malathion

Qi Wei,Fenglan Pan,Guanghua Su,Weifang Jiang,Jiarui Li,Xiaohui Xu,Xuehui Tang,Jinfang Nie,Wenying Jin,Yun Zhang

This work initially reports a simple, instrument-free, quantitative colorimetric assay of organophosphorus pesticides based on the analyte-inhibited in-situ formation of Au nanoparticles (AuNPs) suppressing their Tyndall effect (TE). Malathion was tested as a model analyte of organophosphorus pesticides. L-ascorbic acid-2-phosphate can be dephosphorylated by alkaline phosphatase to generate L-ascorbic acid, which can subsequently reduce chloroauric acid to produce colloidal AuNPs that display a pronounced TE response under irradiation with a laser pointer. In the presence of malathion, the analyte would effectively inhibit catalytic activity of the alkaline phosphatase, preventing the AuNPs’ formation to switch off their TE. The TE is inversely related to the malathion level and can be quantitatively measured by a smartphone. The results showed that this new method enabled a linear concentration range spanning four orders of magnitude (50 nM-15 µM) for malathion quantification. The limit of detection was estimated to be as low as 1.29 nM (3σ). When it was applied to detect real samples (i.e., pond water and tap water), recoveries of malathion ranged from 94.0 to 107.3%. The developed TE-based colorimetric approach offers many advantages including simplicity, low cost, and portable smartphone readout, thus holding great promise for on-site analysis and/or household self-testing of organophosphorus pesticides like malathion.
