Due to the low abundance of miRNAs and the coordinated regulation of multiple miRNAs toward some biological processes, a method that can sensitively and simultaneously detect multiple miRNAs is needed. Here, a strand displacement dual amplification (SDDA) strategy was developed for multiple miRNAs analyses using a unique single strand-double strand-single strand DNA (sdsDNA), which generated by the target recognition probe hybridizing with the site region probe. During the process of SDDA, miRNA hybridized to the target recognition domain of sdsDNA, thereby releasing the site, which triggered the first strand displacement reaction. This strand displacement event resulted in the release of the other site, triggering the second strand displacement reaction. Both two strand displacement reactions in each assembly circle were integrated for amplified signals, which were tested by a fluorescence resonance energy transfer (FRET) assay. NaYF 4 :Yb,Tm/Er UCNPs and Ti 3 C 2 nanosheets were used as the energy donor-acceptor pair. By employing 1:2 ratio of donor-acceptor, the energy transfer efficiency between NaYF 4 :Yb,Tm/Er UCNPs and Ti 3 C 2 nanosheets could reach 90%, which enhanced signal-to-background ratio. Combining the benefits of dual amplification and efficient energy transfer on miRNAs detection, this assay exhibited a linear range from 5 fM to 100 pM for miRNA-21 and miRNA-10b. Moreover, this assay was applied to estimate the expression of miRNA-21 and miRNA-10b in human hepatoma and hepatocyte cell lines. Therefore, this FRET-based SDDA strategy has been used for highly sensitive detection of multiple miRNAs in cell lysate and facilitates the development of efficient biosensor platform.

In this work, a simple and sensitive SERS-based miRNA biosensor was constructed based on porous MOFs nanoparticles and efficient subject-object recognition ability. MOFs as a container can package lots of signal probe neutral red (NR) for the advantages of three dimensional structure and porosity. The partially complementary duplex DNA can as a “lock” to lock up the hole for obtaining a weak Raman signal. In the present of miRNA, miRNA just like a “key” to open the duplex structure with the results of releasing NR. At this time, the released NR can be captured by SERS substrate [email protected] [7] for the subject-object recognition ability to produce a strong Raman signal which was positive correlation to target miRNA. By this way, the proposed SERS biosensor can achieve sensitively and selectively detect miRNA with a detection limit of 0.562 fM. This MOF-based SERS biosensor also be hopeful application for clinical diagnostics.

Heteromultivalent scaffolds with different repeated monomers have great potential in biomedicine, but convenient construction strategies for integrating various functional modules to achieve multiple biological functions are still lacking. Here, taking advantage of the heteromultivalent effect of dendritic nucleic acids and the specific biochemical properties of microRNAs (miRNAs), we assembled novel heteromultivalent nucleic acid scaffolds by biomimetic co-assembly of DNA–RNA building blocks. In our approach, two miRNAs were used to initiate and maintain dendritic structures in an interdependent manner; so, the heteromultivalent nanostructure can only form in the presence of both miRNAs. The proposed nanostructure can be used for one-step analysis of two miRNAs in an AND logic format. Taking miR-18b-5p and miR-342-3p which are associated with Alzheimer's disease as an example, a FRET sensing system was fabricated for the simultaneous analysis of two miRNAs within one hour at picomolar concentration. Further studies show that the designed device may have the potential to distinguish between AD patients and the healthy population by analysis of two miRNAs in CSF (cerebrospinal fluid) samples, suggesting its possible applicability in clinics.

Maintenance hemodialysis (MHD) is currently one of the treatments for uremia, but vascular calcification is the most common complication in MHD patients. Research has shown that peripheral blood microRNA-21 is abnormally expressed in uremic patients with vascular calcification. Therefore, it is necessary to detect miRNAs with sensitivity and specificity for early screening of uremia patients with vascular calcification complications. In this work, a light-activated DNA biosensor was constructed by using a catalytic hairpin assembly (CHA) signal amplification strategy and upconversion nanoparticles (UCNPs) for sensitive and specific determination of miRNA-21. A light-activated DNA hairpin (H1) was modified on the superficial layer of the UCNPs to trigger the analyte miRNA. In the presence of both NIR light and miRNA, H1 can hybridize with another DNA hairpin (H2) labeled with BHQ-2 and Cy5 at its two terminals, enabling fluorescence from “OFF” to “ON”. Furthermore, the released analyte miRNA triggers catalytic assembly, establishing an NIR-activated CHA amplification system. The constructed biosensor was used to detect miRNA-21, with a low detection limit (LOD) of 0.25 nM. Additionally, this biosensor can detect miRNA-21 in serum samples, with recoveries ranging from 96.7%–102.0%, suggesting that the current work provides an opportunity to develop NIR-activated signal amplification platforms for sensitive miRNA detection in biological samples.

Prostate cancer is one of the most common malignancies worldwide and a leading cause of cancer-related death in men. MicroRNAs (miRNAs) have emerged as promising and highly specific biomarkers for its detection. However, conventional miRNA-based biosensors face challenges of limited specificity, high false positives in single-target assays, and inadequate sensitivity. Here we developed an integrated pre-amplified DNA logic gate platform with cascaded signal amplification (IPL-CAFS-PCmi) for catalytic fluorogenic sensing of prostate cancer miRNAs. The system uses an AND-gate architecture, combining rolling circle amplification (RCA) for pre amplification, hybridization chain reaction (HCR) for signal enhancement and HCR-assembled multi-component nucleic acid enzymes (MNAzymes) for final cleavage-based amplification. The platform achieved detection limits of 2.21 fM for miR-141 and 2.66 fM for miR-375, with excellent specificity in discriminating single-base mismatches. In diluted human serum, recovery rates ranged from 98.51% to 99.16%, demonstrating strong robustness. By integrating RCA with HCR-mediated MNAzyme assembly in a programmable DNA logic framework, this design provides multi-level cascade amplification at room temperature, significantly improving both sensitivity and specificity. The platform thus represents a versatile tool with strong potential for precision oncology, clinical diagnostics, and point-of-care testing.

The hybridization and enzymolysis reactions for nucleic acid detection were carried out on the chip surface in the traditional surface plasmon resonance (SPR) biosensors. Herein, we proposed an innovative method for microRNA (miRNA) detection in which the hybridization-enzymolysis recycling reactions were performed in solution. Duplex-specific nuclease (DSN) and streptavidin-modified gold nanoparticles (SA-AuNPs) were employed for enhancing the assay sensitivity. In the absence of miRNA, the biotinylated DNA probe (bio-DNA-bio, biotin tags at both the 3′ and 5′ termini of DNA) was attached to the SA-modified chip through the SA-biotin binding, allowing the capture of SA-AuNPs with the same interaction. As a result, a larger SPR signal was attained. However, in the presence of miRNA, bio-DNA-bio hybridized with miRNA was digested by DSN. In this process, the miRNA strand remained intact and participated in the next hybridization-enzymolysis recycling process. Thus, one miRNA could promote the hydrolysis of many bio-DNA-bio probes and allow the generation of numerous bio-DNA fragments. Meanwhile, the produced bio-DNA competed with the undigested bio-DNA-bio to bind SA on the chip surface. The digestion of bio-DNA-bio and the competitive binding between bio-DNA-bio and bio-DNA led to the attachment of fewer SA-AuNPs and then smaller SPR signals. The change in SPR signal at the concentration as low as 1 fM miRNA has been readily determined. The strategy possessed the advantageous properties of simple operation, fast response, high sensitivity and excellent specificity, serving as a viable means for the fabrication of novel sensing platforms.

Tea is a widely consumed beverage globally, but the tea industry faces a significant waste management challenge. In this study, we developed tea leaf exosome-like nanoparticles (TELNs) with an average size of 274 ± 24.7 nm and a zeta potential of -20.6 ± 0.78 mV, using polyethylene glycol (PEG) 6000 precipitation followed by ultracentrifugation. Structural analysis confirmed that TELNs are composed of lipids, proteins, and RNAs. In vitro assays on HepG-2 cells revealed that TELNs are non-toxic at concentrations up to 300 µg/mL and can be efficiently internalized. TELNs exhibited significant antioxidant capacity and were able to significantly ameliorate H 2 O 2 -induced oxidative stress, increase the viability and reduce the accumulation of ROS in Hepg-2 cells. Notably, TELNs significantly alleviated OA-induced lipid metabolic disorders and hepatocellular injury. Further molecular analysis revealed that TELNs downregulated the expression of miR-21-5p, miR-17-3p, and miR-107, leading to the upregulation of their target genes PPARα, CYP7A1, and CPT-1A, which contributed to the improvement of lipid metabolism. This study is the first to demonstrate the lipid metabolism regulation potential of TELNs, providing new insights into their underlying mechanisms and helping to develop new therapeutic strategies for lipid metabolism-related diseases. Furthermore, it expands the scope of tea use and helps to reuse tea residues. Graphical

The integration of multi-level DNA logic gates for biological diagnosis is far from being fully realized. In particular, the simplification of logical analysis to implement advanced logic diagnoses is still a critical challenge for DNA computing and bioelectronics. Here, we developed a magnetic bead/DNA system to construct a library of logic gates, enabling the sensing of multiplex target miRNAs. In this assay, the miRNA-catalyzed hairpin assembly (CHA) was successfully applied to construct two/three-input concatenated logic circuits with excellent specificity extended to design a highly sensitive multiplex detection system. Significantly, the CHA-based multiplex detection system can distinguish individual target miRNAs (such as miR-21, miR-155, and miR let-7a) under a logic function control, which presents great applications in the development of rapid and intelligent detection. Another novel feature is that the multiplex detection system can be reset by heating the output system and the magnetic separation of the computing modules. Overall, the proposed logic diagnostics with high amplification efficiency is simple, fast, low-cost, and resettable, and holds great promise in the development of biocomputing, multiparameter sensing, and intelligent disease diagnostics.

Herein, the hydrolysis patterns of DNase I against a DNA probe in both single-strand form and DNA/miRNA heteroduplex were experimentally investigated in detail by mass spectrometric technique. Based on the knowledge obtained in the experiments, DNase I-assisted signal amplification strategy was proposed and used to design a mass spectrometric platform for miRNA detection. As a proof of concept, the performance of the proposed platform was evaluated for the detection of miRNA-122 in cell lysate samples. An advanced calibration model was explicitly derived to model the relationships between the concentration of miRNA-122 and the mass spectral signals of mass spectrometrically detected fragments of the DNA probe produced in the DNase I-assisted signal amplification process. With the aid of the advanced calibration model, the proposed mass spectrometric platform achieved quite accurate quantification results for miRNA-122 in cell lysate samples of Huh-7 and HepG-2 cell lines with recovery rates ranging from 98% to 104%. It is noteworthy that the proposed mass spectrometric platform can differentiate the target miRNA from other non-target ones based on their mass spectral response patterns instead of mass spectral intensities, and hence possesses high specificity for the target miRNA. It is reasonable to expect that the combination of mass spectrometry based on DNase I-assisted signal amplification with the advanced chemometric model provides a competitive alternative for designing cost-effective and label-free miRNA detection methods.

Abnormal miRNA expression accompanies prostate cancer (PCa) development, making the simultaneous detection of multiple miRNAs a promising strategy for accurate diagnosis. Herein, we developed a novel one-pot method for multiplex miRNAs detection by integrating quantum dot-labeled hybridization chain reaction (HCR) with polydopamine nanoparticles (PDA NPs) quenching. Target miRNA unfold the quantum dot-labeled hairpin probes to initiate HCR for signal amplification. Subsequently, PDA NPs effectively quench unreacted probes to reduce the nonspecific background signal, while the HCR products retain strong fluorescence. This strategy enables the simultaneous detection of multiple miRNAs under a single excitation wavelength, exhibiting picomolar sensitivity. It demonstrates high specificity, capable of discriminating both homologous family members and single-base mismatches of target miRNAs. Furthermore, the method exhibited reliable performance in the analysis of clinical serum samples. This work provides a robust and promising tool for non-invasive miRNA profiling, which could significantly enhance the precise diagnosis of prostate cancer.

Background Pancreatic cancer continues to be one of the world's most lethal cancers. Chemotherapy resistance in patients with advanced pancreatic cancer often accompany with dismal prognosis, highlighting the need to investigate mechanisms of drug resistance and develop therapies to overcome chemoresistance. Methods This research was filed with the Chinese Clinical Trial Registry (ChiCTR2200061320). In order to isolate primary normal fibroblasts (NFs) and cancer-associated fibroblasts (CAFs) samples of pancreatic ductal adenocarcinoma (PDAC) and paracancerous pancreatic tissue from individuals diagnosed with PDAC were obtained. The exosomes were obtained using ultracentrifugation, and their characteristics were determined by Western blotting, nanoparticle tracking analysis, and transmission electron microscopy. CAF-derived miRNAs were analyzed by RT-qPCR and high-throughput sequencing. Gemcitabine (GEM) was employed to promote ferroptosis, and ferroptosis levels were determined by monitoring lipid reactive oxygen species (ROS), cell survival, and intracellular Fe 2+ concentrations. To assess in vivo tumor response to GEM therapy, a xenograft tumor mouse model was utilized. Results Exosomes derived from CAFs in PDAC did not exhibit innate GEM resistance. CAFs promoted chemoresistance in PDAC cells following GEM treatment by secreting exosomes, and maintaining signaling communication with cancer cells. Mechanistically, miR-3173–5p derived from CAF exosomes sponged ACSL4 and inhibited ferroptosis after uptake by cancer cells. Conclusion This work demonstrates a novel mode of acquired chemoresistance in PDAC and identifies the miR-3173–5p/ACSL4 pathway as a promising treatment target for GEM-resistant pancreatic cancer.

Early diagnosis of acute diseases is restricted by the sensitivity and complex process of sample treatment. Here, an ultrasensitive, rapid, and portable electrochemiluminescence-microfluidic (ECL-M) system is described via sandwich-type immunoassay and surface plasmonic resonance (SPR) assay. Using a sandwich immunoreaction approach, the ECL-M system employs cardiac troponin-I antigen (cTnI) as a detection model with a Ru@SiO 2 NPs labeled antibody as the signal probe. For miR-499-5p detection, gold nanoparticles generate SPR effects to enhance Ru(bpy) 3 2+ ECL signals. The system based on alternating current (AC) electroosmotic flow achieves an LOD of 2 fg mL −1 for cTnI in 5 min and 10 aM for miRNAs in 10 min at room temperature. The point-of-care testing (POCT) device demonstrated 100% sensitivity and 98% specificity for cTnI detection in 123 clinical serum samples. For miR-499-5p, it exhibited 100% sensitivity and 97% specificity in 55 clinical serum samples. Continuous monitoring of these biomarkers in rats' saliva, urine, and interstitial fluid samples for 48 hours revealed observations rarely documented in biotic fluids. The ECL-M POCT device stands as a top-performing system for ECL analysis, offering immense potential for ultrasensitive, rapid, highly accurate, and facile detection and monitoring of acute diseases in POC settings.
