Alginate/pectin aerogel microspheres for controlled release of proanthocyanidins

Kailun Chen,Hui Zhang

Biopolymer aerogel microspheres based on alginate and pectin, were fabricated by crosslinking with divalent cation (Ca 2+ ) and sol-gel method followed by freeze-drying process. The morphology of the alginate/pectin aerogel microspheres examined by scanning electron microscopy indicated that the porosity and pore size increased with the increasing pectin ratio in the aerogels. The aerogels with higher alginate content presented higher stiffness, while the aerogels with higher pectin ratio exhibited the significantly ( p  < 0.05) higher water solubility and equilibrium moisture. Furthermore, proanthocyanidins were encapsulated in these aerogel microspheres and showed a good controlled release, as well described by first-order and Korsmeyer-Peppas models. The radical scavenging activity and ferric reducing antioxidant power results indicated that the encapsulated aerogel microspheres with higher pectin content had a stronger antioxidant activity .

Alkylated pectin: Molecular characterization, conformational change and gel property

Cheng-mei Liu,Xiao-juan Guo,Rui-hong Liang,Wei Liu,Jun Chen

The molecular characterization, conformational change and gel property of alkylated pectins (ALP) prepared by chemical covalent binding with different alkyl chains (C 6 , C 12 , or C 18 ) at different degrees of substitution (DS) (1.35, 2.90 or 4.53) were investigated. Molecular weight of ALP was affected by degradation, alkylation and aggregation. The conformation parameter α (obtained from R g vs M w ) and ρ (R g /R h ) both indicated that original pectin (HMP) and ALP with the lowest DS and the shortest alkyl chain had random coil conformation, while ALP with the greater DS and longer chain had sphere conformation in 0.1 M NaCl. In addition, we observed that the gel strength of ALP with higher DS and longer chain length was higher than HMP, and they were significantly positively correlated with the conformation parameter M w .

Stability of rutin using pectin-chitosan dual coating nanoliposomes

Alkassoumi Hassane Hamadou,Jiyao Zhang,Chen Chao,Bin Xu

This work evaluated the effect of pectin (P) and chitosan (CH) dual coating on rutin (R) loaded nanoliposomes (NLs). Besides R loaded NLs (RNLs), CH coated RNLs (CH-RNLs) and P coated CH-RNLs (P–CH-RNLs) were prepared. The results showed a size increase of NLs due to the polymer coatings, confirming a successful deposition of CH and P onto the surfaces of NLs. A surface charge reversal was observed after coating RNLs and CH-RNLs with CH and P, respectively. The coating resulted in a significant decrease in encapsulation efficiency (EE) from 88.19% in RNLs to 78.89% in CH-RNLs and 73.44% in P–CH-RNLs. Due to protection provided by the P–CH barrier, P–CH-RNLs displayed better thermal, pH, ionic and oxidative stability. NLs were more stable at 4 °C as compared to 25 °C over 8 weeks, and P–CH-RNLs exhibited the best performance overall. This study revealed P–CH coating as a potential route to produce stable NLs for the delivery of hydrophobic rutin.

Effect of different soluble pectin in breadfruit on starch digestibility

Lili Chen,Yuan Hao,Yihan Wang,Gang Wu,Yanjun Zhang,Xu Wang

Breadfruit is rich in starch and pectin. The pectin can alter the digestion process of starch, however, the specific mechanism by which various soluble pectin impact starch digestion remains unclear. The effects of water-soluble pectin (WSP), chelating agent soluble pectin (CSP), and sodium carbonate soluble pectin (SSP) on the digestibility of breadfruit starch were characterized by means of RVA, FTIR, DSC, ITC, SEM and in vitro digestion. The results showed that the three types of pectin could inhibit starch digestion. The addition of CSP increased the resistant starch content from 8.18% to 18.07%, and the addition of SSP increased the slow-digested starch content from 22.55% to 35.24%. The three pectin have different ways in inhibiting starch digestion. They have different principles in inhibiting starch digestion. WSP binds with the dissolved amylose to form a membrane that covers the surface of the starch particles. However, CSP and SSP penetrate into the starch granule during the gelatinization process, blocking the pores on the starch particle surface, thus reducing the starch digestibility.

Different Facets of Lignocellulosic Biomass Including Pectin and Its Perspectives

Shrestha Sarita,Kognou Aristide Laurel Mokale,Zhang Jin,Qin Wensheng

The plant matter, lignocellulosic biomass, is a renewable and inexpensive abundant natural resource in the world. The development of inexhaustible energy rehabilitated from agricultural waste is an alternative for fossil fuel to reduce CO 2 emission and prevent global warming. The amount of waste generated has a direct correlation with the human population. Thus, the waste generated by the community is being added to the environment as the municipal, agricultural waste, and waste produced from forest-based industries. Moreover, there are high possibilities of having environment-friendly valuable bio-based products, including biofuels, biogas, enzymes, and biochar from biomass without competing with the food supply chain. However, only a few or limited kinds of products are produced industrially. This review highlights the significance of lignocellulosic biomass. It describes the different valuable products like biochemicals, biochar, enzymes, single-cell protein, dye dispersant, and bioplastic from lignocellulosic biomass, emphasizing their applications briefly. Besides, this review also highlights the pretreatment of biomass, mainly focusing on biological pretreatment. Natural biomass utilization would lead to solving the energy shortage, food security issues, and obstacles for developing technological solutions in agriculture, agro-processing, and other related manufacturing sectors. Extraction and characterization of a pectin from sea buckthorn peel

Zhu Yulian,Liu Keshan,Yuen Michael,Yuen Tina,Yuen Hywel,Peng Qiang

Sea buckthorn peel is the by-product of the sea buckthorn processing, which contains many bioactive compounds. In this paper, sea buckthorn high methoxyl pectin (SBHMP) was obtained, with a yield of 8% and a light-colored. The SBHMP was a high methoxyl with a degree of esterification of 57.75% and uronic acid content of 65.35%. The structural and morphological characterization of SBHMP were analyzed by high-performance liquid chromatography, Fourier-transform infrared spectroscopy, and scanning electron microscopy. Results showed that SBHMP presented a sheet and layered stacked morphological, and was mainly composed of galacturonic acid, arabinose, galactose, rhamnose, and mannose, which indicated that SBHMP mainly consisted of homogalacturonan (HG) and rhamnogalacturonan-I (RG-I) type pectin polysaccharides. In addition, SBHMP also presented significant gel, thickening, and emulsifying properties. The results exhibited that SBHMP could form jelly-like gels under acid and high sucrose conditions, presenting a shear-thinning behavior and increasing apparent viscosity with the enhancement of pectin and sucrose contents. Besides, SBHMP could form oil-in-water emulsions with pectin concentrations of 1.0–3.0%. When the SBHMP concentrations were 2.0 and 3.0%, the emulsions were stable during 7 days of storage. Findings in this paper demonstrated the potential of SBHMP to be a food thickener and emulsifier and support the in-depth utilization of sea buckthorn by-products.

Structure and microbial-modulating evaluation of a sulfhydryl-modified pectin

Lijuan Hou,Chengxiao Yu,Laiming Zhang,Fuming Zhang,Robert J. Linhardt,Shiguo Chen,Xingqian Ye,Zhiqiang Hou

In this study, we synthesized a modified form of pectin know as sulfhydryl group (-SH) modified pectin (PSH) by linking cysteine's amino groups and pectin's carboxyl group through amide bonds. The -SH amounts varied from 517 μmol/g to 732 μmol/g. Pectin (PEC) modified with -SH groups showed no significant alteration in chain structure, monosaccharide composition and molecular weight. Compared to PEC, PSH had a stronger interaction with mucus, and the strength of this interaction increased with the -SH amounts. In vitro fermentation indicated that PSH was fermentable by human fecal microbiota, and the -SH modified can prolong the overall fermentation time of PEC from 48 h to 72 h, as evidenced by the variations in pH values, total sugar, molecular weight, monosaccharide consumption and short-chain fatty acids production. Furthermore, microbial analysis revealed that PSH significantly increased the abundance of Faecalibacterium and Bifidobacterium while decreasing the abundance of Klebsiella and Collinsella . These results provided a solid foundation for the potential use of -SH modified PEC as a mucoadhesive material for delivering active substances and regulating the effect of PEC on gut microbiota.

Structural modification of raisin pectin fractions by electron beam irradiation

Jiahui Li,Xuan Li,Youlin Xue,Jinfeng Bi

The effect of electron beam irradiation (EBI) on raisin pectin properties remains unclear regarding structural differences across pectin fractions. This study investigated the effects of EBI on the structural changes of three pectin fractions including WSP fraction, CSP fraction and NSP fraction in raisins as well as their correlations with polyphenol release and variations in antioxidant activity. EBI induced differential structural modifications of pectin fractions by disrupting hydrogen and glycosidic bonds, and altering molecular weight, crystallinity, monosaccharide composition and molecular conformation. EBI raised the molecular weight and HG content of WSP, reduced its RG-I ratio, Rg and Rc, leading to molecular aggregation and conformational contraction. Increasing irradiation doses induced de-esterification and molecular weight reduction in CSP, which exhibited maximum linearity and minimal branching at 4 kGy. NSP had the highest structural stability with intact HG and branched RG-I regions. The 4 kGy treatment maximized the release of (+)-catechin, (−)-Epigallocatechin, proanthocyanidins, protocatechuic acid, caffeic acid, quercetin, achieving the optimal antioxidant capacity. EBI degraded the RG-I and HG domains of WSP and the HG region of NSP, facilitating polyphenol release and enhancing the overall antioxidant activity of raisins. This work provides theoretical support for the processing of dried fruits via EBI technology.

Bubble-enhanced ultralight aerogels derived from pectin for oil absorption

Chen Zhibiao,Zhan Bin,Li Shuyi,Wei Dongsong,Zhou Wenting,Fang Zhengping,Liu Yan

With the escalating severity of oily wastewater discharge, the development of novel and efficient adsorption materials is an urgent necessity. Bio-based aerogels, due to their distinctive attributes, including low density, high porosity, sustainability, and safety, have emerged as ideal absorbents for oil spill remediation. In this study, aerogels with ultra-low density were prepared from the renewable biomass material, pectin, through an innovative ice/bubble dual-template method. In this process, with the help of sodium dodecyl sulfate, microbubbles were introduced by high-speed stirring to decrease the density of aerogels. At the same time, phytic acid also played a role in promoting pectin crosslinking and enhancing the flame-retardant properties of aerogels, significantly improving the safety of their transportation and application. After silane modification, the aerogel exhibited ultra-low density (10.91 mg/cm 3 ), exceptional superhydrophobicity (152°), outstanding oil absorption capacity (110 g/g), and flame retardancy. This work underscored the significant and promising prospects of bio-based aerogels in addressing oil pollution challenges, highlighting their potential as optimal candidates for sustainable and effective environmental remediation.

Injectable glycyrrhizinate-pectin hydrogel wound dressing based on natural ingredients

Chunan Lu,Qili Sun,Zimo Li,Yushan Wei,Jialin Yu,Shiman Li,Yansong Wang,Kai Li,Chuqing Tang,Huicheng Cao,Jingle Chen,Qianqian Liu,Xiajun Liang,Shujiang Zhang,Chao Xie,Bin Tang

Wound healing remains a significant clinical challenge, particularly for chronic and non-healing wounds, which impose substantial burdens on patients and healthcare systems. Despite advances in wound care, traditional dressings lack therapeutic properties and can impede the healing process due to their inability to conform to irregular wound geometries and lack of self-healing capabilities. This study addresses these limitations by introducing an innovative, injectable Glycyrrhizic acid ammonium salt/Pectin@Strontium (GAAS/Pec@Sr) hydrogel dressing. The hydrogel is derived from natural ingredients and possesses multifunctional therapeutic properties, including bioabsorbability, self-healing, and the ability to promote blood vessel and collagen regeneration while exhibiting anti-inflammatory and immunomodulatory functions. We developed the GAAS/Pec@Sr hydrogel through a combination of rheological experiments, gene and protein expression studies, and in vivo testing in a rat skin injury model. The GAAS/Pec@Sr hydrogel represents a promising biomaterial for wound healing, offering a novel solution derived entirely from natural products that can significantly contribute to the field of regenerative medicine.

Structural effect of polygalacturonase modified pectin on polyphenol enzymatic oxidation

Wenyue Wang,Xuan Li,Zhongxiang Fang,Jinfeng Bi

Pectin structure plays a vital role in determining the quality of food systems. This study investigated the impact of pectin structural variations on polyphenol oxidase (PPO)-catalyzed catechin oxidation using a series of pectin fractions prepared through polygalacturonase (PG) treatment. PG progressively degraded homogalacturonan (HG) regions, reduced molecular weight, and increased the exposure of hydroxyl and carboxyl groups, thereby converting the pectin matrix from a compact to a disordered state. Among these fractions, pectin enriched in rhamnogalacturonan I (RG-I) with an extended conformation, characterized by longer RG-I chains and fewer branches, exhibited weaker interactions with PPO and catechin, resulting in disordered aggregation and the lowest catalytic efficiency. Integration with molecular docking further revealed that high-HG pectin formed compact hydrophobic networks that restricted substrate access, resulting in high Vmax but low affinity. Conversely, low-HG pectin promoted hydrogen bonding with PPO and catechin, as well as facilitated ordered aggregation. Despite a lower Vmax, it exhibited a markedly reduced Km, resulting in enhanced overall catalytic efficiency. These findings demonstrate that pectin modulates PPO-catalyzed oxidation through steric hindrance, affinity interactions, and aggregation behavior, providing new insights for controlling enzymatic browning in food systems through structural modulation of pectin.

Study on 13C MultiCP/MAS ssNMR Analysis of Tobacco Pectin

Xin Ye,Zhenyu Xu,Da Xu,Jibao Cai,Shuiping Dai,Yuting Luo,Lei Guo,Ying Wang,Jiakun Su,Jun Yang

Background: As one of the most important economic crops, tobacco products have a long history and dominate the development of the world economy. Pectin, as a complex colloidal substance widely present in plant cell walls, its content is an important factor affecting the safety of tobacco smoking. Objective: This study aimed to analyze the content and structure of pectin in tobacco samples. Methods: In this study, tobacco pectin was extracted by ultrasonic-assisted ionic liquid extraction, and the 13C MultiCP/MAS NMR spectral analysis of pectin was conducted. Results: The type of extractant, duration of ultrasonication, extraction temperature, and solid-liquid ratio were optimized. Under the conditions of using 1-Butyl-3-methylimidazolium tetrafluoroborate ([Bmim]BF4) as the extractant, the solid-liquid ratio of 1:20 g/mL, and the ultrasonic power of 600 w for 30 min at 30°C, the yield of 23.7% of tobacco stem pectin and the purity of 54.2% could be obtained. The optimized MultiCP sequence parameters, with 10 CP cycles of 1.0 ms and the repolarization time of 50 ms could obtain high-resolution spectra within a time of 1.0 h. The C-6 peaks of the pectin in spectra were fitted using the spectral deconvolution technique and calculated the methylesterification (DM) of the tobacco pectin, which was generally less than 50% and belonged to the low methyl esterification pectin. The pectin content of the tobacco sample was calculated using the standard curve method with the addition of dimethyl sulfone (DMS) as an internal reference. The results of this method were consistent with the colorimetric method. Conclusion: The 13C MultiCP/MAS NMR method has the advantages of being green, fast, and accurate and provides a new technical tool for quantitative and qualitative studies of cell wall substances in tobacco samples.
