Diabetic cardiomyopathy increases the risk of heart failure independent of coronary artery disease and hypertension. Phloretin (PHL) shows anti-inflammatory effects in macrophages. In this study, we explored the protective effects of PHL on high glucose (HG)-induced injury in diabetic cardiomyopathy in vivo and in vitro . Using streptozotocin-induced diabetic mouse model and incubating cardiac cells line under a HG environment, PHL were evaluated of the activities of anti-inflammation and anti-fibrosis. In the study, PHL treatment ameliorated cardiomyocyte inflammation injury, and reduced fibrosis in vivo and in vitro . PHL also improved cardiac biochemical criterions after 8 weeks of induction of diabetes in C57BL/6 mice. Molecular docking results indicated that silent information regulator 2 homolog 1 (SIRT1) bound to PHL directly and that SIRT1 expression was upregulated in the PHL-treated group in HG-induced H9C2 cells. Protective effect of PHL was been eliminated in silence SIRT1 H9C2 cells. Taken together, these results suggested that PHL suppressed HG-induced cardiomyocyte injury via restoring SIRT1 expression.

This work aimed to study the effect of phloretin on antioxidant profiles through nuclear factor erythroid 2–related 2 (Nrf2)-DNA interactions in the liver of heat-stressed broilers by Cleavage Under Targets and Tagmentation combined with high-throughput sequencing (CUT&Tag-seq). One hundred and sixty 22-day-old Arbor Acres broilers were divided into four groups: a normal temperature group (NT: 22.8–23.4 °C and 46–55.8 % humidity) was fed a basal diet and three high temperature groups (China Summer condition: 29.7–31.7 °C and 77.9–94.8 % humidity) were fed the basal diet supplemented with 0 (HT group), 100 mg/kg (LT group) and 200 mg/kg (PT group) phloretin. Liver samples were taken from 42-day-old broilers. High temperature condition increased ( P ≤ 0.05) the malondialdehyde (MDA) level, but decreased ( P ≤ 0.05) the activities and mRNA expression levels of glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), catalase (CAT), and Nrf2. Dietary 200 mg/kg phloretin decreased ( P ≤ 0.05) the MDA level, but increased ( P ≤ 0.05) the activities and mRNA expression levels of GSH-Px, SOD, CAT and Nrf2 in the liver of heat-stressed broilers. CUT&Tag-seq showed that 4042 DEPs were up-regulated, while 2322 DEPs were down-regulated in the PT group compared with the HT group. The Nrf2-binding gene of DEPs contained SOD3, CAT, SOD2, and p38 mitogen-activated protein kinase. Most TOP20 significant KEGG pathways were associated with “Environmental Information Processing”. Summary, phloretin could improve oxidative damage by Nrf2 and its downstream DNA-sites including genes and Nrf2-bingding non-coding RNAs in the liver of heat-stressed broilers.

During thermal processing, myofibril-bound serine protease (MBSP) can degrade myofibrillar protein (MP), thereby affecting the aquatic product quality. Phloretin, a natural polyphenol, has been shown to delay the thermal degradation of MP. However, its specific mechanism of action remains unclear. To address this, MP (M group) was used as the control to investigate the effects of phloretin on the degradation degree, conformational changes, microstructure, and molecular interactions of MBSP-supplemented MP (M–M group) at 55 °C. SDS-PAGE analysis showed that phloretin could inhibit the degradation of myosin heavy chain induced by MBSP. With increasing phloretin concentration, the myofibrillar fragmentation index of the M–M group decreased more significantly than that of the M group. At 50 μM phloretin, the growth rate of active sulfhydryl in the M–M group (70.17%) was higher than that in the M group (54.91%), while total sulfhydryl in the M–M group (49.06%) was much lower than that in the M group (92.85%). Furthermore, the addition of phloretin caused fluorescence quenching and an initial increase followed by a decrease in α -helix content in the samples. And, the surface height and roughness values of M–M group decreased more significantly. Additionally, phloretin bound to MBSP mainly via hydrogen bonds and van der Waals forces, while binding to myosin primarily through hydrophobic interactions at 55 °C. Notably, phloretin exhibited a stronger binding affinity for MBSP. Overall, phloretin bound to the active site of MBSP, inhibiting its activity and thereby effectively delaying the thermal degradation of MP.

Phloretin exhibits strong antioxidant and anti-ageing properties by inhibiting mitochondrial oxidation of glutamate, succinic acid, and ascorbic acid. However, its clinical application is limited by poor aqueous solubility and low oral bioavailability. To enhance its bioavailability and efficacy, we incorporated phloretin into nano-micelles (phloretin-MM) using the thin film dispersion method. Characterisation revealed that the optimal formulation had TPGS and Pluronic F68 in a 4:1 ratio as the excipients, which resulted in spherical micelles with an average particle size of 33.28 nm and an encapsulation efficiency of 71.2 ± 0.48%. The in vitro release profile showed that the phloretin-MM showed significantly higher cumulative release rates than free phloretin across various pH conditions, while the pharmaceutical analysis in rats indicated that phloretin-MM significantly improved the oral bioavailability of phloretin (about 5 folds) in circulation. Additionally, through the analysis of the staining of zebrafish under light microscopy and the average gray value, it can be concluded that phloretin has anti-ageing drug effect, and phloretin-MM is better than free phloretin. These findings suggest that TPGS/Pluronic F68-modified phloretin-MM could serve as an excellent nano-drug carrier system, potentially enhancing the solubility, bioavailability, and anti-ageing effects of phloretin for broader clinical applications.

The increasingly severe energy shortage and growing environmental problems have accelerated the rapid development of sustainable polymer materials. Hereon, a novel method was proposed for the synthesis of cashew nut shell oil-based waterborne polyurethane (WPUs), which exhibited excellent toughness, transparency, water resistance, UV resistance and corrosion resistance. Specifically, the sorbitan monooleate-based siloxane (MSP) was prepared via a thiol-ene click chemistry reaction. Subsequently, the plant-based phloretin (PRT) and MSP were introduced as polyols into the skeleton of WPUs through the molecular structure design strategy. Series of high bio-based content (91 %) WPUs networks were obtained via pre-polymerization and self-emulsification methods. The properties of their dispersions and films were thoroughly investigated, and it was found that the addition of MSP and PRT enhanced the overall performance of WPUs film. For instance, there was a significant increase in T g . Additionally, they exhibited a maximum tensile strength of 31.5 MPa and a maximum toughness of 53 MJ/m 3 , which were the most outstanding among all reported plant oil-based WPU systems so far, indicating excellent tear resistance. Interestingly, the water absorption rate of PSWPU-40 was reduced to 7.1 %, suggesting good water resistance, which resolved the contradiction between the mechanical properties and water resistance of the current plant oil-based WPU systems. The PSPWU films exhibited promising application prospects, exemplified by its ability to effectively shield the entire UV range. Furthermore, its coating demonstrated outstanding anti-corrosion properties with a maximum anticorrosion efficiency of 97.1 %. It could be attributed to the synergistic effect of a self-crosslinking structure, higher crosslinking density, a tighter network structure, and stronger intermolecular forces (H-bond). This work provided a meaningful guide for the application and development of high-performance bio-based WPU functional coatings.
