Pullulan enhanced dual-network hydrogel electrolyte for high-performance flexible zinc-ion batteries

Runhai Wu,Shaopei Yang,Ran Wang,Yujia Guo,Pengcheng Du

Aqueous zinc-ion batteries (ZIBs) offer significant advantages, including low cost, high capacity, and safety. However, challenges such as zinc dendrites and uncontrollable side reactions hinder their widespread application. Hydrogel electrolytes can effectively inhibit the growth of negative dendrites and reduce side reactions associated with zinc electrode. Moreover, hydrogel electrolytes confer flexible and wearable characteristics to ZIBs, making them suitable for applications in flexible electronics and wearable devices. Herein, we develop a novel dual-network hydrogel electrolyte (PPZ) made by incorporating pullulan (PuL) into polyvinyl alcohol (PVA) hydrogel, enhancing its tensile strength and adhesion performance. The developed PPZ hydrogel electrolyte weakens the solvation of zinc ions, enabling stable and uniform plating/stripping behavior of the Zn metal anode during long-term cycling. The Zn//Zn symmetric cell with PPZ hydrogel electrolyte exhibits outstanding cycling stability (1800 h) at a current density of 0.5 mA cm −2 . Furthermore, the Zn//PPZ//PANI@TOC flexible ZIB maintains a capacity retention rate of 87.42 % after 6,000 cycles at 2 mA cm −2 and undergoes 3,000 bends of 180° with a high capacity retention rate of 87.53 %. This research provides a high-performance dual-network hydrogel electrolyte for designing multifunctional flexible energy storage and wearable electronic devices.

Preparation of multi-functional active packaging film of Galla chinensis waste CDs/pullulan

Baoshan Tang,Xi Wu,Lanxiang Liu,Juan Xu,Jinju Ma,Hong Zhang

In this study, multifunctional green carbon dots (CDs) have been synthesized using Galla chinensis waste (GCW) via hydrothermal method for the first time. An active packaging film has been developed in this work by combining CDs and pullulan (PL), using the solution-casting method. The microscopic morphology revealed that the CDs that were prepared using GCW exhibited good compatibility with PL. In addition, it also led to improvement in the toughness of the PL film (14.01 % to 20.26 %), along with its water vapor permeability value [1.31 to 0.53 (g·mm)/(kPa·h·m 2 )]. The composite films consisting of CDs exhibited good UV blocking rates for the UVA (90.41 %–7.87 %), UVB (87.76 %–0.08 %), and UVC (83.39 %–0 %) spectral ranges. The composite films exhibited strong antioxidant activity , and the clearance of ABTS and DPPH were obtained to be 93.61 % and 86.30 %, respectively. In addition, the composite films showed good antibacterial activity for E. coli and S. aureus , with a high antibacterial rate of up to 99.99 %. Finally, the non-contact preservation of strawberries over a duration of 10 d at room temperature confirmed that the prepared composite film can help preserve the quality of strawberries, as well as extended their shelf-life.

High-resolution 3D printing of modified pullulan elastic hydrogels via solvent-induced shrinkage

Zhaoxuan Feng,Shuangjia Cheng,Shuyu Chen,Jie Zhen,Wenqin Bai

3D-printable hydrogels confront two core bottlenecks: the trade-off among high printing resolution, fabrication efficiency, and structural fidelity, as well as the inherently poor mechanical performance of conventional hydrogel materials. Herein, we fabricated a dual-network photo-crosslinkable pullulan-based hydrogel system, in which the mass ratio of Pul-MA to uHMW Pul-NB and solid content jointly govern the printability and mechanical behaviors of hydrogel inks. A simple ethanol post-treatment induces relatively homogeneous volumetric shrinkage within printed architectures. Specifically, treatment with 75% ( v /v) ethanol shrinks the diameter of hydrogel micro-filaments down to 51.84 ± 0.4% of the original dimension, which greatly improves the feature resolution of printed structures. The ethanol-triggered shrinkage exhibits favourable partial reversibility, which holds promise for programmable dimensional regulation through further optimization of post-treatment cycles, accompanied by simultaneous mechanical reinforcement of hydrogel scaffolds. Complex architectures including bionic auricular models and co-axially printed tubular scaffolds were successfully fabricated to verify the practicability of this strategy. This solvent-induced shrinkage-assisted resolution-enhancing method serves as a hardware-free post-processing strategy for high-precision additive manufacturing of biomaterials, and holds practical value for research fields including tissue engineering, soft robotics and microfluidic devices.

Electrospun pullulan nanofibers containing pterostilbene-hydroxypropyl-β-cyclodextrin inclusion complex: Preparation and characterization

Yuanyuan Bao,Dan Yang,Hui Liu,Shujing Li,Hong Meng

Preservative-free dry masks made from natural active ingredients have attracted much attention in the spirit of safety and environmental protection, which should be in line with the future trend of skin care products. Pterostilbene (PTS) is a natural antioxidant ingredient derived from plants that has received great interest in the field of biomedicine and cosmetics. However, the low water solubility and stability of PTS limiting its wide application. In this study, we first self-assembled Pterostilbene (PTS) and hydroxypropyl-β-cyclodextrin (HP-β-CD) into water-soluble inclusion complexes (IC) to solve the solubility problem of Pterostilbene. The dry mask (PUL-PTS-HP-β-CD-NF) was then successfully produced using electrostatic spinning technology via combining IC with pullulan polysaccharide as co-spinning agents. The PUL-PTS-HP-β-CD-NF masks were detailedly analyzed using SEM, FTIR, and XRD, and the antioxidant capacity, water retention capacity, in vitro release, and safety were comprehensively assessed at the same time. The results showed that the nanofiber mask (PUL-PTS-HP-β-CD-NF) was dense, uniform, and well-distributed, which exhibited a higher level of antioxidant activity compared to isolated PTS counterparts. As the IC increases, the water retention capacity of the PUL-PTS-HP-β-CD-NF mask becomes more evident, and the in vitro dissolving studies further demonstrated that the release of PTS from PUL-PTS-HP-β-CD-NF was concentration-dependent, which exhibited a direct relationship with time and followed a kinetic model of first-order reaction. In addition, the biosafety of the nanofiber mask was confirmed using red blood cell hemolysis. This work not only provide a very novel approach to prepare dry mask but also offer a sustainable way to create environmentally-friendly chemicals.

pH-sensitive and pluronic-modified pullulan nanogels for greatly improved antitumor in vivo

Yan Zheng,XiaoDong Lv,Yong Xu,Xu Cheng,Xin Wang,Rupei Tang

It remains a crucial challenge to achieve efficient cellular uptake in tumor cells for nanoscale drug delivery systems. This work described that two multi-functional pullulan nanogels were prepared by co-polymerization between methacrylated pullulan (Pullulan-M) and different crosslink agents, an acid-labile ortho ester-modified pluronic (L61-MOE) or non-acid-sensitive methacrylated pluronic (L61-M). The prepared nanogels showed a regular spherical structure with the size about 200 nm measured by dynamic light scattering and transmission electron microscopy (TEM). Doxorubicin as a model drug was successfully encapsulated into nanogels. As expected, Pul-L61-MOE showed pH-dependent DOX release, and 25% of DOX was released at pH 7.4 while 84.48% of DOX was released at pH 5.0. In vitro cellular uptake and MTT results indicated that pH-sensitive nanogels (Pul-L61-MOE) displayed higher cellular internalization and cytotoxicity than acid-insensitive nanogels (Pul-L61-M) and free DOX. Flow cytometry assay suggested these nanogels remarkably increased intracellular reactive oxygen species (ROS) level and induced more cell apoptosis by the function of pluronic. Finally, in vivo antitumor results indicated that the multi-functional nanogels exhibit supreme antitumor efficiency, and the tumor growth inhibition (TGI) was 83.37%. Therefore, the pH-sensitive pullulan nanogels can be potential nano-carriers for drug delivery in tumor treatment.

Incorporation of Rutin in Electrospun Pullulan/PVA Nanofibers for Novel UV-Resistant Properties

Yongfang Qian,Mengjie Qi,Laijiu Zheng,Martin W. King,Lihua Lv,Fang Ye

This study aimed to investigate the incorporation of rutin into electrospun pullulan and poly(vinyl alcohol) (PVA) nanofibers to obtain ultraviolet (UV)-resistant properties. The effect of weight ratios between pullulan and PVA, and the addition of rutin on the nanofibers’ morphology and diameters were studied and characterized by scanning electron microscopy (SEM). Fourier transform infrared (FTIR) analysis was utilized to investigate the interaction between pullulan and PVA, as well as with rutin. The results showed that the inclusion of PVA results in the increase in the fiber’s diameter. The addition of rutin had no obvious effect on the fibers’ average diameters when the content of rutin was less than 7.41%. FTIR results indicated that a hydrogen bond formed between pullulan and PVA, also between these polymers and rutin. Moreover, the addition of rutin could enhance the mechanical properties due to its stiff structure and could decrease the transmittance of UVA and UVB to be fewer than 5%; meanwhile, the value of ultraviolet protection factor (UPF) reached more than 40 and 50 when the content of rutin was 4.46% and 5.67%, respectively. Therefore, the electrospun pullulan/PVA/rutin nanofibrous mats showed excellent UV resistance and have potential applications in anti-ultraviolet packaging and dressing materials. Keywords:pullulan;rutin;electrospinning;nanofiber;UV resistance

Fast dissolving oral films for drug delivery prepared from chitosan/pullulan electrospinning nanofibers

Ze-yu Qin,Xi-Wen Jia,Qian Liu,Bao-hua Kong,Hao Wang

In this study, Chitosan/pullulan composite nanofiber fast dissolving oral films (FDOFs) were prepared via electrospinning technology. The ratio of chitosan/pullulan (C/P) had an influence on solution property and nanofiber morphology, with the increase of chitosan, viscosity and conductivity of solutions increased, the morphology obtained by scanning electron microscopy indicated that the diameter of nanofibers decreased initially then increased. The Fourier transform infrared spectra indicated hydrogen bond interactions between chitosan and pullulan molecules. X-ray diffraction analysis proved that electrospinning process decreased the crystallinity of materials. Thermal analysis showed that melting point, degradation temperature and glass transition temperature increased with the addition of chitosan content in the FDOF. Water solubility test proved that the FDOF can dissolve in water completely within 60 s. Finally, in order to prove its practicability in future, a model drug of aspirin was encapsulated in the FDOF successfully.

Biodegradable gelatin/pullulan aerogel modified by a green strategy: Characterization and antimicrobial activity

Zhichao Yang,Chaoyi Shen,Jingshan Rao,Jiawen Li,Xiangzheng Yang,Hui Zhang,Jiangkuo Li,Olaniyi Amos Fawole,Di Wu,Kunsong Chen

The practical application of products prepared on the basis of gelatin (GA) or pullulan (PUL) is greatly limited by their solubility in aqueous solutions and relatively poor mechanical properties . Therefore, modification by chemical cross-linking agents is often needed. However, chemical residues and cytotoxicity limit its application in the food industry. The Maillard reaction , as a green cross-linking method, can be a suitable solution to this problem. In this work, biodegradable GA/PUL composite aerogels were fabricated by a freeze-drying method, and the Maillard reaction was used as a green modification strategy to improve the mechanical properties and solubility resistance of GA/PUL aerogels. The results showed that the Maillard reaction significantly enhanced the water resistance of the GA/PUL composite bioaerogel, resulting in an increase in the water contact angle from 57.45°− 86.42–98.93°− 116.05° at 60 s. In addition, the modified GA/PUL composite bioaerogel had a compression resistance of 1.76–3.01 MPa at a strain of 20 % and thermal insulation properties of 0.064–0.070 W/m∙K, and was completely biodegradable in 10 days. Moreover, the bioaerogel with nisin exhibited antibacterial activity against E. coli and S. aureus . In conclusion, this work effectively expands the application of gelatin-based bioaerogels in the food packaging industry.

Preservation of Pacific white shrimp (Penaeus vannamei) by pullulan and melleolide composite preservatives

Qingyong Zhu,Xin Zhang,Huimin Xu,Qin Qin,Xiaoe Chen,Qihe Chen,Zhengjie Liu

The qualities of Penaeus vannamei that are prone to spoilage have a significant impact on its economic profitability. To extend the shelf life of shrimp, this study formulated a novel composite preservative with pullulan and protoilludane sesquiterpene aryl esters (melleolides). Two melleolide compounds, namely armillarin and armillaripin, were identified using high-performance liquid chromatography-triple quadrupole time-of-flight mass spectrometry (HPLC–Q-TOF-MS/MS) analysis of the ethyl acetate extract of Floccularia luteovirens . The antibacterial activity of melleolides was investigated through the disc diffusion method, and it exhibited the most potent effect against methicillin-resistant Staphylococcus aureus (MRSA) among the tested bacteria, producing an inhibition zone diameter of 15.6 mm. The composite preservative displayed competitive inhibition of polyphenol oxidase, with an IC 50 value achieved at a melleolides content of 6.27%. Furthermore, the effects of the composite preservatives on shrimp quality were evaluated through sensory, colour, pH, TVB-N, APC, and TBARS assessments. The results indicated that S-PM8 treatment group inhibited the growth of the APC in shrimp from 8.98 log CFU/g (S–C) to 7.48 log CFU/g. The TVB-N content was maintained within 28.7 mg/100 g throughout the storage period. The content of TBARS was 0.97 mg MDA/kg at Day 12, which was significantly lower than that of the control group. Additionally, the composite preservatives substantially improved sensory scores and colour retention during storage. These findings demonstrate the potential of pullulan-melleolides composite preservative to extend the shelf life of P. vannamei .

Development of pullulan/gellan gum films loaded with astaxanthin nanoemulsion for enhanced strawberry preservation

Liuchun Yao,Qing Tao,Feng Xian,Zhipeng Chen,Liangliang Huang,Nanjing Zhong,Jing Gao

Innovative packaging materials are essential for preserving food, serving as a substitute for petroleum-based options. In this study, biofilms consisting of pullulan and gellan gum which incorporates astaxanthin nanoemulsion were prepared to extend the shelf-life of strawberries. Hydrophobic deep eutectic solvents (DES) were used as solvents to extract natural astaxanthin from Haematococcus pluvialis . Water-in-oil emulsions were prepared with the DES (containing Ast of 95 μg/mL), tween 20, and water (containing chitosan of 0.3 % and sodium tripolyphosphate of 0.1 %) at a mass ratio of 7:8:85. The average diameter of the astaxanthin nanoemulsion ranged from 167.70 to 351.03 nm, and showed exceptional stability with a retention rate of 95.37 % at 25 °C on the seventh day. Moreover, the nanoemulsion exhibited high antioxidant activity, with an IC50 value of 14.87 μg/mL and high antibacterial effect to E. coli and S. aureus . In the process of preparing the pullulan and gellan gum biofilms, this antioxidant effect and antibacterial effect are maximally preserved. Furthermore, following 20 days of storage with the biofilm, the strawberry experienced a weight loss of 12.48 %, a soluble solid content of 7.4 %, an ascorbic acid content of 0.75 %, and a titrable acidity of 36.45 mg/100 g. The shelf-life of the strawberry was significantly prolonged using the biofilms loaded with astaxanthin nanoemulsion to a minimum of 18 days, surpassing the 12-day shelf-life of those stored with neatfilm. Therefore, the astaxanthin nanoemulsion combined with pullulan and gellan gum biofilms offers an innovative alternative for active food packaging.

Pullulan nanofibrous films incorporated with W/O emulsions via microfluidic solution blow spinning technology

Liying Huang,Di Zhang,Nitong Bu,Yuanbo Zhong,Pingping Tan,Huanglong Lin,Jie Pang,Ruojun Mu

In this work, pullulan (PUL) nanofibrous films incorporated with water-in-oil emulsions (PE) were prepared by microfluidic blowing spinning (MBS). The microstructures of nanofibers were characterized by scanning electron microscopy (SEM), fourier transform infrared (FT-IR), and X-ray diffraction (XRD). With the addition of W/O emulsions, the thermal stability, mechanical, and water barrier properties of PUL nanofibers were improved. Increases in emulsion content significantly affected the antioxidant and antimicrobial properties of nanofibrous films. ABTS and DPPH free radical scavenging rates increased from 10.26 % and 8.57 % to 60.66 % and 57.54 %, respectively. The inhibition zone of PE nanofibers against E. coli and S. aureus increased from 11.00 to 20.00 and from 15.67 to 21.17 mm, respectively. In addition, we investigated the freshness effectiveness of PE nanofibrous films on fresh-cut apples. PE nanofibrous films significantly maintained the firmness, and reduced the weight loss and browning index of the fresh-cut apple, throughout the 4 days of storage. Thus, the PE nanofibrous films exhibited good potential to prolong the shelf life of fresh-cut fruit and promote the development of active food packaging.

A pullulan-based bilayer film for buccal delivery of a GLP-1 peptide analogue

Sandeep Karki,Sahil Malhotra,Muhammad Ijaz,Nicolas Rollet,Eoin D. O'Cearbhaill,Estela Bini,David J. Brayden

The buccal route has re-emerged as an alternative non-injectable route for peptide delivery. However, buccal peptide bioavailability is low, necessitating new formulation strategies. Pullulan, a water-soluble carbohydrate polymer, offers potential as a film-forming agent for buccal peptide delivery. Our aim was to co-administer a glucagon-like peptide-1 receptor agonist (GLP-1 RA) model analogue with an epithelial permeation enhancer, sodium glycodeoxycholate (GDC) in a bilayer film using a pullulan-based mucoadhesive layer. A pullulan–carboxymethyl cellulose mucoadhesive layer was selected, along with a backing layer made from an ammonio-methacrylate copolymer (Eudragit® RLPO). Physicochemical characterization confirmed the suitability of the pullulan-based film for buccal application. A Hilltop Chamber comprising an adhesive-backed cotton pad identified an optimal GLP-1 RA:GDC ratio for peptide permeation across ex vivo porcine buccal mucosae. 200 mM GDC (94.4 mg/mL) enabled ∼6% flux at 3 h using a donor concentration of 50 mg/mL GLP-1 RA (∼1:2). When these concentrations were incorporated into the prototype pullulan films, GLP-1 RA fluxes across mucosae reached ∼4.5% and ∼2% at 3 h for single mucoadhesive-layer and bilayer films, respectively, without overt tissue damage. Integrating GLP-1 RA and GDC into a pullulan–based bilayer film offers a promising carbohydrate polymer platform for enhancing buccal peptide delivery.
