BDE-209 exposure induced male reproductive toxicity with sperm quality decline. However, the role of autophagy in this was unclear. The purpose was to evaluate the protective effect and its potential mechanism of trehalose (Tre, autophagy inducer) on reproductive damage during spermiogenesis induced by BDE-209. We used 2% w/v Tre and 75 mg/kg/d BDE-209 cotreated mice for 42 days. GC-2 spd cells were cotreated with Tre, chloroquine (CQ, inhibition of autophagic flux), compound C (CC, AMPK inhibitor), and BDE-209. Tre intake significantly recovered decrease in sexual organ ratio and poor sperm quality in BDE-209-exposed mice. Supplementation with Tre rescued sperm head malformation by improving aberrant histone-protamine exchange in BDE-209-exposed mice. However, Tre intake couldn't restore the acrosome biogenesis. In addition, Tre supplementation improved testicular damage induced by BDE-209. BDE-209 blocked autophagic flux with increased P62 and LC3BⅡ/Ⅰ levels. Mechanistically, CQ treatment aggravated elevation of P62 and LC3BⅡ/Ⅰ levels induced by BDE-209, otherwise, CC and Tre treatments inhibited the rise in p-AMPK, p-ULK1, P62 and LC3BⅡ/Ⅰ levels induced by BDE-209. Tre supplementation improved reproductive injury in BDE-209-exposed mice by regulating autophagic flow via AMPK-ULK1 signaling pathways, which providing a new theoretical basis and possible therapeutic targets for male reproductive toxicity.

Background Trehalose has many advantages due to its inertness and the ability to stabilize biomolecules. Trehalose synthase can catalyze intramolecular rearrangement of the inexpensive maltose into trehalose in a single step, which represents a simple, fast, and low-cost method for the future industrial production of trehalose. Results In this work, an intelligent visualization method for producing trehalose via trehalose synthase was for the first time established and optimized by corresponding enzymatic hydrolysis with kudzu root starch as the initial raw material. For the first step, kudzu root starch was liquefied by α-amylase at a certain dextrose equivalent value of 19–21, and β-amylase and pullulanase to saccharify for a certain time, a four-factor nine-level experimental design with nine experiments was performed according to the uniform design table U
* 9
(9 4 ) to optimize the yield of maltose. Then, optimization of trehalose conversion ratio from kudzu root starch hydrolysate was carried out with a four-factor 12-level experimental design to forecast the optimal process conditions. By comparing verification tests and predicted results, the optimized operating conditions were pH 7.1, 22 °C, 32.5 % (14,000 U/ml) loading amounts of TreS enzyme and after 24 h, along with a 70.6 % trehalose conversion rate. Conclusions The intelligent visualization method has succeeded in exploiting the conversion process of trehalose from kudzu root starch hydrolysate, which contributes significant benefits to the further commercial production of trehalose.

A Gram-positive exoelectrogenRhodococcus pyridinivoranssp. strain HR-1 was cultivated from leachate-fed microbial fuel cell (MFC) to study the biodegradation effect of pyridine. In the comparison with mixed cultured MFC, HR-1 presented a remarkable electrical capacity with a maximum output of 4.33 W/m3under 30 °C in neutral anolyte with 1 g/L acetate as a substrate. Further, HR-1 demonstrated the environmental resistance as a Gram-positive strain. Microbial metabolism was evident at pH between 5–9 and temperature in the range of 20–40 °C, whereas optimal condition for pyridine degradation was observed at 30 °C. This is the first study reporting the degradation of pyridine in the bio-electrochemical system that achieved a 42% ± 5% degradation rate in a full operation cycle at 2 g/L of the concentration. Considering the nonnegligible internal resistance of HR-1-inoculated MFC, trehalose lipid was also introduced as a bio-surfactant to reduce the charge transfer obstacle between the microbes and the electrodes. The surface morphology illustrated that the strain had a plump shape with a high specific area. Accordingly, bio-surfactant addition promoted the anode biomass (1.2 ± 0.1 mg/cm2to 1.7 ± 0.2 mg/cm2) and achieved a higher degradation rate (68% ± 4%). The feasibility of electrochemical disposal on pyridine and eminent adaptability of strain sp. HR-1 as a Gram-positive exoelectrogen makes MFC a practical approach for real application.Keywords:microbial fuel cells;gram-positive exoelectrogen;pyridine;trehalose lipid;Rhodococcus pyridinivorans

The concentration of small molecules reflects the normality of physiological processes in the human body, making the development of simple and efficient detection equipment essential. In this work, inspired by a facile strategy in point-of-care detection, two devices were fabricated to detect small molecules via photocurrent measurement. A linear response of the photocurrent against the concentration of the small molecules was found. In the first device, metal ions were introduced into gel substrates made by xanthan gum to enhance photocurrent response. N-phenyl-2-naphthylamine was measured when iron or manganese ions were used. L-Phenylalanine was measured when the gel was modified by samarium, iron, cerium, or ytterbium ions. L-(+)-Arabinose was detected via the gels modified by iron or holmium ions. D-(+)-Mannose was detected when the gel was modified by ytterbium, manganese, chromium, or sodium ions. L-Methionine was detected in the gels modified by samarium, zinc, or chromium ions. The second device was based on a paper sheet. A sugar-like molecule was first synthesized, which was then used to modify the paper. The detection was possible since the photocurrent showed a linear trend against the concentration of D-Trehalose. A linear fit was conducted to derive the sensitivity, whose value was found to be 5542.4. This work offers a novel, simple, and environmentally sustainable platform that is potentially useful in remote areas lacking medical devices.

Conductive hydrogels based on natural products are promising for wearable sensors owing to their low cost and good biocompatibility, but typically suffer from poor mechanical strength, limited environmental stability, insufficient antibacterial activity, single functionality, and poor electrical conductivity. Here, we develop a conductive nanocomposite hydrogel (CAT 2 T 1.2 L 0.1 ) featuring trehalose as a natural cryoprotectant—a key design element of our system—along with tannic acid-coated tunicate cellulose nanocrystals (TA@TCNCs) as nano-reinforcements incorporated into a chitosan/acrylic acid network followed by LiCl immersion. The hydrogel integrates dynamic hydrogen bonds, lithium bonds, and Schiff base bonds, achieving high tensile strength (132.4 kPa) and ultrahigh elongation (3996.6%). Trehalose and LiCl synergistically suppress ice crystal formation and water evaporation, enabling stable flexibility and conductivity (4.81–5.0 S/m) from −30 °C to 60 °C. The hydrogel exhibits rapid self-healing (97.1% strain recovery within 12 h), broad-spectrum antibacterial activity (>94% kill rate), and excellent biocompatibility (cell viability 168.5%). As a wearable strain sensor, it enables wireless human motion monitoring and a deep learning-based handwriting recognition system with 93.4% accuracy, demonstrating great potential for extreme-environment flexible electronics.

This study investigated the effect of trehalose coating on the quality and shelf life of Atlantic salmon ( Salmo Salar ) treated by ultra-high pressure (UHP). Key parameters, including microbial safety, texture properties, protein oxidation, and volatile compounds, were analyzed. Results showed that UHP at 200-300 MPa achieved approximately 0.3–2.9 log reductions in microbial counts, while also inducing texture loss and protein oxidation. At 300 MPa, the carbonyl content exceeded 50%. Trehalose coating effectively mitigated texture degradation by restoring hardness to 98% of fresh samples and inhibiting oxidative damage in terms of 13% increased total sulfhydryl groups. Aroma analysis showed that UHP treatment promoted the accumulation of aldehydes derived from lipid oxidation and the beneficial flavor of hexanal and nonanal, while also inhibiting spoilage odor. Trehalose coating further inhibited 2-ethylhexanol and acetic acid, while retaining key flavor ketones of 3-hydroxy-2-butanone. A shelf-life prediction model integrating first-order kinetics and the Arrhenius equation achieved high accuracy (R 2 >0.97), indicating the extended refrigerated shelf life from 3 d to 25 d. This study provides a novel strategy by combining non-thermal treatment and bioprotectants for aquatic product preservation.

Drying preservation of mammalian cells presents a promising alternative to cryopreservation, with intracellular vitrification being the central challenge. Trehalose, a critical desiccation protectant, is essential for achieving this vitrification, but effective intracellular delivery and concentration control remain major hurdles. Current microfluidic methods for generating Giant Unilamellar Vesicles (GUVs, cell models) rely heavily on glycerol. This glycerol dependence fundamentally impedes the study and application of trehalose-mediated vitrification, as glycerol’s low glass transition temperature and difficulty of removal interfere with trehalose’s protective function. This study develops a glycerol-free microfluidic strategy that directly utilizes trehalose as the core component to enable stable generation of monodisperse water-in-octanol-in-water (W/O/W) double emulsions (GUV precursors) with precisely programmable intra- and extra-vesicular trehalose concentrations. Using a PDMS microfluidic chip with a six-way junction, the effects of replacing glycerol with varying trehalose concentrations (0–1.2 M) on double emulsion generation were investigated. Results demonstrate that trehalose effectively compensates for the destabilization caused by glycerol removal by increasing inner aqueous phase viscosity and reducing inner interfacial tension, significantly broadening the stable dripping regime. Droplet size exhibited non-monotonic variation with inner trehalose concentration, while increasing outer aqueous trehalose consistently reduced droplet size. This work is the first to demonstrate that trehalose can fully replace glycerol for generating double emulsions with spatially controlled trehalose loading. This glycerol-free approach overcomes the limitations of conventional methods, providing a vital experimental platform for investigating trehalose-mediated intracellular vitrification mechanisms and advancing trehalose-based anhydrous cell preservation technologies.

Effective moisturizing agents are crucial for improving the performance of rinse-off cosmetic formulations. Herein, we explored the synergistic effects of trehalose and N 2 -(2,3-dihydroxypropyl)arginine hydrochloride (DPAH), a glycerol derivative of L-arginine, on enhancing surface deposition and moisturizing performance. Using various surface-sensitive characterization techniques, we observed significant enhancements in the surface deposition of the trehalose/DPAH mixture compared to trehalose alone or glycerol-based controls. Molecular dynamics simulations further revealed strong intermolecular hydrogen bonds between trehalose and DPAH, contributing to the observed synergistic effects. A moisture-retention study using a model anionic surfactant-based system demonstrated that the trehalose/DPAH mixture achieves higher skin hydration levels and reduced transepidermal water loss compared to trehalose and DPAH alone. These findings offer new insights into the molecular interactions that enhance the surface deposition and moisturizing efficacy of trehalose, presenting promising opportunities for developing advanced cosmetic formulations with optimized moisture-retaining properties.

Heat stress (HS) compromises intestinal barrier integrity and microbiota homeostasis. Trehalose, a nonreducing disaccharide, is well known as a molecular chaperone to prevent protein misfolding under stress. However, whether trehalose supplementation can affect intestinal barrier integrity and microbiota under HS remains unknown. Male C57BL/6 mice (6 week old) were randomly divided into 3 groups (7 mice/group): a normal control group (CON, 23℃), a heat stress group (HS, 42℃) and heat stress + trehalose group (HT, received 2.0% trehalose in drinking water under HS condition). Trehalose supplementation decreased rectal temperature and body weight loss in heat-stressed mice. HS-induced intestinal permeability characterized by increased D-lactate was inhibited by upregulating protein expression of tight junction proteins including occludin and Zo1 in mice administrated by trehalose. Supplementary trehalose significantly ameliorated HS-induced oxidative stress by elevating activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) as well as protein expression of catalase (CAT). Trehalose intervention reversed the up-regulation of endoplasmic reticulum (ER) stress proteins glucose-regulated protein 78 (GRP78), growth arrest and DNA damage-inducible protein 34 (GADD34), phosphorylated eukaryotic initiation factor 2α (p-eif2α), activating transcription factor 4 (ATF4), and C/EBP homologous protein (CHOP) as well as proapoptotic proteins BAX, Cytc, and active-caspase3 in heat-stressed mice. Trehalose supplementation evidently elevated the abundance of beneficial bacteria Lachnospiraceae_NK4A136_group while reducing that of harmful bacteria Bacteroidetes in heat-stressed mice. Taken together, these results revealed that supplementary trehalose could attenuate HS-induced intestinal barrier dysfunction via reducing oxidative stress, reversing ER stress induced apoptosis and gut microbial dysbiosis, suggesting trehalose as a promising dietary additive to counteract the intestinal injury induced by HS in mice.

The risk of autism spectrum disorder (ASD) is elevated by exposure to the organophosphate pesticide chlorpyrifos (CPF) during pregnancy and lactation. As a neurodevelopmental disorder, ASD often co-occurs with depression, exacerbating the social isolation experienced by affected individuals. Trehalose (Tre), a naturally occurring non-reducing sugar, has neuroprotective properties. Nevertheless, the mechanisms by which it exerts protective effects against CPF-induced ASD and depression-like behaviors remain unclear. The objective of study was to investigate the mechanisms underlying Tre combating CPF-induced ASD and depression through network pharmacology and toxicology analyses. In animal studies, Tre was administered to offspring mice exposed to CPF during pregnancy and lactation. Behavioral assessments revealed that Tre alleviated CPF-induced repetitive and stereotypic behaviors, social deficits, and depression-like behaviors in offspring mice. Findings from network pharmacology and toxicology analyses and animal experiments supported the notion that mammalian target of rapamycin-related autophagy and TLR4-related necroptosis signaling may contribute to the protective effects of Tre against CPF-induced hippocampal damage. This research offers valuable information regarding the potential protective effects of Tre against neurodevelopmental disorders linked to environmental chemicals.

The inhibitory effect of water-tailored natural deep eutectic solvents (NADES) constructed from citric acid and trehalose with different amounts on the quality deterioration and oxidation of frozen-thawed (F-T) mirror carp ( Cyprinus carpio L. ) surimi was studied. NADES was obtained by citric acid to trehalose and the effect of moisture addition (v/v) on the structure, physicochemical, and anti-freezing capacity of NADES was assessed. NADES + 10 % H 2 O has relatively low viscosity (25 %) and strong freezing resistance. However, a 50 % H 2 O addition leads to the disappearance of the hydrogen bond. The addition of NADES effectively inhibits water loss, migration, and mechanical damage on F-T surimi. An inhibitory effect of 4 % (w/w) NADES on oxidation was verified by a decrease in carbonyl contents (17.4 %, 8.63 %) and TBARS (37.9 %, 15.2 %) of surimi compared with control ( P < 0.05) and sucrose + sorbitol after 5F-T cycles, suggesting the potential of NADES as a cryoprotectant for the food industry.

Reducing sodium content compromises the texture and flavor of meat products. This study investigated the combined effects of ultrasonic treatment (UT), yeast extract (YE), and trehalose (TH) on myofibrillar protein (MP) gelation and molecular structure under the low-salt (1.5% NaCl) condition. Compared to low-salt control, the UT-YE-TH treatment markedly enhanced the gel functionality ( p < 0.05). Water-holding capacity (WHC) improved from 69.76% (R) to 85.55% (R-UT-YETH), and gel strength increased from 94.75 to 173.54 g ( p < 0.05). Microstructural analysis showed that the combined treatment generated a more continuous and homogeneous protein network. Circular dichroism revealed a decrease in α-helix content (from 13.6% to 2.0%) and an increase in β-turn (from 21.2% to 27.3%). Intrinsic fluorescence and surface hydrophobicity measurements further confirmed enhanced unfolding and intermolecular interactions, accompanied by elevated total and free sulfhydryl contents. Correlation analysis and principal component analysis (PCA) indicated that increased β-turn fractions and greater hydrophobic exposure were associated with improvements in WHC and gel strength. The combined application of UT, YE, and TH effectively improves MP gelation under the low-salt condition by promoting favorable structural transitions, which provides a practical strategy for producing reduced-sodium cured meat products with enhanced quality.
