Photocatalytic activity and mechanism of PCN@GO/AgNPs heterojunction in Rhodamine B degradation

Aijing Li,Xinpeng Chen,Yurong Sun,Jin Cheng,Ping Yao,Tieling Xing,Guoqiang Chen

An essential factor affecting the photocatalytic efficiency of a catalyst is the efficient separation of photogenerated electrons and holes. In the present study, a novel heterojunction material, PCN@GO/AgNPs, was synthesized with the aim of improving the photocatalytic performance. This achievement was accomplished through the establishment of a heterojunction between graphitic phase carbon nitride (PCN) and nano silver (AgNO 3 ). The PCN was prepared from melamine powder calcined at high temperature under nitrogen environment, and was modified by graphene oxide (GO) to form heterojunction with silver nanoparticles, which resulted in a significant improvement in the photocatalytic efficiency of the catalyst. Under ultraviolet(UV)excitation, the catalytic rate constant of PCN@GO/AgNPs heterojunction on RhB dye can reach 10.594 × 10 −2 min −1 . This research represents a promising strategy to increase the surface area of PCN photocatalysts through simply doping noble metal and thus improving photocatalytic performance, which contributes to understand the reaction mechanism of photocatalytic effect.

Improved alkaline fusion method for B isotope and concentration measurements of silicate materials

Yue Cai,Ting Ruan,Ye Li,Baichan Li,Weiwei Zhang,Zhenghui Li,Haizhen Wei,E. Troy Rasbury

Boron (B) is one of the most powerful tracers of fluid-mediated processes. However, challenges still exist for B analysis of silicate materials with the main difficulty in sample dissolution. Cai et al. (2021) reported a novel alkaline fusion method for boron isotope analysis using solid sodium peroxide and glassy carbon crucibles. Here we report a modified and improved version of this method using hydrated sodium peroxide and silver crucibles, which permits a lower flux-to-sample mass ratio (FSMR) (3 : 1), relatively low fusion temperature of 710 °C, and the use of cheaper silver crucibles. This modified method achieves complete sample dissolution while being safer and more efficient as it removes the need for grinding fresh sodium peroxide solid into powder. The B blank of the hydrated sodium peroxide solution can be determined more accurately and further reduced through chromatography. In addition to the improvements in the alkaline fusion procedure, we also propose a new procedure to dissolve the fusion cake, which greatly reduces reagent usage and minimizes the procedural blank as a result. Using these improved techniques, we analyzed B concentrations and B isotope compositions of eight silicate international standards. The results are consistent with published data within analytical uncertainties. Combined with the one-step chromatography using the Amberlite® IRA-743 B-specific resin to purify B and the Neptune plus® MC-ICP-MS for isotopic analysis, this method generates a batch of B concentration and isotope data within 48 hours with minimal need to handle hazardous chemicals. This safe and efficient method will greatly speed up research using B isotopes on silicate materials.

Epimedin B exhibits pigmentation by increasing tyrosinase family proteins expression, activity, and stability

Chen Hong,Yifan Zhang,Lili Yang,Haoyang Xu,Kang Cheng,Zhi Lv,Kaixian Chen,Yiming Li,Huali Wu

Epimedin B (EB) is one of the main flavonoid ingredients present in Epimedium brevicornum Maxim., a traditional herb widely used in China. Our previous study showed that EB was a stronger inducer of melanogenesis and an activator of tyrosinase (TYR). However, the role of EB in melanogenesis and the mechanism underlying the regulation remain unclear. Herein, as an extension to our previous investigation, we provide comprehensive evidence of EB-induced pigmentation in vivo and in vitro and elucidate the melanogenesis mechanism by assessing its effects on the TYR family of proteins (TYRs) in terms of expression, activity, and stability. The results showed that EB increased TYRs expression through microphthalmia-associated transcription factor-mediated p-Akt (referred to as protein kinase B [PKB])/glycogen synthase kinase 3β (GSK3β)/β-catenin, p-p70 S6 kinase cascades, and protein 38 (p38)/MAP kinase (MAPK) and extracellular regulated protein kinases (ERK)/MAPK pathways, after which EB increased the number of melanosomes and promoted their maturation for melanogenesis in melanoma cells and human primary melanocytes/skin tissues. Furthermore, EB exerted repigmentation by stimulating TYR activity in hydroquinone- and N -phenylthiourea-induced TYR inhibitive models, including melanoma cells, zebrafish, and mice. Finally, EB ameliorated monobenzone-induced depigmentation in vitro and in vivo through the enhancement of TYRs stability by inhibiting TYR misfolding, TYR-related protein 1 formation, and retention in the endoplasmic reticulum and then by downregulating the ubiquitination and proteolysis processes. These data conclude that EB can target TYRs and alter their expression, activity, and stability, thus stimulating their pigmentation function, which might provide a novel rational strategy for hypopigmentation treatment in the pharmaceutical and cosmetic industries.

B-Doping-Induced Lattice Expansion of Pd Metallene Nanoribbons for Oxygen Reduction Reaction

Hongjing Wang,Yunju Li,Songliang Liu,Hongjie Yu,Kai Deng,Ziqiang Wang,You Xu,Liang Wang

Pd-based metallene is regarded as an efficient catalyst in the field of oxygen reduction reaction (ORR) because of its fantastic physicochemical features. The morphological structure control, lattice strain engineering, and electronic structure modulation of Pd-based metallene are effective tactics to enhance its electrocatalytic performance. In this work, we fabricate atomically thin B-doped Pd metallene nanoribbons (B-Pd MNRs) for efficient alkaline ORR. The atomically thin nanoribbon structure of B-Pd MNRs can expose many surface atoms as catalytically active sites. Moreover, the incorporation of boron effectively induces the lattice expansion and modulates the electronic structure of Pd, which can synergistically weaken the adsorption of intermediate species on B-Pd MNRs. Therefore, the B-Pd MNRs display excellent activity and durability for ORR. This work opens an avenue to the synthesis of atomically thin heteroatom-doped metallene nanoribbons for energy electrocatalytic applications.

Platelet Membrane-Encapsulated Ginkgolide B Biomimetic Nanoparticles for the Treatment of Ischemic Stroke

Jing-Wen Cui,Hui-Cong Feng,Chang Xu,Ding-Yuan Jiang,Kai-Hua Zhang,Na−Na Gao,Yuan Wang,He Tian,Chang Liu

Stroke is a highly lethal and disabling disease of the central nervous system exacerbated by oxidative stress, inflammation, and ferroptosis. However, the blood–brain barrier makes it extremely difficult for most medications to enter the brain, necessitating extremely high drug concentrations. Thus, we developed platelet-derived nanoparticles to deliver ginkgolide B (GB), a compound that has anti-inflammatory, antioxidative stress, and ferroptosis inhibition properties and assists acute stroke recovery. GB was encapsulated in a platelet membrane (PM) with a particle size of approximately 68.32 ± 3.7 nm, and the encapsulation efficiency reached 79.43 ± 6.3%. Experiments showed that PM-derived nanoparticles with GB (PM-GB) increased the activity of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) and decreased malondialdehyde(MDA) and reactive oxygen species(ROS) levels in the brain. Additionally, the PM-GB group had higher ferroptosis suppressor protein (GPX4, FSP1) expression, approximately 4-folds, with lower levels of the pro-inflammatory cytokines IL-6, IL-1β, and TNF-α, and ferroptosis marker protein (PTGS2) compared with the OGD group. In conclusion, PM-GB reduces ferroptosis and inflammation by inhibiting oxidative stress, which protects neural cells and promotes motor recovery in middle cerebral artery occlusion model rats (MCAO/R).

Efficient Degradation of Rhodamine B Dye through Hand Warmer Heterogeneous Activation of Persulfate

Tiantian Ye,Lihong Liu,Yilin Wang,Jianqiang Zhang,Zhenxing Wang,Cong Li,Haoyu Luo

In this study, an innovative method for RhB (Rhodamine B) degradation in a persulfate (PS) and hand warmer heterogeneous activation system was investigated. The hand warmer showed better catalytic performance and excellent reusability in terms of PS activation. The reaction rate constants of RhB removal in the hand warmer/PS process (0.354 min−1) were much faster than those in other PS- and Fe-related processes (0.010–0.233 min−1) at pH 7. The iron in the hand warmer is the main active ingredient to catalyze PS, and activated carbon, salt, and H+/OH−accelerate the activated reaction due to the formation of micro-batteries in the solution. Moreover, the catalyst of the hand warmer showed excellent stability and reusability with a low level of iron leaching. This new, effective, inexpensive, repeatable, and environmentally friendly catalyst combined with PS has promising prospects for the removal of dyes from industrial wastewater. Keywords:heterogeneous activation;persulfate;hand warmer

Synthesis and characterization of aromatic copolyesters derived from bisphenol B and phenol red

Zhoufeng Wang,Junwei Hu,Bolin Wang,Yingying Liu,Xiubo Long

A series of novel aromatic copolyesters derived from bisphenol B (BPB) and phenol red (PR) were synthesized by interfacial polymerization. The number- and weight-average molecular weight ( M n and M w ) of the obtained amorphous aromatic copolyesters ranged from 1.06–1.80 × 10 4 to 2.01–6.17 × 10 4 g/mol, respectively. Density functional theory calculations showed that the reactivity of BPB was similar to that of bisphenol A. The reaction activation energy of BPB with terephthaloyl and isophthaloyl chloride (TPC and IPC) was 8.23 kcal/mol, and PR with IPC and TPC was 59.30 kcal/mol. Under the influence of the ethyl side group of BPB and the pendant group of PR, the solubility of the aromatic copolymer was improved, and the glass transition temperature ( T g ) was 125.1–195.3°C, while T max could still be maintained above 525.72°C. The optical transmittance of the copolyesters at 450 nm ( T 450 ) was 69.29%–99.41%, and the tensile strength and Young's modulus were in the range of 46.79–58.59 MPa and 1.39–1.86 GPa respectively. All aromatic copolyesters exhibit good mechanical and processability properties and have great application potential as high-performance plastics.

Alisol B regulates AMPK/mTOR/SREBPs via directly targeting VDAC1 to alleviate hyperlipidemia

Gai Gao,Jie Zhao,Jing Ding,Shuyan Liu,Yanyan Shen,Changxin Liu,Huifen Ma,Yu Fu,Jiangyan Xu,Yiran Sun,Xiaowei Zhang,Zhenqiang Zhang,Zhishen Xie

Background The occurrence of hyperlipidemia is significantly influenced by lipid synthesis, which is regulated by sterol regulatory element binding proteins (SREBPs), thus the development of drugs that inhibit lipid synthesis has become a popular treatment strategy for hyperlipidemia. Alisol B (ALB), a triterpenoid compound extracted from Alisma , has been reported to ameliorate NASH and slow obesity. However, the effect of ALB on hyperlipidemia and mechanism are unclear. Purpose To examine the therapeutic impact of ALB on hyperlipidemia whether it inhibits SREBPs to reduce lipid synthesis. Study Design HepG2, HL7702 cells, and C57BL/6J were used to explore the effect of ALB on hyperlipidemia and the molecular mechanism in vivo and in vitro . Methods Hyperlipidemia models were established using western diet (WD)-fed mice in vivo and oleic acid (OA)-induced hepatocytes in vitro . Western blot, real-time PCR and other biological methods verified that ALB regulated AMPK/mTOR/SREBPs to inhibit lipid synthesis. Cellular thermal shift assay (CETSA), molecular dynamics (MD), and ultrafiltration-LC/MS analysis were used to evaluate the binding of ALB to VDAC1. Results ALB decreased TC, TG, LDL-c, and increased HDL-c in blood, thereby ameliorating liver damage. Gene set enrichment analysis (GSEA) indicated that ALB inhibited the biosynthesis of cholesterol and fatty acids. Consistently, ALB inhibited the protein expression of n-SREBPs and downstream genes. Mechanistically, the impact of ALB on SREBPs was dependent on the regulation of AMPK/mTOR, thereby impeding the transportation of SREBPs from endoplasmic reticulum to Golgi apparatus. Further investigations indicated that the activation of AMPK by ALB was independent on traditional CAMKK2 and LKB1. Instead, ALB resulted in a decrease in ATP levels and an increase in the ratios of ADP/ATP and AMP/ATP. CETSA, MD, and ultrafiltration-LC/MS analysis indicated that ALB interacted with VDAC1. Molecular docking revealed that ALB directly bound to VDAC1 by forming hydrogen bonds at the amino acid sites S196 and H184 in the ATP-binding region. Importantly, the thermal stabilization of ALB on VDAC1 was compromised when VDAC1 was mutated at S196 and H184, suggesting that these amino acids played a crucial role in the interaction. Conclusion Our findings reveal that VDAC1 serves as the target of ALB, leading to the inhibition of lipid synthesis, presents potential target and candidate drugs for hyperlipidemia.

LPS adsorption and inflammation alleviation by polymyxin B-modified liposomes for atherosclerosis treatment

Huiwen Liu,Honglan Wang,Qiyu Li,Yiwei Wang,Ying He,Xuejing Li,Chunyan Sun,Onder Ergonul,Füsun Can,Zhiqing Pang,Bo Zhang,Yu Hu

Chronic inflammation is critical in the onset and progression of atherosclerosis (AS). The lipopolysaccharide (LPS) level in the circulation system is elevated in AS patients and animal models, which is correlated with the severity of AS. Inspired by the underlying mechanism that LPS could drive the polarization of macrophages toward the M1 phenotype, aggravate inflammation, and ultimately contribute to the exacerbation of AS, LPS in the circulation system was supposed to be the therapeutic target for AS treatment. In the present study, polymyxin (PMB) covalently conjugated to PEGylated liposomes (PLPs) were formulated to adsorb LPS through specific interactions between PMB and LPS. In vitro , the experiments demonstrated that PLPs could adsorb LPS, reduce the polarization of macrophages to M1 phenotype and inhibit the formation of foam cells. In vivo , the study revealed that PLPs treatment reduced the serum levels of LPS and pro-inflammatory cytokines, decreased the proportion of M1-type macrophages in AS plaque, stabilized AS plaque, and downsized the plaque burdens in arteries, which eventually attenuated the progression of AS. Our study highlighted LPS in the circulation system as the therapeutic target for AS and provided an alternative strategy for AS treatment.

Cry41-Related Mutants against Myzus persicae Based on Its Interaction with Cathepsin B

Liang Jin,Xiao-Di Zhao,Jun-Ao Liao,Li-Jun Fu,Yi Lin

Cry toxins produced by Bacillus thuringiensis (Bt) are toxic to Lepidoptera, Coleoptera, and Diptera, but display very low activity against aphids. Recently, Cry41-related toxin was found to show moderate toxicity against Myzus persicae with the 50% lethal concentration (LC50) of 32.7 μg/mL, and its mode of action was proposed to be the acceleration of the apoptosis of aphid cells by enhancing Cathepsin B activity. This study focused on constructing Cry41-related mutants against M. persicae based on its interaction with Cathepsin B. First, eight key interacting residues in Cry41-related toxin were identified using alanine scanning and site-directed saturation mutagenesis. Subsequently, the positive mutant Cry41-7M protein (mutations of Gly48, Ile59, Lys364, Gln367, Gln377, Tyr378, and Ser400 to Tyr, Ala, Arg, Lys, Lys, Lys, and Ala in Cry41-related toxin, respectively) and the negative mutant Cry41-6A protein (mutations of Gly48, Lys364, Gln367, Gln377, and Tyr378 to Ala and mutation of Pro453 to Glu in Cry41-related toxin) were constructed, expressed, and purified. We then found that Cry41-7M protein performed slightly stronger than Cry41-related toxin in enhancing the enzymatic activity of Cathepsin B, whereas Cry41-6A protein did not affect Cathepsin B activity. A further bioassay showed that the mortality caused by Cry41-7M protein (LC50 = 19.144 μg/mL) was marginally higher than that of Cry41-related toxin, while Cry41-6A protein caused a decreased mortality (LC50 = 42.478 μg/mL). These results are expected to open new avenues for improving Cry aphidicidal activity.

Response of Cupriavidus basilensis B-8 to CuO nanoparticles enhances Cr(VI) reduction

Xu Yan,Mengmeng Song,Mo Zhou,Chunlian Ding,Zhongren Wang,Yunyan Wang,Weichun Yang,Zhihui Yang,Qi Liao,Yan Shi

CuO nanoparticles (NPs) released into aqueous environments induce metal toxicity, which generally exerts negative effects on various organisms and leads to great challenge for wastewater biotreatment. In this study, a promotion effect of CuO NPs on biological process was first found. Cr(VI) reduction by Cupriavidus basilensis B-8 (hereafter B-8) was enhanced in the presence of CuO NPs. The efficiency of Cr(VI) bioreduction was much higher with B-8 and CuO NPs (approximately 100%) than with B-8 (approximately 37.6%) and CuO NPs (39.9–44.7%) alone, indicating a stimulatory effect of CuO NPs on Cr(VI) reduction by B-8. Our material analyses revealed different response mechanisms of B-8 to Cr(VI), with and without CuO NPs. The addition of CuO NPs influenced the interaction of Cr(VI) with the N-, P-, S-, and C-related functional groups of B-8. Transcriptomic analysis indicated that multiple mechanisms, including Cr(VI) uptake and reactive oxygen species detoxification, were induced by Cr(VI). Many genes involved in various metabolic processes were significantly upregulated by the addition of CuO NPs. To a certain extent, the pressure of DNA repairment by B-8 induced by Cr(VI) was also alleviated by the presence of CuO NPs. They contributed to facilitate B-8 growth and enhance Cr(VI) reduction, even with 50 mg/L Cr(VI). This study not only elaborated the mechanisms of bacterial Cr(VI) reduction when enhanced by CuO NPs, but also provided a novel perspective for wastewater biotreatment.

Synthesis and Antifungal Activities of Amphiphilic PDMS-b-QPDMAEMA Copolymers on Rhizoctonia solani

Yaling Lin,Weiqiang Zhong,Chenyun Dong,Chang Zhang,Xixiang Feng,Anqiang Zhang

Cationic polymers are prospective fungicidal agents for inhibiting plant diseases because of the controllability of their structure and properties. This study investigates the effect of the hydrophobic–hydrophilic balance on the antifungal activities of antimicrobial polymers against phytopathogenic fungi (Rhizoctonia solani Kühn AG-1(IA)), the pathogen of rice sheath blight (RShB). A series of polydimethylsiloxane-polymethacrylate block copolymers containing quaternary ammonium salts (PDMS-b-QPDMAEMA, labeled as SnQm; n and m represent 1000th of the molecular weight of the PDMS and QPDMAEMA chain, respectively) were synthesized via anionic ring-opening polymerization and atom transfer radical polymerization (ATRP). The abilities of the quaternary ammonium salts to adsorb onto the surface of R. solani sclerotia and permeate the R. solani sclerotia were investigated on the basis of static water contact angles and fluorescence labeling. The results indicated that the moderately hydrophobic PDMS chain helped stabilize the attachment of the hydrophilic QPDMAEMA chain and then help it penetrate the R. solani sclerotia. Its antifungal properties toward R. solani were characterized by determining its minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) and the inhibition rate of R. solani sclerotia germination. The hydrophobic PDMS chains had a significant influence on the antifungal activities of amphiphilic SnQm against R. solani sclerotia. This work highlights the prospective application of amphiphilic antimicrobial polymers as antifungal agents for inhibiting plant diseases.
