Mechanisms for improving the quality of crucian carp (Carassius auratus) by short-term low-salinity treatment revealed by UPLC-MS and GC–MS

Hong Yawen,Pan Yunyun,Zhu Qijian,Li Yongyong,Yang Huicheng,Lin Bangchu,Dong Zheyun,Lou Yongjiang,Fu Shiqian

Due to high-density farming methods and poor farming environments, freshwater fish often have poor muscle texture and a fishy odor before marketing, which impacts consumer acceptance. Short-term low-salinity seawater treatment of freshwater fish before marketing has been found to improve the quality of the fish, but the mechanisms underlying this phenomenon have not yet been revealed. In this study, metabolomics technique was used based on gas chromatography-mass spectrometry (GC–MS) and ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) to screen different metabolites and predict potential metabolic pathways after treatment with low-salinity seawater. The results showed that 3838 metabolites were presented in all of the carp samples, while 62 and 64 differential metabolites were found in the comparative groups (FW, SW, and CK represented freshwater, seawater and non-transient group, respectively). Additionally, 25 and 26 metabolic pathways were enriched from these groups. Glutaric acid, PC, and creatinine might be potential biomarkers. Seawater aquaculture significantly altered the lipid metabolism, nucleotide metabolism, and amino acid metabolism of carp, enhancing its freshness and having a significant impact on its quality. This study provided insights into the metabolic changes in carp under different rearing conditions and also served as a theoretical basis for salinity treatment before freshwater fish are brought to the market.

Exploring metabolic alterations associated with death from asphyxia and the differentiation of asphyxia from sudden cardiac death by GC-HRMS-based untargeted metabolomics

Kai Zhang,Hui Yan,Ruina Liu,Ping Xiang,Kaifei Deng,Ji Zhang,Ya Tuo,Zhenyuan Wang,Ping Huang

The determination of cause of death is one of the most important tasks in forensic practice. However, asphyxia is a difficult cause of death to determine, especially when the deceased has an underlying disease that can lead to a sudden unexpected death, such as coronary atherosclerotic heart disease (CAHD, which is the leading cause of sudden cardiac death, SCD), because its determination is currently still based on an exclusion strategy. In this study, gas chromatography coupled with high-resolution mass spectrometry (GC-HRMS)-based untargeted metabolomics was employed to obtain the pulmonary metabolic profiles of rats who died from asphyxia and SCD. First, fourteen metabolites were identified to investigate the mechanism of death from asphyxia, and we proposed some explanations that may account for these metabolic alterations, including the perturbation of amino acid metabolism, lipid metabolism, and energy metabolism (TCA cycle). Second, we discovered eight potential biomarkers to differentiate between asphyxia and SCD as the cause of death. The excellent classification performances of the eight individual biomarkers and their combination in fresh lung tissue were observed. Third, we also explored the relative change in the concentration of the eight metabolites and their classification performance in decomposed tissue (at 24 h postmortem). Lactic acid, pantothenic acid, and the combination of the eight biomarkers can be recognized as perfect classifiers to discriminate asphyxia from SCD even when decomposition has occurred. Our results showed that GC-HRMS-based untargeted metabolomics can be used as a promising tool to explore the metabolic alterations of the death process and to determine the cause of death.

Magnetic solid phase extraction using Fe3O4@β-cyclodextrin–lipid bilayers as adsorbents followed by GC-QTOF-MS for the analysis of nine pesticides

Hui Wang,Shaowei Yan,Bo Qu,He Liu,Jie Ding,Nanqi Ren

A rapid and efficient magnetic solid phase extraction (MSPE) method, based on Fe3O4@β-cyclodextrin–lipid bilayer nanocomposites (Fe3O4@β-CD–LB), was built. The MSPE method coupled with GC-QTOF-MS has been designed for the trace detection of nine organochlorine pesticides (OCPs) in soil samples. The Fe3O4@β-CD–LB was successfully synthesized and characterized by vibrating sample magnetometry, Fourier transform infrared spectrometry, transmission electron microscopy, field emission scanning electron microscopy and energy-dispersive X-ray spectroscopy. In this experiment, the OCPs in the soil samples were firstly extracted with n-hexane/dichloromethane (v/v 1 : 1) at high pressure and temperature, and then subjected to MSPE for adsorption and purification. Considering the synergy effect between host–guest inclusion complexation and hydrophobic interaction in the adsorption mechanism, the bi-functionalized magnetic nanocomposites exhibited excellent adsorption properties for the OCPs and natural resistance to interference by macromolecules. Under the optimum experimental conditions, the method showed wide linear ranges with low limits of detection (4.5 to 34 pg g−1). The results showed that the mean recoveries of the OCPs in the soil samples were 78–107% with 3.0–6.1% intra-day relative standard deviation (RSD) and 3.5–6.9% inter-day RSD. The method has been applied for the determination of the OCPs in actual samples, and ΣOCPs ranged between 0 and 2.3 ng g−1 in seven soil samples.

Effects of alkali and alkaline earth metals on the co-pyrolysis of cellulose and high density polyethylene using TGA and Py-GC/MS

Xiaona Lin,Lingshuai Kong,Hongzhen Cai,Qingfa Zhang,Dongmei Bi,Weiming Yi

The effects of the type and content of alkali/alkaline earth metals (AAEMs) on the co-pyrolysis of cellulose/high density polyethylene (HDPE) were studied using TGA and Py-GC/MS. AAEMs showed different effects on the decomposition characteristics of cellulose/HDPE mixtures. During cellulose/HDPE co-pyrolysis, AAEMs present in char could act as a base catalyst, with different catalytic effect on products distribution depending on the types of AAEMs. Na showed stronger catalytic cracking effect to promote alkenes formation compared to the use of K, Ca, and Mg. The comparison of experimental and calculated products distributions of raw cellulose/HDPE confirmed the existence of synergistic effect , which facilitated the cleavage of glycosidic linkages to form anhydrosugars and promoted the scission of HDPE chains to produce hydrocarbons. The effect of potassium on this synergy was also investigated. With increased potassium content, the yields of anhydrosugars and hydrocarbons decreased while the yields of furans and ketones increased, suggesting that potassium inhibited the cleavage of glycosidic bonds and promoted ring-opening reactions. This indicated that addition of potassium inhibited the synergy between cellulose and HDPE.

Zhang Fengsu,Cai Hao,Liu Xunhong,Chen Fei,Yang Nianyun,Hou Ya,Ma Yang,Luo Yiyuan,Liu Juanxiu

To analyze the components of the fatty oil extracted from Antrodia camphorata fungus powder and to evaluate the antioxidant activity of A. camphorata crude oil. A derivatized gas chromatography mass spectrometry (GC–MS) was developed for the quantification of 19 fatty acids in extracts of A. camphorata obtained by supercritical carbon dioxide (SC-CO 2 ). Good separation was obtained under the optimized chromatographic conditions and 19 target compounds after methyl-esterification were identified by GC–MS on a HP-VOC capillary column (60 m × 320 μm × 1.8 μm) with an initial temperature set at 80 °C. The validity of the established method was examined experimentally with good linearity, intra-assay precisions, repeatability, stability and recovery. The antioxidant activity of A. camphorata crude oil was evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging assay. Nineteen fatty acids showed good linearity over the tested ranges (r > 0.9956) and the recovery ranged from 93.47 to 104.89 %. The crude oil extracted from A. camphorata fungus powder also revealed its antioxidant activity. It was the report about simultaneous determination of 19 fatty acids in A. camphorata and antioxidant activity of its crude oil for the first time. The established method might also be utilized for the investigation of edible plant materials and agricultural products containing fatty acids.

Weiwei Wu,Zhiqiang Zheng,Zhihui Wang,Biyun He,Siqing Du,Wen Zeng,Weijiang Sun

White Peony Tea (WPT) is valued for its unique flavor and pleasant sensory effects. However, the specific aroma compounds in WPT contributing to this pleasant sensation and the underlying mechanisms remain unclear. This study integrates GC–MS, computational modeling, and sensory evaluation to systematically explore the key pleasant aroma compounds in WPT and their potential mechanisms. Seven key components were identified as eliciting the pleasant sensation of WPT, including cedrol and β -ionone. These compounds activate human pleasure receptors, influencing neurological pathways to enhance sensory pleasure. Molecular simulations validated the stability of the interactions between the aroma compounds and their targets, specifically cedrol–CHRM4, cedrol–ADORA1, β -ionone–ADORA2A, and cedrol–MAOB. Sensory evaluation revealed pleasantness scores for these aroma compounds between 2.60 and 6.80, supporting the positive effects on sensory experience. This research provides scientific insight into the relationship between tea aroma and sensory pleasure, providing a foundation for the development of tea products.

Molecular dynamics simulation of the effects of base oils in barium-based greases on tribological properties based on IR and GC/MS data

Hao Xu,Run-Song Wang,Yang-Yang Yan,Xing Fan,Xiang Bai,Wen-Long Mo,Xian-Yong Wei

Twelve kinds of base oils were characterized by a series of analytical methods to obtain detailed composition and structure information. Mineral oil and poly alpha olefin oil were mainly composed of long-chain alkanes, while ester oil was mainly composed of ester compounds. Ten of the 12 base oils were selected to synthesize barium lubricating greases . Both base oils and greases were evaluated by molecular simulation , and the corresponding molecular models were constructed based on the structural information of base oils. Grease models constructed by barium 12-hydroxystearate and ester oil exhibited the largest surface adsorption energy and intermolecular interaction energy, respectively, because polar groups in lubricant tend to adsorb on metal surface and form an absorbing film. Comparing physicochemical properties of the synthesized barium lubricating greases. The temperature resistance of grease presents a positive correlation with the simulated intermolecular interaction energy.

Revealing the characteristic aroma and boundary compositions of five pig breeds based on HS-SPME/GC-O-MS, aroma recombination and omission experiments

Xueli Chen,Haoyue Liu,Cong Li,Yujuan Xu,Baocai Xu

To clarify the characteristic aroma compounds and flavor discrepancies of five Chinese typical pig species, headspace-solid phase microextraction gas chromatography–olfactometry-mass spectrometry (HS-SPME/GC-O-MS), electronic nose (E-nose), aroma recombination and omission experiments were used to analyze the characteristic aroma and boundary of five boiled pork. A total of 38 volatile compounds were identified, of which 14 were identified as important odorants with odor-activity values (OAVs) greater than 1. Aroma recombination and omission experiments revealed 8 key characteristic aroma compounds, which significantly contributed to the overall aroma. Sensory evaluation of the recombination model with the 8 aroma compounds scored 3.0 to 4.0 out of 5 points. 12 potential markers were identified to distinguish by principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA), including (E)-2-octenal, 3-ethyl-2-methyl-1,3-hexadiene, (E)-2-heptenal, 2-pentylfuran, cyclooctanol, 1-heptanol, sec -butylamine, D-limonene, N-vinylformamide, 2,3-octanedione, 2-ethylfuran and 3-pentanamine. Alongside benzaldehyde and pentanal, the combinations and fluctuations of these 14 aroma markers were proposed to constitute the aroma boundaries of different pork breeds. The aroma-active substances were able to effectively differentiate different breeds.

Determination of seven fatty acids in plasma of rats after oral administration of Xanthoceras sorbifolia bunge oil by GC-MS and pharmacokinetic study

Guo Shuang,Qiao Xuan,Lv Zihan,Li Lintao,Liang Yutong,Meng Zhiyun,Gan Hui,Gu Ruolan,Wu Zhuona,Dou Guifang,Li Gaoying,Liu Shuchen

This work aims to establish a highly selective, sensitive, and accurate gas chromatography-mass spectrometry (GC-MS) method for detecting concentrations of seven representative fatty acids absorbed into the bloodstream after a single gavage of 10 mL/kg Xanthoceras sorbifolia Bunge ( X.sorbifolia ) oil in rats and conducting pharmacokinetic studies. The plasma samples were processed using two-step derivatization with NaOH and BF 3 -CH 3 OH, with an Agilent HP-88 capillary column serving as the stationary phase. Concentrations of seven fatty acids, namely hexadecanoic acid (palmitic acid, C16:0), octadecenoic acid (stearic acid, C18:0), cis -9-octadecenoic acid (oleic acid, C18:1), cis -9,12-octadecadienoic acid ( linoleic acid, C18:2), cis -11-eicosenoic acid (gondoic acid, C20:1), cis -13-docosaenoic acid (erucic acid, C22:1), and cis -15-tetracosenoic acid (nervonic acid, C24:1), were quantitatively determined under programmed temperature and ion detection mode. The results showed that the seven fatty acids were well separated and exhibited a good linear relationship between concentration and peak area ratio within the detection range (R 2  > 0.99). The precision and accuracy within and between batches were both less than 15%. After derivatization, the plasma samples remained stable at -20 °C, 4 °C, and 10 °C; The matrix effect did not affect the quantitative analysis, and the extraction recovery rate ranged from 100.3% to 107.76%. After gavage administration of X.sorbifolia oil to rats, the levels of unsaturated fatty acids in the blood were significantly increased compared to those in the blank control group. The peak concentration (T max) was 7–8 h, and the mean retention time (MRT) was 10–12 h. This experiment provides a basis for elucidating the potential material basis of the nutritional efficacy of X. sorbifolia oil after oral administration.
