Densities and Viscosities for the Ternary System of Decalin + Methylcyclohexane + Cyclopentanol and Corresponding Binaries at T = 293.15 to 343.15 K

Xiaoyi Chen,Shenda Jin,Yitong Dai,Jianzhou Wu,Yongsheng Guo,Qunfang Lei,Wenjun Fang

Densities (ρ) and viscosities (η) for the ternary system of decalin (1) + methylcyclohexane (2) + cyclopentanol (3) and three corresponding binary systems have been measured over the whole composition range at 11 temperature points from 293.15 K to 343.15 K under atmospheric pressure (0.1 MPa). The excess molar volumes (VmE) and viscosity deviations (Δη) of binary systems have been calculated and further fitted with the Redlich–Kister equation, while corresponding physical data of the ternary system have been correlated via the Clibuka, Singh, Nagata-Tamura, and Redlich–Kister equations. The VmE values are negative for the binary system of decalin (1) + methylcyclohexane (2) with a minimum when the moles of the two components are similar. For the system of decalin (1) + cyclopentanol (2), the VmE values are always positive with a maximun at about x1 = 0.6. At the same time, a sigmoid curve can be observed for the system of methylcyclohexane (1) + cyclopentanol (2). The minimum and maximum appear around x1 = 0.2 and x1 = 0.9, respectively. The Δη values of the three binary systems are all negative and the absolute values decrease with increase in temperature. For the ternary system, the VmE values are partially negative and the Δη values are negative over the entire concentration range. The nonideal behaviors of the mixtures are discussed in the perspective of intermolecular interaction and structural effect.

Maternal Exposure to T-2 Toxin Induces Changes in Antioxidant System and Testosterone Synthesis in the Testes of Mice Offspring

Jiakun Shen,Aneela Perveen,Niaz Kaka,Zhaojian Li,Pengyuan Dai,Chunmei Li

Simple SummaryThis study investigated the effects of maternal T-2 toxin exposure on the development of testis in the mice offspring. The detrimental effects were assessed by testicular weight, antioxidant capacity, and testosterone synthesis and secretion. Studies have shown that the toxin carried by the mother has bad effects on the testicular development of offspring at puberty, affecting the antioxidant system and testosterone synthesis in the testis, but the maternal exposure of T-2 toxin had no significant impact on the testes of offspring after sexual maturity, suggesting the recovery of reproductive function.AbstractT-2 toxin, the most toxic member of trichothecene mycotoxin, is widely distributed in cereals, and has been extensively studied, but few studies focus on the toxicity of maternal exposure to offspring. This study focused on the effects of maternal exposure to T-2 toxin (during gestation and lactation) on the testicular development of mice offspring. Dams were orally administered with T-2 toxin at 0, 0.005, or 0.05 mg/kg body weight from the late stage of gestation to the end of lactation. Testicular samples of the mice offspring were collected on the postnatal day 21, 28, and 56. The results showed significant decreases in body weight and testicular weight on the postnatal day 28. Moreover, significant inhibition of antioxidant system and testosterone synthesis was detected on the postnatal day 28. Furthermore, there were significant decreases in the gene expression levels ofStARand3β-HSD, which are involved in testosterone synthesis. In general, present results demonstrated that maternal exposure to T-2 toxin during gestation and lactation led to bad effects on the capacity of antioxidant system and inhibited testosterone synthesis in testes during pre-puberty with no significant effects on post-puberty.Keywords:maternal exposure;oxidative stress;T-2 toxin;testicular development;testosterone synthesis

Properties of pure 1,1,3,3-tetramethylguanidine imidazole ionic liquid and its binary mixtures with alcohols at T = (293.15 to 313.15) K

Jiayi Chen,Lingxiao Chen,Yingjie Xu

Densities and viscosities of the pure ionic liquid 1,1,3,3-tetramethylguanidine imidazole ([TMG]IM) and its binary mixtures with methanol, ethanol, 1-propanol, and 1-butanol were measured at temperatures from T   =   (293.15 to 313.15)   K. The thermal expansion coefficient, molecular volume, standard entropy, and lattice energy of [TMG]IM were obtained from the experimental density value. The temperature dependence of the viscosity of [TMG]IM was fitted to the fluidity equation. Excess molar volumes V E and viscosity deviations Δ η of the binary mixtures were calculated and fitted to the Redlich–Kister equation with satisfactory results. The result shows that the V E values of the binary mixtures are negative over the whole composition range, while Δ η values have an S-shape deviation. Temperature has little effect on the V E of the systems, but it has significant effect on the Δ η . Furthermore, the absolute values of V E for {[TMG]IM (1)   +   alcohol (2)} systems at the same temperature decrease with increasing carbon alkyl chain of the primary alcohol. Other derived properties, such as the apparent molar volumes, partial molar volumes, excess partial molar volumes, Gibbs free energy of activation for viscous flow, and excess Gibbs free energy of activation for viscous flow of the above-mentioned systems were also calculated.

Measurement and correlation of solubility and solution thermodynamics of 1,3-dimethylurea in different solvents from T = (288.15 to 328.15) K

Peipei Zhu,Yanxin Chen,Meijing Zhang,Ying Bao,Chuang Xie,Baohong Hou,Junbo Gong,Wei Chen

In this work, the solubility of 1,3-dimethylurea form I (DMUI) in seven solvents including water, 2-butanol, 2-propanol, acetone, acetonitrile, ethyl acetate and butyl acetate were measured by a synthetic method at a temperature range from (288.15 to 328.15)   K at atmospheric pressure. For the temperature range investigated, the solubility of DMUI in the selected solvents increased with increasing temperature. Results of these measurements were well-correlated with the modified Apelblat equation, λh equation, van’t Hoff equation and Wilson model. Besides, the thermodynamic data ( i.e. , Gibbs energy, enthalpy, and entropy of mixing) were calculated by using the Wilson model. The chemical potential, solvation free energy and interaction enthalpy of DMU in solution were calculated by COSMO-RS model. This model was also applied to predict DMU solubility which showed good agreement with the experimental values.

Equilibrium study and diversified models of drug Norfloxacin in eight pure organic and binary solvents at T = (278.15–328.15) K

Yemin Yu,Tao Li,Yonghong Hu,Wenge Yang,Yuhao zhang,Renlun Deng,Min Jiang

Research on relevant solid–liquid equilibrium of drug Norfloxacin (NFLX) in different solvents is necessary for an elementary study of industrial implantation. In this work, the solid–liquid equilibrium of NFLX in ethanol, 1-butanol, isopropanol, acetone, acetonitrile , ethyl acetate , N-hexane, N,N-dimethylformamide single pure solvent and in (N,N-dimethylformamide + ethyl acetate) binary solvent mixtures were probed under atmosphere pressure at desired temperatures from 278.15 K to 328.15 K. The Gibbs energy , enthalpy, and entropy of solution were obtained from these solubility data. The modified Apelblat model, the Buchowski–Ksiazaczak λh model, Redich-Kister (CNIBS/R-K) model, Jouyban-Acree model, ideal model and the Van't Hoff analysis were applied to depict and forecast the diversification tendency of solubility. The satisfactory simulation results were obtained. Moreover, the thermodynamic functions for NFLX dissolution from calculation, using the van't Hoff and Gibbs equations, imply that in each studied solvent and in binary solvent mixtures, the process of NFLX dissolution is endothermic. Based on the above analysis, N,N-dimethylformamide is checked as the best solvent and ethyl acetate can be used as an effective anti-solvent in the crystallization process of NFLX.

Cascade STING activation of a ternary immunostimulant for colorectal cancer eradication via photodynamic DNA damage and T regulatory cell inhibition

Yanmei Li,Junmei Nie,Xinyu Li,Ni Yan,Renjiang Kong,Jiaqi Huang,Yeyang Wu,Zhuofeng Li,Yingtao Zhong,Xin Chen,Hong Cheng

Stimulator of interferon genes (STING) plays an essential role in initiating the innate anti-tumor immunity, which could be activated by the cytosolic DNA but suppressed by the immunosuppressive cells. In this work, a self-delivery ternary immunostimulant (designated as ImmSt) is fabricated for colorectal cancer therapy with the immunological cascades of photodynamic DNA damage and T regulatory cell inhibition. Among which, the photosensitizer of Chlorin e6 (Ce6), the STING agonist of 5,6-dimethylxanthine-4-acetic acid (DMXAA) and the phosphatidylinositol 3-kinases (PI3Ks) inhibitor of ZSTK474 could self-assemble into uniform nanomedicine without drug excipient, which enhances the drug stability, bioavailability and pharmacological activities. Importantly, the photodynamic therapy (PDT) effect of ImmSt produces enormous reactive oxygen species to ablate tumor cells, and also triggers the release of cytosolic DNA to amplify the STING signal in combination with the STING agonist. Moreover, ImmSt is able to induce a systemic anti-tumor immunity by the cascade activation of T cells and the decrease of T regulatory cells. In vitro and in vivo results confirm that ImmSt can efficiently eradicate colorectal cancer without severe side effects, which may provide a sophisticated mechanism for immunotherapy in consideration of the immune activation and immunosuppressive factors.

Chaetoglobosin A induces apoptosis in T-24 human bladder cancer cells through oxidative stress and MAPK/PI3K-AKT-mTOR pathway

Jia Song,Jinyu Qiao,Mingxue Chen,Jiahui Li,Jixia Wang,Dayong Yu,Huachuan Zheng,Liying Shi

Chaetoglobosin A (ChA) is an antitumor compound produced by Chaetomium globosum . However, the mechanism of its antitumor effect has been rarely reported. In this study, we evaluated the anti-proliferative effect of ChA on T-24 human bladder cancer cells and explored its mechanism of action. ChA was found to have a good inhibitory effect on T-24 cells by MTT assay with an IC50 value of 48.14 ± 10.25 μΜ. Moreover, it was found to have a migration inhibitory ability and a sustained proliferation inhibitory effect on tumor cells by cell aggregation assay and cell migration assay. The cells morphological changes were determined by Hoechst33342 assay. While Annexin V-FITC/PI double-staining assay also demonstrated that the number of apoptotic cells increased with the increase of drug concentration. Flow cytometry results showed that ChA treatment increased reactive oxygen species (ROS) and decreased mitochondrial membrane potential (MMP) in T-24 cells and inhibited cell mitosis, resulting in an increase in the number of sub-G1 phase cells. Further western blot experiments demonstrated that MAPK and PI3K-AKT-mTOR pathways were activated after drug treatment in addition to endogenous and exogenous apoptotic pathways. The addition of the ROS inhibitor N-acetylcysteine (NAC) upregulated the expression level of Bcl-2 protein, decreased p38 phosphorylation, increased ERK phosphorylation and restored the levels of PI3K and p-mTOR after ChA treatment. These suggest that ChA induces apoptosis by regulating oxidative stress, MAPK, and PI3K-AKT-mTOR signaling pathways in T-24 cells.

In-situ trichosanthin-IL2/pectin dynamic hydrogel activates dendritic cells and reverses T cell exhaustion for post-operative cancer therapy

Zeyun Gu,Guihua Chen,Nan Gao,Shuo Yao,Xiaoru Zhang,Quangang Xu,Wei Xiong,Lin Liu,Qin Liu,Dan Yin,Xiao-Ming Zhu,Yongzhuo Huang

Postoperative recurrence and metastasis of triple-negative breast cancer (TNBC) remain major clinical challenges due to immunosuppression and poor immune recognition of residual cancer cells. Here, we present a novel polysaccharide-protein hydrogel implant, composed of a trichosanthin (TCS)-IL2 fusion protein crosslinked with oxidized pectin, designed for sustained local immunotherapy following tumor resection. TCS is a plant-derived protein with immune-stimulatory properties. This in-situ forming gel (termed TL-pectin gel) delivers the TCS-IL2 (TL) protein to enhance dendritic cell (DC) maturation, improve antigen presentation, and reverse T-cell exhaustion, thereby stimulating tumor-specific cytotoxicity. In a mouse breast cancer model, the TL-pectin hydrogel effectively inhibited local tumor recurrence and reduced lung and brain metastases, demonstrating its potential to improve postoperative outcomes. Mechanistic studies indicated that TCS activated the NF-κB pathway, enhanced cytokine secretion and CD8 + T-cell differentiation, and IL2 maintained T-cell proliferation and activity. The hydrogel showed excellent biocompatibility and functioned as a controlled-release system to form a localized immunostimulatory microenvironment. This study suggests that TL-pectin gel is a promising candidate for clinical translation, offering a potential approach for adjuvant cancer immunotherapy.

NCF4 regulates antigen presentation of cysteine peptides by intracellular oxidative response and restricts activation of autoreactive and arthritogenic T cells

Jing Xu,Chang He,Yongsong Cai,Xipeng Wang,Jidong Yan,Jing Zhang,Fujun Zhang,Vilma Urbonaviciute,Yuanyuan Cheng,Shemin Lu,Rikard Holmdahl

Autoimmune diseases, such as rheumatoid arthritis (RA) and systemic lupus erythematous, are regulated by polymorphisms in genes contributing to the NOX2 complex. Mutations in both Ncf1 and Ncf4 affect development of arthritis in experimental models of RA, but the different regulatory pathways mediated by NOX2-derived reactive oxygen species (ROS) have not yet been clarified. Here we address the possibility that intracellular ROS, regulated by the NCF4 protein (earlier often denoted p40phox) which interacts with endosomal membranes, could play an important role in the oxidation of cysteine peptides in mononuclear phagocytic cells, thereby regulating antigen presentation and activation of arthritogenic T cells. To study the role of NCF4 we used mice with an amino acid replacing mutation (NCF4 R58A ), which is known to affect interaction with endosomal membranes, leading to decreased intracellular ROS production. To study the impact of NCF4 on T cell activation, we used the glucose phosphate isomerase peptide GPI 325-339 , which contains two cysteine residues (325-339c-c). Macrophages from mice with the NCF4 58A mutation efficiently presented the peptide when the two cysteines were intact and not crosslinked, leading to a strong arthritogenic T cell response. T cell priming occurred in the draining lymph nodes (LNs) within 8 days after immunization. Clodronate treatment, which depletes antigen-presenting mononuclear phagocytes, ameliorated arthritis severity, whereas treatment with FYT720, which traps activated T cells in LNs, prohibited arthritis. We conclude that NCF4-dependent intracellular ROS maintains cysteine peptides in an oxidized crosslinked state, which prevents presentation of peptides recognized by non-tolerized T cells and thereby protects against autoimmune arthritis.

Modulation of metal-organic frameworks by anionic surfactants for rapid adsorption of organic pollutants bisphenol A and chrome black T

Wenwen Li,Jihui Li,Jian Han,Kaili Liu,Huixin Zhang,Jianxin Chen

With the accelerated development of industry, which generates large quantities of sewage discharged into the aquatic environment, serious impacts on both environmental and human health. Detecting various organic contaminants in water sources has become common, presenting huge challenges for water treatment. Metal-organic frameworks are characterized by their rich porous structures and high chemical activity, making them ideal adsorbents for removing organic pollutants from water. In this study, sodium dodecylbenzene sulfonate is added to the precursor solution of MIL-101(Cr)-Cl 2 and synthesized hierarchical porous structure adsorbents MIL-SDBS-X, using hydrothermal synthesis to enhance their adsorption capacity, making efficient and rapid removal of bisphenol A(BPA) and chromium black T(EBT), two common organic pollutants in water, which MIL-SDBS-0.7 was the most effective. MIL-SDBS-0.7 exhibites great potential in the adsorption of BPA and EBT for its high adsorption capacity (260.5 mg -1 ·g -1 and 272.3 mg -1 ·g -1 ) and rapid adsorption (equilibrium could be achieved in 15 minutes), The findings reveal adsorption kinetics aligns with the pseudo-second-order model and the Langmuir equation best fits to study adsorption of MIL-SDBS-0.7 to BPA and EBT. Thermodynamic parameters affirm the exothermic and higher spontaneity behavior of the adsorption process . Moreover, the adsorption mechanism of MIL-SDBS-0.7 was explored, illustrating the crucial roles of effects of hydrogen bonding π-π stacking and pore filling in the adsorption of BPA and EBT. Therefore, this work not only provided a convenient strategy for the preparation of efficient and fast MIL-SDBS-0.7 hierarchical porous material but also highlighted the promising potential of synthesized absorbents as an effective adsorbent for organic pollutants in wastewater treatment and environmental remediation.

Solubility Determination and Thermodynamic Modeling of Sodium Thioglycolate in Pure and Binary Solvent Mixtures from T = (293.15 to 333.15) K

Jingyu Wu,Lianzheng Zhang,Dongmei Xu,Jun Gao,Kai Zhang,Meng Kong

The solubility of sodium thioglycolate in five pure solvents of water, methanol, ethanol and isopropyl alcohol, n-propanol and four binary solvent mixtures (water + methanol, water + ethanol, water + isopropyl alcohol, water + n-propanol) was determined within the temperature range from (293.15 to 333.15) K by using the gravimetrical method under atmospheric pressure (p = 0.1 MPa). The solubility of sodium thioglycolate in water, ethanol, isopropyl alcohol, and n-propanol increases with a rise of temperature, but the solubility decreases in methanol with increasing temperature. Meanwhile, in the binary solvent mixtures (water + methanol, water + ethanol, water + isopropyl alcohol, water + n-propanol), the solubility of sodium thioglycolate increases with increasing the mole fraction of water and with temperature. The modified Apelblat equation and van’t Hoff equation were applied to correlate the solubility of sodium thioglycolate in the five pure solvents, while in the binary solvent mixtures, the solubility was correlated by the modified Apelblat equation, CNIBS/R-K model, and Jouyban–Acree model. It was shown that the correlated results were in better agreement with the experimental data. Also, the mixing thermodynamic properties for sodium thioglycolate in the binary solvent mixtures were calculated.

Synthesis of 1-hexanol by highly selective hydrodeoxygenation of 5-hydroxymethylfurfural using Ni/MCM-41 and Pt-WOX/t-ZrO2

Ying Xu,Genkuo Nie,Xiao Jiang,Hongyu Wang,Guihua Yang,Ziyi Yan,Ji-Jun Zou,Hailong Yu,Shitao Yu,Yu Liu

Metal-metal oxide catalysts are robust in selective hydrodeoxygenation of biomass derives. Herein, 1-hexanol in total yield about 76 % is firstly achieved by high selective hydrodeoxygenation of 5-hydroxymethylfurfural over Ni/MCM-41 and Pt-WO X /t-ZrO 2 . By controlling the amount ratio of Ni/Ni 2+ to 0.46, a 99.9 % yield of 2,5-tetrahydrofurandimethanol was achieved from 5-hydroxymethylfurfural by Ni/MCM-41. Subsequently high yield of 76 % of 1-hexanol was obtained by competing pathways of tandem reactions beginning at SN2 reaction at the epoxy atom of 2,5-tetrahydrofurandimethanol to enol/tautomer intermediates and ending with controllable selective hydrogenation. The reaction mechanism is explored and the reaction-oriented regulation is realized by the additional strong BrÖnsted acid sites generated by electron transfer from W to Pt and the appropriate amount of Pt 0 sites of Pt-WO X /t-ZrO 2 , where the ratios of W 5+ /W 6+ and Pt 0 /Pt 2+ are 0.31 and 4.58 respectively. This work provides a new and green method for the synthesis of 1-hexanol and expands the utilization of metal–metal oxide in making value-added products using sustainable feedstocks.
