Design, fabrication and evaluation of redox/pH dual-responsive micelles for tumor targeting delivery and controlled release of paclitaxel

Yingxue Xu,Na Liang,Jia Fu,Zhe Zong,Xueling Long,Shaoping Sun

In this study, a polymer with tumor targeting function and dual redox/pH sensitivity is designed and synthesized by integrating chitosan (CS), Poloxamer 407 (P407), cholesterol (CHO), folic acid (FA), and disulfide bonds into one molecule (P407-CS(FA)-SS-CHO). The polymer can self-assemble in water to form micelles and solubilize paclitaxel (PTX). The mean particle size of PTX-loaded P407-CS(FA)-SS-CHO micelles is 133 nm. In vitro drug release study demonstrates the acid and glutathione (GSH) triggered PTX release behavior of the micelles. In vitro and in vivo experiments demonstrate the excellent tumor growth inhibition efficacy and low systemic toxicity of the PTX-loaded micelles. The P407-CS(FA)-SS-CHO micelles will be potential intelligent drug carriers for PTX delivery.

Controlled release of fishy odors in soluble dietary fiber/surimi mixture by kinetic analysis of real and simulated systems

Jinggui Nie,Chao Xue,Yueqi An,Shanbai Xiong,Qilin Huang

This study aimed to investigate the effects and mechanisms of soluble dietary fiber (SDF), temperature and hydrophobicity on the release of fishy compounds from surimi by the preparation of real and simulated systems. The results of gas chromatography-mass spectrometry revealed 15 fishy compounds in silver carp surimi, and adding 2 wt% SDF to surimi was found to inhibit the release of 14 fishy compounds by 2.16–43.49%. The simulated release indicated that SDF matrices with different concentrations inhibited the release of fishy compounds by different mechanisms. The resistance to the release of fishy compounds was attributed to viscosity and chemisorption in 2 and 5 wt% SDF matrices, while chemisorption and entanglement network in 8 and 10 wt% SDF matrices. Moreover, high temperature and hydrophobicity of fishy compounds could promote their release by decreasing interfacial mass transfer resistance and H-bond interaction, respectively. Additionally, adding SDF changed the release pathway of fishy compounds in surimi, and they were released through the first pathway consisting of surimi intraparticle diffusion, SDF boundary layer diffusion and gas-liquid interfacial diffusion, or through the second pathway additionally including chemisorption and desorption. This study provides new insights into controlling the release of fishy odors in surimi products.

pH-Responsive Micelles Assembled by Three-Armed Degradable Block Copolymers with a Cholic Acid Core for Drug Controlled-Release

Jingjie Feng,Weiqiu Wen,Yong-Guang Jia,Sa Liu,Jianwei Guo

One of the most famous anticancer drugs, paclitaxel (PTX), has often been used in drug controlled-release studies. The polymers derived from bio-compound bile acids and degradable poly(ε-caprolactone) (PCL) form a reservoir and have been used as a drug delivery system with great advantages. Herein, we grafted poly(N,N-diethylaminoethyl methacrylate) and poly(poly(ethylene glycol) methyl ether methacrylate) into the bile acid-derived three-armed macroinitiator CA-(PCL)3, resulting in the amphiphilic block copolymers CA-(PCL-b-PDEAEMA-b-PPEGMA)3. These pH-responsive three-armed block copolymers self-assembled into micelles in aqueous solution and PTX was encapsulated into the micellar core to form PTX-loaded micelles with a drug loading of 29.92 wt %. The micelles were stable in PBS at pH 7.4 and showed a pH-triggered release behavior of PTX under acidic environments, in which 55% of PTX was released at pH 5.0 in 80 h. These cholic acid-based functionalized three-armed block polymers present good biocompatibility, showing great potential for drug controlled-release.Keywords:cholic acid;pH-sensitive;PTX;drug delivery

Exploration on the adsorption mechanism of aminopyrimidines and quinazoline compounds on graphene oxide: Hydrophobicity and structure-controlled release process

Li Li,Pan Chunjiao,Guo Zhongqiu,Liu Bingmi,Pan Hao,Wu Haotian,Liu Yu

In this study, graphene oxide was synthesized using the Hummers method, and stable and homogeneous graphene oxide aqueous solutions were obtained through mechanical stirring and ultrasonic stripping. In conjunction with our previous studies, graphene oxide-loaded insoluble compound delivery systems were prepared to verify the in vivo release profiles of the graphene oxide delivery system. Several insoluble compounds including imatinib, nilotinib, erlotinib, gefitinib, and afatinib were selected for loading and in vitro graphene oxide release assays to study the non-covalent adsorption mechanisms. Computer simulations were employed for validation processes. For in vivo release assays, the T 1/2 values of the poorly water soluble groups were 1.104 ± 0.18 h and the C max was 2.600 ± 2.06 mg/L. In previous assays, compounds with high water solubility supported by graphene oxide were released and detected in vivo. The solubility of the compound and its binding force with the carrier played a crucial role in release. The results of graphene oxide loading experiments showed that the maximum loading and entrapment efficiencies of the insoluble model compounds with similar aromatic rings were comparable. Under basic conditions, the in vitro release rates and maximum release levels of amino pyrimidine were elevated. In contrast, quinazoline release declined. Combined with computer simulations, π–π stacking was identified as the dominant mechanism for adsorption onto graphene oxide. Both hydrogen bonding and cation-π bonds played an auxiliary reinforcing role, and the two were regarded as antagonistic.

Fluorometric turn-on detection of ascorbic acid based on controlled release of polyallylamine-capped gold nanoclusters from MnO2 nanosheets

Tan Qingqing,Kong Weisu,Sun Han,Qin Xia,Qu Fengli

A fluorometric turn-on assay is described for ascorbic acid (AA). It is based on the controlled release of polyallylamine-stabilized gold nanoclusters (polyallylamine-AuNCs) from MnO 2 nanosheets. In an aqueous solution of near-neutral pH value, the positively charged capped AuNCs are adsorbed on the surface of the negatively charged MnO 2 nanosheets. The adsorption leads to the quenching of the fluorescence of the AuNCs. However, in the presence of AA, MnO 2 is reduced to Mn 2+ . This causes the destruction of the MnO 2 nanosheets. As a result, the fluorescence of the polyallylamine-AuNCs at 615 nm is recovered. This method for determination of AA is inexpensive, sensitive, and selective. It works in the 0.01 to 200 μM concentration range and has a 3.2 nM detection limit (for S/ N  = 3). Gold nanoclusters (AuNCs) and polyallylamine can form polyallylamine-AuNCs to enhance the orange fluorescence of AuNCs. MnO 2 nanosheets can absorb polyallylamine-AuNCs, and this results in fluorescence quenching of polyallylamine-AuNCs. Ascorbic acid (AA) can reduce MnO 2 nanosheets, in this results in the fluorescence recovery of polyallylamine-AuNCs.

Coencapsulation of (−)-Epigallocatechin-3-gallate and Quercetin in Particle-Stabilized W/O/W Emulsion Gels: Controlled Release and Bioaccessibility

Xing Chen,David Julian McClements,Jian Wang,Liqiang Zou,Sumeng Deng,Wei Liu,Chi Yan,Yuqing Zhu,Ce Cheng,Chengmei Liu

Particle-stabilized W1/O/W2 emulsion gels were fabricated using a two-step procedure: (i) a W1/O emulsion was formed containing saccharose (for osmotic stress balance) and gelatin (as a gelling agent) in the aqueous phase and polyglycerol polyricinoleate (a lipophilic surfactant) in the oil phase; (ii) this W1/O emulsion was then homogenized with another water phase (W2) containing wheat gliadin nanoparticles (hydrophilic emulsifier). The gliadin nanoparticles in the external aqueous phase aggregated at pH 5.5, which led to the formation of particle-stabilized W1/O/W2 emulsion gels with good stability to phase separation. These emulsion gels were then used to coencapsulate a hydrophilic bioactive (epigallocatechin-3-gallate, EGCG) in the internal aqueous phase (encapsulation efficiency = 65.5%) and a hydrophobic bioactive (quercetin) in the oil phase (encapsulation efficiency = 97.2%). The emulsion gels improved EGCG chemical stability and quercetin solubility under simulated gastrointestinal conditions, which led to a 2- and 4-fold increase in their effective bioaccessibility, respectively.

A Smart Anticorrosive Epoxy Coating Based on Graphene Oxide/Functional Mesoporous Silica Nanoparticles for Controlled Release of Corrosion Inhibitors

Zheng Liu,Biao Zhang,Hao Yu,Zhicai Zhang,Wenjuan Jiang,Zengsheng Ma

When mesoporous nanomaterials and graphene oxide have been used in anti-corrosion coatings, new nanocoatings have attracted greater attention. Adding nano-fillers with special structures and corrosion inhibitors to epoxy coatings has been proven to effectively enhance the corrosion resistance of coatings. However, studies have pointed out that the added corrosion inhibitors are easily degraded by UV light and react with the metal substrate or materials in the coating, resulting in a significant reduction in the service life of the coating. To this end, in this study, the corrosion inhibitor was encapsulated in mesoporous silica with pH response, and the functional silica was composited with graphene oxide to prepare novel graphene oxide/functional mesoporous silica nanoparticles. Coatings with this special filler added have strong anti-corrosion potential and can be applied in marine anti-corrosion contexts, such as containers, in the future. The filler not only has a physical barrier ability but also can effectively prevent the degradation of the inhibitor due to ultraviolet rays. At the same time, in the early stage of corrosion, the release of inhibitors can be effectively controlled by the change in PH to achieve the purpose of preventing corrosion. UV spectrophotometry confirmed the stable encapsulation and controlled release of the inhibitor. Electrochemical-impedance spectroscopy showed that the |Z|0.01 Hzvalue of the smart anti-corrosion epoxy coating was about 10,000 times higher than that of the pure epoxy coating. Through the FT-IR mapping test, it was found that in the area of mechanical damage, the alkaline environment created by the initial corrosion can induce the release of tannic acid and react with common corrosion products to form iron tannins, which effectively inhibits the further occurrence of corrosion. This method provides an effective method for the design of heavy-duty anti-corrosion coatings.Keywords:stimuli-responsive;micro/nano container;in situ polymerization;smart anticorrosion coating

Controlled release and antibacterial properties of PEO/casein nanofibers loaded with Thymol/β-cyclodextrin inclusion complexes in beef preservation

Jinming Dai,Wei Hu,Hongying Yang,Changzhu Li,Haiying Cui,Xiangzhou Li,Lin Lin

There are still many limitations in the application of natural active compounds in meat preservation. Herein, thymol was first inserted into the cavity of β -cyclodextrin ( β -CD) to form a stable inclusion complex (THY/ β -CD-IC). The computational investigation showed that the optimized complexation energy for THY/ β -CD-IC was −12.95 kcal mol −1 . It contributed to the improvement of the thermal stability of thymol in the inclusion compound. Furthermore, the functionalized nanofibers (THY/ β -CD-IC-NFs) loaded with THY/ β -CD-IC were successfully fabricated by electrospinning of the mixture of casein and polyethylene oxide. When dealing with protease-producing bacteria, controllable release of thymol from THY/ β -CD-IC-NFs was achieved through the response of casein to the hydrolysis of bacterial protease. The application results indicated that the prepared THY/ β -CD-IC-NFs had a long-term antimicrobial activity for chilled beef preservation during 7-days storage. The information from this study presents a feasible strategy for the development of natural extracts for use in meat preservation.

A Cu2O-based marine antifouling coating with controlled release of copper ion mediated by amphiphilic PLMA-b-PDMAEMA copolymers

Tongtong Ding,Luyao Xu,Xiangting Liu,Liang Ma,Yingxin Cui,Dalong Li,Xiuhua Sun,Changlu Gao

To achieve prolonged and sustainable release of copper ion in Cu 2 O-containing marine coatings, poly(lauryl methacrylate)-b-poly(2-( N , N -dimethylamino)ethyl methacrylate) (PLMA-b-PDMAEMA) diblock copolymers with different block ratios and molecular weights were synthesized by atom-transfer radical polymerization (ATRP) method. It was proved that the copolymers are able to coordinate with copper ion. And the surface wettability tests showed that the amphiphilic copolymers may undergo structure change in seawater. Taking advantage of the structure transformation and copper coordination abilities, the copolymer was employed to encapsulate Cu 2 O for the preparation of antifouling coatings. A controllable and continuous release of copper ion from Cu 2 O/copolymer coatings was then obtained. More importantly, it showed that higher PLMA/PDMAEMA block ratio exhibited better bactericidal rate. Actually, the bactericidal ability of the coating depends on the time duration of copper ion on the surface instead of the hydrolysis speed of the coating, in other words, the hydrophobic interaction of PLMA with the resin may play a major role in the antifouling performance of the marine coating. Self-polishing coatings Cu 2 O/copolymer/polycaprolactone (PCL) were then prepared and applied for marine field test, the antifouling performances were proved to be comparable to that of the commercially available self-polishing coatings.

Inhibition of ROS activity by controlled release of proanthocyanidins from mesoporous silica nanocomposites effectively ameliorates heterotopic ossification in tendon

Richun Liu,Bo Zhou,Hong Zhang,Yangwu Chen,Chunmei Fan,Tao Zhang,Tian Qin,Jie Han,Shichen Zhang,Xiao Chen,Weiliang Shen,Jiang Chang,Zi Yin

In the inflammatory environment of tendinopathy, tendon stem/progenitor cells (TSPCs) have been demonstrated to aberrantly differentiate into osteochondral lineage causing late-stage heterotopic ossification (HO) of the tendon, which severely damages tissue function and worsens patients’ activities. However, no effective treatment for tendinopathy currently exist because the mechanism underlying the TSPCs abnormal differentiation is unclear. In this article, high levels of reactive oxygen species (ROS) were found in tendinopathy samples. In vitro results revealed that proanthocyanidins (PC) can potentially regulate the differentiation of TSPCs by scavenging excess ROS, which would promote tenogenesis instead of chondrogenesis or osteogenesis. Moreover, in vivo results demonstrated that PC-loaded mesoporous silica nanocomposite (MSNs-PC) hinders the recurrence of HO in repaired tendons owing to the inhibition of oxidative stress, showing anti-inflammatory effects. Our findings indicate a high level of ROS in tendinopathy, which is probably the cause of aberrant differentiation of TSPCs. MSNs-PCs can effectively ameliorate HO of the tendon by scavenging excess ROS, providing a promising avenue to for other ROS-related diseases.

Rheological and pH dependent properties of injectable and controlled release hydrogels based on mushroom hyperbranched polysaccharide and xanthan gum

Ruquan Zhang,Qianru Xu,Yongzhen Tao,Xinyu Wang

Hydrogels with thixotropic and pH-sensitive properties exhibit injectablity and response to external stimulation. Thixotropic and pH-responsive hydrogels were fabricated from xanthan gum (XG) and a hyperbranched β -D-glucan (TM3a) crosslinked with sodium trimetaphosphate (STMP) for injectable and release-controlled application. Thixotropy and creep-recovery tests were carried out to determine injectable and viscoelastic properties of the XG-TM3a-STMP hydrogels. Introduction of TM3a and formation of chemically crosslinking structures resulted in higher yield stress (43.6–168.9 Pa) and elastic modulus (146.9–1245.7 Pa), lower retardation time (32.6–44.4 s), and higher recovery percentage (49.5–66.7%) for XG-TM3a-STMP compared to XG-STMP and XG-TM3a-water. FTIR spectra and SEM observation demonstrated that TM3a disassembled XG aggregates by partially breaking hydrogen bonds, resulting in a relatively thin wall of the grids for XG-TM3a-STMP. The XG-TM3a-STMP hydrogels exhibited pH dependant swelling property and controlled release behavior. The hybrid hydrogels have potential applications in 3D printing or injectable tissue engineering and drug release fields.
